Deep foundations are currently used in engineering practice to solve problems caused by weak geotechnical characteristics of the ground.Impact pile driving is an interesting and viable solution from economic and techn...Deep foundations are currently used in engineering practice to solve problems caused by weak geotechnical characteristics of the ground.Impact pile driving is an interesting and viable solution from economic and technical points of view.However,it is necessary to ensure that the environmental drawbacks,namely ground-borne vibration,are adequately met.For this purpose,the authors propose an axisymmetric finite element method-perfectly matched layer(FEM-PML)approach,where the nonlinear behavior of the soil is addressed through an equivalent linear methodology.Given the complexity of the problem,an experimental test site was developed and fully characterized.The experimental work comprised in-situ and laboratory soil characterization,as well as the measurement of vibrations induced during pile driving.The comparison between experimental and numerical results demonstrated a very good agreement,from which it can be concluded that the proposed numerical approach is suitable for the prediction of vibrations induced by impact pile driving.The experimental database is available as supplemental data and may be used by other researchers in the validation of their prediction models.展开更多

This article investigates the characteristics of shock wave overpressure generated by multi-layer composite charge under different detonation modes.Combining dimensional analysis and the explosion mechanism of the cha...This article investigates the characteristics of shock wave overpressure generated by multi-layer composite charge under different detonation modes.Combining dimensional analysis and the explosion mechanism of the charge,a peak overpressure prediction model for the composite charge under singlepoint detonation and simultaneous detonation was established.The effects of the charge structure and initiation method on the overpressure field characteristics were investigated in AUTODYN simulation.The accuracy of the prediction model and the reliability of the numerical simulation method were subsequently verified in a series of static explosion experiments.The results reveal that the mass of the inner charge was the key factor determining the peak overpressure of the composite charge under single-point detonation.The peak overpressure in the radial direction improved apparently with an increase in the aspect ratio of the charge.The overpressure curves in the axial direction exhibited a multi-peak phenomenon,and the secondary peak overpressure even exceeded the primary peak at distances of 30D and 40D(where D is the charge diameter).The difference in peak overpressure among azimuth angles of 0-90°gradually decreased with an increase in the propagation distance of the shock wave.The coupled effect of the detonation energy of the inner and outer charge under simultaneous detonation improved the overpressure in both radial and axial directions.The difference in peak overpressure obtained from model prediction and experimental measurements was less than 16.4%.展开更多
To analyze the spring disturbance torque caused by motional cable harness in a stabilized platform,the Kirchhoff theory based cable harness model has been previously developed to dynamically simulate the motional cabl...To analyze the spring disturbance torque caused by motional cable harness in a stabilized platform,the Kirchhoff theory based cable harness model has been previously developed to dynamically simulate the motional cable harness.In this paper,this model was validated by comparing the simulation results with the experiment results(both the spring force and the deformed profile of the motional cable harness).In the experiment,a special optical measuring instrument based on binocular vision was developed and the motion and deformation of cable harness were measured.A simplified stabilized platform system was constructed,and the absolute value of spring disturbance force during the motion of this simplified frame was obtained by using a force gauge(0.02N precision).The physical parameters of experimental specimen were also measured.The experimental and simulated results showed good agreement.These results should be useful for better motional cable harness layout design and reliable evaluation of the spring disturbance torque.展开更多
The current research on the evaporation of ferrofuids mainly focuses on the characterization of ultra-low vapor pressure ferrofuids in vacuum and the theoretical analysis of the evaporation process.Few studies have fo...The current research on the evaporation of ferrofuids mainly focuses on the characterization of ultra-low vapor pressure ferrofuids in vacuum and the theoretical analysis of the evaporation process.Few studies have focused on the experimental validation of the proposed evaporation rate equations and on the comparison of the diferences in ferrofuid evaporation.In this study,based on the Bolotov’s model,an evaporation rate equation is deduced from the experimental model.The experimental study included a comparison of the evaporation,magnetic particle volume fraction,temperature,height of the fuid surface from the outlet,and magnetic feld of a kerosene-based ferrofuid and its base carrier liquid.The prepared sample was evaporated in a test tube,and the evaporation rate was calculated by measuring the weight loss of the sample.The experimental results show that the evaporation rate of the base carrier liquid is higher than that of the ferrofuid.The smaller the volume fraction of the magnetic particles,the greater the evaporation rate.The magnetic particles play a key role in preventing evaporation of the base liquid.The higher the temperature,the smaller the deviation of the evaporation rate from the predicted value.The evaporation rates obtained by the two control groups at the height of the fuid surface from the outlet were lower than the predict value.The magnetic feld had a certain promotional efect on the evaporation of the ferrofuid.The experimental results were consistent with the results obtained using Bolotov’s model.This research validates Bolotov’s model and shows that the model is somewhat biased but still responds well to diferent variables.展开更多

