Soybean isoflavones are essential secondary metabolites synthesized in the phenylpropanoid pathway and benefit human health. In the present study, highresolution QTL mapping for isoflavone components was performed using specific-locus amplified fragment sequencing(SLAF-seq) with a recombinant inbred line(RIL) population(F5:7) derived from a cross between two cultivated soybean varieties, Luheidou 2(LHD2) and Nanhuizao(NHZ). Using a high-density genetic map comprising 3541 SLAF markers and the isoflavone contents of soybean seeds in the 200 lines in four environments, 24 stable QTL were identified for isoflavone components, explaining 4.2%–21.2% of phenotypic variation.Of these QTL, four novel stable QTL(qG8, qMD19, qMG18, and qTIF19) were identified for genistin, malonyldaidzin, malonylgenistin, and total isoflavones, respectively. Gene annotation revealed three genes involved in isoflavone biosynthesis(Gm4CL, GmIFR, and GmCHR) and 13 MYB-like genes within genomic regions corresponding to stable QTL intervals, suggesting candidate genes underlying these loci. Nine epistatic QTL were identified for isoflavone components, explaining 4.7%–15.6% of phenotypic variation. These results will facilitate understanding the genetic basis of isoflavone accumulation in soybean seeds. The stable QTL and tightly linked SLAF markers may be used for markerassisted selection in soybean breeding programs. 展开更多

Objective To investigate the dose-dependent toxic effect of soybean isoflavone extracts (SIE) on reproductive development in immature rats. Methods Growing male and female rats (n=50 each, 4 weeks) were divided into five groups fed with a standard cereal-based diet and gastrogavaged daily with 0, 30, 150, 300, and 600 mg SIE / kg body weight, respectively, for 12 weeks. Body weight, organ weights, and serum level of estrogen and testosterone were measured. Results Oral administration of SIE had no effect on food intake but decreased food efficiency ratio (P<0.01). Suppression on body weight gain by SIE was dose-dependent and the effect was greater on male than on female rats (P<0.01). SIE at high doses exhibited hepatotoxicity by increasing a relative liver weight, and also caused a smaller uterus but a greater relative ovary in female rats, while leading to larger relative testis and epididymis in male rats. SIE could decrease progesterone concentrations in female rats, whereas in male rats it reduced not only total testosterone level but also sperm count compared with the control group (P<0.05). Conclusion SIE at a range of 50-1000 times of human intake level affects not only growth but also development of reproductive system in growing rats. 展开更多

A synthetic isoflavone (ISO-S) or genistein was added in culture medium at different concentrations (0, 10, 20, 30, 40, and 80 μmol L-1) to investigate the effects of soybean isoflavones on antioxidative capacity of porcine skeletal muscle satellite cells. After 48 h incubation, the suspension was cryopreserved for the determination of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) activities, and malondialdehyde (MDA) content. The mRNA levels of SOD, CAT, and GSH-Px gene in cells were detected with Taqman fluorescent probe method. The results showed that the content of MDA and the activities and the mRNA levels of SOD of porcine skeletal muscle satellite cells were influenced by supplemented soybean isoflavone (P < 0.05) when adding 10-80 μmol L-1 ISO-S or genistein in the medium. The MDA contents, SOD and CAT activities and their mRNA expression levels of porcine skeletal muscle cells responded quadratically (P < 0.05) as the level of ISO-S or genistein increased. Pre-incubation of porcine skeletal muscle satellite cells with ISO-S or genistein at 10-40 μmol L-1 elevated the activities and the mRNA expression levels of SOD and CAT in cells concurrently and decreased the cellular content of MDA (P < 0.05). The results indicated that pre-incubation of ISO-S or genistein at 10-40 μmol L-1 could improve the antioxidative capacity of porcine skeletal muscle satellite cells. 展开更多

