Secondary Structure Changes and Thermal Stability of Plasma Membrane Proteins of Wheat Roots in Heat Stress

The wheat roots membrane separates the cell from the environment around it and encloses the cell contents. The pro-tein secondary structure and thermal stability of the plasma membrane of wheat root have been characterized in D2O buffer from 20°C to 90°C by Attenuated total reflection Fourier transform infrared spectroscopy (ATR-FTIR). Quantitative analysis of the amide I band (1700 - 1600 cm–1) showed that the plasma membrane proteins contains 41% α-helix, 16% β-sheet, 18% turn, and 25% disorder structures at 20°C. At elevated temperatures from 25°C up to 90°C, the α-helix and the β-sheet structure unfold into turns and the disorder structure, with a major conformational transition occurring at 50°C. There is a rapid decline in H+-ATPase activity of plasma membrane from 35°C to 55°C and it remain very low level H+-ATPase activity of PM from 55°C to 90°C. Therefore the protein conformational transition was one of reasons of loses H+-ATPase activity of plasma membrane. 展开更多

With the two-scale expansion technique proposed by Yoshizawa,the turbulent fluctuating field is expanded around the isotropic field.At a low-order two-scale expansion,applying the mode coupling approximation in the Yakhot-Orszag renormalization group method to analyze the fluctuating field,the Reynolds-average terms in the Reynolds stress transport equation,such as the convective term,the pressure-gradient-velocity correlation term and the dissipation term,are modeled.Two numerical examples:turbulent flow past a backward-facing step and the fully developed flow in a rotating channel,are presented for testing the efficiency of the proposed second-order model.For these two numerical examples,the proposed model performs as well as the Gibson-Launder (GL) model,giving better prediction than the standard k-ε model,especially in the abilities to calculate the secondary flow in the backward-facing step flow and to capture the asymmetric turbulent structure caused by frame rotation. 展开更多

为提高金属材性试验的测量精度和测试效率,在传统拉伸试验方法的基础上引入数字图像相关(Digital Image Correlation, DIC)技术,提出相应形变测量理论及方法。基于拉伸试件的颈缩变形特征,提出链式形变测量理论,该理论通过一种跟随表面... 为提高金属材性试验的测量精度和测试效率,在传统拉伸试验方法的基础上引入数字图像相关(Digital Image Correlation, DIC)技术,提出相应形变测量理论及方法。基于拉伸试件的颈缩变形特征,提出链式形变测量理论,该理论通过一种跟随表面形状变化而自由变形的柔性链条来表征各阶段的试件形变。进一步通过设置多个连续且等尺寸的DIC二阶链式子区,采用DIC亚像素算法得到任意变形时刻的链式子区角点坐标,并进一步得到沿试件长度方向的柔性离散链形变结果,实现对试件各处应变、颈缩极值点等的求解,从而建立了一套完整的金属拉伸试验DIC辅助测试方法。开展了多个标准金属试件的拉伸试验,通过与引伸仪、靶标图像测量等传统方法对比,验证了该文方法对颈缩效应、弹性模量、极限伸长率、真实应力-应变曲线等金属材性指标具备较高测量精度及有效性,同时该文方法能够实现高度自动化测量,可在金属拉伸试验领域进行工业化推广普及。 展开更多

[研究目的]川西坳陷孝泉—丰谷地区须二段砂岩气藏的勘探开发潜力巨大,但该地区埋藏较深且构造复杂、断缝系统多期叠加,使得地应力频繁变化,制约了该区井位轨迹设计与压裂改造的有效实施,故需对该区地应力大小进行精细评价,为工程开发提供建议从而提高产能。[研究方法]基于岩石力学、声发射实验及差应变分析等实验测试方法,并结合常规测井、特殊测井及水力压裂等资料分析,优选了适应于深层块状均质致密砂岩储层的地应力大小实验测试方法,并在单点地应力大小准确评价的基础之上,构建了研究区分构造变形单元分层的单井地应力大小连续测井解释模型,查明了纵向上地应力大小变化结构类型及分布规律。[研究结果]研究表明差应变分析法计算的地应力大小精确度最高,为更能够准确表征深层均质块状致密砂岩地应力大小的实验方法。测试结果显示须二段属于Ⅲ类地应力类型,处于走滑应力状态,存在部分逆冲挤压应力状态;在井点测试的基础上,形成了基于井壁影像反演的地应力大小评价技术;地应力大小结构变化在纵向上分为5种类型,其中南北向(SN)三级以上断层和南北向(SN)褶皱或北东东向(NEE)褶皱变形的高部位以低低高(LLH)型和低应力(LC)型为主,在小规模断层或平缓构造区以高低高(HLH)型或高低低(HLL)型为主。[结论]建议选择低低高(LLH)型地应力剖面进行工程开发,其纵向上可穿透更多含气层,同时避开底层底水,预防生产早期快速见水,故应选择二—三级南北向断层和南北或北东向纵弯褶皱区须二2中上段进行水力压裂改造。 展开更多