Phased array ultrasonic testing (PAUT) as one of the NDE methods is more promising compared with conventional ultrasonic testing (UT) method in terms of inspection speed and flexibility. To incorporate PAUT, the techniques should be qualified, which traditionally is performed by extensive physical experiments. However, with the development of numerical models simulating UT method, it is expected to complement or partly replace the experiments with the intention to reduce costs and operational uncertainties. The models should be validated to ensure its consistency to reality. This validation work can be done by comparing the model with other validated models or corresponding experiments. The purpose of current work focuses on the experimental validation of a numerical model, simSUNDT, developed by the Chalmers University of Technology. Validation is conducted by comparing different data presentations (A-, B- and C-scan) from experimental and simulated results with some well-defined artificial defects. Satisfactory correlations can be observed from the comparisons. After the validation, sound field optimization work aiming at retrieving maximized echo amplitude on a certain defect can be started using the model. This also reveals the flexibility of parametric studies using simulation models.展开更多

A 1.2-MW high power ICP wind tunnel is studied through numerical simulation and experimental validation. The distribution characteristics and interaction mechanism of the flow field and electromagnetic field of the ICP wind tunnel are investigated using the multi-field coupling method of flow, electromagnetic, chemical, and thermodynamic field. The accuracy of the numerical simulation is validated by comparing the experimental results with the simulation results. Thereafter, the wind tunnel pressure, air velocity, electron density, Joule heating rate, Lorentz force, and electric field intensity obtained using the simulation are analyzed and discussed. The results indicate that for the 1.2-MW ICP wind tunnel, the maximum values of temperature, pressure, electron number density, and other parameters are observed during coil heating. The influence of the radial Lorentz force on the momentum transfer is stronger than that of the axial Lorentz force. The electron number density at the central axis and the amplitude and position of the Joule heating rate are affected by the radial Lorentz force. Moreover, the plasma in the wind tunnel is constantly in the subsonic flow state, and a strong eddy flow is easily generated at the inlet of the wind tunnel.展开更多

How to model the contact/impact process accurately and efficiently is one...The impact problem of a flexible multibody system is a non-smooth, high-transient, and strong-nonlinear dynamic process with variable boundary. How to model the contact/impact process accurately and efficiently is one of the main difficulties in many engineering applications.The numerical approaches being used widely in impact analysis are mainly from two fields: multibody system dynamics(MBS) and computational solid mechanics(CSM).Approaches based on MBS provide a more efficient yet less accurate analysis of the contact/impact problems, while approaches based on CSM are well suited for particularly high accuracy needs, yet require very high computational effort. To bridge the gap between accuracy and efficiency in the dynamic simulation of a flexible multibody system with contacts/impacts, a partition method is presented considering that the contact body is divided into two parts, an impact region and a non-impact region. The impact region is modeled using the finite element method to guarantee the local accuracy, while the non-impact region is modeled using the modal reduction approach to raise the global efficiency. A three-dimensional rod-plate impact experiment is designed and performed to validate the numerical results. The principle for how to partition the contact bodies is proposed:the maximum radius of the impact region can be estimated by an analytical method, and the modal truncation orders of the non-impact region can be estimated by the highest frequency of the signal measured. The simulation results using the presented method are in good agreement with the experimental results. It shows that this method is an effec-tive formulation considering both accuracy and efficiency.Moreover, a more complicated multibody impact problem of a crank slider mechanism is investigated to strengthen this conclusion.展开更多

