Sensitive detection and precise quantitation of trace-level crucial biomarkers in a complex sample matrix has become an important area of research.For example,the detection of high-sensitivity cardiac troponin I (hs-c...Sensitive detection and precise quantitation of trace-level crucial biomarkers in a complex sample matrix has become an important area of research.For example,the detection of high-sensitivity cardiac troponin I (hs-cTnI) is strongly recommended in clinical guidelines for early diagnosis of acute myocardial infarction.Based on the use of an electrode modified by single-walled carbon nanotubes (SWCNTs) and a Ru(bpy)32+-doped silica nanoparticle (Ru@SiO2)/tripropylamine (TPA) system,a novel type of electrochemiluminescent (ECL) magnetoimmunosensor is developed for ultrasensitive detection of hs-cTnI.In this approach,a large amount of[Ru(bpy)3]2+is loaded in SiO2(silica nanoparticles) as luminophores with high luminescent efficiency and SWCNTs as electrode surface modification material with excellent electrooxidation ability for TPA.Subsequently,a hierarchical micropillar array of microstructures is fabricated with a magnet placed at each end to efficiently confine a single layer of immunomagnetic microbeads on the surface of the electrode and enable 7.5-fold signal enhancement In particular,the use of transparent SWCNTs to modify a transparent ITO electrode provides a two-order-of-magnitude ECL signal amplification.A good linear calibration curve is developed for hs-cTnI concentrations over a wide range from 10 fg/ml to 10 ng/ml,with the limit of detection calculated as 8.720 fg/ml (S/N=3).This ultrasensitive immunosensor exhibits superior detection performance with remarkable stability,reproducibility,and selectivity.Satisfactory recoveries are obtained in the detection of hs-cTnI in human serum,providing a potentia analysis protocol for clinical applications.展开更多
DNA methylation has been extensively investigated in recent years,not least because of its known relationship with various diseases.Progress in analytical methods can greatly increase the relevance of DNA methylation ...DNA methylation has been extensively investigated in recent years,not least because of its known relationship with various diseases.Progress in analytical methods can greatly increase the relevance of DNA methylation studies to both clinical medicine and scientific research.Microflu-idic chips are excellent carriers for molecular analysis,and their use can provide improvements from multiple aspects.On-chip molecular analysis has received extensive attention owing to its advantages of portability,high throughput,low cost,and high efficiency.In recent years,the use of novel microfluidic chips for DNA methylation analysis has been widely reported and has shown obvious superiority to conventional methods.In this review,wefirst focus on DNA methylation and its applications.Then,we discuss advanced microfluidic-based methods for DNA methylation analysis and describe the great progress that has been made in recent years.Finally,we summarize the advantages that microfluidic technology brings to DNA methylation analysis and describe several challenges and perspectives for on-chip DNA methylation analysis.This review should help researchers improve their understanding and make progress in developing microfluidic-based methods for DNA methylation analysis.展开更多

In about a half century, a huge amount of data and knowledge has been accumulated from theoretical and experimental studies on micromixing. Nevertheless, those results are mostly composites of simplified theoretical and empirical models, and the true nature of interactions of flow inhomogeneity and micro-mixing with chemical reaction has not been fully unveiled. This article reviews the progress in micro-mixing study in chemical reactors to date. A few important topics related to the nature, experimental evaluation, and numerical simulation of micro-mixing are addressed.Some suggestions are given hopefully to motivate more chemical engineers to devote their efforts to better understanding of micro-mixing in chemical reactors.展开更多

The slip and no-slip ratio is not constant and varies irregularly, and viscosity is altered by changing the relaxation time in LBE equation. The slip boundary condition is simulated by specular reflection boundary and the no-slip boundary condition is simulated by bounce back boundary. It has been found that it is feasible to optimize the micro mixer design by combining the viscosity effect and boundary patterned ratio altogether.展开更多

