In this study,the structural characters,antioxidant activities and bile acid-binding ability of sea buckthorn polysaccharides(HRPs)obtained by the commonly used hot water(HRP-W),pressurized hot water(HRP-H),ultrasonic(HRP-U),acid(HRP-C)and alkali(HRP-A)assisted extraction methods were investigated.The results demonstrated that extraction methods had significant effects on extraction yield,monosaccharide composition,molecular weight,particle size,triple-helical structure,and surface morphology of HRPs except for the major linkage bands.Thermogravimetric analysis showed that HRP-U with filamentous reticular microstructure exhibited better thermal stability.The HRP-A with the lowest molecular weight and highest arabinose content possessed the best antioxidant activities.Moreover,the rheological analysis indicated that HRPs with higher galacturonic acid content and molecular weight showed higher viscosity and stronger crosslinking network(HRP-C,HRP-W and HRP-U),which exhibited stronger bile acid binding capacity.The present findings provide scientific evidence in the preparation technology of sea buckthorn polysaccharides with good antioxidant and bile acid binding capacity which are related to the structure affected by the extraction methods. 展开更多

The aquaculture industry has developed significantly over the past few decades and has had a substantial impact on the global food supply and marine fisheries resources.However,some problems arise behind the scenes due to excessive intensive farming,such as slow animal growth,frequent disease,and lipid metabolism disorders.These problems have limited the sustainable development of the aquaculture industry,and a continuable solution is required.The use of fungal polysaccharide appears to provide a solution to these problems.Therefore,different supplemented levels of Poria cocos polysaccharide(PCP)(0,0.4,0.8,1.2,1.6,and 2.0 g/kg,respectively)were fed to spotted sea bass(Lateolabrax maculatus)in similar size(30.28±0.18 g)in current study.The effects of PCP on growth,physiological parameters,and lipid metabolism of spotted sea bass were investigated after a 4-week rearing period.Results showed,fish with PCP intake presented a significantly higher weight gain,specific growth rate,and a significantly lower feed conversion ratio.Significantly higher trypsin activity in liver and intestine were observed in fish with PCP intake.The superoxide dismutase activity in serum and liver of fish with PCP intake were significantly improved,while significantly higher serum total antioxidant capacity and hepatic catalase activity were also observed.However,no significant differences in lysozyme and alkaline phosphatase activity were evident among groups.Fish with PCP intake showed a significantly lower total cholesterol,but no noteworthy change in triglyceride and lipid-metabolismrelated genes expression were observed among groups.Results indicated that intake of PCP has a positive effect on growth and antioxidant capacity of spotted sea bass,but seems to have a limited effect on the non-specific immunity and lipid metabolism of spotted sea bass.Based on the regression analysis results,1.4 g/kg of PCP is the optimal dose for spotted sea bass in size(30.28±0.18 g). 展开更多

The antineoplastic activity of polysaccharide was investigated in Stichopus chlorontus, Isostichopus badionotus, Stichopus horrens and Holothuria lessoni massin. Crude polysaccharide was prepared with enzyme hydrolyzation method and purified by anion exchange chromatography using DEAE-sepharose fast flow column. The effect of polysaccharide on cells apoptosis of SiHa and U87 was examined with cell counting kit-8 colorimetry method. Western blotting was used to analyze related proteins of cellular apoptosis including p53 and Bcl-2. Results showed that there were two main components in each sea cucumber polysaccharide, which could be eluted down by 1.0 mol/LNaCl solution. The four types of polysaccharide in the second component were named as SC-2, IB-2, HLM-2 and SH-2, respectively. They were used for comparing the antineoplastic activity. Results showed that SC-2, IB-2, HLM-2 and SH-2 could promote apoptosis of U87 and SiHa cells. SH-2 and HLM-2 were selected for the subsequent experiment to explore the additional effect of U87 and SiHa cells, The protein expressions of Bcl-2 and p53 decreased considerably with the increase of polysaccharide concentration in U87 cells. In SiHa cells, protein expressions of Bcl-2 and high dosage group of p53 decreased significantly, whereas no obvious decrease was observed in other groups. The polysaccharides are more effective in promoting apoptosis of U87 and SiHa cells from S. horrens and H. lessoni massin than from the rest species. 展开更多

