Background:Chewing Khat(Catha edulis)releases cathine and cathinone,which may reduce appetite via an unknown mechanism.This study investigated the peripheral and central effects of fresh leaves and buds of Catha edulis water extract(CEWE)on appetite biomarkers,gene expression,and body weight,using in vivo,ex vivo,and in silico models.Methods:Rats of both sexes were orally administered CEWE at different doses and durations in three different experiments.Liquid chromatography-mass spectroscopy(LC-MS)-MS was used to detect cathinone and cathine in the murine blood.The effect of Khat on serotonin receptors was studied in isolated rat fundus samples.Docking of the two Khat ligands was performed on G(The 5-hydroxytryptamine-type 2C receptor(5-HT2C)in an agonist-bound active conformation)and H(5-HT2C in an antagonist-bound inactive conformation)proteins to determine which ligands are most likely to act as agonists or antagonists.Results:Significant differences(P<0.05)in body weight were observed between the CEWE-treated groups and the controls over eight weeks.However,the plasma leptin and ghrelin levels did not change significantly(P>0.05).The expression of the ghrelin and leptin genes was also unaffected,but the expression of the 5-hydroxytryptamine(5-HT)gene decreased(P<0.05)with CEWE treatment.CEWE antagonizes 5-HT receptors in isolated rat fundus samples.Docking findings indicated that the khat ligands bound to 5-HT2C receptors.Cathine and cathinone levels in rat plasma were measured.Conclusion:Khat extract may suppress appetite by antagonizing the 5-HT receptors.Further research is required to understand its mechanism and potential applications. 展开更多

Radiographic Equipment and Accessories as a Potential Source of Nosocomial Infection

However, the equipme... Background: Nosocomial infections have become a major challenge in healthcare facilities as they affect the quality of medical care. Radiological imaging plays a crucial role in medical diagnosis. However, the equipment and accessories used increase the risk of transmission of nosocomial bacteria. Objective: This study aims to reveal the extent and nature of microbiological contamination in four hospital diagnostic imaging departments to determine their potential role in the spread of nosocomial bacteria and to evaluate the effectiveness of routine daily disinfection practices in controlling microorganisms in diagnostic imaging departments. Methods & Results: In each department, swabs were taken from the surfaces of selected parts of the equipment and accessories three times a day (early morning, noon, and evening) for five consecutive days. Bacteria were isolated from 65 swabs (36.1% of all samples). The bacteria were isolated 3 times (4.6%) in the morning, 16 times (24.6%) at midday, and 46 times (70.7%) in the evening. The bacteria isolated were Escherichia coli (isolated 34 times;52.3%), Staphylococcus aureus (20 times;30.8%), Staphylococcus epidermidis (6 times;9.3%), and Klebsiella species (5 times;7.7%). Discussion & Conclusion: Findings demonstrated that radiology equipment and accessories are not free of bacteria and further improvements in the sterilization and disinfection of radiology equipment and accessories are needed to protect staff and patients from nosocomial infections. 展开更多

BACKGROUND Bone marrow-derived mesenchymal stem cells(MSCs)show podocyte-protective effects in chronic kidney disease.Calycosin(CA),a phytoestrogen,is isolated from Astragalus membranaceus with a kidney-tonifying effect.CA preconditioning enhances the protective effect of MSCs against renal fibrosis in mice with unilateral ureteral occlusion.However,the protective effect and underlying mechanism of CA-pretreated MSCs(MSCsCA)on podocytes in adriamycin(ADR)-induced focal segmental glomerulosclerosis(FSGS)mice remain unclear.AIM To investigate whether CA enhances the role of MSCs in protecting against podocyte injury induced by ADR and the possible mechanism involved.METHODS ADR was used to induce FSGS in mice,and MSCs,CA,or MSCsCA were administered to mice.Their protective effect and possible mechanism of action on podocytes were observed by Western blot,immunohistochemistry,immunofluorescence,and real-time polymerase chain reaction.In vitro,ADR was used to stimulate mouse podocytes(MPC5)to induce injury,and the supernatants from MSC-,CA-,or MSCsCA-treated cells were collected to observe their protective effects on podocytes.Subsequently,the apoptosis of podocytes was detected in vivo and in vitro by Western blot,TUNEL assay,and immunofluorescence.Overexpression of Smad3,which is involved in apoptosis,was then induced to evaluate whether the MSCsCA-mediated podocyte protective effect is associated with Smad3 inhibition in MPC5 cells.RESULTS CA-pretreated MSCs enhanced the protective effect of MSCs against podocyte injury and the ability to inhibit podocyte apoptosis in ADR-induced FSGS mice and MPC5 cells.Expression of p-Smad3 was upregulated in mice with ADR-induced FSGS and MPC5 cells,which was reversed by MSCCA treatment more significantly than by MSCs or CA alone.When Smad3 was overexpressed in MPC5 cells,MSCsCA could not fulfill their potential to inhibit podocyte apoptosis.CONCLUSION MSCsCA enhance the protection of MSCs against ADR-induced podocyte apoptosis.The underlying mechanism may be related to MSCsCA-targeted inhibition of p-Smad3 in podocytes. 展开更多

