本文建立了皮革制品在酸性汗液中17种元素铝,锑,砷,钡,硼,镉,铬,钴,铜,铅,锰,汞,镍,硒,锶,锡和锌的迁移量测试方法。采用模拟人体汗液即酸性汗液对皮革中的17种元素进行萃取,实验发现17种元素在10~500 ug/L浓度范围内线性相关系数均可... 本文建立了皮革制品在酸性汗液中17种元素铝,锑,砷,钡,硼,镉,铬,钴,铜,铅,锰,汞,镍,硒,锶,锡和锌的迁移量测试方法。采用模拟人体汗液即酸性汗液对皮革中的17种元素进行萃取,实验发现17种元素在10~500 ug/L浓度范围内线性相关系数均可达到0.9999以上,方法检出限在0.03~0.80 mg/kg范围内。对皮革样品进行加标回收率在90.90%~105.26%之间,重复性(n=10)在0.09%-5.73%。此方法准确,快速,可以同时测定皮革制品中17种元素的迁移量。选取50批次的实际样品进行实验研究,结果发现所有样品中砷,铬,铅,镉,锡,铝,硼,钴,硒这九个元素均有检出,而皮革制品在长期与皮肤接触过程中过量的化学元素迁移量会对人体健康造成危害。 展开更多

建立了电感耦合等离子体发射光谱法(ICP-OES)测定渣油中不同形态铁含量的分析方法;采用三氯化铁、环烷酸铁、2-乙基己酸铁、四苯基卟啉氯化铁作为模型化合物,考察萃取方法的可行性及优化萃取条件。结果表明:纯水对三氯化铁的萃取率可达... 建立了电感耦合等离子体发射光谱法(ICP-OES)测定渣油中不同形态铁含量的分析方法;采用三氯化铁、环烷酸铁、2-乙基己酸铁、四苯基卟啉氯化铁作为模型化合物,考察萃取方法的可行性及优化萃取条件。结果表明:纯水对三氯化铁的萃取率可达到100%;采用稀酸作为萃取剂进行萃取后,可将渣油中的铁分为酸提出铁、油中残留铁及其它铁,其中酸提出铁包含水溶性铁、绝大部分石油酸铁以及少量的卟啉铁类化合物;质量分数为5%的盐酸溶液对石油酸铁的萃取效果最好,对环烷酸铁和2-乙基己酸铁的萃取率分别为100.5%和93.2%;适宜的萃取时间为3h。采用所建方法分析实际渣油加氢原料和产物中铁的组成分布,发现其它铁在加氢后大部分消失了,说明这部分铁极易通过加氢反应脱除;而油中残留铁的脱除率远低于总铁脱除率。 展开更多

Various factors/pathways including hormonal regulation have been suggested to control herpes simplex virus type 1 (HSV-1) latency and reactivation. Our computer analysis identified a DNA repeat containing thyroid hormoneresponsive elements (TRE) in the regulatory region of HSV-1 latency-associated transcript (LAT). Thyroid hormone (triiodothyronine, T3) functions via its receptor TR (thyroid hormone receptor), a transcription factor. Present study investigated the roles of TR and T3 in HSV-1 gene expression using cultured neuoroblastoma cell lines. We demonstrated that liganded TR activated LAT transcription, but repressed infected cell protein no. 0 (ICP0) transcription in the presence of LAT TRE. Chromatin immunoprecipitation (CHIP) assays showed that TRs were recruited to LAT TREs independently of T3 and hyperacetylated H4 was associated with the LAT promoter that was transcriptionally active. In addition, ChIP results showed that the chromatin insulator protein CCCTC-binding factor was enriched at the LAT TREs in the presence of TR and T3. In addition, the BRG1 chromatin remodeling complex is found to participate in the T3/TR-mediated LAT activation since overexpression of BRG1 enhanced the LAT transcription and the dominant-negative mutant K785R abolished the activation. This is the first report revealing that TR elicits epigenetic regulation on HSV-1 ICP0 expression in neuronal cells and could have a role in the complex processes of HSV-1 latency/reactivation. 展开更多

As an immediate-early protein of herpes simplex virus, infected-cell polypeptide 0 (ICP0) exhibits complicated interactions with host cells, and its regulatory function on gene expression is of great importance. Since the ICP0 encoding sequence contains many rare codons which are absent in E.coli, and ICP0 is highly unstable in prokaryotic cells, expression of entire ICP0 in prokaryotic cells has never been reported. In order to further investigate the function of ICP0, a recombinant plasmid was constructed by subcloning a cDNA fragment encoding an amino-terminal of 105 residues of the ICP0 protein into pGEX-5x-1 vector. The resulting GST-105 fusion antigen peptide was expressed with high efficiency in E.coli. Antibodies prepared after the immunization of mice with purified fusion protein can recognize not only the denatured ICP0 protein, but also the native ICP0 protein with normal biological conformation. 展开更多