Enzymatic hydrolysis has been widely used to produce bioactive hydrolysates from sea cucumber body wall.Here,inspired by the clarification of Apostichopus japonicus genome,we investigated the enzymatic hydrolysis of sea cucumber body wall by using the omics strategy.Shared proteins,including major yolk proteins,collagens,extracellular matrix glycoproteins and muscle proteins,were released from the body wall by different hydrolysis condition.A portfolio of 216 shared peptides were detected in the peptidome by papain with different hydrolysis time,while 32 shared peptides were detected in the peptidome by differing proteases.Unshared peptides and the relative abundance distribution profiles of shared peptides changed depending on hydrolysis approaches,indicating dynamic changes of peptidome during hydrolysis.Moreover,release of sulfated fucan and fucosylated chondroitin sulfate changed with the hydrolysis condition.The monitoring of dynamic enzymatic hydrolysis process at a molecular scale would contribute to production and quality control of sea cucumber hydrolysates. 展开更多

Sea cucumber is a traditional nutritional food and medicinal resource with many bioactive components in China.Holothuria fuscogliva is a big sea cucumber with a rich of bioactive polysaccharides.To investigate the bioactivities of the polysaccharides from sea cucumber H.fuscogliva,we prepared the sulfated polysaccharides(HfP) from sea cucumber H.fuscogliva using a protease hydrolysis method.Antioxidant activities of HfP were investigated,including hydroxyl radical scavenging activity and superoxide radical scavenging activity.And,the anticoagulant activities of HfP were studied,including the activated partial thromboplastin time(APTT),prothrombin time(PT) and thrombin time(TT).The average molecular weight was 1 867.1 Da,with a sulfate content of 20.7%.In addition,the molar ratio of monosaccharide composition of HfP was Man:Rha:Glc A:Glc:Gal:Xyl:Fuc=0.083 6:0.437:0.134:0:1.182:0.748:1.It had a strong antioxidant activity,the hydroxyl and superoxide radical scavenging activity EC_(50) of HfP was 3.74 and 0.037 mg/mL,respectively.It also showed a good anticoagulant activity in our study.The APTT of HfP was much higher than that of heparin sodium,and the PT and TT of HfP was close to that of heparin sodium at a low concentration.Therefore,HfP shows a good antioxidant and anticoagulant activity and it may become a potential candidate of the natural antioxidant and anticoagulant and will have a good application future in health product or medicine industry. 展开更多

Aim: To evaluate the clinical efficacy of the traditional Chinese medicine sea buckthorn (SBT) in the treatment of patients with nonalcoholic fatty liver disease (NAFLD). Method: 94 patients with NAFLD were randomly divided into two groups: 48 cases of patients received oral sea buckthorn 1.5 g (3 times a day) for three months as the treated group, and 46 cases received only the vehicle for three months as the control group. Serum lipids, transaminase and serum liver fibrosis indices were assessed at baseline and after SBT treatment. All patients underwent liver CT and Fibroscan examination at baseline and after treatment. Results: SBT treatment resulted in a significant decrease in the serum levels of alanine aminotransferase (ALT), LDL-C, hyaluronic acid and collagen type IV. The liver stiffness measurement (LSM) of the treated patients was significantly lower than that in the control or baseline. The CT liver/spleen ratio of the treated patients was also significantly increased. Conclusion: The results of our study demonstrated the beneficial effects of SBT on serum lipids, transaminase, and liver/spleen ratio and liver stiffness in patients with NAFLD, which may be further developed as a promising therapy for the treatment of NAFLD. 展开更多

The objectives of the study are: 1) to estimate the relationship between wild Sea buckthorn (SB) price and Supply, Demand, while some other factors of crude oil price and exchange rate by using simultaneous Supply-Demand and Price system equation and Vector Error Correction Method (VECM);2) to forecast the short-term and long-term SB price;3) to compare and evaluate the price forecasting models. Firstly, the data was analyzed by Ferris and Engle-Granger’s procedure;secondly, both price forecasting methodologies were tested by Pindyck-Rubinfeld and Makridakis’s procedure. The result shows that the VECM model is more efficient using yearly data;a short-term price forecast decreases, and a long-term price forecast is predicted to increase the Mongolian Sea buckthorn market. 展开更多

