Drought has severely affected the yield and quality of commercial crops.The BRI1 family plays an important role in plant response to drought stress,and BRL3 gene plays an important role in the study of drought in Arabidopsis thaliana.In this study,NtBRL3 was constructed as a vector and genetically transformed to obtain‘N.Tobacco K326’overexpression of NtBRL3.The enzyme activities of transgenic tobacco and wild-type tobacco were measured and transcriptome and metabolome analyses were performed.The results showed that the antioxidant enzymes of transgenic tobacco were more active under drought conditions,and 85 significantly differentially metabolites and 106 significantly differentially expressed genes were identified in the metabolome and transcriptome analyses,respectively.Transgenic tobacco NtBRL3ox demonstrated an excessive accumulation of droughtrelated metabolites,sugars such as sucrose and maltotetraose,and amino acids such as proline,compared with WT.We discovered drought-related differential genes in the root transcriptome,among which LOX6,RD22,WSD1,CCD8,and UGT were key genes which play an important role in plant response to drought stress.Our results demonstrate that NtBRL3 overexpression in K326 enhances drought resistance in transgenic tobacco. 展开更多

Monosomic lines of Nicotiana tabacum are helpful to confirm the location of genes on specific chromosomes. In the cross N. nudicaulis and N. tabacum, hybrid seedlings express lethal symptoms, which are controlled by the S subgenome of N. tabacum. To identify the responsible chromosome, we needed to produce chromosome lacking lines (CLLs) of N. tabacum L. “Red Russian” and use them to cross with N. nudicaulis. From a cross of (N. tabacum × N. tomentosiformis) × N. tabacum, 380 BC1 individuals were obtained. Using a Haplo-Q line (a monosomic line lacking the single linkage group 11) and N. tabacum, we found that qPCR is a simple and reliable screening method for CLLs of N. tabacum. The marker PT30342 is located on linkage group 11, and the -Ct value (Ct Actin - Ct PT30342) was 2.0 for a disomic line and was 1.097 for a Haplo-Q line. By the use of flow cytometry, qPCR and chromosome counting together as a screening method, we identified 6 CLLs lacking 2 to 6 chromosomes. Compared with conventional methods, our method is a rapid technique for making and screening CLLs ofthe S or S/T subgenome of N. tabacum. Further, these CLLs will be useful to identify the location of two or more factors on chromosomes controlling a variety of genetic problems affecting breeding. Here, we only made CLLs of the S or S/T subgenome of N. tabacum. We will use the method established in this study to produce CLLs of the T subgenome of N. tabacum, and gather a full set of CLLs of N. tabacum. qPCR could also be applied to the identification of chromosome aberrations in other plants. 展开更多

Investigations into the potential application of nanoparticles acting as nanofungicides in sustainable agriculture are rapidly expanding due to the high antimicrobial properties of these compounds,which do not risk inducing pathogen resistance to fungicides.A detailed understanding of the impact of copper oxide nanoparticles(CuO NPs)on soil-borne phytopathogenic fungi is yet to be obtained.This study aimed to explore the in vitro antifungal activity and control efficacy of CuO NPs applied via irrigation with respect to tobacco black shank(TBS)disease caused by Phytophthora nicotianae.The results revealed that CuO NPs greatly interfered with the reproductive growth process of this fungus,repressing hyphal growth,spore germination and sporangium production.Additionally,morphological damage,intracellular ROS accumulation and increased SOD enzyme activity in hyphae were the antifungicidal mechanisms of these NPs.In pot experiments,treatment with CuO NPs at 100 mg L^(–1)significantly suppressed TBS development,compared with the effect on control plants,and the control efficacy reached 33.69%without inducing phytotoxicity.Exposure to CuO NPs significantly activated a series of defense enzymes,and resistance genes in tobacco can further explain the mechanisms by which CuO NPs suppressed fungal infection.The Cu content in both the leaves and roots of P.nicotianae-infested plants increased by 50.03 and 27.25%,respectively,after treatment with 100 mg L^(–1)CuO NPs,compared with that of healthy plants.In particular,a higher Cu content was observed in infected roots than in leaves.Therefore,this study showed the potential of CuO NPs applied as nanofungicides and as nanoinducers of fungus resistance genes for the management of TBS through inhibition of pathogen infection and stimulation of plant defenses. 展开更多

