Rhodobacter sphaeroides is a gram-negative purple phototroph, used as a model organism for this study due to its genomic complexity and metabolic versatility. Its genome has been completely sequenced, and profiles of the differential gene expression under aerobic, semi-aerobic, and photosynthetic conditions were examined. In this study, we hypothesized that R. sphaeroides will show altered growth characteristics, morphological properties, and gene expression patterns when grown under simulated microgravity. To test that, we measured the optical density and colony-forming units of cell cultures grown under both microgravity and normal gravity conditions. Differences in the cell morphology were observed using scanning electron microscopy (SEM) images by measuring the length and the surface area of the cells under both conditions. Furthermore, we also identified homologous genes of R. spheroides using the differential gene expression study of Acidovorax under microgravity in our laboratory. Growth kinetics results showed that R. sphaeroides cells grown under microgravity experience a shorter log phase and early stationary phase compared to the cells growing under normal gravity conditions. The length and surface area of the cells under microgravity were significantly higher confirming that bacterial cells experience altered morphological features when grown under microgravity conditions. Differentially expressed homologous gene analysis indicated that genes coding for several COG and GO functions, such as metabolism, signal-transduction, transcription, translation, chemotaxis, and cell motility are differentially expressed to adapt and survive microgravity.展开更多

Cell wall invertases(CWINs), which catalyze the irreversibly conversion of sucrose into glucose and fructose in the extracellular space, are key enzymes participating in sucrose allocation in higher plants. Previous studies have shown that CWINs play an essential role in bulblet initiation process in bulbous crops, but the underlying molecular mechanism remains unclear. Here, a CWIN gene of Lilium brownii var. giganteum(Lbg) was identified and amplified from genomic DNA. Quantitative RT-PCR assays revealed that the expression level of LbgCWIN1 was highly upregulated exactly when the endogenous starch degraded in non-sucrose medium during in vitro bulblet initiation in Lbg. Phylogenetic relationship, motif, and domain analysis of LbgCWIN1 protein and CWINs in other plant species showed that all sequences of these CWIN proteins were highly conserved. The promoter sequence of LbgCWIN1 possessed a number of alpha-amylase-, phytohormone-, light-and stress-responsive cis-elements. Meanwhile, β-glucuronidase(GUS) assay showed that the 459 bp upstream fragment from the translational start site displayed maximal promoter activity. These results revealed that LbgCWIN1 might function in the process of in vitro bulblet initiation and be in the response to degradation of endogenous starch.展开更多

Tracking apps that have been adopted in many countries to collect data in a homogeneous and immediate way have made up for the difficulty of collecting data and standardizing evaluation criteria. However, the regulation on the protection of personal data in the health sector and the adoption of the new General Data Protection Regulation in European countries has placed a strong limitation on their use. This has not been the case in non-European countries, where monitoring methodologies have become widespread. The textual analysis presented is based on co-occurrence and multiple correspondence analysis to show the contact tracing methods adopted in different countries in the pandemic period by relating them to the issue of privacy. It also analyzed the possibility of applying Blockchain technology in applications for tracking contagions from COVID-19 and managing health data to provide a high level of security and transparency, including through anonymization, thus increasing user trust in using the apps.展开更多

