The nutrient digestion,absorption and biological activity of bee pollen may be limited due to the complex pollen wall.Here,the effect of superfine grinding technology on the release of nutrients from bee pollen were i...The nutrient digestion,absorption and biological activity of bee pollen may be limited due to the complex pollen wall.Here,the effect of superfine grinding technology on the release of nutrients from bee pollen were investigated,and their antioxidant activities and in vitro digestion were explored in this study.Results showed that the content of nutrients in bee pollen increased after wall disruption.Among them,fat content increased by 22.55%-8.31%,protein content increased by 0.54%-4.91%,starch content increased by 36.31%-48.64%,soluble sugar content increased by 20.57%-29.67%,total phenolic acid content increased by 11.73%-86.98%and total flavonoids content increased by 14.29%-24.79%.At the same time,the antioxidant activity increased by 14.84%-46.00%.Furthermore,the active components such as phenolic compounds in the wall-disruption bee pollen were more readily to be released during the in vitro digestion,and easier to be absorbed because of their higher bioaccessibility.Antioxidant activities during in vitro digestion were also improved in walldisruption bee pollen.These findings provide evidence that bee pollen wall disruption was suggested,thus,it is more conducive to exerting the value of bee pollen in functional foods.展开更多

Poria cocos(PC)is a famous traditional Chinese medicine(TCM)and a widely used healthcare ingredient,which has antiobesity,enhancing immunity and improving sleep effects.Traditionally,only water-soluble poria polysacch...Poria cocos(PC)is a famous traditional Chinese medicine(TCM)and a widely used healthcare ingredient,which has antiobesity,enhancing immunity and improving sleep effects.Traditionally,only water-soluble poria polysaccharide(WSP)is extracted and applied for clinical application,while insoluble polysaccharide(alkali-soluble poria polysaccharide,ASP)is discarded as herb residue.However,the whole PC has also been historically utilized as functional herbal food.Considering the beneficial role of dietary fiber and the traditional use of PC,ASP may also contribute substantially to the therapy function of PC.Compared to WSP,little attention has been paid to ASP and ASP modified product carboxymethyl poria polysaccharide(CMP)which has been used as an antitumor adjuvant drug.In this study,the oil,cholesterol,metal ions and polyphenols adsorption ability,in vitro simulated digestive and the gut microbiota fermentation characteristics of WSP,ASP and CMP were studied to evaluate the functional values of three P.cocos polysaccharides(PCPs).The results showed that all three PCPs had good adsorption capacity on cholesterol,polyphenols and metal ions(Cd^(2+)/Zn^(2+)/Mg^(2+)),among which ASP showed the highest capacity than WSP and CMP.The adsorption capacity of all three PCPs on heavy metal ions(Cd^(2+)/Zn^(2+))was stronger than that of non-heavy metal ions(Mg^(2+));The in vitro digestibility of all three PCPs was very low,but WSP was slightly higher than ASP and CMP;Moreover,the indigestible residue of all three PCPs could improve the richness and diversity of gut microbiota,among which ASP had the greatest influence.In general,ASP and CMP could significantly promote the proliferation of some probiotics and inhibit the growth of some harmful bacteria.The gut microbiota diversity of CMP was reduced,but the richness of probiotics,especially Parabacteroides distasonis was significantly enhanced compared with the ASP group,and the growth of harmful bacteria Klebsiella pneumoniae was inhibited after CMP treatment.The short-chain fatty acids(SCFAs)analysis results showed that all three PCPs could significantly promote the production of acetic acid,propionic acid and the total acid content compared with blank control group,and SCFAs producing activity was positively correlated with the proliferative capacity of probiotics.Taken together,the good adsorption characteristics and gut microbiota regulatory activity of ASP may lay foundation for its lipid-lowering and immune-improving function.Additionally,the probiotic effect of CMP and ASP indicated that except for only use the water extract of PC in clinic,CMP and ASP also can be used in healthcare to take full advantage of this valuable medicine.展开更多

To examine the effect of mechanical processing for plant-based materials on antioxidant properties during digestion,relationships between the size of pulverized plant tissue and changes in antioxidant activities durin...To examine the effect of mechanical processing for plant-based materials on antioxidant properties during digestion,relationships between the size of pulverized plant tissue and changes in antioxidant activities during simulated in vitro digestion were investigated.The citrus peel tissue was pulverized and classified into four powder fractions followed their particle size distributions as 125−180m,180−355m,355−500m and 500−710m.These fractions were regarded as a plant tissue model with various degrees of cell damages.Powder samples were collected during the digestion and measured their particle sizes and colors.Besides,total phenolic content and antioxidant activities of digested fluid were also evaluated.The results showed that the powder color of smaller particles were significantly changed than the bigger ones.The bio-properties of digested fluid for the bigger particle sizes comparatively retained more than smaller ones at the simulated gastric stage.It also showed mostly stable during simulated gastric or small intestinal digestion stage.This suggested that the release of bioactive compounds from plant tissues during digestion could be related to their structural attributes such as degrees of cell damages which could be affected by processing methods and conditions.展开更多
[Objectives]To explore the change rule of polyphenol content and antioxidant activity of coarse old leaves of Yingshan Yunwu Tea in the process of human digestion.[Methods]The coarse and old leaves of Yunwu tea in Yin...[Objectives]To explore the change rule of polyphenol content and antioxidant activity of coarse old leaves of Yingshan Yunwu Tea in the process of human digestion.[Methods]The coarse and old leaves of Yunwu tea in Yingshan,Huanggang,Hubei Province were selected as the research object,and their digestion in vitro was simulated.The total polyphenol content was determined by Folin-phenol reagent colorimetric method,and the DPPH radical scavenging activity and total antioxidant activity were determined.[Results]After simulated gastrointestinal digestion in vitro,the polyphenol content and antioxidant activity of coarse old leaf tea soup showed a downward trend.After gastrointestinal digestion,the polyphenol content in tea infusion separately decreased by 31.8%and 8.5%;the scavenging rate of DPPH free radical was 97%before digestion,decreased to 92%after gastric digestion and 65%after intestinal digestion,which decreased by 5%and 27%,respectively;after gastrointestinal digestion,the total antioxidant capacity of tea soup decreased by 4.7%and 3.1%,respectively.[Conclusions]This study provided a reference for the development and application of coarse old leaves of Yingshan Yunwu tea,and provided a reference for the nutritional value evaluation and comprehensive utilization of coarse old leaves,so as to make the best use of coarse tea leaves and reduce the waste of resources.展开更多

