This study aimed to evaluate the in vivo protective effects and possible mechanism of L. brevis FZU0713 on the disturbance of lipid metabolism in hyperlipidemic rats fed a high-fat diet(HFD). Results showed that oral administration of L. brevis FZU0713 could significantly inhibit obesity, ameliorate the lipid metabolism disorder, including serum/liver biochemical parameters and hepatic oxidative stress in HFD-fed rats. Histopathological result also indicated that dietary intervention of L. brevis FZU0713 could reduce the accumulation of lipid droplets in liver induced by 8 weeks HFD feeding. Furthermore, L. brevis FZU0713 intervention significantly increased the fecal levels of short-chain fatty acids(SCFAs, including acetate, propionate, butyrate, isobutyrate, valerate and isovalerate)in HFD-fed rats, which may be closely related to the changes of intestinal microbial composition and metabolic function. Intestinal microbiota profiling by 16S rRNA gene sequencing revealed that L. brevis FZU0713 intervention significantly altered the relative abundance of Coprococcus, Butyricicoccus, Intestinimonas, Lachnospiraceae FCS020 group, Ruminococcaceae_NK4A214 group, Ruminococcaceae_UCG-005 and UCG-014 at genus levels. Based on Spearman's rank correlation coefficient, serum and liver lipid metabolism related biochemical parameters were positively correlated with genera Ruminococcus, Pediococcus and Lachnospiraceae, but negatively correlated with genera Pseudoflavonifractor, Butyricicoccus and Intestinimonas. Furthermore, liver metabolomics analysis demonstrated that L. brevis FZU0713 had a significant regulatory effect on the composition of liver metabolites in hyperlipidemic rats, especially the levels of some important biomarkers involved in the metabolic pathways of arachidonic acid metabolism, primary bile acid biosynthesis, amino sugar and nucleotide sugar metabolism, taurine and hypotaurine metabolism, biosynthesis of unsaturated fatty acid, fructose and mannose metabolism, tyrosine metabolism, etc. Additionally, oral administration of L. brevis FZU0713 significantly regulated the mR NA levels of liver genes(including Acat2, Acox1, Hmgcr, Cd36, Srebp-1c and Cyp7a1)involved in lipid metabolism and bile acid homeostasis. In conclusion, our findings provide the evidence that L. brevis FZU0713 has the potential to improve disturbance of lipid metabolism by regulating intestinal microflora and liver metabonomic profile. Therefore, L. brevis FZU0713 may be used as a potential probiotic strain to produce functional food to prevent hyperlipidemia.展开更多