文摘BACKGROUND Perianal fistulising Crohn's disease(PFCD)and glandular anal fistula have many similarities on conventional magnetic resonance imaging. However, many patients with PFCD show concomitant active proctitis, but only few patients with glandular anal fistula have active proctitis.AIM To explore the value of differential diagnosis of PFCD and glandular anal fistula by comparing the textural feature parameters of the rectum and anal canal in fat suppression T2-weighted imaging(FS-T2WI).METHODS Patients with rectal water sac implantation were screened from the first part of this study(48 patients with PFCD and 22 patients with glandular anal fistula).Open-source software ITK-SNAP(Version 3.6.0,http://www.itksnap.org/)was used to delineate the region of interest(ROI)of the entire rectum and anal canal wall on every axial section, and then the ROIs were input in the Analysis Kit software(version V3.0.0.R,GE Healthcare)to calculate the textural feature parameters.Textural feature parameter differences of the rectum and anal canal wall between the PFCD group vs the glandular anal fistula group were analyzed using Mann-Whitney U test. The redundant textural parameters were screened by bivariate Spearman correlation analysis, and binary logistic regression analysis was used to establish the model of textural feature parameters. Finally, diagnostic accuracy was assessed by receiver operating characteristic-area under the curve(AUC)analysis.RESULTS In all,385 textural parameters were obtained, including 37 parameters with statistically significant differences between the PFCD and glandular anal fistula groups.Then,16 texture feature parameters remained after bivariate Spearman correlation analysis, including one histogram parameter(Histogram energy);four grey level co-occurrence matrix(GLCM)parameters(GLCM energy_all direction_offset1_SD,GLCM entropy_all direction_offset4_SD,GLCM entropy_all direction_offset7_SD,and Haralick correlation_all direction_offset7_SD);four texture parameters(Correlation_all direction_offset1_SD,cluster prominence_angle 90_offset4,Inertia_all direction_offset7_SD,and cluster shade_angle 45_offset7);five grey level run-length matrix parameters(grey level nonuniformity_angle 90_offset1,grey level nonuniformity_all direction_offset4_SD,long run high grey level emphasis_all direction_offset1_SD,long run emphasis_all direction_offset4_SD,and long run high grey level emphasis_all direction_offset4_SD);and two form factor parameters(surface area and maximum 3D diameter). The AUC,sensitivity, and specificity of the model of textural feature parameters were 0.917,85.42%,and 86.36%,respectively.CONCLUSION The model of textural feature parameters showed good diagnostic performance for PFCD.The texture feature parameters of the rectum and anal canal in FS-T2WI are helpful to distinguish PFCD from glandular anal fistula.

文摘Introduction:Verruca vulgaris is one of the most common low-risk HPV infections and is characterized by excessive proliferation of keratinocytes.Currently, very little genetic information is available regarding verruca vulgaris in the Chinese population. This study aimed to obtain comprehensive transcript information of verruca vulgaris by RNA sequencing.Methods:High-throughput sequencing was performed on three fresh verruca vulgaris samples and adjacent normal skin on the Illumina sequencing platform. The transcriptomes were analyzed using bioinformatics and the differentially expressed genes(DEGs)were verified by immunohistochemistry.Verruca vulgaris exhibited a unique molecular signature. Results:In total,1,643 DEGs were identified in verruca vulgaris compared to normal skin. The functions of the DEGs were studies by Gene Ontology(GO)enrichment,Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis,DEGs Reactome analysis, disease annotation function, and STRING protein-protein interaction(PPI)network analysis. The results revealed 595 GO terms associated with the cell cycle, signal transduction, immune system, signaling molecules, and interaction. The Reactome analysis revealed enrichment in reversible hydration of carbon dioxide and BMP signaling, while the disease annotation function revealed that the enriched DEGs are involved in keratosis disorders. The STRING PPI network showed that the edges with the highest density mainly included the 2′-5′oligoadenylate synthase(OAS)family-related proteins.Furthermore, the M-code analysis found ISG15,IRF7,and OASL were scored as significant modules and their high expression compared to the control was verified by immunohistochemistry. Conclusion:These findings contribute to the genetic information of verruca vulgaris in the Chinese population, revealing that interferon-stimulated genes may play essential roles in verruca vulgaris.

