The PBLs were stimulated into cell cycle with phytohemagglutinin (PH...DNA damage response (DDR) in different cell cycle status of human peripheral blood lymphocytes (PBLs) and the role of H2AX in DDR were investigated. The PBLs were stimulated into cell cycle with phytohemagglutinin (PHA). The apoptotic ratio and the phosphorylation H2AX (S139) were flow cytometrically measured in resting and proliferating PBLs after treatment with camptothecin (CPT) or X-ray. The expressions of γH2AX, Bcl-2, caspase-3 and caspase-9 were detected by Western blotting. DDR in 293T cells was detected after H2AX was silenced by RNAi method. Our results showed that DNA double strand breaks (DSBs) were both induced in quiescent and proliferating PBLs after CPT or X-ray treatment. The phosphorylation of H2AX and apoptosis were more sensitive in proliferating PBLs compared with quiescent lymphocytes (P<0.05). The expression levels of anti-apoptotic proteins Bcl-2 were reduced and cleaved caspase-3 and caspase-9 were increased. No significant changes were observed in CPT-induced apoptosis in 293T cells between H2AX knocking down group and controls. It was concluded that proliferating PBLs were more vulnerable to DNA damage compared to non-stimulated lymphocytes and had higher apoptosis rates. γH2AX may only serve as a marker of DNA damage but exert no effect on apoptosis regulation.展开更多

Methods: CD13 expression in peripheral blood lymphocytes and skin lesions was detected by flow cytometry and immunohistochemical technique, respectively. Results were compared with those of healthy controls. Results: CD13 expression was significantly higher in peripheral blood lymphocytes of patients with advanced psoriasis vulgaris than in that of healthy controls, and in skin lesions than in healthy skin tissues. The expression was mainly in the suprabasal layers of skin lesions, and positively correlated to PASI (R=0.78029). Conclusion: The significantly higher expression of CD13 in peripheral blood lymphocytes and skin lesions of the patients with advanced psoriasis vulgaris probably is related to immunological abnormality, blood vessel abnormality and proliferation of keratinocyte in the pathogenic course of psoriasis. It may be a novel and effective way to treat psoriasis with specific CD13 inhibitors.展开更多

Methods The expression of mRNA representing the seven active MT-1 genes was determined in HPBLs by quantitative RT-PCR before and after exposure to cadmium. Results Basal expressions of MT-1X, and MT-1A in HPBLs were similar to expression of housekeeping gene. In contrast, the basal gene expressions of MT-1H, 1F, 1E, and 1G were a little transcripts in human HPBLs. No signal was detected for MT-1B. There was a sex difference (P<0.05). in basal gene expression of MT-1E. The levels of gene expression of MT-1A, 1E, 1F, 1G, 1H, and 1X increased, but the level of MT-1B did not increase after exposure to cadmium. Conclusions Gene expressions of MT-1G, MT-1H, MT-1F, and MT-1X in HPBLs can be used as a potential biomarker of cadmium exposure.展开更多

Lymphocytes were isolated by Percoll density gradient centrifugation from peripheral blood samples,and electrophysiological characters of the channel were analyzed. In the recorded cells,activated voltage of the channels was-42.5 ± 3.7 mV and the average peak current was 313.12 ± 28.2 pA. The channel was identified as voltage dependent,the current was outward and it could be inhibited by 10 mmol/dm 3 TEA or 5 mmol/dm 3 4-AP,a specific potassium channel inhibitor,identifying the existence of potassium channel in peripheral lymphocytes of the turbot.展开更多

Methods: CD25 percentages (CD25%...Objective: To study expression of membrane receptors of interleukin-2 (CD25) on the peripheral blood lymphocytes (PBL) of patients with colorectal cancer and its clinical significance. Methods: CD25 percentages (CD25%) in PBL of 105 colorectal cancer patients before operation and 100 normal individuals were examined by flow cytometer, and the results were clinically and pathologically analyzed. Results: The mean of CD25% in PBL of the normal individuals was 17.24(5.33, it was significantly lower (p<0.01) than that of the colon cancer patients (21.29(7.95) or rectal cancer patients (21.62(6.11). In contrast to the normal individuals, the means of CD25% in PBL in ulcer type (20.53(6.50) or protruded type (21.56(6.16) colorectal cancer patients were notably elevated (P<0.01). The significant difference (P<0.01) of means of CD25% in PBL was observed between the normal individuals and patients with less than 4 cm mass (22.10(5.43) or 4cm(8cm mass (20.90(6.96). The significant difference (P<0.05) of means of CD25% in PBL was also observed between the normal individuals and patients with greater than 8 cm mass (21.56(5.41). The mean of CD25% in PBL in patients with well differentiation colorectal cancer was 22.20(5.50, it was significantly higher than that in normal individuals (P<0.05). The means of CD25% in PBL in patients with middle or poor differentiation colorectal cancer were 21.30(6.89 and 22.15(5.71 respectively, they were obviously higher than that in normal individuals (P<0.01). The significant difference (P<0.01) of means of CD25% in PBL was present between the colorectal cancer patients without metastatic lymph nodes (22.06(6.90) and normal individuals. The significant difference (P<0.05) of means of CD25% in PBL was present between the colorectal cancer patients with metastatic lymph nodes (20.73(6.40) and normal individuals. The means of CD25% in PBL in colorectal cancer patients in various clinic stages were significantly higher than that in the health subjects (p<0.01). The significant difference (p<0.01) of means of CD25% in PBL was present between the patients whose ages were equal to or less than 60 (21.00(5.76) and normal individuals in the same age group. The significant difference (p<0.05) of means of CD25% in PBL was also present between the patients whose ages were greater than 60 (22.54(7.75) and normal individuals in the same age group. The significant difference (P<0.01) of means of CD25% in PBL was present between the male patients (22.55(7.05) and normal men. The significant difference (p<0.05) of means of CD25% in PBL was also present between the female patients (20.09(5.48) and normal women. Conclusion: The mean of CD25% in PBL of colorectal cancer patients was significantly higher than that in health subjects. Abnormally elevated CD25% were correlative with site of tumor growth, macropathology type of tumor, the degree of tumor differentiation, clinical stage and patient’s age and sex. It may be helpful to detect CD25% in PBL of colorectal cancer patients before operation for diagnosis, immune treatment and judging prognosis.展开更多

