Background Clostridium butyricum(CB)is a probiotic that can regulate intestinal microbial composition and improve meat quality.Rumen protected fat(RPF)has been shown to increase the dietary energy density and provide essential fatty acids. However,it is still unknown whether dietary supplementation with CB and RPF exerts beneficial effects on growth performance and nutritional value of goat meat. This study aimed to investigate the effects of dietary CB and RPF supplementation on growth performance, meat quality, oxidative stability, and meat nutritional value of finishing goats.Thirty-two goats(initial body weight,20.5±0.82 kg)were used in a completely randomized block design with a 2 RPF supplementation(0 vs.30 g/d)×2 CB supplementation(0 vs.1.0 g/d)factorial treatment arrangement. The experiment included a 14-d adaptation and 70-d data and sample collection period. The goats were fed a diet consisted of 400 g/kg peanut seedling and 600 g/kg corn-based concentrate(dry matter basis).Result Interaction between CB and RPF was rarely observed on the variables measured, except that shear force was reduced(P<0.05)by adding CB or RPF alone or their combination;the increased intramuscular fat(IMF)content with adding RPF was more pronounced(P<0.05)with CB than without CB addition. The pH24h(P=0.009),a*values(P=0.007),total antioxidant capacity(P=0.050),glutathione peroxidase activities(P=0.006),concentrations of 18:3(P<0.001),20:5(P=0.003)and total polyunsaturated fatty acids(P=0.048)were increased, whereas the L*values(P<0.001),shear force(P=0.050)and malondialdehyde content(P=0.044)were decreased by adding CB.Furthermore,CB supplementation increased essential amino acid(P=0.027),flavor amino acid(P=0.010)and total amino acid contents(P=0.024)as well as upregulated the expression of lipoprotein lipase(P=0.034)and peroxisome proliferator-activated receptorγ(PPARγ)(P=0.012),and downregulated the expression of stearoyl-CoA desaturase(SCD)(P=0.034).The RPF supplementation increased dry matter intake(P=0.005),averaged daily gain(trend,P=0.058),hot carcass weight(P=0.046),backfat thickness(P=0.006),concentrations of 16:0(P<0.001)and c9-18:1(P=0.002),and decreased the shear force(P<0.001),isoleucine(P=0.049)and lysine content(P=0.003)of meat.In addition, the expressions of acetyl-CoA carboxylase(P=0.003),fatty acid synthase(P=0.038),SCD(P<0.001)and PPARγ(P=0.022)were upregulated due to RPF supplementation, resulting in higher(P<0.001)content of IMF.Conclusions CB and RPF could be fed to goats for improving the growth performance, carcass traits and meat quality, and promote fat deposition by upregulating the expression of lipogenic genes of Longissimus thoracis muscle. 展开更多

Background Diets rich in starch have been shown to increase a risk of reducing milk fat content in dairy goats. While bile acids(BAs)have been used as a lipid emulsifier in monogastric and aquatic animals, their effect on ruminants is not well understood. This study aimed to investigate the impact of BAs supplementation on various aspects of dairy goat physiology, including milk composition, rumen fermentation, gut microbiota, and BA metabolism. Results We randomly divided eighteen healthy primiparity lactating dairy goats(days in milk=100±6 d)into two groups and supplemented them with 0 or 4 g/d of BAs undergoing 5 weeks of feeding on a starch-rich diet. The results showed that BAs supplementation positively influenced milk yield and improved the quality of fatty acids in goat milk.BAs supplementation led to a reduction in saturated fatty acids(C16:0)and an increase in monounsaturated fatty acids(cis-9 C18:1),resulting in a healthier milk fatty acid profile. We observed a significant increase in plasma total bile acid concentration while the proportion of rumen short-chain fatty acids was not affected.Furthermore,BAs supplementation induced significant changes in the composition of the gut microbiota, favoring the enrichment of specific bacterial groups and altering the balance of microbial populations.Correlation analysis revealed associations between specific bacterial groups(Bacillus and Christensenellaceae R-7 group)and BA types, suggesting a role for the gut microbiota in BA metabolism.Functional prediction analysis revealed notable changes in pathways associated with lipid metabolism, suggesting that BAs supplementation has the potential to modulate lipid-related processes. Conclusion These findings highlight the potential benefits of BAs supplementation in enhancing milk production, improving milk quality, and influencing metabolic pathways in dairy goats.Further research is warranted to elucidate the underlying mechanisms and explore the broader implications of these findings. 展开更多