Along the ...Spatial and temporal variation of fish assemblages were investigated seasonally from May 2007 to February 2008across 11 study sites in a subtropical small stream, the Puxi Stream, of the Huangshan Mountain. Along the longitudinal gradientfrom headwater to downstream, fish species richness and abundance increased gradually, but then decreased significantly at thelower reaches. The highest species richness and abundance were observed in August and the lowest in February. Based on analysisof similarities (ANOSIM), fish assemblages were significantly different in spatial variation but not in temporal variation. Althoughdifferences were observed both among sites and among stream orders, the lower R value in order-variation suggestedstream order was not the optimal factor explaining the spatial variation of fish assemblages. In addition, dam construction did notsignificantly alter fish assemblages in the sites adjacent to and immediately downstream to dams. Using cluster analysis andnon-metric Multi Dimensional Scaling analysis (NMS), assemblages were separated into three groups at a Bray-Curtis similarityvalue of 42%: the upper, middle and lower groups. Following analysis of similarity percentages of species contributions (SIMPER),shifts in occurrence or abundance of S. curriculus, Z. platypus, R. bitterling and A.fasciatus contributed most to the differencesamongst the three groups. Standard Deviation Redundancy Analysis (RDA) suggested that habitat structure (such as elevation,substrate, and flow velocity) contributed to the spatial and temporal pattern of fish assemblages in the Puxi Stream. In conclusion,the fish assemblages in Puxi Stream presented significant spatial but not temporal variation. Human disturbance has perhapsinduced the decrease in species diversity in the lower reaches. In experiment 1, metmyoglobin(MetMb), substrate and inhibitors were incubated with mitochondria. Comparsion of the effects of succinate and NADH on MetMb reduction was investigated. The MetMb percentage in sample treated with 8 mol L-1 succinate decreased by about 69% after 3 h incubation, and the effect was inhibited by the addition of 10 mol L-1 electron transfer chain complex II inhibitor malonic acid; the MetMb percentage in samples treated with 2 mol L-1 NADH decreased by 56% and the effect was inhibited by the addition of 0.02 mol L-1 electron transport chain complex I inhibitor rotenone. These results indicated that electron transport chain played an important role in MetMb reduction. Both complex II and complex I take part in the MetMb reduction in mitochondria through different pathways. NADH-MetMb reduction system was less stable than succinateMetMb system. In experiment 2, the beef longissimus dorsi muscle was blended with different concentrations of succinate or NADH. Enhancing patties with higher concentration of succinate or NADH improved colour stability in vacuum packaged samples(P<0.05). These results verified that mitochondria electron transport chain is related to the MetMb reduction in meat system.展开更多
Background:MiRNAs act as pivotal post-transcriptional gene mediators in the regulation of diverse biological processes,including proliferation,development and apoptosis.Our previous study has showed that miR-101-3p is...Background:MiRNAs act as pivotal post-transcriptional gene mediators in the regulation of diverse biological processes,including proliferation,development and apoptosis.Our previous study has showed that miR-101-3p is differentially expressed in dairy goat ovaries compared single with multiple litters.The objective of this research was to explore the potential function and molecular mechanism of miR-101-3p via its target STC1 in goat ovarian growth and development.Results:cDNA libraries were constructed using goat granulosa cells transfected with miR-101-3p mimics and negative control by RNA-sequencing.In total,142 differentially expressed unigenes(DEGs)were detected between two libraries,including 78 down-regulated and 64 up-regulated genes.GO and KEGG enrichment analysis showed the potential impacts of DEGs on ovarian development.STC1 was singled out from DEGs for further research owing to it regulates reproductive-related processes.In vitro,bioinformatics analysis and 3′-UTR assays confirmed that STC1 was a target of miR-101-3p.ELISA was performed to detect the estrogen(E2)and progesterone(P4)levels.CCK8,EdU and flow cytometry assays were performed to detect the proliferation and apoptosis of granulosa cells.Results showed that miR-101-3p regulated STAR,CYP19A1,CYP11A1 and 3β-HSD steroid hormone synthesis-associated genes by STC1 depletion,thus promoted E2 and P4 secretions.MiR-101-3p also affected the key protein PI3K,PTEN,AKT and mTOR in PI3K-AKT pathway by STC1,thereby suppressing proliferation and promoting apoptosis of granulosa cells.In vivo,the distribution and expression levels of miR-101-3p in mouse ovaries were determined through fluorescence in situ hybridisation(FISH).Immunohistochemistry results showed that STC1 expression was suppressed in mouse ovaries in miR-101-3p-agonist and siRNA-STC1 groups.Small and stunted ovarian fragments,decreased numbers of follicles at diverse stages were observed using Hematoxylin-eosin(HE)staining,thereby showing unusual ovarian development after miR-101-3p overexpression or STC1 depletion.Inhibition of miR-101-3p manifested opposite results.Conclusions:Taken together,our results demonstrated a regulatory mechanism of miR-101-3p via STC1 in goat granulosa cells,and offered the first in vivo example of miR-101-3p and STC1 functions required for ovarian development.展开更多
Grapevine(Vitis spp.)is one of the most economically important fruit crops worldwide,and there is considerable interest in improving its major agronomic and enological traits in response to ever-changing agricultural ...Grapevine(Vitis spp.)is one of the most economically important fruit crops worldwide,and there is considerable interest in improving its major agronomic and enological traits in response to ever-changing agricultural environments and consumer demands.Molecular genetic techniques in particular,associated with rapid technological advancements,provide an attractive alternative to conventional breeding approaches for developing new grapevine varieties with enhanced yield performance,quality,stress tolerance and disease resistance.To date,several grapevine varieties have been transformed with genes associated with diverse functions through biolistic bombardment and/or Agrobacterium-mediated transformation,and transgenic grape lines have been obtained using established regeneration systems.Nevertheless,a wide range of factors,including genotype,explant source and culture medium,have been shown to affect the efficiency of plant regeneration.Moreover,the selection and use of acceptor materials,bacterial strain and cell density,selectable markers and selection methods also influence transformation efficiency.This paper provides an overview of recent advances in grapevine regeneration and genetic transformation and in-depth discussion of the major limiting factors,and discusses promising future strategies to develop robust plant regeneration and genetic transformation in grapevine.展开更多

The length of the sequences was 953 bp.The deduced amino acid sequences analysis showed that the sequence contained the complete coding region of a typical NAC gene, including a complete open reading frame which was 939 bp. The study analyzed the expression patterns of NAC transcript gene induced in various PEG(polyethylene glycol) simulated drought condition. Real-time PCR indicated that maize NAC gene can be induced in various PEG to abiotic stress responses, it could express constitutively to adopt the drought condition in short time, and the higher drought, the more early expression, which indicated that NAC transcript gene could be involved in the regulation of the defense response.展开更多

The antimicrobial activity of LS was tested by minimum inhibition concentration (MIC...In this study,the in vitro antimicrobial and antiviral activities of the lysozyme from marine strain S-12-86 (LS) were investigated. The antimicrobial activity of LS was tested by minimum inhibition concentration (MIC) and minimum bactericidal concentration (MBC) method. The inhibiting effects of LS on pseudo rabies virus (PRV) in swine kidney cells (PK-15 cells) were judged by cytopathogenic effect test (CPE). The results showed LS had a broad antimicrobial spectrum against several standard strains including gram-positive bacteria,gram-negative bacteria,fungi,etc. The MIC of LS was 0.25-4.00 mg mL-1 and its MBC was 0.25-8.00 mg mL-1,respectively. Observation under the transmission electron microscope revealed that the cell wall of Candida albicans was distorted seriously,and the cytoplasm with many cavities was asymmetrical after being hydrolyzed by LS. The median cytotoxicity concentration (TC50) of LS was 100.0 μg mL-1,the median effective concentration (EC50) was 0.46 μg mL-1,and the selectivity index (TI = TC50/EC50) was 217. LS could inhibit PRV in PK-15 cells when it was added to cell culture medium at 0,2,4,6,and 8 h after PK-15 cells had been infected by PRV. From the results,we concluded that LS had broad antimicrobial spectrum and good inhibiting effects on PRV.展开更多

