Serum proteins differentially expressed in gestational diabetes mellitus assessed using isobaric tag for relative and absolute quantitation proteomics

BACKGROUND As a well-known fact to the public,gestational diabetes mellitus(GDM)could bring serious risks for both pregnant women and infants.During this important investigation into the linkage between GDM patients and their altered expression in the serum,proteomics techniques were deployed to detect the differentially expressed proteins(DEPs)of in the serum of GDM patients to further explore its pathogenesis,and find out possible biomarkers to forecast GDM occurrence.METHODS Subjects were divided into GDM and normal control groups according to the IADPSG diagnostic criteria.Serum samples were randomly selected from four cases in each group at 24-28 wk of gestation,and the blood samples were identified by applying iTRAQ technology combined with liquid chromatography-tandem mass spectrometry.Key proteins and signaling pathways associated with GDM were identified by bioinformatics analysis,and the expression of key proteins in serum from 12 wk to 16 wk of gestation was further verified using enzyme-linked immunosorbent assay (ELISA).RESULTS Forty-seven proteins were significantly differentially expressed by analyzing the serum samples between the GDMgravidas as well as the healthy ones. Among them, 31 proteins were found to be upregulated notably and the rest16 proteins were downregulated remarkably. Bioinformatic data report revealed abnormal expression of proteinsassociated with lipid metabolism, coagulation cascade activation, complement system and inflammatory responsein the GDM group. ELISA results showed that the contents of RBP4, as well as ANGPTL8, increased in the serumof GDM gravidas compared with the healthy ones, and this change was found to initiate from 12 wk to 16 wk ofgestation.CONCLUSION GDM symptoms may involve abnormalities in lipid metabolism, coagulation cascade activation, complementsystem and inflammatory response. RBP4 and ANGPTL8 are expected to be early predictors of GDM. 展开更多

We systematically calculated the multinucleon transfer reactions of ^(208)Os,^(208)Pt,^(208)Hg,^(208)Pb,^(208)Po,^(208)Rn,^(208)Ra,and ^(132,136) Xe when bombarded on ^(232) Th and ^(248) Cm at Coulomb barrier energies within the dinuclear system model.These results are in good agreement with the available experimental data.The influence of Coulomb and shell effects on actinide production in these reactions has been rigorously studied.We calculated and analyzed the potential energy surface (PES) and total kinetic energy (TKE) mass distributions for the reactions involving ^(208)Hg,^(208)Pb,and ^(208) Po with ^(248) Cm and ^(232)Th.The PES and TKE spectra shed light on the fragment formation mechanisms in multinucleon transfer reactions,with clear indications of isospin and shell effects.The production cross sections for multinucleon transfer products show a strong dependence on isobar projectiles with a mass number A=208.Isobar projectiles with high N/Z ratios are advantageous for generating neutron-rich target-like fragments.Conversely,products induced by isobar projectiles with larger charge numbers tend to shift toward proton-rich regions.The intertwining of the Coulomb potential and shell effect is evident in the production cross sections of actinide isotopes.Drawing from reactions induced by radioactive projectiles,we anticipate the discovery of several new actinide isotopes near the nuclear drip lines,extending our reach into the superheavy nuclei domain. 展开更多

Two 1-D dynamical and isothermal models of cathode gas diffusion layer(GDL) with isobaric and non-isobaric operations for polymer electrolyte fuel cells(PEFCs) were developed and implemented in COMSOL Multiphysics v3.5.The artificial diffusion coefficient was introduced as well to make the numerical computation be stable.In the non-isobaric model,the pressure of gas mixture was obtained by summing up the governing equations of gaseous components,instead of Navier-Stoks equation.Comparison of the two models were carried out with the steady-states and dynamical simulations under given conditions.The corresponding analysis based on the simulated results was also given simultaneously.This paper is contributed to finding the differences between the isobaric and non-isobaric operation in the two-phase model of cathode GDL. 展开更多

