On the Hyperferritinemia and Hereditary Cataract Syndrome

Mutations in the promoter region of ferritin light gene can induce an uncontrolled over expression of this protein. Consequently, ferritin is found in serum at very high levels (~1000 ng/mL) and it accumulates in the crystalline lens, generating cataracts. This entity is known as hyperferritin and hereditary cataract syndrome (HHCS) which is inherited in an autosomal dominant manner. Case Presentation. We describe a family affected by HHCS. The proband was identified among subjects submitted to a biological screening for hemochromatosis. He had very high levels of serum ferritin (~900 ng/mL) with normal transferrin saturation (TS). The proband has a single H63D HFE-gene mutation and normal HAMP-gene. He was submitted to periodical phlebotomies that induced anemia and a decrease in TS but no changes on serum ferritin levels. Analyses of promoter region of ferritin-light chain gene showed a 39 C > T mutation, responsible for HHCS. The proband’s sister carried also this mutation. Both subjects had developed cataracts. Discussion. Similar to the first family described carrying this syndrome and to other cases reported, the proband was erroneously submitted to phlebotomies. Clinical consequences are illustrated in this report. HHCS is an infrequent entity which has to be correctly identified. The unique therapeutic approach to this syndrome must be cataract surgery. 展开更多

AIM To set up cell lines of human hepatocellular carcinoma in nude mice for the research of cell biology and gene therapy. METHODS Xenotransplantation of human hepatoma into nude mice was carried out and the growth rate, histopathology and immunology of the nude mice were studied. The DNA from xenografts were analyzed by HBV gene and PCR amplification of a fragment of p 53 gene exon 7, which were identified by dot blot hybridization, restriction fragments length polymorphism and DNA sequencing. RESULTS hHCC4 and hHCC415 cell lines could be successively transplanted in nude mice and the population doubling time was 7 and 5 days respectively. These strains retained the original characteristics of histopathology, secreting AFP and heteroploid karyotypes in human hepatocellular carcinoma. The fragment of HBV gene was detected in the genomic DNA of both hHCC4 and hHCC15, however only hHCC4 secreted HBsAg. The mutation at 250 code (C→A) and 249 code (G→T) were detected respectively in the genomic DNA of hHCC4 and hHCC15. CONCLUSION The two cell lines are useful material for studying cell biology and gene therapy in human hepatocellular carcinoma and provide molecular biological trace of the relationship between high mortality of hepatoma and AFB1 severe pollution of the daily common foods in this district. 展开更多

This study analyzes and predicts the vibration characteristics of fiberreinforced composite sandwich(FRCS)cylindrical-spherical(CS)combined shells with hexagon honeycomb core(HHC)for the first time based on an analytical model developed,which makes good use of the advantage of the first-order shear deformation theory(FSDT),the multi-segment decomposition technique,the virtual spring technology,the Jacobi-Ritz approach,and the transfer function method.The equivalent material properties of HHC are firstly determined by the modified Gibson’s formula,and the related energy equations are derived for the HHC-FRCS-CS combined shells,from which the fundamental frequencies,the mode shapes,and the forced vibration responses are solved.The current model is verified through the discussion of convergence and comparative analysis with the associated published literature and finite element(FE)results.The effects of geometric parameters of HHC on the dynamic property of the structure are further investigated with the verified model.It reveals that the vibration suppression capability can be greatly enhanced by reducing the ratio of HHC thickness to total thickness and the ratio of wall thickness of honeycomb cell to overall radius,and by increasing the ratio of length of honeycomb cell to overall radius and honeycomb characteristic angle of HHC. 展开更多

Hyperhomoysteinemia as a risk factor for coronary heart diseases in chronic hepatitis C patients

It affects mainly the liver but several extrahepatic manifestations are also accounted. Chronic hepatitis C patients are at an increased risk of developing hepatic steatosis, which share many clinical features with the metabolic syndrome. Hepatic steatosis has also been associated with elevated levels of markers of inflammation such as homocysteine, identified as hyperhomocysteinemia (HHC). HHC due to Methylenetetrahydrofolate Reductase (MTHFR) gene, in particular the C677T polymorphism, was recently associated with coronary heart diseases (CHD) in chronic hepatitis C (CHC) patients. Homocysteine is an intermediate in methionine metabolism, which takes place mainly in the liver metabolism. Deficiencies of micronutrients (folate, vitamin B 6 and possibly vitamin B 12) along with mild hyperhomocysteinemia, perhaps, act synergistically with other classical risk factors to further increase the risk of CHD. Clinical data indicate that HHC is associated with an increased incidence of CHD as well as with the severity of the disease in CHC patients. In conclusion, HHC might be a potential aetiological factor of CHD in CHC patients. The aim of this review is to investigate the progression of coronary heart diseases in chronic hepatitis C patients and correlate with levels of homocysteine in concurrence to genetic defects and nutrient deficiencies. However, future studies need to clarify the mechanistic role of HHC in CHD and CHC as a useful paradigm with most interesting therapeutic implications. 展开更多

Hehui capsule(Hhc)has been used as an assistant treatment for hyperlipidemia.According to the previous studies,miltiorrhiza and Chavica roxburghii may reduce the serum lipid level.This study aims at examining the efficiency of Hehui capsule with regards to treating hyperlipidemia in a randomized control,double-blind clinical trial.In the research,we assigned 104 eligible subjects to Hehui capsule group and placebo groups(52:52,respectively).Subjects used self-control and inter-group controls to compare serum total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL)and other changes before and after the test group and the test group and the control group.During the 45 days intervention,101 cases were actually included,among which 51 cases for test group,and other 50 cases for placebo group.The total effective rate of blood lipid reduction(90.20%)in the test group was significantly higher than that in the control group(26.00%)(P<0.01).Together,the result indicates that Hhc has a certain curative effect on dyslipidemia,with no obvious adverse reactions,which can be developed into a new drug for hyperlipidemia. 展开更多