Phytohemagglutinin( PHA),concanavalin A( Con A) and lipopolysaccharide( LPS) were used as lymphocyte proliferation mitogen,respectively. According to L25( 56) five-factor five-level orthogonal experimental design,the conditions for sturgeons proliferation response of peripheral blood lymphocytes were optimized using enhanced cell counting Kit-8( enhanced CCK-8 or WST-8). Five factors were selected to explore the optimal response conditions,including culture time,culture temperature,cell concentration,fetal bovine serum( FBS) concentration,and mitogen concentration. The results showed that 70% Percoll( 1. 092 g/ml) used as the sturgeon lymphocyte separation solution had the best separating effect. In this study, we aimed to detect the methylation status of GLDC promoter and its diagnosti...Background: Glycine dehydrogenase(GLDC) plays an important role in the initiation and proliferation of several human cancers. In this study, we aimed to detect the methylation status of GLDC promoter and its diagnostic value for hepatitis B virus-associated hepatocellular carcinoma(HBV-HCC). Methods: We enrolled 197 patients, 111 with HBV-HCC, 51 with chronic hepatitis B(CHB), and 35 healthy controls(HCs). The methylation status of GLDC promoter in peripheral mononuclear cells(PBMCs) was identified by methylation specific polymerase chain reaction(MSP). The mRNA expression was examined using real-time quantitative polymerase chain reaction(q PCR). Results: The methylation frequency of the GLDC promoter was significantly lower in HBV-HCC patients(27.0%) compared to that in CHB patients(68.6%) and HCs(74.3%)( P < 0.001). The methylated group had lower alanine aminotransferase level( P = 0.035) and lower rates of tumor node metastasis(TNM) Ⅲ/Ⅳ( P = 0.043) and T3/T4( P = 0.026). TNM stage was identified to be an independent factor for GLDC promoter methylation. GLDC mRNA levels in CHB patients and HCs were significantly lower than those in HBV-HCC patients( P = 0.022 and P < 0.001, respectively). GLDC mRNA levels were significantly higher in HBV-HCC patients with unmethylated GLDC promoters than those with methylated GLDC promoters( P = 0.003). The diagnostic accuracy of alpha-fetoprotein(AFP) combined with GLDC promoter methylation for HBV-HCC was improved compared with that of AFP alone(AUC: 0.782 vs. 0.630, P < 0.001). In addition, GLDC promoter methylation was an independent predictor for overall survival of HBV-HCC patients( P = 0.038). Conclusions: The methylation frequency of GLDC promoter was lower in PBMCs from HBV-HCC patients than that from patients with CHB and HCs. The combination of AFP and GLDC promoter hypomethylation significantly improved the diagnostic accuracy of HBV-HCC.展开更多
The study aimed to detect the expression of the Th1-specific cell surface protein T cell Ig and mucin domain-containing molecule-3(Tim-3)mRNA peripheral blood lymphocytes isolated from asthmaticin patients and to exami...The study aimed to detect the expression of the Th1-specific cell surface protein T cell Ig and mucin domain-containing molecule-3(Tim-3)mRNA peripheral blood lymphocytes isolated from asthmaticin patients and to examine the correlation among Tim-3 mRNA,interleukin-4(IL-4),interferon-γ(IFN-γ)level,and FEV1/FVC(force expiratory volume in one second/forced vital capacity)to explore the role of Tim-3 in the development and progression of asthmatic inflammation.Tim-3 mRNA expression was detected by reverse transcription polymerase chain reaction(RT-PCR).The IL-4 and IFN-γlevels were determined by using enzyme-linked immunosorbent assay(ELISA).The correlation among Tim-3 mRNA,IL-4,IFN-γlevel,and pulmonary ventilatory capacity was analyzed.The expres-sion of Tim-3 mRNA in patients with acute asthma exacerbation was 0.39�0.06,significantly higher than that in patients at the remission stage and controls(0.18�0.05 and 0.07�0.03,P<0.05).The level of IL-4 in patients with acute asthma exacerbation was 68.42�10.54,significantly higher than that in the patients at the remission stage and controls(41.83�9.37 and 32.75�8.16,P<0.05).The level of IL-4 in patients in remission was significantly higher than that in controls(P<0.05).The level of IFN-γin patients with acute asthma exacerbation was 65.74�7.85,significantly lower than that in patients in remission and the control group(120.84�11.62 and 139.65�13.47,P<0.05).The level of IFN-γin patients in asthma remission was significantly lower than that in controls(P<0.05).Tim-3 mRNA expression was positively correlated with the level of IL-4(r=0.68,P<0.05)and negatively with the level of IFN-γand pulmonary ventilatory capacity(r=–0.85,r=–0.76,both P<0.01).The increased expression of Tim-3 mRNA in peripheral blood lymphocytes might be involved in the development and progression of asthmatic inflammation.展开更多

