[Objectives]This study was conducted to analyze changes of color difference and polyphenols and their relationship in the process of threshing and redrying.[Methods]The change trends of color difference parameters L,a,b,H^(0),C and polyphenol contents in tobacco leaves in the middle leaf module and the Pearson correlation between them were analyzed.[Results]During threshing and redrying,L and H^(0)of tobacco showed a decreasing trend,but H^(0)changed slightly;the values of a and C showed an inverted"S"change trend,and the differences between various processes were significant;and the value of b decreased first and then increased.The content of chlorogenic acid and scopoletin decreased first,then increased,and then decreased during threshing and redrying;rutin content showed a significant decreasing trend.The value of L was in an extremely significant positive correlation with rutin content;the value of a was in an extremely significant negative correlation with chlorogenic acid content;b value was also significantly correlated with rutin;and H^(0)was in significant correlation and extremely significant correlation with rutin and total polyphenols,respectively.[Conclusions]This study provides technical support for online detection of tobacco leaf components by the color difference method and for the homogenization of threshing and redrying. 展开更多

Since pure tobacco stalk(TS)biomass pellet fuels tend to slag,five anti-slagging agents were added to the crushed TS to obtain a pellet fuel that could be used in biomass burners to provide heat for tobacco curing.The obtained results revealed that the main component of TS pellet fuel was K_(2)Si_(2)O_(5).During fuel combustion process,additives generated higher melting point silicate compounds by Al–K,Ca–K,and Ca–K elemental structures to replace single K elemental structure of TS,enhancing the anti-slagging efficiency of the pellet fuel from 21.63%to 78.29%and promoting the precipitation of K,Mg,and Na elements in the slag block.By investigating the anti-slagging mechanism pathways of the additives in TS biomass pellet fuels,altering of the structure of silicate ion group pathway was found to improve anti-slagging effects that met the requirements of production formula. 展开更多

In order to effectively reduce the chlorophyll content in flue-cured tobacco, improve the overall quality of tobacco leaves, chlorophyllase gene was cloned from Arabidopsis thaliana. After the expression of the expression vector in E. coil, the recombinant engineering strain was obtained. Afterwards, IPTG (isopropy-β-D-thiogalactopyranoside)was used to induce the goal protein, and the chlorophyllase activity of the recombinant engineering strain was measured, so as to investigate its degradation effect on the chlorophyll in the extracts of tobacco leaves. The results were as follows: (1) the amplified chlorophyllase gene At- CLH1 constructed the expression vector pET28a-AtCLH1 successfully, obtaining the recombinant engineering strain; (2) induced under 30 ℃ for 22 h, the strain could well express the recombinant protein AtCLH1 with 0.5 mmol/L IPTG, and the molecular weight was about 35 kDa; (3) the strain showed good chlorophyllase producing capability, and the activity of the produced chlorophyllase could reach up to 24.9 U/mL, which could degrade the chlorophyll in tobacco extract and had a good application prospect in improving the quality of low quality tobacco; (4) based on the results of orthogonal test, the enzyme extract from the strain was added to the tobacco leaf surface, which could make the degradation rate of chlorophyll in the tobacco leaf reach 17.06% under the temperature of 37 ℃ at the humidity of 75% for 48 h; (5) after treated by the enzyme liquid, the test tobacco showed increase in the content of aromatic substances, enhancement of tobacco fragrance quality and amount, significant decrease of offensive odor and irritation, significant improvement of agreeable aftertaste, making the overall sensory quality of the tobacco leaf significantly improved. 展开更多

Differentiation of Chemisorption and Physisorption of Carbon Dioxide on Imidazolium-type Poly(ionic liquid)Brushes

Amine-functionalized imidazolium-based poly(ionic liquid)brushes on mesoporous silica were synthesized via"grafting through"technique and were applied as model sorbents to understand physisorption and chemisorption of carbon dioxide molecules.The experimental results reveal that the total adsorption capacity of model sorbents derived from temperature programmed desorption(TPD)approach reaches 1.72 mmol·g^-1 that is much higher than the sum of adsorption capacity of bare mesoporous silica and free polymers at 25℃under carbon dioxide partial pressure of 0.2 bar.The proposed physical adsorption and chemical adsorption of carbon dioxide molecules in TPD response curves were validated by volumetric desorption measurement.It is also observed that physical adsorption is completely reversible and chemical adsorption is irreversible.The results demonstrate that the temperature programmed desorption technique is an effective approach to differentiate chemisorption and physisorption of gaseous species on solid sorbents,which is beneficial for understanding of adsorption mechanism and materials design. 展开更多

An acidic extracellular polysaccharide(EPS)was gained from Stropharia rugosoannulata fermentation broth by the alcohol precipitate method.Sulfated derivative of EPS(S-EPS)was gained by chlorosulfonic acid-pyridine method and the conditions were as follows:pyridine as the reaction solvent,temperature 80°C,and reaction time 90 min.The chemical structure and chain conformation of EPS and S-EPS were characterized by FT-IR,NMR and size exclusion chromatography(SEC)coupled with multi-angle laser light scattering(MALLS)detection.FT-IR spectra indicated the sulfated group was linked to the polymer.The EPS mainly consists of mannose,glucose and glucuronic acid,and the glycosidic bond type is mainly(1→4)-linked and(1→6)-linked.C6 and C2 were substituted by sulfate groups.The EPS was degraded into smaller molecular weight(Mw)polysaccharides with the larger molecular size(Rg)after sulfation and S-EPS has better hydrophilicity.Moreover,the sulfated derivative S-EPS was found to have a better scavenging ability on hydroxyl radicals than EPS.It proved that the chemical modification of S.rugosoannulata polysaccharides by sulfation effectively enhanced their antioxidant activity. 展开更多
