Three kinds of new type solid strong acid catalysts S2O 2-8/ZrO2-Al2O3-M2O3(M=Cr,Ce,La) were prepared. Their crystal structure, surface area, acid strength and sulfur content were determined by means of XRD, BET, flow Hammett indicator method and chemical analysis. Their catalytic activities in esterification reaction of acetic acid with n-butanol were studied. The results showed that ZrO2 in the catalysts mainly in tetragonal phase and few in monoclinic phase. The tetragonal phase of ZrO2 and S2O 2-8 are the key factors that guarantee the catalytic activity. Incorporation of appropriate amounts of metallic oxides(Cr2O3,Ce2O3,La2O3) into the catalyst favors the stabilization of sulfur species and surface area, which increase the activity sites on the catalyst. The experimental results showed that three catalysts S2O 2-8/ZrO2-Al2O3(2%)-M2O3(1%)(M=Cr,Ce,La) had higher catalytic activity in mentioned esterification, with the conversion of acetic acid reached 96.8%, 95.7% and 96.1%, respectively. The preparation condition of the catalysts showed great influence on the catalytic activity. 展开更多

Autism spectrum disorders are a group of neurodevelopmental disorders involving more than 1100 genes,including Ctnnd2 as a candidate gene.Ctnnd2knockout mice,serving as an animal model of autis m,have been demonstrated to exhibit decreased density of dendritic spines.The role of melatonin,as a neuro hormone capable of effectively alleviating social interaction deficits and regulating the development of dendritic spines,in Ctnnd2 deletion-induced nerve injury remains unclea r.In the present study,we discove red that the deletion of exon 2 of the Ctnnd2 gene was linked to social interaction deficits,spine loss,impaired inhibitory neurons,and suppressed phosphatidylinositol-3-kinase(PI3K)/protein kinase B(Akt) signal pathway in the prefrontal cortex.Our findings demonstrated that the long-term oral administration of melatonin for 28 days effectively alleviated the aforementioned abnormalities in Ctnnd2 gene-knockout mice.Furthermore,the administration of melatonin in the prefro ntal cortex was found to improve synaptic function and activate the PI3K/Akt signal pathway in this region.The pharmacological blockade of the PI3K/Akt signal pathway with a PI3K/Akt inhibitor,wo rtmannin,and melatonin receptor antagonists,luzindole and 4-phenyl-2-propionamidotetralin,prevented the melatonin-induced enhancement of GABAergic synaptic function.These findings suggest that melatonin treatment can ameliorate GABAe rgic synaptic function by activating the PI3K/Akt signal pathway,which may contribute to the improvement of dendritic spine abnormalities in autism spectrum disorders. 展开更多

Hypoxia promotes proliferation and differentiation of neural stem cells from embryonic day 12 rat brain tissue, but the concentration and time of hypoxic preconditioning are controversial. To address this, we cultured neural stem cells isolated from embryonic day 14 rat cerebral cortex in 5% and 10% oxygen in vitro. MTT assay, neurosphere number, and immunofluorescent staining found that 5% or 10% oxygen preconditioning for 72 hours improved neural stem cell viability and proliferation. With prolonged hypoxic duration (120 hours), the proportion of apoptotic cells increased. Thus, 5% oxygen preconditioning for 72 hours promotes neural stem cell prolif- eration and neuronal differentiation. Our findings indicate that the optimal concentration and duration of hypoxic preconditioning for promoting proliferation and differentiation of neural stem cells from the cerebral cortex are 5% oxygen for 72 hours. 展开更多

目的研究全饥饿状态下大鼠皮质细胞凋亡相关因子Bcl-2、Bax和Caspase-3表达变化情况。方法雄性成年SD大鼠60只,随机分为6组:正常对照组、饥饿3 d、5 d、7 d、9 d和11 d组。采用TUNEL法检测大鼠皮质神经细胞凋亡情况,RT-PCR法检测皮质细... 目的研究全饥饿状态下大鼠皮质细胞凋亡相关因子Bcl-2、Bax和Caspase-3表达变化情况。方法雄性成年SD大鼠60只,随机分为6组:正常对照组、饥饿3 d、5 d、7 d、9 d和11 d组。采用TUNEL法检测大鼠皮质神经细胞凋亡情况,RT-PCR法检测皮质细胞凋亡相关因子Bcl-2、Bax、Caspase-3 mRNA的表达情况,Western blot法检测Bcl-2、Bax及Caspase-3蛋白表达。结果与正常对照组比较,饥饿5 d组皮质区凋亡细胞数增多(P<0.05),7 d、9 d、11 d组均明显增多(P<0.01);饥饿5 d、7 d、9 d、11 d组脑皮质中Bax、Caspase-3表达均上调(P<0.05),7 d、9 d、11 d组较为显著(P<0.01);饥饿3 d、5 d、7 d组Bcl-2表达均上调(P<0.01),5 d组表达量最多,饥饿9 d、11 d组Bcl-2表达下降,11 d组明显下降(P<0.01)。结论短期饥饿应激可能有一定的脑保护作用;长期饥饿应激可能通过上调大鼠皮质细胞Bax、Caspase-3的表达,抑制Bcl-2的表达,破坏了Bcl-2/Bax的平衡,进而诱导细胞凋亡。 展开更多

