OBJECTIVE: To observe the clinical therapeutic effect of modified hemodilution combined therapy applied in elderly patients with acute cerebral thrombosis and analyze the mechanism of this therapeutic method. DESIGN: 1∶1 paired grouping according to gender and controlled observation. SETTING: Department of Internal Medicine, Chengzhanyuan District, First Hospital Affiliated to Zhejiang University. PARTICIPANTS: Totally 90 elderly patients with acute ischemic stroke who received the treatment in the Cadre Ward and Mental Ward, Department of Internal Medicine, Chengzhanyuan District, First Hospital Affiliated to Zhejiang University from March 1996 to June 2004 were recruited. They all met the diagnosis criteria revised by the Fourth Academic Conference of National Cerebrovascular Diseases in 1995 and were diagnosed as acute ischemic stroke by skull CT. They were informed of therapeutic plan and detected items. According to 1∶1 paired principle in gender, 90 enrolled patients were assigned into treated group (n=45) and control group (n=45). There were 39 male and 6 female in the treatment group, and they were aged (76±6)years, ranging from 71 to 84 years, and hospitalized at the 14th to 76th hours after onset. There were 39 male and 6 female in the control group, and they were aged (76±6)years , ranging from 70 to 82 years, and hospitalized at the 16th to 72th hours after onset. METHODS: Therapeutic method: Patients of treated group received modified hemodilution combined therapy. 200 mL whole blood of patients was exchanged with 500 mL dextran-40 (including 20 mL danshen parenteral solution and 32 mg heparin) at the beginning of therapy; From the 2nd day, compound huangqi tea bag (Huangqi mainly, including danshen, honghua, chuanxiong, shishao and a little acetyl salicylic acid) was made, twice a day, 1 bag once. At the same time, the above-mentioned dextran-40 liquid of 500 mL was intravenously injected, once a day, 14 days in total; On the 6th day after therapy, the above-mentioned aseptic autoblood stored in refrigerator at 4 ℃ was transfused back into the patients following pre-treatment of high-concentration oxygenation and ultraviolet irradiation by light quantum instrument. Patients of control group were intravenously injected of 0.4 g venoruton(Traditional Chinese medicine compound parenteral solution for promoting blood circulation and removing blood stasis ) and 50 g/L glucose of 500 mL, 75 mg acetosal was taken orally, once a day, 14 days in total. ② Measurement and observation of index: Blood coagulation index, change of platelet aggregation rate and change of hemorrheology of patients in two groups were monitored before and after therapy. The level of blood lipid of patients in two groups was measured with American Beckman automatic biochemistry analyzer. Blood flow rate of middle cerebral artery of resting electrocardiogram were measured with American HP SONOS 2500 sonoscope. Neuro-dysfunction score revised in the national conference (1995) was used to evaluate the recovery of neurological function of the patients in two groups at the 3rd, 5th, 7th and 14th days after therapy. ③Therapeutic effect and adverse effect were observed at the same time. MAIN OUTCOME MEASURES: ① Changes of coagulation index, blood lipid level and hemorheology; ② Blood flow rate of middle cerebral artery and NDS of patients with acute ischemic stroke in two groups; ③Adverse effect of drug. RESULTS: Totally 90 patients were enrolled in the experiment. One patient from treated group died of hyperosmolar nonketotic diabetic coma of complicated diabetes mellitus. One patient from control group died of severe pulmonary infection. The rest 88 patients entered the stage of result analysis. ① Change of coagulation index and platelet aggregation rate: prothrombin time (PT), activated partial thromboplastin time (APTT) and thrombin time (TT) of patients after therapy were significantly longer than those before therapy in the treated group and those after therapy in control group [After therapy in treated group: (18.4±1.9), (41.8±2.1), (19.7±1.7) s, Before therapy in treated group: (13.4±1.3), (35.8±1.3), (12.5±0.9) s, After therapy in control group: (16.9±1.5), (39.1±1.1), (11.9±2.1) s, P < 0.05];Concentration of fibrinogen (Fbg) after therapy was significantly lower than that before therapy in the treated group and that after therapy in control group[After therapy in treated group: (3.4±0.4) g/L; Before therapy in treated group: (4.3±0.7) g/L; After therapy in control group:(4.0±0.6) g/L; P < 0.05]. Platelet aggregation rate decreased from (37.92 ±0.85)% before therapy to (26.42±1.01)% after therapy (P < 0.01). ②Change of blood lipid level: Levels of total cholesterol (TC), triacylglycerol(TG) and low density lipoprotein cholesterol (LDL-C) of patients after therapy were significantly lower than those before therapy in treated group and those after therapy in control group [After therapy in treated group: (5.2±0.9), (1.9±0.9), (2.08±1.1) mmol/L, before therapy in treated group:(5.9±1.2), (2.8±0.9), (3.94±0.5) mmol/L, After therapy in control group: (6.0±1.1), (2.6±0.8), (3.84±0.9) mmol/L,P < 0.05]. ③ Change of hemorheology index: Hematocrit of patients of treated group was significantly lower after therapy than before therapy [Before therapy: (43.84±4.55)%;After therapy: (40.48±4.02)%;P < 0.05]. ④ Blood flow rate of middle cerebral artery of patients of treated group was significantly lower before therapy than after therapy [(90±1.2),(97±2.1) cm/s,P < 0.01]. ⑤ NDS of patients in treated group was significantly lower than of control group 14 days after therapy. The total effective rate after therapy was significantly higher in the treated group than in the control group (93%,78%, P < 0.05). ⑥There was no obvious adverse effect. CONCLUSION: Modified hemodilution combined therapy can improve hemorheology, decrease hematocrit, increase blood flow rate of middle cerebral artery, so as to improve the impaired clinical neurological function of elderly patients with acute cerebral thrombosis through anticoagulation and antiplatelet aggregative activity as well as regulating blood lipid.展开更多

