The Protective Effects of Flavonoids from Scutellaria Baicalensis Georgi Stems and Leaves on Oligodendrocyte Damage Induced by Aβ1-42

Aim: This study aimed to investigate the protective effects of flavonoids from the stem and leaves of Scutellaria baicalensis Georgi (SSFs) against Aβ<sub>1-42</sub>-induced oligodendrocytes (OL) damage. Methods: Immunofluorescence was used for the detection of myelin-associated glycoprotein (MAG), a characteristic protein of rat oligodendrocytes (OLN-93 cells). To evaluate the potential protective effects of SSFs on OLN-93 cells injured by Aβ<sub>1-42</sub>, an injury model was established by subjecting OLN-93 cells to Aβ<sub>1-42</sub> exposed. Cell morphology was examined using an inverted microscope, while cell viability was assessed using the colorimetric method of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT). Additionally, lactate dehydrogenase (LDH) was measured using the pyruvic acid reduction assay. The Ginkgo biloba leaf extract (GBE) injection was used as a positive control. Results: A total of >95% of the MAG immunofluorescence-positive cells were identified as oligodendrocytes. Gradually increasing concentrations of SSFs impaired the cells, and the maximum nondetrimental dose for OLN-93 cells was 75 mg/L. This study assessed the effects of SSFs on OLN-93 cells damaged by Aβ<sub>1-42</sub>. The results indicated that SSFs significantly improved OLN-93 cell morphological abnormal changes, increased the OLN-93 cell survival rate, and reduced LDH release. Conclusion: SSFs can alleviate Aβ<sub>1-42</sub>-induced damage of OL. 展开更多

The cotton direct seeding after wheat(rape) harvested is under trial and would be the future direction at the Yangtze River Valley region of China.The objective of this study was to quantify the effects of branch and stem architecture on cotton yield and identify the optimal cotton architecture to compensate the yield loss due to the reduction of individual production capacity under high planting density in the direst seeding after wheat harvested cropping system.The characteristics of the stem and branch architecture and the relationships between architecture of the stem and branch with yield formation were studied on eight short season cotton cultivars during 2015 and 2016 cotton growth seasons.Based on the two years results,three cultivars with different architectures of stem and branch were selected to investigate the effect of mepiquat chloride(MC) application on the architecture of the stem and branch,boll retention,and the yield in 2017.Significant differences were observed on plant height,all fruiting nodes to branches ratio(NBR) in the cotton plant,and the curvature of the fruiting branch(CFB) among the studied cultivars.There were three types of stem and fruiting branch structures: Zhong425 with stable and suitable plant height and NBR(about 90 cm and 2.5,respectively),high CFB(more than 10.0),and high boll retention speed and seed cotton yield;Siyang 822 with excessive plant height and NBR,low CFB,and low boll retention speed and seed cotton yield;and other studied cultivars with unstable structure of stem and branch,boll retention speed,and seed cotton yield across years.And MC application could promote the appropriate plant height and NBR and high CFB and thus resulted in high boll retention speed and the yield.The results suggested that the suitable plant height and NBR(about 90 cm and 2.5 respectively),and high CFB(more than 10.0),which was related to both genotype and cultural practice,could promote the higher boll retention speed and seed cotton yield. 展开更多

The accurate assessment of actual tree stem respiration and its relation with temperature plays a considerable role in investigating the forest carbon cycle.An increasing number of research reports have indicated that tree stem respiration determined with the commonlyapplied chamber gas exchange measuring system does not follow expectations regarding temperature relationships.This method is based on the nowadays widely-accepted theory that the respired CO_(2) in a tree stem would all diffuse outward into the atmosphere.However,it neglects partial CO_(2) that is dissolved in the xylem sap and is carried away by the transpirational stream.Scientists have started to realize that the respired CO_(2) measured with the chamber gas exchange method is only a portion of the total stem respiration(CO_(2) efflux),while the other portion,which is sometimes very substantial in quantity(thought to occupy maybe 15%-75%of the total stem respiration),is transported to the upper part of the stem and to the canopy by sap flow.This suggests that the CO_(2) produced by respiration is re-allocated within the stem.Accordingly,the change in CO_(2) efflux could be reflected in the rates of sap flow in addition to its dependence on temperature.Proper methods and instruments are required to quantify the internal and external CO_(2) fluxes in the trunk and their interaction with related environmental factors. 展开更多