Genome DNA was extracted from its conidia using Chelex-100 method. 18S rDNA sequence was amplified by PCR, which was analyzed by Blast after sequencing, and the phylogenetic tree was constructed. [Result] 18S rDNA sequence analysis showed that the pathogen of melon powdery mildew belonged to Podosphaera. Recognition and rejection of foreign eggs are effective defense of hosts against brood parasitism.However,brood parasitism can impose various selection pressures on different geographic populations of the same host sp...Recognition and rejection of foreign eggs are effective defense of hosts against brood parasitism.However,brood parasitism can impose various selection pressures on different geographic populations of the same host species.In a multiple cuckoo system in China,Azure-winged Magpies(Cyanopica cyanus)are parasitized by both Indian Cuckoos(Cuculus micropterus)and Asian Koels(Eudynamys scolopaceus).In this study,egg recognition ability and recognition mechanism of the Azure-winged Magpie were investigated using a population in Fusong,southeastern Jilin,China.The results showed that 55.6%(20/36)of the Azure-winged Magpies correctly rejected quail(Coturnix japonica)eggs in their nests,while 13.9%(5/36)of the individuals experienced rejection costs by wrongly rejecting their own eggs.Azure-winged Magpies could accurately reject the experimental eggs when the number of such eggs in the nests was the same as that of the magpie eggs.However,Azure-winged Magpies do not recognize and reject conspecific eggs(0/28).The present study indicates that the Azure-winged Magpie has moderate egg recognition ability toward non-mimetic quail eggs and shows a true recognition mechanism with rejecting foreign eggs by accurately recognizing their own eggs.However,they cannot recognize conspecific eggs.展开更多

This work first describes a simple approach for the untargeted profiling of volatile compounds for distinguishing between white duck down (WDD) and white goose down (WGD) based on resolution-optimized GC-IMS combined ...This work first describes a simple approach for the untargeted profiling of volatile compounds for distinguishing between white duck down (WDD) and white goose down (WGD) based on resolution-optimized GC-IMS combined with optimized chemometric techniques, namely PCA. The detection method for down samples was established by using GC-IMS. Meanwhile, the reason of unpleasant odors caused by WDD was explained on the basis of the characteristic volatile compounds identification. GC-IMS fingerprinting can be considered a revolutionary approach for a truly fully automatable, cost-efficient, and in particular highly sensitive method. A total of 22 compounds were successfully separated and identified through GC-IMS method, and the significant differences in volatile compounds were observed in three parts of WDD and WGD samples. The most characteristic volatile compounds of WGD belong to aldehydes, whereas carboxylic acids from WDD were detected generated by autoxidation reaction. Meanwhile, the main reason of unpleasant odor generation was possibly attributed to the high concentration of volatile carboxylic acids of WDD. Therefore, the constructed model presents a simple and efficient method of analysis and serves as a basis for down processing and quality control.展开更多
Objective:To investigate the effects of Pien Tze Huang(PZH) on the migration and invasion of HCC cells and underlying molecular mechanism.Methods:Cell counting kit-8(CCK-8) was applied to evaluate the cell viabilities...Objective:To investigate the effects of Pien Tze Huang(PZH) on the migration and invasion of HCC cells and underlying molecular mechanism.Methods:Cell counting kit-8(CCK-8) was applied to evaluate the cell viabilities of SMMC-7721,SK-Hep-1,C3A and HL-7702(6 × 10^(3)cells/well) co-incubated with different concentrations of PZH(0,0.2,0.4,0.6,0.8 mg/mL) for 24 h.Transwell,wound healing assay,CCK-8and Annexin V-FITC/PI staining were conducted to investigate the effects of PZH on the migration,invasion,proliferation and apoptosis of SK-Hep-1 and SMMC-7721 cells(650 μg/mL for SK-Hep-1 cells and 330 μg/mL for SMMC-7721 cells),respectively.In vivo,lung metastasis mouse model constructed by tail vein injection of HCC cells was used for evaluating the anti-metastasis function of PZH.SK-Hep-1 cells(10^(6)cells/200 μL per mice) were injected into B-NDG mice via tail vein.Totally 8 mice were randomly divided into PZH and control groups,4 mice in each group.After 2-d inoculation,mice in the PZH group were administered with PZH(250 mg/kg,daily) and mice in the control group received only vehicle(PBS) from the 2nd day after xenograft to day 17.Transcriptome analysis based on RNA-seq was subsequently used for deciphering anti-tumor mechanism of PZH.Quantitative real-time polymerase chain reaction(qRT-PCR) and Western blot were applied to verify RNA-seq results.Luciferase reporter assay was performed to examine the transcriptional activity of yes-associated protein(YAP).Results:PZH treatment significantly inhibited the migration,invasion,proliferation and promoted the apoptosis of HCC cells in vitro and in vivo(P<0.01).Transcriptome analysis indicated that Hippo signaling pathway was associated with anti-metastasis function of PZH.Mechanical study showed PZH significantly inhibited the expressions of platelet derived growth factor receptor beta(PDGFRB),YAP,connective tissue growth factor(CCN2),N-cadherin,vimentin and matrix metallopeptidase 2(MMP2,P<0.01).Meanwhile,the phosphorylation of YAP was also enhanced by PZH treatment in vitro and in vivo.Furthermore,PZH played roles in inhibiting the transcriptional activity of YAP.Conclusion:PZH restrained migration,invasion and epithelialmesenchymal transition of HCC cells through repressing PDGFRB/YAP/CCN2 axis.展开更多

Although certain sexrelated behavioral responses have been previously characterized in ICR mice, little is known about the effects of sex on morphine-induced behavioral responses in this outbred strain. Therefore, in this study, we investigated the sex differences of morphine-induced locomotion, anxiety-like and social behaviors in ICR mice. After morphine or saline exposure for four consecutive days(twice daily), increased locomotion, more time spent in the central area, as well as attenuated rearing and self-grooming behaviors were found in morphine-treated females in an open field; no differences were found in locomotion and the time spent in the central area between male and female controls. When interacting with the samesex individuals, female controls were engaged in more social investigation, following, body contacting and self-grooming behaviors than controls; morphine exposure reduced contacting and self-grooming behaviors in females; in contrast, these effects were not found in males. These results indicate that female ICR mice are more prosocial and are more susceptible to morphine exposure than males.展开更多

