Low temperature is the most common abiotic stress factors during the eggplant cultivation in solar greenhouses.Melatonin plays important roles in plant resistance to low temperature.However,the role of melatonin in re...Low temperature is the most common abiotic stress factors during the eggplant cultivation in solar greenhouses.Melatonin plays important roles in plant resistance to low temperature.However,the role of melatonin in regulating chilling tolerance and extending the preharvest shelf life of eggplant fruits is still unknown.In this study,we investigated the effects of exogenous melatonin on eggplant plants and fruits in response to low temperature.Under simulated low-temperature conditions,exogenous melatonin significantly relieved the chilling symptoms of seedlings by reducing reactive oxygen species (ROS) and malondialdehyde (MDA) levels and relative leakage rates.These reductions were caused by higher superoxide dismutase (SOD) and catalase (CAT) activities and increased endogenous polyamine and melatonin levels compared with those in untreated seedlings.Notably,the expression levels of SOD,CAT1/2,and polyamine synthesis genes (ADC and ODC) were also increased by 100μmol·L~(-1)melatonin,as well as those of genes involved in melatonin synthesis (TDC,T5H,SNAT,ASMT,and COMT) and cold regulation (COR1,CBFa/b,and ZAT2/6/12).To further investigate the effects of melatonin on eggplant leaves and fruits under natural low temperature conditions,100μmol·L~(-1)melatonin was sprayed on the functional leaves at three days before commodity maturation.Melatonin significantly alleviated chilling injury in the leaves and pericarp and extended the preharvest shelf life of the fruit by increasing the expression of COR1,CBF,ZAT2/6/12,and API5 and decreasing the expression of senescence-related genes (NCED1/2 and SAG12).Therefore,100μmol·L~(-1)melatonin improved chilling tolerance and fruit shelf life by upregulating ZAT2/6/12 to affect ROS-and senescence-related processes,which provides a reference for alleviating cold stress and extending the preharvest fruit shelf life in eggplant.展开更多

Here, anti-H3K2...Histone H3 lysine 27 trimethylation(H3K27me3) is a histone modification associated with transcriptional repression. However, insights into the genome-wide pattern of H3K27me3 in grapevines are limited. Here, anti-H3K27 chromatin immunoprecipitation(ChIP), high-throughput sequencing, and transcriptome analysis were performed using leaves of Vitis amurensis. The leaves were treated at 4°C for 2 h and 24 h and used to investigate changes in H3K27me3 under chilling treatment. The results show that H3K27me3 is well-distributed both in gene regions(-50%) and in the intergenic region(-50%) in the grapevine genome(Vitis vinifera ‘Pinot Noir PN40024'). H3K27me3 was found to be localized in8 368 annotated gene regions in all detected samples(leaves at normal temperature and under chilling treatments) and mainly enriched in gene bodies with the adjacent promoter and downstream areas. The short-term chilling treatments(4°C for 2 h) induced 2 793 gains and 305losses in H3K27me3 modification. Subsequently, 97.3% of the alterations were restored to original levels after 24 h treatment. The ChIP-qPCR for five differential peaks showed similar results to the data for ChIP-seq, indicating that the chilling-induced H3K27me3 modification is reliable.Integrative analysis of transcriptome and ChIP-seq results showed that the expression of H3K27me3 target genes was significantly lower than those of non-target genes, indicating transcriptional repression of H3K27me3 in grapevine leaves. Furthermore, histone methylation alterations were detected in 82 genes and were related to either repression or activation of their expression during chilling stress. The findings provide the genome-wide H3K27me3 patterns in grapevines and shed light on uncovering its regulation in chilling stress responses.展开更多
Chilling-induced accumulation of reactive oxygen species(ROS) is harmful to plants,which usually produce anthocyanins to scavenge ROS as protection from chilling stress.As a tropical crop,cassava is hypersensitive to ...Chilling-induced accumulation of reactive oxygen species(ROS) is harmful to plants,which usually produce anthocyanins to scavenge ROS as protection from chilling stress.As a tropical crop,cassava is hypersensitive to chilling,but the biochemical basis of this hypersensitivity remains unclear.We previously generated Me MYB2-RNAi transgenic cassava with increased chilling tolerance.Here we report that Me MYB2-RNAi transgenic cassava accumulated less ROS but more cyanidin-3-O-glucoside than the wild type under early chilling stress.Under this stress,the anthocyanin biosynthesis pathway was more active in Me MYB2-RNAi lines than in the wild type,and several genes involved in the pathway,including Me TT8,were up-regulated by Me MYB2-RNAi in the transgenic cassava.Me MYB2 bound to the Me TT8 promoter and blocked its expression under both normal and chilling conditions,thereby inhibiting anthocyanin accumulation.Me TT8 was shown to bind to the promoter of Dihydroflavonol 4-reductase(Me DFR-2)and increased Me DFR-2 expression.Me MYB2 appears to act as an inhibitor of chilling-induced anthocyanin accumulation during the rapid response of cassava to chilling stress.展开更多
Mangoes often suffer from low temperature-induced chilling injury (CI) during postharvest cold storage.Therefore,advanced techniques are crucial and in high demand to solve the chilling stress of mango fruit for a hig...Mangoes often suffer from low temperature-induced chilling injury (CI) during postharvest cold storage.Therefore,advanced techniques are crucial and in high demand to solve the chilling stress of mango fruit for a higher value.This study addresses chilling stress modulation by investigating the effects of melatonin treatment on CI,proline metabolism,and related gene expressions of ‘Keitt’ mango during cold storage after dipped in 0 (control),0.1 (MT1),and 0.2 mmol L^(-1)(MT2) melatonin solution for 30 min.The results revealed that melatonin treatment in MT1 significantly reduced CI development and increased proline content in mango fruit during cold storage compared to the control.These changes were along with increases in the activity of critical enzymes as well as the expression of encoding genes involved in proline biosynthesis,such as pyrroline-5-carboxylate synthetase (P5CS),pyrroline-5-carboxylate reductase (P5CR),ornithine D-aminotransferase (OAT),P5CS2,P5CR2,and OAT3.Additionally,proline dehydrogenase (PDH) activity and the expression of the PDH3 gene associated with proline dehydrogenation were lower in MT1-treated mangoes than the controlled group.Thus,melatonin treatment has regulated proline metabolism resulting in the accumulation of proline,subsequently contributing to enhancing the chilling tolerance of‘Keitt’mango fruit.展开更多