BACKGROUND Programmed death 1(PD-1)and CD4^(+)CD25^(+)FoxP3^(+)expression in peripheral blood T-cells has been previously reported in various types of cancer.However,the specific variation tendency during surgery and ...BACKGROUND Programmed death 1(PD-1)and CD4^(+)CD25^(+)FoxP3^(+)expression in peripheral blood T-cells has been previously reported in various types of cancer.However,the specific variation tendency during surgery and chemotherapy,as well as their relationship in gastric cancer patients,still remain unclear.Understanding this aspect may provide some novel insights for future studies on tumor recurrence and tumor immune escape,and also serve as a reference for determining the optimal timing and dose of clinical anti-PD-1 antibodies.AIM To observe and analyze the expression characteristics of peripheral lymphocyte PD-1 and FoxP3^(+)regulatory T cells(FoxP3^(+)Tregs)before and after surgery or chemotherapy in gastric cancer patients.METHODS Twenty-nine stomach cancer patients undergoing chemotherapy after a D2 gastrectomy provided 10 mL peripheral blood samples at each phase of the perioperative period and during chemotherapy.This study also included 29 agematched healthy donors as a control group.PD-1 expression was detected on lymphocytes,including CD4^(+)CD8^(+)CD45RO^(+),CD4^(+)CD45RO^(+),and CD8^(+)CD45RO^(+)lymphocytes as well as regulatory T cells.RESULTS We observed a significant increase of PD-1 expression on immune subsets and a larger number of FoxP3^(+)Tregs in gastric cancer patients(P<0.05).Following D2 gastrectomy,peripheral lymphocytes PD-1 expression and the number of FoxP3^(+)Tregs notably decrease(P<0.05).However,during postoperative chemotherapy,we only observed a decrease in PD-1 expression on lymphocytes in the CD8^(+)CD45RO^(+)and CD8^(+)CD45RO^(+)populations.Additionally,linear correlation analysis indicated a positive correlation between PD-1 expression and the number of CD4^(+)CD45RO^(+)FoxP3high activated Tregs(aTregs)on the total peripheral lymphocytes(r=0.5622,P<0.0001).CONCLUSION The observed alterations in PD-1 expression and the activation of regulatory T cells during gastric cancer treatment may offer novel insights for future investigations into tumor immune evasion and the clinical application of anti-PD-1 antibodies in gastric cancer.展开更多
Oral squamous cell carcinoma(OSCC)is a malignant tumor triggered by the accumulation of multiple gene mutations in oral epithelial cells.Different OSCC-related biomarkers have been reported in circulation in the perip...Oral squamous cell carcinoma(OSCC)is a malignant tumor triggered by the accumulation of multiple gene mutations in oral epithelial cells.Different OSCC-related biomarkers have been reported in circulation in the peripheral blood that support the occurrence and development of OSCC.Recent advances in high-throughput and highly sensitive detection methods have overcome the limitation of the low concentration of most peripheral blood biomarkers.Hence,blood biomarker detection has become an efficient screening tool for the early diagnosis of OSCC.The growing data available in public cancer and gene databases have provided new foundations for OSCC research.In particular,the identification of OSCC biomarkers using bioinformatic tools has shed new light on the underlying mechanisms as well as on the genetic landscape of OSCC.More recently,mRNA targeting therapies have emerged as valuable anticancer treatment strategies,as they allow for the regulation of the expression of certain functional proteins to reverse genetic abnormalities or induce tissue repair.Thus,mRNA-targeting therapies can be used to regulate the expression of antigens,antibodies,or cellular receptors by immune cells.Particularly,anti-cancer cellular immunotherapy carrying specific mRNAs has attracted significant attention in OSCC treatment.Here,we review the present knowledge on the role of peripheral blood mRNAs in the diagnosis,treatment,development,and prognosis of OSCC.Moreover,we address future research prospects of mRNAs in the peripheral blood in OSCC and the opportunities and challenges that may arise in future clinical therapeutic applications.展开更多
Objective:To compare the effect of human chorionic gonadotropin(hCG)-producing peripheral blood mononuclear cells(PBMCs)and PBMCs activated by hCG in vitro and expressions of related immune genes in mouse implantation...Objective:To compare the effect of human chorionic gonadotropin(hCG)-producing peripheral blood mononuclear cells(PBMCs)and PBMCs activated by hCG in vitro and expressions of related immune genes in mouse implantation.Methods:hCG-producing PBMCs(transfected PBMC)and PBMCs activated by hCG in vitro were introduced into isolated mouse endometrial cells,while cell cultures were divided into four groups:the control,PBMC,transfected,and activated PBMC groups.The expression of studied genes(IL-1β,IL-6,Lif,and Vegf)was evaluated and blastocyst attachment on the cocultured cells(isolated endometrial cells and PBMC cells)was monitored in all four groups.Results:Data showed that expression decreased in the PBMC group compared to the treated PBMC(transfected and activated PBMCs)and increased in transfected PBMC compared to the activated PBMC.Attachment and migration of blastocysts were dramatically enhanced in the transfected PBMC group compared to the activated PBMC group(P<0.05).Conclusions:Use of hCG-producing PBMCs(transfected PBMC)has more influence on endometrial receptivity.展开更多
In recent years,Peripheral blood smear is a generic analysis to assess the person’s health status.Manual testing of Peripheral blood smear images are difficult,time-consuming and is subject to human intervention and ...In recent years,Peripheral blood smear is a generic analysis to assess the person’s health status.Manual testing of Peripheral blood smear images are difficult,time-consuming and is subject to human intervention and visual error.This method encouraged for researchers to present algorithms and techniques to perform the peripheral blood smear analysis with the help of computer-assisted and decision-making techniques.Existing CAD based methods are lacks in attaining the accurate detection of abnormalities present in the images.In order to mitigate this issue Deep Convolution Neural Network(DCNN)based automatic classification technique is introduced with the classification of eight groups of peripheral blood cells such as basophil,eosinophil,lymphocyte,monocyte,neutrophil,erythroblast,platelet,myocyte,promyocyte and metamyocyte.The proposed DCNN model employs transfer learning approach and additionally it carries three stages such as pre-processing,feature extraction and classification.Initially the pre-processing steps are incorporated to eliminate noisy contents present in the image by using Histogram Equalization(HE).It is enclosed to improve an image contrast.In order to distinguish the dissimilar class and segmentation approach is carried out with the help of Fuzzy C-Means(FCM)model whereas its centroid point optimality method with Slap Swarm based optimization strategy.Moreover some specific set of Gray Level Co-occurrence Matrix(GLCM)features of the segmented images are extracted to augment the performance of proposed detection algorithm.Finally the extracted features are recorded by DCNN and the proposed classifier has the capability to extract their own features.Based on this the diverse set of classes are classified and distinguished from qualitative abnormalities found in the image.展开更多