One of the strategic methods used to reduce diabetes-associated oxidative stress is to inhibit the carbohydrate-digesting enzymes, thereby decreasing gastrointestinal glucose production. Plant-derived natural antioxidant molecules are considered a therapeutic tool in the treatment of oxidative stress and diabetes. The objective of this study was to identify tartary buckwheat rice flavonoids and evaluate the effect of in vitro digestion on their antioxidant and antidiabetic properties. High performance liquid chromatography(HPLC) analysis indicated the presence of rutin as a major component and quercitrin as a minor component of both digested and non-digested flavonoids. Both extracts showed a significant antioxidant capacity, but digested flavonoids showed reduced activity compared to non-digested. There were some decreases of the antioxidant activities(2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid) diammonium salt(ABTS), 2,2-diphenyl-1-picrylhydrazy(DPPH) radical, and ferric reducing antioxidant power(FRAP)) of digested tartary buckwheat rice flavonoids compared with non-digested. Flavonoids from both groups significantly inhibited reactive oxygen species(ROS) production and α-glucosidase activity. Both digested and non-digested flavonoids markedly increased glucose consumption and glycogen content in Hep G2 cells. Tartary buckwheat rice flavonoids showed appreciable antioxidant and antidiabetic properties, even after digestion. Tartary buckwheat rice appears to be a promising functional food with potent antioxidant and antidiabetic properties.展开更多
Rapeseed meal is a promising food ingredient, but its utilization is limited by the presence of some potentially harmful ingredients, such as glucosinolates. Fermentation is a cost-effective method of detoxication but...Rapeseed meal is a promising food ingredient, but its utilization is limited by the presence of some potentially harmful ingredients, such as glucosinolates. Fermentation is a cost-effective method of detoxication but a food-grade starter culture with glucosinolates degradation capacity is required. In this study, 46 strains of lactic acid bacteria from traditional paocai brines were screened for their ability to glucosinolate degradation. The results showed that more than 50% of the strains significantly degraded glucosinolates. Two strains of Lactiplantibacillus(p7 and s7) with high capacity of glucosinolates degradation through producing enzymes were identified. Then,an optimized condition for rapeseed meal fermentation by p7 was established to degrade glucosinolates, which can achieve about 80% degradation. UPLC/Q-TOF-MS analysis showed that the degradation rate of individual glucosinolates was different and the degradation rate of gluconapin and progoitrin in rapeseed meal can reach more than 90%. Meanwhile, fermentation with p7 can improve safety of rapeseed meal by inhibiting the growth of Enterobacteriaceae and improve its nutritional properties by degrading phytic acid. The in vitro digestion experiments showed that the content of glucosinolates in rapeseed meal decreased significantly during gastric digestion. Meanwhile, fermentation with p7 can greatly improve the release of soluble protein and increase the contents of free essential amino acids, such as lysine(increased by 12 folds) and methionine(increased by 10 folds).展开更多
Background As demand for high quality animal feed continues to raise,it becomes increasingly important to mini-mize the environmental impact of feed production.An appealing sustainable approach to provide feed fractio...Background As demand for high quality animal feed continues to raise,it becomes increasingly important to mini-mize the environmental impact of feed production.An appealing sustainable approach to provide feed fractions is to use organic residues from agro-food industry.In this regard,volatile fatty acids(VFAs)such as acetic,propionic and butyric acids,derived from bioconversion of organic residues can be used as precursors for production of micro-bial protein with ruminant feed inclusion potential.This study aims to investigate the in vitro digestibility of the Asper-gillus oryzae edible fungal biomass cultivated on VFAs-derived from anaerobic digestion of residues.The produced fungal protein biomass,along with hay clover silage and rapeseed meal were subjected to various in vitro assays using two-stage Tilley and Terry(TT),gas,and bag methods to evaluate and compare its digestibility for application in ruminant feed.Results The produced fungal biomass contained a higher crude protein(CP)(41%–49%)and rather similar neutral detergent fiber(NDF)(41%–56%)compared to rapeseed meal.The rumen in vitro dry matter digestibility(IVDMD)of the fungal biomass in the TT method ranged from 82%to 88%(statistically similar to that of the gas method(72%to 85%)).The IVDMD of fungal biomass were up to 26%and 40%greater than that of hay clover silage and rapeseed meal,respectively.The type of substrate and bag method had pronounced effect on the fermentation products(ammonium-N(NH4+-N),total gas and VFAs).Fungal biomass digestion resulted in the highest release of NH4+-N(340–540 mg/L)and the ratio of acetate to propionate ratio(3.5)among subjected substrates.Conclusion The results indicate that gas method can be used as a reliable predictor for IVDMD as well as fermenta-tion products.Furthermore,the high IVDMD and fermentation product observed for Aspergillus oryzae fungal biomass digestion,suggest that the supplementation of fungal biomass will contribute to improving the rumen digestion by providing necessary nitrogen and energy to the ruminant and microbiota.展开更多
Background:The most used protein sources in ruminant nutrition are considered as having negative impacts in terms of environmental sustainability and competition with human nutrition.Therefore,the investigation of alt...Background:The most used protein sources in ruminant nutrition are considered as having negative impacts in terms of environmental sustainability and competition with human nutrition.Therefore,the investigation of alternative and sustainable feedstuffs is becoming a priority in ruminant production systems.Results:This trial was designed to evaluate eight full-fat insect meals(Acheta domesticus–ACD;Alphitobius diaperinus–ALD;Blatta lateralis–BL;Gryllus bimaculatus–GB;Grylloides sygillatus–GS;Hermetia illucens–HI;Musca domestica–MD;and Tenebrio molitor–TM)as potential protein and lipid sources in ruminant nutrition.Fermentation parameters and fatty acids(FA)of rumen digesta after 24-h in vitro ruminal incubation of the tested insect meals were measured and compared with those of three plant-based meals(soybean meal,rapeseed meal and sunflower meal)and fishmeal(FM).Similarly to FM,the insect meals led to a significantly lower total gas production(on average,1.75vs.4.64 mmol/g dry matter-DM),methane production(on average,0.33 vs.0.91 mmol/g DM),volatile FA production(on average,4.12 vs.7.53 mmol/g DM),and in vitro organic matter disappearance(on average,0.32 vs.0.59 g/g)than those observed for the plant meals.The insect meals also led to lower ammonia of rumen fluid,when expressed as a proportion of total N(on average,0.74 vs.0.52 for the plant and insect meals,respectively),which could be an advantage provided that intestinal digestibility is high.Differences in ruminal fermentation parameters between the insect meals could be partially explained by their chitin,crude protein and ether extract contents,as well as by their FA profile.In particular,high content of polyunsaturated FA,or C12:0(in HI),seems to partially inhibit the ruminal fermentations.Conclusions:The tested full-fat insect meals appear to be potentially an interesting protein and lipid source for ruminants,alternative to the less sustainable and commonly used ones of plant origin.The FA profile of the rumen digesta of ACD,ALD,GB,GS and TM,being rich in n-6 polyunsaturated FA,could be interesting to improve the quality of ruminant-derived food products.展开更多

