Protective mechanism of quercetin compounds against acrylamide-induced hepatotoxicity

Quercetin compounds have antioxidant,anti-inflammatory and anticancer pharmacological functions.Longterm exposure to acrylamide(AA)can cause liver injury and endanger human health.However,whether quercetin compounds can attenuate AA-induced liver injury and the specific mechanism are not clear.Here,we studied the mechanism and structure-activity relationship of quercetin compounds in reducing AA-induced hepatotoxicity in vivo and in vitro.In vivo studies found that quercetin-like compounds protect against AAinduced liver injury by reducing oxidative stress levels,activating the Akt/m TOR signaling pathway to attenuate autophagy,and improving mitochondrial apoptosis and endoplasmic reticulum stress-mediated apoptosis.In vitro studies found that quercetin compounds protected Hep G2 cells from AA by attenuating the activation of AA-induced autophagy,lowering reactive oxygen species(ROS)levels by exerting antioxidant effects and thus attenuating oxidative stress,increasing mitochondrial membrane potential(MMP),and improving apoptosis-related proteins,thus attenuating AA-induced apoptosis.Furthermore,the conformational differences between quercetin compounds correlated with their protective capacity against AA-induced hepatotoxicity,with quercetin showing the best protective capacity due to its strongest antioxidant activity.In conclusion,quercetin compounds can protect against AA-induced liver injury through multiple pathways of oxidative stress,autophagy and apoptosis,and their protective capacity correlates with antioxidant activity. 展开更多

Our previous study has revealed that procyanidin A_(1)(A_(1))and its simulated digestive product(D-A,)can alleviate acrylamide(ACR)-induced intestine cell damage.However,the underlying mechanism remains unknown.In this study,we elucidated the molecular mechanism for and D-A_(1) to alleviate ACR-stimulated IPEC-J2 cell damage.ACR slightly activated nuclear factor erythroid 2-related factor 2(Nrf2)signaling and its target genes,but this activation could not reduce intestine cell damage.A_(1) and D-A_(1) could alleviate ACR-induced cell damage,but the effect was abrogated in cells transiently transfected with Nrf2 small interfering RNA(siRNA).Further investigation confirmed that A_(1) and D-A_(1) interacted with Ketch-like ECH-associated protein 1(Keapl),which boosted the stabilization of Nrf2,subsequently promoted the translocation of Nrf2 into the nucleus,and further increased the expression of antioxidant proteins,thereby inhibiting glutathione(GSH)consumption,maintaining redox balance and eventually alleviating ACR-induced cell damage.Importantly,there was no difference between A_(1) and D-A_(1) treated groups,indicating that A_(1) can tolerate gastrointestinal digestion and may be a potential compound to limit the toxicity of ACR. 展开更多

This study was carried out to prepare ZnO nanoparticles incorporated acrylamide grafted chitosan composite film for possible biomedical application especially drug loading in wound healing. ZnO nanoparticles were prepared by co-precipitation method from zinc acetate di-hydrate and incorporated in acrylamide grafted chitosan. FT-IR and TGA of the prepared composite film confirmed the successful incorporation of ZnO nanoparticles in the acrylamide-grafted polymer matrix. SEM images showed that the ZnO nanoparticles were homogeneously distributed on the porous matrix of the composite film. Water uptake and buffer uptake analysis revealed that the composite film could hold water and buffer sufficiently, which facilitated the absorption of exudate from the wound site. Amoxicillin was loaded in the prepared composite film and the maximum loading efficiency was found to be 67.33% with drug concentration of 300 ppm. In vitro studies showed greater antimicrobial activity of drug-loaded composite film compared to both pure film and standard antibiotic disc. Finally, the In vivo mouse model showed maximum healing efficiency compared to conventional gauge bandages because the loading of antibiotic in the film produced a synergistic effect and healing time was reduced. 展开更多

