This paper takes the Lanxi ancient levee as a research object to study the evolution laws of its seepage, displacement and stability before and after reinforcement with the upside-down hanging wells and grouting curtain through numerical simulation methods combined with experiments and observations. The study results indicate that the filled soil is less affected by water level fluctuations and groundwater concentration after reinforcement. A high groundwater level is detrimental to the levee's long-term stability, and the drainage issues need to be fully considered. The deformation of the reinforced levee is effectively controlled since the fill deformation is mainly borne by the upside-down hanging wells. The safety factors of the levee before reinforcement vary significantly with the water level. The minimum value of the safety factors is 0.886 during the water level decreasing period, indicating a very high risk of the instability. While it reached 1.478 after reinforcement, the stability of the ancient levee is improved by a large margin.展开更多

The geometry of hanging drop may play a critical role on the formation of cell spheroids, which, however, has not been explored. In this study, we developed a modified hanging-drop platform that enables the production of cell spheroids in a high-throughput manner by controlling hanging drop geometry with defined spreading ring. The surface tension force is proportional to the spreading ring and gravitational force is determined by the droplet volume, and the geometry can be determined by the balance between surface tension and gravity.β-TC-6 cell spheroids with optimized diameters were fabricated as 3D in vitro islet models. The models show morphology similar to primary islets and have functionality that more closely resembles primary islets than two-dimensional cell culture. The developed platform holds great potential for engineering well-controlled in vitro tissue models for various applications such as physiological and pathological studies, drug screening, as well as transplantation for treatment purpose.展开更多

Drms can be used to capture the general proximity of a site to a dipping fault plane comparing with the rupture distance Drup and the seismogenic distance Dseis. Then, using Drup, Dseis and Drms, the hanging wall/footwall effects on the peak ground acceleration (PGA) during the 1999 Chi-Chi earthquake are evaluated by regression analysis. The logarithm residual shows that the PGA on hanging wall is much greater than that on footwall at the same Drup or Dseis when the Drup or Dseis is used as site-to-source distance measure. In contrast, there is no significant difference between the PGA on hanging wall and that on footwall at the same Drms when Drms is used. This result confirms that the hanging wall/footwall effect is mainly a geometric effect caused by the asymmetry of dipping fault. Therefore, the hanging wall/footwall effect on the near-fault ground motions can be ignored in the future attenuation analysis if the root-mean-square distance Drms is used as the site-to-source distance measure.展开更多

Under the condition that the flat is sub-horizontal, the hanging wall syncline is separated from the half graben by the central dome. And on the dome forms an erosional surface. Both sediments in the half graben and erosional surface on the top of the central dome extended over the dome and entered into the hanging wall syncline with extension going on. Meanwhile, those having entered were overlapped by new sedimentary layers in the hanging wall syncline, so that there is a together-threaded, diachronic unconformity to form in the same epoch stratum. The layers in the hanging wall syncline also have an attribute of migrating laterally and getting tilted with extension. There is no sedimentation on the central dome. But sediments, which came from the half graben, got thicker over the dome in extension.展开更多

文摘Microfluidics is becoming a technology of growing interest for building microphysiological systems with integrated read-out functionalities. Here we present the integration of enzyme-based multi-analyte biosensors into a multi-tissue culture platform for‘body-on-a-chip’applications.The microfluidic platform is based on the technology of hanging-drop networks, which is designed for the formation, cultivation, and analysis of fluidically interconnected organotypic spherical three-dimensional(3D)microtissues of multiple cell types. The sensor modules were designed as small glass plug-ins featuring four platinum working electrodes,a platinum counter electrode, and an Ag/AgCl reference electrode. They were placed directly into the ceiling substrate from which the hanging drops that host the spheroid cultures are suspended. The electrodes were functionalized with oxidase enzymes to enable continuous monitoring of lactate and glucose through amperometry. The biosensors featured high sensitivities of 322±41 nA mM^(−1) mm^(−2) for glucose and 443±37 nA mM^(−1) mm^(−2) for lactate;the corresponding limits of detection were below 10μM.The proposed technology enabled tissue-size-dependent,real-time detection of lactate secretion from single human colon cancer microtissues cultured in the hanging drops.Furthermore, glucose consumption and lactate secretion were monitored in parallel, and the impact of different culture conditions on the metabolism of cancer microtissues was recorded in real-time.