The diagnosis of COPD is changed to acute exacerbation of COPD (AECOPD) when respiratory symptoms become worse, beyond normal day-to-day variations and severely enough that changes in medication are required. Both neutrophils to lymphocyte ratio (NLR) and peripheral blood eosinophilia (PBE) are rapid and relatively inexpensive tests that can be easily applied in the clinical practice for the diagnosis and treatment of AECOPD patients. Furthermore, current studies found that NLR and PBE had a higher accuracy rate than other traditional markers (Leukocyte count and C-reactive protein) for the diagnosis and management of AECOPD. Besides, recent studies determined that NLR and PBE can be used for prediction of future exacerbations in COPD patients. This review aims to explore the current knowledge about the significance of NLR and PBE in AECOPD patients.展开更多

METHODS: We enrolled 70 cancer patients (33 gastric cancer, 37 color...AIM: To assess the absolute number of T-regulatory cells (Tregs; CD4+CD25+Foxp3+) in the peripheral blood of gastric and colorectal cancer patients. METHODS: We enrolled 70 cancer patients (33 gastric cancer, 37 colorectal cancer) and 17 healthy volunteers. The CD3+CD4+ lymphocytes and CD4+CD25+Foxp3+ Tregs in the peripheral blood were analyzed with flow cytometry. The absolute numbers of Tregs were calculated based on the CD4+CD25+Foxp3+ cells percent-age of CD3+CD4+ cells and the absolute numbers of CD3+CD4+ cells per microliter. RESULTS: The mean number of CD4+CD25+Foxp3+ cells per microliter in colorectal cancer patients was 15.7 (SD: 21.8), for gastric cancer patients 12.2 (SD: 14.3), and for controls 17.5 (SD: 11.4). The absolute number of Tregs was significantly lower in gastric cancer patients than in controls (P = 0.026). There was no statistically significant difference for gastric vs colorectal cancer or colorectal cancer vs controls. The absolute number of Tregs was also significantly depressed in N+ vs Ncancer patients [22.0 (27.7) vs 10.1 (9.0), P = 0.013], and in the subgroup of gastric cancer patients [30.3 (27.6) vs 9.6 (8.0), P = 0.003]. No statistical difference was observed in the proportion of Tregs in the CD4+ population between the groups. CONCLUSION: The absolute number of Tregs in peripheral blood of gastric cancer but not colorectal cancer patients was significantly decreased in comparison with that in healthy controls.展开更多

