The clinical role of increase of serum matrix metalloproteinase-8 concentration in patients with acute coronary syndrome

Objective To study the clinical role of the variation of serum matrix metalloproteinase-8 (MMP-8) concentration in patients with acute coronary syndrome (ACS). Methods ELISA method was adopted to detect serum MMP-8 concentration and to observe concentration’s differences and features among 80 selected ACS cases (43 acute myocardial infarction and 37 unstable angina pectoris), 43 stable angina pectoris (SAP) cases and 37 control cases. And meanwhile the atherosclerosis risk factors of each case, such as age, sex, hypertension, body mass index, smoking, family history, diabetes, and hyperlipidemia were collected and analyzed as a whole. Results First, serum MMP-8 concentration reached the highest point in ACS, and there was significant difference between SAP and control groups (P<0.01). Second, serum MMP-8 in AMI was much higher than that in UAP with significant difference (P<0.01). There was no difference between UAP and SAP groups (P>0.05). Third, Logistic regression analysis revealed that serum MMP-8 concentration might be the indicator of ACS (B=4.493, P=0.000), particularly, that of AMI (B=9.961, P=0.000). Fourth, linear correlation and linear regression analysis found that only neutrophil was likely to influence serum MMP-8 concentration (r=0.274, P=0.001). Fifth, in the diagnosis of ACS, the area under ROC curve of MMP-8 was 0.785, the sensitivity and specificity were 68.6% and 76.5%, respectively. Conclusion ① Serum MMP-8 concentration has close relationship with the occurrence of ACS, particularly with AMI; ② Serum MMP-8 concentration may be one of the predicting indicators of ACS and particularly of AMI; ③ Neutrophil may be correlated with serum MMP-8 concentration; ④ MMP-8 is of somewhat valuable in diagnosing ACS. 展开更多

BACKGROUND Increasing data indicated that long noncoding RNAs(lncRNAs)were directly or indirectly involved in the occurrence and development of tumors,including hepatocellular carcinoma(HCC).Recent studies had found that the expression of lncRNA HAND2-AS1 was downregulated in HCC tissues,but its role in HCC progression is unclear.Ultrasound targeted microbubble destruction mediated gene transfection is a new method to overexpress genes.AIM To study the role of ultrasound microbubbles(UTMBs)mediated HAND2-AS1 in the progression of HCC,in order to provide a new reference for the treatment of HCC.METHODS In vitro,we transfected HAND2-AS1 siRNA into HepG2 cells by UTMBs,and detected cell proliferation,apoptosis,invasion and epithelial-mesenchymal transition(EMT)by cell counting kit-8 assay,flow cytometry,Transwell invasion assay and Western blotting,respectively.In addition,we transfected miR-837-5p mimic into UTMBs treated cells and observed the changes of cell behavior.Next,the UTMBs treated HepG2 cells were transfected together with miR-837-5p mimic and tissue inhibitor of matrix metalloproteinase-2(TIMP2)overexpression vector,and we detected cell proliferation,apoptosis,invasion and EMT.In vivo,we established a mouse model of subcutaneous transplantation of HepG2 cells and observed the effect of HAND2-AS1 silencing on tumor formation ability.RESULTS We found that UTMBs carrying HAND2-AS1 restricted cell proliferation,invasion,and EMT,encouraged apoptosis,and HAND2-AS1 silencing eliminated the effect of UTMBs.Additionally,miR-873-5p targets the gene HAND2-AS1,which also targets the 3’UTR of TIMP2.And miR-873-5p mimic counteracted the impact of HAND2-AS1.Further,miR-873-5p mimic solely or in combination with pcDNA-TIMP2 had been transformed into HepG2 cells exposed to UTMBs.We discovered that TIMP2 reversed the effect of miR-873-5p mimic caused by the blocked signalling cascade for matrix metalloproteinase(MMP)2/MMP9.In vivo results showed that HAND2-AS1 silencing significantly inhibited tumor formation in mice.CONCLUSION LncRNA HAND2-AS1 promotes TIMP2 expression by targeting miR-873-5p to inhibit HepG2 cell growth and delay HCC progression. 展开更多