By researching the evolution of the disease in this study, we aimed to summarize our experience of managing a rare patient who underwent five surgeries for keloidal dermatofibroma that developed sequentially in the upper arm and chest and propose a novel treatment for keloidal dermatofibroma. We concluded that keloidal dermatofibroma involving larger areas, high tension sites, and multiple localizations can be treated using the principles of pathological scar management.展开更多

The programs of RevM an 5.3 and Stata 12.0 were used to analyze data.RESULTS: The best corrected visual acuity(BCVA) in combined therapy group were significantly better than those of anti-VEGF monotherapy group at 6,24 and 36 mo,with pooled weighted means differences(WMDs) of 0.12(0.06,0.18),0.25(0.12,0.38) and 0.28(0.13,0.43),respectively. The central retinal thickness(CRT) reductions in combined therapy group were higher than that in antiVEGF monotherapy group at 1,3,6 and 9mo,with pooled WMDs of 63.90(20.41,107.38),33.47(4.69,62.24),30.57(0.12,60.01) and 28.00(2.51,53.49),respectively. The regression rate of polyps in combined therapy group was much higher than that in anti-VEGF monotherapy group [RD: 0.47(0.26,0.68); P<0.0001]. The adverse event retinal hemorrhage did not differ significantly between the two groups.CONCLUSION: Our findings clearly document that antiVEGF combined with PDT is a more effective therapy for PCV compared with anti-VEGF monotherapy. Furthermore, combined therapy does not increase the incidence of retinal hemorrhage.展开更多