The PBLs were stimulated into cell cycle with phytohemagglutinin (PHA). The apoptotic ratio and the phosphorylation H2AX (S139) were flow cytometrically measured in resting and proliferating PBLs after treatment with camptothecin (CPT) or X-ray. The expressions of γH2AX, Bcl-2, caspase-3 and caspase-9 were detected by Western blotting. DDR in 293T cells was detected after H2AX was silenced by RNAi method. Our results showed that DNA double strand breaks (DSBs) were both induced in quiescent and proliferating PBLs after CPT or X-ray treatment. The phosphorylation of H2AX and apoptosis were more sensitive in proliferating PBLs compared with quiescent lymphocytes (P<0.05). The expression levels of anti-apoptotic proteins Bcl-2 were reduced and cleaved caspase-3 and caspase-9 were increased. No significant changes were observed in CPT-induced apoptosis in 293T cells between H2AX knocking down group and controls. It was concluded that proliferating PBLs were more vulnerable to DNA damage compared to non-stimulated lymphocytes and had higher apoptosis rates. γH2AX may only serve as a marker of DNA damage but exert no effect on apoptosis regulation.
文摘Objective: To observe the expression of CD13/APN in peripheral blood lymphocytes and skin lesions of patients with advanced psoriasis vulgaris, and discuss its effect on the pathogenesis of psoriasis. Methods: CD13 expression in peripheral blood lymphocytes and skin lesions was detected by flow cytometry and immunohistochemical technique, respectively. Results were compared with those of healthy controls. Results: CD13 expression was significantly higher in peripheral blood lymphocytes of patients with advanced psoriasis vulgaris than in that of healthy controls, and in skin lesions than in healthy skin tissues. The expression was mainly in the suprabasal layers of skin lesions, and positively correlated to PASI (R=0.78029). Conclusion: The significantly higher expression of CD13 in peripheral blood lymphocytes and skin lesions of the patients with advanced psoriasis vulgaris probably is related to immunological abnormality, blood vessel abnormality and proliferation of keratinocyte in the pathogenic course of psoriasis. It may be a novel and effective way to treat psoriasis with specific CD13 inhibitors.

文摘Objective To study the gene expression of metallothionein 1 (MT-1) isoforms in human peripheral blood lymphocytes (HPBLs). Methods The expression of mRNA representing the seven active MT-1 genes was determined in HPBLs by quantitative RT-PCR before and after exposure to cadmium. Results Basal expressions of MT-1X, and MT-1A in HPBLs were similar to expression of housekeeping gene. In contrast, the basal gene expressions of MT-1H, 1F, 1E, and 1G were a little transcripts in human HPBLs. No signal was detected for MT-1B. There was a sex difference (P<0.05). in basal gene expression of MT-1E. The levels of gene expression of MT-1A, 1E, 1F, 1G, 1H, and 1X increased, but the level of MT-1B did not increase after exposure to cadmium. Conclusions Gene expressions of MT-1G, MT-1H, MT-1F, and MT-1X in HPBLs can be used as a potential biomarker of cadmium exposure.

文摘In order to provide pertinent evidence of ion channel with immune response in the fish,whole cell patch-clamp technique was employed for potassium ion channel study in turbot(Scophthalmus maximus) . Lymphocytes were isolated by Percoll density gradient centrifugation from peripheral blood samples,and electrophysiological characters of the channel were analyzed. In the recorded cells,activated voltage of the channels was-42.5 ± 3.7 mV and the average peak current was 313.12 ± 28.2 pA. The channel was identified as voltage dependent,the current was outward and it could be inhibited by 10 mmol/dm 3 TEA or 5 mmol/dm 3 4-AP,a specific potassium channel inhibitor,identifying the existence of potassium channel in peripheral lymphocytes of the turbot.
文摘Objective: To study expression of membrane receptors of interleukin-2 (CD25) on the peripheral blood lymphocytes (PBL) of patients with colorectal cancer and its clinical significance. Methods: CD25 percentages (CD25%) in PBL of 105 colorectal cancer patients before operation and 100 normal individuals were examined by flow cytometer, and the results were clinically and pathologically analyzed. Results: The mean of CD25% in PBL of the normal individuals was 17.24(5.33, it was significantly lower (p<0.01) than that of the colon cancer patients (21.29(7.95) or rectal cancer patients (21.62(6.11). In contrast to the normal individuals, the means of CD25% in PBL in ulcer type (20.53(6.50) or protruded type (21.56(6.16) colorectal cancer patients were notably elevated (P<0.01). The significant difference (P<0.01) of means of CD25% in PBL was observed between the normal individuals and patients with less than 4 cm mass (22.10(5.43) or 4cm(8cm mass (20.90(6.96). The significant difference (P<0.05) of means of CD25% in PBL was also observed between the normal individuals and patients with greater than 8 cm mass (21.56(5.41). The mean of CD25% in PBL in patients with well differentiation colorectal cancer was 22.20(5.50, it was significantly higher than that in normal individuals (P<0.05). The means of CD25% in PBL in patients with middle or poor differentiation colorectal cancer were 21.30(6.89 and 22.15(5.71 respectively, they were obviously higher than that in normal individuals (P<0.01). The significant difference (P<0.01) of means of CD25% in PBL was present between the colorectal cancer patients without metastatic lymph nodes (22.06(6.90) and normal individuals. The significant difference (P<0.05) of means of CD25% in PBL was present between the colorectal cancer patients with metastatic lymph nodes (20.73(6.40) and normal individuals. The means of CD25% in PBL in colorectal cancer patients in various clinic stages were significantly higher than that in the health subjects (p<0.01). The significant difference (p<0.01) of means of CD25% in PBL was present between the patients whose ages were equal to or less than 60 (21.00(5.76) and normal individuals in the same age group. The significant difference (p<0.05) of means of CD25% in PBL was also present between the patients whose ages were greater than 60 (22.54(7.75) and normal individuals in the same age group. The significant difference (P<0.01) of means of CD25% in PBL was present between the male patients (22.55(7.05) and normal men. The significant difference (p<0.05) of means of CD25% in PBL was also present between the female patients (20.09(5.48) and normal women. Conclusion: The mean of CD25% in PBL of colorectal cancer patients was significantly higher than that in health subjects. Abnormally elevated CD25% were correlative with site of tumor growth, macropathology type of tumor, the degree of tumor differentiation, clinical stage and patient’s age and sex. It may be helpful to detect CD25% in PBL of colorectal cancer patients before operation for diagnosis, immune treatment and judging prognosis.