Effect of 2-m Temperature Data Assimilation in the CMA-MESO 3DVAR System 被引量:1

Assimilation of surface observations including 2-m temperature(T_(2m))in numerical weather prediction(NWP)models remains a challenging problem owing to differences between the elevation of model terrain and that of actual observation stations.NWP results can be improved only if surface observations are assimilated appropriately.In this study,a T_(2m)data assimilation scheme that carefully considers misrepresentation of model and station terrain was established by using the three-dimensional variational data assimilation(3DVAR)system of the China Meteorological Administration mesoscale model(CMA-MESO).The corresponding forward observation operator,tangent linear operator,and adjoint operator for the T_(2m)observations under three terrain mismatch treatments were developed.The T_(2m)data were assimilated in the same method as that adopted for temperature sounding data with additional representative errors,when station terrain was 100 m higher than model terrain;otherwise,the T_(2m)data were assimilated by using the surface similarity theory assimilation operator.Furthermore,if station terrain was lower than model terrain,additional representative errors were stipulated and corrected.Test of a rainfall case showed that the observation innovation and analysis residuals both exhibited Gaussian distribution and that the analysis increment was reasonable.Moreover,it was found that on completion of the data assimilation cycle,T_(2m)data assimilation obviously influenced the temperature,wind,and relative humidity fields throughout the troposphere,with the greatest impact evident in the lower layers,and that both the area and the intensity of rainfall were better forecasted,especially for the first 12hours.Long-term continuous experiments for 2–28 February and 5–20 July 2020,further verified that T_(2m)data assimilation reduced deviations not only in T_(2m)but also in 10-m wind forecasts.More importantly,the precipitation equitable threat scores were improved over the two experimental periods.In summary,this study confirmed that the T_(2m)data assimilation scheme that we implemented in the kilometer-scale CMA-MESO 3DVAR system is effective. 展开更多

OBJECTIVE Glutamatergic projections from prefrontal cortex(PFc) to nucleus accumbens(NAc) regulate the dopamine(DA) release in NAc.However,it is not clear whether this circuit is effective for the reward and motivation of heroin addiction.Our study investigates the effects of metabotropic glutamate receptor 2/3(mGluR2/3) and the projections from ventromedial prefrontal cortex(vmPFc) to the NAc shell on the reward and motivation of heroin-addicted rats.METHODS First,rats were trained to selfadministration for 14 d.On the 15 thday,parts of rats were injected with mGluR 2/3 agonist LY379268(0.1,0.3 and 1.0 mg·kg-1,ip) systematically and another parts of rats were bilaterally microinjected with LY379268(0.3 and 1.0 g·L^(-1))at the volume of 0.5 μL into the ventral tegmental area(VTA),NAc core or NAc shell,respectively.All rats were followed by heroin self-administration testing under fixed ratio 1(FR1) schedule or progressed ratio(PR) schedule to observe the effect of LY379268 on the heroin reward or motivation.Second,rats were injected chemogenetic glutamatergic virus(pAAV-CaMKIIa-hM3 D(Gq)-mCherry or pAOV-CaMKIIa-hM4 D(Gi)-mCherry-3 Flag) or negative control virus in vmPFc,and trained to heroin self-administration for 14 d.On the 15 thday,rats were bilateral y microinjected with clozapine-N-oxide(CNO,1 mmol·L^(-1),0.5 μL) into NAc shell and tested the effect on the heroin reward or motivation.Finally,rats were injected optogenetical glutamatergic virus(AAV2/9-CaM KⅡ-hChR2-EYFP) or negative control virus in vmPFc,implanted 16 channel photoelectrode in ipsilateral NAc shell,and trained to heroin selfadministration for 14 d.On the 15 thday,rats were tested heroin reward under FR1 procedure with blue light stimulation in the wavelength of470 nm,frequency of 25 HZ and power of 5 mW.Each stimulation lasting for 1 h and interval for1 h.The spike changes before and after stimulation in NAc Shel neural nerve was recorded.RESULTS LY379268 cloud dose-dependent attenuated the heroin reward or motivation and the local effective site was mainly in the NAc shell.Chemogenetic results showed activation or inactivation the projection from vmPFc to NAc shell enhanced or attenuated the heroin reward and motivation,respectively.Optogenetical stimulation the same projection also enhanced the heroin reward,and a tonic neuronal firing at the nerve of NAc shell was observed during the light stimulation session.CONCLUSION mGluR2/3 activation in the NAc shell is involved in the inhibition of heroin reward and motivation.Activation the projection from PFc to NAc shell can enhance the effects on heroin reward and motivation. 展开更多
