Lotus(Nelumbo nucifera),an ancient aquatic plant,possesses a unique pharmacological activity that is primarily contributed by benzylisoquinoline alkaloids(BIAs).However,only few genes and enzymes involved in BIA biosy...Lotus(Nelumbo nucifera),an ancient aquatic plant,possesses a unique pharmacological activity that is primarily contributed by benzylisoquinoline alkaloids(BIAs).However,only few genes and enzymes involved in BIA biosynthesis in N.nucifera have been isolated and characterized.In the present study we identified the regiopromiscuity of an O-methyltransferase,designated NnOMT6,isolated from N.nucifera;NnOMT6 was found to catalyze the methylation of monobenzylisoquinoline 6-O/7-O,aporphine skeleton 6-O,phenylpropanoid 3-O,and protoberberine 2-O.We further probed the key residues affecting NnOMT6 activity via molecular docking and molecular dynamics simulation.Verification using site-directed mutagenesis revealed that residues D316,N130,L135,N176A,D269,and E328 were critical for BIA O-methyltransferase activities;furthermore,N323A,a mutant of NnOMT6,demonstrated a substantial increase in catalytic efficiency for BIAs and a broader acceptor scope compared with wild-type NnOMT6.To the best of our knowledge,this is the first study to report the O-methyltransferase activity of an aporphine skeleton without benzyl moiety substitutions in N.nucifera.The study findings provide biocatalysts for the semisynthesis of related medical compounds and give insights into protein engineering to strengthen O-methyltransferase activity in plants.展开更多
A new method for screening and identification 420 pesticide residues in fruits and vegetables by ultra-performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry(UPLC-Q-TOF/MS)were deve...A new method for screening and identification 420 pesticide residues in fruits and vegetables by ultra-performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry(UPLC-Q-TOF/MS)were developed.The samples were extracted with acetonitrile/acetic acid(99:1,V/V),and clean-up by SinChERS-Nano(single-step,cheap,effective,rugged,safe,nano)column,determined by UPLC-Q-TOF/MS.The accurate mass database and MS/MS database which contains 420 pesticides were established,the automatic retrieval of detection results was carried on according to the accurate mass,retention time,isotope ratio,ion fragment information,and so on.Method verification was performed on leeks samples.The results showed that 420 pesticides had good linearity in the range of 0.1-100μg/L,and the correlation coeffificients(R2)was greater than 0.990.The limits of detections(LODs)and limits of quantifications(LOQs)of 420 pesticides were in range of 0.05-2.0 and 0.1-5.0μg/L,respectively.The average spike recoveries at 3 levels were 70.1%to 119.7%,and the relative standard deviations(RSD)were lower than 20%(n=6).With this method,a survey of pesticide residues was conducted for 110 samples of 10 different fruits and vegetables,which provided scientific data for ensuring pesticide residue safety of the fruits and vegetables consumed daily by the public.This method was simple,sensitive and accurate,and could be used for rapid screening of 420 pesticide residues in fruits and vegetables.展开更多

[Methods] The samples were extracted with 80% acetonitrile water(containing 0.1% formic acid), combined with QuEChERS extraction technology and C18 and PSA purification, analyzed by high performance liquid chromatography-mass spectrometry, and quantified by external standard method. The target substances were analyzed on ZORBAX Eclipse C18 chromatographic column using 0.2% formic acid water and 0.2% methanol as mobile phases. The gradient elution mode was used for chromatographic separation and multiple reaction detection. [Results] In the linear range of 0.5-50.0 ng/ml, the linear relationship of the 54 kinds of veterinary drug residues was good, with correlation coefficients(r~2) greater than 0.995, and the detection limits ranged from 0.30 to 1.00 μg/kg. The results showed that the recovery ranged from 75.4% to 118.2% when different matrixes were added for recovery. [Conclusions] This method is simple, efficient, accurate, stable, and highly operable. It is applicable to simultaneous batch screening of veterinary drug residues in animal-derived food, and has high practical application value.展开更多
The tea plant(Camellia sinensis)is rich in polyphenolic compounds.Particularly,flavan-3-ols and proanthocyanidins(PAs)are essential for the flavor and disease-resistance property of tea leaves.The fifth subgroup of R2...The tea plant(Camellia sinensis)is rich in polyphenolic compounds.Particularly,flavan-3-ols and proanthocyanidins(PAs)are essential for the flavor and disease-resistance property of tea leaves.The fifth subgroup of R2R3-MYB transcription factors comprises the primary activators of PA biosynthesis.This study showed that subgroup 5 R2R3-MYBs in tea plants contained at least nine genes belonging to the TT2,MYB5,and MYBPA types.Tannin-rich plants showed an expansion in the number of subgroup 5 R2R3-MYB genes compared with other dicotyledonous and monocot plants.The MYBPA-type genes of tea plant were slightly expanded.qRT–PCR analysis and GUS staining analysis of promoter activity under a series of treatments revealed the differential responses of CsMYB5s to biotic and abiotic stresses.In particular,CsMYB5a,CsMYB5b,and CsMYB5e responded to high-intensity light,high temperature,MeJA,and mechanical wounding,whereas CsMYB5f and CsMYB5g were only induced by wounding.Three genetic transformation systems(C.sinensis,Nicotiana tabacum,and Arabidopsis thaliana)were used to verify the biological function of CsMYB5s.The results show that CsMYB5a,CsMYB5b,and CsMYB5e could promote the gene expression of CsLAR and CsANR.However,CsMYB5f and CsMYB5g could only upregulate the gene expression of CsLAR but not CsANR.A series of site-directed mutation and domain-swapping experiments were used to verify functional domains and key amino acids of CsMYB5s responsible for the regulation of PA biosynthesis.This study aimed to provide insight into the induced expression and functional diversity model of PA biosynthesis regulation in tea plants.展开更多