文摘In this study,sterlet( Acipenser ruthenus) was chosen as the model species of sturgeon,different solutions were used to isolate the sturgeon peripheral blood lymphocytes and study their optimal proliferate response condition. Phytohemagglutinin( PHA),concanavalin A( Con A) and lipopolysaccharide( LPS) were used as lymphocyte proliferation mitogen,respectively. According to L25( 56) five-factor five-level orthogonal experimental design,the conditions for sturgeons proliferation response of peripheral blood lymphocytes were optimized using enhanced cell counting Kit-8( enhanced CCK-8 or WST-8). Five factors were selected to explore the optimal response conditions,including culture time,culture temperature,cell concentration,fetal bovine serum( FBS) concentration,and mitogen concentration. The results showed that 70% Percoll( 1. 092 g/ml) used as the sturgeon lymphocyte separation solution had the best separating effect. The optimal proliferation conditions were as follows: 3. 625 × 10~6 initial cells,20 μg/ml PHA or 50 μg/ml Con A or 10 μg/ml LPS as mitogen,10%-20% FBS,the temperature at 20-25 ℃,and the culture time of 2 d.
文摘Human peripheral blood exposed with 188ReO4 at various radioactivities for 54h was examined to observe the chromosome and chromatin aberrations at metaphases in the mitosis of lymphocytes. The findings indicate that there is an increasing tendency of the aberration yields of both types with increasing 188ReO4 concentration and a dose dependency could be obtained. The aberration yield was best fitted by a quadratic model. Chromatin aberrations, aberration cells and chromosome fragments were fitted into a linear-regression model.

文摘Background: Glycine dehydrogenase(GLDC) plays an important role in the initiation and proliferation of several human cancers. In this study, we aimed to detect the methylation status of GLDC promoter and its diagnostic value for hepatitis B virus-associated hepatocellular carcinoma(HBV-HCC). Methods: We enrolled 197 patients, 111 with HBV-HCC, 51 with chronic hepatitis B(CHB), and 35 healthy controls(HCs). The methylation status of GLDC promoter in peripheral mononuclear cells(PBMCs) was identified by methylation specific polymerase chain reaction(MSP). The mRNA expression was examined using real-time quantitative polymerase chain reaction(q PCR). Results: The methylation frequency of the GLDC promoter was significantly lower in HBV-HCC patients(27.0%) compared to that in CHB patients(68.6%) and HCs(74.3%)( P < 0.001). The methylated group had lower alanine aminotransferase level( P = 0.035) and lower rates of tumor node metastasis(TNM) Ⅲ/Ⅳ( P = 0.043) and T3/T4( P = 0.026). TNM stage was identified to be an independent factor for GLDC promoter methylation. GLDC mRNA levels in CHB patients and HCs were significantly lower than those in HBV-HCC patients( P = 0.022 and P < 0.001, respectively). GLDC mRNA levels were significantly higher in HBV-HCC patients with unmethylated GLDC promoters than those with methylated GLDC promoters( P = 0.003). The diagnostic accuracy of alpha-fetoprotein(AFP) combined with GLDC promoter methylation for HBV-HCC was improved compared with that of AFP alone(AUC: 0.782 vs. 0.630, P < 0.001). In addition, GLDC promoter methylation was an independent predictor for overall survival of HBV-HCC patients( P = 0.038). Conclusions: The methylation frequency of GLDC promoter was lower in PBMCs from HBV-HCC patients than that from patients with CHB and HCs. The combination of AFP and GLDC promoter hypomethylation significantly improved the diagnostic accuracy of HBV-HCC.