Photothermia combined with chemotherapy is anew and effective anti-tumor therapy. However, the drug loading of nanoparticles and increase in performance of photothermalconversion limits the therapeutic effect of combination therapy. In this study, two-dimensional boron (boron, B) nanoparticles wereprepared by ultrasonic exfoliation, and copper sulfide (CuS) nanoparticles and doxorubicin (DOX) were grown on the surface ofthe nanoparticles to form B-CuS-DOX nanoparticles. B-CuS carrier has high DOX drug loading capacity (864mg/g) and goodphotothermal conversion performance (photothermal conversion efficiency at 808nm is 55.8%). At the same time, it can achievedrug release and good photothermal response at near infrared and pH. The nanoparticles designed in this study are expected toprovide an effective chemotherapy-photothermal therapy strategy for tumor therapy in vivo.展开更多

The current study examines the effects of psycho-physiological therapy (stretch therapy combined with breathing exercise) on the sexual desire among heterosexual men. Methods: We used “convenience sampling” for this research;80 people were recruited. For collection of data, we used an identical quasi-experimental design called “nonequivalent control group.” Therapy sessions each lasting (20) 90 to 120 min were carried out on 3 alternate days of week. The volunteers have been selected from heterosexual men with stable relationship, those who married minimum of 6 months, within 20 to 55 years old, who attended to HUSM Family Clinic. Pre-tests, post-tests, and follow-up tests were conducted in a HUSM clinic (Malaysia). For assessment we used Hurlbert Index of Sexual Desire (HISD). Results: The psycho-physiological group had better post-test scores compared to the control group. Also follow-up test scores were marginally better compared to the control group, but this difference did not reach statistical significance. Also age and education level of participants didn’t have any significant effect on this intervention. Conclusion: Psycho-physiological combined therapy including stretching and breathing exercise appears to result in better improvement in sexual desire compared to control group but this improvement is not great. It increases sexual desire of heterosexual men, whenever people follow the therapy.展开更多

The exper...Present research was designated to investigate the hypoglycemic, hypolipidemic and antioxidant activity of the combination of pioglitazone and atorvastatin on long-term alloxan-induced diabetes rats (AIDRs). The experiments were carried out to determine blood glucose level, lipid profile, free radial scavenging activities, superoxide dismutase (SOD) and catalase in liver tissue. In addition, left ventricular (LV) hypertrophy and cardiomyocyte size were also determined by histological analysis. It was found that in short-term induction, pioglitazone significantly reduced the blood glucose level without having any considerable effect on lipid profile and antioxidant enzymes (SOD and catalase) in rats. On the other hand, atorvastatin significantly reduced total cholesterol (TC), triglyceride (TG) and low density lipoprotein cholesterol (LDL-C) with marked increase in the level of high density lipoprotein cholesterol (HDL-C) and improved activity of SOD and catalase enzymes. However, pathological changes of heart and pancreas were not observed after short-term exposure to alloxan in rats. Long-term diabetes induction resulted in LV hypertrophy and prominent shrinkage of islets of Langerhans cells. Treatment with atorvastatin in combination with pioglitazone significantly reduced the LV hypertrophy, TC, TG and LDL level whereas they noticeably increased HDL level, DPPH (1,1-Diphenyl-2-picryl-hydrazyl) free radical scavenging activity, SOD and catalase activity with satisfactory recovery of Langerhans cells. The result demonstrated that combination therapy was more effective than that of mono-therapy for preventing diabetes with cardiovascular diseases (CVD) in rats.展开更多