Medical research on neurologic ailments requires representative animal models to validate treatments before they are translated to human clinical trials.Rodents are the predominant animal model used in neurological research despite limited anatomic and physiologic similarities to humans. As a result, functional testing designed to assess locomotor recovery after neurologic impairment is well established in rodent models.Comparatively, large r,more clinically relevant models have not been as well studied. To achieve similar locomotor testing standardization in larger animals, the models must be accessible to a wide array of researchers.Non-human primates are the most relevant animal model fo r translational research, however ethical and financial barriers limit their accessibility. This review focuses on swine, sheep, and goats as large animal alternatives for transitional studies between rodents and non-human primates. The objective of this review is to compare motor testing and data collection methods used in swine, sheep, and goats to encourage testing standardization in these larger animal models. The PubMed database was analyzed by searching combinations of swine, sheep, and goats, neurologic injuries, and functional assessments.Findings were categorized by animal model, data collection method, and assessment design.Swine and sheep were used in the majority of the studies, while only two studies were found using goats. The functional assessments included open pen analysis, treadmill walking, and guided free walking.Data collection methods included subjective behavioral rating scales and objective tools such as pressure-sensitive mats and image-based analysis software.Overall, swine and sheep were well-suited for a variety of assessment designs, with treadmill walking and guided free walking offering the most consistency across multiple trials.Data collection methods varied, but image-based gait analysis software provided the most robust analysis.Future studies should be conducted to standardize functional testing methods after neurologic impairment in large animals. 展开更多

Understanding how evolutionary pressures related to climate change have shaped the current genetic background of domestic animals is a fundamental pursuit of biology. Here, we generated wholegenome sequencing data from native goat populations in Iraq and Pakistan. Combined with previously published data on modern, ancient(Late Neolithic to Medieval periods), and wild Capra species worldwide, we explored the genetic population structure, ancestry components, and signatures of natural positive selection in native goat populations in Southwest Asia(SWA). Results revealed that the genetic structure of SWA goats was deeply influenced by gene flow from the eastern Mediterranean during the Chalcolithic period, which may reflect adaptation to gradual warming and aridity in the region. Furthermore, comparative genomic analysis revealed adaptive introgression of the KITLG locus from the Nubian ibex(C. nubiana) into African and SWA goats. The frequency of the selected allele at this locus was significantly higher among goat populations located near northeastern Africa. These results provide new insights into the genetic composition and history of goat populations in the SWA region. 展开更多

The Boer goat is one of the top meat breeds in modern animal husbandry and has attracted widespread attention for its unique growth performance. However, the genetic basis of muscle development in the Boer goat remains obscure. In this study,we identified specific structural variants in the Boer goat based on genome-wide selection signals and analyzed the basis of the molecular heredity of related candidate genes in muscle development.A total of9 959 autosomal copy number variations(CNVs) were identified through selection signal analysis in 127 goat genomes.Specifically,we confirmed that the highest signal CNV(HSV) was a chromosomal arrangement containing an approximately 1.11 Mb(CHIR17:60062304-61171840 bp) duplicated fragment inserted in reverse orientation and a 5 362 bp deleted region(CHIR17:60145940-60151302 bp) with overlapping genes(e.g.,ARHGAP10,NR3C2,EDNRA,PRMT9,and TMEM184C).The homozygous duplicated HSV genotype(+/+) was found in 96% of Boer goats but was not detected in Eurasian goats and was only detected in 4% of indigenous African goats. The expression network of three candidate genes(ARHGAP10,NR3C2,and EDNRA)regulating dose transcription was constructed by RNA sequencing. Results indicated that these genes were involved in the proliferation and differentiation of skeletal muscle satellite cells(SMSCs) and their overexpression significantly increased the expression of SAA3.The HSV of the Boer goat contributed to superior skeletal muscle growth via the dose effects of overlapping genes. 展开更多