BACKGROUND Coronary artery disease(CAD)is a major cause of death worldwide,and India contributes to about one-fifth of total CAD deaths.The development of CAD has been linked to the accumulation of Nε-carboxymethyl-lysine(CML)in heart muscle,which correlates with fibrosis.AIM To assess the impact of CML and inflammatory markers on the biochemical and cardiovascular characteristics of CAD patients with and without diabetes.METHODS We enrolled 200 consecutive CAD patients who were undergoing coronary angiography and categorized them into two groups based on their serum glycosylated hemoglobin(HbA1c)levels(group I:HbA1c≥6.5;group II:HbA1c<6.5).We analyzed the levels of lipoproteins,plasma HbA1c levels,CML,interleukin-6(IL-6),tumor necrosis factor alpha(TNF-α),and nitric oxide.RESULTS Group I (81 males and 19 females) patients had a mean age of 54.2 ± 10.2 years, with a mean diabetes duration of4.9 ± 2.2 years. Group II (89 males and 11 females) patients had a mean age of 53.2 ± 10.3 years. Group I had moresevere CAD, with a higher percentage of patients with single vessel disease and greater stenosis severity in the leftanterior descending coronary artery compared to group II. Group I also exhibited a larger left atrium diameter.Group I patients exhibited significantly higher levels of CML, TNF-α, and IL-6 and lower levels of nitric oxide ascompared with group II patients. Additionally, CML showed a significant positive correlation with IL-6 (r = 0.596,P = 0.001) and TNF-α (r = 0.337, P = 0.001) and a negative correlation with nitric oxide (r=-4.16, P = 0.001). Oddsratio analysis revealed that patients with CML in the third quartile (264.43-364.31 ng/mL) were significantlyassociated with diabetic CAD at unadjusted and adjusted levels with covariates. 展开更多

Big Data Analytics with Optimal Deep Learning Model for Medical Image Classification

In recent years,huge volumes of healthcare data are getting generated in various forms.The advancements made in medical imaging are tremendous owing to which biomedical image acquisition has become easier and quicker.Due to such massive generation of big data,the utilization of new methods based on Big Data Analytics(BDA),Machine Learning(ML),and Artificial Intelligence(AI)have become essential.In this aspect,the current research work develops a new Big Data Analytics with Cat Swarm Optimization based deep Learning(BDA-CSODL)technique for medical image classification on Apache Spark environment.The aim of the proposed BDA-CSODL technique is to classify the medical images and diagnose the disease accurately.BDA-CSODL technique involves different stages of operations such as preprocessing,segmentation,fea-ture extraction,and classification.In addition,BDA-CSODL technique also fol-lows multi-level thresholding-based image segmentation approach for the detection of infected regions in medical image.Moreover,a deep convolutional neural network-based Inception v3 method is utilized in this study as feature extractor.Stochastic Gradient Descent(SGD)model is used for parameter tuning process.Furthermore,CSO with Long Short-Term Memory(CSO-LSTM)model is employed as a classification model to determine the appropriate class labels to it.Both SGD and CSO design approaches help in improving the overall image classification performance of the proposed BDA-CSODL technique.A wide range of simulations was conducted on benchmark medical image datasets and the com-prehensive comparative results demonstrate the supremacy of the proposed BDA-CSODL technique under different measures. 展开更多