However, no studi... Kai-Xin-San consists of Ginseng Radix, Polygalae Radix, Acori Tatarinowii Rhizoma, and Poria at a ratio of 3:3:2:2. Kai-Xin-San has been widely used for the treatment of emotional disorders in China. However, no studies have identified the key proteins implicated in response to Kai-Xin-San treatment. In this study, rat models of chronic mild stress were established using different stress methods over 28 days. After 14 days of stress stimulation, rats received daily intragastric administrations of 600 mg/kg Kai-Xin-San. The sucrose preference test was used to determine depression-like behavior in rats, while isobaric tags were used for relative and absolute quantitation-based proteomics to identify altered proteins following Kai-Xin-San treatment. Kai-Xin-San treatment for 2 weeks noticeably improved depression-like behaviors in rats with chronic mild stress. We identified 33 differentially expressed proteins: 7 were upregulated and 26 were downregulated. Functional analysis showed that these differentially expressed proteins participate in synaptic plasticity, neurodevelopment, and neurogenesis. Our results indicate that Kai-Xin-San has an important role in regulating the key node proteins in the synaptic signaling network, and are helpful to better understand the mechanism of the antidepressive effects of Kai-Xin-San and to provide objective theoretical support for its clinical application. Sensory and motor nerve fibers of peripheral nerves have different anatomies and regeneration functions after injury. To gain a clear understanding of the biological processes behind these differences, we used a labeling technique termed isobaric tags for relative and absolute quantitation to investigate the protein profiles of spinal nerve tissues from Sprague-Dawley rats. In response to Wallerian degeneration, a total of 626 proteins were screened in sensory nerves, of which 368 were upregulated and 258 were downregulated. In addition, 637 proteins were screened in motor nerves, of which 372 were upregulated and 265 were downregulated. All identified proteins were analyzed using the Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis of bioinformatics, and the presence of several key proteins closely related to Wallerian degeneration were tested and verified using quantitative real-time polymerase chain reaction analyses. The differentially expressed proteins only identified in the sensory nerves were mainly relevant to various biological processes that included cell-cell adhesion, carbohydrate metabolic processes and cell adhesion, whereas differentially expressed proteins only identified in the motor nerves were mainly relevant to biological processes associated with the glycolytic process, cell redox homeostasis, and protein folding. In the aspect of the cellular component, the differentially expressed proteins in the sensory and motor nerves were commonly related to extracellular exosomes, the myelin sheath, and focal adhesion. According to the Kyoto Encyclopedia of Genes and Genomes, the differentially expressed proteins identified are primarily related to various types of metabolic pathways. In conclusion, the present study screened differentially expressed proteins to reveal more about the differences and similarities between sensory and motor nerves during Wallerian degeneration. The present findings could provide a reference point for a future investigation into the differences between sensory and motor nerves in Wallerian degeneration and the characteristics of peripheral nerve regeneration. Objective:The objective of the study is to examine the possible mechanism by which Guasha(scraping therapy)affects the expression profiles of proteins in a lumbar disc herniation(LDH)rat model using isobaric tags for relative and absolute quantitation(iTRAQ)-based proteomics.Methods:Thirty-six rats were used in this study.LDH rats were subjected to noncompressive LDH surgeries.Rats were randomly divided into the model and Guasha groups.Guasha was applied on alternate days for a total of nine times(three courses).At the end of each course,six rats were randomly selected from each group and their blood samples were collected.iTRAQ labeling was used to examine the mechanism of action of Guasha against LDH.The molecular functions,cellular components,and biological processes were analyzed using gene ontology analysis.The Ingenuity Pathway Analysis database was used to identify canonical pathways involving these proteins.Results:Compared to the model group,198,182,and 170 proteins were identified as differentially expressed at the three respective Guasha treatment courses.Pathways,including focal adhesion kinase signaling,acute-phase response signaling,and the LXR/RXR activation pathway,were closely related to the effects of Guasha in LDH rats.Furthermore,Rac1,Orm1,and Hpx were validated by western blotting,and the results were consistent with the protein expression levels observed using the iTRAQ method.Conclusion:Guasha could not only regulate the pathological changes related to LDH,but also achieve therapeutic effects by stimulating physiological changes.Our results offer a better understanding of the effects of Guasha on LDH. 展开更多

In the past decade,relative proteomic quantification using isobaric labeling technology has developed into a key tool for comparing the expression of proteins in biological samples.Although its multiplexing capacity and flexibility make this a valuable technology for addressing various biological questions,its quantitative accuracy and precision still pose significant challenges to the reliability of its quantification results.Here,we give a detailed overview of the different kinds of isobaric mass tags and the advantages and disadvantages of the isobaric labeling method.We also discuss which precautions should be taken at each step of the isobaric labeling workflow,to obtain reliable quantification results in large-scale quantitative proteomics experiments.In the last section,we discuss the broad applications of the isobaric labeling technology in biological and clinical studies,with an emphasis on thermal proteome profiling and proteogenomics. 展开更多

Using the ground-state mass of ^(52)Ni and two-proton decay energy of ^(54)Zn,the ground-state mass excess of ^(54)Zn is deduced to be-6504(85)keV.This value is about 2 MeV lower than the prediction of the quadratic form of the isobaric multiplet mass equation(IMME).A cubic fit to the existing mass data of the A=54,T=3 isospin multiplet yields a surprisingly large d coefficient of IMME,i.e.,d=18.6(27),being 6.9σdeviated from zero,and the resultant|b/c|ratio significantly deviates from the systematics.This phenomenon is analyzed in this study,and we conclude that the breakdown of the quadratic form of IMME could be likely due to the mis-assignment of the T=3 isobaric analog state(IAS)in the T_(Z)=1 nucleus ^(54)Fe or extremely strong isospin mixing. 展开更多