The aim of this study was to determine the metabolomic differences in postharvest table grapes(‘Red Globe’) ...Changes in the metabolites of table grapes(Vitis vinifera) reportedly occur during postharvest senescence. The aim of this study was to determine the metabolomic differences in postharvest table grapes(‘Red Globe’) after being subjected to different senescence periods. To this end, we used widely targeted metabolomics based on ultra-performance liquid chromatography and tandem mass spectrometry. A total of 135 differential metabolites were identified. During postharvest senescence, the levels of most differential flavonoids(e.g., pelargonidin 3-O-glucoside, quercetin-3-O-glucoside, and cyanidin 3-O-glucoside) and L-aspartic acid decreased, while the levels of phenolic acids(e.g., trans-4-hydroxycinnamic acid methyl ester) and pantothenol increased. During early and late senescence, the levels of most differential lipids, especially LysoPC, as well as those of nucleotides and their derivatives, such as uridine, decreased and increased, respectively. Collectively, the findings of this study provide fundamental insights into the reasonable control of table grape fruit postharvest senescence and lay a solid foundation for further research.展开更多

To determine w...B-3 exopolysaccharide is extracted from the Antarctic psychrophilic bacterium Psychrobacter sp. B-3. We have previously shown that it activates macrophages and affects their immunoregulatory activities. To determine what genes are affected during this process, we detected the genes differentially expressed in cells of RAW264.7 macrophages treated with B-3 exopolysaccharide by transcriptomic analysis. B-3 exopolysaccharide treatment caused differential expression of 420 genes, of which 178 were up-regulated and 242 were down-regulated. These genes were shown to be involved in many aspects of cell function, mainly metabolism and immunity. Genes were enriched in multiple immune-related pathways, and the most significantly enriched genes were involved in antigen processing and presentation pathways. The pathway in which differentially expressed genes were the most significantly enriched was the metabolic pathway; specifically, the expression of many metabolic enzyme genes was altered by B-3 exopolysaccharide treatment. Additionally, the genes involved in metabolisms of amino acids, carbohydrates, lipids and nucleotides, varied to certain degrees. B-3 exopolysaccharide, therefore, appears to directly affect the immune function of RAW264.7 macrophages as an immunostimulant, or to indirectly change intracellular metabolism. This is the first study to determine the effect of an Antarctic psychrophilic bacterial exopolysaccharide on RAW264.7 macrophages. However, the miRNAs of porcine SCs remain largely unknown.Methods: We isolated porcine SCs and conducted small RNA sequencing. By comparing miRNAs in germ cells, we systematically analyzed the miRNA expression pattern of porcine SCs. We screened the highly enriched SC miRNAs and predicted their functions by Gene Ontology analysis. The dual luciferase assay was used to elucidate the regulation of tumor necrosis factor receptor(TNFR)-associated factor 3(TRAF3) by ssc-miR-149.Results: The analysis showed that 18 miRNAs were highly expressed in SCs and 15 miRNAs were highly expressed in germ cells. These miRNAs were predicted to mediate SC and germ cell functions. In addition, ssc-miR-149 played critical roles in SCs by targeting TRAF3.Conclusion: Our findings provide novel insights into the miRNA expression pattern and their regulatory roles of porcine SCs.展开更多

文摘Levilactobacillus brevis FZU0713, a potential probiotic previously isolated from the traditional brewing process of Hongqu rice wine, may have the beneficial effects on improving lipid metabolism. This study aimed to evaluate the in vivo protective effects and possible mechanism of L. brevis FZU0713 on the disturbance of lipid metabolism in hyperlipidemic rats fed a high-fat diet(HFD). Results showed that oral administration of L. brevis FZU0713 could significantly inhibit obesity, ameliorate the lipid metabolism disorder, including serum/liver biochemical parameters and hepatic oxidative stress in HFD-fed rats. Histopathological result also indicated that dietary intervention of L. brevis FZU0713 could reduce the accumulation of lipid droplets in liver induced by 8 weeks HFD feeding. Furthermore, L. brevis FZU0713 intervention significantly increased the fecal levels of short-chain fatty acids(SCFAs, including acetate, propionate, butyrate, isobutyrate, valerate and isovalerate)in HFD-fed rats, which may be closely related to the changes of intestinal microbial composition and metabolic function. Intestinal microbiota profiling by 16S rRNA gene sequencing revealed that L. brevis FZU0713 intervention significantly altered the relative abundance of Coprococcus, Butyricicoccus, Intestinimonas, Lachnospiraceae FCS020 group, Ruminococcaceae_NK4A214 group, Ruminococcaceae_UCG-005 and UCG-014 at genus levels. Based on Spearman's rank correlation coefficient, serum and liver lipid metabolism related biochemical parameters were positively correlated with genera Ruminococcus, Pediococcus and Lachnospiraceae, but negatively correlated with genera Pseudoflavonifractor, Butyricicoccus and Intestinimonas. Furthermore, liver metabolomics analysis demonstrated that L. brevis FZU0713 had a significant regulatory effect on the composition of liver metabolites in hyperlipidemic rats, especially the levels of some important biomarkers involved in the metabolic pathways of arachidonic acid metabolism, primary bile acid biosynthesis, amino sugar and nucleotide sugar metabolism, taurine and hypotaurine metabolism, biosynthesis of unsaturated fatty acid, fructose and mannose metabolism, tyrosine metabolism, etc. Additionally, oral administration of L. brevis FZU0713 significantly regulated the mR NA levels of liver genes(including Acat2, Acox1, Hmgcr, Cd36, Srebp-1c and Cyp7a1)involved in lipid metabolism and bile acid homeostasis. In conclusion, our findings provide the evidence that L. brevis FZU0713 has the potential to improve disturbance of lipid metabolism by regulating intestinal microflora and liver metabonomic profile. Therefore, L. brevis FZU0713 may be used as a potential probiotic strain to produce functional food to prevent hyperlipidemia.
文摘The Qinba areas,located in the Central China,represent 317000 km2,cover 6 provinces.The major mountain of Qinba areas,the Qinling,separates China North and South.This region is one of the areas with the most biodiversity and ecological importance in China and globally,and considered as natural gene bank and bio-resource bank.Qinba mountain areas,together with the Alps in Europe and the Rocky Mountains in North America,are known as the"Three sisters of the earth"by the geological and biological circles.Most of the lands in this region are protected as national park,natural reserve,water resource protection area,etc.These areas have well preserved ecosystem and breeding and habitat area for many endangered species of plants and animals,inculding Panda.In this region,there are more than 4000 species of higher plants,about 2000 species of medicinal plants and animals,200 species of wild ornamental plants and 180 species of economic fungi with edible and medicinal value in those areas.Medicinal plant cultivation,eco-agriculture and ecotourism are the major industries in this region.The State Key Laboratory of Biological Resources and Ecological Environment of Qinba Areas,located in Hanzhong,the central city of Qinba,is focusing on pan-regional biological resources research and rational utilization,ecological protection,and comprehensive and coordinated regional development.

