AIM:To explore the brain mechanism of acupuncture for children with anisometropic amblyopia using the voxelmirror homotopic connectivity(VMHC)analysis method of resting functional magnetic resonance imaging(rs-fMRI)te...AIM:To explore the brain mechanism of acupuncture for children with anisometropic amblyopia using the voxelmirror homotopic connectivity(VMHC)analysis method of resting functional magnetic resonance imaging(rs-fMRI)technology based on clinical effectiveness.METHODS:Eighty children with anisometropic monocular amblyopia were randomly divided into two groups:control(40 cases,1 case of shedding)and acupuncture(40 cases,1 case of shedding)groups.The control group was treated with glasses,red flash,grating,and visual stimulations,with each procedure conducted for 5min per time.Based on routine treatment,the acupuncture group underwent acupuncture of“regulating qi and unblocking meridians to bright eyes”,Jingming(BL1),Cuanzhu(BL2),Guangming(GB37),Fengchi(GB20)acupoints were taken on both sides,with the needle kept for 30min each time.Both groups were treated once every other day,three times per week,for a total of 4wk.After the treatment,the overall curative effect of the two groups and the latency and amplitude changes of P100 wave of pattern visual-evoked potential were counted.At the same time,nine children with left eye amblyopia were randomly selected from the two groups and were scanned with rsfMRI before and after treatment.The differences in the brain regions between the two groups were compared and analyzed with VMHC.RESULTS:Chi-square test showed a notable difference in the total efficiency rate between the acupuncture(94.87%)and control groups(79.49%).Regarding the P100 wave latency and amplitude,the acupuncture group had significantly shorter latency and higher amplitude of P100 wave than the control group.Moreover,the VMHC values of the bilateral temporal lobe,superior temporal gyrus,and middle temporal gyrus were notably increased in the acupuncture group after treatment.CONCLUSION:Acupuncture combined with conventional treatment can significantly improve the corrected visual acuity and optic nerve conduction in children with anisometropic amblyopia.Compared with the conventional treatment,the regulation of acupuncture on the functional activities of the relevant brain areas in the anterior cerebellum may be an effective acupuncture mechanism for anisometropic amblyopia.展开更多
Background:To investigate the effects of acupuncture on post-stroke limb spasm model rats and the underlying mechanism.Methods:A total of 50 Sprague-Dawley rats were randomly divided into three groups,Control group(10...Background:To investigate the effects of acupuncture on post-stroke limb spasm model rats and the underlying mechanism.Methods:A total of 50 Sprague-Dawley rats were randomly divided into three groups,Control group(10),Model group(20)and Zhishen Tiaoxing(ZSTX)acupuncture group(20).Middle cerebral artery occlusion was conducted in SD rats to establish post-stroke limb spasm rats,which were treated with ZSTX acupuncture.Behavioral assays were determined by the Narrow ally test,the limb muscle tension was detected by the BL-420S test system,and infarct volume was assessed after the cerebral infarction by 2,3,5-triphenyltetrazolium chloride staining.Heterogeneous neurotransmitterγ-aminobutyric acid(GABA)and its receptors GABAA and GABAB in the cerebral cortex of the infarct area were determined by immunofluorescence assay.The release of Trkb and K-Cl cotransporter isoform 2 was detected by an immunofluorescence double labeling study.Western Blot was utilized to measure the expression of BDNF and Trkb.Results:The results showed that the behavioral assays in post-stroke limb spasm rats were significantly improved by the treatment of ZSTX acupuncture.14 days of ZSTX acupuncture can effectively inhibit muscle tone and decrease Infarct volume,which was measured with BL-420S biological function experiment system and triphenyltetrazolium chloride.Meanwhile,the results of Double-Label Immunofluorescence Assays showed that ZSTX acupuncture improved the expression of GABA,GABAA,GABAB,BDNF,and K-Cl cotransporter isoform 2.Double-Label Immunofluorescence Assays and WB results showed that 14 days ZSTX acupuncture declined the expression of Trkb.Conclusions:Our results suggest that 14 days of ZSTX acupuncture can significantly improve post-stroke limb spasm.Meanwhile,the pathogenesis of post-stroke limb spasm and the efficacy of ZSTX acupuncture involve metabolic pathways of neurotransmitters,and electro-acupuncture can treat post-stroke limb spasm by regulating BDNF/Trkb-KCC2 signaling pathway.展开更多
