Considering that IBD is a genetic and multifactorial disease, we screened for the distribution dynamism of IBD pathogenic genetic variants (single nucleotide polymorphisms;SNPs) and risk factors in four (4) IBD pediatric patients, by integrating both clinical exome sequencing and computational statistical approaches, aiming to categorize IBD patients in CD and UC phenotype. To this end, we first aligned genomic read sequences of these IBD patients to hg19 human genome by using bowtie 2 package. Next, we performed genetic variant calling analysis in terms of single nucleotide polymorphism (SNP) for genes covered by at least 20 read genomic sequences. Finally, we checked for biological and genomic functions of genes exhibiting statistically significant genetic variant (SNPs) by introducing Fitcon genomic parameter. Findings showed Fitcon parameter as normalizing IBD patient’s population variability, as well as inducing a relative good clustering between IBD patients in terms of CD and UC phenotypes. Genomic analysis revealed a random distribution of risk factors and as well pathogenic SNPs genetic variants in the four IBD patient’s genome, claiming to be involved in: i) Metabolic disorders, ii) Autoimmune deficiencies;iii) Crohn’s disease pathways. Integration of genomic and computational statistical analysis supported a relative genetic variability regarding IBD patient population by processing IBD pathogenic SNP genetic variants as opposite to IBD risk factor variants. Interestingly, findings clearly allowed categorizing IBD patients in CD and UC phenotypes by applying Fitcon parameter in selecting IBD pathogenic genetic variants. Considering as a whole, the study suggested the efficiency of integrating clinical exome sequencing and computational statistical tools as a right approach in discriminating IBD phenotypes as well as improving inflammatory bowel disease (IBD) molecular diagnostic process.展开更多

The virus infects and constitutes irreversible impairment to the nervous system, damages the respiratory system, and marks severe gastrointestinal lesions leading to heavy mortality in short-living birds and substantial losses in layers and breeders. The continuous emergence and evolution of the virus made it inclined to evade the humoral response and indirectly the circumvention of artificial active immunization. Newcastle disease is caused by the orthoavula genus of the paramyxoviridae family and has shown high genetic diversity even in their genotypes while information regarding enzootic trends of the virus is scanty in Pakistan. A total of 40 tracheal samples of NDV were collected from different commercial broiler farms and 11 isolates of NDV were identified. In the current study, we determined the genetic diversity of the Newcastle disease virus based on the partial sequencing of the fusion protein gene available in the NCBI database. Genetic analysis showed that seven isolates belonged to class I genotype VII and four belonged to class II genotype II. Interestingly, two isolates had epidemiological connections with vaccine-like class II genotype II. Our findings, concerning the recent outbreaks of class I genotype VII and class II genotype II of NDV in vaccinated commercial flocks, suggest possible potential partial mutations in the fusion protein gene. Genetic diversity and formation of the new cleavage site in an important neutralizing protein of wild strain are linked with the potency of artificial active immunization and a major cause of vaccine failure.展开更多

文摘[Objectives]The paper was to investigate the relationship between the well-cellar early transplanting of robust seedlings and the prevalence of diseases in the Shiyan tobacco-growing area.[Methods]The relationship between disease occurrence and meteorological factors during the field growth period was examined by analyzing the prevalence of flue-cured tobacco virus diseases, brown spot, and total disease in the Shiyan tobacco-growing area before(2013-2017)and after(2018-2022)the well-cellar early transplanting of robust tobacco seedlings.[Results]The implementation of a well-cellar early transplanting technique of robust seedlings resulted in a reduction in the average incidence of tobacco virus disease, brown spot, and total disease by 0.83%,8.85%,and 7.91%,respectively,in comparison to the incidence observed prior to early transplanting. These findings suggest that early transplanting can significantly reduce the incidence of flue-cured tobacco diseases.Prior to the well-cellar early transplanting of robust tobacco seedlings, there was a significant(including highly significant)positive correlation between the incidence of brown spot and total disease and precipitation in August and September. The incidence of brown spot and total disease in tobacco plants was found to be significantly positively correlated with May precipitation and significantly negatively correlated with May sunshine hours following the well-cellar early transplanting of robust seedlings. The advancement of the transplanting period by 20 d resulted in a reduction in the growing period of tobacco plants in the field under autumn rains(late August to November)in western China. This effectively circumvented the suitable conditions for disease occurrence and can reduce the incidence of disease.[Conclusions]This study offers a framework for enhancing the quality and efficiency of flue-cured tobacco production in the northwest tobacco-growing area of Hubei.

