CHIP相关基因与MPN患者心脑血管事件的风险分析

目的:分析骨髓增殖性肿瘤(MPN)患者不确定潜能的克隆性造血(CHIP)相关基因突变谱和临床特征,探讨CHIP相关基因与其心脑血管事件(CCE)的相关性及可能作用机制。方法:回顾性分析2019年8月-2022年7月首都医科大学附属北京安贞医院血液科收... 目的:分析骨髓增殖性肿瘤(MPN)患者不确定潜能的克隆性造血(CHIP)相关基因突变谱和临床特征,探讨CHIP相关基因与其心脑血管事件(CCE)的相关性及可能作用机制。方法:回顾性分析2019年8月-2022年7月首都医科大学附属北京安贞医院血液科收治的73例MPN患者的临床资料和二代测序结果,采用Logistic回归分析CHIP相关基因、炎症细胞因子对MPN患者CCE的影响。结果:55例(75.3%)MPN患者检出CHIP相关基因,原发性血小板增多症(ET)和真性红细胞增多症(PV)患者CHIP相关基因各突变频率差异无统计学意义。CHIP相关基因突变以单基因形式为主,检出率从高至低依次为JAK2V617F(63.0%,46/73)、ASXL1(16.4%,12/73)、TET2(11.0%,8/73)、DNMT3A(9.6%,7/73)、SRSF2(6.9%,5/73)、SF3B1(4.1%,3/73)、TP53(1.4%,1/73)和PPMID(1.4%,1/73)。年龄>60岁患者CHIP相关基因检出率明显高于≤60岁者[91.7%(33/36)vs 59.5%(22/37)]。27例(37.0%)MPN患者伴CCE(MPN/CCE),2次CCE者5例,均为动脉事件。CCE组患者年龄(62.8±12.8 vs 53.9±15.8岁,P=0.015)、IL-1β水平(17.7±26.0vs 4.3±8.6,P=0.012)、IL-8水平(360.7±598.6 vs 108.3±317.0,P=0.045)、血栓形成史(29.6%vs 2.2%,P=0.020)和CHIP相关基因检出率(88.9%vs 67.4%,P=0.040)高于无CCE组。多因素Logistic回归分析结果显示,年龄(OR=0.917,95%CI:0.843-0.999,P=0.047)、血栓形成史(OR=34.148,95%CI:2.392-487.535,P=0.009)、任何1个CHIP相关基因突变(OR=16.065,95%CI:1.217-212.024,P=0.035)和IL-1β水平升高(OR=0.929,95%CI:0.870-0.992,P=0.027)均是MPN/CCE的独立危险因素;CHIP相关单基因突变与MPN/CCE无关,但DNMT3A(OR=88.717,95%CI:2.690-292.482,P=0.012)、ASXL1(OR=7.941,95%CI:1.045-60.353,P=0.045)突变是PV/CCE的独立危险因素。结论:MPN患者CHIP相关基因突变率高,尤其是60岁以上患者;高龄、血栓形成史、CHIP相关基因突变和IL-1β水平升高是MPN发生CCE的独立危险因素。DNMT3A、ASXL1单基因突变是PV患者CCE的独立危险因素。CHIP相关基因突变及炎症细胞因子IL-1β升高是MPN新的CCE危险因素。 展开更多

Copper-based azide(Cu(N_(3))2 or CuN_(3),CA)chips synthesized by in-situ azide reaction and utilized in miniaturized explosive systems has become a hot research topic in recent years.However,the advantages of in-situ synthesis method,including small size and low dosage,bring about difficulties in quantitative analysis and differences in ignition capabilities of CA chips.The aim of present work is to develop a simplified quantitative analysis method for accurate and safe analysis of components in CA chips to evaluate and investigate the corresponding ignition ability.In this work,Cu(N_(3))2 and CuN_(3)components in CA chips were separated through dissolution and distillation by utilizing the difference in solubility and corresponding content was obtained by measuring N_(3)-concentration through spectrophotometry.The spectrophotometry method was optimized by studying influencing factors and the recovery rate of different separation methods was studied,ensuring the accuracy and reproducibility of test results.The optimized method is linear in range from 1.0-25.0 mg/L,with a correlation coefficient R^(2)=0.9998,which meets the requirements of CA chips with a milligram-level content test.Compared with the existing ICP method,component analysis results of CA chips obtained by spectrophotometry are closer to real component content in samples and have satisfactory accuracy.Moreover,as its application in miniaturized explosive systems,the ignition ability of CA chips with different component contents for direct ink writing CL-20 and the corresponding mechanism was studied.This study provided a basis and idea for the design and performance evaluation of CA chips in miniaturized explosive systems. 展开更多

