Hyperuricemia is a metabolic disorder caused by abnormal purine metabolism,resulting in abnormally high serum uric acid.In this study,a novel Levilactobacillus brevis PDD-5 isolated from salty vegetables was verified with the function of alleviating hyperuricemia.The relevant effects of L.brevis PDD-5 in lowering uric acid were analyzed by in vitro and in vivo experiments.The results showed that the L.brevis PDD-5 has(68.86±15.46)%of inosine uptake capacity and(95.75±3.30)%of guanosine uptake capacity in vitro.Oral administration of L.brevis PDD-5 to hyperuricemia rats reduced uric acid,creatinine,and urea nitrogen in serum,as well as decreased inosine and guanosine levels in the intestinal contents of rats.Analysis of relevant markers in the kidney by ELISA kits revealed that L.brevis PDD-5 alleviated oxidative stress and inflammation.Moreover,the gene expression of uric acid transporter 1(URAT1)and glucose transporter 9(GLUT9)was down-regulated,and the gene expression of organic anion transporter 1(OAT1)was up-regulated after treatment with L.brevis PDD-5.Western blot analysis showed that L.brevis PDD-5 alleviated hyperuricemia-induced kidney injury through the NLRP3 pathway.The se findings suggest that L.brevis PDD-5 can lower uric acid,repair kidney damage,and also has the potential to prevent uric acid nephropathy. 展开更多

Iron deficiency anemia(IDA)is a major global health problem.Tegillarca granosa has been considered as an excellent source of iron given its high content of iron-binding protein,ferritin.The aim of the present study was to determine the physicochemical properties,protein structures,and iron uptake of ferritin extracted from T.granosa,and to evaluate the potential impacts of chitosan glycosylation on these characteristics.Based on Box-Behnken design and response surface methodology,the optimal conditions for glycosylation included a ferritin/chitosan mass ratio of 4:1,a pH of 5.5,a reaction time of 10 min,and a reaction temperature of 50℃.Glycosylation caused decreased surface hydrophobicity and elevated water-holding capacity of ferritin due to the introduction of hydrophilic groups.Additionally,glycosylation improved antioxidant capacity of ferritin by 20.69%–189.66%,likely owing to the protons donated by saccharide moiety to terminate free radical chain reaction.The in vitro digestibility of ferritin was elevated by 22.56%–104.85%after glycosylation,which could be associated with lessβ-sheet content in secondary structure that made the glycosylated protein less resistant to enzymatic digestion.The results of the iron bioavailability in Caco-2 cells revealed that ferritin(78.85–231.77 ngmg^(−1))exhibited better iron bioavailability than FeSO4(51.48–114.37 ngmg^(−1))and the values were further elevated by glycosylation with chitosan(296.23–358.20 ngmg^(−1)),which may be related to the physicochemical properties of ferritin via glycosylation modification.These results provide a basis for the development of T.granosa derived ferritin and its glycosylated products,and can promote the utilization of aquatic resources. 展开更多

As a novel food quality monitoring technology,intelligent freshness indicator has received wide attention in recent years.However,its poor safety and stability are the main problems hindering its practical application.Hence a new pH-sensing indicator based on bromocresol green(BCG)was developed in this study for nondestructive and real-time monitoring the freshness of marine fishes.The indicator was designed with a three-layer structure,using the polytetrafluoroethylene(PTFE)membrane with high hydrophobicity and air permeability as the inner layer to isolate the moisture in the package,BCG-coated filter paper as the colorchanging layer to indicate the freshness of fish,and a transparent unidirectional permeable(TUP)membrane with moisture resistance as the out layer to isolate the moisture in the environment.This contributed to weaken the influence of humidity and prevent dye migration,so as to improve the accuracy and safety of the indicator.Therefore,a highly sensitive and distinguished color variation response to trimethylamine(TMA)standard solution with different concentrations was observed on the indicator.Additionally,the indicator showed a high color stability at different storage temperatures up to 14 days with total color differences(ΔE)less than 5.0.The indicator presented visible color variations from yellow to green then eventually to blue when applied to monitor the freshness of sea bass and salmon stored at 4℃,implying that fish was spoiled.Meanwhile,indicatorsΔE value was significantly positively correlated with total volatile basic nitrogen(TVB-N)and total viable count(TVC)in sea bass and salmon samples.Thus,the pH-sensing indicator can be applied as a cost-effective and promising intelligent indicator for monitoring fish freshness. 展开更多