文摘This paper analyzes the causes of the offshore strength enhancement of Typhoon Tiange (No. 1713) from four aspects: high and low layer divergence, vertical wind shear, southwest monsoon transport, offshore sea temperature and weak cold air. The conclusions are as follows: The high-level airflow divergence enhancement and positive vorticity advection in the offshore, strengthen the middle rise movement of the “Tiange”, and the ascending movement enhances the strength of the “Tiange” by strengthening the lower layer convergence; The high offshore sea temperature and sea temperature are changing temperature, which also has a certain effect on the strengthening of “Tiange” in the offshore; The strengthening of the southwest warm and humid airflow provides sufficient water vapor and energy for the development of “Tiange”, which is beneficial to the maintenance of the typhoon warm heart structure; The intrusion of weak cold air, enhanced the radiation cooperation of the lower typhoon disturbance, which is beneficial to the enhancement of “Tiange”.

文摘Based on theWorld Health Organization(WHO),Meningitis is a severe infection of the meninges, the membranes covering the brain and spinal cord.It is a devastating disease and remains a significant public health challenge. This study investigates a bacterial meningitis model through deterministic and stochastic versions.Four-compartment population dynamics explain the concept, particularly the susceptible population, carrier, infected, and recovered. The model predicts the nonnegative equilibrium points and reproduction number,i.e.,the Meningitis-Free Equilibrium(MFE),and Meningitis-Existing Equilibrium(MEE).For the stochastic version of the existing deterministicmodel, the twomethodologies studied are transition probabilities and non-parametric perturbations. Also, positivity, boundedness, extinction, and disease persistence are studiedrigorouslywiththe helpofwell-known theorems.Standard and nonstandard techniques such as EulerMaruyama, stochastic Euler, stochastic Runge Kutta, and stochastic nonstandard finite difference in the sense of delay have been presented for computational analysis of the stochastic model.Unfortunately, standard methods fail to restore the biological properties of the model,so the stochastic nonstandard finite difference approximation is offered as an efficient,low-cost,and independent of time step size.In addition, the convergence, local, and global stability around the equilibria of the nonstandard computational method is studied by assuming the perturbation effect is zero. The simulations and comparison of the methods are presented to support the theoretical results and for the best visualization of results.

文摘Mature oligodendrocytes form myelin sheaths that are crucial for the insulation of axons and efficient signal transmission in the central nervous system.Recent evidence has challenged the classical view of the functionally static mature oligodendrocyte and revealed a gamut of dynamic functions such as the ability to modulate neuronal circuitry and provide metabolic support to axons.Despite the recognition of potential heterogeneity in mature oligodendrocyte function,a comprehensive summary of mature oligodendrocyte diversity is lacking. We delve into early 20th-century studies by Robertson and Río-Hortega that laid the foundation for the modern identification of regional and morphological heterogeneity in mature oligodendrocytes.Indeed, recent morphologic and functional studies call into question the long-assumed homogeneity of mature oligodendrocyte function through the identification of distinct subtypes with varying myelination preferences.Furthermore, modern molecular investigations, employing techniques such as single cell/nucleus RNA sequencing, consistently unveil at least six mature oligodendrocyte subpopulations in the human central nervous system that are highly transcriptomically diverse and vary with central nervous system region.Age and disease related mature oligodendrocyte variation denotes the impact of pathological conditions such as multiple sclerosis,Alzheimer's disease, and psychiatric disorders.Nevertheless, caution is warranted when subclassifying mature oligodendrocytes because of the simplification needed to make conclusions about cell identity from temporally confined investigations.Future studies leveraging advanced techniques like spatial transcriptomics and single-cell proteomics promise a more nuanced understanding of mature oligodendrocyte heterogeneity.Such research avenues that precisely evaluate mature oligodendrocyte heterogeneity with care to understand the mitigating influence of species, sex, central nervous system region, age, and disease, hold promise for the development of therapeutic interventions targeting varied central nervous system pathology.