Moench) is a major world crop that is a reliable source of fodder and food grain in arid regions. However, unlike other cereals, sorghum grain has low nutritional value, owing mainly to the resistance of its storage proteins(kafirins) to protease digestion. Changing the composition of kafirins or their primary structure may address this problem. To induce mutations in kafirin-encoding genes that were expected to disturb their accumulation in endosperm cells, we used a genome-editing approach. By Agrobacterium-mediated genetic transformation of immature embryos of cv. Avans, we obtained 14 transgenic plants with genetic constructs for site-directed mutagenesis of the k1C5 and g KAF1 genes encoding 22 k Da a-and 28 kDa γ-kafirins, respectively. Sequencing of 5 regenerants obtained by using k1C5-addressing vector revealed two plants with mutations. T_1 progeny of these mutants had higher in vitro digestibility of endosperm proteins(86%–92%), in comparison with the donor Avans(63%–67%). The kernels of these plants had a thick vitreous endosperm. A mutant with increased in vitro protein digestibility and vitreous endosperm, carrying a mutation in the target sequence, was also obtained by use of the gKAF1-addressing vector. Thus, using genome editing technology, we have obtained mutants with improved kafirin digestibility that can be used in sorghum breeding.展开更多
The effects of starch phosphate monoester content(SPC),namely C-3(C3P)and C-6 phosphate monoesters(C6P),on the starch properties were investigated using four potato starches with varied SPC/C3P/C6P and two nonphosphor...The effects of starch phosphate monoester content(SPC),namely C-3(C3P)and C-6 phosphate monoesters(C6P),on the starch properties were investigated using four potato starches with varied SPC/C3P/C6P and two nonphosphorylated maize starches with a similar range of amylose content(AC)as controls.The starch property results showed that a higher SPC is associated with lower turbidity,storage and loss modulus after storage,and water solubility,but higher swelling power(SP)and pasting viscosities.These findings suggested that SPC inhibited molecular rearrangement during storage and starch leaching during heating,and enhanced swelling and viscosities due to increased hydration and water uptake caused by the repulsion effect of phosphate groups and a less ordered crystalline structure.Increased SPC also resulted in lower resistant starch(RS)content in a native granular state but higher RS after retrogradation.Pearson correlations further indicated that SPC/C3P/C6P were positively correlated with peak(r^(2)=0.925,0.873 and 0.930,respectively),trough(r^(2)=0.994,0.968 and 0.988,respectively),and final viscosities(r^(2)=0.981,0.968 and 0.971,respectively).Notably,SPC,mainly C3P,exhibited a significantly positive correlation with SP(r^(2)=0.859)and setback viscosity(r^(2)=0.867),whereas SPC,mainly C6P,showed a weak positive correlation with RS after retrogradation(r^(2)=0.746).However,SPC had no significant correlations with water solubility,turbidity and rheology properties,which were more correlated with AC.These findings are helpful for the food industry to select potato starches with desired properties based on their contents of SPC,C3P,or C6P.展开更多

Qinghai_Xizang Plateau, a unique geographic unit due to high_altitude climate, is one of the most susceptible regions to climate warming. Its ecosystem is very fragile and sensitive to climate change. In order to get a better understanding of the impacts of climate warming on the nutrient contents of herbage grown in Qinghai_Xizang Plateau, a simulative study was implemented at Daban Moutain by using temperature differences resulted from sites selected at different altitudes and nutrient contents and in vitro digestibility were determined for assessing the quality of the grown herbage. There were significant downtrends in crude protein (CP), ether extract (EE) and nitrogen free extract (NFE) contents of herbage along with the increase of temperature. It had a positive correlation between temperature and content of acid detergent fibre (ADF), acid detergent lignin (ADL) in herbage. In vitro digestibility of herbage decreased along with the increase of temperature. The results of this study indicated that climate warming significantly influence nutrient contents and in vitro digestibility of herbage grown in Qinghai_Xizang Plateau. It is suggested that the future climate warming especially the gradual rise of the night temperature could cause negative effect on herbage quality grown in Qinghai_Xizang Plateau by decreasing CP, EE, and NFE contents and increasing some indigestible ingredients such as crude fibre (CF), neutral detergent fibre (NDF), ADF, and ADL. The inhibition of a-glucosidase and glucose-6-phosphatase,two enzymes involved in the carbohydrate metabolism,is an important target to control glycaemia on individuals with type 2 diabetes.In this work we report for ...The inhibition of a-glucosidase and glucose-6-phosphatase,two enzymes involved in the carbohydrate metabolism,is an important target to control glycaemia on individuals with type 2 diabetes.In this work we report for the first time the inhibition of both enzymes by the antihyperglycemic n-butanol extract from Genista tenera(Fabaceae).This extract decreased a-glucosidase and glucose-6-phosphatase activities to 0.97 and 80.25%,respectively,being more effective than acarbose,and phlorizin,the positive controls,which reduced enzymes activities only to 17.39 and 96.06%.Once inflammation and oxidative stress are related to diabetic impairments,the anti-inflammatory activity of the extract was also evaluated,through its inhibitory activity over COX-1 enzyme(47.5%inhibition).Moreover,after induction of oxidative stress by UV radiation,the viability of irradiated rat liver hepatoma cells exposed to the extract was significantly higher(67.82%)than that promoted by ascorbic acid,the positive control(45.05%).In addition,the stability of the extract under gastrointestinal conditions was evaluated by HPLC–DAD-ESI–MS/MS.Flavonoid diglycosides were identified as the main constituents of the extract,and no alterations in the chemical composition nor in the antioxidant activity were observed after in vitro digestion with artificial gastric and pancreatic juices.展开更多

However, a relevant question is whether digestion of fish proteins with gastrointestinal proteases per se generates peptides that also can have health promoting properties and can reduce, e.g., diabetes 2, inflammation and hypertension either in relation to gastrointestinal digestion or as alternative to industrial proteases. The aim of the study was to investigate hydrolysates obtained from in vitro sequential digestion of salmon muscle and skin with gastrointestinal proteases including pepsin, pancreatic and pancreatic + mucosal proteases for their ability to scavenge ABTS^+ radicals and inhibit activity of angiotensin I-converting enzyme (ACE) and dipeptidyl peptidase 4 (DPP-4). Furthermore, it was the aim to study the inhibitory mechanism and stability towards ACE and DPP-4 activity. Analysis of〈 10 kDa hydrolysates showed that gastrointestinal proteases generated peptides with clear radical scavenging activity and DPP-4 and ACE inhibiting activity as well. Hydrolysates from pepsin digestion exhibited the lowest ECso values for radical scavenging activity and ACE inhibition, whereas ECso increased in hydrolysates after subsequent digestion with pancreatic and mucosal proteases. Interestingly, ECso values for the DPP-4 inhibition were hardly affected by sequential digestion. Inhibition modes for the muscle hydrolysates were both competitive and non-competitive, but prolonged incubation showed that the inhibitory properties were unstable and therefore they were probably digested as competitive substrates by gastrointestinal proteases.展开更多