[Methods] Sixty eight kinds of pesticide residues in 415 Chinese chive samples collected from Tangshan area were qualitatively and quantitatively determined by high-performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS) and gas chromatography(GC) in 2020. [Results] The results showed that 41 kinds of pesticide residues were detected in the 415 Chinese chive samples, and the detection rate was 69.4%(288/415), and there was a combination of pesticides in many samples. According to the National Food Safety Standard―Maximum Residue Limits of Pesticides in Food(GB 2763-2019), the residues of 12 pesticides exceeded the maximum residue limits(MRLs), and the unqualified rate was 38.07%(158/415). The highest detection rate of clothianidin was 41.20%(171/415), but there was no MRL in GB 2763-2019. The next was procymidone, the detection rate of which was 35.42%(147/415), and the over-standard rate was 30.12%(125/415). Forbidden and restricted pesticides were detected in some samples. According to the dietary exposure risk assessment, the NEDI/ADI values were all less than 1 and the intake risk was within acceptable range. In Tangshan area, the types of pesticides used in Chinese chive production are complex, and there are risks of multi-residue pollution and use of banned and restricted pesticides and unregistered pesticides. It is suggested that routine monitoring of pesticide residues and management of pesticide use should be strengthened to ensure the quality and safety of agricultural products. [Conclusions] This study provides a theoretical basis for the safe production of Chinese chive and the standardized and rational use of pesticides.展开更多
Prohibited pesticide residues have become one of the main factors affecting the quality and safety of Lycii Fructus,However,rarely studies focus on the rapid determination of these residues.Here,a total of 30 kinds of...Prohibited pesticide residues have become one of the main factors affecting the quality and safety of Lycii Fructus,However,rarely studies focus on the rapid determination of these residues.Here,a total of 30 kinds of prohibited pesticide residues were determined by ultra-performance liquid chromatography tandem triple quadrupole mass spectrometry(UPLC-MS/MS)in five different process ways.Pretreatment methods,chromatographic separation and detection conditions in mass spectrometry were all optimized accordingly.Among the five different pretreatment methods,the first and third solid phase extraction failed to provide high recoveries of sulfosulfuron compounds(both lower than 60%).Recovery of chlorphenamidine by the Quick Easy Cheap Effective Rugged and Safe multiresidue method(QuEChERS)was lower than 60%,which did not meet the requirements of trace determination.The concentrations of 30 prohibited pesticides residues treated by straightforward and solid phase extraction showed good linearity in their corresponding ranges,with correlation coefficients over 0.99.The average recoveries of straightforward ranged from 78.13%to 110.9%,while RSD ranged from 1.3%to 16.9%,albeit poor purification was observed.The recovery yield from solid phase extraction was between 67.75%and 103.08%with RSD value from 0.8%to 14.0%,which met the requirements of trace determination,this method has good precision and stability.These results could be employed to other Traditional Chinese Medicines(TCMs)in detecting prohibited pesticide residues.展开更多
[Objectives]This study was conducted to investigate the pesticide residue status of bananas in Tangshan area and the dietary intake risk of consumers.[Methods]High-performance liquid chromatography-tandem mass spectro...[Objectives]This study was conducted to investigate the pesticide residue status of bananas in Tangshan area and the dietary intake risk of consumers.[Methods]High-performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)was applied to detect 70 pesticide residues in 191 batches of banana samples extracted from Tangshan area,and chronic and acute risk assessments were conducted.[Results]Among the 191 batches of bananas,172 batches of samples were detected to contain agrochemicals,with a detection rate of 90.05%.Seventeen agrochemicals were detected,accounting for 24.29%of the total agrochemicals tested.There was a phenomenon of multiple pesticide residues in a single sample,and high detection rates were found in carbendazim(49.21%),prochloraz(41.88%),pyraclostrobin(34.03%)and imidacloprid(30.37%).According to GB 2763-2021 National Food Safety Standards—Maximum Residue Limits for Pesticides in Food,the over-standard rate of imidacloprid was 17.8%;and the chronic and acute dietary risks of the 17 pesticide residues were all less than 100%.[Conclusions]The pesticide residues in bananas consumed by consumers on a daily basis do not pose unacceptable dietary risks.展开更多
Under the background of a transformation of the global energy structure,coal gasification technology has a wide application prospect,but its by-product,the coal gasification residue(CGR),is still not being efficiently...Under the background of a transformation of the global energy structure,coal gasification technology has a wide application prospect,but its by-product,the coal gasification residue(CGR),is still not being efficiently utilized for recycling.The CGR contains abundant carbon components,which could be applied to the microwave absorption field as the carbon matrix.In this study,Fe/CGR composites are fabricated via a two-step method,including the impregnation of Fe^(3+)and the reduction process.The influence of the different loading capacities of the Fe component on the morphology and electromagnetic properties is studied.Moreover,the loading content of Fe and the surface morphology of the Fe/CGR can be reasonably controlled by adjusting the concentration of the ferric nitrate solution.Meanwhile,Fe particles are evenly inserted on the CGR framework,which expands the Fe/CGR interfaces to enhance interfacial polarization,thus further improving the microwave-absorbing(MA)properties of composites.Particularly,as the Fe^(3+)concentration is 1.0 mol/L,the Fe/CGR composite exhibits outstanding performance.The reflection loss reaches-39.3 dB at 2.5 mm,and the absorption bandwidth covers 4.1 GHz at 1.5 mm.In this study,facile processability,resource recycling,appropriately matched impedance,and excellent MA performance are achieved.Finally,the Fe/CGR composites not only enhance the recycling of the CGR but also pioneer a new path for the synthesis of excellent absorbents.展开更多

