In this paper,we utilized dimethylhydrazine(DMH)-induced colorectal cancer(CC)model rats to explore the effects of casein glycomacropeptide(CGMP)on colorectal cancer.Rats with CC were orally administrated with 10,50,or 100 mg/kg bw d CGMP,or the same volume of phosphatebuffered saline for 15 weeks. The total numbers of aberrant crypt foci(ACF)and crypts per focus in colon were scored using a light microscope at low magnification after the colon was stained with methylene blue solutions. The methylation level of DNA extracted from colon was detected using methylation-specific PCR.The expression of p16 and mucin 2(MUC2)proteins were measured by immunohistochemistry. The results showed that although ACF were found in rats treated with CGMP,their number was significantly decreased compared to that of model rats.In addition, methylation and expression levels of p16 and MUC2 were also inhibited by CGMP,which were more obvious in rats treated with 50 mg/kg bw d CGMP.In conclusion,CGMP has potential application as nutritional therapy for preventing colorectal cancer.©2013 Beijing Academy of Food Sciences.Production and hosting by Elsevier B.V.All rights reserved. 展开更多

Casein glycomacropeptide(CGMP) is a bioactive peptide derived from milk with multiple functions. This study was aimed at evaluating the effects of CGMP as a potential feed additive on growth performance, intestinal morphology, intestinal barrier permeability and inflammatory responses of Escherichia coli K88(E. coli K88) challenged piglets. Eighteen weaning piglets were randomly assigned to three groups.Control group and K88 challenged group received a basal diet, and CGMP treated group received the basal diet supplemented with 1% of CGMP powder. The trail lasted for 12 days, K88 was orally administered to the piglets of K88 challenged group and CGMP treated group on days 8-10. The results showed that the diet containing 1% CGMP significantly alleviated the decrease in average daily gain(P <0.05),increase in pathogenic bacteria amounts in intestinal contents(P < 0.05), intestinal morphology(P > 0.05) and barrier permeability damage(P < 0.05), and acute inflammatory response(P < 0.05)induced by E. coli K88 infection. In conclusion, CGMP supplementation in the diet protected the weaning piglets against E. coli K88 infection. 展开更多

Helicobacter pylori (H. pylori) eradication is considered a necessary step in the management of peptic ulcer disease, chronic gastritis, gastric adenocarcinoma and mucosa associated lymphoid tissue lymphoma. Standard triple therapy eradication regimens are inconvenient and achieve unpredictable and often poor results. Eradication rates are decreasing over time with increase in antibiotic resistance. Fermented milk and several of its component whey proteins have emerged as candidates for complementary therapy. In this context the current review seeks to summarize the current evidence available on their role in H. pylori eradication. Pertinent narrative/systematic reviews, clinical trials and laboratory studies on individual components including fermented milk, yogurt, whey proteins, lactoferrin, α-lactalbumin (α-LA), glycomacropeptide and immunoglobulin were comprehensively searched and retrieved from Medline, Embase, Scopus, Cochrane Controlled Trials Register and abstracts/proceedings of conferences up to May 2013. A preponderance of the evidence available on fermented milk-based probiotic preparations and bovine lactoferrin suggests a beneficial effect in Helicobacter eradication. Evidence for α-LA and immunoglobulins is promising while that for glycomacropeptide is preliminary and requires substantiation. The magnitude of the potential benefit documented so far is small and the precise clinical settings are ill defined. This restricts the potential use of this group as a complementary therapy in a nutraceutical setting hinging on better patient acceptability/compliance. Further work is necessary to identify the optimal substrate, fermentation process, dose and the ideal clinical setting (prevention/treatment, first line therapy/recurrence, symptomatic/asymptomatic, gastritis/ulcer diseases etc.). The potential of this group in high antibiotic resistance or treatment failure settings presents interesting possibilities and deserves further exploration. 展开更多
