Protective effect of camellia oil on H_(2)O_(2)-induced oxidative stress injury in H9C2 cardiomyocytes of rats

Objective: To explore the protective effect of camellia oil against H2O2-induced oxidative stress injury in rat H9C2 cardiomyocytes. Methods: CCK8 method was used to detect the cell survival rate of H9C2 cardiomyocytes treated with different concentrations of H2O2. Normal cultured cells were used as the blank control group, and the cells were treated with 200 μmol/L H2O2 for 24 h. An oxidative stress injury model was constructed as the model group. The cells were pretreated with 1%, 0.1% and 0.01% camellia oil for 24 h, and then H2O2 was added for 24 h as the experimental group. The β-galactosidase senescence staining assay, mitochondrial membrane potential assay, EdU cell proliferation staining assay and scratch assay were used to observe the changes of cell senescence, mitochondrial membrane potential, proliferation, apoptosis and migration in each group. The superoxide dismutase (SOD) activity, lactate dehydrogenase (LDH) activity, and malondialdehyde (MDA) content of the cells in each group were detected by using the kit. Results: The cell viability of H9C2 cardiomyocytes treated with different concentrations of H2O2 was inhibited and positively correlated with the concentration of H2O2 (P<0.01). Compared with the blank control group, the positive rate of cell senescence, MDA content and LDH activity increased in the H2O2 model group (P<0.01);mitochondrial membrane potential, cellular value-added rate, migration rate and SOD activity decreased (P<0.01). Compared with the H2O2 model group, the positive rate of cellular senescence (P<0.01 or P<0.05), MDA content and LDH activity decreased (P< 0.01 or P<0.05);mitochondrial membrane potential increased, cell proliferation rate and migration rate increased (P<0.01 or P<0.05) in the experimental group. Conclusion: Camellia oil can significantly inhibit oxidative stress injury in H9C2 cells and exert cardiomyocyte protective effects. 展开更多

The presentstudy was to investigate the gastroprotective effect of camellia oil against ethanol-induced gastric mucosal injuryin mice. The results showed that camellia oil pretreatment significantly reduced gastric ulcer injury. A remarkableinhibited oxidative stress by reducing the concentration of malondialdehyde and a significant decrease of thelevels of pro-inflammatory factors in gastric tissue were observed in camellia oil pretreated group. In addition,camellia oil improved the diversity of gut microbiota and changed the community structure and composition byincreasing Bacteriodes and Dorea. And the feces metabolomics found that metabolism of cofactors and vitamins,and lipid, carbohydrate and amino acid metabolism were modulated by admiration of camellia oil. Takentogether, the findings of this study suggested that camellia oil could ease the ethanol-induced gastric mucosalinjury via the improvement of anti-oxidant and anti-inflammatory status, as well as the regulation of gutmicrobiota and its metabolites. 展开更多

Administration of camellia oil( 4. 0 and 8. 0 mL/kg/day) significantly shortened the defecation time,increased defecation mass and fecal water content,with efficacies comparable to that of hemp seed oil. Intestinal propulsion test showed that camellia oil significantly enhanced the propelling rates of the charcoal powder in the intestine. Meanwhile,camellia oil also significantly decreased the serum levels of nitric oxide( NO),nitric oxide synthase( NOS) and vasoactive intestinal peptide( VIP) and increased serum substance P in costive animals. These results suggested that camellia oil can largely relieve slowtransit constipation and enhance the gastrointestinal motility through modulation of serum gastrointestinal motility key factors such as NO,NOS and VIP and substance P. 展开更多

In this paper, the crude camellia oil extracted by cold pressing and deacidified oil, decolorized oil, dewaxed oil and finished tea oil obtained in different refining stages were studied. The changes of acid value, peroxide value and free radical scavenging ability of camellia oil in each stage were compared and analyzed.The results showed that the total phenol content of crude oil was the highest, and the total phenol content of alkali refined crude oil decreased significantly. After dewaxing, the total phenol concentration of camellia oil increased by 63.5%. The reason may be that low temperature conditions contributed to the recovery of phenolic structure in camellia oil, and without the influence of alkaline solution, the damage degree of phenolic substances was greatly reduced, thus elevating the total phenol content. The lowest peroxide value of camellia oil was observed in the alkali-refining deacidification stage, which decreased by 29.3%. Crude oil had the highest acid value and concentration of antioxidant substances. After alkali-refining deacidification,the acid value and the concentration of antioxidant substances dropped by 85.3%and 88.9%, respectively. Total phenols had the strongest correlation with acid value,followed by free radical scavenging ability, and the correlation with peroxide value was not obvious. 展开更多