压合衬套二次冷挤压强化数值仿真与实验研究

采用压合衬套二次冷挤压工艺对7050铝合金耳片孔进行挤压强化;通过X射线衍射仪、三坐标测量机和粗糙度仪,研究二次冷挤压对耳片孔内表面的残余应力、塑性变形以及粗糙度的影响,并通过有限元仿真进行验证。实验结果表明:相较于一次冷挤压... 采用压合衬套二次冷挤压工艺对7050铝合金耳片孔进行挤压强化;通过X射线衍射仪、三坐标测量机和粗糙度仪,研究二次冷挤压对耳片孔内表面的残余应力、塑性变形以及粗糙度的影响,并通过有限元仿真进行验证。实验结果表明:相较于一次冷挤压,压合衬套二次冷挤压可有效改善试样两端残余应力的不均匀性。随着二次冷挤压相对挤压量增大,残余压应力峰值和应力层深度都进一步增大,塑性变形程度也更大,同时降低了孔壁表面粗糙度。在相对挤压量为2.5%时,最大残余压应力可达-400.3 MPa,并形成了约14 mm的残余压应力层;试样端面A与端面B内径分别增大了0.56 mm和0.54 mm;孔壁表面粗糙度降低幅度达到71.0%。相较于一次冷挤压,二次冷挤压强化后孔壁沿径向形成了更深的残余压应力层和塑性变形层,显著提高了耳片孔的整体服役性能。 展开更多

压力知觉在护士第二受害者人格特质与负性体验间的中介效应

目的探讨护士第二受害者的人格特质对其在患者安全事件中负性体验的影响,以及压力知觉在负性体验影响机制中的中介作用。方法采用便利抽样法,于2023年3月至4月选择太原市3所三级甲等综合医院近3个月内经历过患者安全事件的560名护士作... 目的探讨护士第二受害者的人格特质对其在患者安全事件中负性体验的影响,以及压力知觉在负性体验影响机制中的中介作用。方法采用便利抽样法,于2023年3月至4月选择太原市3所三级甲等综合医院近3个月内经历过患者安全事件的560名护士作为研究对象,采用一般资料问卷、中国大五人格问卷、第二受害者经历与支持量表、中文版压力知觉量表进行调查,采用Pearson相关性分析护士第二受害者人格特质的神经质维度(以下简称神经质人格)、负性体验与压力知觉间的相关性,依照Bootstrap方法,使用PROCESS插件分析压力知觉在护士第二受害者神经质人格与负性体验间的中介作用。结果回收有效问卷560份,有效回收率100.00%。护士第二受害者人格特质得分从高到低依次为严谨性、宜人性、开放性、神经质、外向性,负性体验得分(40.25±10.64),处于较高水平,压力知觉得分为(30.25±5.98)分,处于较高水平。护士第二受害者神经质人格与负性体验呈正相关(r=0.528,P<0.01),并与压力知觉呈正相关(r=0.594,P<0.01);负性体验与压力知觉呈正相关(r=0.339,P<0.01);护士第二受害者神经质人格对负性体验存在直接效应(β=0.519,95%CI:0.318~0.720),紧张感(压力知觉维度之一)在护士第二受害者神经质人格和负性体验间起部分中介作用,中介效应(β=0.148,95%CI:0.006~0.300),中介作用占比为22.70%。结论护士第二受害者高神经质人格不仅可以直接影响其负性体验,还可以通过紧张感(压力知觉的维度之一)影响其负性体验。护理管理者及护士第二受害者可通过改变高神经质人格及缓解紧张感的角度,减轻其在患者安全事件中的负性体验。 展开更多