The best alternative would be to use human studies, but they are labour-intensive and expensive and can only be used to investigate a limited number of samples. Hence, a number of in vitro: models have been developed to study pre-absorptive processes and factors affecting bioavailability. The question is, however, how well the results obtained by the various methods correlate to each other and to the in vivo situation. In the present paper, we have compared in vivo data from two human studies on differently processed soups containing carrots, tomato and broccoli, with results obtained by in vitro characterisation of the same soups. In vitro bioaccessibility was estimated by a static in vitro digestion investigating matrix release and micellarization of carotenoids and by uptake studies in a human intestinal cell line (Caco-2). In vivo data was obtained from clinical studies measuring total plasma carotenoid concentrations in human subjects after 4 weeks daily consumption of the soups. Comparison of the in vitro and in vivo results indicate that the combination of a two-step in vitro digestion and Caco-2 cells seems to be a useful tool for estimation of β-carotene bioaccessibility and screening of factors governing the release of β-carotene from this type of food. This study was conducted to evaluate the effect of lactic acid bacteria and propionic acid on the fermentation quality, aer- obic stability and in vitro gas production kinetics and digestibility of whole-crop corn bas...This study was conducted to evaluate the effect of lactic acid bacteria and propionic acid on the fermentation quality, aer- obic stability and in vitro gas production kinetics and digestibility of whole-crop corn based totalmixed ration (TMR) silage. Total mixed ration was ensiled with four treatments: (1) no additives (control); (2) an inoculant (Lactobacillus plantarum) (L); (3) propionicacid (P); (4) propionic acid+lactic acid bacteria (PL). All treatments were ensiled in laboratory-scale silos for 45 days, and then subjected to an aerobic stability test for12 days. Further, four TMR silages were incubated in vitro with buffered rumen fluid to study in vitro gas production kinetics and digestibility. The results indicated that all TMR silages had good fermentation characteristics with low pH (〈3.80) and ammonia nitrogen (NH3-N) contents, and high lactic acid contents as well as Flieg points. Addition of L further improved TMR silage quality with more lactic acid production. Addition of P and PL decreased lactic acid and NH3-N contents of TMR silage compared to the control (P〈0.05). After 12 days aerobic exposure, P and PL silages remained stable, but L and the control silages deteriorated as indicated by a reduction in lactic acid and an increase in pH, and numbers of yeast. Compared to the control, addition of L had no effects on TMR silage in terms of 72 h cumulative gas production, in vitro dry matter digestibility, metabolizable energy, net energy for lactation and short chain fatty acids, whereas addition of PL significantly (P〈0.05) increased them. L silage had higher (P〈0.05) in vitro neutral detergent fiber digestibility than the control silage. The results of our study suggested that TMR silage prepared with whole-crop corn can be well preserved with or without additives. Furthermore, the findings of this study suggested that propionic acid is compatible with lactic acid bacteria inoculants, and when used together, although they reduced lactic acid production of TMR silage, they improved aerobic stability and in vitro nutrients digestibility of TMR silage.展开更多

The aim of the current study was to investigate the effects of a lactic acid bacteria inoculant(Lactobacillus plantarum, LP), fibrolytic enzyme(EN), combination of LP and EN(LP+EN) on fermentation quality, nutr...The aim of the current study was to investigate the effects of a lactic acid bacteria inoculant(Lactobacillus plantarum, LP), fibrolytic enzyme(EN), combination of LP and EN(LP+EN) on fermentation quality, nutritive characteristics and in vitro digestibility of total mixed ration(TMR) silages containing 0, 7.5 and 15.0%(on dry matter basis) of rape(Brassica campestris L.) straw(RS)(denoted as CTMR, LTMR and MTMR, respectively). After ensiling for 60 days, TMR silages without additives were well preserved, but MTMR had higher p H than CTMR and LTMR. There were no differences in other parameters of fermentation quality, microbial composition, nutrition and in vitro digestibility between CTMR and LTMR except for yeast and mold number and crude protein(CP) content. CTMR and LTMR silage had higher CP content, in vitro neutral detergent fiber digestibility(IVNDFD) and in vitro acid detergent fiber digestibility(IVADFD), lower acid detergent fiber(ADF) content than MTMR silage. LP and EN decreased p H and increased dry matter(DM) recovery of TMR silages. LP+EN improved the fermentation quality, nutritive characteristics and in vitro digestibility of TMR silages, showed by lower p H, ADF content, higher lactic acid content, in vitro dry matter digestibility(IVDMD) and IVNDFD. Therefore, It was suggested that TMR silage contained 7.5% RS on a DM basis and treated with LP+EN can be as a useful feed for ruminant.展开更多
As a non-thermal processing technology,high hydrostatic pressure(HHP)can be used for starch modification without affecting the quality and flavour constituents.The effect of HHP on starch is closely related to the tre...As a non-thermal processing technology,high hydrostatic pressure(HHP)can be used for starch modification without affecting the quality and flavour constituents.The effect of HHP on starch is closely related to the treatment time of HHP.In this paper,we investigated the impacts of HHP treatment time(0,5,10,15,20,25,30 min)on the microstructure,gelatinization and thermal properties as well as in vitro digestibility of oat starch by scanning electron microscopy,X-ray diffraction,Fourier transform infrared spectroscopy,13C NMR and differential scanning calorimeter.Results showed that 5-min HHP treatment led to deformation and decreases in short-range ordered and doublehelix structures of oat starch granules,and further extending the treatment time to 15 min or above caused the formation of a gelatinous connection zone,increase of particle size,disintegration of short-range ordered and double-helix structures,and crystal structure change from A type to V type,indicating gelatinization occurred.Longer treatment time also resulted in the reduction in both the viscosity and the stability of oat starch.These indicated that HHP treatment time greatly influenced the microstructure of oat starch,and the oat starch experienced crystalline destruction(5 min),crystalline disintegration(15 min)and gelatinization(>15 min)during HHP treatment.Results of in vitro digestibility showed that the rapidly digestible starch(RDS)content declined first after treatment for 5 to 10 min then rose with the time extending from 15 to 30 min,indicating that longer pressure treatment time was unfavourable to the health benefits of oat starch for humans with diabetes and cardiovascular disease.Therefore,the 500-MPa treatment time for oat starch is recommended not more than 15 min.This study provides theoretical guidance for the application of HHP technology in starch modification and development of health foods.展开更多

文摘The nutrient digestion,absorption and biological activity of bee pollen may be limited due to the complex pollen wall.Here,the effect of superfine grinding technology on the release of nutrients from bee pollen were investigated,and their antioxidant activities and in vitro digestion were explored in this study.Results showed that the content of nutrients in bee pollen increased after wall disruption.Among them,fat content increased by 22.55%-8.31%,protein content increased by 0.54%-4.91%,starch content increased by 36.31%-48.64%,soluble sugar content increased by 20.57%-29.67%,total phenolic acid content increased by 11.73%-86.98%and total flavonoids content increased by 14.29%-24.79%.At the same time,the antioxidant activity increased by 14.84%-46.00%.Furthermore,the active components such as phenolic compounds in the wall-disruption bee pollen were more readily to be released during the in vitro digestion,and easier to be absorbed because of their higher bioaccessibility.Antioxidant activities during in vitro digestion were also improved in walldisruption bee pollen.These findings provide evidence that bee pollen wall disruption was suggested,thus,it is more conducive to exerting the value of bee pollen in functional foods.