Heavy metals,polycyclic aromatic hydrocarbons(PAHs),and perfluorinated alkyl substances(PFASs)polluted in 155 samples of two marine crustaceans(Oratosquilla oratoria and Portunus trituberculatus)harvested in the Zhoushan fishing ground in the East China Sea were assessed.The results showed that Pb,Hg,and iAs in the whole edible parts of O.oratoria and P.trituberculatus were at trace levels,far below the limits set by the China’s national standard,whereas the exceeding standard rates of Cd in the whole edible parts of O.oratoria and P.trituberculatus were 98.6%and 75.6%,respectively.Moreover,the average Cd levels and the exceeding standard rates of Cd in the visceral tissues of O.oratoria were significantly higher than those in the muscle tissues(4.64 mg kg^(−1)vs.0.49 mg kg^(−1)and 96.7%vs 30%,respectively).PAHs with high detection frequency in the two marine crustaceans included NA,AC,FL,and PHE.Total contents of 15 PAHs(abbreviated asΣ15 PAHs)in whole edible parts of O.oratoria and P.trituberculatus were 10.75–65.9μg kg^(−1)(mean=18.7μg kg^(−1)and 13.26–181μg kg^(−1)(mean=24.2μg kg^(−1),and those in the muscle and the visceral tissues of P.trituberculatus were 9.58–160μg kg^(−1)(mean=21.1μg kg^(−1)and 18.22–201μg kg^(−1)(mean=32.7μg kg^(−1),respectively.Furthermore,PFASs that were found at high contents included PFOA,PFOS,and PFOSA,and total contents of 23 PFASs(Σ23 PFASs)ranged 0.0162–5.104μg kg^(−1)(mean=0.915μg kg^(−1).Collectively,the present work provides new data on the contamination in two marine crustaceans captured in the Zhoushan fishing ground in the China East Sea,which can be useful for making suggestions on proper consumption amounts of the two marine crustaceans. 展开更多

The freshwater microalga Haematococcus pluvialis accumulates large amounts of fatty acids in response to adverse conditions.However,the key fatty acid desaturase genes in H.pluvialis remain unknown.In this study,we cloned and functionally characterized aΔ12 fatty acid desaturase gene,and designated it as HpFAD2.The open reading frame of HpFAD2 consisted of 1137 base pairs and encoded 378 amino acids.The deduced polypeptide showed 70%identity to other endoplasmic reticulumΔ12 fatty acid desaturases,whereas it had only 44%identity to plastidΔ12 fatty acid desaturases.The PSORT algorithm and phylogenetic analysis further confirmed its affiliation to the endoplasmic reticulumΔ12 fatty acid desaturases.Heterologous expression was performed in Saccharomyces cerevisiae cells transformed with the recombinant plasmid pYES2-HpFAD2.Two additional fatty acids(C16:2 and C18:2)were detected in the yeast transformants.The results indicatedΔ12 desaturation activity and substrate preference for C18:1 over C16:1.The transcriptional levels of H.pluvialis HpFAD2 at different growth stages were measured by quantitative polymerase chain reaction(PCR),indicating that the HpFAD2 transcriptional levels were significantly higher in red cells than those in green cells.Our study brings more insight into the fatty acid biosynthetic pathway of H.pluvialis. 展开更多