Changes in photosynthesis and antioxidant enzymes in chilling acclim...This study investigates the effects of cold-acclimation in conferring chilling tolerance in seedlings of the mulberry(Morus alba) variety ‘Qiuyu’. Changes in photosynthesis and antioxidant enzymes in chilling acclimatized(CA), and non-acclimatized(NA) seedlings were recorded during chilling stress(3 °C) and a recovery period(25 °C) each for 3 days. The results showed that CA plants had higher net photosynthetic rates(P_n), stomatal conductance(G_s), and maximum photochemical efficiency of photosystem Ⅱ(F_v/F_m) in response to chilling stress compared to NA. The seedlings maintained the same trends during the recovery stage. The responses of Q_A reduction degree (1-q_P) and prime electronic transfer rates(F_o) were lower in acclimatized than in non-acclimatized seedlings. Low-temperature acclimation and chilling stress also caused an increase in leaf proline and soluble sugar contents. Leaf malondialdehyde levels were significantly lower while ascorbate peroxidase(APX) activity was significantly higher in acclimatized seedlings, suggesting that elevated osmolytes and APX confer resistance to chilling temperatures. In this study on the response of mulberry seedlings to chilling stress, we also looked at the recovery process. The response to chilling determines whether mulberry leaves can survive under cold temperatures, while the recovery process determines whether photosynthesis can recover as soon as possible to avoid any secondary damage.展开更多

Hongdeng) from a warm winter climate(Shanghai) and cold winter climate(Tai'an, Shandong Province, China) was conducted to explore the reason of low fruit set. The effect of hydrogen cyanamide(HCN) on floral development under warm winter conditions was also investigated. Trees grown in Shanghai with insufficient chilling accumulation exhibited little difference in the progression of microspore development compared to trees in Tai'an that accumulated adequate chilling, but showed substantial delays in ovule and embryo sac development. The growth of nucelli did not proceed beyond the macrospore mother cell and macrospore stages with abortion rates of 13, 15 and 45% by 6, 3 and 0 d before full bloom, respectively. These abnormalities in the ovule and embryo sac in the Shanghai-grown trees were eliminated by HCN application. These results suggest that chilling regulates the development of female floral organs in winter dormancy; therefore, insufficient chilling accumulation, causing abnormality of the female floral organs, restricts the cultivation of sweet cherry in warm winter regions. Interestingly, HCN application, which decreased the chilling requirements for Hongdeng, may be a potential strategy for sweet cherry cultivation in warm winter regions.展开更多
The mechanism of melatonin(MT)induced chilling tolerance in harvested cucumber fruit was investigated at commercial maturity.In this study,cucumber fruits were treated with 100μmol L^(-1) MT at 4℃ and 90% relative h...The mechanism of melatonin(MT)induced chilling tolerance in harvested cucumber fruit was investigated at commercial maturity.In this study,cucumber fruits were treated with 100μmol L^(-1) MT at 4℃ and 90% relative humidity for 15 d of storage.In comparison with the control,cucumber treatment with MT resulted in reduced chilling injury(CI),decreased electrolyte leakage and enhanced firmness.The fruits treated with MT showed higher chlorophyll contents in storage conditions with suppressed chlorophyllase enzyme activity.MT treatment increased arginine decarboxylase(ADC)and ornithine decarboxylase(ODC)enzyme activities.Moreover,enhanced expression of the Cucumis sativus ADC(CsADC)and C.sativus ODC(CsODC)genes resulted in the accumulation of polyamine contents.Similarly,proline levels exhibited higher levels among treated fruits.Meanwhile,the proline synthesizing enzymes △1-pyrroline-5-carboxylate syntheses(P5CS)and ornithine aminotransferase(OAT)were significantly increased,while a catabolic enzyme of proline dehydrogenase(PDH)activity was inhibited by treatment.In addition,MT induced expression of C.sativus OAT(CsOAT)and C.sativus P5CS(CsP5CS)genes.Cucumber fruits treated with MT also exhibited higher γ-aminobutyric acid(GABA)content by enhanced GABA transaminase(GABA-T)and glutamate decarboxylase(GAD)enzyme activities and a higher C.sativus GAD(CsGAD)gene expression.To sum up,the results show that MT treatment enhanced chilling tolerance,which was associated with the regulation of polyamines,as well as proline and γ-aminobutyric acid.展开更多

