This review attempts to delineate the effects and roles of fermented foods on allergic responses(AR),specifically from milk,plant,and meat sources.Evidence for AR alleviation and aggravation were noted for many differ...This review attempts to delineate the effects and roles of fermented foods on allergic responses(AR),specifically from milk,plant,and meat sources.Evidence for AR alleviation and aggravation were noted for many different fermented food groups.Positive outcomes on AR through fermented foods could be linked to microbial hydrolysis of food allergens,improvement in gut microbiota robustness,and modulation of the immune system that promotes a balance between T helper 1(Th1)and Th2 cells.Studies on plant-based,non-protein rich fermented foods tend to show more favourable results compared to those on meat-based or protein-rich group.The usage of specific and known starter cultures are helpful in alleviating AR,as in the case for many yogurt,Kefir or Dahi products.Sufficient fermentation time was also deemed important,exemplified in studies that showed inefficient AR reduction through consumption of fresh cheese.However,formation of new allergens through fermentation of certain meat-based foods,or by using specific fermenting microbes(e.g.Penicillium sp.),is possible.Thus,combination of starter cultures and food substrates must be considered in preventing or eliminating AR from intake of these foods.This review may aid consumers to make informed decision during the consumption of fermented food.展开更多

This review takes stock of the diversity of spices and aromatic herbs, the chemical composition, the different properties and forms of use of six spices and aromatic herbs commonly used in Benin and around the world. These are Zingiber officinalis (ginger), Curcuma longa (curcuma), Syzygium aromaticum (clove) and three aromatic herbs Petroselinum crispum (parsley), Rosmarinus officinalis (rosemary), and Laurus nobilis (laurel). The methodology used is that of documentary research oriented towards the consultation of previous scientific documents that have highlighted the different pharmacological activities of the different species of spices and aromatic plants targeted. It is important to note that more than twenty plant species are used as spices and aromatic plants in Benin and around the world. Chemically, these different spices and aromatic herbs contain certain secondary metabolites such as flavonoids, tannins, coumarins, alkaloids, steroids, terpenes, saponins, and polyphenols. R.Br that are not yet well known. The HPLC-DAD-MS-ESI<sup>+</sup> method was used to investigate the chemical profile, the disc diffusion and microdilution method for the antimicrobial activities, the DPPH and FRAP assays for antioxidant property and SYBR Green I-based growth inhibition assay on Pf 3D7 (CQ-sensitive) strain for antiplasmodial properties. The results indicated significant amounts of hydroxybenzoic acid, flavonol and hydroxycinnamic acid in the two species. The total polyphenols analysis gave 209.0 ± 16.1 mg GAE/100g DW for P. erici-rosenii and 175.3 ± 1.6 mg GAE/100g DW for L. martinicensis while for total flavonoids analysis 33.4 ± 1.8 mg TE/100g DW was found for P. erici-rosenii and 39.6 ± 0.6 mg TE/100g DW for L. martinicensis. Moreover, antimicrobial tests revealed 0.125 mg/mL on E. faecalis ATCC25922 for ethanol extracts P. erici-rosenii and L. martinicensis. The antioxidant assays revealed IC<sub>50</sub> 56.17 μg/mL and 78.43 μg/mL for DPPH assay, and 112.03 μM TE/100mL and 90.46 μM TE/100mL) for FRAP assay for ethanol extracts of P. erici-rosenii and L. martinicensis respectively. Finally, for Antiplasmodial activity, IC<sub>50</sub> was 4.07 μg/mL for P. erici-rosenii and 12.29 μg/mL for L. martinicensis. Therefore, the antimicrobial, the antioxidant, and the antiplasmodial results suggest that the ethanolic extracts of the aerial parts of P. erici-rosenii and L. martinicensis contain compounds with antimicrobial, antioxidant, and antiplasmodial properties, which can be searched in new drug discovery for treatment of infectious diseases.展开更多

It was identified as Penicillium oxalicum based on its morphological characteristics and ITS rDNA gene sequence analysis. The effects of carbon source, nitrogen source and the concentration of methamidophos, temperature and pH on the degradation were investigated. The results showed that the strain could use glucose as carbon source and the methamidophos as sole nitrogen source. The degradation ratio of methamidophos, when the initial concentration was 1.0× 10^-3 mg mL^-1, could reach above 99.9% in 12 incubation days. The strain could use ethanol, glucose, fructose, sucrose, lactose, starch, and dextrin as its carbon and energy source to degrade the methamidophos. The favorable degrading condition of the strain ZHJ6 was in a mineral salt medium at pH 5.0 and 25℃ with 1% glucose, and further studies showed that the strain could degrade folimat, phoxim and glyphosate with glucose as carbon source, but could not degrade chlorpyrifos, phosdrin, trichlorphon, and dichlorvos.展开更多
Objective: To identify mutually regulated proteins in PC-3 and DU145 androgen-independent prostate cancer cell lines treated with 1,5-bis(2-hydroxyphenyl)-1,4-pentadiene-3-one(MS17), and to study the molecular pathway...Objective: To identify mutually regulated proteins in PC-3 and DU145 androgen-independent prostate cancer cell lines treated with 1,5-bis(2-hydroxyphenyl)-1,4-pentadiene-3-one(MS17), and to study the molecular pathways that contributed to the anticancer activity of MS17.Methods: PC-3 and DU145 cells were treated with 3 × EC_(50)(15 μM) concentration of MS17 for 24 h and were subjected to protein expression profiling using two-dimensional gel electrophoresis and protein identification by mass spectrometry.Selected differentially expressed proteins with significant P-value of P<0.05 and fold change over 1.5-folds were filtered through and ontologically classified.Mutually regulated proteins were ranked by fold change and identified as common protein targets of MS17.Results: Profiling data revealed that, the mutually down-regulated proteins included ACTB and ACTG associated with structural molecule activity, ACTN1 with cell cycle, ACTN4 with cell migration, HNRPK with apoptosis, PLST with morphogenesis and TERA with proteolysis.However, the expressions of CH60 and HS71 A respectively associated with response to unfolded protein demonstrated opposing regulation in PC-3 and DU145 cells.Pathway analysis of the differentially expressed proteins in PC-3 cells demonstrated the modulation of top pathways associated with cell-cell adhesion and cytoskeletal organization while in DU145 cells the pathways were associated with proteosomal degradation, regulation of electrolytes and water, regulation control of germ cells and organization of filament assembly/disassembly.Conclusions: The findings of the present study provide an understanding on the anti-tumorigenic activity of MS17 at the proteome level and warrant further research for its potential application for the management and treatment of androgen-independent prostate cancer.展开更多