文摘The in vitro digestion models mimicking the gastrointestinal(GI)tract of general population and lipid indigestion patients(with lower levels of bile salts or pancreatic lipase)were selected to investigate whether diacylglycerols(DAGs)are potential good lipid sources for these patients.Linseed oil-based DAG(LD)and linseed oil(LT)were selected.LD-based emulsion((83.74±1.23)%)had higher lipolysis degree than LT-based emulsion((74.47±1.16)%)when monitoring the GI tract of normal population as previously reported.Indigestion conditions seriously decreased the digestive degree of LT-based emulsion((40.23±2.48)%-(66.50±3.70)%)while showed less influence on LD-based emulsion((64.18±2.41)%-(81.85±3.45)%).As opposed to LT-based emulsion,LD-based emulsion exhibited preference for releasing unsaturated fatty acids(especially oleic acid andα-linolenic acid)due to their different glycerolipid compositions.LD-based emulsion showed potential for providing lipids and nutrients(including essential fatty acids)for lipid indigestion patients.

文摘Poria cocos(PC)is a famous traditional Chinese medicine(TCM)and a widely used healthcare ingredient,which has antiobesity,enhancing immunity and improving sleep effects.Traditionally,only water-soluble poria polysaccharide(WSP)is extracted and applied for clinical application,while insoluble polysaccharide(alkali-soluble poria polysaccharide,ASP)is discarded as herb residue.However,the whole PC has also been historically utilized as functional herbal food.Considering the beneficial role of dietary fiber and the traditional use of PC,ASP may also contribute substantially to the therapy function of PC.Compared to WSP,little attention has been paid to ASP and ASP modified product carboxymethyl poria polysaccharide(CMP)which has been used as an antitumor adjuvant drug.In this study,the oil,cholesterol,metal ions and polyphenols adsorption ability,in vitro simulated digestive and the gut microbiota fermentation characteristics of WSP,ASP and CMP were studied to evaluate the functional values of three P.cocos polysaccharides(PCPs).The results showed that all three PCPs had good adsorption capacity on cholesterol,polyphenols and metal ions(Cd^(2+)/Zn^(2+)/Mg^(2+)),among which ASP showed the highest capacity than WSP and CMP.The adsorption capacity of all three PCPs on heavy metal ions(Cd^(2+)/Zn^(2+))was stronger than that of non-heavy metal ions(Mg^(2+));The in vitro digestibility of all three PCPs was very low,but WSP was slightly higher than ASP and CMP;Moreover,the indigestible residue of all three PCPs could improve the richness and diversity of gut microbiota,among which ASP had the greatest influence.In general,ASP and CMP could significantly promote the proliferation of some probiotics and inhibit the growth of some harmful bacteria.The gut microbiota diversity of CMP was reduced,but the richness of probiotics,especially Parabacteroides distasonis was significantly enhanced compared with the ASP group,and the growth of harmful bacteria Klebsiella pneumoniae was inhibited after CMP treatment.The short-chain fatty acids(SCFAs)analysis results showed that all three PCPs could significantly promote the production of acetic acid,propionic acid and the total acid content compared with blank control group,and SCFAs producing activity was positively correlated with the proliferative capacity of probiotics.Taken together,the good adsorption characteristics and gut microbiota regulatory activity of ASP may lay foundation for its lipid-lowering and immune-improving function.Additionally,the probiotic effect of CMP and ASP indicated that except for only use the water extract of PC in clinic,CMP and ASP also can be used in healthcare to take full advantage of this valuable medicine.

文摘Natural foods,such as whole pulses,are recommended in the dietary guidelines of the US and China.The plant cell wall structure in whole pulses has important implications for the nutritional functionalities of starch.In this study,garbanzo bean cells with varying degrees of cell wall integrity were subjected to dry heat treatment(DHT)and used to elucidate the food structure-starch digestion properties of pulse food.The morphological features suggested that all cell samples do not exhibit remarkable changes after being subjected to DHT.Molecular rearrangement and the crystallite disruption of starch granules entrapped in cells occurred during DHT as assessed by the crystal structure and thermal properties.DHT decreased the inhibitory effects of enzymes of both the soluble and insoluble components,but the digestion rate and extent of slightly and highly damaged cell samples did not exhibit significant differences compared with their native counterparts.We concluded that the starch digestion of pulse cotyledon cells is primarily determined by the intactness of the cellular structure.This study reveals the role of food structure on the ability to retain the desirable nutritional properties of starch after subjection to physical modification.
文摘To examine the effect of mechanical processing for plant-based materials on antioxidant properties during digestion,relationships between the size of pulverized plant tissue and changes in antioxidant activities during simulated in vitro digestion were investigated.The citrus peel tissue was pulverized and classified into four powder fractions followed their particle size distributions as 125−180m,180−355m,355−500m and 500−710m.These fractions were regarded as a plant tissue model with various degrees of cell damages.Powder samples were collected during the digestion and measured their particle sizes and colors.Besides,total phenolic content and antioxidant activities of digested fluid were also evaluated.The results showed that the powder color of smaller particles were significantly changed than the bigger ones.The bio-properties of digested fluid for the bigger particle sizes comparatively retained more than smaller ones at the simulated gastric stage.It also showed mostly stable during simulated gastric or small intestinal digestion stage.This suggested that the release of bioactive compounds from plant tissues during digestion could be related to their structural attributes such as degrees of cell damages which could be affected by processing methods and conditions.

文摘[Objectives]To explore the change rule of polyphenol content and antioxidant activity of coarse old leaves of Yingshan Yunwu Tea in the process of human digestion.[Methods]The coarse and old leaves of Yunwu tea in Yingshan,Huanggang,Hubei Province were selected as the research object,and their digestion in vitro was simulated.The total polyphenol content was determined by Folin-phenol reagent colorimetric method,and the DPPH radical scavenging activity and total antioxidant activity were determined.[Results]After simulated gastrointestinal digestion in vitro,the polyphenol content and antioxidant activity of coarse old leaf tea soup showed a downward trend.After gastrointestinal digestion,the polyphenol content in tea infusion separately decreased by 31.8%and 8.5%;the scavenging rate of DPPH free radical was 97%before digestion,decreased to 92%after gastric digestion and 65%after intestinal digestion,which decreased by 5%and 27%,respectively;after gastrointestinal digestion,the total antioxidant capacity of tea soup decreased by 4.7%and 3.1%,respectively.[Conclusions]This study provided a reference for the development and application of coarse old leaves of Yingshan Yunwu tea,and provided a reference for the nutritional value evaluation and comprehensive utilization of coarse old leaves,so as to make the best use of coarse tea leaves and reduce the waste of resources.