To improve the performance of real-time recombinase polymerase amplification(RPA),a microfluidic system with active mixing is developed to optimize the reaction dynamics.Instead of adopting a single typical reaction c...To improve the performance of real-time recombinase polymerase amplification(RPA),a microfluidic system with active mixing is developed to optimize the reaction dynamics.Instead of adopting a single typical reaction chamber,a specific reactor including a relatively large chamber in center with two adjacent zig-zag channels at two sides is integrated into the microfluidic chip.Active mixing is achieved by driving the viscous reagent between the chamber and the channel back and forth periodically with an outside compact peristaltic pump.To avoid reagent evapora-tion,one end of the reactor is sealed with paraffin oil.A hand-held companion device is developed to facilitate real-time RPA amplification within 20 min.The whole area of the reactor is heated with a resistance heater to provide uniform reaction temperature.To achieve real-time monitoring,a compact fluorescence detection module is integrated into the hand-held device.A smartphone with custom application software is adopted to control the hand-held device and display the real-time fluorescence curves.The performances of two cases with and without active on-chip mixing are compared between each other by detecting African swine fever viruses.It has been demonstrated that,with active on-chip mixing,the amplification efficiency and detection sensitivity can be signifi-cantly improved.展开更多
We propose a nonlinear ultrasonic technique by using the mixed-frequency signals excited Lamb waves to conduct micro-crack detection in thin plate structures.Simulation models of three-dimensional(3D)aluminum plates a...We propose a nonlinear ultrasonic technique by using the mixed-frequency signals excited Lamb waves to conduct micro-crack detection in thin plate structures.Simulation models of three-dimensional(3D)aluminum plates and composite laminates are established by ABAQUS software,where the aluminum plate contains buried crack and composite laminates comprises cohesive element whose thickness is zero to simulate delamination damage.The interactions between the S0 mode Lamb wave and the buried micro-cracks of various dimensions are simulated by using the finite element method.Fourier frequency spectrum analysis is applied to the received time domain signal and fundamental frequency amplitudes,and sum and difference frequencies are extracted and simulated.Simulation results indicate that nonlinear Lamb waves have different sensitivities to various crack sizes.There is a positive correlation among crack length,height,and sum and difference frequency amplitudes for an aluminum plate,with both amplitudes decreasing as crack thickness increased,i.e.,nonlinear effect weakens as the micro-crack becomes thicker.The amplitudes of sum and difference frequency are positively correlated with the length and width of the zero-thickness cohesive element in the composite laminates.Furthermore,amplitude ratio change is investigated and it can be used as an effective tool to detect inner defects in thin 3D plates.展开更多

The micro-chip was composed of p...This paper described a highly sensitive chemiluminescence detection system for micro-chip electrophoresis (MCE) based on luminol-hydrogen peroxide reaction catalyzed by the metal ions. The micro-chip was composed of poly(dimethylsiloxane) (PDMS) and glass, and was fabricated by micro-machining technology. The surface of channels was dynamically modified by polydimethylacrylamide (PDMA) in order to eliminate unhomogeneous electroosmotic flow (EOF) of the PDMS/glass chip, adsorption of molecules, and improve hydrophobicity on PDMS surface. The detection modes, reagent mix procedures and reaction conditions were optimized and the detection limit of 5×10 mol/L for cobalt (II) was achieved by MCE with chemiluminescence detection, which was about four orders of magnitude more sensitive than that reported in the reference.展开更多

Under controlled conditions...We describe the microfabrication of ^85Rb vapour cells using a glass-silicon anodic bonding technique and in situ chemical reaction between rubidium chloride and barium azide to produce Rb. Under controlled conditions, the pure metallic Rb drops and buffer gases were obtained in the cells with a few mm^3 internal volumes during the cell sealing process. At an ambient temperature of 90 ℃ the optical absorption resonance of ^85Rb D1 transition with proper broadening and the corresponding coherent population trapping (CPT) resonance, with a signal contrast of 1.5% and linewidth of about 1.7 kHz, have been detected. The sealing quality and the stability of the cells have also been demonstrated experimentally by using the helium leaking detection and the after-9-month optoelectronics measurement which shows a similar CPT signal as its original status. In addition, the physics package of chip-scale atomic clock (CSAC) based on the cell was realized. The measured frequency stability of the physics package can reach to 2.1 × 10^-10 at one second when the cell was heated to 100 ℃ which proved that the cell has the quality to be used in portable and battery-operated devices.展开更多

A 3D acoustic Doppler velocimeter (micro ADV) was used to measure local flow velocities and Reynolds stress. The results show that hydraulic characteristics in non-vegetation and vegetation layers are totally different. In a region above the vegetation, Reynolds stress distribution is linear, and the measured velocity profile is a classical logarithmic one. Based on the concept of new-riverbed, the river compression parameter representing the impact of vegetation on river is given, and a new assumption of mixing length expression is made. The formula for time-averaged velocity derived from the expression requires less parameters and simple calculation, and is useful in applications.展开更多