为了探究不同围压下花岗岩风化土力学特性性能,选取广西玉林市容县花岗岩风化土作为研究对象,通过对饱和重塑花岗岩风化土进行常偏应力排水剪切(CSD)试验,研究了不同围压下风化土的力学行为过程和破坏机理,并对CSD试验后的试样进行了扫... 为了探究不同围压下花岗岩风化土力学特性性能,选取广西玉林市容县花岗岩风化土作为研究对象,通过对饱和重塑花岗岩风化土进行常偏应力排水剪切(CSD)试验,研究了不同围压下风化土的力学行为过程和破坏机理,并对CSD试验后的试样进行了扫描电镜(SEM)试验,揭示了CSD试验下风化土失效的微观机理。结果表明:在孔压上升初期,试样的轴向应变增长缓慢,偏应力几乎不变;随着孔压的不断升高,平均有效应力下降至失稳点,轴向应变突然增加,偏应力迅速衰减,试样呈现出瞬变破坏的形式;在降雨入渗时,围压程度的变化对土体行为有显著影响,低围压下,试样局部水压力增大,导致鼓胀破坏。而高围压下,试样呈现伴随有软弱面的剪切破坏。 展开更多

目的:探究浓缩生长因子(concentrated growth factor,CGF)在促进下颌阻生第三磨牙拔牙创面愈合中的疗效,及对第二磨牙远中骨缺损深度(osseous defect depth,ODD)、临床附着水平(clinical attachment level,CAL)及疼痛应激指标的影响。方... 目的:探究浓缩生长因子(concentrated growth factor,CGF)在促进下颌阻生第三磨牙拔牙创面愈合中的疗效,及对第二磨牙远中骨缺损深度(osseous defect depth,ODD)、临床附着水平(clinical attachment level,CAL)及疼痛应激指标的影响。方法:选择2022年1月至2022年12月于我院拔除下颌阻生第三磨牙的患者共96例,按照随机数表法将患者随机分为研究组(n=48)及对照组(n=48)。对照组在拔牙后不填入任何植入物,研究组填入CGF凝冻物。比较2组患者的术后视觉模拟评分法(visual analogue scale,VAS)评分、面部肿胀情况、疼痛应激指标[5-羟色胺(5-hydroxytryptamine,5-HT)、半乳糖胺(galactosamine,Gal)、降钙素基因相关肽(calcitonin gene related peptide,CGRP)]含量、ODD、CAL及术后生活质量[术后症状严重程度(postoperative symptom severity,PoSSe)量表]。结果:术后1、3 d,研究组的VAS评分分别为(3.72±0.56)、(2.94±0.43)分,均显著低于对照组的(4.81±0.61)、(3.86±0.57)分(P<0.001);术后研究组的面部肿胀度为(2.79±0.64)mm,低于对照组的(5.46±1.22)mm(P<0.001);2组术后3 d的5-HT、Gal、CGRP水平均高于术前,研究组分别为(2.46±0.68)ng/mL、(9.56±1.42)nmol/mL、(1.76±0.52)ng/mL,均低于对照组的(3.15±0.46)ng/mL、(13.19±1.08)nmol/mL、(2.89±0.44)ng/mL(P<0.001);术后6个月,2组的ODD、CAL水平均低于术后1周,研究组分别为(4.21±0.87)、(0.48±0.12)mm,均低于对照组[(5.75±0.61)、(0.73±0.19)mm,P<0.001];研究组的各项PoSSe评分均低于对照组(P<0.01)。结论:CGF可降低下颌阻生第三磨牙拔牙后患者术后疼痛应激指标水平,改善其拔牙后的疼痛及面部肿胀情况,提高患者术后生活质量。 展开更多