目的:探讨单纯髓核摘除联合应用Isobar半限制性动态固定系统固定治疗单节段腰椎间盘突出症伴节段性不稳的临床效果。方法:2009年1月~2010年12月共收治32例单节段腰椎间盘突出症伴节段性不稳的腰腿痛患者,均有相应节段神经受压引起的... 目的:探讨单纯髓核摘除联合应用Isobar半限制性动态固定系统固定治疗单节段腰椎间盘突出症伴节段性不稳的临床效果。方法:2009年1月~2010年12月共收治32例单节段腰椎间盘突出症伴节段性不稳的腰腿痛患者,均有相应节段神经受压引起的腿痛、麻木、无力,均行保守治疗3~6个月治疗无效。责任节段分别为L3/4节段2例,L4/5节段16例,L5/S1节段14例。均在全麻下行后路腰椎间盘髓核摘除、Isobar半限制性动态固定系统固定手术。采用VAS和ODI评价临床疗效,术后3个月及末次随访行X线正侧位片、动力位片检查及CT扫描观察内固定情况和固定节段活动度。结果:所有患者均完成3年临床随访,平均手术时间70±16min,平均出血量150±42ml。术前VAS评分平均为7.3±2.2分;术后3个月时为2.2±1.0分,末次随访时为1.8±0.9分,均较术前显著减轻(P〈0.05)。ODI术前为(59.7±13.2)%;术后3个月时为(32.6±11.4)%,末次随访时为(30.8±10.6)%,较术前明显改善(P〈0.05)。责任节段术前活动度14°±2.5°,术后3个月时为2.8°±1.5°,末次随访时为2.6°±1.3°,与术前比较差异有显著性(P〈0.05)。所有患者无钉棒断裂,固定节段无再发椎间盘突出。结论:髓核摘除联合Isobar半限制性动态固定系固定治疗单节段腰椎间盘突出症伴节段性不稳能够有效缓解临床症状,同时保留了责任节段部分活动度。 展开更多

目的比较Isobar动态固定联合腰椎融合与椎弓根内固定联合椎间融合术治疗腰椎退行性变的临床疗效。方法选择于2012年11月至2013年11月间在本院接受治疗的腰椎退行性变患者106例作为研究对象,按照随机数表法将所有患者分为观察组和对照组... 目的比较Isobar动态固定联合腰椎融合与椎弓根内固定联合椎间融合术治疗腰椎退行性变的临床疗效。方法选择于2012年11月至2013年11月间在本院接受治疗的腰椎退行性变患者106例作为研究对象,按照随机数表法将所有患者分为观察组和对照组各53例,观察组接受Isobar动态固定联合腰椎融合术治疗,对照组接受椎弓根内固定联合椎间融合术治疗,比较两组患者的腰椎功能及疼痛评分、腰椎总活动度、邻近节段活动度和炎症因子水平等差异。结果观察组患者接受治疗后的Oswestry功能障碍指数(ODI)低于对照组,JOA评分高于对照组,差异均有统计学意义(P<0.05);观察组患者接受治疗后的腰椎总活动度大于对照组,邻近节段活动度小于对照组,差异均有统计学意义(P<0.05);观察组患者接受治疗后的C反应蛋白(CRP)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)等炎症因子水平值均明显低于对照组,差异均有统计学意义(P<0.05)。结论 Isobar动态固定联合腰椎融合治疗可以有效改善腰椎退行性变患者的腰椎功能,优化腰椎活动度,降低全身炎症水平。 展开更多

背景:对于腰椎间盘突出症患者采取后路内固定治疗,可以常规选择后路椎间融合钉棒系统内固定的方式,近年来Isobar非融合内固定系统作为一种半坚强内固定腰椎后路动态钉棒固定系统开始逐渐应用在临床,成为患者的治疗选择方式之一。目的:... 背景:对于腰椎间盘突出症患者采取后路内固定治疗,可以常规选择后路椎间融合钉棒系统内固定的方式,近年来Isobar非融合内固定系统作为一种半坚强内固定腰椎后路动态钉棒固定系统开始逐渐应用在临床,成为患者的治疗选择方式之一。目的:临床对比观察研究分析采取后路Isobar非融合内固定系统和传统椎间融合钉棒系统内固定治疗腰椎间盘突出症的临床疗效。方法:对2011年9月至2012年9月收治解放军第二军医大学附属长海医院骨科进行手术治疗的腰椎间盘突出症患者40例,分为Isobar组和坚强内固定组,各20例,分别采用后路Isobar非融合内固定系统和传统椎间融合钉棒系统内固定进行治疗。结果与结论:两组患者均顺利完成手术,并完成至少2年以上随访,无脱落。与内固定前相比,两组患者内固定后腰腿痛目测类比评分、Oswestry功能障碍指数功能指数均显著改善;且与坚强内固定组相比,Isobar组患者治疗后腰椎活动度较大,而两组患者腰腿痛目测类比评分和Oswestry功能障碍指数接近。说明采用后路Isobar非融合内固定治疗能够获得传统治疗相同的治疗效果,且在腰椎活动度方面更具有优势。 展开更多