BACKGROUND Breast cancer is a multifaceted and formidable disease with profound public health implications.Cell demise mechanisms play a pivotal role in breast cancer pathogenesis,with ATP-triggered cell death attract...BACKGROUND Breast cancer is a multifaceted and formidable disease with profound public health implications.Cell demise mechanisms play a pivotal role in breast cancer pathogenesis,with ATP-triggered cell death attracting mounting interest for its unique specificity and potential therapeutic pertinence.AIM To investigate the impact of ATP-induced cell death(AICD)on breast cancer,enhancing our understanding of its mechanism.METHODS The foundational genes orchestrating AICD mechanisms were extracted from the literature,underpinning the establishment of a prognostic model.Simultaneously,a microRNA(miRNA)prognostic model was constructed that mirrored the gene-based prognostic model.Distinctions between high-and low-risk cohorts within mRNA and miRNA characteristic models were scrutinized,with the aim of delineating common influence mechanisms,substantiated through enrichment analysis and immune infiltration assessment.RESULTS The mRNA prognostic model in this study encompassed four specific mRNAs:P2X purinoceptor 4,pannexin 1,caspase 7,and cyclin 2.The miRNA prognostic model integrated four pivotal miRNAs:hsa-miR-615-3p,hsa-miR-519b-3p,hsa-miR-342-3p,and hsa-miR-324-3p.B cells,CD4+T cells,CD8+T cells,endothelial cells,and macrophages exhibited inverse correlations with risk scores across all breast cancer subtypes.Furthermore,Kyoto Encyclopedia of Genes and Genomes analysis revealed that genes differentially expressed in response to mRNA risk scores significantly enriched 25 signaling pathways,while miRNA risk scores significantly enriched 29 signaling pathways,with 16 pathways being jointly enriched.CONCLUSION Of paramount significance,distinct mRNA and miRNA signature models were devised tailored to AICD,both potentially autonomous prognostic factors.This study's elucidation of the molecular underpinnings of AICD in breast cancer enhances the arsenal of potential therapeutic tools,offering an unparalleled window for innovative interventions.Essentially,this paper reveals the hitherto enigmatic link between AICD and breast cancer,potentially leading to revolutionary progress in personalized oncology.展开更多

Our previous studies showed that tuina regulates relevant regions and indices of the spinal dorsal horn using the Dian, Bo, and Rou method in Yinmen(BL37), Yanglingquan(GB34), and Weizhong(BL40). Treatment prevents muscle atrophy, protects spinal cord neurons, and promotes sciatic nerve repair. The mechanisms of action of tuina for treating peripheral nerve injury remain poorly understood. This study established rat models of sciatic nerve injury using the crushing method. Rats received Chinese tuina in accordance with the principle of "Three Methods and Three Points," once daily for 20 days. Tuina intervention reduced paw withdrawal latency and improved wet weight of the gastrocnemius muscle, as well as promoting morphological recovery of sciatic nerve fibers, Schwann cells, and axons. The protein expression levels of phospho-p38 mitogen-activated protein kinase, tumor necrosis factor-α, and interleukin-1β also decreased. These findings indicate that "Three Methods and Three Points" promoted morphological recovery and improved behavior of rats with peripheral nerve injury.展开更多

Background:To systematically evaluate the efficacy and safety of the Chinese patent medicine Shuxuening injection combined with western medicine in vascular dementia treatment.Methods:Randomized controlled trials of t...Background:To systematically evaluate the efficacy and safety of the Chinese patent medicine Shuxuening injection combined with western medicine in vascular dementia treatment.Methods:Randomized controlled trials of the Chinese patent medicine Shuxuening injection in vascular dementia treatment is searched in the databases of CNKI,Wanfang,VIP,CBM,PubMed,The Cochrane Library,Embase and Web of Science from the establishment time to December 2020.After screen the literature,extract the data and evaluate the bias