Methods:120 cases of patients with chronic bronchitis treated in our hospital were selected and randomly divided to be the therapeutic group and the control group, 60 cases each. Patients in control group were provided some basic treatments, such as spasmolysis, anti-inflammation, asthma relieving, oxygen inhalation and dissolve phlegm. For therapeutic group on this basis, Jin-Shui-Liu-Jun-Jian and Acupoint Application combined therapy was provided. Hemorheology, inflammatory factors and pulmonary functions in the two groups of patients were detected before and after treatment. Results:Differences of hemorheology indexes, inflammatory factors and pulmonary function relevant indexes between the two groups of patients with chronic bronchitis before treatment showed no statistical significance (P>0.05). Compared with prior treatment, hemorheology relevant indexes [whole blood high shear viscosity (WHV), whole blood low shear viscosity (WLV), packed cell volume (PCV), plasma viscosity (PV) and red cell assembling index (RCAI)], inflammatory factors [IL-8 (interleukin-8) and TNF-α(tumor necrosis factor-α)] were significantly decreased and pulmonary function relevant indexes [forced vital capacity (FVC), forced expiratory volume in one second (FEV1.0), peak expiratory flow (PEF), maximal voluntary ventilation (MVV) and expiratory flow rate when exhale vital capacity was 0.50], inflammatory factors (IL-4, IL-2) were significantly increased in the two groups of patients with chronic bronchitis after relevant treatments; The differences between the groups showed statistical significance (P<0.05). After combined therapy provided, pulmonary function relevant indexes (FVC, FEV1.0, PEF, MVV and V0.50) and inflammatory factors (IL-4, IL-2) in therapeutic group were significantly higher than control group after basic therapy. Hemorheology relevant indexes (WHV, WLV, PV, PCV and RCAI) and inflammatory factors (IL-8 and TNF-α) in therapeutic group were significantly lower than control group;Differences between the two groups showed statistical significance (P<0.05). Conclusions: Combination of Jin-Shui-Liu-Jun-Jian and Acupoint Application significantly improved the levels of hemorheology, inflammatory factors and pulmonary functions in patients with chronic bronchitis. It has a great important clinical significance on therapies for patients with chronic bronchitis.展开更多

Clinically, it is mainly characterized by arthralgia, swelling, and motor impairment. Since the knee joint is the load-bearing joint of the human body, and is susceptible to trauma, gonitis tends to have the highest morbidity in the four limbs, which manifests itself arthritis of patella and femur at the early stage; narrowness or disappearance of the medial joint space at the mid stage; and damage of the cartilage accompanied with flexion deformity at the late stage. In recent years, based on the experience of Prof. Cao Yiming, the author has treated 47 cases of gonitis by combined use of Chinese drugs and acupuncture, and obtained satisfactory therapeutic results as reported in the following.展开更多

Methods:Eighty-six patients of non-obesity PCOS, typed as Shen-deficiency with blood stasis Syndrome or Shen-deficiency with Phlegm-Dampness Syndrome by Syndrome Differentiation in traditional Chines medicine, were randomly divided into three groups: (1) The TCM group (n=26) was treated with Chinese drugs for 6 menstrual cycles; (2) The western medicine (WM) group (n=30) was treated with 1 tablet of CPA for 21 days, with the treatment beginning from the 5th day of menstruation. The treatment was given for 3 menstrual cycles by repetitious medication, which stopped and restarted on the 5th day of withdrawal bleeding. Then the ovulation promoting therapy was applied by using clomifene citrate and human chorionic gonadotropin (CC/Hcg) for 3 menstrual cycles; (3) The TCM-WM group (n=30) was treated with the medications used for the above two groups. The menstrual cycle, the volume and duration of the menstruation, as well as the improvement of acne and pilosis [Ferriman-Gallway (F-G) scoring]were observed after 3 cycles ended. Moreover, condition of ovulation was monitored by B-ultrasonography at the 4th-6th cycle and status of pregnancy was observed. Results: Compared with before treatment, the blood level of luteinizing hormone (LH) and testosterone (T) in all 3 groups after treatment significantly decreased (P<0.05), with its ratio to follicle-stimulating hormone (LH/FSH) recovered to normal, but without markedly change in levels of FSH, estradiol (E2) and prolactin (PRL). The menstrual cycle in most patients got regular and acne significantly alleviated (P<0.05), and the improvement of infrequent menstruation and acne was better in the WM group and the TCM-WM group than that in the TCM group, but pilosis showed no significant improvement in all three groups. The periodical ovulation rate in the TCM-WM group (73.1%) and the WM group (68.3%) was significantly higher than that in the TCM group (40%). The pregnancy rate in the TCM-WM group (53.8%) was significantly higher than that in the other two groups (26.1% and 25% respectively, all P<0.05). Conclusion: Using combined therapy of TCM and composite CPA followed by ovulation promoting agents of TCM and WM to treat patients of non-obesity PCOS could relieve the clinical symptoms, improve the abnormal blood level of sex hormones and significantly elevate the pregnancy rate.展开更多