Low temperature severely limits its productivity and geographical distribution in temperate regions of the world. In this study, we investigated the effect of exogenous abscisic Acid(ABA) on chilling tolerance of napier grass(Pennisetum purpureum Schum.) seedlings. Seven-day-old napier grass seedlings were cultured in dd H_2O or ABA solution at different concentrations and exposed to 1 ℃ for different time durations. The chilling injury, membrane stability index(MSI) and proline content were estimated from leaf samples. The results showed that there was obvious morphological injury of leaf blighting and restrained growth for the seedlings under chilling stress, but this damage can be largely reduced(by 2/3) when the seedlings were treated by 100 μmol/L ABA in the culture solution, and that the application of exogenous ABA can help to maintain a good stability of leaf cell membrane as expressed by a high MSI value and a low level of proline in leaf cells. These results suggested that exogenous ABA can significantly alleviate chilling injury in napier grass seedlings by maintaining the stability of leaf cell membrane during chilling stress, and that the chilling tolerance was not ensured by a proline accumulation although a passive accumulation of proline was observed in the seedlings under chilling stress. Our results lay a preliminary foundation for future investigations on the molecular mechanisms of ABA induced chilling or freezing tolerance in napier grass.展开更多
The Dormancy-associated MADS-box(DAM)gene cluster in peach serves as a key regulatory hub on which the seasonal temperatures act and orchestrate dormancy onset and exit,chilling response and floral bud developmental p...The Dormancy-associated MADS-box(DAM)gene cluster in peach serves as a key regulatory hub on which the seasonal temperatures act and orchestrate dormancy onset and exit,chilling response and floral bud developmental pace.Yet,how different temperature regimes interact with and regulate the six linked DAM genes remains unclear.Here,we demonstrate that chilling downregulates DAM1 and DAM3–6 in dormant floral buds with distinct patterns and identify DAM4 as the most abundantly expressed one.We reveal multiple epigenetic events,with tri-methyl histone H3 lysine 27(H3K27me3)induced by chilling specifically in DAM1 and DAM5,a 21-nt sRNA in DAM3 and a ncRNA induced in DAM4.Such induction is inversely correlated with downregulation of their cognate DAMs.We also show that the six DAMs were hypermethylated,associating with the production of 24-nt sRNAs.Hence,the chilling-responsive dynamic of the different epigenetic elements and their interactions likely define distinct expression abundance and downregulation pattern of each DAM.We further show that the expression of the five DAMs remains steadily unchanged or continuously downregulated at the ensuing warm temperature after chilling,and this state of regulation correlates with robust increase of sRNA expression,H3K27me3 and CHH methylation,which is particularly pronounced in DAM4.Such robust increase of repressive epigenetic marks may irreversibly reinforce the chillingimposed repression of DAMs to ensure flower-developmental programming free from any residual DAM inhibition.Taken together,we reveal novel information about genetic and epigenetic regulation of the DAM cluster in peach,which will be of fundamental significance in understanding of the regulatory mechanisms underlying chilling requirement and dormancy release,and of practical application for improvement of plasticity of flower time and bud break in fruit trees to adapt changing climates.展开更多
Syringa species not only have good ornamental properties but also play an important role in the landscaping and environmental purification of cities.To investigate the chilling stress resistance of Syringa oblata Lind...Syringa species not only have good ornamental properties but also play an important role in the landscaping and environmental purification of cities.To investigate the chilling stress resistance of Syringa oblata Lindl.and Syringa reticulata var.mandshurica and provide theoretical grounds for the practical cultivation of Syringa species,in vitro leaves were used to study photosynthetic gas exchange parameters and chlorophyll fluorescence parameters.After nine hours of chilling,decreasing rates of net photosynthesis,stomatal conductance,and transpiration in S.reticulata var.mandshurica leaves were significantly greater than that of the S.oblata,while intercellular CO2 concentrations in S.oblata leaves were higher than those in S.reticulata var.mandshurica.The quantum yield of PSII reaction center(APSII)declined in S.reticulata and light capture efficiency(Fv 0/Fm 0)was stable.However,reduction percentages of Fv 0/Fm 0,APSII,and Fv/Fm in S.oblata were significant higher than those of S.reticulata var.mandshurica.After nine hours of chilling,the relative variable fluorescence of VJ and VI of S.oblata increased and the increasing rate of VJ was greater than VI.In contrast,the change of VJ and VI in S.reticulata var.mandshurica leaves was relatively small.This suggests that chilling primarily damaged the electron transport process of QA to QB at the receptor site of the PSII reaction center.Photosynthetic capacity of S.oblata was more sensitive to chilling stress compared to S.reticulate var.mandshurica,which the limitations were mainly due to non-stomatal factors such as the decrease in electron transport efficiency,activity in the PSII reaction center,and the destruction of the photodamage defense system.展开更多