risk of studies included;tRevMan5.3 was used for met-analysis.Results:Nine randomized controlled trials including 932 patients were included.The results of meta-analysis included:(1)the total effective rate(RR=1.27,95%CI(1.18,1.36),P<0.00001);(2)MMSE score(MD=4.78,95%CI(1.75,7.80),P=0.002);(3)ADL score(MD=8.87,95%CI(6.70,11.05),P<0.00001);(4)NIHSS score(MD=−6.60,95%CI(−7.04,−6.16).P<0.00001).The results of meta-analysis of the test group are better than those in the control group.Conclusion:The Chinese patent medicine Shuxuening injection combined with conventional western medicine has showed some advantages in the total effective rate,MMSE score,ADL score,NIHSS score than conventional western medicine without more side effects in vascular dementia treatment.More randomized,double-blind,large sample clinical studies are needed to confirm the above conclusions.展开更多
Objective:To investigate the mechanism of the effect of Astragalus membranaceus(A.membranaceus)on lung adenocarcinoma at the molecular level to elucidate the specific targets according to the network pharmacology appr...Objective:To investigate the mechanism of the effect of Astragalus membranaceus(A.membranaceus)on lung adenocarcinoma at the molecular level to elucidate the specific targets according to the network pharmacology approach.Methods:The active components of A.membranaceus and their potential targets were col ected from the Traditional Chinese Medicine Systems Pharmacology Database.Lung adenocarcinoma-associated genes were acquired based on Gene Cards,Online Mendelian Inheritance in Man(OMIM),Pharm GKB,and Therapeutic Targets databases.The PI3K/AKT signaling pathway-related genes were obtained using Reactome portal.Networks of"ingredient-target"and"ingredient-target-pathway-disease"were constructed using the Cytoscape3.6.0 software.The relationships among targets were analyzed according protein-protein interaction(PPI)network.Finally,molecular docking was applied to construct the binding conformation between active ingredients and core targets.Cel counting kit 8(CCK8)and Western blot assays were performed to determine the mechanism of the key ingredient of A.membranaceus.Results:A total of 20 active components and their 329 targets,and 7,501 lung adenocarcinoma-related genes and 130 PI3K/AKT signaling pathway-related genes were obtained.According to Venn diagram and PPI network analysis,2 mainly active ingredients,including kaempferol and quercetin,and 6 core targets,including TP53,MAPK1,EGF,AKT1,ERBB2,and EGFR,were identified.The two important active ingredients of A.membranaceus,kaempferol and quercetin,exert the therapeutic effect in lung adenocarcinoma partly by acting on the 6 core targets(TP53,MAPK1,EGF,AKT1,ERBB2,and EGFR)of PI3K/AKT signaling pathway.Expressions of potential targets in lung adenocarcinoma and normal samples were analyzed by using UALCAN portal and found that ERBB2 was overexpressed in lung adenocarcinoma tissues and upregulation of it correlated with clinicopathological characteristics.Final y,quercetin repressed viabilities of lung adenocarcinoma cells by targeting ERBB2 on PI3K/AKT signaling confirmed by CCK8 and Western blot.Conclusion:Our finding unraveled that an active ingredient of A.membranaceus,quercetin,significantly inhibited the lung adenocarcinoma cel s proliferation by repressing ERBB2 level and inactivating the PI3K/AKT signaling pathway.展开更多

OBJECTIVE: To investigate changes in gene expression profiles in the hypothalamus related to the effects of acupuncture at the Renying(ST 9) acupoint in spontaneously hypertensive(SH) rats.METHODS: We randomly divided...OBJECTIVE: To investigate changes in gene expression profiles in the hypothalamus related to the effects of acupuncture at the Renying(ST 9) acupoint in spontaneously hypertensive(SH) rats.METHODS: We randomly divided 18 SH rats into Renying(ST 9) group and model control group, 9 body weight-matched Wistar-Kyoto rats were used as blank controls. Acupuncture was performed manually for 20-min daily over 28 d in the Renying(ST 9) group. Rat Gene 2.0 array technology was used for the determination of gene expression profiles and the screened key genes were verified by real-time quantitative polymerase chain reaction(RT-PCR) analyses.RESULTS: The different groups exhibited differential gene expression: compared with the blank control group, 48 genes were up-regulated and 91 genes were down-regulated in the model group;compared with the model group, 79 genes were up-regulated and 80 genes were down-regulated in Renying(ST 9) group. The RT-PCR results of the key genes including Chi3 l1, Ephx2, Klk1, 5-HT1 A and Cbs were consistent with that of gene chip analysis.CONCLUTION: Acupuncture at Renying(ST 9)could significantly lower the blood pressure of SH rats and affect their hypothalamic gene expression profile. Genes associated with the contraction of vascular smooth muscle and the regulation of inflammation, neurotransmitters may be involved in acupuncture's antihypertensive mechanism.展开更多