背景:双节段椎间盘突出症合并腰椎不稳的患者需要对严重退变节段行坚强固定和融合,但相邻节段会出现运动位移和屈伸角度的过度性增大,出现相邻节段的退变加速,椎体不稳加重,关节突增生、椎间盘突出等情况长期困扰着临床医生及患者。目的... 背景:双节段椎间盘突出症合并腰椎不稳的患者需要对严重退变节段行坚强固定和融合,但相邻节段会出现运动位移和屈伸角度的过度性增大,出现相邻节段的退变加速,椎体不稳加重,关节突增生、椎间盘突出等情况长期困扰着临床医生及患者。目的:对比观察ISObar TTL动态固定系统与坚强钉棒固定系统治疗双节段腰椎椎间盘突出合并腰椎不稳的疗效。方法:纳入130例双节段腰椎椎间盘突出合并腰椎不稳患者,其中L3/4-L4/5患者72例,L4/5-L5/S1患者58例。对退变严重的节段行椎板减压后外侧植骨融合,对退变较轻的节段随机采取ISObar TTL动态固定(68例,设为试验组)和坚强钉棒固定系统(62例,设为对照组)。2组患者对治疗方案均知情同意,且得到医院伦理委员会批准。以死亡终点,至少随访8年,术前及术后1周、4年、8年分别使用改良Pfirrmann分级系统评估椎间盘退变情况,使用目测类比评分系统评估腰腿疼痛改善情况。结果与结论:(1)与术前相比,试验组与对照组患者术后1周、4年、8年的腰痛及腿痛目测类比评分均明显改善(P<0.05)。组间对比,2组术前及术后1周腰痛及腿痛目测类比评分差异无显著性意义(P>0.05);但是术后4,8年试验组腰痛及腿痛测类比评分明显低于对照组(P<0.05);(2)与术前相比,试验组与对照组患者术后1周、4年、8年的融合节段椎间盘退变改良Pfirrmann分级评分无明显变化(P>0.05);组间对比,2组术前及术后1周、4年、8年融合节段椎间盘退变改良Pfirrmann分级评分差异无显著性意义(P>0.05);(3)与术前相比,试验组与对照组患者术后1周、4年、8年的邻近节段椎间盘退变改良Pfirrmann评分均明显改善(P<0.05)。组间对比,2组术前及术后1周邻近节段椎间盘退变改良Pfirrmann评分差异无显著性意义(P>0.05);术后4,8年,试验组邻近节段椎间盘退变改良Pfirrmann评分均显著低于对照组(P<0.05);(4)表明随访8年期间,ISObar TTL动态固定系统与坚强钉棒系统治疗双节段腰椎椎间盘突出症的症状均明显改善;但是术后4,8年ISObar TTL动态固定系统缓解腰腿痛效果更明显,且能够减轻邻近节段椎间盘退变。 展开更多

AIM: To preliminarily test proteomics in aqueous humor in patients with dry age-related macular degeneration(AMD) by using the proteomic technology.METHODS: Aqueous humor samples were collected from patients with or without dry AMD, who underwent cataract surgery. The aqueous samples were analyzed with isobaric tags for relative and absolute quantification(i TRAQ) combined with liquid chromatography tandem mass spectrometry(LC-MS/MS) technology. The differential expressed proteins were analyzed with gene ontology(GO) enrichment, Kyoto Encyclopedia of Genes and Genomes(KEGG) and protein-protein interaction(PPI) network analysis. The data were partly validated by ELISA and Western blot. False discovery rate(FDR) was used for statistical analysis. RESULTS: A total of 244 proteins were detected, in which 38 proteins were up-regulated and 51 were down-regulated significantly in patients with dry AMD compared with that in control groups(FDR value <1.0%). Several proteins, e.g., protein S100-A8(S10 A8), dystroglycan(DAG1), Ig alpha-1 chain C region(IGHA1), carbonic anhydrase 3(CAH3) and alpha-1-acid glycoprotein(A1 AG1) were increased more than 5 times of that in control group. The bioinformatics analysis showed that dry AMD is closely associated with inflammation or immune reaction, oxidative stress, blood coagulation and remodeling of extracellular matrix.CONCLUSION: i TRAQ-based proteomic analysis of aqueous humor demonstrate the differential expressions of proteins between dry AMD and control groups, providing the clues to understand the mechanisms and possible treatments of dry AMD. 展开更多