Objective: To explore the specific pharmacological molecular mechanisms of Kai Xin San(KXS)on treating Alzheimer’s disease(AD) based on network pharmacology and experimental validation. Methods:The chemical compounds...Objective: To explore the specific pharmacological molecular mechanisms of Kai Xin San(KXS)on treating Alzheimer’s disease(AD) based on network pharmacology and experimental validation. Methods:The chemical compounds of KXS and their corresponding targets were screened using the Encyclopedia of Traditional Chinese Medicine(ETCM) database. AD-related target proteins were obtained from MalaCards database and DisGeNET databases. Key compounds and targets were identified from the compound-targetdisease network and protein-protein interaction(PPI) network analysis. Functional enrichment analysis predicted the potential key signaling pathways involved in the treatment of AD with KXS. The binding affinities between key ingredients and targets were further verified using molecular docking. Finally, the predicted key signaling pathway was validated experimentally. Positioning navigation and space search experiments were conducted to evaluate the cognitive improvement effect of KXS on AD rats. Western blot was used to further examine and investigate the expression of the key target proteins related to the predicted pathway. Results: In total, 38active compounds and 469 corresponding targets of KXS were screened, and 264 target proteins associated with AD were identified. The compound-target-disease and PPI networks identified key active ingredients and protein targets. Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway analysis suggested a potential effect of KXS in the treatment of AD via the amyloid beta(Aβ)-glycogen synthase kinase-3 beta(GSK3β)-Tau pathway. Molecular docking revealed a high binding affinity between the key ingredients and targets. In vivo,KXS treatment significantly improved cognitive deficits in AD rats induced by Aβ1-42, decreased the levels of Aβ, p-GSK3β, p-Tau and cyclin-dependent kinase 5, and increased the expressions of protein phosphatase 1alpha(PP1A) and PP2A(P<0.05 or P<0.01). Conclusion: KXS exerted neuroprotective effects by regulating the Aβ-GSK3β-Tau signaling pathway, which provides novel insights into the therapeutic mechanism of KXS and a feasible pharmacological strategy for the treatment of AD.展开更多
Influenza is an acute viral respiratory infection that has caused high morbidity and mortality worldwide.Influenza A virus(IAV)has been found to activate multiple programmed cell death pathways,including ferroptosis.F...Influenza is an acute viral respiratory infection that has caused high morbidity and mortality worldwide.Influenza A virus(IAV)has been found to activate multiple programmed cell death pathways,including ferroptosis.Ferroptosis is a novel form of programmed cell death in which the accumulation of intracellular iron promotes lipid peroxidation,leading to cell death.However,little is known about how influenza viruses induce ferroptosis in the host cells.In this study,based on network pharmacology,we predicted the mechanism of action of Maxing Shigan decoction(MXSGD)in IAV-induced ferroptosis,and found that this process was related to biological processes,cellular components,molecular function and multiple signaling pathways,where the hypoxia inducible factor-1(HIF-1)signaling pathway plays a significant role.Subsequently,we constructed the mouse lung epithelial(MLE-12)cell model by IAV-infected in vitro cell experiments,and revealed that IAV infection induced cellular ferroptosis that was characterized by mitochondrial damage,increased reactive oxygen species(ROS)release,increased total iron and iron ion contents,decreased expression of ferroptosis marker gene recombinant glutathione peroxidase 4(GPX4),increased expression of acyl-CoA synthetase long chain family member 4(ACSL4),and enhanced activation of hypoxia inducible factor-1α(HIF-1α),induced nitric oxide synthase(iNOS)and vascular endothelial growth factor(VEGF)in the HIF-1 signaling pathway.Treatment with MXSGD effectively reduced intracellular viral load,while reducing ROS,total iron and ferrous ion contents,repairing mitochondrial results and inhibiting the expression of cellular ferroptosis and the HIF-1 signaling pathway.Finally,based on animal experiments,it was found that MXSGD effectively alleviated pulmonary congestion,edema and inflammation in IAV-infected mice,and inhibited the expression of ferroptosis-related protein and the HIF-1 signaling pathway in lung tissues.展开更多
BACKGROUND Diabetic kidney disease(DKD)is one of the serious complications of diabetes mellitus,and the existing treatments cannot meet the needs of today's patients.Traditional Chinese medicine has been validated...BACKGROUND Diabetic kidney disease(DKD)is one of the serious complications of diabetes mellitus,and the existing treatments cannot meet the needs of today's patients.Traditional Chinese medicine has been validated for its efficacy in DKD after many years of clinical application.However,the specific mechanism by which it works is still unclear.Elucidating the molecular mechanism of the Nardostachyos Radix et Rhizoma-rhubarb drug pair(NRDP)for the treatment of DKD will provide a new way of thinking for the research and development of new drugs.AIM To investigate the mechanism of the NRDP in DKD by network pharmacology combined with molecular docking,and then verify the initial findings by in vitro experiments.METHODS The Traditional Chinese Medicine Systems Pharmacology(TCMSP)database was used to screen active ingredient targets of NRDP.Targets for DKD were obtained based on the Genecards,OMIM,and TTD databases.The VENNY 2.1 database was used to obtain DKD and NRDP intersection targets and their Venn diagram,and Cytoscape 3.9.0 was used to build a"drug-component-target-disease"network.The String database was used to construct protein interaction networks.Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis and Gene Ontology analysis were performed based on the DAVID database.After selecting the targets and the active ingredients,Autodock software was used to perform molecular docking.In experimental validation using renal tubular epithelial cells(TCMK-1),we used the Cell Counting Kit-8 assay to detect the effect of NRDP on cell viability,with glucose solution used to mimic a hyperglycemic environment.Flow cytometry was used to detect the cell cycle progression and apoptosis.Western blot was used to detect the protein expression of STAT3,p-STAT3,BAX,BCL-2,Caspase9,and Caspase3.RESULTS A total of 10 active ingredients and 85 targets with 111 disease-related signaling pathways were obtained for NRDP.Enrichment analysis of KEGG pathways was performed to determine advanced glycation end products(AGEs)-receptor for AGEs(RAGE)signaling as the core pathway.Molecular docking showed good binding between each active ingredient and its core targets.In vitro experiments showed that NRDP inhibited the viability of TCMK-1 cells,blocked cell cycle progression in the G0/G1 phase,and reduced apoptosis in a concentrationdependent manner.Based on the results of Western blot analysis,NRDP differentially downregulated p-STAT3,BAX,Caspase3,and Caspase9 protein levels(P<0.01 or P<0.05).In addition,BAX/BCL-2 and p-STAT3/STAT3 ratios were reduced,while BCL-2 and STAT3 protein expression was upregulated(P<0.01).CONCLUSION NRDP may upregulate BCL-2 and STAT3 protein expression,and downregulate BAX,Caspase3,and Caspase9 protein expression,thus activating the AGE-RAGE signaling pathway,inhibiting the vitality of TCMK-1 cells,reducing their apoptosis.and arresting them in the G0/G1 phase to protect them from damage by high glucose.展开更多

