However, there are few reports on the influence of electro-acupuncture at the Du meridian on neural stem cell proliferation. OBJECTIVE:To observe the influence of electro-acupuncture at Ren and Du meridians on neural stem cell proliferation in the subventricular zone and altered signal transduction in cerebral ischemia rats. DESIGN,TIME AND SETTING:A randomized, controlled, animal experiment was performed at the Laboratory of Human Anatomy,Medical College of Sun Yat-sen University from May 2006 to February 2008. MATERIALS:Mouse anti-rat bromodeoxyuridine (BrdU) monoclonal antibody was provided by Sigma,USA; mouse anti-rat nestin monoclonal antibody and extracellular signal-regulated protein kinase (ERK) specific inhibitor PD98059 were provided by Calbiochem,Germany; acupuncture needle was provided by Suzhou Acupuncture Supplies,China. METHODS:A total of 126 rats were randomly assigned to four groups:model (n = 36),Du meridian (n = 36),Ren/Du meridian (n = 36),and Ren/Du meridian + PD98059 (n = 18). Rats in the Ren /Du meridian + PD98059 group were observed on days 7 (n = 6) and 14 (n = 12) after cerebral ischemia injury. Rats in the model,Du meridian, and Ren/Du meridian groups were observed on days 7,14,and 28 after cerebral ischemia injury, with 12 rats per group at each time point. Thread occlusion was used to establish middle cerebral artery occlusion models. Electro-acupuncture was performed at Renzhong (DU 26) and Baihui (DU 20) acupoints in the Du meridian group,as well as Chengjiang (RN 24),Guanyuan (RN 4),Renzhong,and Baihui acupoints in the Ren/Du meridian and Ren/Du meridian + PD98059 groups 2 days after model establishment. In addition,electro-acupuncture stimulation with disperse-dense waves was performed, with 30 Hz disperse wave,100 Hz dense wave, and 5 V intensity for 20 minutes. Rats in the Ren/Du meridian + PD98059 group were treated with 0.2 μg PD98059 injection into the subventricular zone,2 μL per rat. Rats in the model group were not treated with electro-acupuncture. MAIN OUTCOME MEASURES:BrdU/nestin immunofluorescent staining was used to detect proliferating neural stem cells in the subventricular zone of cerebral ischemia rats; Western blot was used to determine phosphorylated ERK1 and 2 (pERK1/2) expression in the subventricular zone. RESULTS:On days 14 and 28 after cerebral ischemia, there were significantly more BrdU-positive and BrdU/nestin-positive cells in the Ren /Du meridian group compared with the Du meridian group (P < 0.05). PD98059 decreased the number of BrdU-positive and BrdU/nestin-positive cells induced by electro-acupuncture at the Ren and Du meridians (P < 0.05). On days 7,14,and 28 after treatment,pERK1/2 expression was significantly greater in the Du meridian and Ren/Du meridian groups compared with the model group (P < 0.05). The promoting effect of electro-acupuncture at Ren and Du meridians on ERK1/2 phosphorylation was superior to electro-acupuncture at the Du meridian alone on day 14 after model induction (P < 0.05). However,PD98059 completely abolished the promoting effect of electro-acupuncture at Ren/Du meridians on pERK1/2 expression (P < 0.05). CONCLUSION:Electro-acupuncture at Ren and Du meridians increased proliferation of subventricular zone neural stem cells, which was related to activation of the ERK pathway in a rat model of cerebral ischemia injury.展开更多