Recently,computer assisted diagnosis(CAD)model creation has become more dependent on medical picture categorization.It is often used to identify several conditions,including brain disorders,diabetic retinopathy,and skin cancer.Most traditional CAD methods relied on textures,colours,and forms.Because many models are issue-oriented,they need a more substantial capacity to generalize and cannot capture high-level problem domain notions.Recent deep learning(DL)models have been published,providing a practical way to develop models specifically for classifying input medical pictures.This paper offers an intelligent beetle antenna search(IBAS-DTL)method for classifying medical images facilitated by deep transfer learning.The IBAS-DTL model aims to recognize and classify medical pictures into various groups.In order to segment medical pictures,the current IBASDTLM model first develops an entropy based weighting and first-order cumulative moment(EWFCM)approach.Additionally,the DenseNet-121 techniquewas used as a module for extracting features.ABASwith an extreme learning machine(ELM)model is used to classify the medical photos.A wide variety of tests were carried out using a benchmark medical imaging dataset to demonstrate the IBAS-DTL model’s noteworthy performance.The results gained indicated the IBAS-DTL model’s superiority over its pre-existing techniques. 展开更多

Relevant detection indicator of prethrombotic state in patients with primary hypertension

BACKGROUND Hypertension is a common chronic disease that affects many people worldwide.Only a few reports related to the exploration of relevant indicators of the prethrombotic state in patients with primary hypertension(PH)in clinical settings were available.AIM To detect prethrombotic state-related indicators in patients with PH and analyze their differences in different patient populations to provide a laboratory basis for the clinical prevention and control of hypertensive thrombotic diseases.METHODS The general data of patients with PH who attended the Department of Cardiovascular Medicine,The First Affiliated Hospital of Jiangxi Medical College,from January 2022 to December 2022 were collected retrospectively.The patients were divided into three groups of 40 patients each according to the Grade of PH:Grade 1,Grade 2,and Grade 3 hypertension experimental group.The baseline data of 40 volunteers,who underwent physical examination in our hospital but were not diagnosed with PH during the same period,were included in the control group.The relevant indicators of prethrombotic state of the participants were compared,and mainly included inflammation-related indicators,hemorheology-related indicators,and coagulation function related indicators.The relationship between the aforementioned indicators and the progression of PH was analyzed.RESULTS No significant differences were observed in age,sex,diabetes mellitus,smoking history,drinking history,body mass index,New York Heart Association functional classification,or the course of hypertension among the four groups(P>0.05).The expressions of high-sensitivity C-reactive protein(hs-CRP),thrombomodulin(TM),hematocrit(Hct),erythrocyte sedimentation rate(ESR),P-selectin on platelet surface(CD62P),and fibrinogen(FIB)in the control group were<Grade 1 hypertension group<Grade 2 hypertension group<Grade 3 hypertension group,and the expressions of platelet(PLT),activated partial thromboplastin time(APTT),prothrombin(PT),and plasma thrombin time(TT)in the control group was>Grade 1 hypertension group>Grade 2 hypertension group>Grade 3 hypertension group,and the difference was statistically significant(P<0.05).The results of the multivariate logistic regression model showed that the expression of hs-CRP,TM,Hct,ESR,CD62P,PLT,APTT,PT,TT,and FIB in the included participants was related to the progression of PH.Among these,high expression of hs-CRP,TM,Hct,ESR,CD62P,APTT,PT,and TT,and low expression of PLT and FIB were risk factors for PH(OR>1,P<0.05).The results of the receiver operating characteristic curve analysis showed that the area under the curve of hs-CRP,TM,ESR,CD62P,APTT,PT,TT,and FIB for the prediction of PH were>0.80,and the prediction value was ideal.Linear correlation analysis with bivariate Spearman showed that hs-CRP,TM,Hct,ESR,CD62P,APTT,PT,and TT were positively correlated with each other(r>0,P<0.05);PLT and FIB were negatively correlated with hs-CRP,TM,Hct,ESR,CD62P,APTT,PT,and TT(r<0,P<0.05);and PLT and FIB were positively correlated(r>0,P<0.05).Linear correlation analysis using bivariate Spearman showed that hs-CRP,TM,Hct,ESR,CD62P,and FIB were positively correlated with each other(r>0,P<0.05),whereas PLT,APTT,PT,and TT were negatively correlated with hs-CRP,TM,Hct,ESR,CD62P,and FIB(r<0,P<0.05).There was a positive correlation between PLT,APTT,PT,and TT(r>0,P<0.05).CONCLUSION The relevant indicators of the prethrombotic state in patients with PH,such as hs-CRP,TM,Hct,ESR,CD62P,PLT,APTT,PT,TT,and FIB,showed differences.High expression of hs-CRP,TM,Hct,ESR,CD62P,and FIB,and low expression of PLT,APTT,PT,and TT are the keys to the occurrence,progression,and thrombotic state of PH.Based on the above serum indicators’expression in patients,targeted interventions can be administered to patients with abnormal expression levels to control the progression of their disease and reduce the risk of developing a prethrombotic state. 展开更多