Chronic Toxicity Study in Rats Orally Exposed to Mulberry Sea-Buckthorn Beverage Concentrate

Objective: This study is designed to observe the chronic toxicity after the administration of mulberry sea-buckthorn beverage concentrate for 3 months on rats and to predict the possible adverse effect and the potential toxicity target organs. Method: The rats (SPF level) were randomly divided into high-dose (20 mL/kg BW), middle-dose (10 mL/kg BW), low-dose (5 mL/kg BW) groups and negative control group (20 mL/kg BW of purified water) with 30 rats in each group. Each group was orally given mulberry sea-buckthorn beverage concentrate for 3 months and recovered by stop feeding samples for 2 weeks for a recovery observation. The rats’ general condition, the organ coefficient, the indexes of hematology and blood biochemistry and the histological changes of the main organs were determined. Result: The appearance and behavior of activity in rats showed no anomalies in all these groups and all the rats put on weight during this period. Comparing to the negative control group, no obvious differences were observed in the weekly weight and organ coefficient of each dose group. After 3 months of administration, HGB in both mulberry sea-buckthorn beverage concentrate low-dose group and high-dose group were increased. No significant differences were observed in the indexes of hematology after 2 weeks of recovery. CREA in low-dose, middle-dose and high-dose groups were significantly increased after 3 months of administration and it remained in the high level in middle-dose and high-dose group even after 2 weeks of recovery. No drug-related lesions were observed in the histological changes of major organs. Conclusion: The results show that long term use of mulberry concentrated sea-buckthorn beverage can lead to increased CREA, which suggested kidney toxicity. Although no obvious pathological change was found in kidney, we should pay attention to chronic kidney damage in the further research. 展开更多

2,4,6-Tripyridine-s-triazine(TPTZ)spectrophotometric method was applied to determine the concentrations of dissolved monosaccharides(MCHO),polysaccharides(PCHO),and total carbohydrate(TCHO)in seawater samples collecte... 2,4,6-Tripyridine-s-triazine(TPTZ)spectrophotometric method was applied to determine the concentrations of dissolved monosaccharides(MCHO),polysaccharides(PCHO),and total carbohydrate(TCHO)in seawater samples collected from sea surface to hadal zone and sediment-seawater interface of the Southern Yap Trench in the Western Pacific Ocean.Results show that the concentrations of MCHO,PCHO,and TCHO ranged from 6.3 to 22.3μmol C/L,1.1 to 25.4μmol C/L,and 12.1 to 44.9μmol C/L,respectively,from the euphotic layer to the hadal zone of the trench.At different sampling stations,the concentrations of MCHO,PCHO,and TCHO in the seawater showed complex vertical variation characteristics,but the overall variation trends were decreasing with water depth.In the Southern Yap Trench,the maximum concentration of MCHO in the seawater appeared in the euphotic layer,and the minimum in the hadal zone.The maximum concentration of PCHO appeared in the euphotic layer,and the minimum in the bathypelagic layer.The water layer where the maxima and minima of the average concentration of TCHO appeared was consistent with that of PCHO.PCHO was the major component of TCHO in the seawater of the Southern Yap Trench.In the seawater from the sediment-seawater interface,the concentrations of MCHO,PCHO,and TCHO ranged from 8.4 to 10.6μmol C/L,3.8 to 5.8μmol C/L,and 12.2 to 15.2μmol C/L,respectively,and MCHO was the major component of TCHO.The key factors affecting the concentration and existing forms of dissolved sugars in the seawater of the Southern Yap Trench included photosynthesis,respiration,polysaccharide hydrolysis,adsorption and desorption of particulate matter,trench“funnel effect”,deep ocean currents,sediment resuspension,and etc.This study provided fundamental data about labile organic matter in abyss and hadal zone of marine environment,which is significant for further understanding of deep-sea organic carbon cycle. 展开更多