The results showed that the isoflavone contents and its components of soybean seedare quite differences among the tested materials, the contents of isoflavone and daidzein are controlled not only byadditive effects and but also by non-additive effects, while the content of genistin is dominated by non-additive effects,and genistein, glycitin and daidzin are mainly controlled by additive effects. There are significant differences in thecontents of isoflavone and its components among the combinations derived from different parents. Results also indicatedthat the tested traits are negatively heterosis except for the contens of daidzein and daidzin are positively heterosis basedon the data of the GCA and SCA in average heterosis values. In this research we have a suggestion that soybean varietywith high isoflavone should be used as one of the parents in the breeding program, and it is the best choice that thecombinations crossed between two high isoflavone varieties or a high variety and a low one. 展开更多

Background: Soybean isoflavones are structurally similar to mammalian estrogens and therefore may act as estrogen agonists or antagonists. However, it has not been determined if they have any negative effects on reproductive parameters in male livestock. Therefore, the objective of this study was to evaluate the effects of soybean isoflavones on male reproduction using Chinese mini-pig boars as a model. Fifty Xiang boars were randomly divided into five groups and fed diets containing 0, 125, 250, or 500 ppm soybean isoflavones or 0.5 ppm diethylstilbestrol for 60 days. Results: Dietary supplementation with 250 ppm of soy isoflavones markedly increased the testis index (P<0.05), fructose content (P <0.05), and α-glycosidase content in testicular tissue (P<0.01), as well as increased the number of viable germ cells (P<0.01) and the level of Bcl-2 protein (P <0.01). However, 500 ppm of soybean isoflavones significantly reduced both testis and epididymis indexes (P<0.05) and lactate dehydrogenase levels (P<0.01), as well as reduced serum LH and testosterone levels (P<0.05). High levels of soybean isoflavones also increased malondialdehyde levels (P<0.05), as well as increased the numbers of early and late apoptotic germ cells (P<0.01) and the level of Bax proteins (P<0.05) in the testis. Conclusions: The results of this study indicate that consumption of soy isoflavones at dietary levels up to 250 ppm did not adversely affect reproductive parameters in Chinese mini-pig boars whereas higher levels of soy isoflavones may adversely affect male reproduction. 展开更多

Soybean isoflavone, a type of non-steroid estrogen withsimilar molecular structure and function to estradiol, exhibits estrogen-like characteristics.OBJECTIVE: To investigate the effects of various doses of soybean isoflavone on expression ofERα, NGF, IL-2, and AR in the cerebellum of ovariectomized rat, and to determine whether there is adose-dependent effect.DESIGN, TIME AND SETTING: Controlled trial at the cellular and molecular level. The study wasperformed at the Experimental Animal Engineering Center, College of Veterinary Medicine, SichuanAgricultural University from July 2006 to May 2008.MATERIALS: Soybean isoflavone, comprised of daidzin, genistein and isoflavone, was provided byTaiyuan Yuantai Biochemical Industry, China. The ERα, NGF, IL-2, and AR in situ hybridization kit,rabbit anti-rat ERα, NGF, IL-2, and AR monoclonal antibodies, and SABC kit were purchased fromWuhan Boster Biological Technology, China.METHODS: A total of 50 female, Sprague Dawley rats, aged 3 months, were randomly assigned to5 groups, with 10 animals in each group. With the exception of the sham-operation group (abdominalcavity opening alone), all rats underwent bilateral ovariectomy. At 14 days after surgery, rats in thehigh-, middle-, and low-dose soybean isoflavone groups were subcutaneously injected with 1.5, 1.0,and 0.5 mg/kg soybean isoflavone, respectively, every 2 days for 6 consecutive weeks. Rats in thesham-operation and ovariectomized groups were subcutaneously injected with absolute alcohol(0.5 mL/kg).MAIN OUTCOME MEASURES: Expression levels and distribution of ERα, NGF, IL-2, and AR in thecerebellum were detected by immunohistochemistry and in situ hybridization.RESULTS: Compared with the sham-operation group, immunoreactive products and hybridizationsignals of ERα, NGF, IL-2, and AR were significantly decreased in the cerebellar cortex and nuclei ofovariectomized rats (P< 0.05 or P< 0.01), but increased following soybean isoflavone treatment. Inparticular, levels of the high-dose soybean isoflavone group were almost restored to levels of thesham-operation group (P > 0.05). The immunoreactive products were primarily located in thecytoplasm and neurites, and rarely in the cell membrane and nuclei. However, the hybridizationsignals were predominantly located in the nuclei, but rarely in the cytoplasm, cell membrane, orneurites.CONCLUSION: Soybean isofiavone upregulated ERα, NGF, IL-2, and AR protein and geneexpression in a dose-dependent manner, and played an important role in sustaining and protectingstructure and function of cerebellar neurons. Moreover, the similarity of expression patterns of thesemolecules indicated that they were mutually interactive during the regulation of soybean isoflavoneto the cerebellum. 展开更多