The solubility was determined under low pressures and temperatures as a preliminary study for RPO par- ticle formation using scCO2. The solubility of RPO was 0.5-11.3 mg. (g CO2) -1 and was significantly affected by the pressure and temperature. RPO solubility increased with pressure and decreased with temperature. The Adachi-Lu model showed the best-fit for RPO solubility data with an average relative deviation of 14% with a high coefficient of determination, R2 of 0.9667, whereas the Peng-Robinson equation of state thermodynamic model recorded deviations of 17%-30%.展开更多

A comparison of the sequence of PHY7...The ful length phytase gene of Mitsuokel a jalaludini was successful y cloned and was found to be 1 047 bp in length, with 348 amino acids, and was designated as PHY7 phytase gene. A comparison of the sequence of PHY7 phytase gene of M. jalaludini with various microbial phytase gene sequences showed that it was not similar to those from other bacteria except Selenomonas ruminatium, thus suggesting that they may both express a new class of phytase. The PHY7 phytase gene was subsequently subcloned into bacterial expression vector, pET32a, for expression in Escherichia coli strain Ro-setta-gami. Expression of the recombinant phytase gene was optimised and characterised. The recombinant phytase was estimated to be approximately 55 kDa by SDS-PAGE analysis. The recombinant phytase exhibited optimum activity at 55°C, pH 4.5 and showed good pH stability from pH 3.5 to 5.5 (>78%relative activity). Metal ions such as Ca2+, Mg2+, and K+were found to exert signiifcant stimulatory effect on the recombinant phytase activity while Cu2+, Fe3+, and Zn2+greatly inhibited the enzyme activity. The recombinant phytase showed moderate resistance to trypsin proteolysis, but susceptible to pepsin proteolysis. The results of the study showed that several characteristics of recombinant phytase were slightly different from the native enzyme. Unfavourable characteristics such as reduced pH stability and metal ion effects should be taken into consideration during feed enzyme formulation.展开更多
Objective:To investigate antibiotic resistance profile and characterize Campylobacter jejuni(C.jejuni) isolates using random amplified polymorphic DNA(RAPD) analysis.Methods:Ninety eight C.jejuni isolates from farms a...Objective:To investigate antibiotic resistance profile and characterize Campylobacter jejuni(C.jejuni) isolates using random amplified polymorphic DNA(RAPD) analysis.Methods:Ninety eight C.jejuni isolates from farms and retail outlets were screened against 10 antibiotics commonly used clinically and agriculturally by using disk diffusion method.RAPD analysis was done to characterize 98 C.jejuni isolates.Results:Fifty-one percent of the isolates had multiple antibiotic resistance index 0.2 and below.This indicated that the isolates in the vegetables were not from the high risk environment or extensive farming practices.C.jejuni isolates found resistant towards penicillin G(93%),vancomycin(86%),ampicillin(35%),erythromycin(28%),gentamycin(4%),amikacin(3%),enrofloxacin(1%),norfloxacin(1%) and no resistance towards ciprofloxacin.RAPD clustering analysis showed that the contamination of C.jejuni in vegetables was likely due to cross contamination at retail markets.Conclusions:C.jejuni contamination in vegetables at retail markets was due to cross contamination.Current finding proved that C.jejuni in small scale vegetables production was less expose towards antibiotic abuse.展开更多
Objective: To identify commonly regulated genes in HPV-infected He La and Ca Ski cervical cancer cells treated with curcumin analogue 1,5-bis(2-hydroxyphenyl)-1,4-pentadiene-3-one(MS17) and to explore potential mechan...Objective: To identify commonly regulated genes in HPV-infected He La and Ca Ski cervical cancer cells treated with curcumin analogue 1,5-bis(2-hydroxyphenyl)-1,4-pentadiene-3-one(MS17) and to explore potential mechanisms that underlie its cytotoxic, anti-proliferative and apoptotic activity. Methods: He La and Ca Ski cells were treated with 2×EC50 and 3×EC50 doses of MS17 for 24 h and the RNA extracts were subjected to one-colour microarray-based gene expression profiling. Pair-wise significant genes(false discovery rate-corrected, P<0.05) were analysed for fold change(FC) compared to control samples. Differentially expressed genes with FC≥2.0(up-regulated; FC≥2.0 and down-regulated; FC≤-2.0) compared to the control samples were filtered through and analysed to create a global gene expression profile. Mutually regulated genes were ranked by FC and categorised by gene ontology. Results: Our data indicated dose-dependent regulation by MS17 and identified top 20 mutually upand down-regulated genes each in He La and Ca Ski cells. Amongst these 17 were commonly regulated in both cell lines. These include the up-regulation of CCL26, DEFB103 B, IL1 RL1, LY96, GCNT3, MMP10, MMP3, GADD45 G and HSPA6, and the down-regulation of TENM2, NEBL, KIFC1, CTDSP1, IGFBP5, LTBP1, NREP and MXD3. These genes were associated with key biological functions that were proposed to mediate the anticancer activity of MS17 in cervical cancer cells such as immune response, metabolic processes, proteolysis, programmed cell death, unfolded protein response, cell adhesion, cytoskeletal organisation, phosphatase activity, signal transduction and transcription regulator activity. Conclusions: Identification of seventeen common genes modulated by MS17 could be used as potential therapeutic targets in both cervical cancer cell lines and the findings of this study could be used to present an insight into the potential antitumor activity of MS17 in cervical cancer.展开更多