文摘BACKGROUND: Thrombolysis therapy is not suitable for the elderly patients with acute ischemic stroke who delayed to be diagnosed for more than 3 hours, but traditional medicine is also not very ideal. CONCLUSION: Modified hemodilution combined therapy can improve hemorheology, decrease hematocrit, increase blood flow rate of middle cerebral artery, so as to improve the impaired clinical neurological function of elderly patients with acute cerebral thrombosis through anticoagulation and antiplatelet aggregative activity as well as regulating blood lipid.
文摘Dermatofibromas are benign soft tissue tumors that predominantly affect the limbs, and more rarely the chest.Keloidal dermatofibroma is a rare subtype with distinct clinicopathological features and an aggressive clinical course. By researching the evolution of the disease in this study, we aimed to summarize our experience of managing a rare patient who underwent five surgeries for keloidal dermatofibroma that developed sequentially in the upper arm and chest and propose a novel treatment for keloidal dermatofibroma. We concluded that keloidal dermatofibroma involving larger areas, high tension sites, and multiple localizations can be treated using the principles of pathological scar management.

文摘BACKGROUND Pulmonary epithelioid hemangioendothelioma(P-EHE)is a rare disease. Thus far, consensus on a standard treatment for P-EHE has not been established given its low incidence worldwide.Apatinib combined with chemotherapy with doxorubicin/cyclophosphamide has been used as an effective combination treatment for human malignancies. However, the efficacy of this combination has not been reported in P-EHE cases.CASE SUMMARY We present the case of a 64-year-old woman with chest tightness, cough, and chest pain.Computed tomography showed multiple unresectable pulmonary nodules. She had been misdiagnosed with lung carcinoma and underwent gefitinib treatment at a hospital.Subsequently, the patient underwent a cardiothoracic surgery for further disease investigation.CD31,CD34,and Vimentin expression were detected in the resected nodule specimens by immunohistochemical analyses, and pathological analyses confirmed the diagnosis of P-EHE.Following this, four cycles of apatinib combined with chemotherapy with doxorubicin/cyclophosphamide were initiated. The patient demonstrated stabilization of multiple bilateral nodules and showed a dramatic improvement in the clinical presentation after combination treatment. The patient could not tolerate the side effects of chemotherapy. Therefore, she then continued apatinib monotherapy, which is ongoing to date. The patient was stable at the last follow-up after 24 mo.CONCLUSION Apatinib combined with chemotherapy with doxorubicin/cyclophosphamide may be an effective therapeutic option for P-EHE treatment.
文摘AIM:To evaluate the efficacy and safety of a combined treatment for myopic choroidal neovascularization(CNV)using photodynamic therapy(PDT)and intravitreal bevacizumab and to compare it with intravitreal bevacizumab monotherapy.METHODS:Thirty-four eyes with angiographic evidence of myopic CNV were randomly divided into two groups:17 were treated with one intravitreal bevacizumab injection(1.25 mg)and low-fluence-rate PDT within seven days of the injection(Group A).The other 17 received monotherapy with bevacizumab injections(Group B).Clinical evidence of complications, best corrected visual acuity(BCVA)and fluorescein leakage were evaluated.BCVA and optical coherence tomography(OCT)were evaluated monthly. The timepoints follow-up was established at 6 and 12mo.All patients were retreated following a PRN protocol.RESULTS:A total of 34 eyes of 34 patients(26 women and 8 men)with a mean age of 62.35 years were included. In Group A(17 eyes)the mean BCVA increased from 0.55±0.13 logMAR before the treatment to 0.40±0.09 logMAR at the 12mo follow-up(P<0.01).In Group B(17 eyes)the mean BCVA increased from 0.60±0.11 logMAR before the treatment to 0.55±0.12 logMAR at the 12mo follow-up(P<0.01).There was no statistically significant difference between the two groups in terms of LogMar visual acuity. In Group A the mean number of combined treatments was 1.8±0.11 per patient;in Group B the mean number of intravitreal bevacizumab injections was3.1±0.08 per patient. The number of treatments was significantly fewer in Group A(P<0.01).No local or systemic side effects occurred among any of the patients treated in this study.CONCLUSION:The combination of anti-angiogenic injections and PDT appears to be a safe and effective option for myopic CNV treatment and allows for a significant reduction of intravitreal injections.
文摘AIM: To evaluate the efficacy and safety of anti-vascular endothelial growth factor(VEGF) combined with photodynamic therapy(PDT) versus anti-VEGF monotherapy for polypoidal choroidal vasculopathy(PCV).METHODS: We conducted a Meta-analysis of 9 studies to compare the efficacy and safety between combined therapy and anti-VEGF monotherapy for PCV. Furthermore, combined therapy does not increase the incidence of retinal hemorrhage.
文摘Single chemotherapy is difficult to meet the needs of tumor cure. The nanoparticles designed in this study are expected toprovide an effective chemotherapy-photothermal therapy strategy for tumor therapy in vivo.
文摘Objective: Hypoactive Sexual Desire is characterized as a lack or absence of sexual fantasies and desire for sexual activity. It increases sexual desire of heterosexual men, whenever people follow the therapy.