文摘Spatial and temporal variation of fish assemblages were investigated seasonally from May 2007 to February 2008across 11 study sites in a subtropical small stream, the Puxi Stream, of the Huangshan Mountain. Along the longitudinal gradientfrom headwater to downstream, fish species richness and abundance increased gradually, but then decreased significantly at thelower reaches. The highest species richness and abundance were observed in August and the lowest in February. Based on analysisof similarities (ANOSIM), fish assemblages were significantly different in spatial variation but not in temporal variation. Althoughdifferences were observed both among sites and among stream orders, the lower R value in order-variation suggestedstream order was not the optimal factor explaining the spatial variation of fish assemblages. In addition, dam construction did notsignificantly alter fish assemblages in the sites adjacent to and immediately downstream to dams. Using cluster analysis andnon-metric Multi Dimensional Scaling analysis (NMS), assemblages were separated into three groups at a Bray-Curtis similarityvalue of 42%: the upper, middle and lower groups. Following analysis of similarity percentages of species contributions (SIMPER),shifts in occurrence or abundance of S. curriculus, Z. platypus, R. bitterling and A.fasciatus contributed most to the differencesamongst the three groups. Standard Deviation Redundancy Analysis (RDA) suggested that habitat structure (such as elevation,substrate, and flow velocity) contributed to the spatial and temporal pattern of fish assemblages in the Puxi Stream. In conclusion,the fish assemblages in Puxi Stream presented significant spatial but not temporal variation. Human disturbance has perhapsinduced the decrease in species diversity in the lower reaches. 文摘In two experiments, the effects of succinate and NADH(reduced nicotinamide adenine dinucleotide) on metmyoglobin reductase activity and electron transport chain-linked metmyoglobin reduction were investigated and compared. In experiment 1, metmyoglobin(MetMb), substrate and inhibitors were incubated with mitochondria. Comparsion of the effects of succinate and NADH on MetMb reduction was investigated. The MetMb percentage in sample treated with 8 mol L-1 succinate decreased by about 69% after 3 h incubation, and the effect was inhibited by the addition of 10 mol L-1 electron transfer chain complex II inhibitor malonic acid; the MetMb percentage in samples treated with 2 mol L-1 NADH decreased by 56% and the effect was inhibited by the addition of 0.02 mol L-1 electron transport chain complex I inhibitor rotenone. These results indicated that electron transport chain played an important role in MetMb reduction. Both complex II and complex I take part in the MetMb reduction in mitochondria through different pathways. NADH-MetMb reduction system was less stable than succinateMetMb system. In experiment 2, the beef longissimus dorsi muscle was blended with different concentrations of succinate or NADH. Enhancing patties with higher concentration of succinate or NADH improved colour stability in vacuum packaged samples(P<0.05). These results verified that mitochondria electron transport chain is related to the MetMb reduction in meat system.