目的基于网络药理学—分子对接技术的方法初步探讨沙棘多酚治疗阿尔兹海默症(Alzheimer’s disease,AD)的作用机制。方法检索中国知网、万方数据库查找目前报道的沙棘多酚化学成分,获得沙棘多酚有效活性成分及预测靶基因。通过Cytoscape... 目的基于网络药理学—分子对接技术的方法初步探讨沙棘多酚治疗阿尔兹海默症(Alzheimer’s disease,AD)的作用机制。方法检索中国知网、万方数据库查找目前报道的沙棘多酚化学成分,获得沙棘多酚有效活性成分及预测靶基因。通过Cytoscape 3.8.2软件构建中药—化合物—靶点(基因)网络;采用Gencards、OMIM、DisGeNET等数据库获取AD主要作用靶点,并与化合物基因靶点作韦恩图、明确交集靶点,运用String平台构建蛋白相互作用(PPI)网络模型并挖掘网络中潜在的蛋白质功能模块;通过Metascape平台进行基因本体(GO)功能富集分析及基于京都基因与基因组百科全书(KEGG)通路富集分析,预测其生物过程及作用通路。应用Cytoscape3.8.2软件构建“药物成分-作用靶点-作用通路”网络,运用AutoDock vina软件对核心成分及关键靶点进行分子对接。结果共获得12个沙棘多酚候选化合物、142个成分靶点以及95个沙棘多酚与AD的共同靶点。网络分析结果显示,沙棘多酚调治AD的核心活性成分为阿魏酸、藜芦酸、咖啡酸等,核心靶点包括ALB、EGFR、PTGS2、STAT3、CTNNB1、MMP9、ESR1、TLR4、ICAM1和PPARA。GO和KEGG分析显示,沙棘多酚治疗AD的通路主要富集在化学致癌-受体活化、癌症的通路和糖尿病并发症中的AGE-RAGE信号通路等。其功能主要为氧化还原酶活性、丝氨酸水解酶活性和金属蛋白酶活性等。分子对接研究显示,沙棘多酚的5个关键治疗AD的化合物可与5个核心基因的对接口袋相匹配。结论沙棘多酚可通过多个化合物作用于多个分子靶点和通路发挥对AD的治疗作用。 展开更多

目的:考察主成分为沙棘和花生壳提取物的棘脑通的抗抑郁药效及作用机制。方法:(1)动物水平上,采用慢性不可预知温和应激(CUMS)抑郁模型小鼠,随机分为空白组、模型组、棘脑通给药组(棘脑通低浓度组、棘脑通高浓度组)和氟西汀组。通过体... 目的:考察主成分为沙棘和花生壳提取物的棘脑通的抗抑郁药效及作用机制。方法:(1)动物水平上,采用慢性不可预知温和应激(CUMS)抑郁模型小鼠,随机分为空白组、模型组、棘脑通给药组(棘脑通低浓度组、棘脑通高浓度组)和氟西汀组。通过体重测试、糖水偏嗜实验和悬尾不动时间测试等行为学研究,考察棘脑通的抗抑郁作用。(2)细胞水平上,将不同浓度的棘脑通应用于PC12细胞、SH-SY5Y细胞、BV2细胞和RAW 264.7细胞,研究棘脑通对神经营养因子功能的促进作用,保护神经细胞、诱导神经分化的作用,以及抗氧化、抗炎作用。结果:(1)与空白组相比,模型组小鼠糖水偏嗜率、体重显著降低,悬尾不动时间、强迫游泳不动时间显著延长(P<0.01);棘脑通给药组小鼠糖水偏嗜率较模型组显著升高(P<0.05),棘脑通给药组、氟西汀组小鼠体重较模型组显著升高(P<0.01),棘脑通低、高浓度组小鼠悬尾不动时间较模型组显著缩短(P<0.01、P<0.05),棘脑通给药组小鼠强迫游泳不动时间较模型组显著缩短(P<0.01),上述差异均有统计学意义。(2)棘脑通与神经生长因子(5、50 ng/mL)协同使用时,PC12细胞分化程度、pNF200-Luc表达荧光素酶活性显著提高。棘脑通与脑源性神经营养因子、神经营养因子-3和神经营养因子-4(5 ng/mL)协同使用时,荧光素酶的活性显著提高。100~1 000μg/mL的棘脑通可显著降低β-淀粉样蛋白的聚集。采用叔丁基过氧化氢诱导PC12细胞的氧化损伤后,应用棘脑通可提高PC12细胞活性,且浓度越高,效果越好。采用Erastin诱导SH-SY5Y细胞铁死亡后,应用棘脑通可提高SH-SY5Y细胞活性,且浓度越高,效果越好。将脂多糖用于BV2、RAW 264.7细胞后,应用棘脑通可降低白细胞介素1β的mRNA水平、核因子κB转录活性,且具有浓度依赖性。结论:棘脑通具有抗炎、抗氧化,保护神经细胞,诱导神经微丝表达,增强神经营养因子样的作用;该药通过以上作用发挥抗抑郁效果。 展开更多

