[Methods] 10 rats were randomly selected and fed with normal diet as blank group, and the remaining 30 rats were fed with high-fat diet for 8 weeks. The obesity-induced IR model was induced and divided into two groups: model group and acupressure group. The acupressure group was treated by "acupressure therapy" three times a week for 6 weeks;the model group was taken 3 times at the same time every week, and the supine binding experiment lasted for 10 min for 6 weeks. Morphological observation was carried out before and after the experiment, mainly observing the general conditions and body weight. After the last intervention, the animals were killed and samples were collected. Western Blot method was used to detect the expression of TNF-α and IL-6 protein in adipose tissue of the three groups. [Results] Compared with the blank group, the expression of TNF-α and IL-6 protein in adipose tissue of the model group increased significantly ( P <0.01);compared with the model group, the expression of TNF-α and IL-6 protein in adipose tissue of the acupressure group decreased significantly ( P <0.01). [Conclusions] After intervening in the obesity-induced IR rats with acupressure method, the status of insulin resistance in obese rats was improved by inhibiting the expression of inflammatory factors.展开更多

To explore the role of the nucleus tractus solitarii in the protective mechanism of pre-moxibustion on gastric mucosa, nucleus tractus solitarii were damaged in rats and pre-moxibustion treatment at the Zusanli(ST36) point followed. The gastric mucosa was then damaged by the anhydrous ethanol lavage method. Morphological observations, enzyme linked immunosorbent assays, and western immunoblot analyses showed that gastric mucosa surface lesion and the infiltration of inflammatory cells were significantly ameliorated after pre-moxibustion treatment. Furthermore, the gastric mucosal damage index and somatostatin level were reduced, and epidermal growth factor content in the gastric mucosa and heat-shock protein-70 expression were increased. These results were reversed by damage to the nucleus tractus solitarii. These findings suggest that moxibustion pretreatment at the Zusanli point is protective against acute gastric mucosa injury, and nucleus tractus solitarii damage inhibits these responses. Therefore, the nucleus tractus solitarii may be an important area for regulating the signal transduction of the protective effect of pre-moxibustion on gastric mucosa.展开更多

However, the signaling pathway underlying this effect remains unclear. Rats were intragastrically administered absolute alcohol, causing obvious lesion of the gastric mucosa. Following pre-moxibustion at Zusanli (ST36) for 8 days, the ulcer index decreased to different degrees. The results of an enzyme linked immunosorbent assay and western blotting showed significant upregulation of heat shock protein 70 expression in the gastric mucosa and serum. None out of transection of the spinal cord, damage to the nucleus of the solitary tract, neurotomy of the vagal nerve and neurotomy of the common peroneal nerve affected the decrease in ulcer index or the increase in heat shock protein 70 expression in serum after pre-moxibustion at Zusanli, and heat shock protein 70 expression was obviously decreased in the gastric mucosa. These findings suggest that pre-moxibustion at Zusanli can protect the gastric mucosa against lesioning, and that the mechanism underlying this effect involves its induction of heat shock protein 70 expression. Neural pathways participate in the regulatory effects of moxibustion on heat shock protein 70 expression in the gastric mucosa.展开更多

Methods: Sixty cases with chronic hepatic diseases were randomly divided into a Taichong(LR 3) group and a Ququan(LR 8) group, 30 cases in each group, and were respectively treated with electroacupuncture at Taichong(LR 3) and Ququan(LR 8). The blood flow results of the hepatic artery and portal vein by color ultrasonic test were used as the objective indexes, to compare and observe the hemodynamics changes of the hepatic artery and portal vein in the patients before and after EA. Results: After EA, in Taichong(LR 3) group, the average velocity, the lowest velocity, resistance indexes and the highest velocity of blood flow of the hepatic artery, and the highest velocity, average velocity of blood flow of the portal vein, blood flow volume of the portal vein(PVBF), total blood flow volume of the liver(TLBF), and Doppler blood perfusion indexes of the hepatic artery(DPI) were all statistically different from those before intervention(all P<0.05). In Ququan(LR 8) group, the highest velocity, average velocity, lowest velocity and resistance indexes of blood flow of the hepatic artery, and the highest velocity and average velocity of blood flow of the portal vein, and PVBF were significantly different from those before intervention(P<0.05). The TLBF and DPI were significantly different from those before intervention(P<0.01). All various indexes were not statistically different between the two groups. Conclusion: EA at Taichong(LR 3) and Ququan(LR 8) can speed up blood flow velocity of the portal vein, increase PVBF, increase the TLBF, increase the resistance indexes of the hepatic artery, decrease the blood flow velocity of the hepatic artery, and decrease DPI in the liver.展开更多

