As a result, not only was the functional recombinant ex-pression of heparinase I in E. coli accomplished, but also a linear correlation was obtained between the GFP fluores-cence intensity and heparinase I activity, allowing enzyme activity to be quantified rapidly during the fermentation. 展开更多

Green fluorescent protein (GFP) and its variants /homolog proteins are generally called as GFP-like fluorescent proteins (FPs), which are widely used as visible molecular tools for monitoring a wide range of biological processes due to their capability of simple, accurate and real time quantification. The FPs-based molecular and visible quantification tools are giving more impact on bioprocess engineering, enabling the biomolecule-level dynamic information to be linked with the process-level events. In this review, different applications of FPs in biological engineering with emphasis on rapid molecular bioprocess quantification, such as quantification of the transcription efficiency, the protein production, the protein folding efficiency, the cell concentration, the intracellular microenvironments and so on, would be first introduced. The challenges of using FPs with respect to actual bioprocess applications for the precise quantification including the interaction of FPs and the fused partner proteins, the maturation of FPs, the inner filter effect and sensing technology were then discussed. Finally, the future development for the FPs used in molecular bioprocess quantification would be proposed. 展开更多

A rapid method based on rapid resolution liquid chromatography(RRLC) coupled with a diode array detector(DAD) was developed for the simultaneous determination of six major constituents(magnoflorine,jatrorrhizine,coptisine,palmatine,berberine and evodiamine) in traditional Chinese medicine(TCM) Zuo Jin Pill(ZJP).The satisfactory chromatographic separation was carried on an Eclipse Plus C18 column(1.8 μm i.d.,150 mm×4.6 mm) by linear gradient elution with a mobile phase of acetonitrile-acetate buffer.All the calibration curves show good linearity(r2 >0.9998).The detection limits and quantification limits ranged in 1.4―12 ng and 4.8―30 ng,respectively.The intra-and inter-day precisions were less than 0.63% with accuracies 98.60%―100.78%,and the recoveries were from 99.45% to 100.46%.Furthermore,hierarchical cluster analysis(HCA) was used to evaluate the variation of the herbal prescription.The results demonstrate that this analytical method is simple,sensitive and reliable for rapidly analyzing six major bioactive compounds in ZJPs and is helpful to comprehensively evaluating the quality of this TCM. 展开更多

This paper proposed an efficient research method for high-dimensional uncertainty quantification of projectile motion in the barrel of a truck-mounted howitzer.Firstly,the dynamic model of projectile motion is established considering the flexible deformation of the barrel and the interaction between the projectile and the barrel.Subsequently,the accuracy of the dynamic model is verified based on the external ballistic projectile attitude test platform.Furthermore,the probability density evolution method(PDEM)is developed to high-dimensional uncertainty quantification of projectile motion.The engineering example highlights the results of the proposed method are consistent with the results obtained by the Monte Carlo Simulation(MCS).Finally,the influence of parameter uncertainty on the projectile disturbance at muzzle under different working conditions is analyzed.The results show that the disturbance of the pitch angular,pitch angular velocity and pitch angular of velocity decreases with the increase of launching angle,and the random parameter ranges of both the projectile and coupling model have similar influence on the disturbance of projectile angular motion at muzzle. 展开更多

In this paper,a dynamic modeling method of motor driven electromechanical system is presented,and the uncertainty quantification of mechanism motion is investigated based on this method.The main contribution is to propose a novel mechanism-motor coupling dynamic modeling method,in which the relationship between mechanism motion and motor rotation is established according to the geometric coordination of the system.The advantages of this include establishing intuitive coupling between the mechanism and motor,facilitating the discussion for the influence of both mechanical and electrical parameters on the mechanism,and enabling dynamic simulation with controller to take the randomness of the electric load into account.Dynamic simulation considering feedback control of ammunition delivery system is carried out,and the feasibility of the model is verified experimentally.Based on probability density evolution theory,we comprehensively discuss the effects of system parameters on mechanism motion from the perspective of uncertainty quantization.Our work can not only provide guidance for engineering design of ammunition delivery mechanism,but also provide theoretical support for modeling and uncertainty quantification research of mechatronics system. 展开更多

