The samples were...A rapid and accurate quantitative method of high performance liquid chromatography( HPLC) with fluorescence detector has been developed for the analysis of 18 kinds of amino acids in fresh tea leaves. The samples were minced and mixed,and extracted with ultra pure water at 90℃ for 20 min. The 6-aminoquinolyl N-hydroxy-succinimidyl carbamate( AQC) was used as pre-column derivatization reagent. Gradient HPLC separation was performed on a C_(18) column( Symmetry C_(18),3. 9 mm × 15 cm,4 μm). Good linearity between concentrations and peak areas was achieved in the concentration range of 5. 0-250 μmol/L for 18 kinds of amino acids. The method was validated by the analysis of five replicates. The 18 kinds of amino acid standards were spiked in fresh tea leaf samples and the average recovery rate was 86. 25%-109. 05% with relative standard deviations( n = 5) ranging from 6. 03% to 10. 56%. The limit of detection( LOD) for the analytes was0. 05-1. 27 μmol/L. The method was successfully applied to the analysis of the 18 kinds of amino acids in fresh tea leaves from east Dongting and west Dongting mountains in Suzhou. The results indicate that the method is simple,rapid,precise and reliable.展开更多

The five derivatives were eluted in 28 min with a good reproducibility. Linear range of the calibration graph was 0.08-260 nmol/ml(-1). The relative standard deviations(n=6) are < 5%. Separation of amino acid derivatives was obtained on a reversed-phase C-18 column. Derivatization and chromatographic conditions were optimized. Amino acid derivatives were eluted in 40 min with good reproducibility. The relative standard deviations (n=6) at an analytical concentration of 100 pmol are < 4%. The experimental solubility of CBZ (Carbobenzoxy) derivatized amino acids namely N-CBZ valine, N-CBZ proline, N-CBZ aspartic acid in supercritical carbon dioxide were correlated by Soave-Red- lich-Kwong Equation state...The experimental solubility of CBZ (Carbobenzoxy) derivatized amino acids namely N-CBZ valine, N-CBZ proline, N-CBZ aspartic acid in supercritical carbon dioxide were correlated by Soave-Red- lich-Kwong Equation state based on fugacity determination and group contribution method using extrapolated critical parameters and mixing rules and other two different empirical models proposed by Yu (1994) and Gordillo-coworkers (1999). The SRK EOS prediction showed very high deviation of % AARD of 9% - 59%. The Yu model had three derivatized amino acids with average absolute deviation from 2.04, 8.17, 10.96, while the Gordillo model had 0.245, 1.067 and 1.144 for CBZ-valine, CBZ-proline and CBZ-aspartic acid successively. The correlated values had better fit with Gordillo model. The predictive capability and applicability for these amino acid derivatives for both the models demonstrated with correlation coefficient around 0.99 for all the experimental solubility observed.展开更多

We have developed a novel metal tag, bis(ethylenediamine)-4'-methyl-4-carboxybipyridine-ruthenium N-succinimidyl ester (ECRS) for sensitive analysis of amino acids using high performance liquid chromatography/indu...We have developed a novel metal tag, bis(ethylenediamine)-4'-methyl-4-carboxybipyridine-ruthenium N-succinimidyl ester (ECRS) for sensitive analysis of amino acids using high performance liquid chromatography/inductively coupled plasma mass spectrometry (HPLC/ICP-MS). ECRS is a functional reagent, containing an ester group at one end that can be activated to bind to amino group and a chelated ruthenium at the other. The activated ester was reacted briefly with amino groups under weakly alkaline conditions. The ruthenium was detected sensitively by ICP-MS. ECRS was reacted with 17 proteinogenic amino acids in borate buffer. The derivatives were separated by reversed phase HPLC and identified by quadrupole-based ICP-MS. ECRS was suitable for speciation;low molecular weight compounds containing amino groups. We have thus established a quantitative analytical method for amino acids and dipeptides. The detection limits of branched amino acids (signal-to-noise ratio of 3) were 1.5 nmol·L-1 in the standard solution (100 amol per injection).展开更多

The experimental conditions for the microwave hydrolysis and derivatization were examined and optimized. Several parameters of capillary electrophoresis, such as pH value of background electrolyte, borate concentration and applied voltage were optimized Under the selected conditions, 11 amino acids were completely separated. The real sample was analyzed and the results were satisfactory. Compared with that of conventional heat hydrolysis and derivatization, the analytical time of this method was significantly shortened.展开更多