Ethylene regulates many aspects of plant growth and development.It is perceived by a family of ethylene receptors(ETRs)that have been well described.However,a full understanding of ETR function is complicated by functional redundancy between the receptor isoforms.Here,we characterize a new ETR,SlETR7,that was revealed by tomato genome sequencing.SlETR7 expression in tomato fruit pericarp increases when the fruit ripens and its expression is synchronized with the expression of SlETR1,SlETR2,and SlETR5 which occurs later in the ripening phase than the increase observed for SlETR3,SlETR4,and SlETR6.We uncovered an error in the SlETR7 sequence as documented in the ITAG 3 versions of the tomato genome which has now been corrected in ITAG 4,and we showed that it belongs to sub-family II.We also showed that SlETR7 specifically binds ethylene.Overexpression(OE)of SlETR7 resulted in earlier flowering,shorter plants,and smaller fruit than wild type.Knock-out(KO)mutants of SlETR7 produced more ethylene at breaker(Br)and Br+2 days stages compared to wild type(WT),but there were no other obvious changes in the plant and fruit in these mutant lines.We observed that expression of the other SlETRs is upregulated in fruit of SlETR7 KO mutants,which may explain the absence of obvious ripening phenotypes.Globally,these results show that SlETR7 is a functional ethylene receptor.More work is needed to better understand its specific roles related to the six other tomato ETRs. 展开更多

Therefore, taking intestinal probiotics as the starting point, this article combs the relationship between probiotics and lipid metabolism. Moreover, we discuss the underlying mechanisms of intestinal probiotics regulating lipid metabolism and summarize the therapeutic strategies for abnormal lipids metabolism. This article provides a reference for the further utilization of probiotics in the field of functional foods(food industry). Traditionally,silkworm silk has been used to make high-quality textiles.Nevertheless,various wastes from silk-worm silk textiles that are no longer used are increasing.which is also causing considerable waste and contam-ination.This issue is causing widespread concern in countries that use more silk.Regenerated silk fibroin(RSF)fibers have been shown to be fragile and tender,which prohibits RSF from being widely used as a structural com-ponent.Therefore,enriching the function of silk and enhancing the RSF mechanial properties are important directions to expand the comprehensive utilization of silk products.In the present research,wet spinning was used to create a series of RSF/tungsten disulfide(WS_(2))nanoparticles(NPs)hybrid fiber having distinct WS_(2) nanoparticles concentrations.It was discovered that the temperature of hybrid fibers containing 0.8 wt%RSF/WS_(2) nanoparticles might climb from 20.4℃ to 85.6℃in 1 min and 108.3℃ in 10 min after being exposed to simulated sunlight for a period of one minute and ten minutes.It also had certain antibacterial activity and thermal stability.Fabrics created by hand mixing had outst anding photothermal characteristics under natural sunlight.Further-more,adding WS_(2) nanoparticles might increase the tensile properties of hybrid fibers,which could be caused by the reality that the blending of WS_(2) nanoparticles inhibited the self-assembly of sheets in RSF reaction mixture in a dosage dependent way,as evidenced by the fact that RSF/WSz nanoparticles hybrid fibers had lesser β-sheets material,crystalline nature,and arystalline size.The above performance makes the RSF/WS_(2) nanoparticles hybrid fbers promising candidates for application in photothermal fabrics as well as military dothing. 展开更多

Sucrose metabolism,particularly the decomposition of sucrose by invertase,plays a central role in plant responses to cold stress.Invertase inhibitors(INHs)evolved in higher plants as essential regulators of sucrose metabolism.By limiting invertase activity,INHs keep cellular sugar levels elevated,which provides enhanced protection to plants under stress.Our results showed that the expression of PpVIN2,the only vacuolar invertase(VIN)gene in peach fruit sensitive to chilling temperatures,increases significantly during cold storage,while VIN enzyme activity increases more modestly.We also found that peach fruit transiently overexpressing PpINH1 had decreased VIN activity.Interactions of PpINH1 and PpVIN2 with recombinant proteins were shown by yeast two-hybrid assays and bimolecular fluorescence complementation assays,as well as in vitro.During cold storage,trehalose-treated peach fruit had significantly increased PpINH1 expression,decreased VIN enzyme activity,and significantly higher sucrose content than did untreated fruit.As a result,the treated fruit had enhanced resistance to chilling injury.Collectively,our data show that the post-translational repression of VIN enzyme activity by PpINH1 helps maintain sucrose levels in peach fruit during cold storage,thereby improving resistance to chilling injury. 展开更多