METHODS EA at 1 m A and 3 m A and Mox at 43?℃ and 46?℃ were applied to the Shangjuxu(ST37, bilateral) acupoints in model rats with visceral hypersensitivity. Responses of wide dynamic range neurons in dorsal horns of the spinal cord were observed through the extracellular recordings. Mast cells(MC) activity in the colons of rats were assessed, and 5-hydroxytryptamine(5-HT), 5-hydroxytryptamine 3 receptor(5-HT3R) and 5-HT4Rexpressions in the colons were measured.RESULTS Compared with normal control group, responses of wide dynamic range neurons in the dorsal horn of the spinal cord were increased in the EA at 1 m A and 3 m A groups(1 m A: 0.84 ± 0.74 vs 2.73 ± 0.65, P < 0.001; 3 m A: 1.91 ± 1.48 vs 6.44 ± 1.26, P < 0.001) and Mox at 43?℃ and 46?℃ groups(43?℃: 1.76 ± 0.81 vs 4.14 ± 1.83, P = 0.001; 46?℃: 5.19 ± 2.03 vs 7.91 ± 2.27, P = 0.01). MC degranulation rates and the expression of 5-HT, 5-HT3 R and 5-HT4 R in the colon of Mox 46?℃ group were decreased compared with model group(MC degranulation rates: 0.47 ± 0.56 vs 0.28 ± 0.78, P < 0.001; 5-HT: 1.42 ± 0.65 vs 7.38 ± 1.12, P < 0.001; 5-HT3R: 6.62 ± 0.77 vs 2.86 ± 0.88, P < 0.001; 5-HT4R: 4.62 ± 0.65 vs 2.22 ± 0.97, P < 0.001). CONCLUSION The analgesic effects of Mox at 46?℃ are greater than those of Mox at 43?℃, EA 1 m A and EA 3 m A.展开更多

METHODS: A total of 60 participants met the inclusion criteria were enrolled in a randomized controlled trial with the ratio of 1:1, with 30 cases in the treatment group and 30 cases in the control group. The participants in the treatment group were provided with realEA therapy whereas participants in control group were provided with placebo-EA therapy. Interventions were offered 5 days prior to the surgery, once daily, and for a total of 5 days. The scores of Mini-Mental State Examination(MMSE), and contents of serumal inflammatory cytokines including interleukin 1β(IL-1β) and tumor necrosis factor-α(TNF-α) were observed at 24 hours prior-and posterior-to the surgery respectively for assessing the incidence of POCD among patients. Meanwhile, adverse effects were monitored and recorded. RESULTS:(1) After surgery, both treatment group and control group showed a significant decrease in MMSE global scores(P < 0.001, < 0.001, respectively), and the score in control group decreased more significantly than that in treatment group(P < 0.05);(2) Contents of serumal IL-1β and TNF-α were significantly increased in both groups after 24 hours posterior to the surgery(P < 0.001), and the contents in control group increased more significantly than that in treatment group(P < 0.001);(3) After surgery, the incidence of POCD was 20% in treatment group versus 36.67% in control group. There was no statistical difference between 2 groups(P > 0.05);(4) No serious adverse events were reported in this trial, except 1 patient from treatment group had a slight hematoma after receiving acupuncture. CONCLUSION: EA precondition might reduce cognitive impairments after 24 hours posterior to knee replacement surgery in elderly through inhibiting expression of inflammatory cytokines, including both IL-1β and TNF-α. However, there is insufficient evidence to support that EA precondition could reduce incidence of POCD.展开更多

Methods: Thirty-two Wistar male rats were randomly divided into 4 groups. Mice in the blank control group did not receive any treatment;the remaining groups were applied with para-chlorophenylalanine (PCPA) 300 mg/kg intraperitoneal injection for 2 days. Diazepam group received intraperitoneal injection of Diazepam (0.9 mg/kg, i.p.) one time a day for 5 days, while M group was treated with saline (0.9 mg/kg, i.p.) at the same time. Rats in EA group were given EA treatment, 20 minutes, once a day for 5 days, and rats in remaining groups were put into fixation-machine for the same time everyday, lasting for 5 days. Rats were sacrificed after anesthesia at the 8th day. Real-time PCR was adopted to detect the expression in clock gene Per2 and Bmal1 of each group. Results: Compared with blank control group, the expression of Per2 was significant decreased in PCPA model group (P 0.05). Conclusion: EA can significant up-regulate the expression of Per2 in SD rats, and down-regulate gene Bmal1 expression, and benefiting the weight of rats. Thus, EA is a potentially promising intervention to treat sleep-deprivation.展开更多