The lightness and high strength-to-weight ratio of the magnesium alloy have attracted more interest in various applications.However,micro/nanostructure generation on their surfaces remains a challenge due to the flamm...The lightness and high strength-to-weight ratio of the magnesium alloy have attracted more interest in various applications.However,micro/nanostructure generation on their surfaces remains a challenge due to the flammability and ignition.Motivated by this,this study proposed a machining process,named the ultraprecision diamond surface texturing process,to machine the micro/nanostructures on magnesium alloy surfaces.Experimental results showed the various microstructures and sawtooth-shaped nanostructures were successfully generated on the AZ31B magnesium alloy surfaces,demonstrating the effectiveness of this proposed machining process.Furthermore,sawtooth-shaped nanostructures had the function of inducing the optical effect and generating different colors on workpiece surfaces.The colorful letter and colorful flower image were clearly viewed on magnesium alloy surfaces.The corresponding cutting force,chip morphology,and tool wear were systematically investigated to understand the machining mechanism of micro/nanostructures on magnesium alloy surfaces.The proposed machining process can further improve the performances of the magnesium alloy and extend its functions to other fields,such as optics.展开更多
The mixed forests of the upper Rio Negro at the northern of the Amazon basin grow in oxisol soils that are extremely infertile. These areas exhibit deficiencies in several macro-nutrients, and may also be characterize...The mixed forests of the upper Rio Negro at the northern of the Amazon basin grow in oxisol soils that are extremely infertile. These areas exhibit deficiencies in several macro-nutrients, and may also be characterized by the shortage or toxic excess of some micronutrients. The overall goal of this research is to collect more comprehensive information regarding the micronutrient composition of the upper Rio Negro forests as well as discern the relationship between leaf micro- and macro-nutrients that may contribute to the homeostasis and balance of the ionome. Firstly, the nutrient composition within the oxisol soil and leaf tissues of two top canopy tree species from the mixed forests was determined. We then analyzed the relationship between leaf micronutrient composition with N and P levels of the two species and that of species inhabiting the Amazon caatinga. Extractable soil Zn, B, Mn and Cu were very low in the mixed forest. In contrast, Fe and Al levels were potentially toxic. The analysis of leaf N/P ratios revealed for the first time the co-limitation of N and P in the mixed forest. This contrasts with species from the adjacent Amazon caatinga toposequence that are characterized by strong N limitation. All micronutrients within leaves of species inhabiting the mixed forest were also found to have low concentrations. Moreover, Fe and Al were detected at concentrations well below those reported for accumulator species. This suggested that leaf ion homeostasis was maintained under potentially toxic soil Fe and Al conditions. Leaf micronutrient (Fe, Zn and B) contents mirrored that of leaf N and P contents, and comparable Fe/N, Fe/P, Zn/N, Zn/P, B/N as well as B/P ratios were found across species and forest types. Therefore, forest species exhibited the capability to maintain leaf nutrient balances under soil conditions with deficient or toxic levels of micronutrients.展开更多

文摘We propose a nonlinear ultrasonic technique by using the mixed-frequency signals excited Lamb waves to conduct micro-crack detection in thin plate structures.Simulation models of three-dimensional(3D)aluminum plates and composite laminates are established by ABAQUS software,where the aluminum plate contains buried crack and composite laminates comprises cohesive element whose thickness is zero to simulate delamination damage.The interactions between the S0 mode Lamb wave and the buried micro-cracks of various dimensions are simulated by using the finite element method.Fourier frequency spectrum analysis is applied to the received time domain signal and fundamental frequency amplitudes,and sum and difference frequencies are extracted and simulated.Simulation results indicate that nonlinear Lamb waves have different sensitivities to various crack sizes.There is a positive correlation among crack length,height,and sum and difference frequency amplitudes for an aluminum plate,with both amplitudes decreasing as crack thickness increased,i.e.,nonlinear effect weakens as the micro-crack becomes thicker.The amplitudes of sum and difference frequency are positively correlated with the length and width of the zero-thickness cohesive element in the composite laminates.Furthermore,amplitude ratio change is investigated and it can be used as an effective tool to detect inner defects in thin 3D plates.
文摘This paper described a highly sensitive chemiluminescence detection system for micro-chip electrophoresis (MCE) based on luminol-hydrogen peroxide reaction catalyzed by the metal ions. The micro-chip was composed of poly(dimethylsiloxane) (PDMS) and glass, and was fabricated by micro-machining technology. The surface of channels was dynamically modified by polydimethylacrylamide (PDMA) in order to eliminate unhomogeneous electroosmotic flow (EOF) of the PDMS/glass chip, adsorption of molecules, and improve hydrophobicity on PDMS surface. The detection modes, reagent mix procedures and reaction conditions were optimized and the detection limit of 5×10 mol/L for cobalt (II) was achieved by MCE with chemiluminescence detection, which was about four orders of magnitude more sensitive than that reported in the reference.