由于烟草的分子标记开发和遗传图谱构建十分困难,迄今烟草中有关数量性状基因座(QTL)的定位研究仍非常有限。本研究利用一个由207个株系组成的烤烟DH群体及基于该群体所构建的含有24个连锁群、611个SSR标记,总长为1882.1cM的遗传图谱... 由于烟草的分子标记开发和遗传图谱构建十分困难,迄今烟草中有关数量性状基因座(QTL)的定位研究仍非常有限。本研究利用一个由207个株系组成的烤烟DH群体及基于该群体所构建的含有24个连锁群、611个SSR标记,总长为1882.1cM的遗传图谱,采用复合区间作图方法,对株高(PH)、茎围(SG)、节距(IL)、叶片数(LN)、最大腰叶长(LWL)和最大腰叶宽(WWL)6个与叶片产量有关的农艺性状进行QTL定位分析。共检测到69个QTL,大部分QTL的效应值较小,仅有4个具有较大的效应值,可解释大约15%~20%的表型变异。6个性状之间大多彼此相关。与此相符,在基因组中发现存在许多小区域,每个区域包含2个或2个以上紧密连锁的不同性状的QTL。 展开更多

超量表达 CsADH17 基因提高烟草抗旱性及香气成分分析

乙醇脱氢酶是植物香气物质合成的脂肪酸代谢等途径中的关键酶之一,对茶叶芳香物质的形成有着重要作用,因此探究CsADH17基因有利于茶树的生长发育与品质提升。以本课题组前期筛选的CsADH17基因为研究对象,克隆CsADH17基因并构建植物表达... 乙醇脱氢酶是植物香气物质合成的脂肪酸代谢等途径中的关键酶之一,对茶叶芳香物质的形成有着重要作用,因此探究CsADH17基因有利于茶树的生长发育与品质提升。以本课题组前期筛选的CsADH17基因为研究对象,克隆CsADH17基因并构建植物表达载体,遗传转化烟草,对转基因烟草非生物胁迫下CsADH17的表达量和相关生理指标进行测定与分析。结果表明,对转基因与野生型烟草进行模拟干旱胁迫后,转基因烟草植株在干旱胁迫处理下的CsADH17基因相对表达量比野生型植株高。野生型烟草植株和CsADH17转基因烟草植株在干旱胁迫处理21 d后,超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)和抗坏血酸过氧化物酶(APX)的活性均高于野生型烟草;胁迫处理后,野生型烟草叶片中丙二醛(MDA)含量显著高于转基因烟草。香气成分分析,转基因烟草苯甲醛比野生型含量提高了44.4%,而烟碱含量比野生型提高不到5%。CsADH17基因在烟草干旱胁迫下具有抗旱作用,同时影响烟草的香气成分,为进一步揭示CsADH在茶树生长发育以及抗逆响应中提供参考。 展开更多

烟草西柏烷二萜醇类突变体的理化特征及其突变机制解析

以烟草突变体及其野生型为材料,通过测定农艺性状、腺毛密度、西柏烷二萜醇及其代谢前体物含量、西柏烷二萜醇类代谢关键基因的表达量,分析了突变体的生理生化和代谢差异,并结合基因表达差异初步解析了其突变的生理与分子机制。结果表明... 以烟草突变体及其野生型为材料,通过测定农艺性状、腺毛密度、西柏烷二萜醇及其代谢前体物含量、西柏烷二萜醇类代谢关键基因的表达量,分析了突变体的生理生化和代谢差异,并结合基因表达差异初步解析了其突变的生理与分子机制。结果表明,突变体西柏三烯一醇和西柏三烯二醇含量分别是野生型的2.63倍和4.03倍,均极显著高于野生型;突变体分泌型腺毛密度为757.38条/cm 2,极显著大于野生型;突变体西柏烷二萜醇类代谢前体物IPP和GGPP分别为849.83 ng/g、298.29 ng/g,与野生型间的差异不显著;突变体西柏三烯一醇合成酶基因NtCYC-1、西柏三烯二醇合成酶基因NtCYP71D16的表达量分别是野生型的17.77倍和20.89倍,均极显著高于野生型。研究表明,突变体是一个培育优质、抗虫烤烟新品种的优良材料;其发生突变的原因是NtCYC-1、NtCYP71D16基因发生表达上调,促进了西柏三烯一醇、西柏三烯二醇的合成反应,同时,突变体的分泌型腺毛密度更高,为西柏烷二萜醇类的合成提供了更多的场所,使得西柏烷二萜醇类的含量显著提高。 展开更多

Vacuolar ATPase is the key enzyme in the aspect of plant stress,which contains 13 subunits.In this study,VHA-c which was cloned from the salt-tolerant plants Iris lacteal was transformed into tobacco,and then treated the transgenic tobacco with 0.7% NaCl for 7 days.Detected the MDA content,the SOD activity and the POD activity.Moreover,the transgenic plants displayed obviously better growth than the normal plants under salt stress.These results suggested that VHA-c could confer the stress tolerance through enhancing the POD and the SOD activities,and by protecting membranes from damage by decreasing lipid peroxidation under salt stress. 展开更多