Body weight was determined at birth and then at 3, 6, 9, 12, 18, 24, 36, and...Quantitative, qualitative and phenotypic body measurements were determined on Longling Yellow goats in Longling of Yunnan Province in China. Body weight was determined at birth and then at 3, 6, 9, 12, 18, 24, 36, and 48 mon of age. Body measurements traits including height at wither (HW), body length (BL), heart girth (HG), chest depth (CD), and circumference of cannon bone (CCB) were determined at 3, 6, 9, 12, 18, 24, 36, and 48 mon of age. Phenotypic traits including coat colour, horns type, tip horn torsion, nose type, hair on forehead, wattles, ear lobes, black back line, hair on legs, and black hair on belly were determined at 12, 18, 24, 36, and 48 mon of age. Males were heavier (P〈0.01 or P〈0.05) than females; and except for chest depth at 24 mon of age (P 〉 0.05), all other body measurements were also higher (P 〈 0.01 or P 〈 0.05) in males. HW, BL, CD, HG, and BW increased (P 〈 0.01) with age, except for 48 mon of females (P 〉 0.05); while CCB did not always increase with age. HG had the highest correlation (P〈0.01) with body weights of both sexes. No differences were observed between the two sexes in the colour of the coat, horn type, tip horn torsion, nose type, ear lobes, black back line, and black hair on belly. However, differences exist between the two sexes in the presence of hair on forehead, wattles and long hair on legs. The female goats absence of tip horn torsion had positive effect on all variables measured at 36 and 48 mort of age (P〈0.01 or P〈0.05). The presence of wattles was associated with heavier body weights from 18 mort (P〈0.01) of age onward, and also with longer HW, BL and CD (P〈0.01 or P〈0.05) in female goats. Female goats with ear lobes had heavier body weights at 12, 36 and 48 mon (P〈0.01) and longer CCB at 12 and 24 mon of ages (P〈0.05). Hair on legs and black hair on belly only had partly effect body measurements on 12 mon of Longling Yellow female goats. Results of this study provide valuable information that will assist breeders and genetic improvement specialists when conducting selection and preservation programs in Longling Yellow goats in China.展开更多

Non...Background: Improving the unsaturated fatty acid content of ruminant meat is essential due to the generally saturated nature of fatty acids in ruminant meat and the negative effects this can have on human health. Nonetheless, enhancing the unsaturated fatty acid content of ruminant meat can have adverse effects on the shelf life and quality attributes of the meat. This study assessed the effects of dietary 80 % canola oil and 20 % palm oil blend(CPOB) on fatty acid composition, antioxidants, oxidative spoilage, cholesterol and physicochemical properties of semimembranosus(SM)muscle from goats. Twenty four Boer bucks were randomly assigned to diets containing on dry matter basis 0, 4 and8 % CPOB, fed for 100 d and slaughtered. The carcasses were subjected to a 7 d postmortem refrigerated storage. Al analyses were conducted on the SM muscle.Results: Diet had no effect(P 〉 0.05) on the concentration of free thiol and carbonyl and the band intensity of myosin heavy chain, actin and troponin T. The muscle glycogen, p H, water holding capacity, tenderness, glutathione peroxidase(GPX) activity, total carotenoid, δ-tocopherol, cholesterol and proximate composition did not differ(P 〉 0.05) between diets. The SM muscle from goats fed 4 and 8 % CPOB had lower(P 〈 0.05) concentration of C14:0 and C16:0 and higher(P 〈 0.05) concentration of C18:1 trans-11, C18:1ω-9, C18:3ω-3, C20:5ω-3 and C22:5ω-3 than the SM muscle from the control goats. Dietary CPOB increased(P 〈 0.05) the concentration of α and γ tocopherol and meat redness(a*) on d1 and 4 postmortem. Regardless of diet, antioxidant vitamins, and shear force decreased(P 〈 0.05) while drip loss, lipid and protein oxidation increased(P 〈 0.05) as postmortem storage progressed.Conclusion: Results evince that dietary CPOB can be used as a management tool to enhance the beneficial fatty acids and antioxidant contents of chevon without deleterious effects on its physicochemical properties and shelf life.展开更多
Apart from socio-economic disparities,indigenous people of the Himalayan range in Asia face an increasing trend of suicides.The tragic suicidal events usually go unaddressed,and no strategies are presently in place to...Apart from socio-economic disparities,indigenous people of the Himalayan range in Asia face an increasing trend of suicides.The tragic suicidal events usually go unaddressed,and no strategies are presently in place to mitigate suicides in the future.This study aims to explain the prevailing causes of suicidal ideation to come up with some policy recommendations.Through a preliminary survey,we identified social stigma,social isolation,lack of healthcare facilities,and domestic violence as the potential reasons for suicidal ideation.We identified individuals with suicide ideation for further data collection to test the relationships between the identified variables and suicidal ideation.A quantitative,explanatory and cross-sectional design was used to test the hypotheses.Inferential statistics were applied to the data containing 314 responses,and the results revealed that all the predictive variables influence the formation of suicidal ideation.The study uncovered some new dimensions of social stigma and social isolation that are common among the indigenous people.The results also indicate that some domestic violence is socially acceptable and considered a normal familial matter.Thefindings have theoretical and practical implications.We have identified new dimensions of social stigma,social isolation,and domestic violence that are new additions to the existing bank of literature.It provides insights to encourage public authorities,healthcare providers,and NGO managers to play their role in the right direction to mitigate the problem.This study opens further venues for researchers to investigate the complex issue of suicide more deeply.展开更多