文摘[Objectives]Selenium(Se)-enriched rice is the main type of Se-enriched agricultural product developed in China, and this study aimed to understand the impact of selenium application on the metabolites in rice.[Methods]Se-enriched rice was prepared by foliar application of a sodium selenite aqueous solution, and high-throughput analysis of differential metabolites in Se-enriched rice was conducted based on extensive non-targeted metabolome.[Results]There were significant differences in metabolites between Se-enriched rice and ordinary rice, and a total of 535 differential metabolites were identified.Among them,420 metabolites in Se-enriched rice were upregulated, accounting for 78.5%,far higher than downregulated metabolites. The enrichment differences of three KEGG metabolic pathways, including cysteine and methionine metabolism, zeatin biosynthesis, and arachidonic acid metabolism, reached a significant level, indicating that selenium enrichment had a significant regulatory effect on the metabolism of sulfur-containing amino acids, the synthesis of natural cytokinin zeatin, and arachidonic acid bioactive components in rice.[Conclusions]The results can provide a theoretical basis for the production of Se-enriched rice.
文摘The term “microgravity” is used to describe the “weightlessness” or “zero-g” circumstances that can only be found in space beyond earth’s atmosphere. Differentially expressed homologous gene analysis indicated that genes coding for several COG and GO functions, such as metabolism, signal-transduction, transcription, translation, chemotaxis, and cell motility are differentially expressed to adapt and survive microgravity.
文摘Four numerical schemes are introduced for the analysis of photocurrent transients in organic photovoltaic devices.Themathematicalmodel for organic polymer solar cells contains a nonlinear diffusion-reaction partial differential equation system with electrostatic convection attached to a kinetic ordinary differential equation. To solve the problem,Polynomial-based differential quadrature,Sinc, and Discrete singular convolution are combined with block marching techniques. These schemes are employed to reduce the problem to a nonlinear algebraic system. The iterative quadrature technique is used to solve the reduced problem. The obtained results agreed with the previous exact one and the finite element method.Further, the effects of different times, different mobilities, different densities, different geminate pair distances, different geminate recombination rate constants, different generation efficiencies, and supporting conditions on photocurrent have been analyzed. The novelty of this paper is that these schemes for photocurrent transients in organic polymer solar cells have never been presented before,so the results may be useful for improving the performance of solar cells.
文摘Various data sets showing the prevalence of numerous viral diseases have demonstrated that the transmission is not truly homogeneous. Two examples are the spread of Spanish flu and COVID-19.The aimof this research is to develop a comprehensive nonlinear stochastic model having six cohorts relying on ordinary differential equations via piecewise fractional differential operators.Firstly, the strength number of the deterministic case is carried out. Then, for the stochastic model,we show that there is a critical number RS0 that can predict virus persistence and infection eradication.Because of the peculiarity of this notion,an interesting way to ensure the existence and uniqueness of the global positive solution characterized by the stochastic COVID-19 model is established by creating a sequence of appropriate Lyapunov candidates.Adetailed ergodic stationary distribution for the stochastic COVID-19 model is provided. Our findings demonstrate a piecewise numerical technique to generate simulation studies for these frameworks. The collected outcomes leave no doubt that this conception is a revolutionary doorway that will assist mankind in good perspective nature.
文摘Because of the features involved with their varied kernels, differential operators relying on convolution formulations have been acknowledged as effective mathematical resources for modeling real-world issues. In this paper,we constructed a stochastic fractional framework of measles spreading mechanisms with dual medication immunization considering the exponential decay and Mittag-Leffler kernels. In this approach, the overall population was separated into five cohorts.Furthermore, the descriptive behavior of the system was investigated, including prerequisites for the positivity of solutions, invariant domain of the solution, presence and stability of equilibrium points, and sensitivity analysis. We included a stochastic element in every cohort and employed linear growth and Lipschitz criteria to show the existence and uniqueness of solutions.Several numerical simulations for various fractional orders and randomization intensities are illustrated.
文摘Natural fibers have been extensively researched as reinforcement materials in polymers on account of their environmental and economic advantages in comparison with synthetic fibers in the recent years.Bamboo fibers are renowned for their good mechanical properties, abundance, and short cycle growth. As beams are one of the fundamental structural components and are susceptible to mechanical loads in engineering applications, this paper performs a study on the free vibration and buckling responses of bamboo fiber reinforced composite(BFRC)beams on the elastic foundation.Three different functionally graded(FG)layouts and a uniform one are the considered distributions for unidirectional long bamboo fibers across the thickness. The elastic properties of the composite are determined with the law of mixture.Employing Hamilton’s principle, the governing equations of motion are obtained. The generalized differential quadrature method(GDQM)is then applied to the equations to obtain the results. The achieved outcomes exhibit that the natural frequency and buckling load values vary as the fiber volume fractions and distributions, elastic foundation stiffness values, and boundary conditions(BCs)and slenderness ratio of the beam change.Furthermore,a comparative study is conducted between the derived analysis outcomes for BFRC and homogenous polymer beams to examine the effectiveness of bamboo fibers as reinforcement materials, demonstrating the significant enhancements in both vibration and buckling responses, with the exception of natural frequencies for cantilever beams on the Pasternak foundation with the FG-◇fiber distribution.Eventually, the obtained analysis results of BFRC beams are also compared with those for carbon nanotube reinforced composite(CNTRC)beams found in the literature, indicating that the buckling loads and natural frequencies of BFRC beams are lower than those of CNTRC beams.

