In this study,high performance liquid chromatography(HPLC)and RNA-seq transcriptome sequencing were used to study the changes in soluble sugar components and flavonoids in Prunus persica‘Jinxiangyu’at different deve...In this study,high performance liquid chromatography(HPLC)and RNA-seq transcriptome sequencing were used to study the changes in soluble sugar components and flavonoids in Prunus persica‘Jinxiangyu’at different developmental stages(20–90 d after flowering)and screen the key genes regulating the formation of soluble sugar and flavonoids in the fruits.The results showed that 60–85 d after flowering was the key stage of quality formation of Prunus persica‘Jinxiangyu’,and the content of soluble sugar,soluble solid,fructose,and sucrose in the fruit increased significantly during this period.The sugar content of ripe fruits was mainly fructose and sucrose.The content of kaempferol glycoside was low in the fruit.Quercetin glycoside content was higher in the young fruit stage and decreased with fruit maturity.There were no anthocyanin compounds in the fruit.The expression levels of genes involved in flavonoid metabolism(ANS,DFR,F3H,FLS,4CL1,etc.)were low in the fruit.A total of 181 differentially expressed genes were identified during fruit development to participate in five sugar metabolism pathways,among which the SDH gene had a higher expression level,which continuously rised in the later stage of fruit development.It mainly promoted the accumulation of fructose content in the later stage of fruit development.The expression levels of SPS1,SS,and SS1 genes were continuously up-regulated,which played a key role in sucrose regulation.The higher expression levels of SUS3 and INVA genes in the early stage of fruit development promoted the degradation of sucrose.展开更多

The results showed that A11, A7, and A4 had higher main stalk yield than other combinations.The young stem/leaf ratios of A11, A5, A7, A4, A3, and A1 were in line with the quality requirements for young stem commodity. The soluble sugar content of A2,A8, and A10 was higher than that of CK(Fengyou 737), and the seed yields of A4,A3, A2, A1, A5, and A6 were higher than that of CK. The 11 rapeseed combinations were classified into 3 grades by grey relation analysis and cluster analysis. Two combinations, A4(Y20A×95C4R) and A11(3194A×09-5R), showed the weighted relation degrees higher than 0.95, which were clustered into grade I by cluster analysis. They had good agronomic traits and good performance as both oilseed and vegetable. A8, A5, A3, A7, A2, A10, A6, and A1 were clustered into grade Ⅱ and A9 into grade Ⅲ. The distributions of metals (Cd, Cu, Zn, and Pb) in subcellular fractions (cytosol, debris, and granules) of earthworm Metaphire californica were investigated. The relationship between soil metals and earthworms were analyzed to explain its high plasticity to inhabit in situ contaminated soil of Hunan Province, south China. The concentration of Cd in subcellular compartments showed the same pattern as Cu in the order of cytosol 〉 debris 〉 granules. The distribution of Zn and Pb in earthworms indicated a similar propensity for different subcellular fractions that ranked as granules 〉 debris 〉 cytosol for Zn, and granules 〉 cytosol 〉 debris for Pb. The internal metal concentrations in earthworms increased with the soil metals (p 〈 0.05). Significant positive correlations were found between soil Cd and Cd concentrations in cytosol and debris (p 〈 0.01). Moreover, the soil Pb concentration significantly influenced the Pb concentrations in cytosol and debris (p 〈 0.01), similar to that of Cd. The soil Cu concentrations was only associated with the Cu in granules (p 〈 0.05). Soil Zn concentrations correlated with the Zn concentrations in each subcellular fraction (p 〈 0.05). Our results provide insights into the variations of metals partitioning in earthworms at subcellular levels and the relationships of soil metals, which could be one of the detoxiflcation strategies to adapt the long-term contaminated environment.展开更多
Two-line hybrid breeding can fully utilize heterosis in crops.In thermo-sensitive genic male sterile(TGMS)lines,low critical sterility-inducing temperature(CSIT)is vital to safeguard the production of two-line hybrid ...Two-line hybrid breeding can fully utilize heterosis in crops.In thermo-sensitive genic male sterile(TGMS)lines,low critical sterility-inducing temperature(CSIT)is vital to safeguard the production of two-line hybrid seeds in rice(Oryza sativa),but the molecular mechanism determining CSIT is unclear.Here,we report the cloning of CSIT1,which encodes an E3 ubiquitin ligase,and show that CSIT1 modulates the CSIT of thermo-sensitive genic male sterility 5(tms5)-based TGMS lines through ribosome-associated quality control(RQC).Biochemical assays demonstrated that CSIT1 binds to the 80S ribosomes and ubiquitinates abnormal nascent polypeptides for degradation in the RQC process.Loss of CSIT1 function inhibits the possible damage of tms5 to the ubiquitination system and protein translation,resulting in enhanced accumulation of anther-related proteins such as catalase to suppress abnormal accumulation of reactive oxygen species and premature programmed cell death in the tapetum,thereby leading to a much higher CSIT in the tms5-based TGMS lines.Taken together,our findings reveal a regulatory mechanism of CSIT,providing new insights into RQC and potential targets for future two-line hybrid breeding.展开更多
