BACKGROUND Patients diagnosed with non-small-cell lung cancer with activated epidermal growth factor receptor mutations are more likely to develop leptomeningeal(LM)metastasis than other types of lung cancers and have...BACKGROUND Patients diagnosed with non-small-cell lung cancer with activated epidermal growth factor receptor mutations are more likely to develop leptomeningeal(LM)metastasis than other types of lung cancers and have a poor prognosis.Early diagnosis and effective treatment of leptomeningeal carcinoma can improve the prognosis.CASE SUMMARY A 55-year-old female with a progressive headache and vomiting for one month was admitted to Peking University First Hospital.She was diagnosed with lung adenocarcinoma with osseous metastasis 10 months prior to admittance.epidermal growth factor receptor(EGFR)mutation was detected by genomic examination,so she was first treated with gefitinib for 10 months before acquiring resistance.Cell-free cerebrospinal fluid(CSF)circulating tumor DNA detection by next-generation sequencing was conducted and indicated the EGFR-Thr790Met mutation,while biopsy and cytology from the patient’s CSF and the first enhanced cranial magnetic resonance imaging(MRI)showed no positive findings.A month later,the enhanced MRI showed linear leptomeningeal enhancement,and the cytology and biochemical examination in CSF remained negative.Therefore,osimertinib(80 mg/d)was initiated as a second-line treatment,resulting in a good response within a month.CONCLUSION This report suggests clinical benefit of osimertinib in LM patients with positive detection of the EGFR-Thr790Met mutation in CSF and proposes that the positive findings of CSF circulating tumor DNA as a liquid biopsy technology based on the detection of cancer-associated gene mutations may appear earlier than the imaging and CSF findings and may thus be helpful for therapy.Moreover,the routine screening of chest CT with the novel coronavirus may provide unexpected benefits。展开更多
Objective To establish a highly sensitive serologic method for detecting the circulating antigen of Schistosoma japonicum for the diagnosis of this disease and the evaluation of drug therapy effect.Methods A set of ...Objective To establish a highly sensitive serologic method for detecting the circulating antigen of Schistosoma japonicum for the diagnosis of this disease and the evaluation of drug therapy effect.Methods A set of complex support made of poly vinyl chloride (PVC) film slide, adsorbed with specific antibody, was used to test serum samples collected from cases with schistosomiasis japonica, by adding specific enzyme conjugate and substrate tetra methyl benzidine (TMB) with 3% H 2O 2 in 10∶0.1, developed or prepared in our laboratory. Tests were carried out with the set on cases with malaria, paragonimiasis, clonorchiasis, visceral leishmaniasis and cysticercosis as well as normal individuals as control. Serum sample of 50 μl was used in each test and reacted with reagents at room temperature for 30 minutes. The color of positive reaction was blue while negative reaction was colorless.Results Positive rate among cases with schistosomiasis japonica was 100% (45/45) in acute stage, 94.8% (530/559) in early chronic stage and 52.4% (66/126) in late stage. False positive reaction was found neither among all normal individuals (0/513), nor among cases of 155 with malaria, 120 with clonorchiasis, 24 with visceral leishmaniasis and 110 with cysticercosis, except one among 29 cases with parasgonimiasis (1/29).Conclusions The rapid one step enzyme Immunoassay (EIA) to detect circulating schistosome antigen (CSA) has been well established in our laboratory with high sensitivity and good specificity as well as reproducibility. The reaction result can be read easily even in field conditions without power supply. Moreover, the assay is time saving, simple to handle and suitable for the diagnosis of shistosomiasis japonica and evaluation of drug therapeutic effect in practice in the control program of the disease.展开更多

In this study, aptamer-conjugated upconversion nano- particles (UCNPs) are used for the first time as nanoprobes to recognize tumor cells, which are then enriched by attaching with magnetic nanoparticles (MNPs) and placing in the presence of a magnetic field. Owing to the autofluorescence- free nature of upconversion luminescence imaging, as well as the use of magnetic separation to further reduce background signals, our technique allows for highly sensitive detection and collection of small numbers of tumor cells spiked into healthy blood samples, and shows promise for CTC detection in medical diagnostics.展开更多

Total 23 CFB incineration po...This first nationwide survey was conducted to evaluate the overall performance of the circulating fluidized bed (CFB) incineration of municipal solid waste (MSW) in 2014-2015 in China. Total 23 CFB incineration power plants were evaluated. The data for monthly average flue gas emission of particles, CO, NOx, SO2 and HCl were collected over 12 consecutive months. The data were analyzed to assess the overall performance of CFB incineration by applying the Mahalanobis distance as a multivariate outlier detection method. Although the flue gas emission parameters had met the Chinese national emission standards, there were 11 total outliers (abnormal behavior) detected in 6 out of 23 CFB incineration oower olants from the oersoective of the MSW incineration performance. The results demonstrate that it is more important for a better perlbrmance of CFBs to reduce the lrequencles ot the MSW load changes, rather than the magnitudes of the MSW load changes, particularly reducing the frequencies in the range of 10% and more of the load changes, under the same and stable conditions. Furthermore, the overloading occurs more often than the underloading during the operation of the CFB incineration power plants in China. The frequent overloading is 0% to 30% ot the designed capacity. To achieve the stable performance of CFBs in practice, an appropriately designed MSW storage capacity is suggested to build in a plant to buffer and reduce the frequency of the load changes.展开更多

DNA methylation is an important epigenetic marker known to affect gene expression. cfDNA methylation detection is a very promising approach as abnormal distribution of DNA methylation is one of the hallmarks of many cancers and methylation changes occur early during carcinogenesis. This re?view summarizes the various investigational applications of cfDNA methylation and its oxidized de?rivatives as biomarkers for cancer diagnosis, prenatal diagnosis and organ transplantation monitoring.The review also provides a brief overview of the technologies for cfDNA methylation analysis based on next generation sequencing.展开更多