文摘Gears are pivotal in mechanical drives, and gear contact analysis is a typically difficult problem to solve.Emerging isogeometric analysis(IGA)methods have developed new ideas to solve this problem. In this paper,a threedimensional body parametric gear model of IGA is established, and a theoretical formula is derived to realize single-tooth contact analysis. Results were benchmarked against those obtained from commercial software utilizing the finite element analysis(FEA)method to validate the accuracy of our approach. Our findings indicate that the IGA-based contact algorithmsuccessfullymet theHertz contact test. When juxtaposed with the FEA approach, the IGAmethod demonstrated fewer node degrees of freedomand reduced computational units, all whilemaintaining comparable accuracy.Notably, the IGA method appeared to exhibit consistency in analysis accuracy irrespective of computational unit density, and also significantlymitigated non-physical oscillations in contact stress across the tooth width. This underscores the prowess of IGA in contact analysis. In conclusion,IGA emerges as a potent tool for addressing contact analysis challenges and holds significant promise for 3D gear modeling, simulation, and optimization of various mechanical components.
文摘The fractional-order Boussinesq equations(FBSQe)are investigated in this work to see if they can effectively improve the situation where the shallow water equation cannot directly handle the dispersion wave. The fuzzy forms of analytical FBSQe solutions are first derived using the Adomian decomposition method. It also occurs on the sea floor as opposed to at the functionality.A set of dynamical partial differential equations(PDEs)in this article exemplify an unconfined aquifer flow implication.Thismethodology can accurately simulate climatological intrinsic waves,so the ripples are spread across a large demographic zone. The Aboodh transform merged with the mechanism of Adomian decomposition is implemented to obtain the fuzzified FBSQe in R,R^(n) and(2nth)-order involving generalized Hukuhara differentiability.According to the system parameter,we classify the qualitative features of the Aboodh transform in the fuzzified Caputo and Atangana-Baleanu-Caputo fractional derivative formulations, which are addressed in detail. The illustrations depict a comparison analysis between the both fractional operators under gH-differentiability,as well as the appropriate attributes for the fractional-order and unpredictability factorsσ∈[0,1].A statistical experiment is conducted between the findings of both fractional derivatives to prevent changing the hypothesis after the results are known.Based on the suggested analyses, hydrodynamic technicians,as irrigation or aquifer quality experts, may be capable of obtaining an appropriate storage intensity amount, including an unpredictability threshold.

文摘Lily(Lilium spp.) is an important ornamental flower, which is mainly propagated by bulbs. These results revealed that LbgCWIN1 might function in the process of in vitro bulblet initiation and be in the response to degradation of endogenous starch.

文摘This study proposes a comprehensive, coupled thermomechanical model that replaces local spatial derivatives in classical differential thermomechanical equations with nonlocal integral forms derived from the peridynamic differential operator(PDDO),eliminating the need for calibration procedures. The model employs a multi-rate explicit time integration scheme to handle varying time scales in multi-physics systems.Through simulations conducted on granite and ceramic materials, this model demonstrates its effectiveness. It successfully simulates thermal damage behavior in granite arising from incompatible mineral expansion and accurately calculates thermal crack propagation in ceramic slabs during quenching. To account for material heterogeneity, the model utilizes the Shuffle algorithm andWeibull distribution, yielding results that align with numerical simulations and experimental observations. This coupled thermomechanical model shows great promise for analyzing intricate thermomechanical phenomena in brittle materials.
文摘The literary review presented in the following paper aims to analyze the tracking tools used in different countries during the period of the COVID-19 pandemic. It also analyzed the possibility of applying Blockchain technology in applications for tracking contagions from COVID-19 and managing health data to provide a high level of security and transparency, including through anonymization, thus increasing user trust in using the apps.