Photosynthesis and osmoregulators were analyzed during chilling stress to explore the adaptation mechanisms that underlie the induction of chilling tolerance in response to suboptimal temperature. The relationships of these processes to suboptimal temperature acclimation in watermelon were then determined.Suboptimal temperature-acclimated watermelon plants demonstrated tolerance during chilling stress, as indicated by the decreased electrolyte leakage and malondialdehyde accumulation compared with those non-acclimated watermelon plants. Chilling-induced photoinhibition and reduction in CO2 assimilation rate were alleviated after suboptimal temperature acclimation. The xanthophyll cycle level was enhanced by improving thermal dissipation ability and avoiding light damage. Consequently, the chilling tolerance of suboptimal temperature-acclimated watermelon plants was enhanced. The osmoregulation ability induced by suboptimal temperature acclimation protected watermelon plants against chilling injury because of the accumulation of small molecular substances, such as soluble sugar and proline. The protein levels of Rubisco activase(ClRCA) and the gene expression of the Benson–Calvin cycle simultaneously increased in suboptimal temperature-acclimated watermelon plants during chilling stress. Chilling tolerance in watermelon plants induced by suboptimal temperature acclimation is associated with enhanced photosynthetic adaptability and osmoregulation ability.展开更多
The Mg-3.0Nd-0.2Zn-0.4Zr (NZ30K) alloys were prepared by direct-chill casting (DCC) and sand mould casting (SMC) processes,respectively and their microstructures and mechanical properties were investigated.The results...The Mg-3.0Nd-0.2Zn-0.4Zr (NZ30K) alloys were prepared by direct-chill casting (DCC) and sand mould casting (SMC) processes,respectively and their microstructures and mechanical properties were investigated.The results indicate that casting method plays a remarkable influence on the microstructure and mechanical properties of as-cast NZ30K alloy.The grain size increases from 35-40μm in the billets made by the DCC to about 100-120μm in the billets by the SMC.The aggregation of Mg12Nd usually found at the triple joints of grain boundaries in the billets prepared by SMC while is not observable from the billets by DCC.The tensile strengths and elongations of the billets are 195.2 MPa and 15.5% by DCC,and 162.5 MPa and 3.2% by SMC,respectively.The tensile strength of the alloy by DCC is remarkably enhanced by T6 heat treatment,which reached 308.5 MPa.Fracture surfaces of NZ30K alloy have been characterized as intergranular fracture by SMC and quasi-cleavage fracture by DCC,respectively.展开更多
Peach(Prunus persica Batsch‘Yuhualu’)fruit are sensitive to chilling injury(CI).Proline,polyamine(PA),and nitric oxide(NO)are important small regulators of various metabolic pathways under chilling stress that mitig...Peach(Prunus persica Batsch‘Yuhualu’)fruit are sensitive to chilling injury(CI).Proline,polyamine(PA),and nitric oxide(NO)are important small regulators of various metabolic pathways under chilling stress that mitigate CI.Ethylene is known to promote senescence and CI,while 1-methylcyclopropene(1-MCP)is an antagonist that inhibits the effects of ethylene.However,how1-MCP and ethylene affect proline,PA,and NO levels under chilling stress remains unclear.To address these questions,1-MCP(1μL·L^(−1))and ethylene(1μL·L^(−1))treatments were applied to peach fruit.Fruit were stored at 4°C for 28 d,then moved to 25°C for 3 d immediately after cold storage.Peach fruit exhibited CI symptoms after 7 d of cold storage with enhanced electrolyte leakage and malondialdehyde contents.The 1-MCP treatment significantly(P<0.05)restrained peach CI,and fruit did not exhibit CI symptoms until 14 d of cold storage.Proline and PAs in peach under chilling stress weremostly synthesized from glutamate and arginine,which were catalyzed by1-pyrroline-5-carboxylate synthetase and arginine decarboxylase,respectively.1-MCPtreated fruit exhibited higher proline and PA contents and enhanced chilling tolerance compared to the control,while ethylene-treated fruit had lower proline and PA contents and reduced chilling tolerance.Ethylene-treated fruit,which exhibited more severe CI symptoms compared to the control,had significantly(P<0.05)lower NO contents and NO synthase activities.However,NOmay not be a direct acting factor in 1-MCPinduced chilling tolerance,as 1-MCP-treated fruit had lower NO contents and NO synthase activities compared to the control.In conclusion,proline and PA clearly played direct and important roles in 1-MCP-induced peach chilling tolerance,while NO may not be actively involved.展开更多
Low-temperature storage is extensively used to optimize the postharvest life of various fresh fruits.However,red pitahaya(Hylocereus polyrhizus)fruits are sensitive to chilling injury(CI),which leads to the limitation...Low-temperature storage is extensively used to optimize the postharvest life of various fresh fruits.However,red pitahaya(Hylocereus polyrhizus)fruits are sensitive to chilling injury(CI),which leads to the limitation of low-temperature storage.In this study,red pitahaya fruits were stored at 2,4,6,8,and 10℃,respectively,for 27 days to determine the appropriate storage temperature.During the storage of red pitahaya fruits,storage at 8℃ was more effective in suppressing decay and maintaining quality than other low temperatures.Lowtemperature(2,4,and 6℃)storage decreased weight loss(WL)and maintained higher content of titratable acidity(TA),soluble sugars(SS),and total phenolics(TP)but different degrees of CI were detected.No CI was observed at 8℃ and 10℃.Red pitahay as stored at 8 and 10℃ were associated with better color evaluation,lower electrolyte leakage(EL),respiration rate,and lipoxygenase(LOX)activity,and higher fruit firmness,superoxide dismutase(SOD)activity,and catalase(CAT)activity.However,higher storage temperature(10℃)resulted in higher metabolic activity leading to lower quality and antioxidant capacities compared with 8℃.Therefore,our results demonstrated that red pitahaya stored at 8℃ exhibited a protective effect on fruit quality and resisted CI development during storage.展开更多