Objective:To investigate the effects of melatonin on renal inflammation,oxidative stress,apoptosis,as well as DNA and tissue damage in acrylamide-induced nephrotoxicity in rats.Methods:Fifty male rats were randomly divided into five groups.The control group received distilled water by gastric lavage for 11days and the acrylamide group was administered acrylamide(50 mg/kg,i.g.)for 11 days.The MEL10+ACR and MEL20+ACR groups received intraperitoneal melatonin 10 and 20 mg/kg,respectively,for 11 days,and acrylamide(50 mg/kg,i.g.)was administered 1h after melatonin injection.The MEL20 group was injected with melatonin(20 mg/kg)for 11 days.Kidney function tests were performed and biochemical and inflammatory parameters were determined.In addition,histopathological,immunohistochemical,and immunofluorescence examinations were carried out.Results:Melatonin significantly abated acrylamide-induced rise in serum urea and creatinine levels.Acrylamide caused oxidative stress,inflammation,apoptosis,as well as DNA and tissue damage in the kidneys.Melatonin treatment alleviated acrylamide-induced renal damage by exhibiting antioxidant,anti-inflammatory,and antiapoptotic effects.Moreover,melatonin significantly ameliorated acrylamide-caused histopathological changes in kidney tissue.Conclusions:Melatonin attenuates acrylamide-induced renal oxidative stress,inflammation,apoptosis,and DNA damage in rats. 展开更多

We report simple acrylamide polymerization process to prepare nanocrystalline tin dioxide(SnO_2) particles.A small amount of the so-called premix solution(monofunctional acrylamide and difunctional N,N'-methylenebisacrylamide) was mixed with the aqueous solution of Sn^(4+) in certain proportion,and a few drops of ammonium persulfate solution were added as initiator.When homogenized by magnetic stirring and heated to about 70℃,the mixed solution became a gel.This hydrous gel was directly decomposed by thermal treatment,resulting in obtaining rutile crystalline SnO_2 particles with a narrow size distribution typically in the range of 20 nm~40 nm. 展开更多

Ce0.8Sm0.2O1.9(SDC) nanopowders were synthesized by an acrylamide polymerization process.The XRD results showed that SDC powders prepared at 700 °C possessed a cubic fluorite structure.Transmission electron microscopy(TEM) indicated that the particle sizes of powders were in the range of 10-15 nm.A 98.3% of theoretical density was obtained when the SDC pellets were sintered at 1350 °C for 5 h,indicating that the powders had good sinterability.The conductivity of the sintered SDC ceramics was 0.019 S/cm at 600 °C and the activation energy was only 0.697 eV.Furthermore,a unit cell was fabricated from the powders and the maximum power density of 0.169 W/cm2 was achieved at 700 °C with humidified hydrogen as the fuel and air as the oxidant. 展开更多

Lignin isolated from enzymatic hydrolyzed corn-stalks (EH-lignin) is a renewable natural polymer noted for its versatility and applicability in a vari-ety of uses. Graft copolymerization of EH-lignin with acrylamide (AM) and the application of this copolymer as a flocculant in dye wastewater treatment were studied in this article. The influ-ences of some factors on yield of copolymer and the grafting ratio were investigated and the structure of EH-lignin/AM graft copolymer was characterized by FT-IR. According to the yield and the grafting ratio, the optimum conditions for graft copolymerization were as follows: initiator K2S2O8-Na2S2O3 with a quantity 3 wt% of EH-lignin, mass ratio of AM to EH-lignin was 2~3, reaction time 4h and temperature at 50℃. It was found that the absorption capacity of graft copolymer to two azo-dyes was enhanced with the increase of grafting ratio. Furthermore, the residue concen-tration of EH-lignin/AM graft copolymer remained in the supernatant after flocculation was much lower than that of pure EH-lignin. 展开更多