运用网络药理学和分子对接的分析方法,预测沙棘治疗肝损伤的有效化合物、作用靶点、信号通路和免疫相关作用机制。先用TCMSP和GeneCards等数据库对沙棘的有效活性成分作用靶点以及肝损伤相关疾病靶点进行筛选,找到两者交集靶点,再建立药... 运用网络药理学和分子对接的分析方法,预测沙棘治疗肝损伤的有效化合物、作用靶点、信号通路和免疫相关作用机制。先用TCMSP和GeneCards等数据库对沙棘的有效活性成分作用靶点以及肝损伤相关疾病靶点进行筛选,找到两者交集靶点,再建立药材-化合物-靶点-疾病的网络图。然后利用R语言进行交集靶点的GO功能和KEGG信号通路富集分析,最后通过AutoDock Vina进行分子对接验证。结果表明:共筛选出沙棘22个活性成分,药物与疾病共同靶点有95个,沙棘治疗肝损伤的免疫相关作用机制可能是由槲皮素、山柰酚、异鼠李素等多个成分作用于IL-1β、PTGS2、和NFκBIA等关键靶点,对接结果显示核心成分与免疫相关靶点结合活性较强。沙棘可通过多成分、多靶点、多通路的共同作用,在免疫方面降低机体炎症水平和改善氧化应激状态从而缓解肝损伤。 展开更多

【目的】通过对新疆大果沙棘原浆、籽油及果油呈香物质进行分析,确定其气味特征及产品间相关性,为新疆大果沙棘制品的进一步开发和真伪鉴定提供数据支撑。【方法】采用固相微萃取结合气相色谱-质谱串联技术对新疆大果沙棘原浆、籽油和... 【目的】通过对新疆大果沙棘原浆、籽油及果油呈香物质进行分析,确定其气味特征及产品间相关性,为新疆大果沙棘制品的进一步开发和真伪鉴定提供数据支撑。【方法】采用固相微萃取结合气相色谱-质谱串联技术对新疆大果沙棘原浆、籽油和果油这3种沙棘主要制品的香气进行研究,表征每种沙棘制品的呈香组分及其香气活度,探究3种沙棘产品的关键香气组分,对其进行归类和分析,明确每种沙棘产品的香气特征,并通过聚类热图和相关性分析初步探究3种沙棘制品的香气组分之间的关联性。【结果】3种新疆大果沙棘制品共有64种呈香物质,沙棘原浆最多,其次是沙棘果油,沙棘籽油最少;沙棘原浆和果油关键呈香物质以酯类为主,籽油以醇类为主。沙棘原浆与果油的香气特征相似,均以果味和酒味为主,沙棘籽油以果味、花香和油脂味为主。新疆大果沙棘原浆、籽油和果油挥发性成分含量丰富,香气独特。沙棘原浆与果油相关性更高,沙棘籽油与另外两种制品相关性较低。【结论】研究结果可为后续沙棘产品深加工开发提供一定的理论基础。 展开更多