There was significant difference between these two groups (p【0. 05).The motor evoked potential was recorded two weeks after spinal cord injury. The latency of theshort latency peak was 15. 61±4. 98 mS and the amplitude was 10. 61 0. 59 mV in the treatmentgroup; and 21. 7±5. 02 mS and 0. 56±0. 32 mV in the control group. Both latency and amplitudewere significant different between the two groups (P【0. 05, P【0. 05). The Anti-NF-H (neuro-filament) was determined at 2 and 4 weeks respectively after spinal cord injury. The more NF-Hpositive labeling fibers were found in the treatment group than in the control group (P【0. 01 ).展开更多

We observed that glutamate transporter mRNA expression decreased significantly in the area penumbra of cerebral cortex at 12h following focal cerebral ischemia. EA did not change glutamate transporter mRNA expression in normal rat brain but prevented the decrease of glutamate transporter mRNA expression in the area penumbra of cerebral cortex following focal cerebral ischemia. The results suggest that the protective effect of EA on ischemic neuronal death may bhe related to the increase of uptake of glutamate by its transporter.展开更多

EA decreases cortical excitability and increases its inhibitory process. Electric stimulation of subcortical nuclei: caudate n., cuneiform n. of midbrain RF, substantia nigra and superior colliculus of rats with petit mal epilepsy can suppress SW, and EA effect becomes low after destruction of the latter at two sites. Stimulation and destruction applied to reticular n. of thalamus elicit reverse effect. Microinjection of drugs affecting neurotransmitters: HC3, haloperidal, PCA and 3-MP into certain parts of brain leads to prolonged SW and reduced EA effect. Injection of muscimol shortens SW duration and reinforces EA effect, while baclofen induces reverse result. AP5 causes shortening of SW duration. The contents of GABA, glycine and taurine are increased markedly in sensory-motor area and optic area of the cortex after EA, while glutamate is decreased. The GABA contents of thalamic reticular n. and reticular part of substantia nigra are also obviously elevated.展开更多

