Two winter wheat(Triticum aestivum L. ) cultivars, large-spike type Yumai66 amd small-spike type Yumai49, were used to study the activities of enzymes involved in starch synthesis in the kernel during grain filling. Starch accumulated faster in the kernel of Yumai49 than Yumai66 up to 25 d after anthesis, thereafter starch accumulated faster in the kernel of Yumai66. Starch accumulation in Yumai66 peaked at 20 -25 d after anthesis, while in Yumai49 starch accumulation peaked at 15 -20 d after anthesis and 25 -30 d after anthesis. The first peak was much higher than that of the second. Sucrose content and sucrose synthase activity peaked at 20 and 15 d after anthesis in Yumai66 and Yumai49, respectively. The sucrose content and sucrose synthase activity in Yumai66 were higher than that in Yunai49 during grain filling. ADP-glucose pyrophosphorylase and starch branching enzyme activity in the kernel of Yumai66 peaked at 20 d after anthesis, while soluble starch synthase activity peaked at 10 and 20 d after anthesis. The second peak was much higher than that of the first. 展开更多

Soil microorganisms play important roles in nitrogen transformation. The aim of this study was to characterize changes in the activity of nitrogen transformation enzymes and the abundance of nitrogen function genes in rhizosphere soil aerated using three different methods(continuous flooding(CF), continuous flooding and aeration(CFA), and alternate wetting and drying(AWD)). The abundances of amoA ammonia-oxidizing archaea(AOA) and ammonia-oxidizing bacteria(AOB), nirS, nirK, and nifH genes, and the activities of urease, protease, ammonia oxidase, nitrate reductase, and nitrite reductase were measured at the tillering(S1), heading(S2), and ripening(S3) stages. We analyzed the relationships of the aforementioned microbial activity indices, in addition to soil microbial biomass carbon(MBC) and soil microbial biomass nitrogen(MBN), with the concentration of soil nitrate and ammonium nitrogen. The abundance of nitrogen function genes and the activities of nitrogen invertase in rice rhizosphere soil were higher at S2 compared with S1 and S3 in all treatments. AWD and CFA increased the abundance of amoA and nifH genes, and the activities of urease, protease, and ammonia oxidase, and decreased the abundance of nirS and nirK genes and the activities of nitrate reductase and nitrite reductase, with the effect of AWD being particularly strong. During the entire growth period, the mean abundances of the AOA amoA, AOB amoA, and nifH genes were 2.9, 5.8, and 3.0 higher in the AWD treatment than in the CF treatment, respectively, and the activities of urease, protease, and ammonia oxidase were 1.1, 0.5, and 0.7 higher in the AWD treatment than in the CF treatment, respectively. The abundances of the nirS and nirK genes, and the activities of nitrate reductase and nitrite reductase were 73.6, 84.8, 10.3 and 36.5% lower in the AWD treatment than in the CF treatment, respectively. The abundances of the AOA amoA, AOB amoA, and nifH genes were significantly and positively correlated with the activities of urease, protease, and ammonia oxidase, and the abundances of the nirS and nirK genes were significantly positively correlated with the activities of nitrate reductase. All the above indicators were positively correlated with soil MBC and MBN. In sum, microbial activity related to nitrogen transformation in rice rhizosphere soil was highest at S2. Aeration can effectively increase the activity of most nitrogen-converting microorganisms and MBN, and thus promote soil nitrogen transformation. 展开更多

Starting from sissotrin (1), a natural isoflavonoid isolated from Trifolium baccarinii (Fabaceae), one new semisynthetic derivative, 6-nitrobiochanin A (1b) and two known derivatives, 8-nitrobiochanin A (1a) and 2",3",4",6"-tetraacetylsissotrin (1c) have been obtained after performing nitration and acetylation reactions. Their structures were assigned after interpretation of their spectrometric (HR-ESI-MS) and spectroscopic (NMR 1D and 2D) data and by comparison with those reported in the literature. The substrate as well as the semisynthetic derivatives were evaluated for their antibacterial activities against six strains. The results reveal that they are inactive or weakly active on the strains tested with the exception of 8-nitrobiochanin A (1a) which showed moderate activity (MIC = 62.5 μg∙mL−1) on Staphylococcus aureus ATCC 43300. 展开更多

