Laboratory bioassays were performed to determine the lethal doses and lethal time of pyrethrins (a botanical insecticide) on 4th instar larvae of spruce bud-worm using larval dip assay. Results show that the LT50 values(time of 50% larval mortality)for spruce budworm at the pyrethrins concentra-tions of 12.5, 25, 50, 100, and 200 μg·L-1 were 94.78, 45.54, 20.36, 14.39 and 11.37 h, respectively. The percentage of cumulative mortality at the pyrethrins concentrations of 12.5, 25, 50, 100, 200 μg·L-1 was approxi-mately 50%, 67%, 93%, 100% and 100% within 120 h, respectively. The LC50 value (concentration of 50% larval mortality) for the 4th instar lar-vae was 16.1μg·L-1. Thus, larval mortality of spruce budworm increased in a concentration-dependent manner, and lethal time decreased with increasing pyrethin concentrations. These findings suggest that pyrethrins have a potential in controlling spruce budworm populations.展开更多
The ichnovirus TrIV, transmitted by the endoparasitic wasp Tranosema rostrale to its lepidopteran host during oviposition, replicates asymptomatically in wasp ovaries and causes physiological dysfunctions in parasitiz...The ichnovirus TrIV, transmitted by the endoparasitic wasp Tranosema rostrale to its lepidopteran host during oviposition, replicates asymptomatically in wasp ovaries and causes physiological dysfunctions in parasitized caterpillars. The need to identify ichnoviral genes responsible for disturbances induced in lepidopteran hosts has provided the impetus for the sequencing and annotation of ichnovirus genomes, including that of TrIV. In the latter, 86 putative genes were identified, including 35 that could be assigned to recognized ichnoviral gene families. With the aim of assessing the relative importance of each TrIV gene, as inferred from its level of expression, and evaluating the accuracy of the gene predictions made during genome annotation, the present study builds on an earlier qPCR quantification of transcript abundance of TrIV rep ORFs, in both lepidopteran and wasp hosts, extending it to other gene families as well as to a sample of unassigned ORFs. We show that the majority (91%) of putative ORFs assigned to known gene families are expressed in infected larvae, while this proportion is lower (67%) for a sample taken among the remaining ORFs. Selected members of the TrV and rep gene families are shown to be transcribed in infected larvae at much higher levels than genes from any other TrIV gene family, pointing to their likely involvement in host subjugation. In wasp ovaries, the transcriptional profile is dominated by a rep gene and a member of a newly described gene family encoding secreted proteins displaying a novel cysteine motif, which we identified among previously unassigned ORFs.展开更多

A leaf disc method used to evaluate the impact of pesticides on leafrollers resulted in high levels of leaf-abandonment of first and second instar larvae. Inserting leaf petioles into water soaked Oasis foam resulted in a significant reduction in overall leaf weight and diminished the appearance of leaf quality. When leaf petioles were inserted into water-filled Eppendorf vials, leaf weight increased and overall leaf quality appeared to be good. We opted to adopt this latter whole-leaf bioassay method, which though very time consuming, provided a robust assessment of OBLR life history parameters against different apple genotypes. Across all apple genotypes evaluated, male larvae developed faster than females to pupation as well as to adulthood, while females developed faster as pupae and were heavier than male pupae. OBLR larvae reared on leaves from different apple genotypes displayed significant variability in development time, pupal weight, larval survivorship, and number of offspring produced. We utilized three indices to characterize host acceptance, sub-lethal effects, and lethal effects of OBLR to apple genotypes. Our results indicated that ‘Lady’, ‘Viking’, and ‘Northern Spy’ show some of the highest levels of resistance against OBLR, while ‘Granny Smith’ was quite susceptible to OBLR and could be used in future studies identifying apple genotypes with relative resistance to OBLR.展开更多

However, they are also known to act as long-range insect attractants, as they are volatile and permeate forest airspaces. Moreover, they are lipid soluble and can be absorbed into plant epicuticular waxes. We test their role in short-range host plant choice by both adult females and larvae of a folivorous forest pest (Choristoneura fumiferana). We conducted laboratory assays testing the responses of Eastern spruce budworm to an artificial monoterpene mix (α-pinene, β-pinene, limonene, myrcene) and to white spruce (Picea glauca) epicuticular waxes in closed arenas. Ovipositing females pre- ferred filter paper discs treated with P. glauca waxes to controls, and preferred the waxes + monoterpenes treatment to waxes alone. However, females showed no preference between the monoterpene-treated disc and the control when presented without waxes. Feeding larvae prefered wax discs to control discs. They also consumed discs treated with realistic monoterpene concentrations and wax preferentially over wax-only discs, but showed no preference between extremely high monoterpene concentrations and wax-only controls. In an insect-free assay, P. glauca epicuticular wax decreased monoterpene volatilization. These results suggest that P. glauca waxes and realistic concentrations of monoterpenes are stimulatory to both egg-laying females and feeding larvae, and that their effects are synergistic.展开更多