On-demand droplet sorting is extensively applied for the efficient manipulation and genome-wide analysis of individual cells.However,state-of-the-art microfluidic chips for droplet sorting still suffer from low sorting speeds,sample loss,and labor-intensive preparation procedures.Here,we demonstrate the development of a novel microfluidic chip that integrates droplet generation,on-demand electrostatic droplet charging,and high-throughput sorting.The charging electrode is a copper wire buried above the nozzle of the microchannel,and the deflecting electrode is the phosphate buffered saline in the microchannel,which greatly simplifies the structure and fabrication process of the chip.Moreover,this chip is capable of high-frequency droplet generation and sorting,with a frequency of 11.757 kHz in the drop state.The chip completes the selective charging process via electrostatic induction during droplet generation.On-demand charged microdroplets can arbitrarilymove to specific exit channels in a three-dimensional(3D)-deflected electric field,which can be controlled according to user requirements,and the flux of droplet deflection is thereby significantly enhanced.Furthermore,a lossless modification strategy is presented to improve the accuracy of droplet deflection or harvest rate from 97.49% to 99.38% by monitoring the frequency of droplet generation in real time and feeding it back to the charging signal.This chip has great potential for quantitative processing and analysis of single cells for elucidating cell-to-cell variations. 展开更多

Trace amines(TAs)are metabolically related to catecholamine and associated with cancer and neurological disorders.Comprehensive measurement of TAs is essential for understanding pathological processes and providing proper drug intervention.However,the trace amounts and chemical instability of TAs challenge quantification.Here,diisopropyl phosphite coupled with chip two-dimensional(2D)liquid chromatography tandem triple-quadrupole mass spectrometry(LC-QQQ/MS)was developed to simultaneously determine TAs and associated metabolites.The results showed that the sensitivities of TAs increased up to 5520 times compared with those using nonderivatized LC-QQQ/MS.This sensitive method was utilized to investigate their alterations in hepatoma cells after treatment with sorafenib.The significantly altered TAs and associated metabolites suggested that phenylalanine and tyrosine metabolic pathways were related to sorafenib treatment in Hep3B cells.This sensitive method has great potential to elucidate the mechanism and diagnose diseases considering that an increasing number of physiological functions of TAs have been discovered in recent decades. 展开更多

目的 本课题组前期构建了一种正交泛素传递方法,并发现染色体浓缩调节蛋白2(RCC2)是泛素连接酶CHIP的潜在底物。本实验旨在验证CHIP是否能介导RCC2的泛素化及降解,以期通过CHIP来泛素化调控RCC2的表达和功能。方法 首先构建CHIP及其突... 目的 本课题组前期构建了一种正交泛素传递方法,并发现染色体浓缩调节蛋白2(RCC2)是泛素连接酶CHIP的潜在底物。本实验旨在验证CHIP是否能介导RCC2的泛素化及降解,以期通过CHIP来泛素化调控RCC2的表达和功能。方法 首先构建CHIP及其突变体质粒,验证CHIP与RCC2是否有相互作用;然后构建pLVX-RCC2-FLAG重组质粒并转染进CHIP敲减细胞株(shCHIP)、HEK293细胞株、CHIP过表达细胞株(OE CHIP),通过免疫共沉淀下拉富集RCC2-FLAG并检测其泛素化水平;接下来在HEK293细胞中转染不同量的CHIP,以及设置CHX chase蛋白稳定性实验,检测RCC2的蛋白水平变化;最后利用泛素突变体K48R-Ub及K11R-Ub进行CHIP介导RCC2上形成泛素链的实验检测。结果 在HEK293细胞中,CHIP能够介导RCC2的泛素化,并在RCC2上形成K48和K11多聚泛素链,促进其通过蛋白酶体发生降解。结论 泛素连接酶CHIP能够介导RCC2的泛素化及降解,为实现通过CHIP来泛素化调控RCC2提供了理论基础。 展开更多