Commercially availa...This study was aimed to determine sufficient frying time to reduce the number of Listeria monocytogenes present in chicken burger patties to non-detectable level which is fit for human consumption. Commercially available chicken burger patties were artificially contaminated with L. monocytogenes at level of approximately 9 log CFU/ml. The contaminated chicken burger patties were cooked for 0, 2, 4, 5, 8, and 10 minutes to determine survival of L. monocyto-genes. Results demonstrated a linear correlation between mean log reduction of L. monocytogenes and frying time. L. monocytogenes was not detected in chicken burger patties that were cooked for 6 minutes and above. As a result from this study, it is suggested that a minimum frying time for burger patties is 6 minutes. This can be treated as a safety measure to avoid consequences of consumption of undercooked burger patties.展开更多
BACKGROUND Cachexia is responsible for the low quality of life in pancreatic adenocarcinoma(PDAC).The rapid disease progression and patient deterioration seems related to perineural invasion,but the relationship betwe...BACKGROUND Cachexia is responsible for the low quality of life in pancreatic adenocarcinoma(PDAC).The rapid disease progression and patient deterioration seems related to perineural invasion,but the relationship between cachexia and perineural invasion for the evolution of the disease has been rarely studied.As perineural invasion is difficult to be highlighted,a biomarker such as the neurotrophic factor Midkine(MK)which promotes the neuronal differentiation and the cell migration could be helpful.Also,Activin(ACV)has been described as cachexia related to PDAC.However,their role for assessing and predicting the disease course in daily practice is not known.AIM To assess the relationship between perineural invasion and cachexia and their biomarkers,MK and ACV,respectively,and their prognostic value.METHODS This study included prospectively enrolled patients with proven adenocarcinoma and a matched group of controls without any malignancies.Patients with other causes of malnutrition were excluded.The plasma levels of ACV and MK were analyzed using western blotting and were correlated with the clinicopathological features and survival data.These results were validated by immunohistochemical analyses of the pancreatic tumor tissue of the patients included in the study and a supplementary group of surgically resected specimens from patients with a benign disease.RESULTS The study comprised 114 patients with PDAC,125 controls and a supplementary group of 14 benign pancreatic tissue samples.ACV and MK were both overexpressed more frequently in the plasma of patients with PDAC than in the controls(63% vs 32% for ACV,P<0.001;47%vs 16%for MK,P<0.001),with similar levels in pancreatic tissue the MK protein expression was closely related to the advanced clinical stage(P=0.006),the presence of metastasis(P=0.04),perineural invasion(P=0.03)and diabetes(P=0.002),but with no influence on survival.No correlation between clinicopathological factors and ACV expression was noted.Cachexia,present in 19%of patients,was unrelated to ACV or MK level.Higher ACV expression was associated with a shorter survival(P=0.008).CONCLUSION The MK was a biomarker of perineural invasion,associated with tumor stage and diabetes,but without prognostic value as ACV.Cachexia was unrelated to perineural invasion,ACV level or survival.展开更多
AIM: To investigate the mechanistic action of brewers' rice in regulating the Wnt/nuclear factor-kappa B(NF-κB)/Nrf2-signaling pathways during colon carcinogenesis in male Sprague-Dawley rats.METHODS: Male Spragu...AIM: To investigate the mechanistic action of brewers' rice in regulating the Wnt/nuclear factor-kappa B(NF-κB)/Nrf2-signaling pathways during colon carcinogenesis in male Sprague-Dawley rats.METHODS: Male Sprague-Dawley rats were randomly divided into the following five groups(six rats in each group):(G1) normal,(G2) azoxymethane(AOM) alone,(G3) AOM + 10%(weight(w)/weight(w)) brewers' rice,(G4) AOM + 20%(w/w) brewers' rice, and(G5) AOM + 40%(w/w) brewers' rice. They were intraperitoneally administered 15 mg/kg body weight of AOM in saline once weekly over a twoweek period and treated with an American Institute of Nutrition(AIN)-93 G diet containing 10%, 20%, and 40%(w/w) brewers' rice. The m RNA levels of glycogen synthase kinase 3β(GSK 3β), β-catenin, key inflammation markers, nuclear factor E2-related factor 2(Nrf2), and heme oxygenase-1(HO-1)-dependent transcriptional activity were assessed by quantitative real-time polymerase chain reaction analyses. The colon superoxide dismutase, malondialdehyde, and nitric oxide levels were also analyzed to assess the antioxidant effect of these treatments. The results were analyzed using one-way analysis of variance(ANOVA), and a P value of < 0.05 was considered significant.RESULTS: The overall analyses demonstrated that the dietary administration of brewers' rice in AOM-induced rat colon carcinogenesis resulted in the transcriptional upregulation of GSK 3β, inducible nitric oxide synthase(i NOS), Nrf2, and HO-1. We discovered that the dietary administration of brewers' rice downregulated the β-catenin and NF-κB m RNA levels. A significant reduction in β-catenin expression was found in the groups administered with 20%(0.611 ± 0.034) and 40%(0.436 ± 0.045)(w/w) brewers' rice compared with that of the group treated with AOM alone(1.000 ± 0.064)(P < 0.05). The NF-κB expression was significantly lower between the AOM-alone group(1.000 ± 0.048) and those groups fed with diets containing 10%(w/w) brewers' rice(0.255 ± 0.022), 20%(w/w) brewers' rice(0.450 ± 0.045), or 40%(w/w) brewers' rice(0.541 ± 0.027)(P < 0.05). Brewers' rice improved the antioxidant levels, indicating that brewers' rice can enhance effective recovery from oxidative stress induced by AOM.CONCLUSION: Our results provide evidence that brewers' rice can suppress colon cancer via the regulation of Nrf2 expression and the inhibition of the Wnt/NF-κB signaling pathways.展开更多
文摘This review attempts to delineate the effects and roles of fermented foods on allergic responses(AR),specifically from milk,plant,and meat sources.Evidence for AR alleviation and aggravation were noted for many different fermented food groups.Positive outcomes on AR through fermented foods could be linked to microbial hydrolysis of food allergens,improvement in gut microbiota robustness,and modulation of the immune system that promotes a balance between T helper 1(Th1)and Th2 cells.Studies on plant-based,non-protein rich fermented foods tend to show more favourable results compared to those on meat-based or protein-rich group.The usage of specific and known starter cultures are helpful in alleviating AR,as in the case for many yogurt,Kefir or Dahi products.Sufficient fermentation time was also deemed important,exemplified in studies that showed inefficient AR reduction through consumption of fresh cheese.However,formation of new allergens through fermentation of certain meat-based foods,or by using specific fermenting microbes(e.g.Penicillium sp.),is possible.Thus,combination of starter cultures and food substrates must be considered in preventing or eliminating AR from intake of these foods.This review may aid consumers to make informed decision during the consumption of fermented food.

文摘Meat products are an important part in our daily diet,providing valuable nutrients for the human body.However,heating processes cause the meat to become more appetizing with changes in texture,appearance,flavor,and chemical properties by the altering of protein structure and other ingredients.As one kind of cooking-induced contaminants,heterocyclic aromatic amines(HAAs)are widely present in protein aceous food products with strong carcinogenic and mutagenic properties.In order to promote the safety of traditional meat products,this review focused on the formation,metabolism,biological monitoring and inhibitory mechanism of HAA.An overview of the formation pathways,hazards,and control methods of HAAs during food processing in recent years was studied,aiming to provide some valuable information for exploring effective methods to inhibit the production of associated hazards during food processing.Systematic selection of different types of flavonoids to explore their effects on the formation of HAAs in an actual barbecue system can provide theoretical reference for effectively controlling the formation of HAAs and reducing their harm to human health.