The boundary condition is a crucial factor affecting the permeability variation due to suffusion.An experimental investigation on the permeability of gap-graded soil due to horizontal suffusion considering the boundar...The boundary condition is a crucial factor affecting the permeability variation due to suffusion.An experimental investigation on the permeability of gap-graded soil due to horizontal suffusion considering the boundary effect is conducted,where the hydraulic head difference(DH)varies,and the boundary includes non-loss and soil-loss conditions.Soil samples are filled into seven soil storerooms connected in turn.After evaluation,the variation in content of fine sand(ΔR_(f))and the hydraulic conductivity of soils in each storeroom(C_(i))are analyzed.In the non-loss test,the soil sample filling area is divided into runoff,transited,and accumulated areas according to the negative or positive ΔR_(f) values.ΔR_(f) increases from negative to positive along the seepage path,and Ci decreases from runoff area to transited area and then rebounds in accumulated area.In the soil-loss test,all soil sample filling areas belong to the runoff area,where the gentle-loss,strengthened-loss,and alleviated-loss parts are further divided.ΔR_(f) decreases from the gentle-loss part to the strengthened-loss part and then rebounds in the alleviated-loss part,and C_(i) increases and then decreases along the seepage path.The relationship between ΔR_(f) and Ci is different with the boundary condition.Ci exponentially decreases with ΔR_(f) in the non-loss test and increases with ΔR_(f) generally in the soil-loss test.展开更多
Deepsea mining has been proposed since the 1960s to alleviate the lack of resources on land.Vertical hydraulic transport of collected ores from the seabed to the sea surface is considered the most promising method for...Deepsea mining has been proposed since the 1960s to alleviate the lack of resources on land.Vertical hydraulic transport of collected ores from the seabed to the sea surface is considered the most promising method for industrial applications.In the present study,an indoor model test of the vertical hydraulic transport of particles was conducted.A noncontact optical method has been proposed to measure the local characteristics of the particles inside a vertical pipe,including the local concentration and particle velocity.The hydraulic gradient of ore transport was evaluated with various particle size distributions,particle densities,feeding concentrations and mixture flow velocities.During transport,the local concentration is larger than the feeding concentration,whereas the particle velocity is less than the mixture velocity.The qualitative effects of the local concentration and local fluid velocity on the particle velocity and slip velocity were investigated.The local fluid velocity contributes significantly to particle velocity and slip velocity,whereas the effect of the local concentration is marginal.A higher feeding concentration and mixture flow velocity result in an increased hydraulic gradient.The effect of the particle size gradation is slight,whereas the particle density plays a crucial role in the transport.展开更多