Forest wastes generated after thinning, land clearing, and logging operations, such as wood debris, tree trimmings, barks, sawdust, wood chips, and black liquor, among others, can serve as potential fuels for energy production in Louisiana. This paper aims to evaluate the potential annual volumes of forest wastes established on detailed and existing data on the forest structure in the rural-urban interface of Louisiana. It also demonstrates the state’s prospects of utilizing forest wastes to produce bio-oils. The data specific to the study was deduced from secondary data sources to obtain the annual average total residue production in Louisiana and estimate the number of logging residues available for procurement for bioenergy production. The total biomass production per year was modeled versus years by polynomial regression curve fitting using Microsoft Excel. Results of the model show that the cumulative annual total biomass production for 2025 and 2030 in Louisiana is projected to be 80000000 Bone Dry Ton (BDT) and 16000000 (BDT) respectively. The findings of the study depict that Louisiana has a massive biomass supply from forest wastes for bioenergy production. Thus, the potential for Louisiana to become an influential player in the production of bio-based products from forest residues is evident. The author recommends that future research can use Geographic Information Systems (GIS) to create maps displaying the potential locations and utilization centers of forest wastes for bioenergy production in the state.展开更多
To ensure the safety of the commercially available chenpi,a convenient and fast analytical method was developed for the determination of 133 pesticide residues in chenpi using gas chromatography-tandem mass spectromet...To ensure the safety of the commercially available chenpi,a convenient and fast analytical method was developed for the determination of 133 pesticide residues in chenpi using gas chromatography-tandem mass spectrometry(GC-MS/MS).In this study,different extraction solvents,redissolution solvents and adsorbents were tested according to the recovery and purification effect to obtain a modified QuEChERS method.The samples were extracted with acetonitrile.During the clean-up step,octadecyl-modified silica(C18)and graphitized carbon black(GCB)were selected,and aminopropyl(NH2)was used instead of primary secondary amine(PSA)because of its weaker ion exchange capacity which had little effect on the recovery of ditalimfos.Samples were quantified by matrix-matched calibration with internal standards.All pesticides showed good linearity in the respective range,both with values of r2>0.99.The average recoveries of the pesticides spiked samples ranged from 70.0% to 112.2% with the RSDs of 0.2%–14.4%.The modified QuEChERS method was validated and applied to twenty real samples.Five pesticides were found in eight batches,but no pesticide exceeded the maximum residue limits(MRL,MRL reference to European commission).展开更多

The aroma of peach fruit is predominantly determined by the accumulation of γ-decalactone and ester compounds.A previous study showed that the biosynthesis of these aroma compounds in peach fruit is catalyzed by PpAA...The aroma of peach fruit is predominantly determined by the accumulation of γ-decalactone and ester compounds.A previous study showed that the biosynthesis of these aroma compounds in peach fruit is catalyzed by PpAAT1,an alcohol acyltransferase.In this work,we investigated the key active site residues responsible for γ-decalactone and ester biosynthesis.A total of 14 candidate amino acid residues possibly involved in internal esterification and 9 candidate amino acid residues possibly involved in esterification of PpAAT1 were assessed via site-directed mutagenesis.Analyses of the in vitro enzyme activities of PpAAT1 and its site-directed mutant proteins(PpAAT1-SMs)with different amino acid residue mutations as well as the contents of γ-decalactone in transgenic tobacco leaves and peach fruits transiently expressing PpAAT1 and PpAAT1-SMs revealed that site-directed mutation of H165 in the conserved HxxxD motif led to lost enzymatic activity of PpAAT1 in both internal esterification and its reactions,whereas mutation of the key amino acid residue D376 led to the total loss ofγ-decalactone biosynthesis activity of PpAAT1.Mutations of 9 and 7 other amino acid residues also dramatically affected the enzymatic activity of PpAAT1 in the internal esterification and esterification reactions,respectively.Our findings provide a biochemical foundation for the mechanical biosynthesis ofγ-decalactone and ester compounds catalyzed by PpAAT1 in peach fruits,which could be used to guide the molecular breeding of new peach species with more favorable aromas for consumers.展开更多