文摘Background:Diabetic kidney disease(DKD)is a microvascular complication of diabetes mellitus and is the main cause of end-stage renal failure.Suoquan pills(SQP)has a variety of pharmacological activities and multiple therapeutic effects, and it is used clinically as a basic formula for the treatment of DKD.Methods:Public databases were used to identify SQP compounds and the potential targets of SQP and DKD.A drug-component-therapeutic target network was constructed.Protein-protein interaction network analysis,Gene Ontology functional analysis, and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis were used to analyse the potential molecular mechanisms of SQP based on common targets of drugs and diseases.Molecular docking simulations were conducted to confirm the binding abity of the core compounds to key targets. The efficacy and predicted molecular mechanisms of SQP were validated using cell counting kit-8 assay, flow cytometry, and western blotting with HK-2 cells as a model. Results:Network pharmacology analysis showed that 26 compounds and 207 potential targets of SQP were involved in the treatment of DKD;boldine, denudatin B,pinocembrin, kaempferoid, and quercetin were considered core compounds, and epidermal growth factor receptor(EGFR)and proto-oncogene,non-receptor tyrosine kinase(SRC)were considered key targets.Gene Ontology enrichment analysis indicated that protein phosphorylation and negative regulation of apoptotic processes are important biological processes in the treatment of DKD by SQP.Molecular docking confirmed the excellent binding abilities of boldine, denudatin B,kaempferide, and quercetin to EGFR and SRC.The results of in vitro experiments showed that treatment with an ethanolic extract of SQP significantly protected HK-2 cells from high glucose-induced cell damage. In addition, the SQP ethanol extract inhibited the phosphorylation of EGFR and SRC,suppressed the apoptosis rate, and regulated apoptosis-related proteins in HK-2 cells under high glucose stress. Conclusion:This study systematically and intuitively illustrated the possible pharmacological mechanisms of SQP against DKD through multiple components, targets, and signalling pathways, especially the inhibition of EGFR and SRC phosphorylation and apoptosis.
文摘Inflammatory bowel diseases (IBD) are complex multifactorial disorders that include Crohn’s disease (CD) and ulcerative colitis (UC). Considering as a whole, the study suggested the efficiency of integrating clinical exome sequencing and computational statistical tools as a right approach in discriminating IBD phenotypes as well as improving inflammatory bowel disease (IBD) molecular diagnostic process.
文摘Newcastle disease (ND) virus is a leading threat to commercial and domestic poultry in Pakistan. Genetic diversity and formation of the new cleavage site in an important neutralizing protein of wild strain are linked with the potency of artificial active immunization and a major cause of vaccine failure.
文摘Recent medical literature shows that the application of artificial intelligence(AI)models in gastrointestinal pathology is an exponentially growing field, with pro-mising models that show very high performances.Regarding inflammatory bowel disease(IBD),recent reviews demonstrate promising diagnostic and prognostic AI models. However, studies are generally at high risk of bias(especially in AI models that are image-based).The creation of specific AI models that improve diagnostic performance and allow the establishment of a general prognostic fo-recast in IBD is of great interest,as it may allow the stratification of patients into subgroups and,in turn, allow the creation of different diagnostic and therapeutic protocols for these patients.Regarding surgical models, predictive models of post-operative complications have shown great potential in large-scale studies. In this work, the authors present the development of a predictive algorithm for early post-surgical complications in Crohn's disease based on a Random Forest model with exceptional predictive ability for complications within the cohort. The pre-sent work, based on logical and reasoned, clinical, and applicable aspects, lays a solid foundation for future prospective work to further develop post-surgical prognostic tools for IBD.The next step is to develop in a prospective and mul-ticenter way,a collaborative path to optimize this line of research and make it applicable to our patients.