文摘Low temperature is the most common abiotic stress factors during the eggplant cultivation in solar greenhouses.Melatonin plays important roles in plant resistance to low temperature.However,the role of melatonin in regulating chilling tolerance and extending the preharvest shelf life of eggplant fruits is still unknown.In this study,we investigated the effects of exogenous melatonin on eggplant plants and fruits in response to low temperature.Under simulated low-temperature conditions,exogenous melatonin significantly relieved the chilling symptoms of seedlings by reducing reactive oxygen species (ROS) and malondialdehyde (MDA) levels and relative leakage rates.These reductions were caused by higher superoxide dismutase (SOD) and catalase (CAT) activities and increased endogenous polyamine and melatonin levels compared with those in untreated seedlings.Notably,the expression levels of SOD,CAT1/2,and polyamine synthesis genes (ADC and ODC) were also increased by 100μmol·L~(-1)melatonin,as well as those of genes involved in melatonin synthesis (TDC,T5H,SNAT,ASMT,and COMT) and cold regulation (COR1,CBFa/b,and ZAT2/6/12).To further investigate the effects of melatonin on eggplant leaves and fruits under natural low temperature conditions,100μmol·L~(-1)melatonin was sprayed on the functional leaves at three days before commodity maturation.Melatonin significantly alleviated chilling injury in the leaves and pericarp and extended the preharvest shelf life of the fruit by increasing the expression of COR1,CBF,ZAT2/6/12,and API5 and decreasing the expression of senescence-related genes (NCED1/2 and SAG12).Therefore,100μmol·L~(-1)melatonin improved chilling tolerance and fruit shelf life by upregulating ZAT2/6/12 to affect ROS-and senescence-related processes,which provides a reference for alleviating cold stress and extending the preharvest fruit shelf life in eggplant.

文摘Histone H3 lysine 27 trimethylation(H3K27me3) is a histone modification associated with transcriptional repression. However, insights into the genome-wide pattern of H3K27me3 in grapevines are limited. Here, anti-H3K27 chromatin immunoprecipitation(ChIP), high-throughput sequencing, and transcriptome analysis were performed using leaves of Vitis amurensis. The leaves were treated at 4°C for 2 h and 24 h and used to investigate changes in H3K27me3 under chilling treatment. The results show that H3K27me3 is well-distributed both in gene regions(-50%) and in the intergenic region(-50%) in the grapevine genome(Vitis vinifera ‘Pinot Noir PN40024'). H3K27me3 was found to be localized in8 368 annotated gene regions in all detected samples(leaves at normal temperature and under chilling treatments) and mainly enriched in gene bodies with the adjacent promoter and downstream areas. The short-term chilling treatments(4°C for 2 h) induced 2 793 gains and 305losses in H3K27me3 modification. Subsequently, 97.3% of the alterations were restored to original levels after 24 h treatment. The ChIP-qPCR for five differential peaks showed similar results to the data for ChIP-seq, indicating that the chilling-induced H3K27me3 modification is reliable.Integrative analysis of transcriptome and ChIP-seq results showed that the expression of H3K27me3 target genes was significantly lower than those of non-target genes, indicating transcriptional repression of H3K27me3 in grapevine leaves. Furthermore, histone methylation alterations were detected in 82 genes and were related to either repression or activation of their expression during chilling stress. The findings provide the genome-wide H3K27me3 patterns in grapevines and shed light on uncovering its regulation in chilling stress responses.

文摘Chilling-induced accumulation of reactive oxygen species(ROS) is harmful to plants,which usually produce anthocyanins to scavenge ROS as protection from chilling stress.As a tropical crop,cassava is hypersensitive to chilling,but the biochemical basis of this hypersensitivity remains unclear.We previously generated Me MYB2-RNAi transgenic cassava with increased chilling tolerance.Here we report that Me MYB2-RNAi transgenic cassava accumulated less ROS but more cyanidin-3-O-glucoside than the wild type under early chilling stress.Under this stress,the anthocyanin biosynthesis pathway was more active in Me MYB2-RNAi lines than in the wild type,and several genes involved in the pathway,including Me TT8,were up-regulated by Me MYB2-RNAi in the transgenic cassava.Me MYB2 bound to the Me TT8 promoter and blocked its expression under both normal and chilling conditions,thereby inhibiting anthocyanin accumulation.Me TT8 was shown to bind to the promoter of Dihydroflavonol 4-reductase(Me DFR-2)and increased Me DFR-2 expression.Me MYB2 appears to act as an inhibitor of chilling-induced anthocyanin accumulation during the rapid response of cassava to chilling stress.

