Growing evidence has revealed the existence of CSCs in various types of cancers and suggested that CSCs differentiate into diverse lineage cells that contribute to tumor progression. We may be able to overcome the limitations of cancer treatment with a comprehensive understanding of the biological features and mechanisms underlying therapeutic resistance in CSCs. This review provides an overview of the properties, biomarkers, and mechanisms of resistance shown by CSCs. Recent findings on metabolic features, especially fatty acid metabolism and ferroptosis in CSCs, are highlighted, along with promising targeting strategies. Targeting CSCs is a potential treatment plan to conquer cancer and prevent resistance and relapse in cancer treatment.展开更多

Methods; A total of 27 cases of autism children were subjected into this study. By using a SPECT, the cerebral images were collected before and after acupuncture treatment and analyzed according to the recommended methods in 《Clinical Nuclear Medicine》 for assessing the state of blood flow, radioactivity quantity distribution and radioactivity count in bilateral hemispheres. 'JIN's three-needling' was employed. The acupuncture treatment was given once every other day, with 4 months being a therapeutic course and an interval of one month between two courses. Results: After acupuncture treatment, of the 22 cases, 20 had remarkable improvement and 2 had improvement in cerebral blood flow, with the total effective rate being 90.8%. Before the treatment there were significant differences between the left and right cerebrum (P<0. 001), and between the left and right frontal lobes in radioactive areas (P<0.01); however, after treatment, no differences were found between them (P>0.05). After treatment, the radioactivity count in the whole brain decreased significantly in comparison with that of pretreatment (P<0.01). It indicates the improvement of cerebral blood flow and cellular metabolism after the treatment. Conclusion: Acupuncture can significantly improve cerebral blood flow in autism children.展开更多

文摘Cancer stem cells(CSCs) are a small subset of cells in cancers that are thought to initiate tumorous transformation and promote metastasis, recurrence, and resistance to treatment. Targeting CSCs is a potential treatment plan to conquer cancer and prevent resistance and relapse in cancer treatment.

文摘Immune checkpoint inhibitors(ICIs)are used to relieve and refuel anti-tumor immunity by blocking the interaction, transcription, and translation of co-inhibitory immune checkpoints or degrading co-inhibitory immune checkpoints.Thousands of small molecule drugs or biological materials, especially antibody-based ICIs,are actively being studied and antibodies are currently widely used.Limitations, such as anti-tumor efficacy, poor membrane permeability, and unneglected tolerance issues of antibody-based ICIs,remain evident but are thought to be overcome by small molecule drugs.Recent structural studies have broadened the scope of candidate immune checkpoint molecules,as well as innovative chemical inhibitors.By way of comparison, small molecule drug-based ICIs represent superior oral bioavailability and favorable pharmacokinetic features.Several ongoing clinical trials are exploring the synergetic effect of ICIs and other therapeutic strategies based on multiple ICI functions, including immune regulation,anti-angiogenesis,and cell cycle regulation. In this review we summarized the current progression of small molecule ICIs and the mechanism underlying immune checkpoint proteins, which will lay the foundation for further exploration.

文摘Objective: To observe the effect of acupuncture on images in autism children. Conclusion: Acupuncture can significantly improve cerebral blood flow in autism children.
文摘To observe effect of moxibustion at Zusanli (足三里 ST 36) and Liangmen (梁门 ST 21 ) on expression of heat shock protein 70 (HSP70) in gastric mucosa of the rat of stress ulcer (SU) to explore the mechanism of moxibustion in resisting oxidative injury of the gastric mucosa. Methods: Sixty SD rats were evenly randomized into 4 groups, a blank group, a model group, an acupoint moxibustion group and a non-acupoint moxibustion group. 文摘Therapies that complement free radical scavenging are an important approach for treating aging in the brain. In the present study, two formulations of moxa cone moxibustion were applied at acupoints Zusanfi (ST 36) and Xuanzhong (GB 39), and at acupoints Baihui (DU 20) and Guanyuan (RN 4), in D-galactose-induced senile mice. The results revealed that moxa cone moxibustion improved total superoxide dismutase and Cu/Zn-superoxide dismutase activity in the homogenates of the cerebral tissue, as well as ameliorating deficits in neuronal morphology and neuronal density in the cerebral cortex and hippocampal CA3. Moxa cone moxibustion also enhanced learning and memory functions of senile mice. Moxa cone moxibustion at Zusanli, Xuanzhong, Baihuiand Guanyuan acupoints can thus be used to complement free radical scavengers, with efficacy that is equal to that of electroacupuncture at Zusanliand Xuanzhong, and superior to that of nimodipine treatment.

文摘Pain is a complex physiological and psychological activity, involving at least three dimensions, including pain sensation, pain emotion, and pain cognition.Acupuncture can clearly relieve the pain sensation of patients and improve pain emotion and pain cognition induced by pain;acupuncture participates in the multi-dimensional regulation of pain through brain regions of the limbic system such as anterior cingulate cortex(ACC),amygdala(AMY),and hippocampus.By analyzing relevant literature,it has been found that the regulation of acupuncture on pain emotion is mainly related to the activation of pertinent opioid receptors in the ACC,the decrease of the expression of extracellular signal-regulated kinase(ERK),and the promotion of the expression of glutamic acid(Glu)A1,metabotropic glutamate receptor-1(mGluR1),andγ-aminobutyric acid aminobutyric acid(GABA)B2 protein in the AMY.The regulation of acupuncture on pain cognition is mainly related to the elevation of the expression of protein kinase A(PKA)and phospho-p38 mitogen-activated protein kinase(phospho-p38 MAPK)and the inhibition of cyclic adenosine monophosphate(cAMP)/PKA/cAMP response element-binding protein(CREB)signaling pathway in the ACC.
文摘Objective:To observe the efficacy of needling the Xi-Cleft points of yang meridians plus topical surrounding needling and pregabalin in treating postherpetic neuralgia(PHN)affecting the head and face and its influence on quality of life(QOL).Methods:Eighty patients with PHN affecting the head and face were randomized into an acupuncture-medication group and a Western medication group by their visiting sequence, with 40 cases in each group. The Western medication group took pregabalin capsules orally,75 mg each time, twice a day.The dosage could increase to 150 mg each time and twice a day within 1 week based on the efficacy and tolerance. The treatment lasted for 5 weeks. The acupuncture-medication group was given additional surrounding needling at the herpes zoster on the head and face, and Xi-Cleft points of yang meridians were chosen according to the affected area for acupuncture, once every other day,for 5 weeks in total. Results:Prior to treatment, there were no significant differences in the visual analog scale(VAS)and QOL scores between the two groups(both P>O,05).After treatment,VAS and QOL scores dropped significantly in both groups(all P<0.01),and were significantly lower in the acupuncture-medication group than in the Western medication group(both P<0.01).The total effective rate was 67.5%in the Western medication group, versus 92.5%in the acupuncture-medication group, and the b etwee n-group difference was statistically significant(P<0.05).Conclusion:Based on oral administration of pregabalin, needling the Xi-Cleft points of yang meridians and topical surrounding needling can reduce PHN,notably improve patients'QOL,and produce more significant efficacy than oral administration of pregabalin alone。