It was tried to microencapsulate camellia oil using heterocoagulation between fatty acid dissolved in camellia oil and chitosan dissolved in the continuous water phase. Oleic acid as a fatty acid was dissolved in camellia oil in order to certainly form the microcapsule shell made from oleic acid and chitosan. The microcapsules were observed with optical microscope and characterized about the diameters, ζ-potential, FTIR analysis and adhesion feature on human hair. Microcapsules with the mean diameter in the range from ca. 1.5 μm to 4.5 μm could be prepared with the preparation method presented in this study. The oil droplets of camellia oil charged negatively to be -54.6 mV and the microcapsules charged positively to be 59.6 mV. The microcapsules adhered well on the negatively charged human hair and were kept stably before and after drying at room temperature for 24 h and blowing. 展开更多

Camellia oil is an edible vegetable oil with high value of nutrition and health protection function such as antioxidant and adjusting blood fat. In this study, a simple, rapid and effective HPTLC method was developed for analyzing the composition and antioxidant constituents of camellia oil. The HPTLC was performed on G60 plate with n-hexane-diethyl ether-acetic acid (6:4:0.1, v/v/v) as mobile phase combined with two coloration methods (ethanol containing 10% phosphomolybdic acid, ethanol containing 0.03% DPPH) and scanning densitometry technique. The unsaturated fatty glyceride, free fatty acids, sterols and lipids including triolein, oleic acid, ergosterin, β-sitosterol, tocopherol and phospholipids in camellia oils were determined and performed densitometrically at λs1 = 620 nm and λs2 = 517 nm. The results show that the main components of different samples of camellia oil are similar, however the contents are diverse. The antioxidative test shows that camellia oil has obvious antioxidant capability as olive oil, especially the pressed virgin oil. Therefore, this non-derivatization HPTLC method can be used for composition and antioxidative capacity determination of camellia oils. 展开更多

Plasticizers are essential additives in the processing of polyvinyl chloride(PVC),with phthalate plasticizers being widely used.However,these conventional plasticizers have been shown to be harmful to human health and environmentally unfriendly,necessitating the exploration of eco-friendly bio-based alternatives.In this study,Camellia oleifera seed oil,a specialty resource in China,was utilized as a raw material and reacted with 4,4′-Methylenebis(N,N-diglycidylaniline)(AG-80)to synthesize Phenyl Camellia seed Oil Ester(PCSOE).PCSOE was employed as a plasticizer to prepare modified PVC films with varying concentrations,with the conventional plasticizer dioctyl phthalate(DOP)serving as a control.Experimental results demonstrate that PSCOE-plasticized PVC films exhibit enhanced hydrophilicity,tensile strength,and thermal stability compared to DOP-modified PVC films.The contact angle of PSCOE-plasticized PVC films ranges from 66.26°to 78.48°,which is generally lower than the contact angle of DOP-modified PVC films at 78.40°,indicating improved hydrophilicity due to the modification with PCSOE.The tensile strength of PSCOE-plasticized PVC films ranges from 17.73 to 20.17 MPa,all surpassing the value of 16.41 MPa for DOP-modified PVC films.Moreover,the temperatures corresponding to 5%,10%,and 50%weight loss for PVC samples modified with PCSOE are higher than those for DOP.Hence,PCSOE presents a viable alternative to DOP as a plasticizer for PVC materials. 展开更多

Camellia oil and olive oil with superior economic value are easily adulterated with other,cheaper oils.It is difficult to identify both oils by traditional methods because of their similar fatty acid profiles.In the present study,a novel method for qualitative and quantitative analysis ofβ-sitosterol using GC/MS and GC/FID was developed to identify camellia oil and olive oil.The method validation ofβ-sitosterol analysis showed good linearity and satisfactory values for recovery,accuracy,precision,and repeatability.The linear regression coefficient(R^(2))of the calibration curve was 0.9985.An acceptable limit of detection(0.36 mg/100 g)and limit of quantification(1.20 mg/100 g)were achieved.The spiked recoveries were 95.0%to 100.3%.The relative standard deviation(RSD)of within-day precision was less than 3.26%,and the RSD of retention times and peak areas for repeatability were within 0.03%and 1.08%,respectively.The contents ofβ-sitosterol in virgin camellia oil and virgin olive oil were in the range of 14.1–30.2 mg/100 g and 94.3–173.2 mg/100 g,respectively,indicating that theβ-sitosterol content in the former is seven times that in the latter,andβ-sitosterol could be a potential marker for the authentication and adulteration detection of both oils. 展开更多