Spiking neural networks(SNNs)utilize brain-like spatiotemporal spike encoding for simulating brain functions.Photonic SNN offers an ultrahigh speed and power efficiency platform for implementing high-performance neuromorphic computing.Here,we proposed a multi-synaptic photonic SNN,combining the modified remote supervised learning with delayweight co-training to achieve pattern classification.The impact of multi-synaptic connections and the robustness of the network were investigated through numerical simulations.In addition,the collaborative computing of algorithm and hardware was demonstrated based on a fabricated integrated distributed feedback laser with a saturable absorber(DFB-SA),where 10 different noisy digital patterns were successfully classified.A functional photonic SNN that far exceeds the scale limit of hardware integration was achieved based on time-division multiplexing,demonstrating the capability of hardware-algorithm co-computation. 展开更多

In this work,an automated microfluidic chip that uses negative pressure to sample and analyze solutions with high temporal resolution was developed.The chip has a T-shaped channel for mixing the sample with a fluorescent indicator,a flow-focusing channel for generating droplets in oil,and a long storage channel for incubating and detecting the droplets.By monitoring the fluorescence intensity of the droplets,the device could detect changes in solution accurately over time.The chip can generate droplets at frequencies of up to 42 Hz with a mixing ratio of 1:1 and a temporal resolution of 3–6 s.It had excellent linearity in detecting fluorescein solution in the concentration range 1–5μM.This droplet microfluidic chip provides several advantages over traditional methods,including high temporal resolution,stable droplet generation,and faster flow rates.This approach could be applied to monitoring calcium ions with a dynamic range from 102 to 107 nM and a detection limit of 10 nM. 展开更多

Emodin is an effective component of rhubarb with positive pharmacological effects on human health.However,it is also toxic to different cells or tissues to varying degrees.The effects of emodin on glomerular endothelial cells(GECs)remain to be tested,and the documented works were always performed in vitro and hardly reflect the real physiological situation.To study the effects of emodin on GECs in a biomimetic environment,we utilized a microfluidic chip to assess the physiological reaction of human renal glomerular endothelial cells to various concentrations of emodin in this work.The results showed that emodin caused cytotoxicity,impaired glomerular filtration barrier integrity to macromolecules,and increased barrier permeability in a dose-dependent manner.With the increase in emodin concentration,the concentration of the pro-inflammatory cytokine tumor necrosis factor-α,interleukin(IL)-6,transforming growth factor-β1,and monocyte chemoattractant protein(MCP-1)increased while the production of inflammatory cytokine IL-6 first increased and then decreased with the increase in emodin concentration.Our findings shed new light on emodin-induced nephrotoxicity and provide insights for the application of microfluidic chip devices to reveal drug-cell interactions. 展开更多

Diabetes mellitus is a global health problem resulting from islet dysfunction or insulin resistance.The mechanisms of islet dysfunction are still under investigation.Islet hormone secretion is the main function of islets,and serves an important role in the homeostasis of blood glucose.Elucidating the detailed mechanism of islet hormone secretome distortion can provide clues for the treatment of diabetes.Therefore,it is crucial to develop accurate,real-time,laborsaving,high-throughput,automated,and cost-effective techniques for the sensing of islet secretome.Microfluidic chips,an elegant platform that combines biology,engineering,computer science,and biomaterials,have attracted tremendous interest from scientists in the field of diabetes worldwide.These tiny devices are miniatures of traditional experimental systems with more advantages of timesaving,reagent-minimization,automation,high-throughput,and online detection.These features of microfluidic chips meet the demands of islet secretome analysis and a variety of chips have been designed in the past 20 years.In this review,we present a brief introduction of microfluidic chips,and three microfluidic chipsbased islet hormone sensing techniques.We focus mainly on the theory of these techniques,and provide detailed examples based on these theories with the hope of providing some insights into the design of future chips or whole detection systems. 展开更多