文摘Dietary flavonoids are abundant in natural plants and possess multiple pharmacological and nutritional activities.In this study,apigenin,luteolin,and baicalein were chosen to evaluate their anti-diabetic effect in high-glucose and dexamethasone induced insulin-resistant(IR)HepG2 cells.All flavonoids improves the glucose consumption and glycogen synthesis abilities in IR-HepG2 cells via activating glucose transporter protein 4(GLUT4)and phosphor-glycogen synthase kinase(GSK-3β).These fl avonoids signifi cantly inhibited the production of reactive oxygen species(ROS)and advanced glycation end-products(AGEs),which were closely related to the suppression of the phosphorylation form of NF-κB and P65.The expression levels of insulin receptor substrate-1(IRS-1),insulin receptor substrate-2(IRS-2)and phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)pathway in IR-HepG2 cells were all partially activated by the fl avonoids,with variable effects.Furthermore,the intracellular metabolic conditions of the fl avonoids were also evaluated.
文摘Spices and aromatic plants are products of plant origin used in food. They are used for the preparation of remedies, for seasoning dishes or for preserving food. This review takes stock of the diversity of spices and aromatic herbs, the chemical composition, the different properties and forms of use of six spices and aromatic herbs commonly used in Benin and around the world. These are Zingiber officinalis (ginger), Curcuma longa (curcuma), Syzygium aromaticum (clove) and three aromatic herbs Petroselinum crispum (parsley), Rosmarinus officinalis (rosemary), and Laurus nobilis (laurel). The methodology used is that of documentary research oriented towards the consultation of previous scientific documents that have highlighted the different pharmacological activities of the different species of spices and aromatic plants targeted. It is important to note that more than twenty plant species are used as spices and aromatic plants in Benin and around the world. Chemically, these different spices and aromatic herbs contain certain secondary metabolites such as flavonoids, tannins, coumarins, alkaloids, steroids, terpenes, saponins, and polyphenols. This diversity of secondary metabolites alone or in a possible synergy may be responsible for many beneficial properties attributed to spices and aromatic herbs.
文摘This study has been conducted to analyze the chemical composition and antimicrobial, antioxidant and antiplasmodial properties of ethanol extracts of Pycnostachys erici-rosenii R.E.Fr and Leucas martinicensis (Jack.) R.Br that are not yet well known. The HPLC-DAD-MS-ESI<sup>+</sup> method was used to investigate the chemical profile, the disc diffusion and microdilution method for the antimicrobial activities, the DPPH and FRAP assays for antioxidant property and SYBR Green I-based growth inhibition assay on Pf 3D7 (CQ-sensitive) strain for antiplasmodial properties. The results indicated significant amounts of hydroxybenzoic acid, flavonol and hydroxycinnamic acid in the two species. The total polyphenols analysis gave 209.0 ± 16.1 mg GAE/100g DW for P. erici-rosenii and 175.3 ± 1.6 mg GAE/100g DW for L. martinicensis while for total flavonoids analysis 33.4 ± 1.8 mg TE/100g DW was found for P. erici-rosenii and 39.6 ± 0.6 mg TE/100g DW for L. martinicensis. Moreover, antimicrobial tests revealed 0.125 mg/mL on E. faecalis ATCC25922 for ethanol extracts P. erici-rosenii and L. martinicensis. The antioxidant assays revealed IC<sub>50</sub> 56.17 μg/mL and 78.43 μg/mL for DPPH assay, and 112.03 μM TE/100mL and 90.46 μM TE/100mL) for FRAP assay for ethanol extracts of P. erici-rosenii and L. martinicensis respectively. Finally, for Antiplasmodial activity, IC<sub>50</sub> was 4.07 μg/mL for P. erici-rosenii and 12.29 μg/mL for L. martinicensis. Therefore, the antimicrobial, the antioxidant, and the antiplasmodial results suggest that the ethanolic extracts of the aerial parts of P. erici-rosenii and L. martinicensis contain compounds with antimicrobial, antioxidant, and antiplasmodial properties, which can be searched in new drug discovery for treatment of infectious diseases.

文摘One methamidophos-degrading fungus strain, named as ZHJ6, was isolated from the soils contaminated with methamidophos. It was identified as Penicillium oxalicum based on its morphological characteristics and ITS rDNA gene sequence analysis. The effects of carbon source, nitrogen source and the concentration of methamidophos, temperature and pH on the degradation were investigated. The results showed that the strain could use glucose as carbon source and the methamidophos as sole nitrogen source. The degradation ratio of methamidophos, when the initial concentration was 1.0× 10^-3 mg mL^-1, could reach above 99.9% in 12 incubation days. The strain could use ethanol, glucose, fructose, sucrose, lactose, starch, and dextrin as its carbon and energy source to degrade the methamidophos. The favorable degrading condition of the strain ZHJ6 was in a mineral salt medium at pH 5.0 and 25℃ with 1% glucose, and further studies showed that the strain could degrade folimat, phoxim and glyphosate with glucose as carbon source, but could not degrade chlorpyrifos, phosdrin, trichlorphon, and dichlorvos.

文摘Objective: To identify mutually regulated proteins in PC-3 and DU145 androgen-independent prostate cancer cell lines treated with 1,5-bis(2-hydroxyphenyl)-1,4-pentadiene-3-one(MS17), and to study the molecular pathways that contributed to the anticancer activity of MS17.Methods: PC-3 and DU145 cells were treated with 3 × EC_(50)(15 μM) concentration of MS17 for 24 h and were subjected to protein expression profiling using two-dimensional gel electrophoresis and protein identification by mass spectrometry.Selected differentially expressed proteins with significant P-value of P<0.05 and fold change over 1.5-folds were filtered through and ontologically classified.Mutually regulated proteins were ranked by fold change and identified as common protein targets of MS17.Results: Profiling data revealed that, the mutually down-regulated proteins included ACTB and ACTG associated with structural molecule activity, ACTN1 with cell cycle, ACTN4 with cell migration, HNRPK with apoptosis, PLST with morphogenesis and TERA with proteolysis.However, the expressions of CH60 and HS71 A respectively associated with response to unfolded protein demonstrated opposing regulation in PC-3 and DU145 cells.Pathway analysis of the differentially expressed proteins in PC-3 cells demonstrated the modulation of top pathways associated with cell-cell adhesion and cytoskeletal organization while in DU145 cells the pathways were associated with proteosomal degradation, regulation of electrolytes and water, regulation control of germ cells and organization of filament assembly/disassembly.Conclusions: The findings of the present study provide an understanding on the anti-tumorigenic activity of MS17 at the proteome level and warrant further research for its potential application for the management and treatment of androgen-independent prostate cancer.