Objective:To explore the possible effects of naringin on acrylamide-induced nephrotoxicity in rats.Methods:Sprague-Dawley rats weighing 200-250 g were randomly divided into five groups.The control group was given intragastric(i.g.)saline(1 mL)for 10 d.The acrylamide group was given i.g.acrylamide in saline(38.27 mg/kg titrated to 1 mL)for 10 d.The treatment groups were administered with naringin in saline(50 and 100 mg/kg,respectively)for 10 d and given i.g.acrylamide(38.27 mg/kg)1 h after naringin injection.The naringin group was given i.g.naringin(100 mg/kg)alone for 10 d.On day 11,intracardiac blood samples were obtained from the rats when they were under anesthesia,after which they were euthanized.Urea and creatinine concentrations of blood serum samples were analyzed with an autoanalyzer.Enzyme-linked immunosorbent assay was used to quantify malondialdehyde,superoxide dismutase,glutathione,glutathione peroxidase,catalase,tumor necrosis factor-α,nuclear factor-κB,interleukin(IL)-33,IL-6,IL-1β,cyclooxygenase-2,kidney injury molecule-1,mitogen-activated protein kinase-1,and caspase-3 in kidney tissues.Renal tissues were also evaluated by histopathological and immunohistochemical examinations for 8-OHdG and Bcl-2.Results:Naringin attenuated acrylamide-induced nephrotoxicity by significantly decreasing serum urea and creatinine levels.Naringin increased superoxide dismutase,glutathione,glutathione peroxidase,and catalase activities and decreased malondialdehyde levels in kidney tissues.In addition,naringin reduced the levels of inflammatory and apoptotic parameters in kidney tissues.The histopathological assay showed that acrylamide caused histopathological changes and DNA damage,which were ameliorated by naringin.Conclusions:Naringin attenuated inflammation,apoptosis,oxidative stress,and oxidative DNA damage in acrylamide-induced nephrotoxicity in rats. 展开更多

The preparation and application of functional hydrogels based on natural polysaccharides have always been a hot research topic.In this study,using acrylamide(AM)monomer,N,N'-methylene bisacrylamide(MBA)as crosslinking agent,potassium persulfate(K2S2O8)as initiator,in the presence of natural polysaccharide sodium alginate(SA),the PAM/SA hydrogel was prepared by free radical polymerization.Fourier transform infrared spectroscopy(FT-IR),swelling performance tests,scanning electron microscope(SEM),thermogravimetric analysis(TGA),UV-visible spectrophotometer,mechanical property measurements were carried out to analyze the composition,morphology,and performance of the hydrogels.The swelling behavior,dye adsorption performance,and the mechanical properties of PAM/SA hydrogels before and after Fe^(3+)adsorption were studied.The experimental results showed that the introduction of SA with 4.7%,7.8%,and 10.3%effectively improved the mechanical and dye adsorption properties of PAM composite hydrogels.The adsorption capacity of PAM/4.7%SA and PAM/10.3%SA hydrogels at equilibrium can reach 40.01 and 44.02 mg/g for methylene blue,which is higher than the value 13.58 mg/g of pure PAM hydrogel.The compressive strength of pure PAM hydrogel is 0.124 MPa.When the SA content is 4.7%,7.8%,and 10.3%,the compressive strength of the PAM/SA hydrogel was corresponding to 0.130 MPa,0.134 MPa,and 0.152 MPa,respectively.Fe^(3+)was introduced into the PAM/SA hydrogels,and PAM/SA/Fe^(3+)double-network hydrogels with excellent mechanical properties could be prepared by adjusting the SA content(4.7%,7.8%,and 10.3%),soaking time(1 h,2 h,3 h,4 h,5 h,6 h),and Fe^(3+)concentration(4.76%,7.41%,9.09%,and 13.04%).Under the same Fe^(3+)concentration of 9.09%and adsorption time of 4 h,the compressive strengths of the PAM/4.7%SA,PAM/7.8%SA,and PAM/10.3%SA hydrogels could reach 0.354 MPa,0.767 MPa,and 0.778 MPa,respectively. 展开更多

A new classification of the different types of fullerene-containing polymers is presented according to their different properties and applications they exhibit in a variety of fields. 13C NMR and Raman studies of a series of polymeric samples of fullerene-grafted poly (acrylamide), which were prepared by systematic variation of concentration of fullerene and acrylamide, are described. 13C NMR spectral analysis of the polymeric samples showed a peak for fullerene at 143 δppm and for poly (acrylamide) between 170 and 180 δppm and Raman spectral analysis of the poly-meric samples gave the Raman band for fullerene between 1470 cm-1 and poly (acrylamide) at 2800 cm-1. The Tg value, obtained from DSC results, showed a high glass transition temperature at 100.94°C revealing the presence of fullerene in the polymeric matrix. TGA analysis shows that polymer is thermally stable up to 340°C. 展开更多