Background:SCNT(somatic cell nuclear transfer)is of great significance to biological research and also to the livestock breeding. However, the survival rate of the SCNT cloned animals is relatively low compared to other transgenic methods. This indicates the potential epigenetic variations between them.DNA methylation is a key marker of mammalian epigenetics and its alterations will lead to phenotypic differences. In this study,ASMT(acetylserotonin-Omethyltransferase)ovarian overexpression transgenic goat was produced by using SCNT.To investigate whether there are epigenetic differences between cloned and WT(wild type)goats,WGBS(whole-genome bisulfite sequencing)was used to measure the whole-genome methylation of these animals. Results:It is observed that the different m Cp G sites are mainly present in the intergenic and intronic regions between cloned and WT animals, and their CG-type methylation sites are strongly correlated.DMR(differentially methylated region)lengths are located around 1000 bp,mainly distributed in the exonic, intergenic and intronic functional domains.A total of 56 and 36 DMGs(differentially methylated genes)were identified by GO and KEGG databases, respectively.Functional annotation showed that DMGs were enriched in biological-process,cellularcomponent,molecular-function and other signaling pathways.A total of 10 identical genes related to growth and development were identified in GO and KEGG databases. Conclusion:The differences in methylation genes among the tested animals have been identified.A total of 10 DMGs associated with growth and development were identified between cloned and WT animals. The results indicate that the differential patterns of DNA methylation between the cloned and WT goats are probably caused by the SCNT.These novel observations will help us to further identify the unveiled mechanisms of somatic cell cloning technology, particularly in goats. 展开更多

Background: Liver has important immune function during fetal development and after birth. However, the effect of maternal malnutrition on immune function of the fetal liver is rarely reported. In this study, twelve pregnant goats(Xiangdong black goat,at d 45 of gestation) were assigned to the control group(fed 100% of nutritional requirements) and the restriction group(fed 60% of the intake of the control group) during gestation from d 55 to100.Fetal goats were harvested at d 100 of gestation and immune indexes and amino acid profiles of the umbilical cord blood and liver Toll-like receptors(TLRs) signaling pathways were measured. Results: Maternal body weight in the restriction group was lower than the control group(P < 0.05).Maternal feed intake restriction decreased(P < 0.05) heart weight, heart index, alkaline phosphatase and serum amyloid protein A in the umbilical cord blood(UCB).Moreover, only histidine was decreased in the restricted group(P = 0.084),and there were no differences in other amino acids contents in the UCB between the two groups(P > 0.05).The TLR2 and TLR4 mRNA expression in the fetal liver in the restriction group was greater(P < 0.05) than that in the control group.Furthermore, the mRNA expression levels of myeloid differentiation primary response 88(MyD88),TNF receptor associated factor 6,nuclear factor kappa B subunit 1,NFKB inhibitor alpha,IFN-β,TGF-β,TNF-α and IL-1β in the restricted group were upregulated(P < 0.05),and the expression of TLR3(P = 0.099) tended to be higher in the restricted group. However, protein levels of TLR2,TLR4,IκBα,phosphorylated IκBα,phosphorylated IκBα/total IκBα,TRIF and MyD88 were not affected(P > 0.05) by maternal intake restriction. Conclusions: These results revealed that the restriction of maternal feed intake influenced the development of heart and hepatic protein synthesis at the acute phase of fetal goats and upregulated the mRNA expression of genes involved in MyD88-dependent signaling pathways and of target cytokines. 展开更多

Endometrial development is a complicated process involving numerous regulatory factors.Circular RNAs(circRNAs)have been known as a member of the naturally occurring non-coding RNA family, and are reportedly crucial for a variety of physiological processes. This study investigated the circRNA landscape of non-pregnant endometrium of dairy goats during estrus.Non-pregnant endometrial samples of goats at estrus day 5(Ed5)and estrus day 15(Ed15)were used to methodically analyze the circRNA landscape using strand-specific Ribo-Zero RNA-Seq.A total of 2331 differentially expressed(P<0.05)circRNAs(DEciRs)between Ed5 and Ed15 were discovered in the goat endometrium. It was found that Nipped-B-like(NIPBL)and calcium responsive transcription factor(CARF)may participate in the development of the endometrium by decreasing(P<0.05)the levels of their circRNA-transcript forms.Furthermore,Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses of DEciR host genes(hgDEciRs)revealed that tight junctions and GTPases may be involved in endometrial development during the estrus cycle.A total of 2331 DEciRs were discovered in the endometrium at Ed5 and Ed15.Based on GO and KEGG enrichment analyses,it could be inferred that tight junctions and GTPases are likely to play an important role in the development of goat endometrium during the estrus cycle. This circRNA study greatly enhances our knowledge of global trends in the development of non-pregnant endometrium during the estrus cycle in goats;these results help us to better understand the molecular regulation of endometrial development in dairy goats. 展开更多