Green alga Enteromorpha clathrata(E.clathrata)contains a variety of bioactive compounds,including polysaccharides,polyphenols and fat-soluble pigments etc.,among which polyphenols exhibit a wide range of medicinal properties.E.clathrata polyphenols(ECPs)have shown various biological activities such as antioxidant,anti-inflammatory and antidiabetic effects;however,the potential of ECPs as an anti-cancer reagent remains unclear.The aim of this study was to investigate the anti-tumor activity and underlying mechanisms of ECPs on hepatocellular carcinoma.The cytotoxicity of Hepa1-6 cells was determined by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)and lactate dehydrogenase(LDH)assay.Flow cytometry and fluorescence microscope analysis of cell apoptosis after annexin V-fluorescein isothiocyanate(FITC)/propidium iodide(PI)staining.2',7'-dichlorodihydrofluorescein diacetate(DCFH-DA)assay was used for intracellular reactive oxygen species(ROS)detection.caspase-9 activity was determined using cspase-9 colorimetric assay.Mitochondrial transmembrane potential(Δψm)was measured using JC-1.Western blot and quantitative real-time PCR(qPCR)were used to assess the expressions of the apoptosis regulators Bax,Bcl-2,cytochrome c and caspase-3.It was found that ECPs showed a dose-dependent cytotoxicity against Hepa1-6 cells by inducing apoptosis.The apoptosis in ECPs-treated Hepa1-6 cells was accompanied by the loss of mitochondrial membrane potential,elevated ROS generation,increased release of mitochondrial cytochrome c,and up-regulation of caspase-9 and caspase-3.The expressions of Bax(pro-apoptotic molecule)and Bcl-2(apoptosis suppressor)were up-regulated and down-regulated,respectively,at both mRNA and protein levels.These molecular alterations revealed that ECPs caused apoptosis of cells through the mitochondrial pathway,suggesting that ECPs are potential candidates to be developed for liver cancer treatment. 展开更多

The quality changes of shelled Pacific oysters(Crassostrea gigas)were examined in relation to the effects of superchilling storage at-1 C for 28 d by measuring changes in biochemical properties(microbial analysis,adenosine triphosphate(ATP)-related compounds,pH,free amino acids)and sensory evaluations in this study.The results indicated that microorganism growth was significantly inhibited during superchilling storage.Adenosine diphosphate(ADP)and adenosine monophosphate(AMP)accumulated while ATP rapidly decreased in the adductor muscle.ATP and ADP were the primary components in the other 3 tissues including mantle,gill,and body trunk of oysters,and they remained relatively stable over time.The pH and adenylate energy charge in the adductor muscle could be utilized as freshness indicators for shelled oysters.However,there were no significant differences(P>0.05)among the free amino acids during whole storage.According to the sensory evalu-ations,oysters could be alive and tolerated up to 21 d at-1℃storage.The study demonstrated that superchilling storage at-1℃could better maintain the eating quality of shelled oysters and the shelf life was extended to 21 d. 展开更多