文摘Background:MiRNAs act as pivotal post-transcriptional gene mediators in the regulation of diverse biological processes,including proliferation,development and apoptosis.Our previous study has showed that miR-101-3p is differentially expressed in dairy goat ovaries compared single with multiple litters.The objective of this research was to explore the potential function and molecular mechanism of miR-101-3p via its target STC1 in goat ovarian growth and development.Results:cDNA libraries were constructed using goat granulosa cells transfected with miR-101-3p mimics and negative control by RNA-sequencing.In total,142 differentially expressed unigenes(DEGs)were detected between two libraries,including 78 down-regulated and 64 up-regulated genes.GO and KEGG enrichment analysis showed the potential impacts of DEGs on ovarian development.STC1 was singled out from DEGs for further research owing to it regulates reproductive-related processes.In vitro,bioinformatics analysis and 3′-UTR assays confirmed that STC1 was a target of miR-101-3p.ELISA was performed to detect the estrogen(E2)and progesterone(P4)levels.CCK8,EdU and flow cytometry assays were performed to detect the proliferation and apoptosis of granulosa cells.Results showed that miR-101-3p regulated STAR,CYP19A1,CYP11A1 and 3β-HSD steroid hormone synthesis-associated genes by STC1 depletion,thus promoted E2 and P4 secretions.MiR-101-3p also affected the key protein PI3K,PTEN,AKT and mTOR in PI3K-AKT pathway by STC1,thereby suppressing proliferation and promoting apoptosis of granulosa cells.In vivo,the distribution and expression levels of miR-101-3p in mouse ovaries were determined through fluorescence in situ hybridisation(FISH).Immunohistochemistry results showed that STC1 expression was suppressed in mouse ovaries in miR-101-3p-agonist and siRNA-STC1 groups.Small and stunted ovarian fragments,decreased numbers of follicles at diverse stages were observed using Hematoxylin-eosin(HE)staining,thereby showing unusual ovarian development after miR-101-3p overexpression or STC1 depletion.Inhibition of miR-101-3p manifested opposite results.Conclusions:Taken together,our results demonstrated a regulatory mechanism of miR-101-3p via STC1 in goat granulosa cells,and offered the first in vivo example of miR-101-3p and STC1 functions required for ovarian development.

文摘Grapevine(Vitis spp.)is one of the most economically important fruit crops worldwide,and there is considerable interest in improving its major agronomic and enological traits in response to ever-changing agricultural environments and consumer demands.Molecular genetic techniques in particular,associated with rapid technological advancements,provide an attractive alternative to conventional breeding approaches for developing new grapevine varieties with enhanced yield performance,quality,stress tolerance and disease resistance.To date,several grapevine varieties have been transformed with genes associated with diverse functions through biolistic bombardment and/or Agrobacterium-mediated transformation,and transgenic grape lines have been obtained using established regeneration systems.Nevertheless,a wide range of factors,including genotype,explant source and culture medium,have been shown to affect the efficiency of plant regeneration.Moreover,the selection and use of acceptor materials,bacterial strain and cell density,selectable markers and selection methods also influence transformation efficiency.This paper provides an overview of recent advances in grapevine regeneration and genetic transformation and in-depth discussion of the major limiting factors,and discusses promising future strategies to develop robust plant regeneration and genetic transformation in grapevine.

文摘In order to study NAC transcription factor gene in maize(Zea mays), a completed c NAC encoding the NAC-like gene homologue named as Zm NAC was cloned from the maize inbred line Zheng 58 using a pair of specific primers, which were designed to cover the full coding region according to the reported NAC gene family. The length of the sequences was 953 bp.The deduced amino acid sequences analysis showed that the sequence contained the complete coding region of a typical NAC gene, including a complete open reading frame which was 939 bp. The study analyzed the expression patterns of NAC transcript gene induced in various PEG(polyethylene glycol) simulated drought condition. Real-time PCR indicated that maize NAC gene can be induced in various PEG to abiotic stress responses, it could express constitutively to adopt the drought condition in short time, and the higher drought, the more early expression, which indicated that NAC transcript gene could be involved in the regulation of the defense response.
文摘In this study,the in vitro antimicrobial and antiviral activities of the lysozyme from marine strain S-12-86 (LS) were investigated. The antimicrobial activity of LS was tested by minimum inhibition concentration (MIC) and minimum bactericidal concentration (MBC) method. The inhibiting effects of LS on pseudo rabies virus (PRV) in swine kidney cells (PK-15 cells) were judged by cytopathogenic effect test (CPE). The results showed LS had a broad antimicrobial spectrum against several standard strains including gram-positive bacteria,gram-negative bacteria,fungi,etc. The MIC of LS was 0.25-4.00 mg mL-1 and its MBC was 0.25-8.00 mg mL-1,respectively. Observation under the transmission electron microscope revealed that the cell wall of Candida albicans was distorted seriously,and the cytoplasm with many cavities was asymmetrical after being hydrolyzed by LS. The median cytotoxicity concentration (TC50) of LS was 100.0 μg mL-1,the median effective concentration (EC50) was 0.46 μg mL-1,and the selectivity index (TI = TC50/EC50) was 217. LS could inhibit PRV in PK-15 cells when it was added to cell culture medium at 0,2,4,6,and 8 h after PK-15 cells had been infected by PRV. From the results,we concluded that LS had broad antimicrobial spectrum and good inhibiting effects on PRV.

文摘[Objective]The aim was to isolate and identify an endophytic bacteria strain with antimicrobial activity from Davidia involucrate Brail.[Method]Endophytic strain with antibiosis ability was isolated from D.involucrate Brail by using cylinder-plate method.Then,it was identified through physiological and biochemical tests,16S rDNA homology analysis as well as some gene-specific sequence analysis.[Result]B221 stain had antimicrobial activity against a variety of rice plant pathogens,and it was identified as Bacillus subtilis.[Conclusion]This study enriches the research on endophyte within D.involucrate Brail,application of Bacillus bio-control,and therefore has laid a good foundation for the development of fungus used in biological control of crop pathogens.