Early diagnosis of pregnancy plays an important role to minimize reproductive losses in farm animals. There are several methods for pregnancy diagnosis like profiling of reproductive hormones(such as progesterone and estrone sulfate),but sometimes they provide false-positive results.Embryo specific pregnancy markers, which delineate the presence and viability of the embryo, are considered as perfect for pregnancy determination.Pregnancy-associated glycoproteins are distinguished as the best indicator for the determination of early pregnancy, fetal number, and birth weight of kids.Pregnancy-associated glycoproteins are structurally correlated to aspartic proteinase and are communicated in the external epithelial cell layer of the placenta. They have been found to share about half amino acid sequence identity with pepsinogen, pepsin, cathepsin D and E.Dislike different individuals from aspartic proteinase family, numerous pregnancy-associated glycoproteins appear to be latent compound as a result of amino acid substitutions in and around the catalytic site. This review is to discuss the scope and prospects of pregnancy-associated glycoproteins as a pregnancy marker in farm animals, more specifically in goats. 展开更多

Haemonchus contortus is a blood-sucking abomasal helminth of small ruminants responsible for major economic losses to small farmers worldwide. Widespread resistance to synthetic anthelminthics has stimulated a need for alternative strategies of parasite control, among which is the use of medicinal plants with natural anthelminthic properties. This study assessed in vitro the efficacy of infused and macerated aqueous extract as well ethanolic extract of Moringa oleifera against fresh eggs, embryonated eggs, L1 and L2 larvae of H. contortus. For this purpose, five different concentrations (0.625, 1.25, 2.5, 3.75 and 5 mg/ml) were prepared from dry extracts via serial dilutions with distilled water. Fresh eggs obtained from artificially infected goat feces were exposed to these different concentrations for 48 hours, while embryonated eggs and larvae were exposed for 6 and 24 hours respectively. Distilled water and 1.5% DMSO were used as negative control. The results were expressed in terms of mean inhibition percentage of egg embryonation, mean inhibition percentage of egg hatch and mean percentage of larval mortality. An overview of results revealed that ethanolic leaf extract of M. oleifera was most efficient on eggs by inhibiting 60.3% ± 8.2% and 92.8% ± 6.2% eggs embryonation at 3.75 and 5 mg/ml respectively with a significant difference (P 0.05), which contributed to obtaining the lowest LC50 value of 0.985 mg/ml. This extract also inhibited 99% ± 2% egg hatching of H. contortus at 5 mg/ml with an LC50 value of 1.7 mg/ml. Concerning activity on larvae, the ethanolic extract was also most potent against them by inducing 98.8% ± 2.5% and 100% ± 0% mortality of L1 and L2 larvae at 5 mg/ml respectively. Infused aqueous extract was more efficient on eggs than on larvae with an IC50 value less than 2 mg/ml and an LC50 value more than 3.5 mg/ml. Macerated aqueous extract showed good activity against the four developmental stages with LC50 values ranging from 2.08 mg/ml for L2 larvae to 2.92 mg/ml for L1 larvae and 2.37 to 2.52 mg/ml for embryonated and fresh eggs respectively. The current study showed that all three extracts of M. oleifera tested possessed potential ovicidal and larvicidal activities against H. contortus. However, further in vivo studies are necessary to validate the anthelminthic property of this plant. 展开更多