文摘Oxidative stress and diabetes have a tendency to alter protein, lipid, and DNA moieties. One of the strategic methods used to reduce diabetes-associated oxidative stress is to inhibit the carbohydrate-digesting enzymes, thereby decreasing gastrointestinal glucose production. Plant-derived natural antioxidant molecules are considered a therapeutic tool in the treatment of oxidative stress and diabetes. The objective of this study was to identify tartary buckwheat rice flavonoids and evaluate the effect of in vitro digestion on their antioxidant and antidiabetic properties. High performance liquid chromatography(HPLC) analysis indicated the presence of rutin as a major component and quercitrin as a minor component of both digested and non-digested flavonoids. Both extracts showed a significant antioxidant capacity, but digested flavonoids showed reduced activity compared to non-digested. There were some decreases of the antioxidant activities(2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid) diammonium salt(ABTS), 2,2-diphenyl-1-picrylhydrazy(DPPH) radical, and ferric reducing antioxidant power(FRAP)) of digested tartary buckwheat rice flavonoids compared with non-digested. Flavonoids from both groups significantly inhibited reactive oxygen species(ROS) production and α-glucosidase activity. Both digested and non-digested flavonoids markedly increased glucose consumption and glycogen content in Hep G2 cells. Tartary buckwheat rice flavonoids showed appreciable antioxidant and antidiabetic properties, even after digestion. Tartary buckwheat rice appears to be a promising functional food with potent antioxidant and antidiabetic properties.

文摘Rapeseed meal is a promising food ingredient, but its utilization is limited by the presence of some potentially harmful ingredients, such as glucosinolates. Fermentation is a cost-effective method of detoxication but a food-grade starter culture with glucosinolates degradation capacity is required. In this study, 46 strains of lactic acid bacteria from traditional paocai brines were screened for their ability to glucosinolate degradation. The results showed that more than 50% of the strains significantly degraded glucosinolates. Two strains of Lactiplantibacillus(p7 and s7) with high capacity of glucosinolates degradation through producing enzymes were identified. Then,an optimized condition for rapeseed meal fermentation by p7 was established to degrade glucosinolates, which can achieve about 80% degradation. UPLC/Q-TOF-MS analysis showed that the degradation rate of individual glucosinolates was different and the degradation rate of gluconapin and progoitrin in rapeseed meal can reach more than 90%. Meanwhile, fermentation with p7 can improve safety of rapeseed meal by inhibiting the growth of Enterobacteriaceae and improve its nutritional properties by degrading phytic acid. The in vitro digestion experiments showed that the content of glucosinolates in rapeseed meal decreased significantly during gastric digestion. Meanwhile, fermentation with p7 can greatly improve the release of soluble protein and increase the contents of free essential amino acids, such as lysine(increased by 12 folds) and methionine(increased by 10 folds).

文摘Background As demand for high quality animal feed continues to raise,it becomes increasingly important to mini-mize the environmental impact of feed production.An appealing sustainable approach to provide feed fractions is to use organic residues from agro-food industry.In this regard,volatile fatty acids(VFAs)such as acetic,propionic and butyric acids,derived from bioconversion of organic residues can be used as precursors for production of micro-bial protein with ruminant feed inclusion potential.This study aims to investigate the in vitro digestibility of the Asper-gillus oryzae edible fungal biomass cultivated on VFAs-derived from anaerobic digestion of residues.The produced fungal protein biomass,along with hay clover silage and rapeseed meal were subjected to various in vitro assays using two-stage Tilley and Terry(TT),gas,and bag methods to evaluate and compare its digestibility for application in ruminant feed.Results The produced fungal biomass contained a higher crude protein(CP)(41%–49%)and rather similar neutral detergent fiber(NDF)(41%–56%)compared to rapeseed meal.The rumen in vitro dry matter digestibility(IVDMD)of the fungal biomass in the TT method ranged from 82%to 88%(statistically similar to that of the gas method(72%to 85%)).The IVDMD of fungal biomass were up to 26%and 40%greater than that of hay clover silage and rapeseed meal,respectively.The type of substrate and bag method had pronounced effect on the fermentation products(ammonium-N(NH4+-N),total gas and VFAs).Fungal biomass digestion resulted in the highest release of NH4+-N(340–540 mg/L)and the ratio of acetate to propionate ratio(3.5)among subjected substrates.Conclusion The results indicate that gas method can be used as a reliable predictor for IVDMD as well as fermenta-tion products.Furthermore,the high IVDMD and fermentation product observed for Aspergillus oryzae fungal biomass digestion,suggest that the supplementation of fungal biomass will contribute to improving the rumen digestion by providing necessary nitrogen and energy to the ruminant and microbiota.

文摘Background:The most used protein sources in ruminant nutrition are considered as having negative impacts in terms of environmental sustainability and competition with human nutrition.Therefore,the investigation of alternative and sustainable feedstuffs is becoming a priority in ruminant production systems.Results:This trial was designed to evaluate eight full-fat insect meals(Acheta domesticus–ACD;Alphitobius diaperinus–ALD;Blatta lateralis–BL;Gryllus bimaculatus–GB;Grylloides sygillatus–GS;Hermetia illucens–HI;Musca domestica–MD;and Tenebrio molitor–TM)as potential protein and lipid sources in ruminant nutrition.Fermentation parameters and fatty acids(FA)of rumen digesta after 24-h in vitro ruminal incubation of the tested insect meals were measured and compared with those of three plant-based meals(soybean meal,rapeseed meal and sunflower meal)and fishmeal(FM).Similarly to FM,the insect meals led to a significantly lower total gas production(on average,1.75vs.4.64 mmol/g dry matter-DM),methane production(on average,0.33 vs.0.91 mmol/g DM),volatile FA production(on average,4.12 vs.7.53 mmol/g DM),and in vitro organic matter disappearance(on average,0.32 vs.0.59 g/g)than those observed for the plant meals.The insect meals also led to lower ammonia of rumen fluid,when expressed as a proportion of total N(on average,0.74 vs.0.52 for the plant and insect meals,respectively),which could be an advantage provided that intestinal digestibility is high.Differences in ruminal fermentation parameters between the insect meals could be partially explained by their chitin,crude protein and ether extract contents,as well as by their FA profile.In particular,high content of polyunsaturated FA,or C12:0(in HI),seems to partially inhibit the ruminal fermentations.Conclusions:The tested full-fat insect meals appear to be potentially an interesting protein and lipid source for ruminants,alternative to the less sustainable and commonly used ones of plant origin.The FA profile of the rumen digesta of ACD,ALD,GB,GS and TM,being rich in n-6 polyunsaturated FA,could be interesting to improve the quality of ruminant-derived food products.