文摘AIM:To assess the efficacy of combined transcatheter arterial chemoembolization(TACE)and percutaneous microwave coagulation therapy(PMCT)for small hepatocellular carcinoma(HCC). METHODS:Thirty-five patients with a total of 41 HCC nodules(≤3 cm in diameter)were treated with TACE followed by computed tomograghy(CT)-guided percutaneous microwave coagulation therapy(PMCT) within 1-3 wk. RESULTS:By biopsies and enhanced CT scans, complete necrosis of the tumor and 3-5 mm of the surrounding non-cancerous area were observed in 34 foci.In seven foci, incomplete necrosis of the surrounding parenchyma was observed.Serum alpha- fetoprotein(AFP)levels returned to normal 10 d after treatment in 25 patients who originally had high serum AFP levels. The follow-up period was 6-31 mo,and all patients remained alive. One patient had a recurrence in the subsegments of the liver, and another patient had a recurrence near the original lesion. CONCLUSION:Combined therapy with TACE and PMCT is a safe and effective treatment without severe complications for small HCC.
文摘Present research was designated to investigate the hypoglycemic, hypolipidemic and antioxidant activity of the combination of pioglitazone and atorvastatin on long-term alloxan-induced diabetes rats (AIDRs). The result demonstrated that combination therapy was more effective than that of mono-therapy for preventing diabetes with cardiovascular diseases (CVD) in rats.
文摘The study developed a mild method to prepare partially reduced graphene oxide(pRGO).Through the non-covalent interaction of pRGO with nucleic acid aptamers AS1411 and indocytinine green(ICG),photothermal and photosensitive nanocomposites pRGO-AS1411-ICG(pRAI)were constructed. The complex structure and morphological characteristics of pRGO and pRAI were used by fourier transform infrared spectroscopy(FTIR),raman spectroscopy(Raman),X-ray photoelectron spectroscopy(XPS),ultraviolet-visible spectroscopy(Uv-vis),and transmission electron microscope(TEM).And the effect of EDS phototherapeutic back heat therapy on tumor cells was carried out in cell experiments. The results showed that pRAI with targeting of AS1411 can effectively kill tumor cells through the dual effects of photothermal therapy(PTT)and photodynamic therapy(PDT).
文摘Objective:To investigate the influencing mechanism of Jin-Shui-Liu-Jun-Jian and Acupoint Application combined therapy on hemorheology, inflammatory factors and pulmonary functions in patients with chronic bronchitis, thus to offer relevant help for clinical therapies on patients with chronic bronchitis. It has a great important clinical significance on therapies for patients with chronic bronchitis.
文摘Osteoarthritis, a nonspecific inflammatory lesion, is a commonly seen joint disease. Cao Yiming, the author has treated 47 cases of gonitis by combined use of Chinese drugs and acupuncture, and obtained satisfactory therapeutic results as reported in the following.
文摘Objective: To evaluate the effect of combined therapy of Chinese herbal medicine and compound cyproterone acetate (CPA) in treating non-obesity polycystic ovarian syndrome (PCOS) and to explore its mechanism in improving withdrawal ovulation. Conclusion: Using combined therapy of TCM and composite CPA followed by ovulation promoting agents of TCM and WM to treat patients of non-obesity PCOS could relieve the clinical symptoms, improve the abnormal blood level of sex hormones and significantly elevate the pregnancy rate.