文摘As 3D in vitro tissue models become more pervasive, their built-in nutrient, metabolite, compound, and waste gradients increase biological relevance at the cost of analysis simplicity.Investigating these gradients and the resulting metabolic heterogeneity requires invasive and time-consuming methods. An alternative is using electrochemical biosensors and measuring concentrations around the tissue model to obtain size-dependent metabolism data. With our hanging-dropintegrated enzymatic glucose biosensors,we conducted current measurements within hanging-drop compartments hosting spheroids formed from the human colorectal carcinoma cell line HCT116.We developed a physics-based mathematical model of analyte consumption and transport,considering(1)diffusion and enzymatic conversion of glucose to form hydrogen peroxide(H_(2)O_(2))by the glucose-oxidase-based hydrogel functionalization of our biosensors at the microscale;(2)H_(2)O_(2)oxidation at the electrode surface, leading to amperometric H_(2)O_(2)readout;(3)glucose diffusion and glucose consumption by cancer cells in a spherical tissue model at the microscale;(4)glucose and H_(2)O_(2)transport in our hangingdrop compartments at the macroscale;and(5)solvent evaporation, leading to glucose and H_(2)O_(2)upconcentration.Our model relates the measured currents to the glucose concentrations generating the currents. The low limit of detection of our biosensors(0.4±0.1μM),combined with our current-fitting method, enabled us to reveal glucose dynamics within our system.By measuring glucose dynamics in hanging-drop compartments populated by cancer spheroids of various sizes,we could infer glucose distributions within the spheroid, which will help translate in vitro 3D tissue model results to in vivo.
文摘Adaptive higher-order finite element methods(hp-FEM)are well known for their potential of exceptionally fast(exponential)convergence. However, most hp-FEM codes remain in an academic setting due to an extreme algorithmic complexity of hp-adaptivity algorithms. This paper aims at simplifying hpadaptivity for H(curl)-conforming approximations by presenting a novel technique of arbitrary-level hanging nodes. The technique is described and it is demonstrated numerically that it makes adaptive hp-FEM more efficient compared to hp-FEM on regular meshes and meshes with one-level hanging nodes.
文摘Hanging wall syncline and central dome are special extension structures, developing over the hanging wall in an extensional ramp-flat fault. But sediments, which came from the half graben, got thicker over the dome in extension.

文摘Mechanical damage induced by vibration during harvesting and post-harvest handling could decrease the quality, quantity, and shelf life of the fresh grape cluster.Usually, fresh grape clusters are harvested by gripping and cutting from the main rachis in the present robotic harvesting system, then transported towards the basket during post-harvest handling. However, serious cluster vibration and corresponding berry falling may occur during the robotic transportation of hanging grape clusters. Therefore, this study was designed to perform experimental and theoretical hanging force analysis to explore the vibration mechanism of hanging grape clusters during robotic transportation.A lead screw lathe with an attached linear actuator was used to investigate the effects of four different speeds(0.4,0.6,0.8,1.0 m/s)with four acceleration levels(6,8,10,12 m/s2)on the vibration of the hanging grape cluster.By the experiments, the peak hanging force of the grape cluster at the start, constant speed, and stop phase of the actuator was recorded using a single axis force sensor, and the cluster’s swing angle was measured with a digital camera. The experimental results showed a linear relationship between the swing angle and hanging force of the cluster at the start and stop phase of the actuator. The multi-stage cluster’s vibration during robotic transportation was observed, and the behavior of cycled damping after a sudden stop of the actuator was found. The simulated results of hanging force of grape cluster in damping phase were agreed with experimental results with R2 more than 0.90 at an optimum acceleration of 10 m/s2.To conclude, this research provides theoretical basics for understanding the complex vibration mechanism of the hanging cluster fruits during speedy robotic transportation operations with low-loss of berry drop both on industrial and farm levels.
文摘Violence against the neck can result in a range of macromorphological and micromorphological findings. However, the forensic relevance of the carotid sinus in cases of violence against the neck remains controversial. In this follow-up study of 22 cases of suicidal and accidental strangulations, carotid bifurcations were examined histologically for morphological changes implying direct trauma, including haemorrhage and immunohistochemical expression of heat-shock proteins 27,60,and 70 and aquaporin-3.These cases were compared with a control group(82 cases)without neck compression or head trauma and with variable causes of death. No relevant histopathological findings implying direct trauma of the carotid bifurcation were found. No cases showed positive aquaporin-3 staining and only five cases showed positive heat-shock protein-27 staining, all of which were hangings.Without massive trauma of the carotid bifurcation, histological alterations cannot be expected.Without signs of rapid death, findings of acute circulatory failure, macromorphological and micromorphological findings of neck compression, and reliable markers indicating relevant impact on the carotid bifurcation the diagnosis of a lethal reflex cannot be verified.