文摘Mangoes often suffer from low temperature-induced chilling injury (CI) during postharvest cold storage.Therefore,advanced techniques are crucial and in high demand to solve the chilling stress of mango fruit for a higher value.This study addresses chilling stress modulation by investigating the effects of melatonin treatment on CI,proline metabolism,and related gene expressions of ‘Keitt’ mango during cold storage after dipped in 0 (control),0.1 (MT1),and 0.2 mmol L^(-1)(MT2) melatonin solution for 30 min.The results revealed that melatonin treatment in MT1 significantly reduced CI development and increased proline content in mango fruit during cold storage compared to the control.These changes were along with increases in the activity of critical enzymes as well as the expression of encoding genes involved in proline biosynthesis,such as pyrroline-5-carboxylate synthetase (P5CS),pyrroline-5-carboxylate reductase (P5CR),ornithine D-aminotransferase (OAT),P5CS2,P5CR2,and OAT3.Additionally,proline dehydrogenase (PDH) activity and the expression of the PDH3 gene associated with proline dehydrogenation were lower in MT1-treated mangoes than the controlled group.Thus,melatonin treatment has regulated proline metabolism resulting in the accumulation of proline,subsequently contributing to enhancing the chilling tolerance of‘Keitt’mango fruit.

文摘Inoculation can reduce the chill depth of a chilled iron,and therefore influence the microstructure and properties of the iron.In this paper,the effect of rare earth Lanthanun (La) on the microstructure and properties of a chilled iron camshaft was studied.The results show that the La addition efficiently enhances the mechanical properties,yet with the unfavorable effect of decreasing the chilled depth and hardness.Moreover,La promotes graphite concentration and results in large graphite size,as well as A-type graphite.It is also found that excessive La destroys the interconnection and directivity of ledeburite.According to the experimental results,the optimum adding content of La should be no more than 0.02wt.%.

文摘This study investigates the effects of cold-acclimation in conferring chilling tolerance in seedlings of the mulberry(Morus alba) variety ‘Qiuyu’. Changes in photosynthesis and antioxidant enzymes in chilling acclimatized(CA), and non-acclimatized(NA) seedlings were recorded during chilling stress(3 °C) and a recovery period(25 °C) each for 3 days. The results showed that CA plants had higher net photosynthetic rates(P_n), stomatal conductance(G_s), and maximum photochemical efficiency of photosystem Ⅱ(F_v/F_m) in response to chilling stress compared to NA. The seedlings maintained the same trends during the recovery stage. The responses of Q_A reduction degree (1-q_P) and prime electronic transfer rates(F_o) were lower in acclimatized than in non-acclimatized seedlings. Low-temperature acclimation and chilling stress also caused an increase in leaf proline and soluble sugar contents. Leaf malondialdehyde levels were significantly lower while ascorbate peroxidase(APX) activity was significantly higher in acclimatized seedlings, suggesting that elevated osmolytes and APX confer resistance to chilling temperatures. In this study on the response of mulberry seedlings to chilling stress, we also looked at the recovery process. The response to chilling determines whether mulberry leaves can survive under cold temperatures, while the recovery process determines whether photosynthesis can recover as soon as possible to avoid any secondary damage.

文摘The microscopic investigation of the floral development of sweet cherry(Prunus avium L. cv. Hongdeng) from a warm winter climate(Shanghai) and cold winter climate(Tai'an, Shandong Province, China) was conducted to explore the reason of low fruit set. The effect of hydrogen cyanamide(HCN) on floral development under warm winter conditions was also investigated. Trees grown in Shanghai with insufficient chilling accumulation exhibited little difference in the progression of microspore development compared to trees in Tai'an that accumulated adequate chilling, but showed substantial delays in ovule and embryo sac development. The growth of nucelli did not proceed beyond the macrospore mother cell and macrospore stages with abortion rates of 13, 15 and 45% by 6, 3 and 0 d before full bloom, respectively. These abnormalities in the ovule and embryo sac in the Shanghai-grown trees were eliminated by HCN application. These results suggest that chilling regulates the development of female floral organs in winter dormancy; therefore, insufficient chilling accumulation, causing abnormality of the female floral organs, restricts the cultivation of sweet cherry in warm winter regions. Interestingly, HCN application, which decreased the chilling requirements for Hongdeng, may be a potential strategy for sweet cherry cultivation in warm winter regions.

文摘The mechanism of melatonin(MT)induced chilling tolerance in harvested cucumber fruit was investigated at commercial maturity.In this study,cucumber fruits were treated with 100μmol L^(-1) MT at 4℃ and 90% relative humidity for 15 d of storage.In comparison with the control,cucumber treatment with MT resulted in reduced chilling injury(CI),decreased electrolyte leakage and enhanced firmness.The fruits treated with MT showed higher chlorophyll contents in storage conditions with suppressed chlorophyllase enzyme activity.MT treatment increased arginine decarboxylase(ADC)and ornithine decarboxylase(ODC)enzyme activities.Moreover,enhanced expression of the Cucumis sativus ADC(CsADC)and C.sativus ODC(CsODC)genes resulted in the accumulation of polyamine contents.Similarly,proline levels exhibited higher levels among treated fruits.Meanwhile,the proline synthesizing enzymes △1-pyrroline-5-carboxylate syntheses(P5CS)and ornithine aminotransferase(OAT)were significantly increased,while a catabolic enzyme of proline dehydrogenase(PDH)activity was inhibited by treatment.In addition,MT induced expression of C.sativus OAT(CsOAT)and C.sativus P5CS(CsP5CS)genes.Cucumber fruits treated with MT also exhibited higher γ-aminobutyric acid(GABA)content by enhanced GABA transaminase(GABA-T)and glutamate decarboxylase(GAD)enzyme activities and a higher C.sativus GAD(CsGAD)gene expression.To sum up,the results show that MT treatment enhanced chilling tolerance,which was associated with the regulation of polyamines,as well as proline and γ-aminobutyric acid.