Objectives:To study the acupoint specificity through the comprehensive evaluation of the acupuncture effect on rat model with the middle cerebral artery occlusion(MCAO).Methods:Duplicated MCAO model by Zea-longa's...Objectives:To study the acupoint specificity through the comprehensive evaluation of the acupuncture effect on rat model with the middle cerebral artery occlusion(MCAO).Methods:Duplicated MCAO model by Zea-longa's thread ligation was assessed by Zausinger's six-point method,and rats with 1-3 scores were chosen to be grouped.According to the randomized and controlled principle,rats were divided into 6 basic control groups(including normal group,sham group,model control group,model group without intervention,Nimodipine group,lateral-to-Renzhong group)and 6 acupuncture groups[including Neiguan(PC6)group,Weizhong(BL40)group,Chize(LU5)group,Sanyinjiao(SP6)group,Renzhong(DU6)group and non-acupoint group];in each acupoint or non-acupoint 9 different parameter combinations were respectively set by orthogonal intersection method.There were in total 60 groups,and each group had 12 rats.The rats were treated by acupuncture with the lifting-thrusting manipulation every 12 h,in total 6 times.Indices of neurobehavioral scores,cerebral blood flow,infarction rate,microcirculation,and light microscope,etc.were measured.In order to comprehensively evaluate the acupuncture effect to research the specificity of acupoint,a cluster analysis was made with SPSS17.0 for the comprehensive effect scores of the samples computed according to the comprehensive evaluation models,and then sorting and classification were made for the comprehensive effect scores.Results:In the acupuncture groups every acupoint had different therapeutic effect due to different acupuncture parameter combinations;among the acupuncture groups the orders of curative effect were as followings:Neiguan group was of exceptional result,Weizhong group and Sanyinjiao group were of valid results in upper level,Chize group and Renzhong group were of valid results in lower level,and non-acupoint group was of invalid result;the therapeutic result of acupoint group was superior to non-acupoint group;and the curative effect of acupuncture group was also superior to basic control group.Conclusion:The acupoint has the specificity of acupuncture effect,and the acupoint specificity is relative.展开更多
Objective:To observe the specificity of reinforcing and reducing methods of mild moxibustion and explore its clinical significance.Methods:Thirty healthy students were selected and given reinforcing of mild moxibustio...Objective:To observe the specificity of reinforcing and reducing methods of mild moxibustion and explore its clinical significance.Methods:Thirty healthy students were selected and given reinforcing of mild moxibustion on the left Neiguan(PC 6)and reducing on the right Neiguan(PC 6)respectively.Temperature of the acupoint skin surface and the temperature-time curve were recorded by a digital thermometer.Results:In application of mild moxibustion on Neiguan(PC 6),there was a statistical difference in the temperature-increase section between reinforcing and reducing methods(P<0.01).The temperature-increasing section of the temperature-time curve in the reducing method was obviouslysteeper than that in the reinforcing method.In the period of0.5-3 min during moxibustion,the temperature of Neiguan(PC 6)was remarkably higher in the reducing than in the reinforcing(P<0.01).In the application of the reducing,the time for the temperature of Neiguan(PC 6)to increase to 40℃was remarkably advanced(P<0.05)and the rate of burning pain in the subjects was remarkably higher in the reducing method than in the reinforcing method(P<0.001).Conclusion:In comparison with the reinforcing method,the acupoint temperature increased more quickly in the application of reducing method and can produce obvious hot stimulation in a short period of time.In the application of the reinforcing method,the acupoint temperature increased mildly and could enhance the tolerance of the organism to the high temperature.展开更多

