Heat stress in pigs and broilers:role of gut dysbiosis in the impairment of the gut-liver axis and restoration of these effects by probiotics,prebiotics and synbiotics

Heat stress is one of the most challenging stressors for animal production due to high economic losses resulting from impaired animal’s productivity,health and welfare.Despite the fact that all farm animal species are susceptible to heat stress,birds and pigs are particularly sensitive to heat stress due to either lacking or non-functional sweat glands.Con-vincing evidence in the literature exists that gut dysbiosis,a term used to describe a perturbation of commensal gut microbiota,develops in broilers and pigs under heat stress.Owing to the protective role of commensal bacteria for the gut barrier,gut dysbiosis causes a disruption of the gut barrier leading to endotoxemia,which contributes to the typical characteristics of heat stressed broilers and growing and growing-finishing pigs,such as reduced feed intake,decreased growth and reduced lean carcass weight.A substantial number of studies have shown that feeding of probiotics,prebiotics and synbiotics is an efficacious strategy to protect broilers from heat stress-induced gut barrier disruption through altering the gut microbiota and promoting all decisive structural,biochemical,and immunologi-cal elements of the intestinal barrier.In most of the available studies in heat stressed broilers,the alterations of gut microbiota and improvements of gut barrier function induced by feeding of either probiotics,prebiotics or synbiot-ics were accompanied by an improved productivity,health and/or welfare when compared to non-supplemented broilers exposed to heat stress.These findings indicate that the restoration of gut homeostasis and function is a key target for dietary interventions aiming to provide at least partial protection of broilers from the detrimental impact of heat stress conditions.Despite the fact that the number of studies dealing with the same feeding strategy in heat stressed pigs is limited,the available few studies suggest that feeding of probiotics might also be a suitable approach to enhance productivity,health and welfare in pigs kept under heat stress conditions. 展开更多

BACKGROUND Dementia is a chronic progressive neurological disease affecting millions of people worldwide,and represents a relevant economic burden for healthcare systems.Although its pathogenesis is still unknown,recent findings have reported that a dysregulated gut-brain axis communication,a fundamental relationship mediated by several host and microbial molecules,is associated with cognitive disorders.In addition,gut microbiota manipulation reduces neuroinflammation,improving cognitive function by restoring the functional gut-brain axis.AIM To better define the effects of probiotics,prebiotics,synbiotics,and fecal microbiota transplant(FMT)on cognitive function.METHODS We performed a literature search of human randomized clinical trials to examine the effects of the administration of probiotics,prebiotics,synbiotics,or FMT on cognition outcomes in healthy or sick people of every age,sex,and nationality.We systematically searched Embase,Medline/PubMed,Cochrane Library,central and clinicaltrials.gov databases with a combination of comprehensive terms related to cognition and gut microbiota manipulation.Then we carefully reviewed and synthesized the data by type of study design and setting,characteristics of the studied population,kind of intervention(strain type or mixture type,dosage,and frequency of administration),control treatment,inclusion and exclusion criteria,follow-up duration,and cognitive or memory outcomes.RESULTS After examining the titles and abstracts,the initial literature screening identified 995 articles,but we added 23 papers in our systematic review.The analyses of these selected studies highlighted that both probiotic supplementation and FMT improved cognitive function regardless of the type and posology of administration and the adopted cognitive tests and questionnaires.We found that most of the studies conducted in healthy people showed a significant positive effect of the intervention on at least one of the performed cognitive tests.Regarding unhealthy subjects,while FMT and especially probiotic administration had multiple beneficial effects on different cognitive functions,supplementation with prebiotics did not provide any cognitive improvement.CONCLUSION Probiotic supplementation and FMT may represent a promising strategy to restore gut eubiosis and enhance the cognitive functions of healthy people and patients with neurological disorders. 展开更多