目的探讨热休克蛋白70羧基末端反应蛋白(CHIP)介导的8-氧代鸟嘌呤-DNA糖基化酶(OGG)1在老年性白内障发展过程中的作用。方法培养SRA01/04永生化人晶状体上皮细胞,分为空白对照组、空载体质粒转染组、CHIP过表达质粒转染组、细胞氧化损... 目的探讨热休克蛋白70羧基末端反应蛋白(CHIP)介导的8-氧代鸟嘌呤-DNA糖基化酶(OGG)1在老年性白内障发展过程中的作用。方法培养SRA01/04永生化人晶状体上皮细胞,分为空白对照组、空载体质粒转染组、CHIP过表达质粒转染组、细胞氧化损伤模型组。免疫沉淀实验检测OGG1蛋白的泛素化修饰及与E3泛素连接酶CHIP的结合能力。使用手持中波紫外线检测灯照射建立细胞氧化损伤模型,通过转染CHIP质粒提升其表达,Western印迹检测细胞中OGG1蛋白表达变化。结果免疫沉淀实验结果显示,晶状体上皮细胞中存在OGG1蛋白的泛素化修饰;UbiBrowser软件预测CHIP可能是引发OGG1泛素化修饰的E3泛素连接酶。Western印迹检测结果显示,CHIP能够与OGG1相互结合,提示OGG1可能参与了OGG1蛋白的泛素化修饰过程。在中波紫外线照射10 min时,晶状体上皮细胞中CHIP蛋白相对表达量明显低于其他中波紫外线照射时间;在中波紫外线照射20~40 min时,随着照射时间的增加,晶状体上皮细胞中CHIP蛋白相对表达量逐渐增加,差异有统计学意义(P<0.05)。根据后续实验建立的细胞氧化损伤模型均选用中波紫外线照射10 min后,再孵育24 h。CHIP过表达质粒转染组晶状体上皮细胞中CHIP蛋白相对表达量明显高于空载体质粒转染组和空白对照组(P<0.05);细胞氧化损伤模型组晶状体上皮细胞中OGG1蛋白相对表达量明显高于空白对照组(P<0.01);CHIP过表达质粒转染组上述指标明显低于细胞氧化损伤模型组、空载体质粒转染组(P<0.05)。结论E3泛素连接酶CHIP可诱发OGG1泛素化降解,造成晶状体上皮细胞中损伤性DNA聚集,导致老年性白内障的发生与发展。 展开更多

湖羊lncRNA-269的鉴定及转录调控分析

[目的]本文旨在研究湖羊长链非编码RNA-269(long no-coding RNA-269,lncRNA-269)序列特征、表达模式和转录调控,为后续开展lncRNA-269的功能研究提供理论基础。[方法]以湖羊为研究对象,采用5′/3′RACE结合PCR扩增方法获得其序列全长,利用RT-qPCR方法鉴定其在湖羊组织中的表达模式,并利用PCR方法扩增其启动子区,鉴定其启动子区特征,分析其转录调控情况。[结果]湖羊lncRNA-269全长3 146 bp,组织表达谱显示其在湖羊各组织中广泛表达,且在健康卵泡中显著高表达,其表达水平与NR5A1 mRNA水平和血清中雌二醇(E_(2))水平呈正相关。荧光素酶活性分析发现在lncRNA-269启动子区的-359~-17 nt有1个转录激活基序,在-1 485~-942 nt有1个转录抑制基序。JASPAR软件预测发现在lncRNA-269转录抑制启动子区域存在FOXO3、PDX1等转录因子结合位点,在转录激活区域有SMAD4、YY1等转录因子结合位点。过表达SMAD4可显著提升lncRNA-269启动子区荧光素酶活性,染色质免疫沉淀(ChIP)-PCR试验进一步确认了SMAD4特异性与lncRNA-269启动子区结合。[结论]湖羊lncRNA-269在健康卵泡中显著高表达,其启动子区存在一个转录激活区域和一个转录抑制区域,转录因子SMAD4可通过与lncRNA-269启动子区结合显著上调lncRNA-269的启动子活性。 展开更多