文摘Stipa breviflora seeds,collected from Inner Mongolia,were sowed on MS medium containing 0,50,100,150,200 mmol/L NaC l,to investigate the effects of salt stress on the physiological phenotype and antioxidant enzyme activity of S.breviflora seedlings.The results showed the germination rate,root length and fresh weight of seedlings were all influenced by salinity.Germination rate,root length and fresh weight of seedlings were significantly inhibited by 100,150 and50 mmol/L NaC l,and were 62%,1.30 cm,0.299 g,respectively.With NaC l content increasing,the trend of superoxide dismutase( SOD) and peroxidase( POD) activity was rising at first and then falling,and the activity of SOD and POD reached the maximum at 150 and 100 mmol/L,48.6 and 6 766 U/g,respectively.The activity of catalase( CAT) continued to increase and reached the maximum at 150 mmol/L,299 U/g.The isozyme zymogram showed that SOD 2-4,POD 1-2,and CAT 1-2 were positively correlated with salinity tolerance of S.breviflora seedlings.

文摘[Objective] The paper was to measure and analyze pathogen of 18S rDNA sequence of the pathogen of melon powdery mildew from lands overlaid with sands. [Method]The melon powdery mildew was isolated from infected plants of "Yujinxiang", a major melon variety cultivated in lands overlaid with sands in the middle arid area of Ningxia. Genome DNA was extracted from its conidia using Chelex-100 method. 18S rDNA sequence was amplified by PCR, which was analyzed by Blast after sequencing, and the phylogenetic tree was constructed. [Result] 18S rDNA sequence analysis showed that the pathogen of melon powdery mildew belonged to Podosphaera. [Conclusion] The study provided reference for biocontrol and disease-resistance breeding against melon powdery mildew.

文摘Recognition and rejection of foreign eggs are effective defense of hosts against brood parasitism.However,brood parasitism can impose various selection pressures on different geographic populations of the same host species.In a multiple cuckoo system in China,Azure-winged Magpies(Cyanopica cyanus)are parasitized by both Indian Cuckoos(Cuculus micropterus)and Asian Koels(Eudynamys scolopaceus).In this study,egg recognition ability and recognition mechanism of the Azure-winged Magpie were investigated using a population in Fusong,southeastern Jilin,China.The results showed that 55.6%(20/36)of the Azure-winged Magpies correctly rejected quail(Coturnix japonica)eggs in their nests,while 13.9%(5/36)of the individuals experienced rejection costs by wrongly rejecting their own eggs.Azure-winged Magpies could accurately reject the experimental eggs when the number of such eggs in the nests was the same as that of the magpie eggs.However,Azure-winged Magpies do not recognize and reject conspecific eggs(0/28).The present study indicates that the Azure-winged Magpie has moderate egg recognition ability toward non-mimetic quail eggs and shows a true recognition mechanism with rejecting foreign eggs by accurately recognizing their own eggs.However,they cannot recognize conspecific eggs.

文摘Strategies aimed at defining,discovering,and developing alternatives to traditional antibiotics will underlie the development of sustainable agricultural systems.Among such strategies,antimicrobial peptides(AMPs)with broad-spectrum antimicrobial activity and multifaceted mechanisms of action are recognized as ideal alternatives in the post-antibiotic era.In particular,AMPs derived from microbes with active metabolisms that can adapt to a variety of extreme environments have long been sought after.Consequently,this review summarizes information on naturally occurring AMPs,including their biological activity,antimicrobial mechanisms,and the preparation of microbial-derived AMPs;it also outlines their applications and the challenges presented by their use in the agroindustry.By dissecting the research results on microbial-derived AMPs of previous generations,this study contributes valuable knowledge on the exploration and realization of the applications of AMPs in sustainable agriculture.
文摘This work first describes a simple approach for the untargeted profiling of volatile compounds for distinguishing between white duck down (WDD) and white goose down (WGD) based on resolution-optimized GC-IMS combined with optimized chemometric techniques, namely PCA. The detection method for down samples was established by using GC-IMS. Meanwhile, the reason of unpleasant odors caused by WDD was explained on the basis of the characteristic volatile compounds identification. GC-IMS fingerprinting can be considered a revolutionary approach for a truly fully automatable, cost-efficient, and in particular highly sensitive method. A total of 22 compounds were successfully separated and identified through GC-IMS method, and the significant differences in volatile compounds were observed in three parts of WDD and WGD samples. The most characteristic volatile compounds of WGD belong to aldehydes, whereas carboxylic acids from WDD were detected generated by autoxidation reaction. Meanwhile, the main reason of unpleasant odor generation was possibly attributed to the high concentration of volatile carboxylic acids of WDD. Therefore, the constructed model presents a simple and efficient method of analysis and serves as a basis for down processing and quality control.

文摘Objective:To investigate the effects of Pien Tze Huang(PZH) on the migration and invasion of HCC cells and underlying molecular mechanism.Methods:Cell counting kit-8(CCK-8) was applied to evaluate the cell viabilities of SMMC-7721,SK-Hep-1,C3A and HL-7702(6 × 10^(3)cells/well) co-incubated with different concentrations of PZH(0,0.2,0.4,0.6,0.8 mg/mL) for 24 h.Transwell,wound healing assay,CCK-8and Annexin V-FITC/PI staining were conducted to investigate the effects of PZH on the migration,invasion,proliferation and apoptosis of SK-Hep-1 and SMMC-7721 cells(650 μg/mL for SK-Hep-1 cells and 330 μg/mL for SMMC-7721 cells),respectively.In vivo,lung metastasis mouse model constructed by tail vein injection of HCC cells was used for evaluating the anti-metastasis function of PZH.SK-Hep-1 cells(10^(6)cells/200 μL per mice) were injected into B-NDG mice via tail vein.Totally 8 mice were randomly divided into PZH and control groups,4 mice in each group.After 2-d inoculation,mice in the PZH group were administered with PZH(250 mg/kg,daily) and mice in the control group received only vehicle(PBS) from the 2nd day after xenograft to day 17.Transcriptome analysis based on RNA-seq was subsequently used for deciphering anti-tumor mechanism of PZH.Quantitative real-time polymerase chain reaction(qRT-PCR) and Western blot were applied to verify RNA-seq results.Luciferase reporter assay was performed to examine the transcriptional activity of yes-associated protein(YAP).Results:PZH treatment significantly inhibited the migration,invasion,proliferation and promoted the apoptosis of HCC cells in vitro and in vivo(P<0.01).Transcriptome analysis indicated that Hippo signaling pathway was associated with anti-metastasis function of PZH.Mechanical study showed PZH significantly inhibited the expressions of platelet derived growth factor receptor beta(PDGFRB),YAP,connective tissue growth factor(CCN2),N-cadherin,vimentin and matrix metallopeptidase 2(MMP2,P<0.01).Meanwhile,the phosphorylation of YAP was also enhanced by PZH treatment in vitro and in vivo.Furthermore,PZH played roles in inhibiting the transcriptional activity of YAP.Conclusion:PZH restrained migration,invasion and epithelialmesenchymal transition of HCC cells through repressing PDGFRB/YAP/CCN2 axis.

