The physical function of even a person getting on a wheelchair could be tested in all subjects. Blood was collected by venipuncture and the serum 1, 25-hydroxy vitamin D (1, 25OHD) concentration was measured. The Mini-Mental State Examination (MMSE) and the Montreal Cognitive Assessment-Japanese version (MoCA-J) was used for the cognitive function test. Physical function was measured objectively using the Timed UP and Go (TUG) and 4-m walking test (4MWS). A significant positive correlation was found between serum 1, 25OHD and MMSE or MoCA-J. It is expected that an elderly person can maintain a mean serum 1, 25OHD level of about 100 pg/mL for preventing early cognitive disorder. In the present study, a significant positive correlation was found between urinary 25-hydroxy vitamin D (25OHD)/creatinine and MMSE or MoCA-J. Our results showed that urinary 25OHD might be a useful biomarker for predicting cognitive disorder. There was a significant negative correlation between serum 1, 25OHD and TUG or 4MWS. These findings suggest that serum 1, 25OHD levels might serve as a useful index to improve cognitive and physical functional impairment.展开更多

Male Sprague-Dawley rats were randomly separated into 3 groups: the control...We studied the effects of exercise on muscle mitochondria, and lipid and glycogen content in non-alcoholic steatohepatitis (NASH) model rats. Male Sprague-Dawley rats were randomly separated into 3 groups: the control group was fed standard chow;the NASH group was fed a methionine-choline-deficient high-fat diet (MCD);the NASH-exercise group was fed the MCD and exercised three times a week. Exercise training consisted of continuous running for thirty minutes at a 13 m/min, 6° slope on a motor-driven rodent treadmill for 6 weeks. Mitochondria content in NASH group decreased in the both fiber types compared with those of the control group. As compared between the NASH and NASH-exercise groups, however, exercise not only promoted significant improvements in liver fibrosis and cirrhosis and triglyceride (TG) content but also increased mitochondria content in type I muscle fiber in particular. These data suggest that exercise improved hepatic steatosis in NASH model rats and can prevent the progression of NASH.展开更多

S1P1 and S1P2 were originally identifie...Sphingosine-1-phosphate (S1P) is a blood-borne lipid mediator with pleiotropic biological activities. S1P acts via the specific cell surface G-protein-coupled receptors, S1P1-5. S1P1 and S1P2 were originally identified from vascular endothelial cells (ECs) and smooth muscle cells, respectively. Emerging evidence shows that S1P plays crucial roles in the regulation of vascular functions, including vascular formation, barrier protection and vascular tone via S1P1, S1P2 and S1P3. In particular, S1P regulates vascular formation through multiple mechanisms; S1P exerts both positive and negative effects on angiogenesis and vascular maturation. The positive and negative effects of S1P are mediated by S1P1 and S1P2, respectively. These effects of S1P1 and S1P2 are probably mediated by the S1P receptors expressed in multiple cell types including ECs and bone-marrow-derived cells. The receptor-subtype-specific, distinct effects ofS1P favor the development of novel therapeutic tactics for antitumor angiogenesis in cancer and therapeutic angiogenesis in ischemic diseases.展开更多

The aim of this study is to clarify the utility of its self-checklist, “loco-check,” as a tool for estimating the physical dysfunction of elderly people. Methods: Subjects were 1124 community-dwelling Japanese people, 557 men and 567 women, aged 40-89 years. Information about the seven “loco-check” items was obtained from present inquiry sheets. Physical functions were examined by grip strength, knee extension strength, walking speed and one-leg standing time with open eyes. The averages of these test values, controlled for age and BMI, were compared between the “loco-check” (+) group and the “loco-check” (-) group. Also we examined about the trend of decline of physical function, together with SF36 physical function subscale score, as the number of the items chosen increased. Results: Adjusted average values of all four physical function examinations in the “lococheck” (+) group were significantly lower than those of the “loco-check” (-) group (all, p . Also the adjusted average values of the majority of four tests were significantly lower in those who checked each of the “loco-check” items than those who did not, for most of the items. It was also revealed that the more items subjects checked, the lower the adjusted average values were, except for one-leg standing time. To prevent LS, it is important to increase muscle mass and muscle strength in middle-age by continuous resistance training. A total of 38 men and women were assessed at baseline and 6 months. Body composition, physical strength and salivary cortisol and cortisone were analyzed. The exercise intervention program was performed by individual muscle endurance level. Body weight, muscle weight and basal metabolism were increased after exercise intervention. The 30-second sit-up test and 3-minute walking were increased, and the 10-time sit-to-stand was decreased significantly. This may be related to increase of leg and abdominal muscular strength. The exercise intervention program increased salivary 11β-hydroxysteroid dehydrogenase type 2 (11β-HSD2) activities significantly. These results suggested that 11β-HDS2 became the index for the increase of muscular strength to prevent LS.展开更多

