Based on clinical presentations, Traditional Chinese Medicine (TCM) classifies healthy people into nine constitutions: Balanced, Qi, Yang or Yin deficiency, Phlegm-dampness, Damp-heat, Blood stasis, Qi stagnation, and Inherited special constitutions. In particular, Yang and Yin deficiency constitutions exhibit cold and heat aversion, respectively. However, the intrinsic molecular characteristics of unbal- anced phenotypes remain unclear. To determine whether gene expression-based clustering can reca- pitulate TCM-based classification, peripheral blood mononudear cells (PBMCs) were collected from Chinese Han individuals with Yang/Yin deficiency (n = 12 each) and Balanced (n = 8) constitutions, and global gene expression profiles were determined using the Affymetrix HC-UI33A Plus 2.0 array. Notably, we found that gene expression-based classifications reflected distinct TCM-based subtypes. Consistent with the clinical observation that subjects with Yang deficiency tend toward obesity, series-clustering analysis detected several key lipid metabolic genes (diacylglycerol acyltransferase (DGAT2), acyl-CoA synthetase (ACSL1), and ATP-hinding cassette subfamily A member 1 (ABCAI)) to be down- and up- regulated in Yin and Yang deficiency constitutions, respectively. Our findings suggest that Yin]Yang deficiency and Balanced constitutions are unique entities in their mRNA expression profiles. Moreover, the distinct physical and clinical characteristics of each unbalanced constitution can be explained, in part, by specific gene expression signatures.展开更多
Objective: To study the effects of Bushen Yisui Capsule(补肾益髓胶囊, BSYSC) on the oligodendrocyte lineage genes(Olig) 1 and Olig2 in C57BL/6 mice with experimental autoimmune encephalomyelitis(EAE) in order t...Objective: To study the effects of Bushen Yisui Capsule(补肾益髓胶囊, BSYSC) on the oligodendrocyte lineage genes(Olig) 1 and Olig2 in C57BL/6 mice with experimental autoimmune encephalomyelitis(EAE) in order to explore the remyelination effect of BSYSC. Methods: The mice were randomly divided into normal control(NC), EAE model(EAE-M), prednisone acetate(PA, 6 mg/kg), BSYSC high-dose(3.02 g/kg) and BSYSC low-dose(1.51 g/kg) groups. The mice were induced by immunization with myelin oligodendrocyte glycoprotein(MOG) 35-55. The neurological function scores were assessed once daily. The pathological changes in mice brains were observed with hematoxylin-eosin(HE) staining and transmission electron microscope(TEM). The protein expressions of myelin basic protein(MBP), Olig1 and Olig2 in brains were measured by immunohistochemistry. The m RNA expressions of Olig1 and Olig 2 was also determined by quantitative real-time polymerase chain reaction. Results: Compared with the EAE-M mice,(1) the neurological function scores were significantly decreased in BSYSC-treated mice on days 22 to 40(P〈0.01);(2) the inflammatory cells and demyelination in brains were reduced in BSYSC-treated EAE mice;(3) the protein expression of MBP was markedly increased in BSYSC-treated groups on day 18 and 40 respectively(P〈0.05 or P〈0.01);(4) the protein expression of Olig1 was increased in BSYSC(3.02 g/kg)-treated EAE mice on day 40(P〈0.01). Protein and m RNA expression of Olig2 was increased in BSYSC-treated EAE mice on day 18 and 40(P〈0.01). Over past decades plant tissue culture has emerged as an alternative of whole plant cultivation in the production of valuable secondary metabolites.Adventitious roots culture of Panax ginseng and Echinacea purpure has...Over past decades plant tissue culture has emerged as an alternative of whole plant cultivation in the production of valuable secondary metabolites.Adventitious roots culture of Panax ginseng and Echinacea purpure has reached the scale of 1-10 kL.Some molecular biological techniques,such as transgenic technology and genetic stability are increasingly used in the studies on plant tissue cultures.The studies on elicitors have deepened into the induction mechanism,including signal molecules,functional genes,and so on.More and more biological elicitors,such as A.niger and yeast are used to increase the active compounds in plant tissue cultures.We also discussed the application of synthetic biology in the studies on biosynthesis of artemisinin,paclitaxel,and tanshinon.The studies on active ingredients biosynthesis of medicinal plants provide unprecedented possibilities to achieve mass production of active ingredients.Plant tissue cultures can not only produce active ingredients but also as experimental materials for biosynthesis.In order to improve the contents of active compounds in medicinal plants,following aspects could be carried out gene interference or gene silencing,gene overexpression,combination with chemical synthesis,application of elicitors,and site-directed mutagenesis of the key enzymes.展开更多
Objective:To determine the impact of adjunctive Buchang Naoxintong Capsule(步心脑心通胶囊,NXT) on dual antiplatelet therapy in patients with cytochrome P450 2C19*2(CYP2C19*2) polymorphism undergoing percutaneou...Objective:To determine the impact of adjunctive Buchang Naoxintong Capsule(步心脑心通胶囊,NXT) on dual antiplatelet therapy in patients with cytochrome P450 2C19*2(CYP2C19*2) polymorphism undergoing percutaneous coronary intervention(PCI).Methods:Ninety patients with CYP2C19*2 polymorphism were enrolled,and their genotypes were confirmed by polymerase chain reaction(PCR).The patients were randomly assigned to receive either adjunctive NXT(triple group,45 cases) or dual antiplatelet therapy(dual group,45 cases) using a computer-generated randomization sequence and sealed envelopes.Platelet function was assessed at baseline and 7 days after treatment with conventional aggregometry.Subsequent major adverse cardiovascular events(MACE,including sudden cardiac arrest and acute coronary syndrome) were recorded during a 12-month followup.Results:Baseline platelet function measurements were similar in both groups.After 7 days,percent inhibitions of maximum platelet aggregation and late platelet aggregation were significantly greater in the triple versus dual group(42.3%±16.0%vs.20.8%±15.2%,P〈0.01,and 54.7%±18.3%vs.21.5%±29.2%,P〈0.01,respectively).During the 12-month follow-up,the rate of subsequent MACE(6/45) was significantly lower in the triple group compared with the dual group(14/45;P〈0.05).Conclusion:Adjunctive NXT to maintenance dose clopidogrel(75 g) could enhance the antiplatelet effect and decrease subsequent MACE in patients with the CYP2C19'2polymorphism undergoing PCI.展开更多