文摘Background:To investigate the effects of acupuncture on post-stroke limb spasm model rats and the underlying mechanism.Methods:A total of 50 Sprague-Dawley rats were randomly divided into three groups,Control group(10),Model group(20)and Zhishen Tiaoxing(ZSTX)acupuncture group(20).Middle cerebral artery occlusion was conducted in SD rats to establish post-stroke limb spasm rats,which were treated with ZSTX acupuncture.Behavioral assays were determined by the Narrow ally test,the limb muscle tension was detected by the BL-420S test system,and infarct volume was assessed after the cerebral infarction by 2,3,5-triphenyltetrazolium chloride staining.Heterogeneous neurotransmitterγ-aminobutyric acid(GABA)and its receptors GABAA and GABAB in the cerebral cortex of the infarct area were determined by immunofluorescence assay.The release of Trkb and K-Cl cotransporter isoform 2 was detected by an immunofluorescence double labeling study.Western Blot was utilized to measure the expression of BDNF and Trkb.Results:The results showed that the behavioral assays in post-stroke limb spasm rats were significantly improved by the treatment of ZSTX acupuncture.14 days of ZSTX acupuncture can effectively inhibit muscle tone and decrease Infarct volume,which was measured with BL-420S biological function experiment system and triphenyltetrazolium chloride.Meanwhile,the results of Double-Label Immunofluorescence Assays showed that ZSTX acupuncture improved the expression of GABA,GABAA,GABAB,BDNF,and K-Cl cotransporter isoform 2.Double-Label Immunofluorescence Assays and WB results showed that 14 days ZSTX acupuncture declined the expression of Trkb.Conclusions:Our results suggest that 14 days of ZSTX acupuncture can significantly improve post-stroke limb spasm.Meanwhile,the pathogenesis of post-stroke limb spasm and the efficacy of ZSTX acupuncture involve metabolic pathways of neurotransmitters,and electro-acupuncture can treat post-stroke limb spasm by regulating BDNF/Trkb-KCC2 signaling pathway.

文摘BACKGROUND Breast cancer is a multifaceted and formidable disease with profound public health implications.Cell demise mechanisms play a pivotal role in breast cancer pathogenesis,with ATP-triggered cell death attracting mounting interest for its unique specificity and potential therapeutic pertinence.AIM To investigate the impact of ATP-induced cell death(AICD)on breast cancer,enhancing our understanding of its mechanism.METHODS The foundational genes orchestrating AICD mechanisms were extracted from the literature,underpinning the establishment of a prognostic model.Simultaneously,a microRNA(miRNA)prognostic model was constructed that mirrored the gene-based prognostic model.Distinctions between high-and low-risk cohorts within mRNA and miRNA characteristic models were scrutinized,with the aim of delineating common influence mechanisms,substantiated through enrichment analysis and immune infiltration assessment.RESULTS The mRNA prognostic model in this study encompassed four specific mRNAs:P2X purinoceptor 4,pannexin 1,caspase 7,and cyclin 2.The miRNA prognostic model integrated four pivotal miRNAs:hsa-miR-615-3p,hsa-miR-519b-3p,hsa-miR-342-3p,and hsa-miR-324-3p.B cells,CD4+T cells,CD8+T cells,endothelial cells,and macrophages exhibited inverse correlations with risk scores across all breast cancer subtypes.Furthermore,Kyoto Encyclopedia of Genes and Genomes analysis revealed that genes differentially expressed in response to mRNA risk scores significantly enriched 25 signaling pathways,while miRNA risk scores significantly enriched 29 signaling pathways,with 16 pathways being jointly enriched.CONCLUSION Of paramount significance,distinct mRNA and miRNA signature models were devised tailored to AICD,both potentially autonomous prognostic factors.This study's elucidation of the molecular underpinnings of AICD in breast cancer enhances the arsenal of potential therapeutic tools,offering an unparalleled window for innovative interventions.Essentially,this paper reveals the hitherto enigmatic link between AICD and breast cancer,potentially leading to revolutionary progress in personalized oncology.

