AIM: To compare the clinical manifestation of cytomegalovirus(CMV) retinitis and microvascular retinopathy(MVR) in patients with acquired immunodeficiency syndrome(AIDS) in China.METHODS: A total of 93 consecutive patients with AIDS, including 41 cases of CMV retinitis and 52 cases of MVR were retrospectively reviewed. Highly active antiretroviral therapy(HAART) status was recorded. HIV and CMV immunoassay were also tested. CD4+ T-lymphocyte count and blood CMV-DNA test were performed in all patients. Aqueous humor CMV-DNA test was completed in 39 patients. Ophthalmological examinations including best corrected visual acuity(BCVA, by International Standard Vision Chart), intraocular pressure(IOP), slit-lamp biomicroscopy, indirect ophthalmoscopy were performed.RESULTS: In MVR group, the anterior segment examination was normal in all patients with a mean BCVA of 0.93±0.13. Blood CMV-DNA was 0(0, 269000) and 42 patients(80.77%) did not receive HAART. In CMV retinitis group, 13 patients(31.71%) had anterior segment abnormality. The mean BCVA was 0.64±0.35 and blood CMV-DNA was 3470(0, 1 450000). Nineteen patients(46.34%) had not received HAART. MVR group and CMV retinitis group the positive rates of aqueous CMV-DNA were 0 and 50%, respectively.Two patients with MVR progressed to CMV retinitis during the follow-up period.CONCLUSION: In comparison of CMV, patients with MVR have relatively mild visual function impairment. Careful ophthalmological examination and close follow-up are mandatory, especially for patients who have systemic complications, positive CMV-DNA test and without received HAART. 展开更多

Objective:To evaluate humoral immunity and peripheral blood T lymphocyte subset levels after Mycoplasma pneumoniae infection. Methods:118 cases of children with Mycoplasma pneumoniae infection were selected as observation group, and 98 cases of healthy children receiving medical examination in our hospital during the same period were selected as control group. The differences in humoral immunity, T lymphocyte subsets, anti-inflammatory and pro-inflammatory factors, Th1/Th2 balance state, etc were compared between two groups of children. Results:Serum IgA level of observation group was lower than that of control group (P<0.05), and IgM level was higher than that of control group (P<0.05). Peripheral blood CD3+and CD4+T lymphocyte levels as well as CD4+/CD8+ratio were lower than those of control group (P<0.05), while CD8+T lymphocyte level was higher than that of control group (P<0.05). Serum pro-inflammatory factors IL-8 and TNF-αlevels were higher than those of control group (P<0.05), while anti-inflammatory factors IL-10 and IL-13 levels were lower than those of control group (P<0.05). Serum IL-4, IFN-γand IL-4/IFN-γlevels were higher than those of control group (P<0.05). Conclusions:Humoral immune and cellular immune suppression, high inflammatory state and so on after Mycoplasma pneumoniae infection are the important mechanisms leading to disease progression. 展开更多

Objective:To investigate effect of bifidobacterium triple viable powder on inflammatory factors and T lymphocyte subsets in patients with ulcerative colitis.Method:80 patients with ulcerative colitis were selected and divided into observation group and control group, 40 cases in each group. The control group was given dietary guide, mesalazine, vitamin C and other comprehensive treatment;The observation group was given bifidobacterium triple viable powder adjuvant therapy on the basis of conventional treatment. The level of IL-6, CRP, TNF-α and T lymphocyte subsets in the two groups was compared before treatment (T0), after treatment for 1 month (T1), after treatment for 3 months (T2).Results: (1) The levels of IL-6, CRP and TNF-α at T1 serum were lower than those in T0 in both groups, the levels of serum IL-6, CRP and TNF-α at T2 were lower than those of T0 and T1, these three levels in different time points were statistical significant difference;The levels of serum IL-6, CRP and TNF-α in observation group at T1 and T2 were significantly lower than those in control group, there was significant difference in both groups;(2)The levels of CD3+, CD4+, CD4+/CD8+ at T1 were higher than T0 in two groups, The levels of CD3+, CD4+, CD4+/CD8+ at T2 were superior than at T0 and T1. The level of CD8+ at T1 was below at T0, The level of CD8+ at T2 was inferior than those in T0 and T1, The levels of CD3+,CD4+,CD8+ and CD4+/CD8+ at different time points were significant difference. The levels of CD3+, CD4+, CD4+ /CD8+ in the observation group were significantly higher than those in the control group T1 and T2, CD8+ level was significantly lower than that in the control group at T1 and T2, difference between the two groups was statistically significant.Conclusion:Bifidobacterium triple viable powder adjuvant treatment ulcerative colitis was good for inhibiting body inflammatory response and improve immune function. 展开更多

Genetic factors and gene polymorphisms leading to the onset of autoimmune response in autoimmune hepatitis(AIH)are still not full elucidated.Since the CTLA-4 molecule is a key modulator of the lymphocytes responses we hypothezied that deficiencies or mutations in the gene encoding CTLA4 protein may be involved in AIH susceptibility and trigger the autoimmune response.We investigated 3 distinct polymorphic sites(t49A>G,CT60 G>A and e318C>T)of the CTLA4 gene in 50 AIH patients and 100 healthy controls using the KASP genotyping technology.A significant positive association with AIH susceptibility was found for the GG genotype in t49 position of the CTLA4 gene which was significantly higher in AIH patients compared to controls(28%vs 9%,pZ0.003,ORZ3.93[1.56e9.88]).The CTLA4 A/A genotype in position CT60 was more significantly frequent in controls comparing to AIH patients and could be considered as a protective genotype for the tunisian patients.CTLA4 genotyping in position318 did not show any statistically significant difference in genotype or allele distribution.The CTLA4 gene polymorphism in position t49 is associated to AIH susceptibility in the Tunisian population.Mutation in the CTLA4 gene may lead to a modification of the CTLA4 protein structure that could have functional relevance in AIH pathogenesis and onset. 展开更多

Effects of Shikonin on the Proliferation and Activation of T Lymphocytes

Objective: To investigate the effects of Chinese herbal monomer shikonin which has the function of cooling blood and detoxification on abnormally activated T lymphocyte and its related signal pathways and to elucidate the role of shikonin in the pathological state of psoriasis and its mechanism of treating psoriasis. Materials and Methods: Jurkat E6?1 T lymphocytes were activated with phorbol ester and ionomycin. The shikonin concentration of 0.5–2 μg/mL was applied to the cells, and the proliferation of T lymphocytes was detected by Cell Counting Kit?8 assay. Flow cytometry was used to measure CD69 expression on the cell membrane and intracellular free calcium ion concentration([Ca^(2+)]i); enzyme?linked immunosorbent assay was used to detect the levels of interleukin(IL)?2, interferon?γ(IFN?γ), and(TNF?α) released by activated T lymphocytes; quantitative polymerase chain reaction and Western blot were used to observe the expression of nuclear transcription factor m RNA and protein, respectively. Results: Different concentrations of shikonin could significantly inhibit cell proliferation, CD69 expression, and secretion of Th1 cytokines. In addition, shikonin could effectively reduce the [Ca^(2+)]i and protein kinase C phosphorylation proteins. Besides that, shikonin could significantly reduce the nuclear transcription factor nuclear factor of activated T lymphocytes mRNA expression, downregulate c?Jun mRNA and protein expression, and inhibit NF?κB protein expression. All the above indicators show a certain dose–effect relationship. Conclusions: Shikonin can exert immune regulation by inhibiting the function of overactivated T lymphocytes. Hence, this study provides experimental basis for the mechanism and application prospect of shikonin in the treatment of psoriasis. 展开更多
