In the absence of a Differentiation of Infected from Vaccinated Animal (DIVA) assay for bovine Brucellosis, it becomes difficult to assess whether the anti-Brucella antibody response in an animal is due to vaccination or infection. We compared the anti-Brucella antibody titers of naturally Brucellosis affected unvaccinated cows, previously vaccinated infected cows, normal healthy vaccinated cows and healthy unvaccinated calves. The titers of anti-Brucella antibodies were estimated by indirect ELISA. The mean titer (log10) was found to be 1.518 ± 0.005 in case of naturally Brucellosis affected cattle which had been vaccinated during calf hood. The mean titer in case of naturally infected cattle which had never been vaccinated was 1.5441 ± 0.005. The mean titer in healthy unaffected cattle vaccinated during calf hood was 1.504 ± 0.002 and that of unvaccinated healthy calves was 0.560 ± 0.016. It was interesting to find that the antibody titers in naturally affected cattle which had never been vaccinated were very significantly (p < 0.01) higher than those of Brucellosis affected cows which had been vaccinated during calf hood. The titer in vaccinated infected cattle was very significantly (p < 0.01) higher than that of uninfected vaccinated cows.展开更多

During our surveillance of chicken from 2019 to 2021 in Shandong Province, China, we isolated 11 H9N2AIVs. Phylogenetic analyses showed that the eight gene segments of the 11 isolates were closely related to several sublineages of Eurasian lineage: BJ/94-like clades(HA and NA genes), G1-like clades(PB2 and M genes), and SH/F/98-like clades(PB1, PA, NP and NS genes). The isolates showed mutation sites that preferentially bind to humanlike receptors(HA) and mammalian fitness sites(PB2, PB1 and PA), as well as mutations in antigen and drug resistance sites. Moreover, studies with mice revealed four isolates with varying levels of pathogenicity. The average antibody titer of the H9N2 AIVs was 8.60 log2. Based on our results, the epidemiological surveillance of H9N2 AIVs should be strengthened.展开更多

This knowledge was lost to confirmation bias and uncontrolled studies that only looked at Epstein-Barr virus antibodies. The latter led to a long American fad of misdiagnosing major affective disorders as a “chronic Epstein-Barr virus syndrome.” This misdiagnosis even became part of the American popular culture during the 1980s. Mortality rates of the different groups were plotted against their respective mean PHA antibody titers. A group with zero antibody titer had a mortality rate of 75% while those with PHA antibody titers of 185.6, 243.2, 256 and 307.2 had mortality rates of 40%, zero, zero and zero respectively. Linear equation generated for a line of best fit of the graph of mortality rates of the chicks on their IBD antibody titers gave antibody titer (X) at which mortality (Y) would be zero as 300. A mortality of 75% and the high antibody level needed to protect chicks suggest that the isolate may be a hypervirulent strain.展开更多

Design of experiment (DoE) was applied to investigate the effect of process parameters (pH, temperature shift and dissolve oxygen (DO)) on protein titer. The key metabolites connecting the critical process parameters (CPPs) with monoclonal antibody production were identified by different chemometrics tools. Finally, the biological events of marker metabolites relating with titer improvement were concluded. pH and temperature shift were identified as CPPs that affect the target protein titer. A series of metabolites influenced by the altered CPPs and correlated with protein titer were screened by principal component analysis (PCA) and Pearson' correlation test. The marker metabolites and their pathways linking CPPs to target protein titer in different culture phases were summarized. Metabolomics and chemometrics are promising data-driven tools to shine light into the biological black box between the bioprocess parameters and process performance.展开更多

The results showed that IgE, IgG or IgGl had no significant effect on tumor incidence; the high IgE level could extend the survival time of 3-Methylcholanthrene injected mice (P【0.05) and survival time of tumor bearing mice (P【0.01), while IgG or IgGl could not. The data from this study did not show that IgE, IgG or IgGl had any significant effect on the tumor size and the tumor growth rate. The probable mechanism involved was discussed.展开更多

Atypical chorlocarcinoma is different from the regular choriocarcinoma in both histopathology and clinical manifestations. Morphologically, the tumor cells are predominantly cyto-trophoblast and clinically, the hCG titer is very low which is undetectable by routine biological or immunologic assays. Therefore, It Is not infrequently misdiagnosed as sarcoma, squamous cell carcinoma or clear cell carcinoma of the uterus. A comprehensive review of the past history and clinical manifestations is most essential to obtain a correct diagnosis.展开更多

文摘To determine passive haemagglutination (PHA) antibody titer that would protect chicks against Nigerian isolates of the Infectious Bursa Disease Virus (IBDV), five groups of chicks aged 30 days which had different antibody titers were challenged with a Nigerian isolate of virulent IBDV. A mortality of 75% and the high antibody level needed to protect chicks suggest that the isolate may be a hypervirulent strain.