文摘Deep foundations are currently used in engineering practice to solve problems caused by weak geotechnical characteristics of the ground.Impact pile driving is an interesting and viable solution from economic and technical points of view.However,it is necessary to ensure that the environmental drawbacks,namely ground-borne vibration,are adequately met.For this purpose,the authors propose an axisymmetric finite element method-perfectly matched layer(FEM-PML)approach,where the nonlinear behavior of the soil is addressed through an equivalent linear methodology.Given the complexity of the problem,an experimental test site was developed and fully characterized.The experimental work comprised in-situ and laboratory soil characterization,as well as the measurement of vibrations induced during pile driving.The comparison between experimental and numerical results demonstrated a very good agreement,from which it can be concluded that the proposed numerical approach is suitable for the prediction of vibrations induced by impact pile driving.The experimental database is available as supplemental data and may be used by other researchers in the validation of their prediction models.
文摘Piles in a group experience additional displacements in soil due to pile-to-pile interactions apart from those resulting from the external loading.The effect of these interactions determined assuming soil as an elastic and/or viscoelastic material on pile head impedance functions of the pile group is studied by relating the group stiffness to the static stiffness of a single pile.However,the prevailing elastic solutions may misestimate the resulting pile group response due to the lack of consideration for either soil(material)and/or soil-pile interface nonlinearities.It is well established that soil behaves nonlinearly under moderate-to-high loading amplitudes,and besides,the soil-pile interface nonlinearity can exist even at small loading amplitudes.This study addresses the effects of these nonlinearities on the vertical impedance functions of a 3×3-pile group using numerical methods by direct analyses and superposition using pile-to-pile interaction factors.The numerical results are validated using scaled model tests under 1 g conditions.The results highlight the overestimation of pile-to-pile interactions in the pile group when assuming elastic soil conditions.The cases either by direct analyses or superposition approach involving soil and soil-pile interface nonlinearities agree well with the experimental pile group responses under close-to-elastic and nonlinear conditions.

文摘This article investigates the characteristics of shock wave overpressure generated by multi-layer composite charge under different detonation modes.Combining dimensional analysis and the explosion mechanism of the charge,a peak overpressure prediction model for the composite charge under singlepoint detonation and simultaneous detonation was established.The effects of the charge structure and initiation method on the overpressure field characteristics were investigated in AUTODYN simulation.The accuracy of the prediction model and the reliability of the numerical simulation method were subsequently verified in a series of static explosion experiments.The results reveal that the mass of the inner charge was the key factor determining the peak overpressure of the composite charge under single-point detonation.The peak overpressure in the radial direction improved apparently with an increase in the aspect ratio of the charge.The overpressure curves in the axial direction exhibited a multi-peak phenomenon,and the secondary peak overpressure even exceeded the primary peak at distances of 30D and 40D(where D is the charge diameter).The difference in peak overpressure among azimuth angles of 0-90°gradually decreased with an increase in the propagation distance of the shock wave.The coupled effect of the detonation energy of the inner and outer charge under simultaneous detonation improved the overpressure in both radial and axial directions.The difference in peak overpressure obtained from model prediction and experimental measurements was less than 16.4%.

文摘To analyze the spring disturbance torque caused by motional cable harness in a stabilized platform,the Kirchhoff theory based cable harness model has been previously developed to dynamically simulate the motional cable harness.In this paper,this model was validated by comparing the simulation results with the experiment results(both the spring force and the deformed profile of the motional cable harness).In the experiment,a special optical measuring instrument based on binocular vision was developed and the motion and deformation of cable harness were measured.A simplified stabilized platform system was constructed,and the absolute value of spring disturbance force during the motion of this simplified frame was obtained by using a force gauge(0.02N precision).The physical parameters of experimental specimen were also measured.The experimental and simulated results showed good agreement.These results should be useful for better motional cable harness layout design and reliable evaluation of the spring disturbance torque.

文摘The current research on the evaporation of ferrofuids mainly focuses on the characterization of ultra-low vapor pressure ferrofuids in vacuum and the theoretical analysis of the evaporation process.Few studies have focused on the experimental validation of the proposed evaporation rate equations and on the comparison of the diferences in ferrofuid evaporation.In this study,based on the Bolotov’s model,an evaporation rate equation is deduced from the experimental model.The experimental study included a comparison of the evaporation,magnetic particle volume fraction,temperature,height of the fuid surface from the outlet,and magnetic feld of a kerosene-based ferrofuid and its base carrier liquid.The prepared sample was evaporated in a test tube,and the evaporation rate was calculated by measuring the weight loss of the sample.The experimental results show that the evaporation rate of the base carrier liquid is higher than that of the ferrofuid.The smaller the volume fraction of the magnetic particles,the greater the evaporation rate.The magnetic particles play a key role in preventing evaporation of the base liquid.The higher the temperature,the smaller the deviation of the evaporation rate from the predicted value.The evaporation rates obtained by the two control groups at the height of the fuid surface from the outlet were lower than the predict value.The magnetic feld had a certain promotional efect on the evaporation of the ferrofuid.The experimental results were consistent with the results obtained using Bolotov’s model.This research validates Bolotov’s model and shows that the model is somewhat biased but still responds well to diferent variables.