教育、社会资本对中国农村家庭代际流动的影响——基于CHIP数据的实证分析 被引量:20

良好的代际流动可以缩小中国城乡收入差距,家庭成员的教育和社会资本是中国农村家庭代际流动的重要通道。文章基于2013年中国家庭收入调查(CHIP)数据,从教育和社会资本的角度出发,探讨两者对中国农村家庭代际流动的影响机理及区域差异... 良好的代际流动可以缩小中国城乡收入差距,家庭成员的教育和社会资本是中国农村家庭代际流动的重要通道。文章基于2013年中国家庭收入调查(CHIP)数据,从教育和社会资本的角度出发,探讨两者对中国农村家庭代际流动的影响机理及区域差异。研究发现:男性子代的受教育水平对农村家庭代际流动具有显著的正向影响,而女性子代的受教育水平并未显著影响代际流动;尽管父代受教育水平并不直接影响家庭代际流动,但它在男性子代的代际流动中发挥了完全中介作用;社会资本在农村家庭代际流动中的作用也存在性别差异,男性子代通过社会资本路径求职能够促进其向上的代际流动,而社会资本在女性子代的代际流动中并未发挥显著作用;进一步的区域比较研究发现,农村地区整体上的代际流动不明显,且一级地区、二级地区均呈现微弱向下的流动趋势,三级地区呈现微弱向上的流动趋势。由此提出提升农村教育质量;维护农村家庭的社会网络和加强农村乡风文明建设;拓宽农民增收渠道,为农民提供更多非农就业信息等建议。 展开更多

目的:利用基因芯片研究Carboxyl terminus of Hsc70-interacting protein(CHIP)对肾间质纤维化TGF-β1/Smads信号通路的抑制性调节。方法:将18只SPF级SD大鼠中6只行unilateral ureteral occlusion(UUO)模型,6只植入弹性管脂肪垫加压梗... 目的:利用基因芯片研究Carboxyl terminus of Hsc70-interacting protein(CHIP)对肾间质纤维化TGF-β1/Smads信号通路的抑制性调节。方法:将18只SPF级SD大鼠中6只行unilateral ureteral occlusion(UUO)模型,6只植入弹性管脂肪垫加压梗阻,7天后再通定为reverse of unilateral ureteral occlusion(RUUO)组,6只UUO在7天处死留取左肾组织,另外6只为假手术组。6只RUUO大鼠在再通7天后处死,留取左侧肾组织,运用West-ern blotting检测肾组织CHIP的表达,免疫组化检测肾组织中α-SMA、CD68的阳性表达面积,定制芯片研究TGF-β1/Smads信号通路的基因变化。结果:UUO组TGF-β1、smad2、smad3、COL1a2、JunB基因显著上调,和假手术组比较上调超过1.5倍,肾组织中的α-SMA、CD68表达面积和假手术组比较有显著统计学意义P<0.01。而RUUO组CHIP表达量较UUO组更是显著增强P<0.01,TGF-β1、smad2、smad3、COL1a2、JunB基因显著下调,和UUO组比较下调超过1.5倍,肾组织中的α-SMA、CD68表达面积减小和UUO组比较有显著统计学意义P<0.01。结论:发现CHIP的过量表达能显著降低细胞内,TGF-β1、Smad2/3、COL1a2、JunB基因水平,Western blot结果也表明CHIP蛋白表达能明显降低Smads介导的基因转录活性,进一步的基因芯片结果显示CHIP蛋白能明显下调TGF-β1所诱导的下游基因JunB的表达。结果提示CHIP蛋白能抑制性调节TGF-β1/Smads信号通路,抑制肾纤维化。 展开更多