Background:Soybean has long been utilized in the realm of traditional Chinese medicine.One of its extracts,soybean glycolipids,serves as a vital by-product of soybean oil refining,but its chemical composition and pharmacological potential have yet to be fully elucidated.Methods:In this study,the chemical components were isolated,and the inhibitory effects of these isolates were explored in different zebrafish inflammatory models by survival rate,Histological examination assay and quantitative Real-time PCR assay.The cytotoxicity of daidzin in RAW264.7 cells was evaluated by cell viability assay,and the effect of daidzin on the release of inflammatory cytokines in RAW264.7 cells was detected by enzyme linked immunosorbent assay(ELISA).Western blotting,immunofluorescence assay and alpha7 nicotinic acetylcholine receptors siRNA transfection assay were used to further explore the anti-inflammatory mechanism of daidzin.Results:Four compounds(verticilloside,soya-cerebroside I,soya-cerebroside II and daidzin)were firstly isolated from the soybean glycolipids,among which verticilloside and daidzin inhibited the lipopolysaccharide,CuSO4-and tail cut-stimulated zebrafish inflammation.Noticeably,daidzin exhibited anti-inflammatory activities by increasing the survival rate,alleviating the inflammatory cells infiltration,and down-regulating the expression of pro-inflammatory cytokines and nuclear factor kappa-B,NF-kappa-B inhibitor alpha,and signal transducer and activator of transcription3 in zebrafish.Moreover,daidzin decreased the secretion of IL-6 and TNF-α,inhibited the nuclear translocations of nuclear factor kappa-B p65 and p-signal transducer and activator of transcription3 as well as the NF-kappa-B inhibitor alpha phosphorylation at Ser32 in RAW 264.7 cells.More importantly,it elevated the expression level of alpha7 nicotinic acetylcholine receptors in both zebrafish and RAW 264.7 cells,and the inhibitory effect of daidzin was attenuated after the addition of alpha7 nicotinic acetylcholine receptors siRNA.Conclusion:Our study revealed that daidzin inhibited inflammation by activating the cholinergic anti-inflammatory pathway and further inhibiting the nuclear factor kappa-B and signal transducer and activator of transcription3 signaling.At the same time,it also promotes the recycling of crude soybean glycolipids and supports the potential use of daidzin as a functional food or natural dietary anti-inflammatory agent. 展开更多

Rhizobia, crucial for nitrogen fixation in leguminous plants, play a vital role in soybean cultivation. This study, conducted in Mexico, a major soybean importer, aimed to identify bacteria from nodules of five soybean varieties in high-production regions. Multilocus sequence analysis (MLSA) was employed for enhanced species resolution. The study identified six Bradyrhizobium species: Bradyrhizobium japonicum USDA 110, Bradyrhizobium japonicum USDA 6, Bradyrhizobium elkanii USDA 76, Bradyrhizobium neotropicale, Bradyrhizobium lablabi, and Bradyrhizobium icense. Bradyrhizobium japonicum USDA 110 predominated in the soils, displaying symbiotic preference for the Huasteca 400 variety. However, phylogenetic analysis didn't reveal a clear association between strains, soil, and soybean variety. This research sheds light on the diversity of rhizobia in Mexican soybean cultivation, contributing to the understanding of symbiotic relationships in soybean production systems. 展开更多