Vascular endothelial growth factor and its mimic peptide KLTWQELYQLKYKGI(QK)are widely used as the most potent angiogenic factors for the treatment of multiple ischemic diseases.However,conventional topical drug delivery often results in a burst release of the drug,leading to transient retention(inefficacy)and undesirable diffusion(toxicity)in vivo.Therefore,a drug delivery system that responds to changes in the microenvironment of tissue regeneration and controls vascular endothelial growth factor release is crucial to improve the treatment of ischemic stroke.Matrix metalloproteinase-2(MMP-2)is gradually upregulated after cerebral ischemia.Herein,vascular endothelial growth factor mimic peptide QK was self-assembled with MMP-2-cleaved peptide PLGLAG(TIMP)and customizable peptide amphiphilic(PA)molecules to construct nanofiber hydrogel PA-TIMP-QK.PA-TIMP-QK was found to control the delivery of QK by MMP-2 upregulation after cerebral ischemia/reperfusion and had a similar biological activity with vascular endothelial growth factor in vitro.The results indicated that PA-TIMP-QK promoted neuronal survival,restored local blood circulation,reduced blood-brain barrier permeability,and restored motor function.These findings suggest that the self-assembling nanofiber hydrogel PA-TIMP-QK may provide an intelligent drug delivery system that responds to the microenvironment and promotes regeneration and repair after cerebral ischemia/reperfusion injury. 展开更多

Periodontitis is an inflammatory disease of the periodontium. Any imbalance between the matrix metalloproteinases (MMPs) secreted by neutrophils and tissue inhibitors initiates the destruction of collagen in gum tissue, leading to chronic periodontitis. This study aimed to correlate salivary levels of MMP-8 and periodontal parameters of chronic periodontitis to establish MMP-8 as a noninvasive marker for the early diagnosis of chronic periodontitis. The study involved 40 subjects visiting the periodontic OPD of Dr. Ziauddin Ahmad Dental College and Hospital, located in Aligarh, U.P., India, from 2011 to 2012. The subjects were divided into two groups: group I consisted of 20 periodontally healthy subjects (controls) while group II consisted of 20 patients with chronic periodontitis. Chronic periodontitis was assessed on the basis of several periodontal parameters, including pocket probing depth (PPD), clinical attachment level (CAL), gingival index (GI), and plaque index (PI). Around 3 ml of unstimulated and whole expectorated saliva was collected for MMP-8 estimation by ELISA using Quantikine human total MMP-8 immunoassay kits. Data were analyzed using STATISTICA (Windows version 6) software. Salivary MMP-8 levels of groups I and II were 190.91 ± 143.89 ng/ml and 348.26± 202.1 ng/ml, respectively. The MMP-8 levels and periodontal status (PPD, CAL, GI, and PI) of groups I and II showed positive and significant correlations (for PPD, r = 0.63, P 〈 0.001; for CAL, r = 0.54, P 〈 0.001; for GI, r = 0.49, P 〈 0.001; and for PI, r = 0.63, P 〈 0.001). The results of this study demonstrate elevated concentrations of MMP-8 in individuals with chronic periodontitis. 展开更多

目的 探究固定正畸矫治病人唾液基质金属蛋白酶8(MMP-8)、基质金属蛋白酶9(MMP-9)水平的变化及临床意义。方法 选取2019年6月至2021年12月于保定市第二医院接受固定正畸矫治成年病人80例及健康受试者40例为研究对象。对病人固定矫治前(... 目的 探究固定正畸矫治病人唾液基质金属蛋白酶8(MMP-8)、基质金属蛋白酶9(MMP-9)水平的变化及临床意义。方法 选取2019年6月至2021年12月于保定市第二医院接受固定正畸矫治成年病人80例及健康受试者40例为研究对象。对病人固定矫治前(T1)、固定矫治1周(T2)、固定矫治1个月(T3)及健康受试者探诊出血(BOP)、菌斑指数(PLI)进行检测;收集健康受试者及病人T1、T2、T3时期唾液,采用ELISA检测唾液中MMP-8、MMP-9水平;流式细胞术对唾液白细胞含量进行检测;分别对MMP-8、MMP-9与牙龈BOP相关性进行分析;分离唾液白细胞,检测白细胞MMP-8、MMP-9表达水平变化。结果T2、T3时病人BOP[(21.36±8.79)%、(13.06±5.80)%]、PLI[(2.53±0.43)分、(1.89±0.39)分]及唾液中MMP-8[(0.43±0.11)μg/L、(0.32±0.10)μg/L]、MMP-9[(3.64±0.76)μg/L、(2.02±0.50)μg/L]、白细胞水平[(17 893.71±505.49)个、(8 532.18±421.89)个],显著高于T1水平[(5.05±2.11)%、(0.71±0.25)分、(0.16±0.08)μg/L、(0.25±0.13)μg/L、(2 308.66±178.04)个,P<0.05];唾液MMP-8、MMP-9水平与牙龈BOP、PLI呈正相关;进一步发现T2、T3时白细胞MMP-8、MMP-9表达水平显著高于T1水平(P<0.05)。受试者操作特征(ROC)曲线显示唾液MMP-8及MMP-9水平能较好预测牙龈炎程度。结论 正畸病人唾液MMP-8、MMP-9水平与牙龈炎病情呈正相关,唾液MMP-8、MMP-9可作为评估正畸病人牙龈炎的潜在生物标志物。 展开更多