目的研究不同剖宫产手术时机对瘢痕子宫再次妊娠产妇应激反应及分娩风险的影响。方法选取100例瘢痕子宫再次妊娠产妇作为研究对象,根据剖宫产时机不同分为对照组(37周≤孕周<39周)与观察组(孕周≥39周),各50例。所有产妇均进行剖宫... 目的研究不同剖宫产手术时机对瘢痕子宫再次妊娠产妇应激反应及分娩风险的影响。方法选取100例瘢痕子宫再次妊娠产妇作为研究对象,根据剖宫产时机不同分为对照组(37周≤孕周<39周)与观察组(孕周≥39周),各50例。所有产妇均进行剖宫产手术,比较两组分娩前后应激指标、妊娠结局、并发症发生情况。结果产后两组去甲肾上腺素(NA)、皮质醇(Cor)以及醛固酮(ALD)指标均高于产前,且观察组低于对照组(P<0.05)。观察组产后出血量少于对照组,产后住院时间短于对照组,新生儿Apgar评分高于对照组,新生儿呼吸系统疾病发生率低于对照组(P<0.05)。观察组并发症发生率4.00%低于对照组的18.00%(P<0.05)。结论孕周≥39周为瘢痕子宫再次妊娠产妇的良好剖宫产手术时机,其可有效减少手术应激反应,改善妊娠结局的同时,降低相关并发症发生风险,值得临床推广。 展开更多

However, it is difficult to directly observe a rapidly growing de novo aneurysm in a patient. To investigate possible associations between hemodynamic parameters and the formation and growth of intracranial aneurysms, the present study constructed a computational model of a case with an internal carotid artery aneurysm and an anterior communicating artery aneurysm, based on the CT angiography findings of a patient. To simulate the formation of the anterior communicating artery aneurysm and the growth of the internal carotid artery aneurysm, we then constructed a model that virtually removed the anterior communicating artery aneurysm, and a further two models that also progressively decreased the size of the internal carotid artery aneurysm. Computational simulations of the fluid dynamics of the four models were performed under pulsatile flow conditions, and wall shear stress was compared among the different models. In the three aneurysm growth models, increasing size of the aneurysm was associated with an increased area of low wall shear stress, a significant decrease in wall shear stress at the dome of the aneurysm, and a significant change in the wall shear stress of the parent artery. The wall shear stress of the anterior communicating artery remained low, and was significantly lower than the wall shear stress at the bifurcation of the internal carotid artery or the bifurcation of the middle cerebral artery. After formation of the anterior communicating artery aneurysm, the wall shear stress at the dome of the internal carotid artery aneurysm increased significantly, and the wall shear stress in the upstream arteries also changed significantly. These findings indicate that low wall shear stress may be associated with the initiation and growth of aneurysms, and that aneurysm formation and growth may influence hemodynamic parameters in the local and adjacent arteries. 展开更多

The velocity field and the associated shear stress corresponding to the longitudinal oscillatory flow of a generalized second grade fluid, between two infinite coaxial circular cylinders, are determined by means of the Laplace and Hankel transforms. Initially, the fluid and cylinders are at rest and at t = 0+ both cylinders suddenly begin to oscillate along their common axis with simple harmonic motions having angular frequencies Ω1 and Ω2. The solutions that have been obtained are presented under integral and series forms in terms of the generalized G and R functions and satisfy the governing differential equation and all imposed initial and boundary conditions. The respective solutions for the motion between the cylinders, when one of them is at rest, can be obtained from our general solutions. Furthermore, the corresponding solutions for the similar flow of ordinary second grade fluid and Newtonian fluid are also obtained as limiting cases of our general solutions. At the end, the effect of different parameters on the flow of ordinary second grade and generalized second grade fluid are investigated graphically by plotting velocity profiles. 展开更多

GH4169 superalloy stress relaxation test was investigated to study its characteristics of stress relaxation curves at various temperatures( 550,650,and 750 ℃). These curves presented jointly two distinct stages,the stage of inner stress relaxing quickly,and the stage of inner stress relaxing slowly and closing to the stress relaxation limit. And these curves obtained could be fitted by second order exponential decay function well. Based on the experimental stress relaxation curves,the relationship between stress relaxation rate and time were derived, which showed that the higher relaxation temperature and the greater initial rate of stress relaxation. The whole process presented two different stages,the stage of stress relaxation rate falling rapidly and the stage of stress relaxation rate slowing down and tending to be constant. Mechanical models of residually stressed fibre-reinforced solids,which do not resist bending,have been developed in the literature.However,in some residually stressed fibre-reinforced elastic solids,resistance to fibre bending is significant,and the mechanical behavior of such solids should be investigated.Hence,in this paper,we model the mechanical aspect of residually stressed elastic solids with bending stiffness due to fibre curvature,which up to the authors’knowledge has not been mechanically modeled in the past.The proposed constitutive equation involves a nonsymmetric stress and a couple-stress tensor.Spectral invariants are used in the constitutive equation,where each spectral invariant has an intelligible physical meaning,and hence they are useful in experiment and analysis.A prototype strain energy function is proposed.Moreover,we use this prototype to give results for some cylindrical boundary value problems. 展开更多