文摘Napier grass, an important forage crop with potentials in multi-purpose applications, is widely grown throughout the tropics and subtropics. Low temperature severely limits its productivity and geographical distribution in temperate regions of the world. In this study, we investigated the effect of exogenous abscisic Acid(ABA) on chilling tolerance of napier grass(Pennisetum purpureum Schum.) seedlings. Seven-day-old napier grass seedlings were cultured in dd H_2O or ABA solution at different concentrations and exposed to 1 ℃ for different time durations. The chilling injury, membrane stability index(MSI) and proline content were estimated from leaf samples. The results showed that there was obvious morphological injury of leaf blighting and restrained growth for the seedlings under chilling stress, but this damage can be largely reduced(by 2/3) when the seedlings were treated by 100 μmol/L ABA in the culture solution, and that the application of exogenous ABA can help to maintain a good stability of leaf cell membrane as expressed by a high MSI value and a low level of proline in leaf cells. These results suggested that exogenous ABA can significantly alleviate chilling injury in napier grass seedlings by maintaining the stability of leaf cell membrane during chilling stress, and that the chilling tolerance was not ensured by a proline accumulation although a passive accumulation of proline was observed in the seedlings under chilling stress. Our results lay a preliminary foundation for future investigations on the molecular mechanisms of ABA induced chilling or freezing tolerance in napier grass.

文摘The Dormancy-associated MADS-box(DAM)gene cluster in peach serves as a key regulatory hub on which the seasonal temperatures act and orchestrate dormancy onset and exit,chilling response and floral bud developmental pace.Yet,how different temperature regimes interact with and regulate the six linked DAM genes remains unclear.Here,we demonstrate that chilling downregulates DAM1 and DAM3–6 in dormant floral buds with distinct patterns and identify DAM4 as the most abundantly expressed one.We reveal multiple epigenetic events,with tri-methyl histone H3 lysine 27(H3K27me3)induced by chilling specifically in DAM1 and DAM5,a 21-nt sRNA in DAM3 and a ncRNA induced in DAM4.Such induction is inversely correlated with downregulation of their cognate DAMs.We also show that the six DAMs were hypermethylated,associating with the production of 24-nt sRNAs.Hence,the chilling-responsive dynamic of the different epigenetic elements and their interactions likely define distinct expression abundance and downregulation pattern of each DAM.We further show that the expression of the five DAMs remains steadily unchanged or continuously downregulated at the ensuing warm temperature after chilling,and this state of regulation correlates with robust increase of sRNA expression,H3K27me3 and CHH methylation,which is particularly pronounced in DAM4.Such robust increase of repressive epigenetic marks may irreversibly reinforce the chillingimposed repression of DAMs to ensure flower-developmental programming free from any residual DAM inhibition.Taken together,we reveal novel information about genetic and epigenetic regulation of the DAM cluster in peach,which will be of fundamental significance in understanding of the regulatory mechanisms underlying chilling requirement and dormancy release,and of practical application for improvement of plasticity of flower time and bud break in fruit trees to adapt changing climates.
文摘Syringa species not only have good ornamental properties but also play an important role in the landscaping and environmental purification of cities.To investigate the chilling stress resistance of Syringa oblata Lindl.and Syringa reticulata var.mandshurica and provide theoretical grounds for the practical cultivation of Syringa species,in vitro leaves were used to study photosynthetic gas exchange parameters and chlorophyll fluorescence parameters.After nine hours of chilling,decreasing rates of net photosynthesis,stomatal conductance,and transpiration in S.reticulata var.mandshurica leaves were significantly greater than that of the S.oblata,while intercellular CO2 concentrations in S.oblata leaves were higher than those in S.reticulata var.mandshurica.The quantum yield of PSII reaction center(APSII)declined in S.reticulata and light capture efficiency(Fv 0/Fm 0)was stable.However,reduction percentages of Fv 0/Fm 0,APSII,and Fv/Fm in S.oblata were significant higher than those of S.reticulata var.mandshurica.After nine hours of chilling,the relative variable fluorescence of VJ and VI of S.oblata increased and the increasing rate of VJ was greater than VI.In contrast,the change of VJ and VI in S.reticulata var.mandshurica leaves was relatively small.This suggests that chilling primarily damaged the electron transport process of QA to QB at the receptor site of the PSII reaction center.Photosynthetic capacity of S.oblata was more sensitive to chilling stress compared to S.reticulate var.mandshurica,which the limitations were mainly due to non-stomatal factors such as the decrease in electron transport efficiency,activity in the PSII reaction center,and the destruction of the photodamage defense system.