Obesity is a complex disease with multiple contributing factors.One of the most intensely studied factors during the past decade has been the gut microbiota,which is the community of all microbes in the intestinal tract.The gut microbiota,via energy extraction,inflammation,and other actions,is now recognized as an important player in the pathogenesis of obesity.Dysbiosis,or an imbalance in the microbial community,can initiate a cascade of metabolic disturbances in the host.Early life is a particularly important period for the development of the gut microbiota,and perturbations such as with antibiotic exposure can have long-lasting consequences for host health.In early life and throughout the life span,diet is one of the most important factors that shape the gut microbiota.Although diets high in fat and sugar have been shown to contribute to dysbiosis and disease,dietary fiber is recognized as an important fermentative fuel for the gut microbiota and results in the production of short-chain fatty acids that can act as signaling molecules in the host.One particular type of fiber,prebiotic fiber,contributes to changes in the gut microbiota,the most notable of which is an increase in the abundance of Bifidobacterium.This review highlights our current understanding of the role of gut microbiota in obesity development and the ways in which manipulating the microbiota through dietary means,specifically prebiotics,could contribute to improved health in the host,including musculo skeletal health. 展开更多

Hepatocellular carcinoma(HCC)is the most prevalent primary malignancy in patients suffering from chronic liver diseases and cirrhosis.Recent attention has been paid to the involvement of the gut-liver axis(GLA)in HCC pathogenesis.This axis results from a bidirectional,anatomical and functional relationship between the gastrointestinal system and the liver.Moreover,the complex network of interactions between the intestinal microbiome and the liver plays a crucial role in modulation of the HCC-tumor microenvironment,contributing to the pathogenesis of HCC by exposing the liver to pathogen-associated molecular patterns,such as bacterial lipopolysaccharides,DNA,peptidoglycans and flagellin.Indeed,the alteration of gut microflora may disturb the intestinal barrier,bringing several toll-like receptor ligands to the liver thus activating the inflammatory response.This review explores the new therapeutic opportunities that may arise from novel insights into the mechanisms by which microbiota immunomodulation,represented by probiotics,and prebiotics,affects HCC through the GLA. 展开更多

Cancer is the leading cause of death and strategies for chemoprevention are important to reduce mortality and morbidity. Probiotics are gaining attention to use as preventive agents. Efficacy of their use as chemopreventive agents was established through research. This review focused on the mechanisms of prebiotics and probiotics action against cancer. Benefits of probiotics against cancer are attributed to competitive exclusion of pathogenic bacteria, direct physical binding to carcinogens, altering intestinal environment to modulate the production enzymes, antioxidant activity and immune modulation. Prebiotics are indigestible food components that could promote the growth of probiotics. Chemopreventive properties of prebiotics are due to their production of short chain fatty acids and enhancing the immunity of the host. Anticarcinogenic properties of pre- and probiotics result from a combination of events rather from a single event. 展开更多

Xylooligosaccharides (XOS) are more effective than other oligosaccharides such as fructo-oligosaccharides as dietary supplements. Chemical methods are preferred to produce XOS mixtures with a wide DP range, while enzymatic methods are preferred in the food or pharmaceutical industries to reduce formation of degradation products. With the growing importance of making fuels fromlignocellulosic biomass (LCM) and the increasing demand for XOS, more opportunities are emerging to utilize xylan-rich component generated in biorefinery into high-value products such as XOS that could further lower the cost of LCM derived biofuels. 展开更多

Effects of common prebiotics on iron status and production of colonic short-chain fatty acids in anemic rats

Prebiotics may enhance iron absorption,and one plausible mechanism involves the production of shortchain fatty acids(SCFA)in the colon by intestinal microflora.The objectives of this study were to determine the effects of common commercially-available prebiotics including fructooligosaccharide(FOS),inulin,FOS-inulin mixture,galactooligosaccharide(GOS),and lactulose on the iron status of anemic rats,and to monitor changes in the production of colonic SCFA.Anemic Sprague-Dawley rats receiving a lowiron diet(12μg Fe/g diet)were supplemented with or without prebiotics(5%m/V in drinking water)for 5 weeks.Hemoglobin concentration in rats supplemented with GOS after 3 weeks(4.3 g/dL)was significantly higher than rats without supplementation(3.7 g/dL),while FOS also significantly increased hemoglobin concentration after 4 weeks(4.1 g/dL vs.3.7 g/dL).All other prebiotics showed no effects.Anemic rats showed lower overall SCFA production in the colon than normal rats,and only FOS signifi cantly increased the production of the three main SCFA(acetic acid,propionic acid and isobutyric acid)identifi ed in anemic rats,with other prebiotics showing no noticeable trends.Our results suggest that GOS and FOS may slightly improve iron status of anemic rats,but the role of SCFA in the colon is not clear. 展开更多