文摘The solubility of red palm oil (RPO) in supercritical carbon dioxide (scCO2) was determined using a dynamic method at 8.5-25 MPa and, 313.15-333.15 K and at a fixed scCO2 flow rate of 2.9 g. mn -1 using a full factorial design. The solubility was determined under low pressures and temperatures as a preliminary study for RPO par- ticle formation using scCO2. The solubility of RPO was 0.5-11.3 mg. (g CO2) -1 and was significantly affected by the pressure and temperature. RPO solubility increased with pressure and decreased with temperature. The Adachi-Lu model showed the best-fit for RPO solubility data with an average relative deviation of 14% with a high coefficient of determination, R2 of 0.9667, whereas the Peng-Robinson equation of state thermodynamic model recorded deviations of 17%-30%.

文摘The ful length phytase gene of Mitsuokel a jalaludini was successful y cloned and was found to be 1 047 bp in length, with 348 amino acids, and was designated as PHY7 phytase gene. A comparison of the sequence of PHY7 phytase gene of M. jalaludini with various microbial phytase gene sequences showed that it was not similar to those from other bacteria except Selenomonas ruminatium, thus suggesting that they may both express a new class of phytase. The PHY7 phytase gene was subsequently subcloned into bacterial expression vector, pET32a, for expression in Escherichia coli strain Ro-setta-gami. Expression of the recombinant phytase gene was optimised and characterised. The recombinant phytase was estimated to be approximately 55 kDa by SDS-PAGE analysis. The recombinant phytase exhibited optimum activity at 55°C, pH 4.5 and showed good pH stability from pH 3.5 to 5.5 (>78%relative activity). Metal ions such as Ca2+, Mg2+, and K+were found to exert signiifcant stimulatory effect on the recombinant phytase activity while Cu2+, Fe3+, and Zn2+greatly inhibited the enzyme activity. The recombinant phytase showed moderate resistance to trypsin proteolysis, but susceptible to pepsin proteolysis. The results of the study showed that several characteristics of recombinant phytase were slightly different from the native enzyme. Unfavourable characteristics such as reduced pH stability and metal ion effects should be taken into consideration during feed enzyme formulation.

文摘Objective:To investigate antibiotic resistance profile and characterize Campylobacter jejuni(C.jejuni) isolates using random amplified polymorphic DNA(RAPD) analysis.Methods:Ninety eight C.jejuni isolates from farms and retail outlets were screened against 10 antibiotics commonly used clinically and agriculturally by using disk diffusion method.RAPD analysis was done to characterize 98 C.jejuni isolates.Results:Fifty-one percent of the isolates had multiple antibiotic resistance index 0.2 and below.This indicated that the isolates in the vegetables were not from the high risk environment or extensive farming practices.C.jejuni isolates found resistant towards penicillin G(93%),vancomycin(86%),ampicillin(35%),erythromycin(28%),gentamycin(4%),amikacin(3%),enrofloxacin(1%),norfloxacin(1%) and no resistance towards ciprofloxacin.RAPD clustering analysis showed that the contamination of C.jejuni in vegetables was likely due to cross contamination at retail markets.Conclusions:C.jejuni contamination in vegetables at retail markets was due to cross contamination.Current finding proved that C.jejuni in small scale vegetables production was less expose towards antibiotic abuse.

文摘Objective: To identify commonly regulated genes in HPV-infected He La and Ca Ski cervical cancer cells treated with curcumin analogue 1,5-bis(2-hydroxyphenyl)-1,4-pentadiene-3-one(MS17) and to explore potential mechanisms that underlie its cytotoxic, anti-proliferative and apoptotic activity. Methods: He La and Ca Ski cells were treated with 2×EC50 and 3×EC50 doses of MS17 for 24 h and the RNA extracts were subjected to one-colour microarray-based gene expression profiling. Pair-wise significant genes(false discovery rate-corrected, P<0.05) were analysed for fold change(FC) compared to control samples. Differentially expressed genes with FC≥2.0(up-regulated; FC≥2.0 and down-regulated; FC≤-2.0) compared to the control samples were filtered through and analysed to create a global gene expression profile. Mutually regulated genes were ranked by FC and categorised by gene ontology. Results: Our data indicated dose-dependent regulation by MS17 and identified top 20 mutually upand down-regulated genes each in He La and Ca Ski cells. Amongst these 17 were commonly regulated in both cell lines. These include the up-regulation of CCL26, DEFB103 B, IL1 RL1, LY96, GCNT3, MMP10, MMP3, GADD45 G and HSPA6, and the down-regulation of TENM2, NEBL, KIFC1, CTDSP1, IGFBP5, LTBP1, NREP and MXD3. These genes were associated with key biological functions that were proposed to mediate the anticancer activity of MS17 in cervical cancer cells such as immune response, metabolic processes, proteolysis, programmed cell death, unfolded protein response, cell adhesion, cytoskeletal organisation, phosphatase activity, signal transduction and transcription regulator activity. Conclusions: Identification of seventeen common genes modulated by MS17 could be used as potential therapeutic targets in both cervical cancer cell lines and the findings of this study could be used to present an insight into the potential antitumor activity of MS17 in cervical cancer.