With XRFS (X-ray fluorescence spectrometry...INAA (instrumental neutron activation analysis) was used to determine the contents of trace elements in coal samples from 11 workable Permian coal seams in Huainan coalfield. With XRFS (X-ray fluorescence spectrometry), the geochemical composition of its combustion residues from Tianjiaan Power Plant and Luohe Power Plant was analyzed. In addition, the electron microprobe was employed to study the chemical composition of some fly ashes and bottom ashes. Compared with those of the coals around the world, most trace elements of the Huainan coal are characterized by a normal abundance. Like most coals worldwide, the elements Se, S, As, Sb, Br, U and Cl are enriched in Huainan coal according to dark’s value, while the other elements tend to be diluted. The large fluctuation in the contents of trace elements in this kind of coal demonstrates great differences in coal seams. On the other hand, the contents of these elements are very similar in different bottom and fly ash samples. The elements with low organic affinity tend to concentrate in fly and bottom ash than in coal, especially those elements associated with Fe. However, the elements with strong organic affinity tend to be volatile during the combustion. Moreover, the abundance of the elements in fly ash and bottom ash changes with different densities and with different particle sizes. The bottom and fly ashes consist of glass, lime, mullite, coal residues and ferrite minerals. Some residual chars contain higher contents of volatile elements, such as S, P, Cl and As, indicating that the ash containing more residual chars has a strong adsorption of some hazardous elements, suggesting a possible utilization of power plant ash in the field of water cleaning.展开更多

Biomass estimations can help design energy strategies to reduce fossil fuels dependency. Because of the land property distribution in Mexico,biomass estimations should consider not only the physical availability, but also the willingness of landowners to extract such raw materials.Methods: This study presents a methodological approach for evaluating the potential use of forest residues as a feedstock to generate bioenergy in northern Mexico. Remote sensing and field forest inventory were used to estimate the quantity and distribution of forest residues. In addition, a discrete choice analysis evaluated landowners’ preferences towards bioenergy development, including the most important factors that influence their willingness to extract their products and the expected price.Results and conclusions: Considering both physical and socio-economic aspects, results showed that about59,000 metric tons per year could be available in the study area. The vast majority of landowners surveyed are willing to extract forest residues, as long as they are presented with extraction plans with the highest income. However, many showed concerns about the environmental impacts this activity can have on soils,plants, and fauna. These results can help evaluate the potential of these resources for bioenergy development.展开更多

文摘Lotus(Nelumbo nucifera),an ancient aquatic plant,possesses a unique pharmacological activity that is primarily contributed by benzylisoquinoline alkaloids(BIAs).However,only few genes and enzymes involved in BIA biosynthesis in N.nucifera have been isolated and characterized.In the present study we identified the regiopromiscuity of an O-methyltransferase,designated NnOMT6,isolated from N.nucifera;NnOMT6 was found to catalyze the methylation of monobenzylisoquinoline 6-O/7-O,aporphine skeleton 6-O,phenylpropanoid 3-O,and protoberberine 2-O.We further probed the key residues affecting NnOMT6 activity via molecular docking and molecular dynamics simulation.Verification using site-directed mutagenesis revealed that residues D316,N130,L135,N176A,D269,and E328 were critical for BIA O-methyltransferase activities;furthermore,N323A,a mutant of NnOMT6,demonstrated a substantial increase in catalytic efficiency for BIAs and a broader acceptor scope compared with wild-type NnOMT6.To the best of our knowledge,this is the first study to report the O-methyltransferase activity of an aporphine skeleton without benzyl moiety substitutions in N.nucifera.The study findings provide biocatalysts for the semisynthesis of related medical compounds and give insights into protein engineering to strengthen O-methyltransferase activity in plants.