文摘In safety dominant industries, nondestructive evaluation (NDE) is crucial in quality assurance and assessment. Phased array ultrasonic testing (PAUT) as one of the NDE methods is more promising compared with conventional ultrasonic testing (UT) method in terms of inspection speed and flexibility. To incorporate PAUT, the techniques should be qualified, which traditionally is performed by extensive physical experiments. However, with the development of numerical models simulating UT method, it is expected to complement or partly replace the experiments with the intention to reduce costs and operational uncertainties. The models should be validated to ensure its consistency to reality. This validation work can be done by comparing the model with other validated models or corresponding experiments. The purpose of current work focuses on the experimental validation of a numerical model, simSUNDT, developed by the Chalmers University of Technology. Validation is conducted by comparing different data presentations (A-, B- and C-scan) from experimental and simulated results with some well-defined artificial defects. Satisfactory correlations can be observed from the comparisons. After the validation, sound field optimization work aiming at retrieving maximized echo amplitude on a certain defect can be started using the model. This also reveals the flexibility of parametric studies using simulation models.

文摘We take the established inductively coupled plasma(ICP) wind tunnel as a research object to investigate the thermal protection system of re-entry vehicles. A 1.2-MW high power ICP wind tunnel is studied through numerical simulation and experimental validation. The distribution characteristics and interaction mechanism of the flow field and electromagnetic field of the ICP wind tunnel are investigated using the multi-field coupling method of flow, electromagnetic, chemical, and thermodynamic field. The accuracy of the numerical simulation is validated by comparing the experimental results with the simulation results. Thereafter, the wind tunnel pressure, air velocity, electron density, Joule heating rate, Lorentz force, and electric field intensity obtained using the simulation are analyzed and discussed. The results indicate that for the 1.2-MW ICP wind tunnel, the maximum values of temperature, pressure, electron number density, and other parameters are observed during coil heating. The influence of the radial Lorentz force on the momentum transfer is stronger than that of the axial Lorentz force. The electron number density at the central axis and the amplitude and position of the Joule heating rate are affected by the radial Lorentz force. Moreover, the plasma in the wind tunnel is constantly in the subsonic flow state, and a strong eddy flow is easily generated at the inlet of the wind tunnel.

文摘The impact problem of a flexible multibody system is a non-smooth, high-transient, and strong-nonlinear dynamic process with variable boundary. How to model the contact/impact process accurately and efficiently is one of the main difficulties in many engineering applications.The numerical approaches being used widely in impact analysis are mainly from two fields: multibody system dynamics(MBS) and computational solid mechanics(CSM).Approaches based on MBS provide a more efficient yet less accurate analysis of the contact/impact problems, while approaches based on CSM are well suited for particularly high accuracy needs, yet require very high computational effort. To bridge the gap between accuracy and efficiency in the dynamic simulation of a flexible multibody system with contacts/impacts, a partition method is presented considering that the contact body is divided into two parts, an impact region and a non-impact region. The impact region is modeled using the finite element method to guarantee the local accuracy, while the non-impact region is modeled using the modal reduction approach to raise the global efficiency. A three-dimensional rod-plate impact experiment is designed and performed to validate the numerical results. The principle for how to partition the contact bodies is proposed:the maximum radius of the impact region can be estimated by an analytical method, and the modal truncation orders of the non-impact region can be estimated by the highest frequency of the signal measured. The simulation results using the presented method are in good agreement with the experimental results. It shows that this method is an effec-tive formulation considering both accuracy and efficiency.Moreover, a more complicated multibody impact problem of a crank slider mechanism is investigated to strengthen this conclusion.

文摘A modified one-dimensional transient hygrothermal model for multilayer wall was proposed using air humidity ratio and temperature as the driving potentials.The solution for the governing equations was obtained numerically by implementing the finite-difference scheme.To evaluate the accuracy of the model,a test system was built up to measure relative humidity and temperature within a porous wall and compare with the prediction of the model.The prediction results have good agreement with the experimental results.For the interface close to indoor side,the maximum deviation of temperature between calculated and test data is 1.87 K,and the average deviation is 0.95 K;the maximum deviation of relative humidity is 11.4%,and the average deviation is 5.7%.For the interface close to outdoor side,the maximum deviation of temperature between prediction and measurement is 1.78 K,and the average deviation is 1.1 K;the maximum deviation of relative humidity is 9.9%,and the average deviation is 4.2%.