BACKGROUND Type 2 diabetes mellitus(T2DM)is a metabolic disease of impaired glucose utilization.Imbalance in generation and elimination of free radicals generate oxidative stress which modulates glucose metabolism and insulin regulation,resulting in the occurrence and progression of diabetes and associated complications.Antioxidant supplements in T2DM can be seen as a potential preventive and effective therapeutic strategy.AIM To compare randomized controlled trials(RCTs)in which antioxidants have been shown to have a therapeutic effect in T2DM patients.METHODS We systematically searched the electronic database PubMed by keywords.RCTs evaluating the effect of antioxidant therapy on glycaemic control as well as oxidant and antioxidant status as primary outcomes were included.The outcomes considered were:A reduction in blood glucose;changes in oxidative stress and antioxidant markers.Full-length papers of the shortlisted articles were assessed for the eligibility criteria and 17 RCTs were included.RESULTS The administration of fixed-dose antioxidants significantly reduces fasting blood sugar and glycated hemoglobin and is associated with decreased malondialdehyde,advanced oxidation protein products,and increased total antioxidant capacity.CONCLUSION Antioxidant supplements can be a beneficial approach for the treatment of T2DM. 展开更多

Effect of False Windows on Light Exposure and Sleep Quality in Hospitalized Patients

Purpose: We aimed to investigate the effects of installing false windows next to hospital beds without windows on the amount of light received by patients and their sleep quality. Methods: The study included patients admitted to the Department of Neurology at our hospital between September 2020 and August 2021. An Actigraph device was fitted to patients’ wrist and their beds to measure the amount of light received and sleep quality. Patients were divided into three groups: bed with a window, aisle bed with a false window, and aisle bed without a window. Mean sleep efficiency (%), mean steps (per day), and the amount of light (lux) received by the patients and beds were measured. Results: Valid data were obtained for 48 participants (median age, 66.5 years). There were 23 patients in beds with a window, 13 patients in aisle beds without a false window, and 12 in aisle beds with a false window. No statistically significant differences were found in terms of mean sleep efficiency, number of steps taken, and the amount of light received by the patients (P > 0.05);however, difference in the mean amount of light received by the beds at the location of the bed was statistically significant (P Conclusion: The amount of light that the patient receives is not necessarily affected by the location of the bed or the presence of a false window. 展开更多

The Effects of Energy Drink Consumption on Kidney and Liver Function: A Comparative Study

The current study aimed to investigate the potential health impacts associated with Energy Drink (ED) consumption and its effects on general health, liver function, and kidney function among students at Jazan University. EDs contain a variety of ingredients such as taurine, arginine, caffeine, acidity regulators, B vitamins, vitamin C, herbal extracts, and sugar. Previous studies have reported that EDs contain more caffeine content more a cup of coffee. Fifty-seven volunteer female students from the same socioeconomic background were eligible for the study based on their ED consumption habits. The students were divided into four groups: the control group (1) which did not consume EDs (19 students), group (2) which consumed 2 - 3 cans per week (19 students), group (3) which consumed 4 - 7 cans per week (14 students), and group (4) which consumed more than 7 cans per week (5 students). Five milliliters of venous blood were drawn from all subjects to determine ALT, AST, and GGT levels for liver function tests and creatinine, urea, uric acid, and BUN levels for kidney function tests. The results of the kidney function analysis showed that uric acid levels significantly increased (P 0.05) in all groups compared to the control group. The liver function analysis illustrated that, compared to the control group, only AST was significantly increased (P < 0.05) in groups 1 and 2. There was no significance in ALT and GGT in all groups. Overall, the study found that long-term ED consumption had a significant relationship to increased uric acid and AST concentrations, while urea and BUN decreased significantly. Further research is needed to understand the long-term health implications of ED consumption. 展开更多