文摘Tuina is a traditional Chinese treatment for sensory disturbances caused by peripheral nerve injury and related diseases. Our previous studies showed that tuina regulates relevant regions and indices of the spinal dorsal horn using the Dian, Bo, and Rou method in Yinmen(BL37), Yanglingquan(GB34), and Weizhong(BL40). Treatment prevents muscle atrophy, protects spinal cord neurons, and promotes sciatic nerve repair. The mechanisms of action of tuina for treating peripheral nerve injury remain poorly understood. This study established rat models of sciatic nerve injury using the crushing method. Rats received Chinese tuina in accordance with the principle of "Three Methods and Three Points," once daily for 20 days. Tuina intervention reduced paw withdrawal latency and improved wet weight of the gastrocnemius muscle, as well as promoting morphological recovery of sciatic nerve fibers, Schwann cells, and axons. The protein expression levels of phospho-p38 mitogen-activated protein kinase, tumor necrosis factor-α, and interleukin-1β also decreased. These findings indicate that "Three Methods and Three Points" promoted morphological recovery and improved behavior of rats with peripheral nerve injury.

文摘Obesity is a common clinical disease,in order to take a healthy and simple way to treat it,the volatile properties of traditional Chinese medicine play a role in lifting weights.Aromatherapy uses the four qi and five flavors of traditional Chinese medicine as a typical representative.In order to explore the mechanism of action of aromatherapy in depth,through the etiology and pathogenesis of obesity,the historical origin of modern times,related organs,pathogenesis,and the research progress of aromatherapy in the treatment of obesity,the classification of aromatherapy,and the related research of aromatic drugs,it is found that aromatherapy has a large number of significant clinical effects in modern times.It can be used to treat obesity independently under the guidance of the basic theory of Chinese medicine,and can also be combined with other Chinese medicines to form prescription drug treatment,and in combination with other ways to treat obesity.However,at present,this method of treating obesity has not received widespread attention,and it still needs to be studied and promoted by scholars.

文摘Non-alcoholic fatty liver disease(NAFLD)is a common chronic liver disease characterized by diffuse hepatic steatosis.With the improvement of people's living standard,the incidence rate of NAFLD has been increasing,which has become one of the global health problems in 21st Century.However,there is no specific drug or standard treatment for NAFLD,which brings challenges to treatment.Acupuncture,moxibustion,massage and other external therapies based on the characteristics of traditional Chinese medicine have obvious curative effect in the clinical treatment of NAFLD,but the mechanism has not been systematically explained,which makes the clinical promotion evidence insufficient.This paper aims to summarize the researches on the treatment of NAFLD by external therapies of traditional Chinese medicine in recent years,and analyze its possible mechanism,so as to provide a scientific theoretical basis for future basic experiments and clinical research,and form a set of standardized clinical diagnosis and treatment scheme with the characteristics of traditional Chinese medicine.

文摘Bronchial asthma(asthma for short)is a common chronic respiratory disease inchildren,which has adverse effects on children's physical and mental health.Nuclear factorκB(NF-κB)signaling pathway is an important transcription factor in airway inflammation,and plays an important role in airway inflammation and airway remodeling in asthma.Clinical studies have found that traditional Chinese medicine has positive effects in the treatment of asthma.Hence,this paper aims to review the literatures on the treatment of asthma through traditional Chinese medicine compound and single herbs and extracts by regulating the NF-κB signaling pathway for nearly a decade.It is found that traditional Chinese medicine can inhibit airway inflammation,relieve airway smooth muscle spasm and reduce airway resistance through the regulation of NF-κB signaling pathway,so as to play a key role in the treatment of asthma,and provide a scientific basis for the treatment of asthma with traditional Chinese medicine.