文摘A new method for screening and identification 420 pesticide residues in fruits and vegetables by ultra-performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry(UPLC-Q-TOF/MS)were developed.The samples were extracted with acetonitrile/acetic acid(99:1,V/V),and clean-up by SinChERS-Nano(single-step,cheap,effective,rugged,safe,nano)column,determined by UPLC-Q-TOF/MS.The accurate mass database and MS/MS database which contains 420 pesticides were established,the automatic retrieval of detection results was carried on according to the accurate mass,retention time,isotope ratio,ion fragment information,and so on.Method verification was performed on leeks samples.The results showed that 420 pesticides had good linearity in the range of 0.1-100μg/L,and the correlation coeffificients(R2)was greater than 0.990.The limits of detections(LODs)and limits of quantifications(LOQs)of 420 pesticides were in range of 0.05-2.0 and 0.1-5.0μg/L,respectively.The average spike recoveries at 3 levels were 70.1%to 119.7%,and the relative standard deviations(RSD)were lower than 20%(n=6).With this method,a survey of pesticide residues was conducted for 110 samples of 10 different fruits and vegetables,which provided scientific data for ensuring pesticide residue safety of the fruits and vegetables consumed daily by the public.This method was simple,sensitive and accurate,and could be used for rapid screening of 420 pesticide residues in fruits and vegetables.
文摘[Objectives]This study was conducted to establish a rapid and effective method for simultaneous extraction of 54 kinds of veterinary drug residues in animal-derived food, including sulfonamides, quinolones, tetracyclines, malachite greens, penicillins, nitroimidazoles, tranquilizers and macrolides, by HPLC-MS. [Methods] The samples were extracted with 80% acetonitrile water(containing 0.1% formic acid), combined with QuEChERS extraction technology and C18 and PSA purification, analyzed by high performance liquid chromatography-mass spectrometry, and quantified by external standard method. The target substances were analyzed on ZORBAX Eclipse C18 chromatographic column using 0.2% formic acid water and 0.2% methanol as mobile phases. The gradient elution mode was used for chromatographic separation and multiple reaction detection. [Results] In the linear range of 0.5-50.0 ng/ml, the linear relationship of the 54 kinds of veterinary drug residues was good, with correlation coefficients(r~2) greater than 0.995, and the detection limits ranged from 0.30 to 1.00 μg/kg. The results showed that the recovery ranged from 75.4% to 118.2% when different matrixes were added for recovery. [Conclusions] This method is simple, efficient, accurate, stable, and highly operable. It is applicable to simultaneous batch screening of veterinary drug residues in animal-derived food, and has high practical application value.

文摘The tea plant(Camellia sinensis)is rich in polyphenolic compounds.Particularly,flavan-3-ols and proanthocyanidins(PAs)are essential for the flavor and disease-resistance property of tea leaves.The fifth subgroup of R2R3-MYB transcription factors comprises the primary activators of PA biosynthesis.This study showed that subgroup 5 R2R3-MYBs in tea plants contained at least nine genes belonging to the TT2,MYB5,and MYBPA types.Tannin-rich plants showed an expansion in the number of subgroup 5 R2R3-MYB genes compared with other dicotyledonous and monocot plants.The MYBPA-type genes of tea plant were slightly expanded.qRT–PCR analysis and GUS staining analysis of promoter activity under a series of treatments revealed the differential responses of CsMYB5s to biotic and abiotic stresses.In particular,CsMYB5a,CsMYB5b,and CsMYB5e responded to high-intensity light,high temperature,MeJA,and mechanical wounding,whereas CsMYB5f and CsMYB5g were only induced by wounding.Three genetic transformation systems(C.sinensis,Nicotiana tabacum,and Arabidopsis thaliana)were used to verify the biological function of CsMYB5s.The results show that CsMYB5a,CsMYB5b,and CsMYB5e could promote the gene expression of CsLAR and CsANR.However,CsMYB5f and CsMYB5g could only upregulate the gene expression of CsLAR but not CsANR.A series of site-directed mutation and domain-swapping experiments were used to verify functional domains and key amino acids of CsMYB5s responsible for the regulation of PA biosynthesis.This study aimed to provide insight into the induced expression and functional diversity model of PA biosynthesis regulation in tea plants.

文摘[Objectives]This study was conducted to understand the status of pesticide residues and dietary intake risk of Chinese chives in Tangshan area. [Methods] Sixty eight kinds of pesticide residues in 415 Chinese chive samples collected from Tangshan area were qualitatively and quantitatively determined by high-performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS) and gas chromatography(GC) in 2020. [Results] The results showed that 41 kinds of pesticide residues were detected in the 415 Chinese chive samples, and the detection rate was 69.4%(288/415), and there was a combination of pesticides in many samples. According to the National Food Safety Standard―Maximum Residue Limits of Pesticides in Food(GB 2763-2019), the residues of 12 pesticides exceeded the maximum residue limits(MRLs), and the unqualified rate was 38.07%(158/415). The highest detection rate of clothianidin was 41.20%(171/415), but there was no MRL in GB 2763-2019. The next was procymidone, the detection rate of which was 35.42%(147/415), and the over-standard rate was 30.12%(125/415). Forbidden and restricted pesticides were detected in some samples. According to the dietary exposure risk assessment, the NEDI/ADI values were all less than 1 and the intake risk was within acceptable range. In Tangshan area, the types of pesticides used in Chinese chive production are complex, and there are risks of multi-residue pollution and use of banned and restricted pesticides and unregistered pesticides. It is suggested that routine monitoring of pesticide residues and management of pesticide use should be strengthened to ensure the quality and safety of agricultural products. [Conclusions] This study provides a theoretical basis for the safe production of Chinese chive and the standardized and rational use of pesticides.
文摘Prohibited pesticide residues have become one of the main factors affecting the quality and safety of Lycii Fructus,However,rarely studies focus on the rapid determination of these residues.Here,a total of 30 kinds of prohibited pesticide residues were determined by ultra-performance liquid chromatography tandem triple quadrupole mass spectrometry(UPLC-MS/MS)in five different process ways.Pretreatment methods,chromatographic separation and detection conditions in mass spectrometry were all optimized accordingly.Among the five different pretreatment methods,the first and third solid phase extraction failed to provide high recoveries of sulfosulfuron compounds(both lower than 60%).Recovery of chlorphenamidine by the Quick Easy Cheap Effective Rugged and Safe multiresidue method(QuEChERS)was lower than 60%,which did not meet the requirements of trace determination.The concentrations of 30 prohibited pesticides residues treated by straightforward and solid phase extraction showed good linearity in their corresponding ranges,with correlation coefficients over 0.99.The average recoveries of straightforward ranged from 78.13%to 110.9%,while RSD ranged from 1.3%to 16.9%,albeit poor purification was observed.The recovery yield from solid phase extraction was between 67.75%and 103.08%with RSD value from 0.8%to 14.0%,which met the requirements of trace determination,this method has good precision and stability.These results could be employed to other Traditional Chinese Medicines(TCMs)in detecting prohibited pesticide residues.