文摘The temperature drop of plants from the optimal requirements can increase tolerance to severe chilling stress. Photosynthesis and osmoregulators were analyzed during chilling stress to explore the adaptation mechanisms that underlie the induction of chilling tolerance in response to suboptimal temperature. The relationships of these processes to suboptimal temperature acclimation in watermelon were then determined.Suboptimal temperature-acclimated watermelon plants demonstrated tolerance during chilling stress, as indicated by the decreased electrolyte leakage and malondialdehyde accumulation compared with those non-acclimated watermelon plants. Chilling-induced photoinhibition and reduction in CO2 assimilation rate were alleviated after suboptimal temperature acclimation. The xanthophyll cycle level was enhanced by improving thermal dissipation ability and avoiding light damage. Consequently, the chilling tolerance of suboptimal temperature-acclimated watermelon plants was enhanced. The osmoregulation ability induced by suboptimal temperature acclimation protected watermelon plants against chilling injury because of the accumulation of small molecular substances, such as soluble sugar and proline. The protein levels of Rubisco activase(ClRCA) and the gene expression of the Benson–Calvin cycle simultaneously increased in suboptimal temperature-acclimated watermelon plants during chilling stress. Chilling tolerance in watermelon plants induced by suboptimal temperature acclimation is associated with enhanced photosynthetic adaptability and osmoregulation ability.

文摘The Mg-3.0Nd-0.2Zn-0.4Zr (NZ30K) alloys were prepared by direct-chill casting (DCC) and sand mould casting (SMC) processes,respectively and their microstructures and mechanical properties were investigated.The results indicate that casting method plays a remarkable influence on the microstructure and mechanical properties of as-cast NZ30K alloy.The grain size increases from 35-40μm in the billets made by the DCC to about 100-120μm in the billets by the SMC.The aggregation of Mg12Nd usually found at the triple joints of grain boundaries in the billets prepared by SMC while is not observable from the billets by DCC.The tensile strengths and elongations of the billets are 195.2 MPa and 15.5% by DCC,and 162.5 MPa and 3.2% by SMC,respectively.The tensile strength of the alloy by DCC is remarkably enhanced by T6 heat treatment,which reached 308.5 MPa.Fracture surfaces of NZ30K alloy have been characterized as intergranular fracture by SMC and quasi-cleavage fracture by DCC,respectively.

文摘Peach(Prunus persica Batsch‘Yuhualu’)fruit are sensitive to chilling injury(CI).Proline,polyamine(PA),and nitric oxide(NO)are important small regulators of various metabolic pathways under chilling stress that mitigate CI.Ethylene is known to promote senescence and CI,while 1-methylcyclopropene(1-MCP)is an antagonist that inhibits the effects of ethylene.However,how1-MCP and ethylene affect proline,PA,and NO levels under chilling stress remains unclear.To address these questions,1-MCP(1μL·L^(−1))and ethylene(1μL·L^(−1))treatments were applied to peach fruit.Fruit were stored at 4°C for 28 d,then moved to 25°C for 3 d immediately after cold storage.Peach fruit exhibited CI symptoms after 7 d of cold storage with enhanced electrolyte leakage and malondialdehyde contents.The 1-MCP treatment significantly(P<0.05)restrained peach CI,and fruit did not exhibit CI symptoms until 14 d of cold storage.Proline and PAs in peach under chilling stress weremostly synthesized from glutamate and arginine,which were catalyzed by1-pyrroline-5-carboxylate synthetase and arginine decarboxylase,respectively.1-MCPtreated fruit exhibited higher proline and PA contents and enhanced chilling tolerance compared to the control,while ethylene-treated fruit had lower proline and PA contents and reduced chilling tolerance.Ethylene-treated fruit,which exhibited more severe CI symptoms compared to the control,had significantly(P<0.05)lower NO contents and NO synthase activities.However,NOmay not be a direct acting factor in 1-MCPinduced chilling tolerance,as 1-MCP-treated fruit had lower NO contents and NO synthase activities compared to the control.In conclusion,proline and PA clearly played direct and important roles in 1-MCP-induced peach chilling tolerance,while NO may not be actively involved.

文摘Low-temperature storage is extensively used to optimize the postharvest life of various fresh fruits.However,red pitahaya(Hylocereus polyrhizus)fruits are sensitive to chilling injury(CI),which leads to the limitation of low-temperature storage.In this study,red pitahaya fruits were stored at 2,4,6,8,and 10℃,respectively,for 27 days to determine the appropriate storage temperature.During the storage of red pitahaya fruits,storage at 8℃ was more effective in suppressing decay and maintaining quality than other low temperatures.Lowtemperature(2,4,and 6℃)storage decreased weight loss(WL)and maintained higher content of titratable acidity(TA),soluble sugars(SS),and total phenolics(TP)but different degrees of CI were detected.No CI was observed at 8℃ and 10℃.Red pitahay as stored at 8 and 10℃ were associated with better color evaluation,lower electrolyte leakage(EL),respiration rate,and lipoxygenase(LOX)activity,and higher fruit firmness,superoxide dismutase(SOD)activity,and catalase(CAT)activity.However,higher storage temperature(10℃)resulted in higher metabolic activity leading to lower quality and antioxidant capacities compared with 8℃.Therefore,our results demonstrated that red pitahaya stored at 8℃ exhibited a protective effect on fruit quality and resisted CI development during storage.