Root exudates serve as crucial mediators for information exchange between plants and soil, and are an important evolutionary mechanism for plants’adaptation to environmental changes. In this study,15 different abiotic stress models were established using various stress factors, including drought(D),high temperature(T),nitrogen deficiency(N),phosphorus deficiency(P),and their combinations. We investigated their effects on the seedling growth of Salvia miltiorrhiza Bunge and the activities of Solid-Urease(S-UE),Solid-Nitrite Reductase(S-NiR),Solid-Nitrate Reductase(S-NR),Solid-Phosphotransferase(S-PT),and Solid-Catalase(S-CAT),as well as the contents of polysaccharides in the culture medium. The results showed that the growth of S.miltiorrhiza was inhibited under 15 stress conditions.Among them,13 stress conditions increased the root-shoot ratio. These 15 stress conditions significantly reduced the activity of S-NR,two combinations significantly improved the activity of S-NIR,they were synergistic stresses of high temperature and nitrogen deficiency(TN),and synergistic stresses of drought and nitrogen deficiency(DN)(p<0.05).The activity of S-UE was significantly improved under N,D,T,synergistic stresses of drought and high temperature(DT),DN,synergistic stresses of drought and phosphorus deficiency(DP),and synergistic stresses of high temperature, nitrogen, and phosphorus deficiency(TNP)stress conditions(p<0.05).Most stress combinations reduced the activity of S-PT,but D and T significantly improved it.(p<0.05).The N,DN,and TN stress conditions significantly reduced S-CAT activity. The P,DT,and synergistic stresses of drought, high temperature, and phosphorus deficiency(DTP)significantly decreased the total polysaccharide content of the soil(p<0.05).The research suggested that abiotic stress hindered the growth of S.miltiorrhiza and altered the behavior of root secretion.Roots regulated the secretion of several substances in response to various abiotic stresses, including soil nitrogen cycle enzymes, phosphorus transport-related enzymes, and antioxidant enzymes. In conclusion, plants regulate the utilization of rhizosphere substances in response to abiotic stresses by modulating the exudation of soil enzymes and polysaccharides by the root system. At the same time, soil carbon sequestration was affected by the adverse environment, which restricted the input of organic matter into the soil. 展开更多

Whether climate change or anthropogenic activities play a more pivotal role in regulating vegetation growth on the Tibetan Plateau is still controversial.A better understanding on grassland changes at a fine scale may provide important guidance for local government policy and grassland management.Using two of the most reliable satellite NDVI products(MODIS NDVI and SPOT NDVI),we evaluated the dynamic of grasslands in the Zhegucuo valley on the southern Tibetan Plateau from 2000 to 2020,and analyzed its driving factors and relative influences of climate change and anthropogenic activities. Here, the key indicators of climate change were assumed to be precipitation and temperature. The main results were:(1)the grassland NDVI in Zhegucuo valley did not reflect a significant temporal change during the last 21 years. The variation of precipitation during the early growing season(GSP)resembled that of NDVI,and the GSP was positively correlated with NDVI.At the pixel level, the partial correlation analysis showed that 37.79%of the pixels depicted a positive relationship between GSP and NDVI,while 11.32%of the pixels showed a negative relationship between temperature during the early growing season(GST)and NDVI.(2)In view of the spatial distribution, the areas mainly controlled by GSP were generally distributed in the southern part, while those affected by GST stood in the eastern part, mainly around the Zhegucuo lake where most population in Cuomei County settled down.(3)Decreasing NDVI trends were mainly occurred in alpine steppe at lower elevations rather than alpine meadow at higher elevations.(4)The residual trend(RESTREND)analysis further indicated that the anthropogenic activities played a more pivotal role in regulating the annual changes of NDVI rather than climate factors in this area.Future studies should pay more attention on climate extremes rather than the simple temporal trends. Also, the influence of human activities on alpine grassland needs to be accessed and fully considered in future sustainable management. 展开更多