沙棘是一种药食资源,在民间常应用于治疗胃部疾病。已有研究表明沙棘果乙酸乙酯萃取物具有较强的抗幽门螺杆菌活性。为探究沙棘乙酸乙酯萃取物抑制幽门螺杆菌的化学成分及抑制机理,试验采用高效液相色谱法测定其化学成分,并测定生物膜... 沙棘是一种药食资源,在民间常应用于治疗胃部疾病。已有研究表明沙棘果乙酸乙酯萃取物具有较强的抗幽门螺杆菌活性。为探究沙棘乙酸乙酯萃取物抑制幽门螺杆菌的化学成分及抑制机理,试验采用高效液相色谱法测定其化学成分,并测定生物膜形成率、细胞膜通透性、膜蛋白荧光光谱、细胞膜完整性、脲酶活性和细胞微观结构等探究其抑菌机制。结果表明,沙棘乙酸乙酯萃取物中苹果酸含量最高,为872.97 mg/g,其次是芦丁、异鼠李素、槲皮素等黄酮类物质;沙棘乙酸乙酯萃取物可抑制生物膜形成,抑制率达到77.71%;细胞膜通透性增加2.45倍;膜蛋白结构发生变化,色氨酸和酪氨酸残基的亲水增强;脲酶活性下降87.89%;细胞内蛋白质和核酸的泄漏量达98.72%;扫描电镜结果表明萃取物会导致细胞结构变形,表面有褶皱。以上结果说明,沙棘乙酸乙酯萃取物中含有苹果酸和多种黄酮类物质,通过多种方式有效抑制幽门螺杆菌的生长繁殖,该结果为进一步分析沙棘中抑制幽门螺杆菌的活性物质奠定了基础。 展开更多

目的:优化沙棘中总黄酮提取工艺,明确其抗氧化性能。方法:以沙棘为研究对象,以沙棘黄酮得率为指标值,使用乙醇溶液作为提取剂,采用超声波辅助萃取法,对提取时间(A)、提取温度(B)、乙醇体积分数(V乙醇/V总)(C)、料液比(D)进行单因素试验... 目的:优化沙棘中总黄酮提取工艺,明确其抗氧化性能。方法:以沙棘为研究对象,以沙棘黄酮得率为指标值,使用乙醇溶液作为提取剂,采用超声波辅助萃取法,对提取时间(A)、提取温度(B)、乙醇体积分数(V乙醇/V总)(C)、料液比(D)进行单因素试验和响应面分析,考察4个因素对总黄酮得率的影响,并对沙棘黄酮的体外抗氧化活性进行检测。结果:在提取时间23 min、提取温度80℃、50%乙醇、料液比1∶20条件下沙棘黄酮得率最高,为2.316 mg/g。各因素对沙棘黄酮得率的影响大小依次为:料液比、乙醇体积分数、提取时间、提取温度。体外抗氧化结果表明,沙棘黄酮具有很好的DPPH自由基和羟自由基清除能力,当沙棘黄酮粗提取物质量浓度达到5 mg/mL时,沙棘黄酮对DPPH自由基的清除率达87.5%,对羟自由基的清除率达到了77.7%。结论:沙棘中含有丰富的黄酮类物质,且具有较好的抗氧化性能。 展开更多

为研制黑枸杞沙棘发酵乳,以生牛乳、黑枸杞和沙棘为主要原料,以感官评分和乳酸菌总数为评价指标,采用单因素、模糊数学评价和响应面法研究黑枸杞沙棘发酵乳制作工艺,并在最优工艺基础上添加不同比例鸡枞菌多糖,以提高发酵乳品质;测定添... 为研制黑枸杞沙棘发酵乳,以生牛乳、黑枸杞和沙棘为主要原料,以感官评分和乳酸菌总数为评价指标,采用单因素、模糊数学评价和响应面法研究黑枸杞沙棘发酵乳制作工艺,并在最优工艺基础上添加不同比例鸡枞菌多糖,以提高发酵乳品质;测定添加前后发酵乳DPPH·清除率等指标,研究鸡枞菌多糖对发酵乳品质的影响。结果表明,发酵乳最佳制备工艺为接种量5.33%,白糖添加量8.45%,CMC添加量0.56%,沙棘汁添加量3.00%,黑枸杞汁添加量4.61%。感官评分93.5分,乳酸菌总数20.92×10^(8)CFU/mL。此条件下的发酵乳组织细腻、无乳清析出、有光泽、具有发酵乳固有风味。DPPH·清除率随着多糖添加量增大而提高,发酵后乳饮料DPPH·清除率均高于发酵前。鸡枞菌多糖最适宜添加量为0.08%,此时发酵乳DPPH·清除率提升5.12%,蛋白质含量增高15.1%,酸度提升9.3%。研究为黑枸杞沙棘发酵乳的发展提供可靠数据参考。 展开更多