The use of Levamisole hydrochloride as an immunostimulant has been successful in the control of certain diseases such as Derzsy’s disease in chicken as well as in increasing lymphocytes and weight gain in pigs. However, its use as immunostimulant in the prevention of Peste des petits ruminants (PPR) in goats has not been investigated. In this study, the use of Levamisole in enhancing immune response to PPR vaccination including its effects on weight gain was investigated among groups of goats (groups A, B, C and D). Virus neutralization test was used to determine the antibody profile in vaccinated goats while the total and differential leucocyte counts and body mass index (BMI) were determined using standard procedures. Goats in group A (Levamisole primed and PPR vaccinated) seroconverted to more than 3-folds of the initial pre-vaccination geometric mean titre (GMT) of neutralizing antibody beginning from second week post-vaccination and remained high throughout the period of the experiment. Similarly, there was significant increase (p 0.05) in BMI of animals in other groups throughout the period of the experiment. The results of this study further confirm the immunostimulatory effect of Levamisole when used in combination with a vaccine. 展开更多

Neospora caninum (N. caninum), an intracellular protozoan parasite, causes abortion and neonatal mortality in many herbivores including sheep and goats. The aim of this study was to estimate the Seroprevalence of N. caninum in sheep and goats in Grenada, West Indies. Sera were collected from 138 sheep and 138 goats from all six parishes of Grenada and Carriacou, a small island within Grenada. Animals were identified by gender. Sera were tested for antibodies for N. caninum using an indirect Enzyme Linked Immunosorbant Assay (ELISA) multispecies Kit from Pourquire Laboratorie (IDvet, France). Antibodies to N. caninum were detected in 18 of 138 sheep (13%, 95% confidence interval (CI): 7.39% to 18.61%) and 8 of 138 goats (5.8%, 95% CI: 1.9% to 9.7%). Although seropositivity was high in female sheep and in male goats, there was no statistical difference in seropositivity of males and females in both species of animals (p > 0.05, χ2). The results of this study revealed that sheep and goats in Grenada were exposed to N. caninum and dogs might be considered as a source of infection. There was a need to educate the farmers, possessing small ruminants, about the transmission cycle of N. caninum and methods of prevention, to minimize the exposure of their flocks. This is the first report on seroprevalence of N. caninum in Grenada, West Indies. 展开更多

A new protocol to evaluate and improve Aardi goats' reproductive performance was implemented during summer and winter months. A total of 60 healthy mature Aardi goats were used (30 and 30 in the summer and winter, respectively). All were synchronized using an ovsynch protocol and then naturally mated (NM) to mature tested bucks. Conception rates and caPAG were measured and pregnancy was verified by ultrasonography. Results:Regardless of season, however, on day 23 and 35 post-NM, caPAGs were higher in sera of pregnant (P<0.05) than in non-pregnant goats, while there was no difference in caPAG concentrations on day 15. On day 23 and 35 post-NM, serum caPAG concentrations were higher (P<0.001) in the pregnant winter group than in the pregnant summer group, while no differences were found in non-pregnant goats. Measuring serum caPAG concentrations proved to be easy and accurate in assessing early stages of pregnancy (as early as 23 days post-NM) during both summer and winter seasons. Conclusions: It can be concluded that pregnancy rates were not affected by seasonal differences. The modified ovsynch program can be advantageous and worthwhile for its ease of use. When we add the accuracy of caPAG tests, such a program will help in detecting pregnancies in Aardi goats as early as 23 days post-NM, regardless of the season. 展开更多

The histological changes after tibial lengthening with metaphyseal osteotomywere observed in 30 goats. It was found that(1)a slow lengthening rate of 1 mm/d couldenable the tissues to adapt the mechanical traction and consequently increase thelengthening limitation;(2)the newly-formed bone in the lengthened area might be due to acombined osteogenesis of endochondral, intramembranous, and fibrous tissue ossification;and(3)the osteogenetic process could be distinguished into 3 stages as hemorrhage andorganization of the hematocele, callus formation, and remodelling of the newly-formedbones. 展开更多