文摘[Objectives]This study was conducted to investigate the pesticide residue status of bananas in Tangshan area and the dietary intake risk of consumers.[Methods]High-performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)was applied to detect 70 pesticide residues in 191 batches of banana samples extracted from Tangshan area,and chronic and acute risk assessments were conducted.[Results]Among the 191 batches of bananas,172 batches of samples were detected to contain agrochemicals,with a detection rate of 90.05%.Seventeen agrochemicals were detected,accounting for 24.29%of the total agrochemicals tested.There was a phenomenon of multiple pesticide residues in a single sample,and high detection rates were found in carbendazim(49.21%),prochloraz(41.88%),pyraclostrobin(34.03%)and imidacloprid(30.37%).According to GB 2763-2021 National Food Safety Standards—Maximum Residue Limits for Pesticides in Food,the over-standard rate of imidacloprid was 17.8%;and the chronic and acute dietary risks of the 17 pesticide residues were all less than 100%.[Conclusions]The pesticide residues in bananas consumed by consumers on a daily basis do not pose unacceptable dietary risks.

文摘Under the background of a transformation of the global energy structure,coal gasification technology has a wide application prospect,but its by-product,the coal gasification residue(CGR),is still not being efficiently utilized for recycling.The CGR contains abundant carbon components,which could be applied to the microwave absorption field as the carbon matrix.In this study,Fe/CGR composites are fabricated via a two-step method,including the impregnation of Fe^(3+)and the reduction process.The influence of the different loading capacities of the Fe component on the morphology and electromagnetic properties is studied.Moreover,the loading content of Fe and the surface morphology of the Fe/CGR can be reasonably controlled by adjusting the concentration of the ferric nitrate solution.Meanwhile,Fe particles are evenly inserted on the CGR framework,which expands the Fe/CGR interfaces to enhance interfacial polarization,thus further improving the microwave-absorbing(MA)properties of composites.Particularly,as the Fe^(3+)concentration is 1.0 mol/L,the Fe/CGR composite exhibits outstanding performance.The reflection loss reaches-39.3 dB at 2.5 mm,and the absorption bandwidth covers 4.1 GHz at 1.5 mm.In this study,facile processability,resource recycling,appropriately matched impedance,and excellent MA performance are achieved.Finally,the Fe/CGR composites not only enhance the recycling of the CGR but also pioneer a new path for the synthesis of excellent absorbents.

文摘[Objectives]The paper was to establish a rapid screening method of pesticide residues in strawberry based on HPLC-QTOF/MS SWATH acquisition mode.[Methods]The strawberry sample was extracted with acetonitrile,purified with anhydrous magnesium sulfate,sodium chloride and primary secondary amine(PSA)powder,and then loaded on the machine.The full information spectrum was collected by positive ion mode and screened by comparing with the spectrum library,and 20 kinds of pesticide residues were quantitatively analyzed.[Results]The detection limits of 20 pesticide residues were 0.0002-0.0100 mg/kg,and they showed good linearity in the range of 0.0005-0.1000μg/mL(R 2≥0.995).The recoveries ranged from 62.8%to 106.3%.[Conclusions]This method is rapid,simple,sensitive and non-targeted,and can be used for rapid screening of multiple pesticide residues in strawberry.