文摘Background:To systematically evaluate the efficacy and safety of the Chinese patent medicine Shuxuening injection combined with western medicine in vascular dementia treatment.Methods:Randomized controlled trials of the Chinese patent medicine Shuxuening injection in vascular dementia treatment is searched in the databases of CNKI,Wanfang,VIP,CBM,PubMed,The Cochrane Library,Embase and Web of Science from the establishment time to December 2020.After screen the literature,extract the data and evaluate the bias risk of studies included;tRevMan5.3 was used for met-analysis.Results:Nine randomized controlled trials including 932 patients were included.The results of meta-analysis included:(1)the total effective rate(RR=1.27,95%CI(1.18,1.36),P<0.00001);(2)MMSE score(MD=4.78,95%CI(1.75,7.80),P=0.002);(3)ADL score(MD=8.87,95%CI(6.70,11.05),P<0.00001);(4)NIHSS score(MD=−6.60,95%CI(−7.04,−6.16).P<0.00001).The results of meta-analysis of the test group are better than those in the control group.Conclusion:The Chinese patent medicine Shuxuening injection combined with conventional western medicine has showed some advantages in the total effective rate,MMSE score,ADL score,NIHSS score than conventional western medicine without more side effects in vascular dementia treatment.More randomized,double-blind,large sample clinical studies are needed to confirm the above conclusions.

文摘Objective:To investigate the mechanism of the effect of Astragalus membranaceus(A.membranaceus)on lung adenocarcinoma at the molecular level to elucidate the specific targets according to the network pharmacology approach.Methods:The active components of A.membranaceus and their potential targets were col ected from the Traditional Chinese Medicine Systems Pharmacology Database.Lung adenocarcinoma-associated genes were acquired based on Gene Cards,Online Mendelian Inheritance in Man(OMIM),Pharm GKB,and Therapeutic Targets databases.The PI3K/AKT signaling pathway-related genes were obtained using Reactome portal.Networks of"ingredient-target"and"ingredient-target-pathway-disease"were constructed using the Cytoscape3.6.0 software.The relationships among targets were analyzed according protein-protein interaction(PPI)network.Finally,molecular docking was applied to construct the binding conformation between active ingredients and core targets.Cel counting kit 8(CCK8)and Western blot assays were performed to determine the mechanism of the key ingredient of A.membranaceus.Results:A total of 20 active components and their 329 targets,and 7,501 lung adenocarcinoma-related genes and 130 PI3K/AKT signaling pathway-related genes were obtained.According to Venn diagram and PPI network analysis,2 mainly active ingredients,including kaempferol and quercetin,and 6 core targets,including TP53,MAPK1,EGF,AKT1,ERBB2,and EGFR,were identified.The two important active ingredients of A.membranaceus,kaempferol and quercetin,exert the therapeutic effect in lung adenocarcinoma partly by acting on the 6 core targets(TP53,MAPK1,EGF,AKT1,ERBB2,and EGFR)of PI3K/AKT signaling pathway.Expressions of potential targets in lung adenocarcinoma and normal samples were analyzed by using UALCAN portal and found that ERBB2 was overexpressed in lung adenocarcinoma tissues and upregulation of it correlated with clinicopathological characteristics.Final y,quercetin repressed viabilities of lung adenocarcinoma cells by targeting ERBB2 on PI3K/AKT signaling confirmed by CCK8 and Western blot.Conclusion:Our finding unraveled that an active ingredient of A.membranaceus,quercetin,significantly inhibited the lung adenocarcinoma cel s proliferation by repressing ERBB2 level and inactivating the PI3K/AKT signaling pathway.

