There are many active substances in Atlantic cod(Gadus morhua)explaining the variety of biological activities.In order to study the immunomodulatory activity and the mechanism of Atlantic cod peptides at the cellular level.In this study,cod peptides were isolated by 80%ethanol extraction method,the isolated ethanol-soluble cod peptides(CP-ES)were investigated and their immunomodulatory activity was verified.Additionally,CP-ES showed lower molecular weight and more hydrophobic amino acids.CP-ES could promote the proliferation of spleen lymphocytes and T lymphocytes in mice,suggesting that CP-ES may regulate adaptive immunity.It promoted the release of NO and the expression of iNOS,TNF-α,IL-6 and IL-1βgenes in macrophages,suggesting that CP-ES may regulate innate immunity.CP-ES could promote the expression of TLR2 gene,and the peptides identified in CP-ES were docked with TLR2 to predict the peptides playing a major role in CP-ES.These results suggested that CP-ES may regulate the immune activity of both innate and adaptive lines. 展开更多

Natural nano-hydroxyapatite(HA)was extracted from Pacific cod(Gadus macrocephalus)bone with a thermostable col-lagenolytic protease in the present study.Conditions for the enzymatic reaction were optimized to be 60℃and pH 7.0,and a desir-able extraction efficiency was achieved by using the crude collagenolytic protease.Dynamic light scattering,transmission electron microscopy and energy-dispersive X-ray analysis revealed that nano-HA are anionic spherical(about 110nm)particles mainly com-prised of calcium and phosphorus at an approximate ratio of 5:3.As evaluated with the mouse ex vivo intestinal segments,the extracted nano-HA displayed comparable level of intestinal bioavailability to the positive control CaCl_(2).By treating with inhibitors(NaN3,ami-loride)and low temperature(4℃),clathrin-mediated endocytosis was assumed to involve the intestinal absorption of nano-HA.Over-all,the application of thermostable collagenolytic protease is proved to be a promising alternative method for nano-HA extraction from natural resource with improved ecological and biological value. 展开更多

采用分步降温法冷冻保存太平洋鳕(Gadus macrocephalus)精液,并用扫描电镜和透射电镜技术研究了精子的超微结构损伤。分析了添加剂(蛋黄)及五种抗冻剂(PG(丙二醇)、DMSO(二甲基亚砜)、EG(乙二醇)、GLY(甘油)、MeOH(甲醇))不同浓度(8%、... 采用分步降温法冷冻保存太平洋鳕(Gadus macrocephalus)精液,并用扫描电镜和透射电镜技术研究了精子的超微结构损伤。分析了添加剂(蛋黄)及五种抗冻剂(PG(丙二醇)、DMSO(二甲基亚砜)、EG(乙二醇)、GLY(甘油)、MeOH(甲醇))不同浓度(8%、10%、12%、14%、16%、18%)对太平洋鳕精子冷冻保存效果的影响。实验结果表明,蛋黄能够显著提高太平洋鳕冷冻保存效果(P<0.05);12%PG中添加10%蛋黄保存的冻融精子运动率最高(85.87%±1.6%),显著高于其他冻存组(P<0.05)。添加蛋黄的12%浓度的DMSO组解冻后精子运动速率较高,平均直线速度、平均曲线速度、平均路径速度分别达到了(120.39±20.78)μm/s、(120.39±20.78)μm/s、(155.64±12.02)μm/s,与其他各实验组差异显著(P<0.05)。通过扫描电镜和透射电镜观察新鲜和冻融后的鳕鱼精子发现,鲜精中75.5%精子形态结构正常、24.5%精子形态结构异常;运动率最高的冻精中67%精子形态结构正常、33%精子形态结构异常。超低温保存对太平洋鳕精子结构产生了显著影响,细胞膜破裂、线粒体肿胀变形,鞭毛脱落、断裂。 展开更多

Transferrin gene polymorphisms and population genetic studies of Atlantic cod(Gadus morhua)