文摘A deeper understanding of the biological events occurring when bioprocess parameters changed will be of great value in improving the monoclonal antibodies (mAbs) production. Metabolomics and chemometrics are promising data-driven tools to shine light into the biological black box between the bioprocess parameters and process performance.

文摘Neurotrophic factors are able to promote neuronalsurvival and prevent neuronal death induced by variousinjuries and have potential uses in the treatment ofneuronal degenerative diseases and neuropathy. Due tothe existence of brain blood barrier, neurotrophic factorscan not be ased in CNS diseases directly. Gene therapy isa good candidate to overcome this problem.

文摘BACKGROUND Reactivation of hepatitis B virus(HBV)during anticancer treatment is a critical issue. When treating patients with solid tumors,it is unclear whether specific cancer types or treatments affect HBV reactivation in hepatitis B surface antigen(HBsAg)-negative and hepatitis B core antibody(HBcAb)-positive patients, socalled de novo hepatitis B patients. The risk of de novo hepatitis B may vary based on different background factors.AIM To determine the frequency and risk factors for de novo hepatitis B during solid tumor treatment.METHODS This retrospective cohort study comprised 1040 patients without HBsAgs and with HBcAbs and/or hepatitis B surface antibodies(HBsAbs). The patients were treated for solid cancer from 2008 to 2018 at the National Kyushu Cancer Center and underwent HBV DNA measurements.Patient characteristics and disease and treatment information were investigated.HBV DNA measurements were performed using TaqMan polymerase chain reaction(PCR).To identify the risk factors associated with HBV DNA expression, the age, sex, original disease, pathology, treatment method, presence or absence of hepatitis C virus(HCV),and HBsAb and/or HBcAb titers of all subjects were investigated. In patients with HBV DNA,the time of appearance, presence of HBsAgs and HBsAbs at the time of appearance, and course of the subsequent fluctuations in virus levels were also investigated.RESULTS Among the 1040 patients,938 were HBcAb positive, and 102 were HBcAb negative and HBsAb positive.HBV DNA expression was observed before the onset of treatment in nine patients(0.9%)and after treatment in 35 patients(3.7%),all of whom were HBcAb positive. The HBV reactivation group showed significantly higher median HBcAb values[9.00(8.12-9.89)vs 7.22(7.02-7.43),P=0.0001]and significantly lower HBsAb values(14 vs 46,P=0.0342)than the group without reactivation.Notably, the reactivated group showed a significantly higher proportion of cancers in organs related to digestion and absorption(79.0%vs 58.7%,P=0.0051).A high HBcAb titer and cancers in organs involved in digestion and absorption were identified as independent factors for HBV reactivation(multivariate analysis,P=0.0002 and P=0.0095).The group without HBsAbs tended to have a shorter time to reactivation(day 43 vs day 193),and the frequency of reactivation within 6 mo was significantly higher in this group(P=0.0459)than in the other group.CONCLUSION A high HBcAb titer and cancers in organs involved in digestion and absorption are independent factors that contribute to HBV reactivation during solid tumor treatment.
文摘The relationship between murine IgE, IgG, and IgG1 level and birosarcoma induced by 3-Methyl-cholanthrene in NIH mice was studied. The probable mechanism involved was discussed.
文摘Hepatitis A virus, HM-175 strain, adapted to low temperature was cultivated in BSC-1 cell culture.Replication of the virus was detected by radioimmunoassay(RIA), radioimmunofocus assay (RIFA) and dot blot hybridization. The results showed that, of the 7 low temperature-adapted(LTA) strains studied, strain E, F and D gave the highest titer in BSC-1 cell culture. This culture system was proved to be suitable for the study of the characteristic of temperature mutants of HAV.

文摘Atypical choriocarcinoma b a rare type of gestational trophoblastic disease and only 3 cases were diagnosed among a total of 696 patients with chorlocarcinoma admitted to our hospital from 1949 through 1985. A comprehensive review of the past history and clinical manifestations is most essential to obtain a correct diagnosis.
文摘Several features of adenoviruses make them anattractive option as a vehicle to transfer genes intoprimary malignant neoplasms in vivo. These viruseshave low pathogenicity in humans and are notneurotoxic. In addition, high titers of the virus can beachieved to allow higher levels of gene transfectionefficiency than the other vector systems. In vivotumorigenicity of G422 glioblastoma cells transfectedwith IL-2 and/or IL-3 genes decreased significantly inour privious report. In this study, recombinantadenoviruses were used to evaluate the therapeuticpotential of combined IL-2/IL-3 gene therapy in thetreatment of established subcutaneous tumor model ofG422 glioblastoma. Murine IL-2, IL-3 recombinantadenoviruses (2×10~8 pfu) were injected directly