文摘[Objectives]A high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)method was established for the determination of 14β-receptor agonist residues in mutton.[Methods]Samples were hydrolyzed byβ-glucuronidase and extracted with 5%acetic acid-acetonitrile(1:99,V/V)solution.An Eclipse plus C 18 column was used for separation,and the MRM mode was used for qualitative analysis,and the external standard method was used for quantitative analysis of matrix standard solutions.[Results]Under the optimal conditions,the retention time of the 14 kinds ofβ-receptor agonists ranged from 1.0 to 9.5 min.When the mass concentration was in the range of 0.05-0.50μg/ml,the linear relationship ofβ-receptor agonists was good,with correlation coefficients(r)≥0.9992.The detection limits of the method were in the range of 0.04-0.87μg/kg,and the quantitative limits were in the range of 0.35-1.86μg/kg.The average recovery values were in the range of 82.8%-108.9%,with RSDs(n=6)in the range of 1.9%-6.7%.[Conclusions]The method is simple,sensitive,reproducible,accurate,and can be used for simultaneous determination of the 14 kinds ofβ-receptor agonist residues in mutton.
文摘Louisiana is endowed with forest resources. Forest wastes generated after thinning, land clearing, and logging operations, such as wood debris, tree trimmings, barks, sawdust, wood chips, and black liquor, among others, can serve as potential fuels for energy production in Louisiana. This paper aims to evaluate the potential annual volumes of forest wastes established on detailed and existing data on the forest structure in the rural-urban interface of Louisiana. It also demonstrates the state’s prospects of utilizing forest wastes to produce bio-oils. The data specific to the study was deduced from secondary data sources to obtain the annual average total residue production in Louisiana and estimate the number of logging residues available for procurement for bioenergy production. The total biomass production per year was modeled versus years by polynomial regression curve fitting using Microsoft Excel. Results of the model show that the cumulative annual total biomass production for 2025 and 2030 in Louisiana is projected to be 80000000 Bone Dry Ton (BDT) and 16000000 (BDT) respectively. The findings of the study depict that Louisiana has a massive biomass supply from forest wastes for bioenergy production. Thus, the potential for Louisiana to become an influential player in the production of bio-based products from forest residues is evident. The author recommends that future research can use Geographic Information Systems (GIS) to create maps displaying the potential locations and utilization centers of forest wastes for bioenergy production in the state.
文摘To ensure the safety of the commercially available chenpi,a convenient and fast analytical method was developed for the determination of 133 pesticide residues in chenpi using gas chromatography-tandem mass spectrometry(GC-MS/MS).In this study,different extraction solvents,redissolution solvents and adsorbents were tested according to the recovery and purification effect to obtain a modified QuEChERS method.The samples were extracted with acetonitrile.During the clean-up step,octadecyl-modified silica(C18)and graphitized carbon black(GCB)were selected,and aminopropyl(NH2)was used instead of primary secondary amine(PSA)because of its weaker ion exchange capacity which had little effect on the recovery of ditalimfos.Samples were quantified by matrix-matched calibration with internal standards.All pesticides showed good linearity in the respective range,both with values of r2>0.99.The average recoveries of the pesticides spiked samples ranged from 70.0% to 112.2% with the RSDs of 0.2%–14.4%.The modified QuEChERS method was validated and applied to twenty real samples.Five pesticides were found in eight batches,but no pesticide exceeded the maximum residue limits(MRL,MRL reference to European commission).

文摘This study established a method for the simultaneous determination of 74 pesticide residues in Panax notoginseng by QuEChERS pretreatment method coupled with GC-MS/MS,and carried out pesticide residue analysis on 20 batches of market samples in China.The samples were extracted with acetonitrile,cleaned up with primary secondary amine(PSA)and octadecylsilane(C18)and determined by GC-MS/MS in multiple reaction monitoring(MRM)mode.Matrix-matched calibration was recommended to combat the matrix effect.A good linearity was observed in the range of 10−500 ng/mL with correlation coefficients≥0.9950.The mean recoveries for most of the pesticides were in the range of 70%−120%with RSD<20%.The limits of detection ranged 0.28–2.00μg/kg,while the limits of quantification were 0.94–6.65μg/kg.Following the application of“top-down”approach,the expanded measurement uncertainty for all the analytes was<30%.The proposed method was successfully applied to determine pesticide residues in 20 market samples in China,where 9 pesticides were detected and quintozene exceeded the criteria domestically and abroad.

文摘In this experiment,a liquid chromatography tandem mass spectrometry method was built to determine 15 pesticide residues in Chinese cabbage and cucumber samples based on online turbulent flow chromatography purification.After modified quick,easy,cheap,effective,rugged,and safe(QuEChERS)extraction,extracts were directly injected to the TLX(TurboFlow Liquid Xcalibur)system and brought to TurboFlow™columns for on-line purification and then transferred to analytical column for further separation and analysis.TurboFlow™columns types,transfer flow rate,and transfer time were optimized.Limits of detection and limits of quantification of the method obtained for 15 pesticide residues were ranged between 0.2–1.0μg/kg and 0.5–2.0μg/kg in Chinese cabbage and cucumber samples.Recoveries of pesticide residues were in range of 75.3%–103.7%.Matrix effects for 15 pesticides were in range of 5.6%–106.6%.The developed method has been successfully used for the determination of 15 pesticide residues in real samples.