文摘The study aimed to detect the expression of the Th1-specific cell surface protein T cell Ig and mucin domain-containing molecule-3(Tim-3)mRNA peripheral blood lymphocytes isolated from asthmaticin patients and to examine the correlation among Tim-3 mRNA,interleukin-4(IL-4),interferon-γ(IFN-γ)level,and FEV1/FVC(force expiratory volume in one second/forced vital capacity)to explore the role of Tim-3 in the development and progression of asthmatic inflammation.Tim-3 mRNA expression was detected by reverse transcription polymerase chain reaction(RT-PCR).The IL-4 and IFN-γlevels were determined by using enzyme-linked immunosorbent assay(ELISA).The correlation among Tim-3 mRNA,IL-4,IFN-γlevel,and pulmonary ventilatory capacity was analyzed.The expres-sion of Tim-3 mRNA in patients with acute asthma exacerbation was 0.39�0.06,significantly higher than that in patients at the remission stage and controls(0.18�0.05 and 0.07�0.03,P<0.05).The level of IL-4 in patients with acute asthma exacerbation was 68.42�10.54,significantly higher than that in the patients at the remission stage and controls(41.83�9.37 and 32.75�8.16,P<0.05).The level of IL-4 in patients in remission was significantly higher than that in controls(P<0.05).The level of IFN-γin patients with acute asthma exacerbation was 65.74�7.85,significantly lower than that in patients in remission and the control group(120.84�11.62 and 139.65�13.47,P<0.05).The level of IFN-γin patients in asthma remission was significantly lower than that in controls(P<0.05).Tim-3 mRNA expression was positively correlated with the level of IL-4(r=0.68,P<0.05)and negatively with the level of IFN-γand pulmonary ventilatory capacity(r=–0.85,r=–0.76,both P<0.01).The increased expression of Tim-3 mRNA in peripheral blood lymphocytes might be involved in the development and progression of asthmatic inflammation.

文摘BACKGROUND Programmed death 1(PD-1)and CD4^(+)CD25^(+)FoxP3^(+)expression in peripheral blood T-cells has been previously reported in various types of cancer.However,the specific variation tendency during surgery and chemotherapy,as well as their relationship in gastric cancer patients,still remain unclear.Understanding this aspect may provide some novel insights for future studies on tumor recurrence and tumor immune escape,and also serve as a reference for determining the optimal timing and dose of clinical anti-PD-1 antibodies.AIM To observe and analyze the expression characteristics of peripheral lymphocyte PD-1 and FoxP3^(+)regulatory T cells(FoxP3^(+)Tregs)before and after surgery or chemotherapy in gastric cancer patients.METHODS Twenty-nine stomach cancer patients undergoing chemotherapy after a D2 gastrectomy provided 10 mL peripheral blood samples at each phase of the perioperative period and during chemotherapy.This study also included 29 agematched healthy donors as a control group.PD-1 expression was detected on lymphocytes,including CD4^(+)CD8^(+)CD45RO^(+),CD4^(+)CD45RO^(+),and CD8^(+)CD45RO^(+)lymphocytes as well as regulatory T cells.RESULTS We observed a significant increase of PD-1 expression on immune subsets and a larger number of FoxP3^(+)Tregs in gastric cancer patients(P<0.05).Following D2 gastrectomy,peripheral lymphocytes PD-1 expression and the number of FoxP3^(+)Tregs notably decrease(P<0.05).However,during postoperative chemotherapy,we only observed a decrease in PD-1 expression on lymphocytes in the CD8^(+)CD45RO^(+)and CD8^(+)CD45RO^(+)populations.Additionally,linear correlation analysis indicated a positive correlation between PD-1 expression and the number of CD4^(+)CD45RO^(+)FoxP3high activated Tregs(aTregs)on the total peripheral lymphocytes(r=0.5622,P<0.0001).CONCLUSION The observed alterations in PD-1 expression and the activation of regulatory T cells during gastric cancer treatment may offer novel insights for future investigations into tumor immune evasion and the clinical application of anti-PD-1 antibodies in gastric cancer.