Due to their high specificity and reliability, monoclonal antibodies (mAbs) have emerged as useful diagnostic and therapeutic tools for a variety of diseases. As a result, several scientists have jumped right into developing Ab-based assays for the identification of SARS-CoV-2 and Ab drugs for use as COVID-19 therapy agents. Since the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein is essential for viral infection and has a known precise structure, it has become a key target for the creation of therapeutic antibodies. The use of Ab cocktails is anticipated to be a key component of an efficient COVID-19 treatment plan since SARS-CoV-2 is an RNA virus with a high mutation rate, particularly when subjected to the selection pressure of aggressively applied preventive vaccinations and neutralizing Abs. Furthermore, SARS-CoV-2 infection could provoke an overzealous immune response, leading to a cytokine storm that accelerates the onset of a severe disease. Abs to counteract cytokine storms are also actively being researched as COVID-19 therapies. Abs are now used in SARS-CoV-2 detection assays, including immunoglobulin and antigen tests, in addition to their use as medicines. In order to stop the spread of COVID-19, such Ab-based detection tests are essential surveillance tools. In this article, we’ll go over several important ideas related to mAb-based COVID-19 pandemic detection tests and treatments. Objective: To understand the role of hybridoma technology in therapeutic implications. 1) To study the basic concepts and options in hybridoma technology;2) To study the applications of hybridoma technology;3) To explore how hybridoma technology is applied in diagnostic histopathology. Method: For this method generally there is use of mouse or mammals are transfect with the Ags to find out the formation of antibody afterwards isolate the antibody which has been formed after injecting the antigens for a number of weeks. Following are the steps for mAbs: Step 1: In this step immunization of mouse is done;Step 2: Spleen is used for the isolation of B cells;Step 3: Cultivation of cancerous cells;Step 4: Merging of B cells with Myeloma cells;Step 5: This step cell lines are separated;Step 6: in the next step screening the suitable cell lines;Step 7: observation of multiplication in vitro as well as in vivo;Step 8: Harvesting. Discussion: Now a day there are many diseases which has been cured easily at the mean time it’s very difficult to diagnose and get the treatment. In this article,a Susceptible-Exposed-Infectious-Recovered(SEIR)epidemic model is considered.The equilibrium analysis and reproduction number are studied.The conventional models have made assumptions of homogeneity in disease transmission that contradict the actual reality.However,it is crucial to consider the heterogeneity of the transmission rate when modeling disease dynamics.Describing the heterogeneity of disease transmission mathematically can be achieved by incorporating fuzzy theory.A numerical scheme nonstandard,finite difference(NSFD)approach is developed for the studied model and the results of numerical simulations are presented.Simulations of the constructed scheme are presented.The positivity,convergence and consistency of the developed technique are investigated using mathematical induction,Jacobean matrix and Taylor series expansions respectively.The suggested scheme preserves all these essential characteristics of the disease dynamical models.The numerical and simulation results reveal that the proposed NSFD method provides an adequate representation of the dynamics of the disease.Moreover,the obtained method generates plausible predictions that can be used by regulators to support the decision-making process to design and develop control strategies.Effects of the natural immunity on the infected class are studied which reveals that an increase in natural immunity can decrease the infection and vice versa. 展开更多