Recent studies on tropical cyclone(TC)intensity change indicate that the development of a vertically aligned TC circulation is a key feature of its rapid intensification(RI),however,understanding how vortex alignment occurs remains a challenging topic in TC intensity change research.Based on the simulation outputs of North Atlantic Hurricane Wilma(2005)and western North Pacific Typhoon Rammasun(2014),vortex track oscillations at different vertical levels and their associated role in vortex alignment are examined to improve our understanding of the vortex alignment during RI of TCs with initial hurricane intensity.It is found that vortex tracks at different vertical levels oscillate consistently in speed and direction during the RI of the two simulated TCs.While the consistent track oscillation reduces the oscillation tilt during RI,the reduction of vortex tilt results mainly from the mean track before RI.It is also found that the vortex tilt is primarily due to the mean vortex track before and after RI.The track oscillations are closely associated with wavenumber-1 vortex Rossby waves that are dominant wavenumber-1 circulations in the TC inner-core region.This study suggests that the dynamics of the wavenumber-1 vortex Rossby waves play an important role in the regulation of the physical processes associated with the track oscillation and vertical alignment of TCs. 展开更多

The reasonable quantification of the concrete freezing environment on the Qinghai–Tibet Plateau(QTP) is the primary issue in frost resistant concrete design, which is one of the challenges that the QTP engineering managers should take into account. In this paper, we propose a more realistic method to calculate the number of concrete freeze–thaw cycles(NFTCs) on the QTP. The calculated results show that the NFTCs increase as the altitude of the meteorological station increases with the average NFTCs being 208.7. Four machine learning methods, i.e., the random forest(RF) model, generalized boosting method(GBM), generalized linear model(GLM), and generalized additive model(GAM), are used to fit the NFTCs. The root mean square error(RMSE) values of the RF, GBM, GLM, and GAM are 32.3, 4.3, 247.9, and 161.3, respectively. The R^(2) values of the RF, GBM, GLM, and GAM are 0.93, 0.99, 0.48, and 0.66, respectively. The GBM method performs the best compared to the other three methods, which was shown by the results of RMSE and R^(2) values. The quantitative results from the GBM method indicate that the lowest, medium, and highest NFTC values are distributed in the northern, central, and southern parts of the QTP, respectively. The annual NFTCs in the QTP region are mainly concentrated at 160 and above, and the average NFTCs is 200 across the QTP. Our results can provide scientific guidance and a theoretical basis for the freezing resistance design of concrete in various projects on the QTP. 展开更多

The 2D sandwich model serves as a potent tool in exploring the influence of surface geometry on the combustion attributes of Ammonium perchlorate/Hydroxyl-terminated polybutadiene(AP/HTPB)propellant under rapid pressure decay.The thickness of the sandwich propellant is derived from slicing the 3D random particle packing,an approach that enables a more effective examination of the micro-flame structure.Comparative analysis of the predicted burning characteristics has been performed with experimental studies.The findings demonstrate a reasonable agreement,thereby validating the precision and soundness of the model.Based on the typical rapid depressurization environment of solid rocket motor(initial combustion pressure is 3 MPa and the maximum depressurization rate is 1000 MPa/s).A-type(a flatter surface),B-type(AP recesses from the combustion surface),and C-type(AP protrudes from the combustion surface)propellant combustion processes are numerically simulated.Upon comparison of the evolution of gas-phase flame between 0.1 and 1 ms,it is discerned that the flame strength and form created by the three sandwich models differ significantly at the beginning stage of depressurization,with the flame structures gradually becoming harmonized over time.Conclusions are drawn by comparison extinction times:the surface geometry plays a pivotal role in the combustion process,with AP protrusion favoring combustion the most. 展开更多