为揭示烟草脱落酸(ABA)受体基因NtPYR1和NtPYL9的生物学功能,以拟南芥PYR1和PYL9基因为探针,在NCBI中通过同源比对查找普通烟草表达序列标签(Expressed sequence tag,EST)序列,将EST序列进行拼接并设计特异性引物,从普通烟草红花大金元c... 为揭示烟草脱落酸(ABA)受体基因NtPYR1和NtPYL9的生物学功能,以拟南芥PYR1和PYL9基因为探针,在NCBI中通过同源比对查找普通烟草表达序列标签(Expressed sequence tag,EST)序列,将EST序列进行拼接并设计特异性引物,从普通烟草红花大金元c DNA文库中克隆出NtPYR1和NtPYL9基因,其开放阅读框长分别为678 bp和564 bp,分别编码225个和187个氨基酸。NtPYR1和NtPYL9蛋白都具有一个保守的疏水性结构域,同属于PYR/PYL/RCAR ABA受体基因家族,NtPYR1和NtPYL9与番茄同源蛋白的相似性分别达到79.10%和95.24%。遗传聚类分析结果显示:NtPYR1和NtPYL9与番茄和马铃薯同源基因的遗传进化距离最近,说明PYR1和PYL9在茄科植物中较为保守。定量PCR对烟草不同组织表达模式分析表明,NtPYR1和NtPYL9在烟草不同组织中均有表达,分别在子叶和须根中的表达量最高。NtPYR1和NtPYL9表达均受ABA抑制,同时受H2O2诱导,说明两基因可能参与烟草的抗逆生理过程。 展开更多

Quality Characteristics of Tobacco Leaves with Different Aromatic Styles from Guizhou Province,China 被引量:8

The relationships between chemical components and quality indexes were studied in the tobacco leaves with different aromatic styles. A total of 16 chemical components, 4 quality indexes, and 6 smoking quality indexes from 366 tobacco leaf samples with 4 different types of aroma from Guizhou Province, China, were subjected to principal component analysis and stepwise regression analysis. The tobacco leaves with different types of aroma showed remarkable difference in the contents of chemical components, quality indexes, and smoking quality indexes. The first principal factors (carbohydrates and nitrogen-containing compounds) of the chemical composition of the leaf were similar among different types of aroma, which showed that the quality of the leaf was mainly influenced by carbohydrates and nitrogen-containing compounds and their ratios. The factors for the second through the seventh principal components varied largely among various aromatic types, suggesting the contribution of other chemical components to the leaf quality. In addition, the smoking quality of four different aromatic leaves showed significant correlation with the different chemical components. The quality of tobacco leaves with different types of aroma was influenced by multiple factors, especially ecological conditions and culture techniques, which may provide guidance for directive cultivation of high-quality tobacco leaves. 展开更多

The constitutive promoter of cauliflower mosaic virus 35S (CaMV 35S) is a polar unidirectional promoter and is widely used in plant genetic engineering. In the present study, the unidirectional CaMV 35S promoter has been modified to a bi-directional promoter by fusing its minimal promoter element to the 5′end of CaMV 35S promoter in the opposite orientation. To qualitatively and quantitatively esti-mate its bi-directional transcriptional function and activity, two visible reporter genes, gusA (β-glucuronidase, GUS) and gfp (green fluo-rescent protein, GFP), were fused to the two ends of the promoter in bi-orientations ending with NOS terminator sequences, respectively. Stable expression of gusA and gfp genes in transgenic tobacco (Nicotiana tabacum L.) was visulized by histochemically staining for GUS and fluorescence microscopic observation under UV for GFP in transgenic plants. The expression of two reporter genes showed that the constructed bi-directional promoter did have the bi-directional transcriptional function in both expected orientations. The quantitative estimation of GUS and GFP were determined on a HITACHI F1000 Fluorescence Spectrophotometer with various wavelengths of excita-tion and emission. The GUS activity varied from 8 to 250 pmol 4-MU / min / mg protein and the GFP content varied from 0.9 to 1.8 μg / mg protein in various lines of transgenic tobacco plants. Higher GUS activity generally coupled with lower GFP content, and vice versa. 展开更多

Ethiopian guizotia showed very high K-enrichment capability atdifferent soil K levels, and the K content in its dry matter was over 110 mg kg-1 when soil K was fullysupplied, and about 60 mg kg-1 when no K fertilizer was applied. For alligator alternanthera, the capabilityto accumulate K was closely related with its growth medium. When it was grown on soils, both the K contentand K uptake rate of the plant were similar to those of tobacco. Evident K dep1etion was observed in therhizosphere of all plant species, and the depletion rate was related to the capability of K enrichment of plant. 展开更多