This comprehensive review elucidates the complex interplay between gut microbiota and constipation in Parkinson’s disease(PD),a prevalent non-motor symptom contributing significantly to patients’morbidity.A marked alteration in the gut microbiota,predominantly an increase in the abundance of Proteobacteria and Bacteroidetes,is observed in PD-related constipation.Conventional treatments,although safe,have failed to effectively alleviate symptoms,thereby necessitating the development of novel therapeutic strategies.Microbiological interventions such as prebiotics,probiotics,and fecal microbiota transplantation(FMT)hold therapeutic potential.While prebiotics improve bowel movements,probiotics are effective in enhancing stool consistency and alleviating abdominal discomfort.FMT shows potential for significantly alleviating constipation symptoms by restoring gut microbiota balance in patients with PD.Despite promising developments,the causal relationship between changes in gut microbiota and PD-related constipation remains elusive,highlighting the need for further research in this expanding field. 展开更多

The goal of prebiotic applications from different sources is to improve the gut ecosystem where the host and microbiota can benefit from prebiotics.It has already been recognized that prebiotics have potential roles in the gut ecosystem because gut microbiota ferment complex dietary macronutrients and carry out a broad range of functions in the host body,such as the production of nutrients and vitamins,protection against pathogens,and maintenance of immune system balance.The gut ecosystem is very crucial and can be affected by numerous factors consisting of dietary constituents and commensal bacteria.This review focuses on recent scientific evidence,confirming a beneficial effect of prebiotics on animal health,particularly in terms of protection against pathogenic bacteria and increasing the number of beneficial bacteria that may improve epithelial cell barrier functions.It has also been reviewed that modification of the gut ecosystem through the utilization of prebiotics significantly affects the intestinal health of animals.However,the identification and characterization of novel potential prebiotics remain a topical issue and elucidation of the metagenomics relationship between gut microbiota alteration and prebiotic substances is necessary for future prebiotic studies. 展开更多

Osteoporosis is the most common bone disorder,characterized by low bone mineral density and microarchitectural deterioration of the bone tissue,which increases the susceptibility to fracture.In the past decade,emerging research findings reported the implication of gut microbiota on bone health and osteoporosis pathology.Osteoporotic patients or individuals with a lower bone mineral density exhibit an alteration of the gut microbiota at several taxonomic levels.Additional reports demonstrate that gut microbiota regulates bone metabolism through the modulation of the gut function(mineral availability and absorption,gut integrity),the immune system,and the endocrine system.Thus,based on the vital role of gut microbiota on bone health,it has emerged as a novel therapeutic target for the prevention of bone loss and the treatment of osteoporosis.Microbial-based functional food ingredients,such as probiotics,prebiotics,synbiotics,and fermented foods,have been developed to alter the gut microbiota composition and function and thus,to provide benefits to the host bone health.Despite promising initial results,microbial-based therapies are still under investigation.Moreover,additional animal studies and clinical trials are needed to understand the interactions between gut microbiota and bone metabolism before further applications. 展开更多