In this study,as the plasticizer,Camellia oleifera seed-oil-based cyclohexyl ester(COSOCE)was prepared by the reaction of cyclohexene oxide and refined C.oleifera seed oil(RCOSO)obtained by acidification hydrolysis after saponification.In addition,the structure of the target product was confirmed by Fourier transform infrared(FTIR)spectroscopy,nuclear magnetic resonance(NMR)spectroscopy,and Raman spectroscopy.COSOCE was used as plasticizer-modified polyvinyl chloride(PVC)membranes.The structure of the COSOCE-modified PVC membranes were characterized by Raman spectroscopy and scanning electron microscopy(SEM).The properties of the COSOCE-modified PVC membrane were characterized by contact angle measurements,universal testing machine,thermogravimetric analysis(TGA),and differential scanning calorimetry(DSC).The results revealed that(1)The COSOCE-modified PVC membranes exhibit a good microscopic morphology.Combined with energy-dispersive X-ray spectroscopy(EDS)and contact angle measurement results,the COSOCE-modified PVC membranes are confirmed to be a hydrophilic material.(2)The modified PVC membrane with 60%COSOCE exhibited the best mechanical properties.The tensile strength reached 23.56±2.94 MPa.(3)COSOCE-modified PVC material exhibited better thermal stability,with a loss rate of less than 75%at the end of the first decomposition stage.Compared with that of the dioctyl-phthalate(DOP)-modified PVC membrane,the initial decomposition temperature of PVC was increased by 1.17°C–8.17°C,and the residual rate was increased by 0.67%–5.75%.The carbon–carbon double bond in the COSOCE molecular structure can remove the free radicals generated during the degradation of PVC material and slow down the decomposition rate of PVC.In addition,the double bond can be cross-linked partially with the PVC molecular chain containing the conjugated polyene structure,thereby increasing the movement resistance of the PVC molecular chain segment.Hence,COSOCE can replace DOP as a PVC plasticizer. 展开更多

Camellia oil has become an important plant oil in China in recent years,but its effects on non-alcoholic fatty liver disease(NAFLD)have not been documented.In this study,the effects of camellia oil,soybean oil,and olive oil on NAFLD were evaluated by analyzing the fatty acid profiles of the plant oils,the serum lipids and lipoproteins of rats fed different oils,and by cytological and ultrastructural observation of the rats’hepatocytes.Analysis of fatty acid profiles showed that the polyunsaturated fatty acid(PUFA)n-6/n-3 ratio was 33.33 in camellia oil,12.50 in olive oil,and 7.69 in soybean oil.Analyses of serum lipids and lipoproteins of rats showed that the levels of total cholesterol and low-density lipoprotein cholesterol in a camellia oil-fed group(COFG)were lower than those in an olive oil-fed group(OOFG)and higher than those in a soybean oil-fed group(SOFG).However,only the difference in total cholesterol between the COFG and SOFG was statistically significant.Cytological observation showed that the degree of lipid droplet(LD)accumulation in the hepatocytes in the COFG was lower than that in the OOFG,but higher than that in the SOFG.Ultrastructural analysis revealed that the size and number of the LDs in the hepatocytes of rats fed each of the three types of oil were related to the degree of damage to organelles,including the positions of nuclei and the integrity of mitochondria and endoplasmic reticulum.The results revealed that the effect of camellia oil on NAFLD in rats was greater than that of soybean oil,but less than that of olive oil.Although the overall trend was that among the three oil diets,those with a lower n-6/n-3 ratio were associated with a lower risk of NAFLD,and the effect of camellia oil on NAFLD was not entirely related to the n-6/n-3 ratio and may have involved other factors.This provides new insights into the effect of oil diets on NAFLD. 展开更多