文摘Objective: To explore the specific pharmacological molecular mechanisms of Kai Xin San(KXS)on treating Alzheimer’s disease(AD) based on network pharmacology and experimental validation. Methods:The chemical compounds of KXS and their corresponding targets were screened using the Encyclopedia of Traditional Chinese Medicine(ETCM) database. AD-related target proteins were obtained from MalaCards database and DisGeNET databases. Key compounds and targets were identified from the compound-targetdisease network and protein-protein interaction(PPI) network analysis. Functional enrichment analysis predicted the potential key signaling pathways involved in the treatment of AD with KXS. The binding affinities between key ingredients and targets were further verified using molecular docking. Finally, the predicted key signaling pathway was validated experimentally. Positioning navigation and space search experiments were conducted to evaluate the cognitive improvement effect of KXS on AD rats. Western blot was used to further examine and investigate the expression of the key target proteins related to the predicted pathway. Results: In total, 38active compounds and 469 corresponding targets of KXS were screened, and 264 target proteins associated with AD were identified. The compound-target-disease and PPI networks identified key active ingredients and protein targets. Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway analysis suggested a potential effect of KXS in the treatment of AD via the amyloid beta(Aβ)-glycogen synthase kinase-3 beta(GSK3β)-Tau pathway. Molecular docking revealed a high binding affinity between the key ingredients and targets. In vivo,KXS treatment significantly improved cognitive deficits in AD rats induced by Aβ1-42, decreased the levels of Aβ, p-GSK3β, p-Tau and cyclin-dependent kinase 5, and increased the expressions of protein phosphatase 1alpha(PP1A) and PP2A(P<0.05 or P<0.01). Conclusion: KXS exerted neuroprotective effects by regulating the Aβ-GSK3β-Tau signaling pathway, which provides novel insights into the therapeutic mechanism of KXS and a feasible pharmacological strategy for the treatment of AD.

文摘Influenza is an acute viral respiratory infection that has caused high morbidity and mortality worldwide.Influenza A virus(IAV)has been found to activate multiple programmed cell death pathways,including ferroptosis.Ferroptosis is a novel form of programmed cell death in which the accumulation of intracellular iron promotes lipid peroxidation,leading to cell death.However,little is known about how influenza viruses induce ferroptosis in the host cells.In this study,based on network pharmacology,we predicted the mechanism of action of Maxing Shigan decoction(MXSGD)in IAV-induced ferroptosis,and found that this process was related to biological processes,cellular components,molecular function and multiple signaling pathways,where the hypoxia inducible factor-1(HIF-1)signaling pathway plays a significant role.Subsequently,we constructed the mouse lung epithelial(MLE-12)cell model by IAV-infected in vitro cell experiments,and revealed that IAV infection induced cellular ferroptosis that was characterized by mitochondrial damage,increased reactive oxygen species(ROS)release,increased total iron and iron ion contents,decreased expression of ferroptosis marker gene recombinant glutathione peroxidase 4(GPX4),increased expression of acyl-CoA synthetase long chain family member 4(ACSL4),and enhanced activation of hypoxia inducible factor-1α(HIF-1α),induced nitric oxide synthase(iNOS)and vascular endothelial growth factor(VEGF)in the HIF-1 signaling pathway.Treatment with MXSGD effectively reduced intracellular viral load,while reducing ROS,total iron and ferrous ion contents,repairing mitochondrial results and inhibiting the expression of cellular ferroptosis and the HIF-1 signaling pathway.Finally,based on animal experiments,it was found that MXSGD effectively alleviated pulmonary congestion,edema and inflammation in IAV-infected mice,and inhibited the expression of ferroptosis-related protein and the HIF-1 signaling pathway in lung tissues.