文摘[Objectives]To establish a method for the determination of heavy metals and pesticide residues in Dendrobium officinale produced in Guangxi and its culture substrate.[Methods]The contents of five heavy metals(Pb,Cd,As,Hg and Cu)were determined by the atomic absorption spectrophotometric(AAS)method;residues of 9 organochlorine pesticides(α-BHC,β-BHC,γ-BHC,δ-BHC,p,p′-DDT,p,p′-DDE,p,p′-DDD,o,p′-DDT and pentachloronitrobenzene)were determined by gas chromatography(GC).[Results]The contents of five heavy metals in the medicinal materials of D.officinale produced in Guangxi were 0.96,0.21,0.021 mg/kg,and not detected,and 0.35 mg/kg.The contents of five heavy metals in the culture substrate were 0.03,0.19,0.21,0.004 and 1.80 mg/kg,respectively;the content of 9 organochlorine pesticide residues in the sample was not detected.[Conclusions]The detection results of heavy metal content and organochlorine pesticide residues in D.officinale produced in Guangxi and its substrate are consistent with the limit requirements of the Chinese Pharmacopoeia.This experiment is expected to provide reliable data reference for ensuring the safety of the drug resources of D.officinale in Guangxi.

文摘The aroma of peach fruit is predominantly determined by the accumulation of γ-decalactone and ester compounds.A previous study showed that the biosynthesis of these aroma compounds in peach fruit is catalyzed by PpAAT1,an alcohol acyltransferase.In this work,we investigated the key active site residues responsible for γ-decalactone and ester biosynthesis.A total of 14 candidate amino acid residues possibly involved in internal esterification and 9 candidate amino acid residues possibly involved in esterification of PpAAT1 were assessed via site-directed mutagenesis.Analyses of the in vitro enzyme activities of PpAAT1 and its site-directed mutant proteins(PpAAT1-SMs)with different amino acid residue mutations as well as the contents of γ-decalactone in transgenic tobacco leaves and peach fruits transiently expressing PpAAT1 and PpAAT1-SMs revealed that site-directed mutation of H165 in the conserved HxxxD motif led to lost enzymatic activity of PpAAT1 in both internal esterification and its reactions,whereas mutation of the key amino acid residue D376 led to the total loss ofγ-decalactone biosynthesis activity of PpAAT1.Mutations of 9 and 7 other amino acid residues also dramatically affected the enzymatic activity of PpAAT1 in the internal esterification and esterification reactions,respectively.Our findings provide a biochemical foundation for the mechanical biosynthesis ofγ-decalactone and ester compounds catalyzed by PpAAT1 in peach fruits,which could be used to guide the molecular breeding of new peach species with more favorable aromas for consumers.

文摘INAA (instrumental neutron activation analysis) was used to determine the contents of trace elements in coal samples from 11 workable Permian coal seams in Huainan coalfield. With XRFS (X-ray fluorescence spectrometry), the geochemical composition of its combustion residues from Tianjiaan Power Plant and Luohe Power Plant was analyzed. In addition, the electron microprobe was employed to study the chemical composition of some fly ashes and bottom ashes. Compared with those of the coals around the world, most trace elements of the Huainan coal are characterized by a normal abundance. Like most coals worldwide, the elements Se, S, As, Sb, Br, U and Cl are enriched in Huainan coal according to dark’s value, while the other elements tend to be diluted. The large fluctuation in the contents of trace elements in this kind of coal demonstrates great differences in coal seams. On the other hand, the contents of these elements are very similar in different bottom and fly ash samples. The elements with low organic affinity tend to concentrate in fly and bottom ash than in coal, especially those elements associated with Fe. However, the elements with strong organic affinity tend to be volatile during the combustion. Moreover, the abundance of the elements in fly ash and bottom ash changes with different densities and with different particle sizes. The bottom and fly ashes consist of glass, lime, mullite, coal residues and ferrite minerals. Some residual chars contain higher contents of volatile elements, such as S, P, Cl and As, indicating that the ash containing more residual chars has a strong adsorption of some hazardous elements, suggesting a possible utilization of power plant ash in the field of water cleaning.
文摘Background: Forest residues can be a feasible alternative for converting energy into fuels, electricity, or heat.Compared to other second-generation bioenergy sources, they do not compete for food, are relatively cheap,abundant in forest-rich areas, and more importantly their energy balance is close to zero. Biomass estimations can help design energy strategies to reduce fossil fuels dependency. Because of the land property distribution in Mexico,biomass estimations should consider not only the physical availability, but also the willingness of landowners to extract such raw materials.Methods: This study presents a methodological approach for evaluating the potential use of forest residues as a feedstock to generate bioenergy in northern Mexico. Remote sensing and field forest inventory were used to estimate the quantity and distribution of forest residues. In addition, a discrete choice analysis evaluated landowners’ preferences towards bioenergy development, including the most important factors that influence their willingness to extract their products and the expected price.Results and conclusions: Considering both physical and socio-economic aspects, results showed that about59,000 metric tons per year could be available in the study area. The vast majority of landowners surveyed are willing to extract forest residues, as long as they are presented with extraction plans with the highest income. However, many showed concerns about the environmental impacts this activity can have on soils,plants, and fauna. These results can help evaluate the potential of these resources for bioenergy development.