Drinking good quality water is essential for better health. It is therefore essential to assess the radiological quality of all water consumed in the District of Abidjan in order to prevent related hazards. Thus, the objective of this study was to assess the risk of cancer due to the ingestion of alpha and beta emitting radionuclides in the different types of water consumed in the region. A total of 63 water samples with 43 tap water samples, 5 bottled mineral water and 15 sachet water samples was collected and taken to GAEC laboratory for analysis. The low background Gas-less Automatic Alpha/Beta counting system (Canberra iMaticTM) was used to determine alpha and beta activity concentrations. Activity concentrations of both gross alpha and gross beta obtained in water sample were respectively lower than the WHO recommended limits of 0.1 Bq/l and 1 Bq/l. Also, the annual effective dose and total equivalent effective dose found in mineral bottled water samples were higher than in other types of water. The assessment of radiological lifetime risk has shown values of cancer risk due to ingestion alpha and beta emitters lower than recommended limit. These results indicate that there is no health hazard associated to consumption of water in the District of Abidjan. 展开更多

Background: Self-care tasks or activities of daily living (ADL)’s performance are often disrupted after an older adult sustains an injury or illness, which can be a determining factor for discharge placement after hospitalization, or it can determine the level of assistance required after discharge from a skilled nursing facility (SNF). We believe that comprehensive rehabilitation can improve a patient’s overall functioning during their short term stay in SNFs. The purpose of this study is to determine if an individual’s level of independence improved after admission to a skilled nursing facility and procurement of rehabilitation services. Methods: This study retrospectively reviewed 4612 nursing home patients in California who underwent rehabilitative services at skilled nursing facilities to determine how the patients responded to their environments, therapy interventions, and any other additional supportive measures. The records of patients who were admitted for short-term rehabilitation were reviewed in a blinded fashion, looking specifically at Section GG of the Minimum Data Set (MDS). Self-Care Section GG scores recorded by rehabilitative staff provide objective data and measure patients’ levels of assistance that were required. Each activity is scored from 1, being completely dependent, to 6, being entirely independent, with varying levels of assistance with scores in between. The admission scores versus the discharge scores can be compared to determine if a patient improved their level of functioning upon discharge. Results: Utilizing the Pearson’s correlation coefficient, a strong correlation of improvement in ADL performance on Section GG between the admission and discharge scores was identified, implying significant improvement in functional independence upon discharge. The average percent improvement for Managed Care patients was ~35.4% and ~39.4% for Medicare patients. Conclusions: The results support the benefits of rehabilitation services in skilled nursing facilities, and the data suggests that admission scores can be used as a predictive tool for functional outcomes. 展开更多

Objective:To investigate the relationship between triterpenoid saponin content and antioxidant, antimicrobial, and α-glucosidase inhibitory activities of 70%ethanolic,butanolic,aqueous,supernate and precipitate extracts of Juglans regia leaves. Methods:Triterpenoid saponins of different Juglans regia leaf extracts were measured by the vanillin method.Antioxidant activity was evaluated against DPPH and ABTS free radicals. We also assessed α-glucosidase inhibitory and antimicrobial activities of the leaf extracts.Pearson’s correlation coefficient was evaluated to determine the correlation between the saponin content and biological activities. Results:The butanolic extract was most effective against DPPH with an IC50of 6.63μg/mL,while the aqueous extract showed the highest scavenging activity against ABTS free radical with an IC50of 42.27μg/mL.Pearson’s correlation analysis indicated a strong negative correlation (r=-0.956) between DPPH radical scavenging activity (IC50) and the saponin content in the samples examined. In addition, the aqueous extract showed the best α-glucosidase inhibitory activity compared with other extracts. All the extracts had fair antibacterial activity against Bacillus subtilis,Escherichia coli, and Klebsiella pneumoniae except for the aqueous extract. Conclusions:Juglans regia extracts show potent antioxidant, antimicrobial, and α-glucosidase inhibitory activities. There is a correlation between saponin levels in Juglans regia leaf extracts and the studied activities. However, additional research is required to establish these relationships by identifying the specific saponin molecules responsible for these activities and elucidating their mechanisms of action. 展开更多