文摘BACKGROUND Diabetic kidney disease(DKD)is one of the serious complications of diabetes mellitus,and the existing treatments cannot meet the needs of today's patients.Traditional Chinese medicine has been validated for its efficacy in DKD after many years of clinical application.However,the specific mechanism by which it works is still unclear.Elucidating the molecular mechanism of the Nardostachyos Radix et Rhizoma-rhubarb drug pair(NRDP)for the treatment of DKD will provide a new way of thinking for the research and development of new drugs.AIM To investigate the mechanism of the NRDP in DKD by network pharmacology combined with molecular docking,and then verify the initial findings by in vitro experiments.METHODS The Traditional Chinese Medicine Systems Pharmacology(TCMSP)database was used to screen active ingredient targets of NRDP.Targets for DKD were obtained based on the Genecards,OMIM,and TTD databases.The VENNY 2.1 database was used to obtain DKD and NRDP intersection targets and their Venn diagram,and Cytoscape 3.9.0 was used to build a"drug-component-target-disease"network.The String database was used to construct protein interaction networks.Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis and Gene Ontology analysis were performed based on the DAVID database.After selecting the targets and the active ingredients,Autodock software was used to perform molecular docking.In experimental validation using renal tubular epithelial cells(TCMK-1),we used the Cell Counting Kit-8 assay to detect the effect of NRDP on cell viability,with glucose solution used to mimic a hyperglycemic environment.Flow cytometry was used to detect the cell cycle progression and apoptosis.Western blot was used to detect the protein expression of STAT3,p-STAT3,BAX,BCL-2,Caspase9,and Caspase3.RESULTS A total of 10 active ingredients and 85 targets with 111 disease-related signaling pathways were obtained for NRDP.Enrichment analysis of KEGG pathways was performed to determine advanced glycation end products(AGEs)-receptor for AGEs(RAGE)signaling as the core pathway.Molecular docking showed good binding between each active ingredient and its core targets.In vitro experiments showed that NRDP inhibited the viability of TCMK-1 cells,blocked cell cycle progression in the G0/G1 phase,and reduced apoptosis in a concentrationdependent manner.Based on the results of Western blot analysis,NRDP differentially downregulated p-STAT3,BAX,Caspase3,and Caspase9 protein levels(P<0.01 or P<0.05).In addition,BAX/BCL-2 and p-STAT3/STAT3 ratios were reduced,while BCL-2 and STAT3 protein expression was upregulated(P<0.01).CONCLUSION NRDP may upregulate BCL-2 and STAT3 protein expression,and downregulate BAX,Caspase3,and Caspase9 protein expression,thus activating the AGE-RAGE signaling pathway,inhibiting the vitality of TCMK-1 cells,reducing their apoptosis.and arresting them in the G0/G1 phase to protect them from damage by high glucose.
文摘A method of analysis for the simultaneous determination of methylisothiazolinone (MI), methylchloroisothiazolinone (CMI), benzisothiazolinone (BIT) and Bronopol (BNP) in washing-up liquid was established. The method consisted of a gradient HPLC analysis at three different wavelengths. The four compounds could be analyzed with good precision and accuracy.

文摘The boundary condition is a crucial factor affecting the permeability variation due to suffusion.An experimental investigation on the permeability of gap-graded soil due to horizontal suffusion considering the boundary effect is conducted,where the hydraulic head difference(DH)varies,and the boundary includes non-loss and soil-loss conditions.Soil samples are filled into seven soil storerooms connected in turn.After evaluation,the variation in content of fine sand(ΔR_(f))and the hydraulic conductivity of soils in each storeroom(C_(i))are analyzed.In the non-loss test,the soil sample filling area is divided into runoff,transited,and accumulated areas according to the negative or positive ΔR_(f) values.ΔR_(f) increases from negative to positive along the seepage path,and Ci decreases from runoff area to transited area and then rebounds in accumulated area.In the soil-loss test,all soil sample filling areas belong to the runoff area,where the gentle-loss,strengthened-loss,and alleviated-loss parts are further divided.ΔR_(f) decreases from the gentle-loss part to the strengthened-loss part and then rebounds in the alleviated-loss part,and C_(i) increases and then decreases along the seepage path.The relationship between ΔR_(f) and Ci is different with the boundary condition.Ci exponentially decreases with ΔR_(f) in the non-loss test and increases with ΔR_(f) generally in the soil-loss test.

文摘Deepsea mining has been proposed since the 1960s to alleviate the lack of resources on land.Vertical hydraulic transport of collected ores from the seabed to the sea surface is considered the most promising method for industrial applications.In the present study,an indoor model test of the vertical hydraulic transport of particles was conducted.A noncontact optical method has been proposed to measure the local characteristics of the particles inside a vertical pipe,including the local concentration and particle velocity.The hydraulic gradient of ore transport was evaluated with various particle size distributions,particle densities,feeding concentrations and mixture flow velocities.During transport,the local concentration is larger than the feeding concentration,whereas the particle velocity is less than the mixture velocity.The qualitative effects of the local concentration and local fluid velocity on the particle velocity and slip velocity were investigated.The local fluid velocity contributes significantly to particle velocity and slip velocity,whereas the effect of the local concentration is marginal.A higher feeding concentration and mixture flow velocity result in an increased hydraulic gradient.The effect of the particle size gradation is slight,whereas the particle density plays a crucial role in the transport.