In this study, we investigated whether electro-acupuncture(EA) affects the CAP in COPD.Methods: Sprague-Dawley rats were induced into COPD through exposure to cigarette smoke combined with lipopolysaccharide. EA treatment was applied at Zusanli(ST36) and Feishu(BL13) points for 30 min/d for 7 d. Seventy-two rats were randomly divided into six study groups, including normal, normal + EA,normal + α-bungarotoxin(a-BGT)(the antagonist of the nicotinic acetylcholine receptor a7 subunit(a7 nAChR)) + EA, COPD, COPD + EA, and COPD + α-BGT + EA. Lung function, pathology and vagus nerve discharge were tested. The levels of acetylcholine(ACh), acetylcholinesterase(AChE), interleukin-6(IL-6) and tumor necrosis factor-a(TNF-a) in bronchoalveolar lavage fluid(BALF) and lung tissue were measured by enzyme-linked immunosorbent assay. The mRNA and protein expression and immunoreactivity of a7 nAChR and its postreceptor inflammation signal pathway, including janus kinase 2(JAK2), signal transducers and activators of transcription 3(STAT3), nuclear factor-KB(NF-κB), were observed by quantitative reverse transcription-polymerase chain reaction, Western blot and immunohistochemistry. Results: Compared with normal rats, there were a significant decline in lung function and discharge of the vagus nerve(P < 0.01), a marked sign of lung inflammation and an increase of ACh, AChE, IL-6 and TNF-a level in BALF or lung tissue(P < 0.05, P < 0.01) and higher expression of a7 nAChR, JAK2, STAT3 and NF-κB(P<0.05, P<0.01) in the COPD rats. In rats receiving EA, the lung function and vagal discharge were enhanced(P < 0.01), lung inflammation was improved and the levels of ACh, AChE, IL-6 and TNF-a were decreased(P<0.01). Further, the expression of a7 nAChR, JAK2, STAT3 and NF-κB was downregulated(P < 0.05, P < 0.01). However, the above effects of EA were blocked in rats injected with α-BGT(P < 0.01).Conclusion: EA treatment can reduce the lung inflammatory response and improve lung function in COPD, which may be related to its involvement in the regulation of CAP.展开更多

Additionally, Tokyo Wistar rats were used as the control group. Changes in blood pressure were recorded in different groups of rats before and after the corresponding treatment. Hematoxylin and eosin(HE) staining was used to study basic morphological changes, and immunohistochemistry was used to determine GLUT1 expression in the hypothalamus. Further,PET-CT was utilized to elucidate the antihypertensive mechanism after acupuncture at the Taichong(LR 3) acupoints.RESULTS: PET-CT indicated activation of the hypothalamus. Measurement of blood pressure showed that acupuncture at the Taichong(LR 3) acupoints lowered blood pressure. HE staining did not show any significant pathological changes, although differences in cell number were observed. Immunohistochemical analysis indicated a GLUT1 downregulation in the SHRs of the Taichong(LR 3) acupuncture group after the treatment.CONCLUSION: Acupuncture at Taichong(LR 3) acupoints lowered blood pressure in SHRs, with possible mechanisms being changes in cell number and GLUT1 expression in the hypothalamus.展开更多

文摘[Objectives] To study the effect of "acupressure therapy" on anti-inflammatory mechanism in obesity-induced IR rats. [Conclusions] After intervening in the obesity-induced IR rats with acupressure method, the status of insulin resistance in obese rats was improved by inhibiting the expression of inflammatory factors.

文摘Previous studies have shown that somatic sensation by acupuncture and visceral nociceptive stimulation can converge in the nucleus tractus solitarii where neurons integrate signals impacting on the function of organs. Therefore, the nucleus tractus solitarii may be an important area for regulating the signal transduction of the protective effect of pre-moxibustion on gastric mucosa.

文摘Studies have shown that pre-moxibustion protects the gastric mucosa by up-regulating the expres- sion of heat shock protein 70. Neural pathways participate in the regulatory effects of moxibustion on heat shock protein 70 expression in the gastric mucosa.

文摘Objective: Acupuncture has a definite therapeutic effect on chronic obstructive pulmonary disease(COPD), and the cholinergic anti-inflammatory pathway(CAP) has been shown to be involved in regulation of inflammation. However, the above effects of EA were blocked in rats injected with α-BGT(P < 0.01).Conclusion: EA treatment can reduce the lung inflammatory response and improve lung function in COPD, which may be related to its involvement in the regulation of CAP.

文摘OBJECTIVE: To evaluate the effects of Taichong(LR3) acupuncture points(acupoints) on the expression of glucose transporter protein 1(GLUT1) in the hypothalamus of spontaneously hypertensive rats(SHRs) as measured by combined positron emission tomography and computed tomography(PETCT).METHODS: Spontaneously hypertensive rats(SHR)were divided into model, Taichong(LR 3) acupuncture, and sham groups. Immunohistochemical analysis indicated a GLUT1 downregulation in the SHRs of the Taichong(LR 3) acupuncture group after the treatment.CONCLUSION: Acupuncture at Taichong(LR 3) acupoints lowered blood pressure in SHRs, with possible mechanisms being changes in cell number and GLUT1 expression in the hypothalamus.