目的观察微种植体支抗用于骨性错牙合畸形患者的疗效及对血清基质金属蛋白酶8(MMP-8)、基质金属蛋白酶9(MMP-9)、一氧化氮(NO)、胰岛素样生长因子-1(IGF-1)水平的影响。方法前瞻性选取2022年2月至2023年12月在沧州市中心医院进行诊治的... 目的观察微种植体支抗用于骨性错牙合畸形患者的疗效及对血清基质金属蛋白酶8(MMP-8)、基质金属蛋白酶9(MMP-9)、一氧化氮(NO)、胰岛素样生长因子-1(IGF-1)水平的影响。方法前瞻性选取2022年2月至2023年12月在沧州市中心医院进行诊治的骨性错牙合畸形患者84例作为研究对象,按照随机数字表法将其分为观察组(n=42)和对照组(n=42)。观察组采用微种植体支抗治疗,对照组采用口外弓支抗治疗,统计两组治疗前和治疗6个月后的口腔结构、上下颌骨角度[上齿槽座角(SNA)、下齿槽座角(SNB)、上齿槽座点及鼻根点与下齿槽座点形成角(ANB)]、血清MMP-8、MMP-9、NO、IGF-1水平,记录两组美学满意度及并发症发生情况。结果在口腔结构方面,观察组磨牙移位为(3.21±0.84)mm,显著低于对照组[(5.32±1.11)mm],上中切牙凸距差、中切牙倾角差分别为(4.24±1.66)mm、(26.42±4.38)°,均显著高于对照组[(2.45±1.21)mm、(12.44±3.05)°],差异均有统计学意义(P<0.05)。治疗6个月后,观察组SNA、ANB分别为(72.01±2.19)°、(4.68±0.78)°,显著低于对照组[(76.49±2.21)°、(5.37±0.82)°],SNB为(78.65±2.36)°,显著高于对照组[(76.07±2.44)°],差异均有统计学意义(P<0.05)。治疗6个月后,观察组血清MMP-8、MMP-9、NO、IGF-1水平分别为(94.16±10.84)μmol/L、(74.49±9.87)pg/mL、(55.72±8.03)μmol/L、(643.85±26.32)ng/mL,显著低于对照组[(112.51±11.02)μmol/L、(86.52±10.01)pg/mL、(67.96±8.41)μmol/L、(703.36±26.96)ng/mL],差异均有统计学意义(P<0.05)。在美学满意度方面,观察组总满意度为92.86%,明显高于对照组(76.19%),差异有统计学意义(P<0.05)。在并发症方面,观察组总发生率为9.52%,明显低于对照组(30.95%),差异有统计学意义(P<0.05)。结论微种植体支抗治疗骨性错牙合畸形疗效较好,可以有效改善患者口腔结构和上下颌骨角度,调节血清MMP-8、MMP-9、NO、IGF-1水平,且美学满意度高,并发症少,值得临床推广应用。 展开更多