文摘Gender and genetic strain are two prominent variants that influence drug abuse. Although certain sexrelated behavioral responses have been previously characterized in ICR mice, little is known about the effects of sex on morphine-induced behavioral responses in this outbred strain. Therefore, in this study, we investigated the sex differences of morphine-induced locomotion, anxiety-like and social behaviors in ICR mice. After morphine or saline exposure for four consecutive days(twice daily), increased locomotion, more time spent in the central area, as well as attenuated rearing and self-grooming behaviors were found in morphine-treated females in an open field; no differences were found in locomotion and the time spent in the central area between male and female controls. When interacting with the samesex individuals, female controls were engaged in more social investigation, following, body contacting and self-grooming behaviors than controls; morphine exposure reduced contacting and self-grooming behaviors in females; in contrast, these effects were not found in males. These results indicate that female ICR mice are more prosocial and are more susceptible to morphine exposure than males.

文摘Changes in the metabolites of table grapes(Vitis vinifera) reportedly occur during postharvest senescence. The aim of this study was to determine the metabolomic differences in postharvest table grapes(‘Red Globe’) after being subjected to different senescence periods. To this end, we used widely targeted metabolomics based on ultra-performance liquid chromatography and tandem mass spectrometry. A total of 135 differential metabolites were identified. During postharvest senescence, the levels of most differential flavonoids(e.g., pelargonidin 3-O-glucoside, quercetin-3-O-glucoside, and cyanidin 3-O-glucoside) and L-aspartic acid decreased, while the levels of phenolic acids(e.g., trans-4-hydroxycinnamic acid methyl ester) and pantothenol increased. During early and late senescence, the levels of most differential lipids, especially LysoPC, as well as those of nucleotides and their derivatives, such as uridine, decreased and increased, respectively. Collectively, the findings of this study provide fundamental insights into the reasonable control of table grape fruit postharvest senescence and lay a solid foundation for further research.

文摘B-3 exopolysaccharide is extracted from the Antarctic psychrophilic bacterium Psychrobacter sp. B-3. We have previously shown that it activates macrophages and affects their immunoregulatory activities. To determine what genes are affected during this process, we detected the genes differentially expressed in cells of RAW264.7 macrophages treated with B-3 exopolysaccharide by transcriptomic analysis. B-3 exopolysaccharide treatment caused differential expression of 420 genes, of which 178 were up-regulated and 242 were down-regulated. These genes were shown to be involved in many aspects of cell function, mainly metabolism and immunity. Genes were enriched in multiple immune-related pathways, and the most significantly enriched genes were involved in antigen processing and presentation pathways. The pathway in which differentially expressed genes were the most significantly enriched was the metabolic pathway; specifically, the expression of many metabolic enzyme genes was altered by B-3 exopolysaccharide treatment. Additionally, the genes involved in metabolisms of amino acids, carbohydrates, lipids and nucleotides, varied to certain degrees. B-3 exopolysaccharide, therefore, appears to directly affect the immune function of RAW264.7 macrophages as an immunostimulant, or to indirectly change intracellular metabolism. This is the first study to determine the effect of an Antarctic psychrophilic bacterial exopolysaccharide on RAW264.7 macrophages. Our findings provide an important reference for research into the regulation of macrophage immune function by different polysaccharides.

文摘Background: Sertoli cells(SCs) create a specialized environment to support and dictate spermatogenesis.MicroRNAs(miRNAs), a kind of ~ 22 nt small noncoding RNAs, have been reported to be highly abundant in mouse SCs and play critical roles in spermatogenesis. However, the miRNAs of porcine SCs remain largely unknown.Methods: We isolated porcine SCs and conducted small RNA sequencing. By comparing miRNAs in germ cells, we systematically analyzed the miRNA expression pattern of porcine SCs. We screened the highly enriched SC miRNAs and predicted their functions by Gene Ontology analysis. The dual luciferase assay was used to elucidate the regulation of tumor necrosis factor receptor(TNFR)-associated factor 3(TRAF3) by ssc-miR-149.Results: The analysis showed that 18 miRNAs were highly expressed in SCs and 15 miRNAs were highly expressed in germ cells. These miRNAs were predicted to mediate SC and germ cell functions. In addition, ssc-miR-149 played critical roles in SCs by targeting TRAF3.Conclusion: Our findings provide novel insights into the miRNA expression pattern and their regulatory roles of porcine SCs.