Comparison and analysis of physicochemical properties and oxidative stability of 10 brands of camellia oils were conducted in this study.Results showed that more attention should be paid to the iodine(IV),saponification(SV)and peroxide(PV)values of camellia oils during sampling inspection as they were more likely to be out the range of quality standards.Regression analysis between physicochemical indices and oxidative stability showed individual index could not affect the stability of camellia oils significantly(p>0.05).However,very high correlations were found between physical indices such as optical rotation(OR)and turbidity(R=-0.929),turbidity and color(R=-0.930).High correlations were found between chemical indices such as IV and moisture and volatile matter(MVM)(R=-0.853),IV and PV(R=0.831),MVM and PV(R=-0.809).Package with nitrogen could retard the oxidation of camellia oil.These results may be useful for rapid evaluation,differentiation and quality improvement of camellia oils. 展开更多

To determine the age of oil-tea camellia trees, regression equations including Logistic, Mitscherlich, Gompertz, Korf, and Richards were used to calculate accumulative growth rate using basal trunk disc and investigate the relations between the age of oil-tea camellia trees and their growth rate of secondary trunk. The Gompertz equation Y=71.296 1exp (-3.874 4exp (-0.006 4t)) was the most optimal equation to simulate the accumulative growth rate of basal trunk disc. This equation could be used to estimate the age of oil-tea camellia trees that grow under similar environmental conditions. The Korf equation Y=576.900 1exp (-4.153 0x -0.314 2 ) was the best equation to describe the relation between the age and growth rate of different secondary trunks. With the adjustment coefficient and average growth of different secondary trunk discs, it is possible to predict the age of ancient oil-tea camellia trees that grow under similar environmental conditions. In addition, taking three or more discs from the same diameter group and calculating their average growth rate could lead to more accurate results. For trees that grow in different areas, environmental conditions should be carefully considered when using the above two equations to predict the age of ancient oil-tea camellia trees. 展开更多

This paper studied effects of different picking time and different geographical provenances on oil content of Camellia oleifera. The results showed that different picking time had significant effects on the oil content of cold dew seeds and frost's descent seeds. With the delay of picking time,the oil content of cold dew seeds,frost's descent seeds,dry seeds and oil content of seed kernels were significantly increased.There was a significantly positive correlation between the oil content of fresh fruit of cold dew seeds and frost's descent seeds and the dry seed yield of fresh seeds and the oil content of fresh fruit( P < 0. 05),and the correlation coefficient was greater than 0. 85. Besides,the oil content of seed kernels of cold dew seeds was closely correlated with the seed yield of fresh seeds,oil content of fresh fruit,and oil content of fresh seeds. In the process of C. oleifera breeding,the thin coat,high seed yield and high oil content can be taken as the key research directions for future breeding. The oil content of ordinary C. oleifera seeds in 18 counties( cities) was 36. 42%-63. 33%,indicating that there were significant differences in the oil content of C. oleifera in different geographical provenances. In conclusion,according to the study of different picking time,the recommended picking time of cold dew seeds in Hunan area is around October 10,while the best picking time of frost's descent seeds is about October 30. Objectives:Harvesting fruits prematurely is a serious problem in many Camellia oleifera‘Huaxin’orchards.Systematic views of dynamic alterations in central metabolism associated with the oil content and oil quality of this cultivar at different harvesting dates were investigated.Materials and Methods:Oil trait analysis was performed in conjunction with data-independent acquisition proteomics analysis using kernels harvested at three time points.Samples were taken from different years and locations to verify oil trait changes,and parallel reaction monitoring(PRM)was performed for some pivotal proteins to validate changes in their abundance.Results:Samples harvested at 6 d after the recommended harvesting date had the highest contents and best fatty acid composition of Camellia oil.A total of 614 differentially accumulated proteins were detected in three pairwise comparisons.Six days prior to the recommended harvesting date,the maximum expression of ribonucloprotein,helicase ATP-binding domain-containing protein,and glycerol kinase and the highest seed moisture content were observed.The highest mitogen-activated protein kinase 6,very-long-chain 3-oxoacyl-CoA reductase and stearoyl-CoA desaturase expression appeared on the recommended harvesting date.A delayed effect was observed for proteomic and oil trait data.Oil trait and PRM validations indicated that the results were highly reliable.Conclusions:The current data strongly suggest that in Guizhou Province,‘Huaxin’cannot be harvested earlier than the recommended harvesting date;appropriately delaying the harvesting time can improve the ultimate production and quality of Camellia oil. 展开更多