目的探究无托槽隐形矫治器与固定矫治器对口腔正畸治疗患者口腔微生物菌群和龈沟液可溶性细胞间黏附分子-1(sICAM-1)、基质金属蛋白酶-8(MMP-8)水平的影响。方法前瞻性纳入2020年2月至2023年3月在山西省儿童医院接受正畸治疗的106例患... 目的探究无托槽隐形矫治器与固定矫治器对口腔正畸治疗患者口腔微生物菌群和龈沟液可溶性细胞间黏附分子-1(sICAM-1)、基质金属蛋白酶-8(MMP-8)水平的影响。方法前瞻性纳入2020年2月至2023年3月在山西省儿童医院接受正畸治疗的106例患者为研究对象,按照随机数字表法对患者分为对照组和研究组,每组各53例。对照组采用固定矫治器,研究组采用无托槽隐形矫治器。比较两组患者矫正前和矫正后3个月口腔微生物菌群情况(卟啉单胞菌、具核梭杆菌、福塞斯坦氏菌和中间型普里沃菌)、牙周健康情况(牙龈指数、龈沟出血指数、菌斑指数及龈沟探诊深度)、龈沟液sICAM-1和MMP-8水平、患者满意度及并发症情况。结果矫正后3个月,两组各菌群水平均较矫正前升高,但研究组卟啉单胞菌、具核梭杆菌、福塞斯坦氏菌、中间型普里沃菌水平分别为0.29±0.06、0.33±0.06、0.33±0.08、0.27±0.06,均低于对照组(0.35±0.07、0.44±0.08、0.45±0.09、0.37±0.09),差异均有统计学意义(P<0.05)。两组组内矫正后3个月的牙龈指数和龈沟探诊深度与矫正前比较,以及组间矫正后3个月的牙龈指数和龈沟探诊深度比较,差异均无统计学意义(P>0.05);两组矫正后3个月的龈沟出血指数及菌斑指数均较矫正前升高,且研究组出血指数及菌斑指数分别为(0.65±0.06)、(0.99±0.12)分,均低于对照组[(0.72±0.08)、(1.23±0.15)分],差异均有统计学意义(P<0.05)。矫正后3个月,两组龈沟液sICAM-1水平均较矫正前升高,MMP-8水平均较矫正前降低,研究组龈沟液sICAM-1、MMP-8水平分别为(166.26±21.02)、(22.12±3.39)ng/mL,均低于对照组[(178.52±23.05)、(26.77±4.12)ng/mL],差异均有统计学意义(P<0.05)。研究组患者矫正后3个月的咀嚼、固定、美观、舒适性、方便程度、言语功能的满意度均高于对照组,差异均有统计学意义(P<0.05)。研究组患者牙龈红肿、牙龈萎缩、局部拥挤等总并发症发生率为7.55%,低于对照组(22.64%),差异有统计学意义(P<0.05)。结论相比固定矫治器矫正,采用无托槽隐形矫治器矫正对口腔微生物菌群和龈沟液中sICAM-1、MMP-8水平具有积极影响,能够提高患者的满意度,降低牙周疾病的发生。 展开更多

目的 检测慢性牙髓炎患者龈沟液白介素-6 (IL-6)、基质金属蛋白酶-8 (MMP-8)、基质金属蛋白酶抑制剂-1(TIMP-1)水平,并分析其与患者牙髓活力的关系。方法 选取106例慢性牙龈炎患者(观察组)和106例健康志愿者(对照组)。取龈沟液测定IL-6... 目的 检测慢性牙髓炎患者龈沟液白介素-6 (IL-6)、基质金属蛋白酶-8 (MMP-8)、基质金属蛋白酶抑制剂-1(TIMP-1)水平,并分析其与患者牙髓活力的关系。方法 选取106例慢性牙龈炎患者(观察组)和106例健康志愿者(对照组)。取龈沟液测定IL-6、MMP-8、TIMP-1水平,比较两组差异;比较观察组不同临床特征患者龈沟液IL-6、MMP-8、TIMP-1水平。牙髓活力电测仪测定牙髓活力,比较不同牙髓活力患者龈沟液IL-6、MMP-8、TIMP-1水平,Pearson法分析观察组牙髓电活力(EPT)值与龈沟液IL-6、MMP-8、TIMP-1水平的相关性。结果 观察组龈沟液IL-6、MMP-8均高于对照组[(61.28±8.11) ng/L vs.(38.15±7.04) ng/L、(33.04±6.02) ng/mL vs.(20.05±4.18) ng/mL](P<0.05),TIMP-1水平低于对照组[(1.35±0.30) ng/mL vs.(2.04±0.41) ng/mL](P<0.05);观察组病程≥12个月、合并糖尿病患者龈沟液IL-6、MMP-8水平均高于病程<12个月、未合并糖尿病患者(P<0.05),TIMP-1水平均低于病程<12个月、未合并糖尿病患者(P<0.05);中度、重度疼痛患者龈沟液IL-6、MMP-8水平均高于轻度疼痛(P<0.05),且重度疼痛均高于中度疼痛(P<0.05),而TIMP-1水平变化趋势正相反;牙髓活力下降、无活力患者龈沟液IL-6、MMP-8水平均高于活力正常(P<0.05),且牙髓无活力均高于活力下降(P<0.05),而TIMP-1水平变化趋势正相反;观察组患牙EPT与龈沟液IL-6、MMP-8水平均呈正相关(r=0.715、0.689, P<0.05),与TIMP-1水平呈负相关(r=-0.673, P<0.05)。结论 慢性牙龈炎患者龈沟液IL-6、MMP-8水平均升高,TIMP-1水平下降,且与病程、疼痛程度、合并糖尿病、牙髓活力均有关。 展开更多