文摘Sorghum(Sorghum bicolor(L.) Moench) is a major world crop that is a reliable source of fodder and food grain in arid regions. However, unlike other cereals, sorghum grain has low nutritional value, owing mainly to the resistance of its storage proteins(kafirins) to protease digestion. Changing the composition of kafirins or their primary structure may address this problem. To induce mutations in kafirin-encoding genes that were expected to disturb their accumulation in endosperm cells, we used a genome-editing approach. By Agrobacterium-mediated genetic transformation of immature embryos of cv. Avans, we obtained 14 transgenic plants with genetic constructs for site-directed mutagenesis of the k1C5 and g KAF1 genes encoding 22 k Da a-and 28 kDa γ-kafirins, respectively. Sequencing of 5 regenerants obtained by using k1C5-addressing vector revealed two plants with mutations. T_1 progeny of these mutants had higher in vitro digestibility of endosperm proteins(86%–92%), in comparison with the donor Avans(63%–67%). The kernels of these plants had a thick vitreous endosperm. A mutant with increased in vitro protein digestibility and vitreous endosperm, carrying a mutation in the target sequence, was also obtained by use of the gKAF1-addressing vector. Thus, using genome editing technology, we have obtained mutants with improved kafirin digestibility that can be used in sorghum breeding.

文摘The effects of starch phosphate monoester content(SPC),namely C-3(C3P)and C-6 phosphate monoesters(C6P),on the starch properties were investigated using four potato starches with varied SPC/C3P/C6P and two nonphosphorylated maize starches with a similar range of amylose content(AC)as controls.The starch property results showed that a higher SPC is associated with lower turbidity,storage and loss modulus after storage,and water solubility,but higher swelling power(SP)and pasting viscosities.These findings suggested that SPC inhibited molecular rearrangement during storage and starch leaching during heating,and enhanced swelling and viscosities due to increased hydration and water uptake caused by the repulsion effect of phosphate groups and a less ordered crystalline structure.Increased SPC also resulted in lower resistant starch(RS)content in a native granular state but higher RS after retrogradation.Pearson correlations further indicated that SPC/C3P/C6P were positively correlated with peak(r^(2)=0.925,0.873 and 0.930,respectively),trough(r^(2)=0.994,0.968 and 0.988,respectively),and final viscosities(r^(2)=0.981,0.968 and 0.971,respectively).Notably,SPC,mainly C3P,exhibited a significantly positive correlation with SP(r^(2)=0.859)and setback viscosity(r^(2)=0.867),whereas SPC,mainly C6P,showed a weak positive correlation with RS after retrogradation(r^(2)=0.746).However,SPC had no significant correlations with water solubility,turbidity and rheology properties,which were more correlated with AC.These findings are helpful for the food industry to select potato starches with desired properties based on their contents of SPC,C3P,or C6P.
文摘The increasing trend of air temperature along with the climate warming has been accepted gradually by scientists and by the general public. Qinghai_Xizang Plateau, a unique geographic unit due to high_altitude climate, is one of the most susceptible regions to climate warming. Its ecosystem is very fragile and sensitive to climate change. In order to get a better understanding of the impacts of climate warming on the nutrient contents of herbage grown in Qinghai_Xizang Plateau, a simulative study was implemented at Daban Moutain by using temperature differences resulted from sites selected at different altitudes and nutrient contents and in vitro digestibility were determined for assessing the quality of the grown herbage. There were significant downtrends in crude protein (CP), ether extract (EE) and nitrogen free extract (NFE) contents of herbage along with the increase of temperature. It had a positive correlation between temperature and content of acid detergent fibre (ADF), acid detergent lignin (ADL) in herbage. In vitro digestibility of herbage decreased along with the increase of temperature. The results of this study indicated that climate warming significantly influence nutrient contents and in vitro digestibility of herbage grown in Qinghai_Xizang Plateau. It is suggested that the future climate warming especially the gradual rise of the night temperature could cause negative effect on herbage quality grown in Qinghai_Xizang Plateau by decreasing CP, EE, and NFE contents and increasing some indigestible ingredients such as crude fibre (CF), neutral detergent fibre (NDF), ADF, and ADL. This, consequently, decreases the ruminant assimilation ability.

文摘The inhibition of a-glucosidase and glucose-6-phosphatase,two enzymes involved in the carbohydrate metabolism,is an important target to control glycaemia on individuals with type 2 diabetes.In this work we report for the first time the inhibition of both enzymes by the antihyperglycemic n-butanol extract from Genista tenera(Fabaceae).This extract decreased a-glucosidase and glucose-6-phosphatase activities to 0.97 and 80.25%,respectively,being more effective than acarbose,and phlorizin,the positive controls,which reduced enzymes activities only to 17.39 and 96.06%.Once inflammation and oxidative stress are related to diabetic impairments,the anti-inflammatory activity of the extract was also evaluated,through its inhibitory activity over COX-1 enzyme(47.5%inhibition).Moreover,after induction of oxidative stress by UV radiation,the viability of irradiated rat liver hepatoma cells exposed to the extract was significantly higher(67.82%)than that promoted by ascorbic acid,the positive control(45.05%).In addition,the stability of the extract under gastrointestinal conditions was evaluated by HPLC–DAD-ESI–MS/MS.Flavonoid diglycosides were identified as the main constituents of the extract,and no alterations in the chemical composition nor in the antioxidant activity were observed after in vitro digestion with artificial gastric and pancreatic juices.
文摘Research on marine bioactive peptides has mainly focused on characterization of peptides in hydrolysates prepared with commercial industrial enzymes and the usefulness of such hydrolysates in health and functional foods. However, a relevant question is whether digestion of fish proteins with gastrointestinal proteases per se generates peptides that also can have health promoting properties and can reduce, e.g., diabetes 2, inflammation and hypertension either in relation to gastrointestinal digestion or as alternative to industrial proteases. The aim of the study was to investigate hydrolysates obtained from in vitro sequential digestion of salmon muscle and skin with gastrointestinal proteases including pepsin, pancreatic and pancreatic + mucosal proteases for their ability to scavenge ABTS^+ radicals and inhibit activity of angiotensin I-converting enzyme (ACE) and dipeptidyl peptidase 4 (DPP-4). Furthermore, it was the aim to study the inhibitory mechanism and stability towards ACE and DPP-4 activity. Analysis of〈 10 kDa hydrolysates showed that gastrointestinal proteases generated peptides with clear radical scavenging activity and DPP-4 and ACE inhibiting activity as well. Hydrolysates from pepsin digestion exhibited the lowest ECso values for radical scavenging activity and ACE inhibition, whereas ECso increased in hydrolysates after subsequent digestion with pancreatic and mucosal proteases. Interestingly, ECso values for the DPP-4 inhibition were hardly affected by sequential digestion. Inhibition modes for the muscle hydrolysates were both competitive and non-competitive, but prolonged incubation showed that the inhibitory properties were unstable and therefore they were probably digested as competitive substrates by gastrointestinal proteases.