This paper deals with the rotational flow of a generalized second grade fluid, within a circular cylinder, due to a torsional shear stress. The fractional calculus approach in the constitutive relationship model of a second grade fluid is introduced. The velocity field and the resulting shear stress are determined by means of the Laplace and finite Hankel transforms to satisfy all imposed initial and boundary conditions. The solutions corresponding to second grade fluids as well as those for Newtonian fluids are obtained as limiting cases of our general solutions. The influence of the fractional coefficient on the velocity of the fluid is also analyzed by graphical illustrations. 展开更多

The velocity field and the associated shear stress corresponding to the torsional oscillatory flow of a second grade fluid, between two infinite coaxial circular cylinders, are determined by means of the Laplace and Hankel transforms. At time t = 0, the fluid and both the cylinders are at rest and at t = 0 + , cylinders suddenly begin to oscillate around their common axis in a simple harmonic way having angular frequencies ω 1 and ω 2 . The obtained solutions satisfy the governing differential equation and all imposed initial and boundary conditions. The solutions for the motion between the cylinders, when one of them is at rest, can be obtained from our general solutions. Furthermore, the corresponding solutions for Newtonian fluid are also obtained as limiting cases of our general solutions. 展开更多

The areas affected by the L1 transfer to L2,such as phonotactics and stress,appear to have different degrees of erasability through training and practice.This paper attempts a theoretical analysis to account for empirical observations of L1 transfer in diphthong-coda coalescence and in sentence stress for Chinese learners of English in an American accent training course.Using the framework and tableau method of Optimality Theory(Prince and Smolensky 1993),the phonological paradigm that views languages’differences as symptomatic of differences in rankings of a set of universal violable constraints,this paper posits the underlying ranking at work in(a)the segmental case where[aŋ]is the observed output for/aʊn/;(b)the suprasegmental case where no stress contrast is observed for a pair of phonologically similar sentences with contrastive stress.It is argued that,in the segmental case,the coalescence of the second vowel of a diphthong with an alveolar nasal coda results from the high ranking of markedness constraints,including Complex Syllable;and in the suprasegmental case,sentence stress is greatly affected by changes in the ranking of the constraint culminativity.Aside from shedding light on the understudied issue of diphthong-coda coalescence,this research also contributes to the discussion of the comparative erasability of L1 transfer for the two areas and demonstrates how a deduction-based theoretical method may actually have practical implications for guiding the design of L2 teaching methodology of English pronunciation as well as for promoting native English speakers’clearer understanding of Chinese learners of English on the global stage. 展开更多

The USM-θ model of Bingham fluid for dense two-phase turbulent flow was developed, which combines the second-order moment model for two-phase turbulence with the particle kinetic theory for the inter-particle collision. In this model, phases interaction and the extra term of Bingham fluid yield stress are taken into account. An algorithm for USM-θ model in dense two-phase flow was proposed, in which the influence of particle volume fraction is accounted for. This model was used to simulate turbulent flow of Bingham fluid single-phase and dense liquid-particle two-phase in pipe. It is shown USM-θ model has better prediction result than the five-equation model, in which the particle-particle collision is modeled by the particle kinetic theory, while the turbulence of both phase is simulated by the two-equation turbulence model. The USM-θ model was then used to simulate the dense two-phase turbulent up flow of Bingham fluid with particles. With the increasing of the yield stress, the velocities of Bingham and particle decrease near the pipe centre. Comparing the two-phase flow of Bingham-particle with that of liquid-particle, it is found the source term of yield stress has significant effect on flow. 展开更多