文摘Glioblastoma(GBM)is the most aggressive malignant brain tumor.Due to the infiltration and heterogeneity of GBM,the obstruction of the blood-brain barrier(BBB)and the unique immunosuppressive mechanism,it is hard to achieve significant effects of GBM treatment. Here,a kind of chemotactic nanomotor that loaded with glucose oxidase(GOx)and carboxylated cisplatin(Pt(Ⅳ))prodrug on the L-arginine-derived polymer is proposed. The nanomotors are driven by catalysis of glucose decomposition and the positive chemotaxis towards the GBM microenvironment where inducible nitric oxide synthase and reactive oxygen species are highly expressed. This facilitates the BBB crossing and GBM targeting of the nanomotors. In addition, the released nitric oxide(NO)during propulsion as well as the loaded GOx and Pt(Ⅳ)can exert combined NO/starvation/chemotherapy.Meanwhile,it is able to induce and enhance the immune response through multiple pathways, thus better coping with the complexities of GBM treatment.

文摘In the present study,we investigated the synergistic effects of targeted methotrexate-selenium nanostructure containing Myc decoy oligodeoxynucleotides along with X-irradiation exposure as a combination therapy on LNCaP prostate cancer cells.Myc decoy ODNs were designed based on the promoter of Bcl-2 gene and analyzed by molecular docking and molecular dynamics assays.ODNs were loaded on the synthesized Se@BSA@Chi-MTX nanostructure. The physicochemical characteristics of nanostructures were determined by FTIR,DLS,UV-vis,TEM,EDX,in vitro release, and hemolysis tests.Subsequently, the cytotoxicity properties of them with and without X-irradiation were investigated by uptake,MTT,cell cycle, apoptosis, and scratch assays on the LNCaP cell line. The results of DLS and TEM showed negative charge(−9 mV)and nanometer size(40 nm)for Se@BSA@Chi-DEC-MTX NPs,respectively. The results of FTIR,UV-vis,and EDX showed the proper interaction of different parts and the correct synthesis of nanoparticles. The results of hemolysis showed the hemocompatibility of this nanoparticle in concentrations less than 6 mg/mL.The ODNs release from the nanostructures showed a pH-dependent manner, and the release rate was 15%higher in acidic pH.The targeted Se@BSA@Chi-labeled ODN-MTX NPs were efficiently taken up by LNCaP cells by targeting the prostate-specific membrane antigen(PSMA).The significant synergistic effects of nanostructure(containing MTX drug)treatment along with X-irradiation showed cell growth inhibition, apoptosis induction(~57%),cell cycle arrest(G2/M phase),and migration inhibition(up to 90%)compared to the control. The results suggested that the Se@BSA@Chi-DEC-MTX NPs can potentially suppress the cell growth of LNCaP cells. This nanostructure system can be a promising approach for targeted drug delivery and chemoradiotherapy in prostate cancer treatment.