文摘Presently, there is no clear consensus on the best approach to estimate carotenoid bioavailability. The best alternative would be to use human studies, but they are labour-intensive and expensive and can only be used to investigate a limited number of samples. Hence, a number of in vitro: models have been developed to study pre-absorptive processes and factors affecting bioavailability. The question is, however, how well the results obtained by the various methods correlate to each other and to the in vivo situation. In the present paper, we have compared in vivo data from two human studies on differently processed soups containing carrots, tomato and broccoli, with results obtained by in vitro characterisation of the same soups. In vitro bioaccessibility was estimated by a static in vitro digestion investigating matrix release and micellarization of carotenoids and by uptake studies in a human intestinal cell line (Caco-2). In vivo data was obtained from clinical studies measuring total plasma carotenoid concentrations in human subjects after 4 weeks daily consumption of the soups. Comparison of the in vitro and in vivo results indicate that the combination of a two-step in vitro digestion and Caco-2 cells seems to be a useful tool for estimation of β-carotene bioaccessibility and screening of factors governing the release of β-carotene from this type of food. For lycopene the in vitro and in vivo results were less consistent, suggesting that reliable prediction of lycopene bioavailability might be more problematic.

文摘This study was conducted to evaluate the effect of lactic acid bacteria and propionic acid on the fermentation quality, aer- obic stability and in vitro gas production kinetics and digestibility of whole-crop corn based totalmixed ration (TMR) silage. Total mixed ration was ensiled with four treatments: (1) no additives (control); (2) an inoculant (Lactobacillus plantarum) (L); (3) propionicacid (P); (4) propionic acid+lactic acid bacteria (PL). All treatments were ensiled in laboratory-scale silos for 45 days, and then subjected to an aerobic stability test for12 days. Further, four TMR silages were incubated in vitro with buffered rumen fluid to study in vitro gas production kinetics and digestibility. The results indicated that all TMR silages had good fermentation characteristics with low pH (〈3.80) and ammonia nitrogen (NH3-N) contents, and high lactic acid contents as well as Flieg points. Addition of L further improved TMR silage quality with more lactic acid production. Addition of P and PL decreased lactic acid and NH3-N contents of TMR silage compared to the control (P〈0.05). After 12 days aerobic exposure, P and PL silages remained stable, but L and the control silages deteriorated as indicated by a reduction in lactic acid and an increase in pH, and numbers of yeast. Compared to the control, addition of L had no effects on TMR silage in terms of 72 h cumulative gas production, in vitro dry matter digestibility, metabolizable energy, net energy for lactation and short chain fatty acids, whereas addition of PL significantly (P〈0.05) increased them. L silage had higher (P〈0.05) in vitro neutral detergent fiber digestibility than the control silage. The results of our study suggested that TMR silage prepared with whole-crop corn can be well preserved with or without additives. Furthermore, the findings of this study suggested that propionic acid is compatible with lactic acid bacteria inoculants, and when used together, although they reduced lactic acid production of TMR silage, they improved aerobic stability and in vitro nutrients digestibility of TMR silage.

文摘The objective of this study was to investigate the effect of lactic acid bacteria(LAB) inoculums on fermentation quality and in vitro digestibility of corn stover silage.Corn stover was ensiled without(control) or with Lactobacillus plantarum(LP),Enterococcus faecalis(EF),and Enterococcus mundtii(EM) for 45 days.The fermentation characteristics were assessed,and subsequent in vitro dry matter digestibility(DM-D),neutral detergent fiber digestibility(NDF-D),volatile fatty acids(VFA),methane(CH4) production,cellulolytic bacteria proportions and their activities per corn stover silage were also determined.There was no significant difference(P>0.05) among the silage pH,lactic acid,crude protein(CP),water soluble carbohydrates(WSC) and lignocelluloses contents of different treatments.The relative proportions of Ruminococcus flavefaciens and Fibrobacter succinogenes,carboxymethyl-ocellulose and β-glycosidase activities,DM-D,NDF-D,and VFA production of in vitro incubation was higher(P<0.05) for silages inoculated with LP and EF than those of the control silage.Silage inoculated with LP showed the lowest(P<0.05) CH4 production per unit yield of VFA,which was positively corresponded to the lowest(P<0.05) ratio of acetate to propionate.In summary,the ensiling fermentation quality and subsequent utilization of corn stover silage were efficiently improved by inoculated with L.plantarum.

文摘The aim of the current study was to investigate the effects of a lactic acid bacteria inoculant(Lactobacillus plantarum, LP), fibrolytic enzyme(EN), combination of LP and EN(LP+EN) on fermentation quality, nutritive characteristics and in vitro digestibility of total mixed ration(TMR) silages containing 0, 7.5 and 15.0%(on dry matter basis) of rape(Brassica campestris L.) straw(RS)(denoted as CTMR, LTMR and MTMR, respectively). After ensiling for 60 days, TMR silages without additives were well preserved, but MTMR had higher p H than CTMR and LTMR. There were no differences in other parameters of fermentation quality, microbial composition, nutrition and in vitro digestibility between CTMR and LTMR except for yeast and mold number and crude protein(CP) content. CTMR and LTMR silage had higher CP content, in vitro neutral detergent fiber digestibility(IVNDFD) and in vitro acid detergent fiber digestibility(IVADFD), lower acid detergent fiber(ADF) content than MTMR silage. LP and EN decreased p H and increased dry matter(DM) recovery of TMR silages. LP+EN improved the fermentation quality, nutritive characteristics and in vitro digestibility of TMR silages, showed by lower p H, ADF content, higher lactic acid content, in vitro dry matter digestibility(IVDMD) and IVNDFD. Therefore, It was suggested that TMR silage contained 7.5% RS on a DM basis and treated with LP+EN can be as a useful feed for ruminant.

文摘As a non-thermal processing technology,high hydrostatic pressure(HHP)can be used for starch modification without affecting the quality and flavour constituents.The effect of HHP on starch is closely related to the treatment time of HHP.In this paper,we investigated the impacts of HHP treatment time(0,5,10,15,20,25,30 min)on the microstructure,gelatinization and thermal properties as well as in vitro digestibility of oat starch by scanning electron microscopy,X-ray diffraction,Fourier transform infrared spectroscopy,13C NMR and differential scanning calorimeter.Results showed that 5-min HHP treatment led to deformation and decreases in short-range ordered and doublehelix structures of oat starch granules,and further extending the treatment time to 15 min or above caused the formation of a gelatinous connection zone,increase of particle size,disintegration of short-range ordered and double-helix structures,and crystal structure change from A type to V type,indicating gelatinization occurred.Longer treatment time also resulted in the reduction in both the viscosity and the stability of oat starch.These indicated that HHP treatment time greatly influenced the microstructure of oat starch,and the oat starch experienced crystalline destruction(5 min),crystalline disintegration(15 min)and gelatinization(>15 min)during HHP treatment.Results of in vitro digestibility showed that the rapidly digestible starch(RDS)content declined first after treatment for 5 to 10 min then rose with the time extending from 15 to 30 min,indicating that longer pressure treatment time was unfavourable to the health benefits of oat starch for humans with diabetes and cardiovascular disease.Therefore,the 500-MPa treatment time for oat starch is recommended not more than 15 min.This study provides theoretical guidance for the application of HHP technology in starch modification and development of health foods.