OBJECTIVE: To observe the effects of electro-acupuncture on STAT3 expression and nuclear translocation in hippocampal CA1 region of rat models of brain ischemia/reperfusion. DESIGN: Randomized and controlled observation. SETTING: Staff Room of Acupuncture and Moxibustion, Department of Acupuncture and Bone Injury, Hubei College of Traditional Chinese Medicine; Tongji Medical College, Huazhong University of Science and Technology. MATERIALS: Seventy-two healthy SD rats, of clean degree and either gender, weighing (200±20) g, were provided by the Experimental Animal Center of Hubei College of Traditional Chinese Medicine. STAT3 monoclonal antibody was purchased from Santa Cruz Company, USA, and G-6805 electro-acupuncture instrument was purchased from Shanghai Medical Electronic Instruments Factory. METHODS: This experiment was carried out in the comprehensive laboratory of Department of Acupuncture and Bone Injury, Hubei College of Traditional Chinese Medicine between September 2005 and February 2006. Seventy-two rats were randomly divided into 4 groups: ① control group(n =6): Untouched. ② Sham-operation group (n =18): Artery was isolated, but without inserting thread bolt.③ Model group (n =24): Rat models of local brain ischemia/reperfusion were established with modified suture occlusion. ④Electro-acupuncture group (n =24): Dazhui and bilateral Neiguan points were selected for electro-acupuncture treatment. No. 28 acupuncture needle of 3.33 cm was used in the treatment. A G-6085 electro-acupuncture instrument with continuous wave, frequency of 120 times/min, intensity of 1 mA, 30 min/time, was used. Acupuncture was conducted firstly at ischemia/reperfusion 3 hours, then once every 12 hours. STAT3 positive nuclear translocation in hippocampal CA1 region of rats was observed with immunohistochemical method at 24, 48 and 72 hours after brain ishcemia/reperfusion, and then STAT3 positive cells were counted. MAIN OUTCOME MEASURES: STAT3 positive cells and nuclear translocation in hippocampal CA1 region of rats in each group. RESULTS: All the 72 rats were involved in the result analysis. ①In the control group and sham-operation group, STAT3 positive cells with light cytoplasm and nucleus were decreased , and nuclear translocation was not found. ② In the model group, STAT3 positive cells were mostly found in the cytoplasm of the hippocampal CA1 region at the ischemic side of rats after ischemia/reperfusion 24 hours. They were significantly more than those in the sham-operation group and control group [(18.00±2.68), (9.00±1.35), (8.00±1.22) cells/ mm2, P < 0.01], but cells with nuclear reaction were fewer; At ischemia/reperfusion 48 and 72 hours, STAT3 positive cells were increased, and they were significantly more than those of sham-operation group [(25.00±3.23), (35.00±3.52) cells/mm2, (13.00±1.93), (12.00±1.24) cells/mm2, P < 0.01]. Positive cells with nuclear reaction were found dark-stained. ③At ischemia/reperfusion 24, 48 and 72 hours, STAT3 positive cells were strongly expressed in hippocampal CA1 region at ischemic side of rats of electro-acupuncture group, and they were significantly more than those of model group [(25±3.52), (50±6.31), (75±8.09) cells/mm2, P < 0.01]. STAT3 positive cells were gradually enhanced with time, and considerable STAT3 nuclear positive reaction cells were found. CONCLUSION: Electro-acupuncture can activate STAT3 protein expression in hippocampal tissue of rats with local brain ischemia/reperfusion, promote STAT3 nuclear translocation and function its neuroprotective effect.展开更多

文摘BACKGROUND:Studies have shown that electro-acupuncture at the Ren meridian could improve proliferation of subventricular zone neural stem cells in cerebral-ischemic rats. CONCLUSION:Electro-acupuncture at Ren and Du meridians increased proliferation of subventricular zone neural stem cells, which was related to activation of the ERK pathway in a rat model of cerebral ischemia injury.

文摘OBJECTIVE: To investigate that whether electro-acupuncture(EA) precondition can reduce the incidence of postoperative cognitive dysfunction(POCD) following knee replacement and its safety in elderly. However, there is insufficient evidence to support that EA precondition could reduce incidence of POCD.
文摘Objective: The objective is to observe the treatment effect of electro-acupuncture (EA) on core circadian clock gene Per2 and Bmal1 expression in hypothalamus of sleep-deprivation (SD) rats. Thus, EA is a potentially promising intervention to treat sleep-deprivation.
文摘44 cats were used in this experiment. The spinal cords of 40 animals were injuredat about L1 level by using Allen’s method (400g/cm) and randomly divided into 2 groups: A)electro-acupuncture treatment group (n=20) and B) control group(n=20). 2 weeks after spinalcord injury, 80% of animals in the treatment group were survival and 45% of animals in the con-trol group were survival. The more NF-Hpositive labeling fibers were found in the treatment group than in the control group (P【0. 01 ).
文摘In this study, in situ hybridization histochemistry technique was used to observe the effect of electro-acupuncture (EA) on glutamate transporter mRNA expression following focal cerebral ischemia. The results suggest that the protective effect of EA on ischemic neuronal death may bhe related to the increase of uptake of glutamate by its transporter.