文摘Quantitative, qualitative and phenotypic body measurements were determined on Longling Yellow goats in Longling of Yunnan Province in China. Body weight was determined at birth and then at 3, 6, 9, 12, 18, 24, 36, and 48 mon of age. Body measurements traits including height at wither (HW), body length (BL), heart girth (HG), chest depth (CD), and circumference of cannon bone (CCB) were determined at 3, 6, 9, 12, 18, 24, 36, and 48 mon of age. Phenotypic traits including coat colour, horns type, tip horn torsion, nose type, hair on forehead, wattles, ear lobes, black back line, hair on legs, and black hair on belly were determined at 12, 18, 24, 36, and 48 mon of age. Males were heavier (P〈0.01 or P〈0.05) than females; and except for chest depth at 24 mon of age (P 〉 0.05), all other body measurements were also higher (P 〈 0.01 or P 〈 0.05) in males. HW, BL, CD, HG, and BW increased (P 〈 0.01) with age, except for 48 mon of females (P 〉 0.05); while CCB did not always increase with age. HG had the highest correlation (P〈0.01) with body weights of both sexes. No differences were observed between the two sexes in the colour of the coat, horn type, tip horn torsion, nose type, ear lobes, black back line, and black hair on belly. However, differences exist between the two sexes in the presence of hair on forehead, wattles and long hair on legs. The female goats absence of tip horn torsion had positive effect on all variables measured at 36 and 48 mort of age (P〈0.01 or P〈0.05). The presence of wattles was associated with heavier body weights from 18 mort (P〈0.01) of age onward, and also with longer HW, BL and CD (P〈0.01 or P〈0.05) in female goats. Female goats with ear lobes had heavier body weights at 12, 36 and 48 mon (P〈0.01) and longer CCB at 12 and 24 mon of ages (P〈0.05). Hair on legs and black hair on belly only had partly effect body measurements on 12 mon of Longling Yellow female goats. Results of this study provide valuable information that will assist breeders and genetic improvement specialists when conducting selection and preservation programs in Longling Yellow goats in China.
文摘Background: Improving the unsaturated fatty acid content of ruminant meat is essential due to the generally saturated nature of fatty acids in ruminant meat and the negative effects this can have on human health. Nonetheless, enhancing the unsaturated fatty acid content of ruminant meat can have adverse effects on the shelf life and quality attributes of the meat. This study assessed the effects of dietary 80 % canola oil and 20 % palm oil blend(CPOB) on fatty acid composition, antioxidants, oxidative spoilage, cholesterol and physicochemical properties of semimembranosus(SM)muscle from goats. Twenty four Boer bucks were randomly assigned to diets containing on dry matter basis 0, 4 and8 % CPOB, fed for 100 d and slaughtered. The carcasses were subjected to a 7 d postmortem refrigerated storage. Al analyses were conducted on the SM muscle.Results: Diet had no effect(P 〉 0.05) on the concentration of free thiol and carbonyl and the band intensity of myosin heavy chain, actin and troponin T. The muscle glycogen, p H, water holding capacity, tenderness, glutathione peroxidase(GPX) activity, total carotenoid, δ-tocopherol, cholesterol and proximate composition did not differ(P 〉 0.05) between diets. The SM muscle from goats fed 4 and 8 % CPOB had lower(P 〈 0.05) concentration of C14:0 and C16:0 and higher(P 〈 0.05) concentration of C18:1 trans-11, C18:1ω-9, C18:3ω-3, C20:5ω-3 and C22:5ω-3 than the SM muscle from the control goats. Dietary CPOB increased(P 〈 0.05) the concentration of α and γ tocopherol and meat redness(a*) on d1 and 4 postmortem. Regardless of diet, antioxidant vitamins, and shear force decreased(P 〈 0.05) while drip loss, lipid and protein oxidation increased(P 〈 0.05) as postmortem storage progressed.Conclusion: Results evince that dietary CPOB can be used as a management tool to enhance the beneficial fatty acids and antioxidant contents of chevon without deleterious effects on its physicochemical properties and shelf life.

文摘Apart from socio-economic disparities,indigenous people of the Himalayan range in Asia face an increasing trend of suicides.The tragic suicidal events usually go unaddressed,and no strategies are presently in place to mitigate suicides in the future.This study aims to explain the prevailing causes of suicidal ideation to come up with some policy recommendations.Through a preliminary survey,we identified social stigma,social isolation,lack of healthcare facilities,and domestic violence as the potential reasons for suicidal ideation.We identified individuals with suicide ideation for further data collection to test the relationships between the identified variables and suicidal ideation.A quantitative,explanatory and cross-sectional design was used to test the hypotheses.Inferential statistics were applied to the data containing 314 responses,and the results revealed that all the predictive variables influence the formation of suicidal ideation.The study uncovered some new dimensions of social stigma and social isolation that are common among the indigenous people.The results also indicate that some domestic violence is socially acceptable and considered a normal familial matter.Thefindings have theoretical and practical implications.We have identified new dimensions of social stigma,social isolation,and domestic violence that are new additions to the existing bank of literature.It provides insights to encourage public authorities,healthcare providers,and NGO managers to play their role in the right direction to mitigate the problem.This study opens further venues for researchers to investigate the complex issue of suicide more deeply.

文摘The poultry industry is experiencing rapid growth worldwide.This accelerated growth has led to multiple food fraud incidents across the food supply chain,which consequently created a demand for precise determination of quality poultry production.This increase in demand for precise poultry production quality has necessitated advanced solutions.Metabolomics has emerged as a viable solution by offering detailed differentiation of biochemical indicators throughout the poultry supply chain.Additionally,this study provides a means to address risk factors affecting the poultry industry without compromising animal welfare,which is a critical concern.This review focuses on important issues related to poultry product quality assessment.Food adulteration has escalated in recent years as it is driven by the increasing focus on consuming high-quality and nutritious food.However,there is no specifc guideline for such determinations,especially when appearance,texture,and taste can be manipulated by substituting for food components.Metabolomics can pave the way for a deeper understanding of existing and novel biochemical indicators responsible for determining the quality of poultry meat and eggs.This approach holds the potential to enhance the overall quality of poultry meat and egg products while also preventing food fraud.