文摘OBJECTIVE: To investigate changes in gene expression profiles in the hypothalamus related to the effects of acupuncture at the Renying(ST 9) acupoint in spontaneously hypertensive(SH) rats.METHODS: We randomly divided 18 SH rats into Renying(ST 9) group and model control group, 9 body weight-matched Wistar-Kyoto rats were used as blank controls. Acupuncture was performed manually for 20-min daily over 28 d in the Renying(ST 9) group. Rat Gene 2.0 array technology was used for the determination of gene expression profiles and the screened key genes were verified by real-time quantitative polymerase chain reaction(RT-PCR) analyses.RESULTS: The different groups exhibited differential gene expression: compared with the blank control group, 48 genes were up-regulated and 91 genes were down-regulated in the model group;compared with the model group, 79 genes were up-regulated and 80 genes were down-regulated in Renying(ST 9) group. The RT-PCR results of the key genes including Chi3 l1, Ephx2, Klk1, 5-HT1 A and Cbs were consistent with that of gene chip analysis.CONCLUTION: Acupuncture at Renying(ST 9)could significantly lower the blood pressure of SH rats and affect their hypothalamic gene expression profile. Genes associated with the contraction of vascular smooth muscle and the regulation of inflammation, neurotransmitters may be involved in acupuncture's antihypertensive mechanism.

文摘Objectives:To study the acupoint specificity through the comprehensive evaluation of the acupuncture effect on rat model with the middle cerebral artery occlusion(MCAO).Methods:Duplicated MCAO model by Zea-longa's thread ligation was assessed by Zausinger's six-point method,and rats with 1-3 scores were chosen to be grouped.According to the randomized and controlled principle,rats were divided into 6 basic control groups(including normal group,sham group,model control group,model group without intervention,Nimodipine group,lateral-to-Renzhong group)and 6 acupuncture groups[including Neiguan(PC6)group,Weizhong(BL40)group,Chize(LU5)group,Sanyinjiao(SP6)group,Renzhong(DU6)group and non-acupoint group];in each acupoint or non-acupoint 9 different parameter combinations were respectively set by orthogonal intersection method.There were in total 60 groups,and each group had 12 rats.The rats were treated by acupuncture with the lifting-thrusting manipulation every 12 h,in total 6 times.Indices of neurobehavioral scores,cerebral blood flow,infarction rate,microcirculation,and light microscope,etc.were measured.In order to comprehensively evaluate the acupuncture effect to research the specificity of acupoint,a cluster analysis was made with SPSS17.0 for the comprehensive effect scores of the samples computed according to the comprehensive evaluation models,and then sorting and classification were made for the comprehensive effect scores.Results:In the acupuncture groups every acupoint had different therapeutic effect due to different acupuncture parameter combinations;among the acupuncture groups the orders of curative effect were as followings:Neiguan group was of exceptional result,Weizhong group and Sanyinjiao group were of valid results in upper level,Chize group and Renzhong group were of valid results in lower level,and non-acupoint group was of invalid result;the therapeutic result of acupoint group was superior to non-acupoint group;and the curative effect of acupuncture group was also superior to basic control group.Conclusion:The acupoint has the specificity of acupuncture effect,and the acupoint specificity is relative.

文摘Objective:To observe the specificity of reinforcing and reducing methods of mild moxibustion and explore its clinical significance.Methods:Thirty healthy students were selected and given reinforcing of mild moxibustion on the left Neiguan(PC 6)and reducing on the right Neiguan(PC 6)respectively.Temperature of the acupoint skin surface and the temperature-time curve were recorded by a digital thermometer.Results:In application of mild moxibustion on Neiguan(PC 6),there was a statistical difference in the temperature-increase section between reinforcing and reducing methods(P<0.01).The temperature-increasing section of the temperature-time curve in the reducing method was obviouslysteeper than that in the reinforcing method.In the period of0.5-3 min during moxibustion,the temperature of Neiguan(PC 6)was remarkably higher in the reducing than in the reinforcing(P<0.01).In the application of the reducing,the time for the temperature of Neiguan(PC 6)to increase to 40℃was remarkably advanced(P<0.05)and the rate of burning pain in the subjects was remarkably higher in the reducing method than in the reinforcing method(P<0.001).Conclusion:In comparison with the reinforcing method,the acupoint temperature increased more quickly in the application of reducing method and can produce obvious hot stimulation in a short period of time.In the application of the reinforcing method,the acupoint temperature increased mildly and could enhance the tolerance of the organism to the high temperature.