Amoebiasis, classified as the third intestinal parasitic infection, represents a public health problem in low-income countries where hygiene and sanitation conditions are poor. With the resurgence of resistant pathogenic strains as well as ancestral considerations in developing countries such as Cameroon, many people rely on medicinal plants to treat a plethora of diseases. This work aimed to highlight the anti-amoebic and anti-oxidant potential of Codiaeum variegatum extracts and fractions. The anti-amoebic potential of C. variegatum was assayed on the polyxenic culture of the clinical isolates of E. histolytica. Then, the anti-oxidant potential of the ethanolic/hydroethanolic extracts and fractions was evaluated through DPPH radical scavenging, iron reduction (FRAP), lipid peroxidation inhibitory potential and total antioxidant capacity tests followed by the determination of phenolic compound and flavonoid content. It was found that the fractionation process decreased the amoebicidal activities of C. variegatum leaf extracts. However hydroethanolic extract (CI<sub>50</sub>: 10.08 ± 0.42, 5.18 ± 0.09, 5.18 ± 0.09 μg/mL respectively after 24, 48 and 72 hours) was more active than ethanolic extract (CI<sub>50</sub>: 15.59 ± 6.17;9.61 ± 2.37;6.26 ± 3.22 μg/mL respectively after 24, 48 and 72 hours). Interestingly, the activities of hydroethanolic extract were significantly non-different compared to metronidazole CI<sub>50</sub>: 8.42 ± 0.44, 6.45 ± 0.22 and 3.42 ± 0.33 μg/mL, respectively after 24, 48 and 72 hours). Ethanolic extract and EF5 showed higher Phenolic compound contents and higher antioxidant activity than hydroethanolic extract and other fractions through DPPH radical scavenging power (EC50 = 311.50 ± 4.12 μg/mL) and total antioxidant capacity (44 ± 0.07 mgEAA/gF). However, these activities are significantly lower than those of ascorbic acid (EC50 = 31.20 ± 4.39 μg/mL, and 61.34 ± 4.42 μg/mL respectively). This low antioxidant activity was confirmed by poor phenolic and flavonoid compounds contents found in the extracts and fractions. The present result brings a new approach to the ethnopharmacological uses of C. variegatum against dysentery in cases associated with Amoebiasis in Cameroun. 展开更多

Objective:To green synthesize and characterize copper nanoparticles(Cu NPs)using Astragalus sinicus,as well as evaluate the acaricidal,larvacidal,and repellent activities of Cu NPs against Hyalomma anatolicum(H.anatolicum),one of the most prevalent ticks infesting cattle in Saudi Arabia.Methods:Cu NPs were green synthesized by adding the Astragalus sinicus extract to a copper sulfate solution.The acaricidal,larvicidal,and repellent activities of Cu NPs against H.anatolicum were assessed via the adult immersion test,the larval packet test,and the vertical movement behavior of tick larvae,respectively.The effects of Cu NPs on acetylcholinesterase as well as oxidative enzyme activities were examined.Results:The green synthesized Cu NPs displayed a spherical form with a size range of 15-75 nm.After exposure of adult H.anatolicum to different concentrations of Cu NPs,the viability rate of adult H.anatolicum and the mean number,weight,and hatchability of eggs were noticeably reduced,in comparison to the control group(P<0.001).In addition,the viability rate of larvae considerably declined(P<0.001)with the LC_(50)and LC_(90)values of 11.30 and 20.34μg/m L,respectively.The maximum repellent activity of Cu NPs was observed at 50,100,and 200μg/m L with complete repellent activity after 60,120,and 180 min of exposure,respectively.Cu NPs,mainly at 1/2 LC_(50)and LC_(50)concentrations,markedly suppressed the acetylcholinesterase activity of the larval stage of H.anatolicum(P<0.001).Moreover,Cu NPs,mainly at LC_(50)dose,significantly elevated malondialdehyde level while declining glutathione-S-transferase level in H.anatolicum larvae(P<0.001).Conclusions:Cu NPs show potent acaricidal,larvicidal,and repellent activities against adults and larvae of H.anatolicum.However,further studies must be performed to clarify the precise mechanisms and the efficacy of Cu NPs in practical use. 展开更多

In the present study, we carried out further detailed analysis on the involvement of Dectin-2, a CLR that senses high mannose polysaccharide, in innate immune responses induced by influenza virus hemagglutinin (HA). Treatment of HA with periodate or PNGase F induced lower interleukin (IL)-12p40 secretion by conventional dendritic cells (DCs) compared with the untreated group. In contrast, treatment with O-glycosidase did not affect cytokine production. Green fluorescent protein expression in canonical Dectin-2-transducing cells was approximately 3% - 12% following HA stimulation, except with the A/H1N1pdm09 subtype HA. This expression was markedly reduced in cells possessing mutated amino acids in the carbohydrate recognition domain of Dectin-2, especially following stimulation with HA derived from the A/H3N2 subtype. Interferon (IFN)-α production from CD11c<sup>+</sup>Siglec-H<sup>+</sup>PDCA-1<sup>+</sup> plasmacytoid DCs was significantly increased in Dectin-2 knockout mice compared with wild-type mice upon stimulation with HA except for the B/Yamagata lineage HA. These results suggested that Dectin-2 is involved in initiating inflammatory responses via mannose polysaccharide on HA. However, other mechanisms may function in the antiviral response, including the type I IFN axis. 展开更多