为了提高北方地区黄精栽培收益,以黄精单作+覆盖遮阳网为对照,以烤烟、玉米、白蜡树与黄精间作为处理,连续4 a调查黄精成活率、株高和病虫害发生情况,第4年收获黄精时,检测土壤酶活性,调查黄精及间作作物的产量及综合产值,从而筛选适合... 为了提高北方地区黄精栽培收益,以黄精单作+覆盖遮阳网为对照,以烤烟、玉米、白蜡树与黄精间作为处理,连续4 a调查黄精成活率、株高和病虫害发生情况,第4年收获黄精时,检测土壤酶活性,调查黄精及间作作物的产量及综合产值,从而筛选适合与黄精间作的经济作物。结果表明:黄精与玉米、烤烟、白蜡树间作,能不同程度提高黄精产量,优化黄精生长环境;4种种植模式中,黄精与烤烟间作模式的黄精主要病虫害(叶斑病、白绢病、地老虎、蛴螬等)发病率低,同时,土壤酸性磷酸酶和蔗糖酶活性增加,对土壤修复有一定的积极作用;黄精和烤烟间作时,黄精产量(干重)达459.7kg/667m^(2),明显高于遮阳网处理(314.6 kg/667m^(2)),总收益达32057.4元/667m^(2)。由此表明,黄精和烤烟间作有利于提升黄精产量,总收益高,二者间作能合理利用光照和土壤资源,降低黄精连作对土壤造成的不利影响,值得在生产上推广和应用。 展开更多

为研究干旱胁迫对烟草幼苗根、茎、叶生理特性的影响,对烟草幼苗进行20%PEG6000干旱胁迫处理,于胁迫处理2 h、24 h后进行根、茎、叶3个部位的取样,通过测定干旱胁迫下烟草幼苗的生理指标了解其抗旱特性。结果表明:根、茎、叶3个部位的... 为研究干旱胁迫对烟草幼苗根、茎、叶生理特性的影响,对烟草幼苗进行20%PEG6000干旱胁迫处理,于胁迫处理2 h、24 h后进行根、茎、叶3个部位的取样,通过测定干旱胁迫下烟草幼苗的生理指标了解其抗旱特性。结果表明:根、茎、叶3个部位的超氧化物歧化酶(superoxide dismutase,SOD)活性随干旱胁迫时间的增加都呈现出先增加后降低的趋势;根、茎、叶3个部位的过氧化物酶(peroxidase,POD)活性随干旱胁迫时间的增加呈现出不同的变化;烟苗根和茎的过氧化氢酶(catalase,CAT)活性呈现出增加的趋势,叶片的CAT活性随胁迫时间的增加呈现出先升高后降低的趋势;随干旱胁迫时间的增加,根的丙二醛(malondialdehyde,MDA)含量呈现出先增加后降低的变化趋势,烟苗茎和叶的MDA含量随胁迫时间的增加而增加,在胁迫24 h时达到1.09和6.76 nmol/g。生理指标相关性分析表明烟草幼苗中CAT活性与MDA含量呈极显著正相关关系(P<0.01),相关系数为0.91。该研究结果可为进一步研究烟草抗旱性研究提供理论依据。 展开更多

烟草叶部病害是烟草生产中的主要病害,严重影响烟叶的产量和品质,易流行造成重大经济损失。2020—2021年对湖南省各烟区的烟草病害进行了调查,在郴州烟区烟叶上发现了一种新的叶部病害。为了明确引起该叶部病害的病原菌,采集了具有典型... 烟草叶部病害是烟草生产中的主要病害,严重影响烟叶的产量和品质,易流行造成重大经济损失。2020—2021年对湖南省各烟区的烟草病害进行了调查,在郴州烟区烟叶上发现了一种新的叶部病害。为了明确引起该叶部病害的病原菌,采集了具有典型症状的烟草病叶进行病原菌的分离、纯化、菌落形态观察、分子生物学鉴定以及致病性测定。结果表明,该叶部病害是由高粱附球菌Epicoccum sorghinum引起的叶斑病,该病害侵染烟草叶片,病斑形状为椭圆形或不规则形,呈淡褐色或深褐色,病斑周围有明显的黄色晕圈。据查这是湖南省首次发现由E. sorghinum引起的烟草叶斑病。 展开更多

YABBY基因家族是种子植物特有的一类转录因子,具有C 2C 2锌指结构域和YABBY结构域,该家族在植物的发育过程中起重要调控作用。为研究烟草YABBY基因家族的成员及其作用,利用生物信息学技术从烟草基因组数据库中鉴定YABBY基因,并分析其蛋白理化性质、构建系统发育树、分析启动子顺势作用元件等。结果表明:在烟草基因组中一共鉴定到7个NtYABBYs基因,分为5个亚家族,均无跨膜结构,定位于细胞核内,启动子顺式作用分析表明NtYABBY基因含有与植物生长发育、植物激素、防御与胁迫等相关的顺势作用元件。该结果可为烟草NtYABBYs基因家族的进一步研究提供理论基础。 展开更多