The postoperative anal exhaust time, first meal time, early activity time, pain time, abdominal drainage tube removal time, hospitalization time and complication rate were compared between the two groups. Aim: This study evaluates the impact of Enhanced Recovery After Surgery (ERAS) nursing on postoperative complications and quality of life in patients undergoing robot-assisted minimally invasive esophagectomy (RAMIE). Methods: A total of 150 patients who underwent RAMIE from January 2020 to January 2022 at our hospital were randomly assigned to either the observation group or the control group, with 75 patients in each. The control group received standard perioperative management and nursing care, while the observation group was treated with ERAS nursing strategies. Interventions continued until discharge, and outcomes such as postoperative complications, quality of life, and nutritional status were compared between the groups. Results: The observation group exhibited a significantly lower incidence of postoperative adverse reactions compared to the control group (P ionally, all dimension scores of the Short-Form 36 Health Survey (SF-36), including the total score, were higher in the observation group (P < 0.05). Under the influence of anthropogenic and climate change,the problems caused by urban heat island(UHI)has become increasingly prominent.In order to promote urban sustainable development and improve the quality of human settlements,it is significant for exploring the evolution characteristics of urban thermal environment and analyzing its driving forces.Taking the Landsat series images as the basic data sources,the winter land surface temperature(LST)of the rapid urbanization area of Fuzhou City in China was quantitatively retrieved from 2001 to 2021.Combing comprehensively the standard deviation ellipse model,profile analysis and GeoDetector model,the spatio-temporal evolution characteristics and influencing factors of the winter urban thermal environment were systematically analyzed.The results showed that the winter LST presented an increasing trend in the study area during 2001–2021,and the winter LST of the central urban regions was significantly higher than the suburbs.There was a strong UHI effect from 2001 to 2021with an expansion trend from the central urban regions to the suburbs and coastal areas in space scale.The LST of green lands and wetlands are significantly lower than croplands,artificial surface and unvegetated lands.Vegetation and water bodies had a significant mitigation effect on UHI,especially in the micro-scale.The winter UHI had been jointly driven by the underlying surface and socio-economic factors in a nonlinear or two-factor interactive enhancement mode,and socio-economic factors had played a leading role.This research could provide data support and decision-making references for rationally planning urban layout and promoting sustainable urban development. 展开更多

Genetic Programming (GP) is an important approach to deal with complex problem analysis and modeling, and has been applied in a wide range of areas. The development of GP involves various aspects, including design of genetic operators, evolutionary controls and implementations of heuristic strategy, evaluations and other mechanisms. When designing genetic operators, it is necessary to consider the possible limitations of encoding methods of individuals. And when selecting evolutionary control strategies, it is also necessary to balance search efficiency and diversity based on representation characteristics as well as the problem itself. More importantly, all of these matters, among others, have to be implemented through tedious coding work. Therefore, GP development is both complex and time-consuming. To overcome some of these difficulties that hinder the enhancement of GP development efficiency, we explore the feasibility of mutual assistance among GP variants, and then propose a rapid GP prototyping development method based on πGrammatical Evolution (πGE). It is demonstrated through regression analysis experiments that not only is this method beneficial for the GP developers to get rid of some tedious implementations, but also enables them to concentrate on the essence of the referred problem, such as individual representation, decoding means and evaluation. Additionally, it provides new insights into the roles of individual delineations in phenotypes and semantic research of individuals. 展开更多

Surface-enhanced Raman scattering(SERS)spectroscopy has been employed as a rapid analysis technology for food security inspection recently.Nowadays,it is still a great challenge to rapidly quantify multiple trace antibiotics potentially abused in aquaculture industry.In this work,a magnetic Ti_(3)C_(2)T_(x)/Fe_(3)O_(4)/Ag substrate was prepared for the development of a reliable rapid SERS quantification method for multiple trace sulfonamides in aquatic products.This magnetic substrate had good uniformity,reproducibility,stability and SERS activity.Moreover,this substrate could integrate the magnetic separation-enrichment and matrix clean-up without cross contamination,which endowed it with good selectivity and antiinterference capability during real sample analysis.The electromagnetic enhancement and chemical enhancement mechanism of this magnetic substrate were studied in detail to reveal its good separationenrichment performance and SERS activity.Finally,a rapid SERS quantification method was established and practically applied for trace phthalic sulfathiazole(PST)and silver sulfadiazine(SSD)in aquatic products by using Ti_(3)C_(2)T_(x)/Fe_(3)O_(4)/Ag magnetic substrates.Trace PST and SSD could be actually detected and quantified as 55.9μg/kg and 64.0μg/kg in aquatic products,respectively.Good recoveries of 83.9%-116%with relative standard deviations(RSDs)of 0.5%-3.2%for PST and 80.2%-102%with RSDs of 1.3%-5.8%for SSD were obtained.This work proposed an efficient and reliable method for rapid quantification of trace multiple sulfonamides in complex aquatic samples during food security inspection. 展开更多