目的探讨麝香保心丸联合阿托伐他汀治疗急性冠脉综合征(ACS)患者的疗效及对血脂、血清基质金属蛋白酶8(MMP-8)和基质金属蛋白酶组织抑制因子1(TIMP-1)的影响。方法160例ACS患者随机分为观察组(n=80)和对照组(n=80)。对照组给予阿托伐他... 目的探讨麝香保心丸联合阿托伐他汀治疗急性冠脉综合征(ACS)患者的疗效及对血脂、血清基质金属蛋白酶8(MMP-8)和基质金属蛋白酶组织抑制因子1(TIMP-1)的影响。方法160例ACS患者随机分为观察组(n=80)和对照组(n=80)。对照组给予阿托伐他汀治疗,观察组在对照组基础上给予麝香保心丸治疗。比较两组冠脉介入疗效,总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)及高密度脂蛋白胆固醇(HDL-C)等血脂指标水平,MMP-8、TIMP-1、C反应蛋白(CRP)和白细胞介素6(IL-6)等炎症因子水平,左心室收缩末期容积(LVESV)、左心室舒张末期容积(LVEDV)、左心室舒张末期容积指数(LVEDVI)及左心室射血分数(LVEF)等心功能指标水平,以及30 d主要心血管不良事件(MACE)发生情况。结果两组冠脉介入治疗总有效率比较,差异无统计学意义(χ^(2)=1.56,P>0.05);治疗后,观察组TC、TG、LDL-C水平均明显低于对照组,HDL-C水平明显高于对照组(t分别=2.30、2.03、2.57、-3.64,P均<0.05),观察组血脂达标率为43.33%,明显高于对照组血脂达标率25.00%(χ^(2)=4.48,P<0.05);治疗后,观察组MMP-8、CRP、IL-6水平及LVESV、LVEDV、LVEDVI均明显低于对照组,TIMP-1水平、LVEF明显高于对照组(t分别=2.29、2.19、8.55、2.17、2.29、2.37、-2.97、-2.40,P均<0.05);两组30 d MACE发生率比较,差异无统计学意义(χ^(2)=0.12,P>0.05)。结论麝香保心丸联合阿托伐他汀治疗ACS患者可有效改善患者血脂、心功能及降低炎症水平,利于稳定斑块。 展开更多