拉曼光谱对茶油三元体系掺伪检测研究

该研究采用拉曼光谱技术对茶油三元体系掺伪进行定量检测研究,通过对比不同预处理方法、建模方法及优化算法的优劣,确定最优的大豆油、玉米油、茶油的多元掺伪检测模型。利用一阶微分、二阶微分、多元散射矫正、标准正态变换等不同预处... 该研究采用拉曼光谱技术对茶油三元体系掺伪进行定量检测研究,通过对比不同预处理方法、建模方法及优化算法的优劣,确定最优的大豆油、玉米油、茶油的多元掺伪检测模型。利用一阶微分、二阶微分、多元散射矫正、标准正态变换等不同预处理方法消除外界因素对光谱的影响,竞争性自适应重加权算法提取特征光谱波段,通过偏最小二乘回归和支持向量机建立茶油掺伪检测模型,分别采用网格搜索法和粒子群算法对支持向量机进行优化。基于标准正态变换预处理后所建立模型效果最佳,大豆油和茶油的最佳预测模型为基于粒子群算法优化的支持向量机,玉米油的最佳预测模型为基于网格搜索法优化的支持向量机,大豆油、玉米油和茶油的预测集决定系数R2和预测均方根误差分别为0.9986、0.9994、0.9999和2.73%、1.62%、0.40%。该研究确定了最优的大豆油、玉米油、茶油的多元掺伪检测模型,针对市场茶油的掺伪检测,基于拉曼光谱分析和优化算法的支持向量机模型为茶油的无损快速定量检测提供了一定的参考和借鉴。 展开更多

加热氧化对山茶油热力学及流变学特性的影响

目的探究加热氧化对山茶油热力学及流变学特性的影响。方法山茶油经加热氧化处理后,运用差示扫描量热技术(differential scanning calorimetry,DSC)、旋转流变技术,研究其DSC氧化稳定性,熔融结晶特性及黏度随剪切速率、温度变化等特性... 目的探究加热氧化对山茶油热力学及流变学特性的影响。方法山茶油经加热氧化处理后,运用差示扫描量热技术(differential scanning calorimetry,DSC)、旋转流变技术,研究其DSC氧化稳定性,熔融结晶特性及黏度随剪切速率、温度变化等特性。结果山茶油加热时间越长,其DSC热力学氧化稳定性越低,当加热120 min后较初始山茶油的氧化起始温度降低了8.3℃,加热氧化后它的熔融温度及熔融焓降低,主要结晶峰消失。山茶油加热氧化后流动性变差,表现为黏度升高,加热120 min的山茶油较初始山茶油黏度提高了2.8倍,山茶油氧化程度越高,其黏度随温变化的速率越快。结论加热氧化降低了山茶油的热力学稳定性及流变学稳定性,本研究为山茶油精深加工的热力学性能及流变性能提供了一定的理论支撑。 展开更多

目的:基于超高效液相色谱-四极杆飞行时间质谱(ultra-high performance liquid chromatography-quadrupole time-of-flightmassspectrometry,UPLC-Q/TOF-MS)技术,利用代谢组学方法探讨山茶油干预的阿尔茨海默病(Alzheimer’s disease,AD... 目的:基于超高效液相色谱-四极杆飞行时间质谱(ultra-high performance liquid chromatography-quadrupole time-of-flightmassspectrometry,UPLC-Q/TOF-MS)技术,利用代谢组学方法探讨山茶油干预的阿尔茨海默病(Alzheimer’s disease,AD)小鼠尿液的代谢组学变化。方法:将小鼠随机分为正常对照组、模型组、阳性药组、山茶油组,除对照组外其余各组小鼠均由β-淀粉样蛋白25-35肽诱导AD,并取尿液样本。尿液沉淀蛋白后进行UPLC-Q/TOF-MS分析,获得小鼠尿液代谢物数据,使用SIMCA-P14.1软件进行主成分分析(principalcomponent analysis,PCA)和正交偏最小二乘判别分析,使用HMBD、MassBank、MetaboAnalyst、京都基因与基因组百科全书在线数据库对生物标志物进行鉴定、通路富集分析和代谢网络的构建。结果:在PCA得分图中,山茶油组样本点远离模型组样本点区域,更接近正常对照组样本点区域。鉴定得到48个生物标志物,主要涉及的通路有牛磺酸和次牛磺酸代谢,精氨酸和脯氨酸代谢,半胱氨酸和蛋氨酸代谢,丙氨酸、天冬氨酸和谷氨酸代谢等。结论:山茶油通过调节能量代谢、氨基酸代谢等改善了AD小鼠体内代谢,为山茶油干预AD的进一步研究提供参考。 展开更多