Treatment group (n = 7) consisted of a 500 IU/day vitamin D3 corresponding to twice of daily requirement for 6 months. Blood was collected by venipuncture and the serum 25-hydroxy vitamin D (25 OHD), 1,25-hydroxy vitamin D (1,25 OHD), Ca and PTH concentration were measured. The Mini-Mental State Examination and the Montreal Cognitive Assessment-Japanese version were used for the cognitive function test. Physical function was measured objectively using the Timed Up and Go (TUG), 4-mgait speed test (4 MGS), 5-repetition sit-to-stand (5 STS), 30-second chair stand test (CS-30) and measurement of upper grip strength. Our results show that a 6-month intake of vitamin D3 increased serum 25 OHD within the insufficiency and sufficiency levels without changing 1,25 OHD, Ca and PTH concentration. And it significantly decreased 4 MGS without changing TUG, 5 STS, CS-30, upper grip strength and cognitive function. Serum 25 OHD cut-off values for skeletal muscle index, grip strength and 4 MGS in diagnosis of sarcopenia are 18.6 - 23.4 ng/mL. These findings suggest that serum 25 OHD levels (23.4 ng/mL) might improve moving capacity, lower limb muscle strength, and physical balance functional impairment.展开更多

Gastroscopic examination revealed multiple gastric ulcers near t...A spiral bacterium (SH9), morphologically different from Helicobacter pylori (H. pylori), was found in a 62-year-old woman’s gastric mucosa. Gastroscopic examination revealed multiple gastric ulcers near the pyloric ring; mapping gastric biopsy showed mild mononuclear infiltration with large lymphoid follicles in the antrum, without corpus atrophy. Urea breath test and H. pylori culture were negative, but Giemsa staining of biopsies revealed tightly coiled bacteria that immunostained with anti-H. pylori antibody. Sequencing of SH9 16S rRNA and the partial urease A and B subunit genes showed that the former sequence had highest similarity (99%; 1302/1315 bp) to Helicobacter heilmannii (H. heilmannii) sensu stricto (H. heilmannii s.s.) BC1 obtained from a bobcat, while the latter sequence confirmed highest similarity (98.3%; 1467/1493 bp) to H. heilmannii s.s. HU2 obtained from a human. The patient was diagnosed with multiple gastric ulcers associated with H. heilmannii s.s. infection. After triple therapy (amoxicillin, clarithromycin, and lansoprazole) with regimen for eradicating H. pylori, gastroscopy showed ulcer improvement and no H. heilmannii s.s. upon biopsy.展开更多
Pomegranate fruit and its derived products are rich sources of bioactive compounds.They have many biological activities.Pomegranate molasses(PM)is a thick,traditional Middle Eastern syrup used in many recipes for Leba...Pomegranate fruit and its derived products are rich sources of bioactive compounds.They have many biological activities.Pomegranate molasses(PM)is a thick,traditional Middle Eastern syrup used in many recipes for Lebanese and international cuisines.It is a highly nutritious product which makes it of great interest.The purpose of this study was to assess the phytochemical composition,antioxidant and anti-diabetic properties of homemade and commercial PM consumed in Lebanon using samples collected from households in rural areas(n=4)and the market(n=28).The objectives of this study were to determine their total phenolic content using the Follin-Ciocalteu method,their total flavonoid content using aluminum chloride method,their antioxidant activity using DPPH radical scavenging as well as ferrous ion chelating assays and their anti-diabetic activity usingα-amylase andα-glucosidase inhibitory activities with acarbose,a known anti-diabetic drug,used as the standard reference.Homemade PM samples exhibited a higher antioxidant activity than commercial samples,with the most active homemade PM sample having IC_(50)values of 0.09 mg/mL(DPPH radical scavenging assay)and 46.78%ferrous ion chelating assay).Homemade PM samples also showed higher anti-diabetic activity than commercial samples,with the most active homemade PM sample having an IC_(50)values of 0.63 mg/mL(α-amylase inhibitory activity)and of 0.41 mg/mL(α-glucosidase inhibitory activity)and being almost as active as acarbose with both enzymes(1.5 times).Significant inverse strong/moderate correlations were observed between total phenolic content/total flavonoid content and the IC_(50)value of DPPH radical scavenging assay,indicating positive associations between total phenolic and flavonoid contents and antioxidant activity.In addition,significant negative moderate correlations were observed between total phenolic/flavonoid contents andα-amylase inhibitory activity,indicating positive associations between total phenolic and flavonoid contents and anti-diabetic activity.展开更多