Objective:To detect single nucleotide polymorphisms(SNPs)in the cod transferrin gene by comparing the sequences from Norwegian(North East Atlantic Ocean)and Canadian(North West Atlantic Ocean)specimen,and to quantify the genetic variation and differentiation in East and West Atlantic cod populations.Methods:cDNA sequences between individuals of Canadian(North West Atlantic Ocean)and Norwegian(North East Atlantic Ocean)origin were aligned.Allele frequencies of the SNPs were used to discriminate the different Atlantic cod populations in West/East Atlantic Ocean,and the Baltic Sea.Results:The sequence alignment detected 19 SNPs,of which 18 of them resulted in amino acid changes in the transferrin protein.Nonsynonymous to synonymous site substitution ratio(dn/ds)was by far greater than 1 providing an evidence for the existence of positive selection.The West Atlantic cod populations showed high values of heterozygosity and the Baltic populations were found to be inbred.Conclusions:This study identified and indicated transferrin gene polymorphisms that can be used for population differentiations. 展开更多

驯化野生大头鳕(Gadus macrocephalus Tilesius)作为亲鱼,通过自然受精和人工授精两种方法获得受精卵,自然受精卵通过虹吸法收集。受精卵孵化条件为水温6~8℃,盐度32~33,pH7.8~8.2,微充气。在奥林巴斯SZ61解剖镜下对大头鳕的胚胎发育... 驯化野生大头鳕(Gadus macrocephalus Tilesius)作为亲鱼,通过自然受精和人工授精两种方法获得受精卵,自然受精卵通过虹吸法收集。受精卵孵化条件为水温6~8℃,盐度32~33,pH7.8~8.2,微充气。在奥林巴斯SZ61解剖镜下对大头鳕的胚胎发育进行了观察,并使用CCD图像传感器拍照,描述了各发育时期的发育时序和形态特征。大头鳕受精卵为圆球形端黄卵,弱黏性,无油球,卵膜上具龟裂结构,卵子直径0.95~1.12mm,n=30,卵黄均匀透明。类似于大部分硬骨鱼类,其胚胎发育可分为胚盘形成期、卵裂期、桑椹期、囊胚期、原肠期、神经胚期、器官发生期、尾芽期、肌肉效应期。在水温6~8℃,盐度32~33的条件下,受精卵历时约336h完成胚胎发育过程,孵化出膜。在大头鳕胚胎发育过程中,具有4个不同于其他硬骨鱼类的地方:(1)发现早期分裂球具有不规则现象;(2)很多胚胎发育过程不出现柯氏囊,有柯氏囊的胚胎仅占10%左右,且出现晚,消失快;(3)胚胎孵化为仔鱼与发育进程关联性不大,孵化的仔鱼有3种主要形态;(4)胚胎各个时期都有较高死亡率,以原肠期最为严重。 展开更多

通过对江鳕(LotalataLinnaeus)和大头鳕(GadusmacrocephalusTilexius)的形态学比较研究发现,江鳕和大头鳕都有胆囊、幽门盲囊,肝脏较大。脊柱和附肢骨骼种类相同,区别仅在于骨块的数量和嵴的发达程度上,尾都是等尾型。

Early life history affects fish size mainly by indirectly regulating the growth during each stage:a case study in a demersal fish

The complex life histories of demersal fishes are artificially separated into multiple stages along with changes in morphology and habitat.It is worth exploring whether the phenotypes expressed earlier and later during the life cycle are related or decoupled.The life stages of first year Pacific cod(Gadus macrocephalus)were tracked over different hatch years and regions to test whether the early life history had a long-lasting effect on subsequent growth.We further explored the contribution of growth in the early and subsequent life history stages to body size at the end of each stage.In addition to the accessory growth centre and the first annual ring,the other two checks on the otolith possibly related to settlement and entering deeper waters were identified in 75 Pacific cod individuals.The direct and indirect relationships among the life history stages was interpreted based on path analysis.The results showed that growth prior to the formation of the accessory growth centre had a significant effect on the absolute growth of the fish before and after settlement and migration to deep water.However,there was no or moderate evidence that early growth affected the body size at each stage,which was mainly regulated by growth during the stage.This study supports the lasting effect of early growth and clarifies that it affects size mainly by indirectly regulating staged growth.Quantifying the phenotype relationships and identifying the internal mechanisms form the basis for assessing population dynamics and understanding the processes behind the changes. 展开更多