文摘Objective:To determine? the anlihypercholesterolemic effects of kenaf seed samples and compare with the commercial hypocholesterolemic drug on serum lipids profiles and malondialdehyde(MDA) level in the rat.Methods:Kenaf seed oil(KSO),microencapsulated kenaf seed oil(MKSO),kenaf seed extract(KSE) and defatted kenaf seed meal(DKSM) were prepared and phytocHemicals screening on these samples were done prior in viro study.Phenolic compounds in KSF.were quantified using high performance liquid chromatography.There were 40(divided in eight diet groups of 5) male Sprague dawley rats adapted lo normal standard diet or hypercholesterolemic diet(HD) with or without the treatment of these kenaf samples for 32 days.Results:All the kenaf samples exhibited to contain most of the major phytocliemicals.KSE possessed gallic acid,tannic acid,catechin.benzaldehyde.benzoic acid,syringic acid,sinapic acid,ferulic acid,naringin acid,and protocatechuic acid.The significant higher(P<0.05) serum total cholesterol,low density lipoprotein cholesterol and MDA levels in HD group without treatment than the normal control group suggested the hypercholesterolemia was induced by the Incorporation of cholesterol into diet.KSE exhibited higher cholesterol-lowering properties due to the significant lower(P<0.05)in serum triglycerides,total cholesterol and MDA levels.KSF showed the highest efficiency of cholesterol-lowering activity,followed by KSO.MKSO and DKSM.Conclusions:DKSM.MKSO.KSO and KSE appeared lo have comparable anti-hypercholesterolemic effect with the commercial hypocholesterolemic drug.Hence,kenaf seed could be used as an alternative natural source lo replace the synthetic hypocholosterolemic drugs.
文摘This study was aimed to determine sufficient frying time to reduce the number of Listeria monocytogenes present in chicken burger patties to non-detectable level which is fit for human consumption. Commercially available chicken burger patties were artificially contaminated with L. monocytogenes at level of approximately 9 log CFU/ml. The contaminated chicken burger patties were cooked for 0, 2, 4, 5, 8, and 10 minutes to determine survival of L. monocyto-genes. Results demonstrated a linear correlation between mean log reduction of L. monocytogenes and frying time. L. monocytogenes was not detected in chicken burger patties that were cooked for 6 minutes and above. As a result from this study, it is suggested that a minimum frying time for burger patties is 6 minutes. This can be treated as a safety measure to avoid consequences of consumption of undercooked burger patties.

文摘BACKGROUND Cachexia is responsible for the low quality of life in pancreatic adenocarcinoma(PDAC).The rapid disease progression and patient deterioration seems related to perineural invasion,but the relationship between cachexia and perineural invasion for the evolution of the disease has been rarely studied.As perineural invasion is difficult to be highlighted,a biomarker such as the neurotrophic factor Midkine(MK)which promotes the neuronal differentiation and the cell migration could be helpful.Also,Activin(ACV)has been described as cachexia related to PDAC.However,their role for assessing and predicting the disease course in daily practice is not known.AIM To assess the relationship between perineural invasion and cachexia and their biomarkers,MK and ACV,respectively,and their prognostic value.METHODS This study included prospectively enrolled patients with proven adenocarcinoma and a matched group of controls without any malignancies.Patients with other causes of malnutrition were excluded.The plasma levels of ACV and MK were analyzed using western blotting and were correlated with the clinicopathological features and survival data.These results were validated by immunohistochemical analyses of the pancreatic tumor tissue of the patients included in the study and a supplementary group of surgically resected specimens from patients with a benign disease.RESULTS The study comprised 114 patients with PDAC,125 controls and a supplementary group of 14 benign pancreatic tissue samples.ACV and MK were both overexpressed more frequently in the plasma of patients with PDAC than in the controls(63% vs 32% for ACV,P<0.001;47%vs 16%for MK,P<0.001),with similar levels in pancreatic tissue the MK protein expression was closely related to the advanced clinical stage(P=0.006),the presence of metastasis(P=0.04),perineural invasion(P=0.03)and diabetes(P=0.002),but with no influence on survival.No correlation between clinicopathological factors and ACV expression was noted.Cachexia,present in 19%of patients,was unrelated to ACV or MK level.Higher ACV expression was associated with a shorter survival(P=0.008).CONCLUSION The MK was a biomarker of perineural invasion,associated with tumor stage and diabetes,but without prognostic value as ACV.Cachexia was unrelated to perineural invasion,ACV level or survival.

文摘AIM: To investigate the mechanistic action of brewers' rice in regulating the Wnt/nuclear factor-kappa B(NF-κB)/Nrf2-signaling pathways during colon carcinogenesis in male Sprague-Dawley rats.METHODS: Male Sprague-Dawley rats were randomly divided into the following five groups(six rats in each group):(G1) normal,(G2) azoxymethane(AOM) alone,(G3) AOM + 10%(weight(w)/weight(w)) brewers' rice,(G4) AOM + 20%(w/w) brewers' rice, and(G5) AOM + 40%(w/w) brewers' rice. They were intraperitoneally administered 15 mg/kg body weight of AOM in saline once weekly over a twoweek period and treated with an American Institute of Nutrition(AIN)-93 G diet containing 10%, 20%, and 40%(w/w) brewers' rice. The m RNA levels of glycogen synthase kinase 3β(GSK 3β), β-catenin, key inflammation markers, nuclear factor E2-related factor 2(Nrf2), and heme oxygenase-1(HO-1)-dependent transcriptional activity were assessed by quantitative real-time polymerase chain reaction analyses. The colon superoxide dismutase, malondialdehyde, and nitric oxide levels were also analyzed to assess the antioxidant effect of these treatments. The results were analyzed using one-way analysis of variance(ANOVA), and a P value of < 0.05 was considered significant.RESULTS: The overall analyses demonstrated that the dietary administration of brewers' rice in AOM-induced rat colon carcinogenesis resulted in the transcriptional upregulation of GSK 3β, inducible nitric oxide synthase(i NOS), Nrf2, and HO-1. We discovered that the dietary administration of brewers' rice downregulated the β-catenin and NF-κB m RNA levels. A significant reduction in β-catenin expression was found in the groups administered with 20%(0.611 ± 0.034) and 40%(0.436 ± 0.045)(w/w) brewers' rice compared with that of the group treated with AOM alone(1.000 ± 0.064)(P < 0.05). The NF-κB expression was significantly lower between the AOM-alone group(1.000 ± 0.048) and those groups fed with diets containing 10%(w/w) brewers' rice(0.255 ± 0.022), 20%(w/w) brewers' rice(0.450 ± 0.045), or 40%(w/w) brewers' rice(0.541 ± 0.027)(P < 0.05). Brewers' rice improved the antioxidant levels, indicating that brewers' rice can enhance effective recovery from oxidative stress induced by AOM.CONCLUSION: Our results provide evidence that brewers' rice can suppress colon cancer via the regulation of Nrf2 expression and the inhibition of the Wnt/NF-κB signaling pathways.