Background High dietary carbohydrates can spare protein in rainbow trout(Oncorhynchus mykiss)but may affect growth and health.Inulin,a prebiotic,could have nutritional and metabolic effects,along with anti-inflammatory properties in teleosts,improving growth and welfare.We tested this hypothesis in rainbow trout by feeding them a 100%plant-based diet,which is a viable alternative to fishmeal and fish oil in aquaculture feeds.In a two-factor design,we examined the impact of inulin(2%)as well as the variation in the carbohydrates(CHO)/plant protein ratio on rainbow trout.We assessed the influence of these factors on zootechnical parameters,plasma metabolites,gut microbiota,production of short-chain fatty acids and lactic acid,as well as the expression of free-fatty acid receptor genes in the mid-intestine,intermediary liver metabolism,and immune markers in a 12-week feeding trial.Results The use of 2%inulin did not significantly change the fish intestinal microbiota,but interestingly,the high CHO/protein ratio group showed a change in intestinal microbiota and in particular the beta diversity,with 21 bacterial genera affected,including Ralstonia,Bacillus,and 11 lactic-acid producing bacteria.There were higher levels of butyric,and valeric acid in groups fed with high CHO/protein diet but not with inulin.The high CHO/protein group showed a decrease in the expression of pro-inflammatory cytokines(il1b,il8,and tnfa)in liver and a lower expression of the genes coding for tight-junction proteins in mid-intestine(tjp1a and tjp3).However,the 2%inulin did not modify the expression of plasma immune markers.Finally,inulin induced a negative effect on rainbow trout growth performance irrespective of the dietary carbohydrates.Conclusions With a 100%plant-based diet,inclusion of high levels of carbohydrates could be a promising way for fish nutrition in aquaculture through a protein sparing effect whereas the supplementation of 2%inulin does not appear to improve the use of CHO when combined with a 100%plant-based diet. 展开更多

This study was to identify the effects of prebiotics supplemented in infant formula on enzyme activity and phosphate uptake in the small intestine of Sprague Dawley(SD) rats. Forty-eight healthy SD rats at15 days old(a week before weaning) with similar weight were randomly divided into 3 groups: A(control group), B, C, with 16 rats per group. Rats in groups A, B, C were fed a standard infant formula, the standard infant formula supplemented with oligosaccharides, and the standard infant formula supplemented with polysaccharides, respectively. The feeding test was conducted for 28 d. Compared with group A, the results showed the following: 1) the activities of sucrose and lactase in the small intestine were significantly increased in SD rats of group C(P < 0.05); 2) the relative expressions of lactase gene in the anterior and posterior segments of the small intestine were significantly increased by 1.68 and 2.26 in SD rats of group C(P < 0.05), and the relative expression of Mgam gene in the posterior segment of the small intestine was significantly increased by 0.99 in SD rats of group C(P < 0.05); 3) the relative expressions of Na/Pi-IIb gene in the anterior and posterior segments of the small intestine were significantly increased by 1.85 and 2.28 in SD rats of group C(P < 0.05). These results indicate that the supplementation of infant formula with prebiotics can promote enzyme activity in the small intestine by increasing the relative expression of enzyme gene or by decreasing intestinal injury, and can increase the relative expression of Na/Pi-IIb gene. The effect of polysaccharides is better than that of oligosaccharides. 展开更多

Pure wheat malt beer (PWMB) with potentially functional properties may attract consumer interest.To explore the functional properties of non-starch polysaccharides (NSPs) in PWMB,NSPs were extracted from PWMB and separated into PWMB non-starch polysaccharides-1 (PNSP-1) and PWMB non-starch polysaccharides-2 (PNSP-2) by Q-Sepharose Fast Flow column.PNSP-1 exhibited more effects on increasing the viscosity of beers,which had higher purity of 92.58% containing 88.53% arabinoxylan with arabinose/xylose (A/X) 0.60 and 2.78% β-glucan but no glucomannan and mannan;while PNSP-2 showed greater effects on promoting beer haze and foam retention,which had more diverse NSPs including 46.03% arabinoxylan with A/X 1.13,41.47% arabinogalactan,10.26% glucomannan,and 2.70% mannan but no β-glucan.Diversity of sugar composition resulted in a better antioxidant capacity of PNSP-2.NSPs proliferated Lactobacillus casei LC6117 and LC0331,but did not promote L.plantarum LP-28 and L.fermentium LF-2H.PNSP-2 was also utilized by L.rhamnosus SRG.PNSP-2 had better effects on prebiotics activities.PNSP,PNSP-1 and PNSP-2 all exhibited mild inhibitory effects on α-glucosidase and lipase but not on α-amylase,and showed a binding capacity to fat and cholesterol.This work is of great reference significance for improving the viscosity,haze,foam retention and health value of wheat beer. 展开更多