文摘Rett syndrome is a progressive neurodevelopmental disorder that lacks effective treatments.Although deep-brain stimulation can alleviate some symptoms in Rett model mice, this interventional manipula-tion requires deliberate surgical operations. Here,we report that electro-acupuncture stimulation(EAS)can ameliorate symptoms of an Mecp2-knockout rat model of Rett syndrome from the remote acupoints Baihui(GV 20),Yongquan(KI 1),and Shenmen(HT 7).We find that EAS not only prolongs the survival time of Rett rats, but also improves their behavior ability, including locomotion, motor coordination, and social interaction.Neural activation was observed in the substantia nigra of the midbrain, corpus striatum, and cerebral cortex of wild-type and Rett model rats,as reflected by the increased expression of the c-Fos protein.Hence,EAS provides a potential promising therapeutic tool for treating neurodevel-opmental diseases.
文摘119 patients with gastroduodenal diseases were observed, clinically and experi-mentally,on the effect of electro-acupuncture at the auricular acupoint"Gastric area" on gastrogram, and the effect was compared with that of Heart area and Ear tip points. There was a significant differ-ence in the amplitudes and frequencies of gastrogram of patients between pre-and post-acupuncture(P【0.01),with good diphasic regulation;the effect of acupuncturing at"Gastric area" is the mostsignificant as compared with that at "Heart" and "Ear tip".The experiment demonstrates that thereis a relationship between the "Gastric area" and stomach, and indicates that there is relative specificityof "Gastric Area" to stomach. This provides the experimental foundation for the auricular acupointtherapy in clinic.
文摘Electro- acupuncture (EA) applied to acupoints of GV channel suppresses spike- wave discharges(SW) of PTZ-induced petit mal epilepsy in rats. The GABA contents of thalamic reticular n. and reticular part of substantia nigra are also obviously elevated.

文摘BACKGROUND: It has been found in recent years that STAT3 widely distributes in nervous system, including hippocampal CA1-3 region, dentate gyrus and cerebral neocortex, etc. Ischemic brain injury can cause the release of some cytokines and growth factors, while electro-acupuncture may have multi-level, multi-channel and multi-target protective and interventional effects on ischemic brain injury. CONCLUSION: Electro-acupuncture can activate STAT3 protein expression in hippocampal tissue of rats with local brain ischemia/reperfusion, promote STAT3 nuclear translocation and function its neuroprotective effect.
文摘OBJECTIVE:To explore the prevention of urine retention after subabdominal surgery by electro-acupuncture Yin Ling Quan(SP 9),Zhao Hai(KI 6) acupoints with the assistance of subarachnoid acupoint anesthesia by observing the postoperative voiding time and urine volume of postoperative patients undergoing Electroacupuncture plus subarachnoid block anesthesia.METHODS:60 cases of preoperative expected for subarachnoid block surgery patients were randomly divided into two groups, the treatment group and the control group, with 30 cases in each group. The control group underwent routine subarachnoid anesthesia after surgery without acupuncture treatment group; The treatment group was treated with acupuncture bilateral Yin Ling Quan and Zhaohai acupoints 15 minutes before anesthesia, and then received unilateral electric anesthesia instrument treatment, the intensity of the patient was tolerable(generally 1.5 ~ 2.0 m A),and the routine subarachnoid block anesthesia was performed; The first urination time, urine volume, lower abdominal pain and assistant catheterization were recorded in the two groups.RESULTS:The first postoperative voiding time of the treatment group was significantly earlier compared with that of the control group, there was significant difference(P<0.05);the amount of urine was significantly more than that of the control group in reservation time, there was significant difference(P<0.05);the abdominal pain was light in the treatment group, there was significant difference(P<0.05);the cases of assisted catheterization of the control group were significantly more thanthose of the treatment group, there was significant difference(P< 0.05).CONCLUSION:Electro-acupuncture Yin Ling Quan(SP 9),Zhao Hai(KI 6) acupoints with the assistance of subarachnoid acupoint anesthesia could significantly reduce the urine retention after subabdominal surgery in postoperative patients with ubarachnoid block anesthesiau, accelerate the recovery of bladder function and promote early postoperative urination in addition to relieving the abdominal symptoms after operation.