The role of ZBTB7A in estrogen receptor ...ZBTB7A, a member of the POZ/BTB and Krüppel (POK) family of transcription factors, has been shown to have a context-dependent role in cancer development and progression. The role of ZBTB7A in estrogen receptor alpha (ERα)-positive breast cancer is largely unknown. Approximately 70% of breast cancers are classified as ERα-positive. ERα carries out the biological effects of estrogen and its expression level dictates response to endocrine therapies and prognosis for breast cancer patients. In this study, we find that ZBTB7A transcriptionally regulates ERα expression in ERα-positive breast cancer cell lines by binding to the ESR1 promoter leading to increased transcription of ERα. Inhibition of ZBTB7A in ERα-positive cells results in decreased estrogen responsiveness as demonstrated by diminished estrogen-response element-driven luciferase reporter activity, induction of estrogen target genes, and estrogen-stimulated growth. We also report that ERα potentiates ZBTB7A expression via a post-translational mechanism, suggesting the presence of a positive feedback loop between ZBTB7A and ERα, conferring sensitivity to estrogen in breast cancer. Clinically, we find that ZBTB7A and ERα are often co-expressed in breast cancers and that high ZBTB7A expression correlates with improved overall and relapse-free survival for breast cancer patients. Importantly, high ZBTB7A expression predicts a more favorable outcome for patients treated with endocrine therapies. Together, these findings demonstrate that ZBTB7A contributes to the transcriptional program maintaining ERα expression and potentially an endocrine therapy-responsive phenotype in breast cancer.展开更多

文摘Pomegranate fruit and its derived products are rich sources of bioactive compounds.They have many biological activities.Pomegranate molasses(PM)is a thick,traditional Middle Eastern syrup used in many recipes for Lebanese and international cuisines.It is a highly nutritious product which makes it of great interest.The purpose of this study was to assess the phytochemical composition,antioxidant and anti-diabetic properties of homemade and commercial PM consumed in Lebanon using samples collected from households in rural areas(n=4)and the market(n=28).The objectives of this study were to determine their total phenolic content using the Follin-Ciocalteu method,their total flavonoid content using aluminum chloride method,their antioxidant activity using DPPH radical scavenging as well as ferrous ion chelating assays and their anti-diabetic activity usingα-amylase andα-glucosidase inhibitory activities with acarbose,a known anti-diabetic drug,used as the standard reference.Homemade PM samples exhibited a higher antioxidant activity than commercial samples,with the most active homemade PM sample having IC_(50)values of 0.09 mg/mL(DPPH radical scavenging assay)and 46.78%ferrous ion chelating assay).Homemade PM samples also showed higher anti-diabetic activity than commercial samples,with the most active homemade PM sample having an IC_(50)values of 0.63 mg/mL(α-amylase inhibitory activity)and of 0.41 mg/mL(α-glucosidase inhibitory activity)and being almost as active as acarbose with both enzymes(1.5 times).Significant inverse strong/moderate correlations were observed between total phenolic content/total flavonoid content and the IC_(50)value of DPPH radical scavenging assay,indicating positive associations between total phenolic and flavonoid contents and antioxidant activity.In addition,significant negative moderate correlations were observed between total phenolic/flavonoid contents andα-amylase inhibitory activity,indicating positive associations between total phenolic and flavonoid contents and anti-diabetic activity.

文摘ZBTB7A, a member of the POZ/BTB and Krüppel (POK) family of transcription factors, has been shown to have a context-dependent role in cancer development and progression. The role of ZBTB7A in estrogen receptor alpha (ERα)-positive breast cancer is largely unknown. Approximately 70% of breast cancers are classified as ERα-positive. ERα carries out the biological effects of estrogen and its expression level dictates response to endocrine therapies and prognosis for breast cancer patients. In this study, we find that ZBTB7A transcriptionally regulates ERα expression in ERα-positive breast cancer cell lines by binding to the ESR1 promoter leading to increased transcription of ERα. Inhibition of ZBTB7A in ERα-positive cells results in decreased estrogen responsiveness as demonstrated by diminished estrogen-response element-driven luciferase reporter activity, induction of estrogen target genes, and estrogen-stimulated growth. We also report that ERα potentiates ZBTB7A expression via a post-translational mechanism, suggesting the presence of a positive feedback loop between ZBTB7A and ERα, conferring sensitivity to estrogen in breast cancer. Clinically, we find that ZBTB7A and ERα are often co-expressed in breast cancers and that high ZBTB7A expression correlates with improved overall and relapse-free survival for breast cancer patients. Importantly, high ZBTB7A expression predicts a more favorable outcome for patients treated with endocrine therapies. Together, these findings demonstrate that ZBTB7A contributes to the transcriptional program maintaining ERα expression and potentially an endocrine therapy-responsive phenotype in breast cancer.