Two phenolic acids P-hydroxy benzoic acid and cinnamic acid were designated as four concentrations (0, 50μmol/L, 100μmol/L, 150μmol/L) to investigate the effects of phenoic acids on the growth and the activities of membrane protective enzymes of cucumber seedlings. The results showed that both phenolic acids inhibited the seedlings growth. The inhibitory effects were increased with the concentration of phenolic acids increasing and the time of treatment prolonging. Seedlings treated with A150 (P-hydroxy benzoic acid, 150μmol/L), B50 (cinnamic acid, 50 μmol/L), B100 (cinnamic acid,100μmol/L), B150 (cinnamic acid, 150μmol/L) showed significantly shorter in plant height , smaller in leaf area. and lighter in fresh weight. The inhibitory effect of cinnamic acid was comparatively stronger than that of P-hydroxy benzoic acid. For protective enzymes system, compared to control, the POD activity increased at all concentrations of P-hydroxy benzoic acid during the treatment but increased at first then decreased before increased again at last at all concentrations of cinnamic acid . In the case of CAT, its activity increased at first, then decreased, and increased again at lower concentrations of phenolic acids. However, at higher concentrations the activities decreased at first, then increased a little, decreased continuously at last. In addition, the treatments of phenolic acids led to an increase then a decreaseof SOD activity and an increase of MDA content in the seedlings. All above indicated the accumulating of free radicalsand destruction of protective enzymes at higher concentrations of phenolic acids. 展开更多

Under the combined influence of climate change and human activities, vegetation ecosystem has undergone profound changes. It can be seen that there are obvious differences in the evolution patterns and driving mechanisms of vegetation ecosystem in different historical periods. Therefore,it is urgent to identify and reveal the dominant factors and their contribution rates in the vegetation change cycle.Based on the data of climate elements(sunshine hours, precipitation and temperature),human activities(population intensity and GDP intensity)and other natural factors(altitude, slope and aspect),this study explored the spatial and temporal evolution patterns of vegetation NDVI in the Yellow River Basin of China from 1989 to 2019 through a residual method,a trend analysis, and a gravity center model, and quantitatively distinguished the relative actions of climate change and human activities on vegetation evolution based on Geodetector model. The results showed that the spatial distribution of vegetation NDVI in the Yellow River Basin showed a decreasing trend from southeast to northwest.During 1981-2019,the temporal variation of vegetation NDVI showed an overall increasing trend. The gravity centers of average vegetation NDVI during the study period was distributed in Zhenyuan County,Gansu Province, and the center moved northeastwards from 1981 to 2019.During 1981-2000 and 2001-2019,the proportion of vegetation restoration areas promoted by the combined action of climate change and human activities was the largest.During the study period(1981-2019),the dominant factors influencing vegetation NDVI shifted from natural factors to human activities. These results could provide decision support for the protection and restoration of vegetation ecosystem in the Yellow River Basin. 展开更多

White rust caused by Puccinia horiana is a destructive disease of chrysanthemum plants. To better understand the resistance mechanisms of composite species to this disease, the leaf cuticular traits, antioxidant and defensive enzymes activities of immune(Chrysanthemum makinoi var.wakasaense)and highly susceptible(Ajania shiwogiku var.kinokuniense)species were compared.Trichome density of two species was markedly different, negatively associated with plant resistance to P.horiana.Total wax load in C.makinoi var.wakasaense was two times more than that in A.shiwogiku var.kinokuniense.The wax composition in immune one was abundant in esters and primary alcohols.Superoxide dismutase(SOD,EC 1.15.1.1),peroxidase(POD,EC 1.11.1.7),polyphenoloxidase(PPO,EC 1.14.18.1 or EC 1.10.3.2)and phenylalanine ammonia lyase(PAL,EC 4.3.1.5)activitieswere investigated. In C.makinoi var.wakasaense, the activity of SOD and POD increased rapidly after inoculation, whichmight be non-host induced reactive oxygen species(ROS)activated antioxidant enzymes, however SOD and POD remained a low and steady level in the highly susceptible one after inoculation.Quick increase in PPO activities after inoculation was observed in both species, however it remained higher in C.makinoi var.wakasaense at the late period of inoculation.PAL in C.makinoi var.wakasaense was induced after pathogen inoculation, but not in A.shiwogiku var.kinokuniense, suggesting that these two enzymes might contribute to the resistance to P.horiana. 展开更多