文摘Oral squamous cell carcinoma(OSCC)is a malignant tumor triggered by the accumulation of multiple gene mutations in oral epithelial cells.Different OSCC-related biomarkers have been reported in circulation in the peripheral blood that support the occurrence and development of OSCC.Recent advances in high-throughput and highly sensitive detection methods have overcome the limitation of the low concentration of most peripheral blood biomarkers.Hence,blood biomarker detection has become an efficient screening tool for the early diagnosis of OSCC.The growing data available in public cancer and gene databases have provided new foundations for OSCC research.In particular,the identification of OSCC biomarkers using bioinformatic tools has shed new light on the underlying mechanisms as well as on the genetic landscape of OSCC.More recently,mRNA targeting therapies have emerged as valuable anticancer treatment strategies,as they allow for the regulation of the expression of certain functional proteins to reverse genetic abnormalities or induce tissue repair.Thus,mRNA-targeting therapies can be used to regulate the expression of antigens,antibodies,or cellular receptors by immune cells.Particularly,anti-cancer cellular immunotherapy carrying specific mRNAs has attracted significant attention in OSCC treatment.Here,we review the present knowledge on the role of peripheral blood mRNAs in the diagnosis,treatment,development,and prognosis of OSCC.Moreover,we address future research prospects of mRNAs in the peripheral blood in OSCC and the opportunities and challenges that may arise in future clinical therapeutic applications.
文摘Objective:To compare the effect of human chorionic gonadotropin(hCG)-producing peripheral blood mononuclear cells(PBMCs)and PBMCs activated by hCG in vitro and expressions of related immune genes in mouse implantation.Methods:hCG-producing PBMCs(transfected PBMC)and PBMCs activated by hCG in vitro were introduced into isolated mouse endometrial cells,while cell cultures were divided into four groups:the control,PBMC,transfected,and activated PBMC groups.The expression of studied genes(IL-1β,IL-6,Lif,and Vegf)was evaluated and blastocyst attachment on the cocultured cells(isolated endometrial cells and PBMC cells)was monitored in all four groups.Results:Data showed that expression decreased in the PBMC group compared to the treated PBMC(transfected and activated PBMCs)and increased in transfected PBMC compared to the activated PBMC.Attachment and migration of blastocysts were dramatically enhanced in the transfected PBMC group compared to the activated PBMC group(P<0.05).Conclusions:Use of hCG-producing PBMCs(transfected PBMC)has more influence on endometrial receptivity.
文摘In recent years,Peripheral blood smear is a generic analysis to assess the person’s health status.Manual testing of Peripheral blood smear images are difficult,time-consuming and is subject to human intervention and visual error.This method encouraged for researchers to present algorithms and techniques to perform the peripheral blood smear analysis with the help of computer-assisted and decision-making techniques.Existing CAD based methods are lacks in attaining the accurate detection of abnormalities present in the images.In order to mitigate this issue Deep Convolution Neural Network(DCNN)based automatic classification technique is introduced with the classification of eight groups of peripheral blood cells such as basophil,eosinophil,lymphocyte,monocyte,neutrophil,erythroblast,platelet,myocyte,promyocyte and metamyocyte.The proposed DCNN model employs transfer learning approach and additionally it carries three stages such as pre-processing,feature extraction and classification.Initially the pre-processing steps are incorporated to eliminate noisy contents present in the image by using Histogram Equalization(HE).It is enclosed to improve an image contrast.In order to distinguish the dissimilar class and segmentation approach is carried out with the help of Fuzzy C-Means(FCM)model whereas its centroid point optimality method with Slap Swarm based optimization strategy.Moreover some specific set of Gray Level Co-occurrence Matrix(GLCM)features of the segmented images are extracted to augment the performance of proposed detection algorithm.Finally the extracted features are recorded by DCNN and the proposed classifier has the capability to extract their own features.Based on this the diverse set of classes are classified and distinguished from qualitative abnormalities found in the image.
文摘Chronic obstructive pulmonary disease (COPD) is a chronic, progressive respiratory disease and the third leading cause of respiratory disease mortality. The diagnosis of COPD is changed to acute exacerbation of COPD (AECOPD) when respiratory symptoms become worse, beyond normal day-to-day variations and severely enough that changes in medication are required. Both neutrophils to lymphocyte ratio (NLR) and peripheral blood eosinophilia (PBE) are rapid and relatively inexpensive tests that can be easily applied in the clinical practice for the diagnosis and treatment of AECOPD patients. Furthermore, current studies found that NLR and PBE had a higher accuracy rate than other traditional markers (Leukocyte count and C-reactive protein) for the diagnosis and management of AECOPD. Besides, recent studies determined that NLR and PBE can be used for prediction of future exacerbations in COPD patients. This review aims to explore the current knowledge about the significance of NLR and PBE in AECOPD patients.

文摘AIM: To assess the absolute number of T-regulatory cells (Tregs; CD4+CD25+Foxp3+) in the peripheral blood of gastric and colorectal cancer patients. METHODS: We enrolled 70 cancer patients (33 gastric cancer, 37 colorectal cancer) and 17 healthy volunteers. The CD3+CD4+ lymphocytes and CD4+CD25+Foxp3+ Tregs in the peripheral blood were analyzed with flow cytometry. The absolute numbers of Tregs were calculated based on the CD4+CD25+Foxp3+ cells percent-age of CD3+CD4+ cells and the absolute numbers of CD3+CD4+ cells per microliter. RESULTS: The mean number of CD4+CD25+Foxp3+ cells per microliter in colorectal cancer patients was 15.7 (SD: 21.8), for gastric cancer patients 12.2 (SD: 14.3), and for controls 17.5 (SD: 11.4). The absolute number of Tregs was significantly lower in gastric cancer patients than in controls (P = 0.026). There was no statistically significant difference for gastric vs colorectal cancer or colorectal cancer vs controls. The absolute number of Tregs was also significantly depressed in N+ vs Ncancer patients [22.0 (27.7) vs 10.1 (9.0), P = 0.013], and in the subgroup of gastric cancer patients [30.3 (27.6) vs 9.6 (8.0), P = 0.003]. No statistical difference was observed in the proportion of Tregs in the CD4+ population between the groups. CONCLUSION: The absolute number of Tregs in peripheral blood of gastric cancer but not colorectal cancer patients was significantly decreased in comparison with that in healthy controls.