Drought stress limits agricultural productivity worldwide.Identifying and characterizing genetic components of drought stress-tolerance networks may improve crop resistance to drought stress.We show that the regulatory module formed by miR166 and its target gene,ATHB14-LIKE,functions in the regulation of drought tolerance in soybean(Glycine max).Drought stress represses the accumulation of miR166,leading to upregulation of its target genes.Optimal knockdown of miR166 in the stable transgenic line GmSTTM166 conferred drought tolerance without affecting yield.Expression of ABA signaling pathway genes was regulated by the miR166-mediated regulatory pathway,and ATHB14-LIKE directly activates some of these genes.There is a feedback regulation between ATHB14-LIKE and MIR166 genes,and ATHB14-LIKE inhibits MIR166 expression.These findings reveal that drought-triggered regulation of the miR166-mediated regulatory pathway increases plants drought resistance,providing new insights into drought stress regulatory network in soybean. 展开更多

Soybean(Glycine max)stands as a globally significant agricultural crop,and the comprehensive assembly of its genome is of paramount importance for unraveling its biological characteristics and evolutionary history.Nevertheless,previous soybean genome assemblies have harbored gaps and incompleteness,which have constrained in-depth investigations into soybean.Here,we present Telomere-to-Telomere(T2T)assembly of the Chinese soybean cultivar Zhonghuang 13(ZH13)genome,termed ZH13-T2T,utilizing PacBio Hifi and ONT ultralong reads.We employed a multi-assembler approach,integrating Hifiasm,NextDenovo,and Canu,to minimize biases and enhance assembly accuracy.The assembly spans 1,015,024,879 bp,effectively resolving all 393 gaps that previously plagued the reference genome.Our annotation efforts identified 50,564 high-confidence protein-coding genes,707 of which are novel.ZH13-T2T revealed longer chromosomes,421 not-aligned regions(NARs),112 structure variations(SVs),and a substantial expansion of repetitive element compared to earlier assemblies.Specifically,we identified 25.67 Mb of tandem repeats,an enrichment of 5S and 48S rDNAs,and characterized their genotypic diversity.In summary,we deliver the first complete Chinese soybean cultivar T2T genome.The comprehensive annotation,along with precise centromere and telomere characterization,as well as insights into structural variations,further enhance our understanding of soybean genetics and evolution. 展开更多

Background Two studies were designed to determine standard ileal crude protein(CP)and amino acid(AA)digestibility of soybean meal(SBM)from different origins fed to non-pregnant and pregnant sows.Seven solvent-extracted SBMs from soybeans produced in the USA,Brazil,and China were selected.In Exp.1,eight different diets were created:a nitrogen(N)-free diet and 7 experimental diets containing SBM from different origins as the only N source.Eight non-pregnant,multiparous sows were arranged in an 8×8 Latin square design(8 periods and 8 diets).In Exp.2,the diet formula was the same as in Exp.1.Eight gestating sows(parity 3)were assigned to 4 different diets in a replicated 4×3 Youden square design(three periods and four diets)in mid-gestation and again in late-gestation stages.Results When fed to non-pregnant and late-gestating sows,the standardized ileal digestibility(SID)of CP and most AAs from different SBM were not significantly different(P>0.05).When fed to mid-gestating sows,the SID values for Arg,His,Lys,Phe,Cys,Gly,Ser,and Tyr in SBM 1 were lower than in SBM 4 and 5(P<0.05),whereas SID for Leu from SBM 5 was higher than in SBM 1 and 4(P<0.05).SID values for Ile,Ala,and Asp from SBM 4 were lower than in SBM 1 and 5(P<0.05).Sows had significantly greater SID values for Lys,Ala,and Asp during mid-gestation when compared with late-gestation stages(P<0.05).Mid-gestating sows had greater SID value for Val and lower SID value for Tyr when compared with non-pregnant and late-gestating sows(P<0.01),whereas non-pregnant sows had significantly greater SID value for Met when compared with gestating sows(P<0.01).Conclusions When fed to mid-gestating sows,the SID values for most AAs varied among SBM samples.The SID values for Lys,Met,Val,Ala,Asp,and Tyr in SBM were affected by sow gestation stages.Our findings provide a cornerstone for accurate SBM use in sow diets. 展开更多