A study was conducted in three cluster regions in Kenya where the Alpine,Toggenburg and Saanen dairy goat breeds, respectively, were kept. The objective was to determine the breeds’relative performance for use as a basis of their recommendation to farmers.Formal questionnaires were used to obtain information on farm sizes, dairy goat sources, reasons for keeping the dairy goats, goat milk production, amount of feed offered to the goats and the constraints faced.Further information on the actual milk production and live weights of the milking does was collected directly from the farms using hired recorders. Results indicated that the dairy goats were fed between 6 kg/goat/day and 17 kg/goat/day of forage. They had a low production average of 1.4 L milk/goat/day with no clear cut breed differences.Toggenburg milking does were significantly(p<0.05)heavier than the other breeds(48.3 vs.38.0 and 39.0 kg for the Alpine and Saanen dairy goats, respectively). The milk production and goat live weights were below their reported potential.Feed shortage in the dry season was a major constraint. It was concluded that farmers could keep any of the three breeds. It was recommended that data on the comparative performance from the three breeds be generated to guide on farmer choices.Governments receiving donor support on agricultural endeavours should put in place the technical and policy mechanisms to support the ventures after the donor exits, and also ensure recommendations by different donors on a technical issue are consistent and complementary. 展开更多

West African Dwarf (WAD) goats represent one of the predominant small ruminant breeds raised by resource-limited households in southern Nigeria as well as in the humid west and central Africa. This breed of goats is renowned for its adaptation to hot and humid environments, high fertility and prolificacy under backyard systems where they are raised almost with zero investment. On account of its contributions to household income and food security in southern Nigeria, there is the need for a detailed phenotypic and genetic characterization as well as the design of breeding strategies for its conservation through sustainable utilization. A critical challenge, however, is that there are no national breeding policies in most of the countries where WAD goats are raised. The objectives of this paper are: to review literature reports on trait-level information and estimates of genetic parameters for some economic characters of WAD goats;and to discuss a communal breeding scheme for its conservation through sustainable utilization. The presentation shows that WAD goats represent a highly diverse population, with particular reference to qualitative and performance-related traits, reflecting the role of traditional institutions in the maintenance of animal diversity in local populations. A genetic improvement programme involving dispersed open nucleus breeding scheme was discussed. The scheme involves backyard WAD goat units in villages as base population and dispersed communally managed schemes that maintain a population of bucks only, as nucleus units. Critical issues such as the definition of breeding goals by farmers, as well as communal project ownership, are emphasized. The ultimate goal of the breeding programme is to create opportunities for community-based management of WAD goat genetic resources for the benefit of backyard goat farmers in the region. 展开更多

The purpose of the study was to establish the structure of monitored animal farms and determine reproductive parameters of Sahelian goats in the Guera Region, in Central Chad. Data were collected through longitudinal survey in 27 observatory farms during three years using goats identified by numbered ear tags. The herds were composed on average of about 71% of females. The age pyramid covering all years showed a significant number of males aged between 0 - 1 year and perceptible fall thereafter. However, some stability was observed in the class of the 2 - 8 years old among females. The frequencies of accumulated kiddings showed that 44% of births occurred in the rainy season against 28% respectively in the cool dry and hot dry seasons. Obtained reproduction parameters were: a sex ratio of 0.51 in favor of males, a kidding interval of 10.6 months with significant differences (P < 0.05) according to the rank of birth, an age at first kidding of 16.5 months for females born in monitoring and an age of 20.3 months for all first-kidding, giving an apparent fertility rate of 145%, a fecundity rate of 116% and an abortion rate of 2%. The composition and structure of herds have highlighted the importance of females to ensure the reproducibility of goat production systems. The determined reproduction parameters are references for actions to improve goat rearing in this area. 展开更多

This article explores a technique for measuring the energy (NEL) value of feeds without respiration calorimetry or slaughter. The objectives were to compare results obtained from goats with those from cows, and to describe factors which limited the precision of these NEL estimates. One lactating Alpine doe and one lactateing Holstein cow were assigned to each of six different sequences of three 56-d feeding treatments consisting of low, medium, and high doses of the basal diet, rice bran, or hominy feed. This resulted in 30 observations of the basal diet and 12 of each byproduct feed for each species and utilized a total of 18 cows and 18 does. The NEL values of the basal diet, rice bran, and hominy feed were calculated as the sum of milk energy, change in body energy, and estimated fasting heat production per kilogram of feed dry matter. Milk energy was determined by bomb calorimetry and body energy from live body weight and deuterium oxide space. The NEL (MJ/kg) determined in this manner were basal diet, 5.73 and 5.98;rice bran, 7.11 and 7.07;and hominy feed, 6.99 and 8.20 for cows and goats, respectively. 展开更多