In this study, it was to investigate the swelling performance of novel biohybrid composite hydrogel sorbents containing acrylamide/potassium 3-sulfopropyl methacrylate/sodium alginate/bentonite in water and binary mixtures of water-solvent. Novel hydrogels were synthesized with free radical solution polymerization by using ammonium persulfate/N,N,N’,N’-tetramethylethylenediamine as redox initiating pair in presence of poly(ethylene glycol) diacrylate as crosslinker. Swelling experiments were performed in water and binary mixtures of water-solvent (acetone, methanol and tetrahydrofuran) at 25°C, gravimetrically. Some swelling and diffusion properties of the hydrogels were calculated and they were discussed for the biohybrid/hybrid hydrogel systems prepared under various formulations. It has been seen the lower equilibrium percentage swelling ratio values (62% - 124%) in all solvent compositions in comparison with the equilibrium percentage swelling ratio values in water (718% - 2055%). Consequently, the hydrogel systems developed in this study could serve as a potential device for water and water-solvent binary mixtures. 展开更多

Two different methods have been used to introduce—COONa groups into polymer chains of the gels: (1) by partial hydrolysis of acrylamide homopolymer gel; (2) by direct copolymerization and crosslinking of acrylamide and sodium acrylate in aqueous solutions. It was found that the gels obtained in different ways had different properties, the swelling character of the gels obtained by partial hydrolysis were more sensitive to pH of swelling aqueous media, in order to explain these differences, <sup>13</sup>C-NMR techniques were used to investigate the sequence distribution of monomer units of both gels. 展开更多

Polymerization of acrylamide initiated by ceric ammonium nitrate alone has been studied in aqueous medium. The effects of UV light irradiation on the initial rates of polymerization, the activation energy and on the polymer molecular weights have been investigated. Compared with that in the dark, the rate of polymerization under UV light was accelerated to eleven times higher, and the overall activation energy was lowered markedly. 展开更多

It has been reported that acrylamide can cause DNA damage and cytotoxicity. The present study aimed to investigate the potential mechanism of human hepatocarcinoma HepG2 cell proliferation induced by acrylamide and to explore the antagonistic effects of a natural polyphenol curcumin against acrylamide via miR-21. The results indicated that acrylamide(≤100μmol/L) significantly increased HepG2 cell proliferation and miR-21 expression. In addition, acrylamide reduced the PTEN expression in protein level, while induced the expressions of p-AKT, EGFR and cyclin D1. The PI3 K/AKT inhibitor decreased p-AKT protein expression and inhibited the proliferation of HepG2 cells. In addition, curcumin effectively reduced acrylamide-induced HepG2 cell proliferation and induced apoptosis through the expression of miR-21. In conclusion, the results showed that acrylamide increased HepG2 cell proliferation via upregulating miR-21 expression, which may be a new target for the treatment and prevention of cancer. 展开更多

A sensitive, precise, accurate and simple in house validated Liquid Chromatography and Tandem mass spectrometric (LC-MS/MS) analytical method was developed for the quantification of acrylamide in potato chips, a probable carcinogenic and neurotoxic compound in fried food items. Acrylamide was extracted and cleaned up by QuEChERS method, then analyzed by triple quadrupole mass spectrophotometer using positive electrospray-ionization mode and d3-Acrylamide internal standard. The analyte and internal standard were separated on a reversed-phase C18 column (100 × 2.0 mm, 2.5 μm) by gradient elution with water and methanol containing 0.1% formic acid as mobile phase. The analytical method was fully validated by assessment of linearity, specificity, precision, accuracy, selectivity and robustness with relative standard deviation of less than 4%. The calibration curves were found linear with correlation coefficient (r2) of 0.9997 over the concentration range 5 - 500 μg/L. The recoveries of Acrylamide in the concentration of 250, 400, and 500 μg/L were 97% to 104%. Based on the signal to noise ratio, the lower limit of detection (LOD) and lower limit of quantification (LOQ) were achieved 2.0 μg/L and 4.0 μg/L respectively. The proposed method was also applied to other available most popular 24 types of food (265 number of sample) first time in Bangladesh. The highest level of acrylamide has been found in fried products with the range of 197.04 μg/L to 114.63 μg/L, protein-rich food lies between 79.76 to 89.14 μg/L whereas baked food products exhibited less content in the range of 35.23 to 51.17 μg/L. 展开更多