Drought is one of the abiotic stresses limiting the production of soybean(Glycine max).Elucidation of the genetic and molecular basis of the slow-wilting(SW)trait of this crop offers the prospect of its genetic improvement.A panel of 188 accessions and a set of recombinant inbred lines produced from a cross between cultivars Liaodou 14 and Liaodou 21 were used to identify quantitative-trait loci(QTL)associated with SW.Plants were genotyped by Specific-locus amplified fragment sequencing and seedling leaf wilting was assessed under three water-stress treatments.A genome-wide association study identified 26 SW-associated single-nucleotide polymorphisms(SNPs),including three located in a 248-kb linkage-disequilibrium(LD)block on chromosome 2.Linkage mapping revealed a major-effect QTL,qSW2,associated with all three treatments and adjacent to the LD block.Fine mapping in a BC_(2)F_(3) population derived from a backcross between Liaodou 21 and R26 confined qSW2 to a 60-kb interval.Gene expression and sequence variation analysis identified the gene Glyma.02 g218100,encoding an auxin transcription factor,as a candidate gene for qSW2.Our results will contribute significantly to improving drought-resistant soybean cultivars by providing genetic information and resources. 展开更多

Enzymatic hydrolysis of proteins can enhance their emulsifying properties and antioxidant activities.However,the problem related to the hydrolysis of proteins was the generation of the bitter taste.Recently,high hydrostatic pressure(HHP)treatment has attracted much interest and has been used in several studies on protein modification.Hence,the study aimed to investigate the effects of enzymatic hydrolysis by Corolase PP under different pressure treatments(0.1,100,200,and 300 MPa for 1-5 h at 50℃)on the emulsifying property,antioxidant activity,and bitterness of soybean protein isolate hydrolysate(SPIH).As observed,the hydrolysate obtained at 200 MPa for 4 h had the highest emulsifying activity index(47.49 m^(2)/g)and emulsifying stability index(92.98%),and it had higher antioxidant activities(44.77%DPPH free radical scavenging activity,31.12%superoxide anion radical scavenging activity,and 61.50%copper ion chelating activity).At the same time,the enhancement of emulsion stability was related to the increase of zeta potential and the decrease of mean particle size.In addition,the hydrolysate obtained at 200 MPa for 4 h had a lower bitterness value and showed better palatability.This study has a broad application prospect in developing food ingredients and healthy foods. 展开更多

This study was to explore the functional mechanism of rare earth regulating soybean leaves and the characteristics and functions of differentially expressed proteins under the regulation of rare earth. In this study, Dongnong 42 was used as material, and 30 mg·L^(-1) CeCl_(3) solution was sprayed on soybean leaves at the seedling stage. Tandem mass tag(TMT) quantitative proteomics technique and bioinformatics analysis were used to identify soybean leaf proteins. A total of 8 510 proteins were identified, and 127 differentially expressed proteins(DEPs) in response to rare earth cerium regulation were identified, among which 64 were upregulated and 63 were down-regulated. The gene ontology(GO) annotation indicated that DEPs were mainly involved in metabolic process, cellular process, response to stimulus, biological regulation, and response to a stimulus;DEPs in cell module categories were mainly involved in cells, cell part, organelle, membrane, membrane part, organelle par, and protein-containing complex;DEPs in molecular functional categories were mainly involved in catalytic activity, binding and antioxidant activity. Kyoto encyclopedia of genes and genomes(KEGG) pathway significantly enriched starch and sucrose metabolism, glycolysis/gluconeogenesis, galactose metabolism, pentose phosphate pathway, and MAPK signaling pathway-plant. These DEPs were mainly involved in photosynthesis, glucose metabolism and stress response. Forty-six differential protein interaction networks were identified by protein interaction network analysis. This experiment provided a reference for studies of the mechanism of rare earth cerium regulating soybean leaf function from the proteomic perspective. 展开更多

In order to realize genomic selection breeding for FLS resistance of soybean, least absolute shrinkage and selection operator(LASSO) regression and stepwise regression were combined, and a genomic selection model was established for 40 002 SNP markers covering soybean genome and relative lesion area of soybean FLS. As a result, 68 molecular markers controlling soybean FLS were detected accurately, and the phenotypic contribution rate of these markers reached 82.45%. In this study, a model was established, which could be used directly to evaluate the resistance of soybean FLS and to select excellent offspring. This research method could also provide ideas and methods for other plants to breeding in disease resistance. 展开更多