文摘We describe the microfabrication of ^85Rb vapour cells using a glass-silicon anodic bonding technique and in situ chemical reaction between rubidium chloride and barium azide to produce Rb. Under controlled conditions, the pure metallic Rb drops and buffer gases were obtained in the cells with a few mm^3 internal volumes during the cell sealing process. At an ambient temperature of 90 ℃ the optical absorption resonance of ^85Rb D1 transition with proper broadening and the corresponding coherent population trapping (CPT) resonance, with a signal contrast of 1.5% and linewidth of about 1.7 kHz, have been detected. The sealing quality and the stability of the cells have also been demonstrated experimentally by using the helium leaking detection and the after-9-month optoelectronics measurement which shows a similar CPT signal as its original status. In addition, the physics package of chip-scale atomic clock (CSAC) based on the cell was realized. The measured frequency stability of the physics package can reach to 2.1 × 10^-10 at one second when the cell was heated to 100 ℃ which proved that the cell has the quality to be used in portable and battery-operated devices.

文摘By choosing a PVC slice to simulate flexible vegetation, we carried out experiments in an open channel with submerged flexible vegetation. A 3D acoustic Doppler velocimeter (micro ADV) was used to measure local flow velocities and Reynolds stress. The results show that hydraulic characteristics in non-vegetation and vegetation layers are totally different. In a region above the vegetation, Reynolds stress distribution is linear, and the measured velocity profile is a classical logarithmic one. Based on the concept of new-riverbed, the river compression parameter representing the impact of vegetation on river is given, and a new assumption of mixing length expression is made. The formula for time-averaged velocity derived from the expression requires less parameters and simple calculation, and is useful in applications.

文摘The lightness and high strength-to-weight ratio of the magnesium alloy have attracted more interest in various applications.However,micro/nanostructure generation on their surfaces remains a challenge due to the flammability and ignition.Motivated by this,this study proposed a machining process,named the ultraprecision diamond surface texturing process,to machine the micro/nanostructures on magnesium alloy surfaces.Experimental results showed the various microstructures and sawtooth-shaped nanostructures were successfully generated on the AZ31B magnesium alloy surfaces,demonstrating the effectiveness of this proposed machining process.Furthermore,sawtooth-shaped nanostructures had the function of inducing the optical effect and generating different colors on workpiece surfaces.The colorful letter and colorful flower image were clearly viewed on magnesium alloy surfaces.The corresponding cutting force,chip morphology,and tool wear were systematically investigated to understand the machining mechanism of micro/nanostructures on magnesium alloy surfaces.The proposed machining process can further improve the performances of the magnesium alloy and extend its functions to other fields,such as optics.
文摘The mixed forests of the upper Rio Negro at the northern of the Amazon basin grow in oxisol soils that are extremely infertile. These areas exhibit deficiencies in several macro-nutrients, and may also be characterized by the shortage or toxic excess of some micronutrients. The overall goal of this research is to collect more comprehensive information regarding the micronutrient composition of the upper Rio Negro forests as well as discern the relationship between leaf micro- and macro-nutrients that may contribute to the homeostasis and balance of the ionome. Firstly, the nutrient composition within the oxisol soil and leaf tissues of two top canopy tree species from the mixed forests was determined. We then analyzed the relationship between leaf micronutrient composition with N and P levels of the two species and that of species inhabiting the Amazon caatinga. Extractable soil Zn, B, Mn and Cu were very low in the mixed forest. In contrast, Fe and Al levels were potentially toxic. The analysis of leaf N/P ratios revealed for the first time the co-limitation of N and P in the mixed forest. This contrasts with species from the adjacent Amazon caatinga toposequence that are characterized by strong N limitation. All micronutrients within leaves of species inhabiting the mixed forest were also found to have low concentrations. Moreover, Fe and Al were detected at concentrations well below those reported for accumulator species. This suggested that leaf ion homeostasis was maintained under potentially toxic soil Fe and Al conditions. Leaf micronutrient (Fe, Zn and B) contents mirrored that of leaf N and P contents, and comparable Fe/N, Fe/P, Zn/N, Zn/P, B/N as well as B/P ratios were found across species and forest types. Therefore, forest species exhibited the capability to maintain leaf nutrient balances under soil conditions with deficient or toxic levels of micronutrients.