目的探讨血清白细胞介素8(IL-8)、基质金属蛋白酶2(MMP-2)水平在非小细胞肺癌(NSCLC)患者胸腔镜肺段切除术预后评估中的价值。方法选取101例行胸腔镜肺段切除术的NSCLC患者。收集所有患者的一般资料,检测其术前血清IL-8和MMP-2水平。对... 目的探讨血清白细胞介素8(IL-8)、基质金属蛋白酶2(MMP-2)水平在非小细胞肺癌(NSCLC)患者胸腔镜肺段切除术预后评估中的价值。方法选取101例行胸腔镜肺段切除术的NSCLC患者。收集所有患者的一般资料,检测其术前血清IL-8和MMP-2水平。对所有患者随访3年,根据是否出现复发分为复发组和未复发组。采用二元Logistic回归分析评估影响NSCLC患者复发的危险因素。采用受试者工作特征(ROC)曲线评价IL-8、MMP-2判断NSCLC患者复发的效能。采用Kaplan-Meier生存曲线和Log-rankχ^(2)检验评估NSCLC患者的复发情况。结果复发组术前血清IL-8、MMP-2水平显著高于未复发组(P<0.05)。以IL-8和MMP-2的均值分别作为临界值,将所有患者分为高表达组和低表达组。二元Logistic回归分析结果显示,术前血清IL-8≥87.52 ng/L和MMP-2≥477.86 ng/mL均是NSCLC患者复发的危险因素[比值比(OR)值分别为24.50、23.56,95%可信区间(CI)分别为18.94~37.23、18.01~38.29]。ROC曲线分析结果显示,术前IL-8和MMP-2判断NSCLC患者复发的曲线下面积(AUC)分别为0.834、0.858。Kaplan-Meier生存曲线分析结果显示,IL-8高表达组和MMP-2高表达组累积复发率分别显著高于IL-8低表达组和MMP-2低表达组(P<0.001)。结论术前血清IL-8、MMP-2水平可作为评估NSCLC患者胸腔镜肺段切除术预后的生物标志物。 展开更多

AIM:To evaluate the levels of preoperative serum matrix metalloproteinase-1 (MMP-1) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in gastric cancer.METHODS:One hundred gastric cancer patients who underwent gastrectomy were enrolled in this study.The serum concentrations of MMP-1 and TIMP-1 in these patients and in fifty healthy controls were determined using an enzyme-linked immunosorbent assay.RESULTS:Higher serum MMP-1 and TIMP-1 levels were observed in patients than in controls (P < 0.001).Serum MMP-1 and TIMP-1 levels were positively associated with morphological appearance,tumor size,depth of wall invasion,lymph node metastasis,liver metastasis,perineural invasion,and pathological stage.They were not significantly associated with age,gender,tumor location,or histological type.CONCLUSION:Increased MMP-1 and TIMP-1 were associated with gastric cancer.Although these markers are not good markers for diagnosis,these markers show in advanced gastric cancer. 展开更多

AIM: To evaluate the expression of matrix metalloproteinase-9 (MMP-9) and its clinical significance in esophageal squamous cell carcinoma (ESCC). METHODS: The expression of MMP-9 in 208 cases of ESCC was detected by immunohistochemistry (IHC) and its clinical significance in ESCC especially the relationship with the clinicopathological parameters was analyzed. RESULTS: The percentage of positive cases for MMP-9 detected by IHC was 49.0%. MMP-9 was mainly expressed in the cytoplasm of cancer cells especially in the invasive front. Only weak expression was detected in the stromal cells and no expression in non-cancerous mucosa. The expression of MMP-9 was positively correlated with poorer differentiation (P= 0.001<0.01), existence of vessel permeation (P= 0.027<0.05) and lymph node metastasis (P=0.027<0.05). CONCLUSION: The expression of MMP-9 correlates with the cancer cell differentiation, vessel permeation and lymph node metastasis. It may be a novel biomarker for the diagnosis and treatment of ESCC. 展开更多