Vegan consumers look for fresh,tasty,and nutritious products that use eco/animal-friendly ingredients.Prebiotics are substrates selectively utilized by host microorganisms conferring a health benefit.This study aimed to overview the potentially vegan products added with prebiotics,their health benefits,and the main technological and sensory challenges.The main potentially vegan products added with prebiotics studied were based on water-soluble plant-based extracts(fermented beverages,frozen desserts,cheese analog),fruits(fruit juices,fruit jelly,and ready to eat fruits),and cereals(snack bars,granola bars,bread,and cookies).The main prebiotic compounds used were inulin,fructooligosaccharides,oligofructose,polydextrose,and xylooligosaccharides.The effects of prebiotic on food products’,host health,and technological and sensory properties depended on the prebiotic type and dosage,food matrix,and subject type.The positive health effects were related to skin health,gut microbiota improvement,hypocholesterolemic impact,reduction of the risk of metabolic diseases,and improvements in postprandial blood glucose and insulin response.Prebiotics could improve vegan products’technological and sensory properties,such as fermentation time,texture and rheological properties,storage length,sensory acceptance,and probiotic survival.Furthermore,they could increase the concentration and/or bioaccessibility of phenolic compounds and vitamins.However,it is of paramount importance the determination of prebiotic stability.The trends direct to new prebiotic compounds based on microalgae,fruit and cereal by-products,and seeds.This review provides industries and researchers with important information about prebiotics and vegan products. 展开更多

Intestinal failure-associated liver disease(IFALD)is a common hepatobiliary complication resulting from long-term parenteral nutrition(PN)in patients with intestinal failure.The spectrum of IFALD ranges from cholestasis,steatosis,portal fibrosis,to cirrhosis.Development of IFALD is a multifactorial process,in which gut dysbiosis plays a critical role in its initiation and progression in conjunction with increased intestinal permeability,activation of hepatic immune responses,and administration of lipid emulsion.Gut microbiota manipulation including pre/probiotics,fecal microbiota transplantation,and antibiotics has been studied in IFALD with varying success.In this review,we summarize current knowledge on the taxonomic and functional changes of gut microbiota in preclinical and clinical studies of IFALD.We also review the function of microbial metabolites and associated signalings in the context of IFALD.By providing microbiota-targeted interventions aiming to optimize PN-induced liver injury,our review provides perspectives for future basic and translational investigations in the field. 展开更多

Synergistic effects of alginate oligosaccharide and cyanidin-3-O-glucoside on the amelioration of intestinal barrier function in mice

Emerging evidence shows that dietary oligosaccharides are important prebiotics that can improve intestinal flora,while dietary polyphenols can act directly on intestinal cells.However,information about their synergistic effects on gut health is still limited.In this study,alginate oligosaccharide(AOS)and cyanidin-3-O-glucoside(C3G)were selected as a common marine plant oligosaccharide and terrestrial plant polyphenol,respectively,to study their effects on intestinal health.The results show that,in comparison to their individual applications,the combination of AOS and C3G(mass ratio,3:1)displayed a stronger ability to up-regulate the expression of tight junction proteins,while enhanced intestinal epithelial barrier was also observed and levels of mucin-2 andβ-defensins were simultaneously increased in the intestinal mucus.Interestingly,the secretion of immunoglobulin A and immune-related cytokines were approximately doubled by the AOS+C3G mixture.In addition,the AOS+C3G mixture was found to be more conducive to the positive transformation of intestinal flora,which stimulated the growth of beneficial bacteria Akkermansia,Lachnospiraceae and Feacalibaculum while inhibiting the growth of harmful bacteria Helicobacter and Turicibacter.The data generated herein thus suggests that dietary oligosaccharides and dietary polyphenols may be more beneficial to intestinal health when applied in combination than their individual effects alone. 展开更多