为建立一种特异性强、灵敏度高且简单方便的诺如病毒GⅡ.6核酸快速检测方法,下载诺如病毒GⅡ.6基因组序列,用Mega7.0对基因组进行比对分析获得高度保守序列;基于保守序列设计诺如病毒GⅡ.6的反转录重组酶和聚合酶等温检测(RT-RAPID)技... 为建立一种特异性强、灵敏度高且简单方便的诺如病毒GⅡ.6核酸快速检测方法,下载诺如病毒GⅡ.6基因组序列,用Mega7.0对基因组进行比对分析获得高度保守序列;基于保守序列设计诺如病毒GⅡ.6的反转录重组酶和聚合酶等温检测(RT-RAPID)技术扩增引物、荧光探针、crRNA和报告分子;优化RT-RAPID的反应引物、反应温度和反应时间及Cas12a检测用的crRNA,并分析RT-RAPID-Cas12a方法的特异性和灵敏度。基于RT-RAPID荧光法和RT-RAPID-Cas12a荧光法的诺如病毒GⅡ.6亚型核酸快速检测方法能在1 h内完成检测并得出结果,灵敏度分别为10 copies/μL和0.5 copies/μL,且两个方法均与诺如病毒GⅡ.17型、鼻病毒、偏肺病毒和博卡病毒无交叉反应。2个检测方法与RT-qPCR阳性样本一致率均为90%,阴性样本一致率均为100%。论文建立的基于RT-RAPID和Cas12a的诺如病毒GⅡ.6核酸快速检测方法均可高效快速地检测出诺如病毒GⅡ.6,具有灵敏度高、特异性强、操作便捷且快速等特点。 展开更多

目的探讨Rapid软件、CT血管造影(CTA)和数字减影血管造影(DSA)对侧支循环评估的一致性。方法选择2020年1月至2022年4月重庆医科大学附属永川医院收治的单侧大脑中动脉狭窄的患者75例,患者发病后接受CTA、CTP和DSA检查,由Rapid软件自动... 目的探讨Rapid软件、CT血管造影(CTA)和数字减影血管造影(DSA)对侧支循环评估的一致性。方法选择2020年1月至2022年4月重庆医科大学附属永川医院收治的单侧大脑中动脉狭窄的患者75例,患者发病后接受CTA、CTP和DSA检查,由Rapid软件自动计算评估侧支循环,CTA和DSA侧支循环血流评分分别由2名医师双盲评估,以DSA评分为金标准,判断CTA及Rapid软件评估的准确性。应用Kappa检验评估DSA、CTA与Rapid软件血流评分的一致性。结果Rapid软件与DSA、CTA血流评分一致性好(r_(s)=0.729,κ=0.516;r_(s)=0.744,κ=0.543),CTA与DSA血流评分一致性好(κ=0.768,r_(s)=0.857)。Rapid软件和CTA判定侧支循环良好的灵敏度和特异度分别为83.33%、60.87%和100.00%、76.81%;Rapid软件和CTA判定侧支循环中等的灵敏度和特异度分别为46.67%、100.00%和86.67%、100.00%;Rapid软件和CTA判定侧支循环较差的灵敏度和特异度分别为66.67%、70.67%和62.50%、96.08%。结论CTA具有较高的灵敏度与特异度,且与DSA有很高的相关性。Rapid软件的灵敏度与特异度较低,但与DSA有较好的一致性,因此也可以作为评估侧支循环的检查手段。 展开更多