AIM: To examine the expression of metalloproteinase-1 (MMP-1) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in the colonic mucosa of patients with ulcer- ative colitis (UC). METHODS: Reverse transcription-polymerase chain re- action (RT-PCR) and immunohistochemistry were used to study the expression of MMP-1 and TIMP-1 at both mRNA and protein levels in patients with UC and con- trols. The relationship between MMP-1 mRNA, TIMP-1 mRNA, MMP-1 mRNA/TIMP-1 mRNA ratio and the sever- ity of clinical symptoms of the patients with UC were also analyzed. RESULTS: The expression of MMP-1 mRNA and TIMP-1 mRNA in the ulcerated and inflamed colonic mucosa was signifi cantly higher than that in the non-inflamed colonic mucosa (P < 0.001), but there was no statistically signif i- cant difference in the non-inflamed colonic mucosa of UC patients and normal controls (P > 0.05). The mRNA ex- pression of MMP-1 and TIMP-1 in ulcerated colonic mu- cosa of UC patients was increased by 80-fold and 2.2-fold, respectively when compared with the normal controls. In the inflamed colonic mucosa, the increase was 30-fold and 1.6-fold, respectively. Immunohistochemical analy- sis showed that among the ulcerated, inflamed, and non-inflamed colonic mucosae of UC patients and the normal controls, the positive rate of MMP-1 expression was 87%, 87%, 40% and 35% respectively, and the positive rate of TIMP-1 expression was 89%, 89%, 80% and 75%, respectively. Furthermore, the expression of MMP-1 mRNA, TIMP-1 mRNA and the MMP-1 mRNA/ TIMP-1 mRNA ratio were correlated with the severity of clinical symptoms (P <0.05).CONCLUSION: Excessive expression of MMP-1 in the diseased colonic mucosa causes excessive hydrolysis of the extracellular matrix (ECM) and ulceration in UC pa-tients. MMP-1 mRNA, TIMP-1 mRNA and MMP-1 mRNA/ TIMP-1 mRNA ratio can be used as biomarkers to judge the severity of clinical symptoms in patients with UC. AIM:To investigate the inhibitory effects of RNA interference (RNAi) on expression of matrix metalloproteinase-2 (MMP-2) gene and invasiveness and adhesion of human pancreatic cancer cell line,BxPC-3.METHODS:RNAi was performed using the vector (pGPU6)-based small interference RNA (siRNA) plasmid gene silence system to specifically knock down MMP-2 expression in pancreatic cancer cell line,BxPC-3. Four groups of different specific target sequence in coding region of MMP-2 and one non-specific sequence were chosen to construct four experimental siRNA plasmids of pGPU6-1,pGPU6-2,pGPU6-3 and pGPU6-4,and one negative control siRNA plasmid of pGPU6 (-). MMP-2 expression was measured by reverse transcription polymerase chain reaction (RT-PCR) and Western blot. Cell proliferation and apoptosis were examined by methyl thiazolyl tetrazolium (MTT) and flow cytometry,respectively. The abilities of adhesion and invasion were detected by cell adhesion assay and cell invasion assay using Transwell chambers.RESULTS:The expression of MMP-2 was inhibited and the inhibitory effects of different sequence varied. pGPU6-1 group had the most efficient inhibitory effect,followed by pGPU6-2 and pGPU6-3 groups.Invasiveness and adhesion were more significantly reduced in pGPU6-1,pGPU6-2 and pGPU6-3 groups as compared with pGPU6 (-) and blank control groups. However,no difference concerning cell proliferation and apoptosis was observed after transfection between experiment groups and control groups.CONCLUSION:RNAi against MMP-2 successfully inhibited the mRNA and protein expression of MMP-2 in the pancreatic cancer cell line,BxPC-3,leading to a potent suppression of tumor cell adhesion and invasion without affecting cell proliferation and apoptosis. These findings suggest that the RNAi approach towards MMP-2 may be an effective therapeutic strategy for the clinical management of pancreatic tumor. 展开更多

To explore the role of the matrix metalloproteinase-9 (MMP-9) polymorphism in colorectal cancer (CRC) in a northeast Chinese population.METHODS: Genotyping of MNP-9-1562C〉T and 279R〉Q polymorphisms was carried out on blood samples from 137 colorectal cancer patients and 199 controls using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Multivariate logistic regression models were used to calculate adjusted odds ratios (OR) and 95% confidence intervals (95% CI).RESULTS: The distribution of IVllVlP-9 -2562C〉T and 279 R〉Q genotype was not significantly associated with the risk of CRC. However, the risk of Ilymph node metastasis of CRC was increased in patients with the -1562T allele (OR = 2.601; 95% CI = 1.160-5.835; P = 0.022). The frequency of MMP-9 279RR + RQ genotype was higher than the QQ genotype among CRC patients younger than sixty years old (OR = 0.102, 95% CI = 0.013-0.812; P = 0.012).CONCLUSION: Our results indicated that the MMP-9- 1562C〉T polymorphism affects lymph node metastasis of CRC. In addition, the MMP-9 279R allele may lead to a younger age of onset of colorectal cancer. 展开更多