Microbial pathogens include bacteria, viruses, fungi, and parasites and together account for a significant percentage of acute and chronic human diseases. In addition to understanding the mechanisms by which various pathogens cause human disease, research in microbial pathogenesis also addresses mechanisms of antimicrobial resistance and the development of new antimicrobial agents and vaccines. Answering fundamental questions regarding host-microbe interactions requires an interdisciplinary approach, including microbiology, genomics, informatics, molecular and cellular biology, biochemistry, immunology, epidemiology, environment and interaction between host and microbe. Studies investigating the direct effects of pollutants on respiratory tract infections are very vast, but those interested in the role of a pre-existing disease and effects of the exposure on the response to secondary stresses are few. In an experimental study at concentrations of air pollutants found in urban environments, frank toxicological responses are rarely observed, however, exposure to secondary stress like the respiratory challenge with infectious bacteria can exacerbate the response of the experimental host. The models like experimental, mechanical, and mathematical are the most abstract, but they allow analysis and logical proofs in a way that other approaches do not permit. The present review is mostly concerned with these model representations particularly with a novel mathematical model explaining the interaction between pathogen and immunity including the equivalence point. 展开更多

Electroencephalogram(EEG)is a method of capturing the electrophy-siological signal of the brain.An EEG headset is a wearable device that records electrophysiological data from the brain.This paper presents the design and fab-rication of a customized low-cost Electroencephalogram(EEG)headset based on the open-source OpenBCI Ultracortex Mark IV system.The electrode placement locations are modified under a 10–20 standard system.The fabricated headset is then compared to commercially available headsets based on the following para-meters:affordability,accessibility,noise,signal quality,and cost.First,the data is recorded from 20 subjects who used the EEG Headset,and signals were recorded.Secondly,the participants marked the accuracy,set up time,participant comfort,and participant perceived ease of set-up on a scale of 1 to 7(7 being excellent).Thirdly,the self-designed EEG headband is used by 5 participants for slide changing.The raw EEG signal is decomposed into a series of band sig-nals using discrete wavelet transform(DWT).Lastly,thesefindings have been compared to previously reported studies.We concluded that when used for slide-changing control,our self-designed EEG headband had an accuracy of 82.0 percent.We also concluded from the results that our headset performed well on the cost-effectiveness scale,had a reduced setup time of 2±0.5 min(the short-est among all being compared),and demonstrated greater ease of use. 展开更多

Objective YAP1 plays a dual role as an oncogene and tumor suppressor gene in several tumors;differentiating between these roles may depend on the YAP1 phosphorylation pattern.The specific function of YAP1 in B cell acute lymphoblastic leukemia(B-ALL),however,is currently unclear.Thus,in the present study,the role of YAP1 in B-ALL was investigated using relevant cell lines and patient datasets.Methods The effects of shRNA-mediated knockdown on YAP1 and LATS1 levels in the NALM6 and MOLT-4 cell lines were examined using Western blotting,quantitative real-time polymerase chain reaction,flow cytometry,immunostaining,and nude mouse subcutaneous tumorigenesis experiments.Gene expression levels of Hippo pathway-related molecules before and after verteporfin(VP)treatment were compared using RNA-Seq to identify significant Hippo pathway-related genes in NALM6 cells.Results Patients with ALL showing high YAP1 expression and low YAP1-Ser127 phosphorylation levels had worse prognoses than those with low YAP1 protein expression and high YAP1-Ser127 phosphorylation levels.YAP1-Ser127 phosphorylation levels were lower in NALM6 cells than in MOLT-4 and control cells;YAP1 was distributed in the nuclei in NALM6 cells.Knockdown of YAP1 inhibited MOLT-4 and NALM6 cell proliferation and arrested the NALM6 cell cycle in the G0/G1 phase.Before and after VP treatment,the expression of the upstream gene LATS1 was upregulated;its overexpression promoted YAP1-Ser127 phosphorylation.Further,YAP1 was distributed in the plasma.Conclusion LATS1 may downregulate YAP1-Ser127 phosphorylation and maintain B-ALL cell function;thus,VP,which targets this axis,may serve as a new therapeutic method for improving the outcomes for B-ALL patients. 展开更多