Shade tolerance is essential for soybeans in inter/relay cropping systems.A genome-wide association study(GWAS)integrated with transcriptome sequencing was performed to identify genes and construct a genetic network governing the trait in a set of recombinant inbred lines derived from two soybean parents with contrasting shade tolerance.An improved GWAS procedure,restricted two-stage multi-locus genome-wide association study based on gene/allele sequence markers(GASM-RTM-GWAS),identified 140 genes and their alleles associated with shade-tolerance index(STI),146 with relative pith cell length(RCL),and nine with both.Annotation of these genes by biological categories allowed the construction of a protein–protein interaction network by 187 genes,of which half were differentially expressed under shading and non-shading conditions as well as at different growth stages.From the identified genes,three ones jointly identified for both traits by both GWAS and transcriptome and two genes with maximum links were chosen as beginners for entrance into the network.Altogether,both STI and RCL gene systems worked for shade-tolerance with genes interacted each other,this confirmed that shadetolerance is regulated by more than single group of interacted genes,involving multiple biological functions as a gene network. 展开更多

Resuits indicated that genetic factors of isoflavone contents in F2 population inherited quantitatively. Isoflavone content of F1, F2 seeds normally trended. There were heterosis in F1, F2 of most combinations, and also heterobeltiosis in part of the crosses. The broad sense heritability of F2 was higher in parts of the crosses. It predicted the selection might be carried out preliminarily in F2 hybrids. There was significant positive correlation between hybrids and mid-parent. 展开更多

It is important to determine the isoflavone components by high-performance liquid chromatography (HPLC) for the molecular assistant selection of isoflavone in soybean. Based on the standard samples of 12 isoflavone components, the isoflavone components were analyzed using the determination of absorbance peaks method by HPLC. The results showed that there were different maximum ultraviolet (UV) absorbance for the aglycones of daidzein, glycitein, and genistein, which were at 250, 257, and 260 nm, respectively. A linear gradient elution of acetonitrile (13-30%) containing 0.1% acetic acid as a mobile phase was applied on a YMC-C18 column at 35℃. The 12 isoflavone components were determined using the UV detector by HPLC. We concluded that this is a rapid and precise method which adapted to determine the large numbers of samples with microanalysis. 展开更多

Previous research showed that foliar boron (B) application at flowering or seed-fill growth stages altered seed protein, oil, and fatty acids. The objective of this research was to investigate the effects of foliar B fertilizer on seed phenolics (phenol, lignin, and isoflavones) and sugars concentrations. A repeated greenhouse experiment was conducted on soybean [(Glycine max(L.) Merr.)] under watered and water-stressed conditions. Soybean plants were divided into different sets, and each set was subjected to one of the following treatments: W = plants were watered with no foliar B;WB = plants were watered and received foliar B;WS = plants were water-stressed with no foliar B;WSB = plants were waterstressed and received foliar B. Foliar B was applied at rate of 0.45 kg/ha twice at flowering and twice at seed-fill stages. The results showed that total phenol and lignin concentrations were higher in seed collected from water-stressed plants compared with those collected from watered plants whether B was applied or not. The higher total phenol and lignin concentration in seed collected of water-stressed plants may be due to B-deficiency in plant tissues. Application of B resulted in higher concentrations of total seed B and isoflavones under watered and water-stressed plants. Higher cell wall B was higher in water-stressed plants than in watered plants, having an opposite trend to total B. Application of B resulted in higher seed sucrose in watered and water-stressed plants, but raffinose and stachyose were significantly higher under water-stressed plants. The research demonstrated that foliar B fertilizer altered seed phenol, lignin, isoflavones, and sugars, suggesting that B involved in phenolics and sugar metabolism. The higher cell wall B in waterstressed plants than in watered plants supports previous research that B has mainly a structural role. The higher sucrose resulting from foliar B in watered plants is desirable as sucrose contributes to seed quality. The increase of raffinose and stachyose concentrations in seed of water-stressed plants is undesirable as raffinose, and especially stachyose may be involved in water stress/drought tolerance. The current knowledge would help soybean breeders select for higher phenolic compounds and desirable sugars for higher seed qualities under drought conditions. 展开更多