To clarify the effect of aluminum stress on the quality of Enteromorpha prolifera(E.prolifera)and to explore the mechanism of the combination of aluminum and E.prolifera,we analyzed changes in the nutrients,micromorphology,element distribution,and spectrum of E.prolifera treated with different concentrations of aluminum(0,0.2,2.0,and 20.0μmol· L^(-1))using scanning electron microscopy-energy dispersive spectroscopy(SEM-EDX)and Fourier-transform infrared spectroscopy(FT-IR).The biomass,protein,dietary fiber,and ash contents of E.prolifera initially increased and then subsequently decreased with an increasing concentration of aluminum.Meanwhile,the total amount of amino acids decreased.Scanning the surface of E.prolifera by SEM-EDX revealed that a high concentration of aluminum damaged the cells of E.prolifera.Additionally,the content of aluminum on the surface of E.prolifera cells increased and the absorption of other elements was also affected.The FT-IR analysis showed that aluminum might combine with the functional groups at the 3408 cm^(-)1,2928 cm1,and 1072 cm^(-)1 peaks in E.prolifera and alter the characteristic of the different absorption peaks. 展开更多

Herein,the 2,4-dinitrophenyl functional group acting as the thiophenol reactive site was introduced into a carbazole-chalcone fluorophore to synthesize probe-CCF2,which could result in a remarkable increase in fluorescence when reacting with thiophenols.The selectivity and accuracy of probe-CCF2 were investigated with thiophenols,hydrosulphide salt,aliphatic thiols,glutathione,cysteine,anions and metal cations.Probe-CCF2 exhibited a detection limit of 37 nmol/L(R^(2)=0.9951),a remarkable Stokes shift of approximately 130 nm,and a brief response time of 9 min with a remarkable increase in fluorescence of 90-fold.Probe-CCF2 was applied for thiophenol detection in water samples and imaging in living cells successfully,with high sensitivity and excellent selectivity. 展开更多

Scallop adductor muscle has great popularity for raw consumption in Japan,but conventionally shucking live scallops with high vitality by knife requires skill and is time-consuming.Flash-boil shucking is a convenient shucking method in sushi restaurants for live scallops.However,the effects of this treatment on the quality of its main edible part,the adductor muscle,are not clear.This study was conducted to investigate the impact of flash-boil shucking on the quality of raw-edible scallop adductor muscle,focusing on flavor-enhancing components and ultrastructure.Thermal treatment could contribute to the formation of flavor-enhancing nucleotides in the external layer of the adductor muscle.Adenosine monophosphate content increased with the boiling time,which significantly increased to 1.47±0.42μmol/g in the 30 s-boiled group compared with 0.38±0.13μmol/g in the control group(p<0.05).The rapid adenosine monophosphate accumulation after boiling resulted in a faster accumulation of inosine and hypoxanthine during subsequent refrigeration,which could bring bitterness to aquatic products.Flash-boil shucking had no discernible effect on the proportion of free amino acids relevant to the flavor of scallop adductor muscle,despite the possibility of a reduction in total free amino acids.The external layer of the scallop adductor muscle could be hardened after flash-boil shucking.At the ultrastructural level,muscle fibers tend to aggregate transversely and contract longitudinally depending on the heating time.Flash-boil shucking is ideal for consuming the raw-edible scallop adductor muscle promptly.Flash-boil shucking treatment is recommended for households,restaurants,and small factories for treating scallops with high vitality. 展开更多

Microbial communities in marine habitats are regarded as underexplored reservoirs for discovering new natural products with potential application.However,only a few microbes in nature can be cultivated in the laboratory.This has led to the development of a variety of isolation and cultivation methods,and in situ cultivation is one of the most popular.Diverse in situ cultivation methods,with the same basic principle,have been applied to a variety of environmental samples.Compared with conventional approaches,these new methods are able to cultivate previously uncultured and phylogenetically novel microbes,many with biotechnological potential.This review introduces the various in situ cultivation methods for the isolation of previously uncultured microbial species and their potential for marine microbial resource mining.Furthermore,studies that investigated the key and previously unidentified mechanisms of growing uncultivated microorganisms by in situ cultivation,which will shed light on the understanding of microbial uncultivability,were also reviewed. 展开更多
