Gibberellins(GAs)play a crucial role in growth and development of the tomato fruit.Previously published studies focusing on the effect of GAs on tomato fruits used chemical treatments,constitutive overexpression or si...Gibberellins(GAs)play a crucial role in growth and development of the tomato fruit.Previously published studies focusing on the effect of GAs on tomato fruits used chemical treatments,constitutive overexpression or silencing of GA biosynthetic and catabolic genes globally throughout the plant.Fruit-specific overexpression of GA catabolic enzyme genes GA2-oxidases(GA2oxs),however,may provide an alternative method to study the role of endogenous GAs on the fruit development.In this study,we have identified 11 SlGA2ox proteins in tomato that are classified into three subgroups.Motif analysis and multiple sequence alignments have demonstrated that all SlGA2oxs,except SlGA2ox10,have similar motif compositions and high-sequence conservation.Quantitative reverse transcription-PCR analysis has showed that SlGA2oxs exhibit differential expression patterns in tomato fruits at different developmental stages.When the fruit-specific promoter TFM7 was used to control the expression of SlGA2ox1,we observed no changes in growth and development of vegetative organs.However,fruit weight,seed number and germination rate were significantly affected.We also treated tomato fruits with GA biosynthesis inhibitor and observed phenotypes similar to those of the transgenic fruits.Furthermore,we have demonstrated that expression of cell expansion and GA responsive genes were downregulated in transgenic tomato fruits,supporting that overexpression of the SlGA2ox1 leads to reduction in endogenous GAs.This study provides additional evidence that endogenous GAs and the SlGA2ox1 gene play an important role in controlling on fruit weight,seed development and germination in tomato plant.展开更多
Objective:To determine the seroprevalence of Toxoplasma gondii(T.gondii) infection in dogs and cats in Zhenjiang City,Jiangsu Province.Eastern China,and to evaluate the main associated risk factors relating to exposur...Objective:To determine the seroprevalence of Toxoplasma gondii(T.gondii) infection in dogs and cats in Zhenjiang City,Jiangsu Province.Eastern China,and to evaluate the main associated risk factors relating to exposure to 71 gondii in this region.Methods:Sera from 160 clogs and 116 cats from Zhenjiang City were tested for anti-T.gondii antibodies using EUSA.The seropositivity by area of activity,sex and age was analyzed.Results:Overall.21 dogs(13.l%) and 24 cats(20.7%) had antibodies to T.gondii.The infection rate in stray dogs(38.7%) and cats(28.6%! was significantly higher(P<0.05) than in household dogs(6.9%) and cats(18.2%).The seroprevalence in male clogs(14.8%) and cats(21.05%) were slighlly higher than their female counterparts(11.4%in dogs and 20.0%in cats),but were not significantly differenent(P>0.05).A high proportion of dogs at 3 to 6 years of age were positive to T.gondii(20.0%)while cats with relatively high seropositivity rates were at 0 to 1 year of age(33.3%).Conclusions:The prevalence of T.gondii infection in dogs and cats in Zhenjiang City was high,which is probably the main source of T.gondii infection in this area.展开更多

User interface includes basic information interface,job information interface,camera monitoring interface,and fault diagnosis interface. Finally,it made evaluation of the user interface from color,indicator lamp,dial,and pointer. Results indicate that the designed user interface layout conforms to cognition mentality and operation habits and easy to get familiar and grasp; graphical interface is vivid and easy to stimulate pleasure of drivers in operation; interface color matching is coordinated; the layout of controls is hierarchical and logic,and operating mode is consistent with Windows system.展开更多

After inoculation of TP-1 for 14 d,water-soaked necrotic lesions about 3 mm in diameter were observed,which were similar to natural infections,and the original strain was obtained after re-isolation. Morphology and sequencing identification confirmed that TP-1 was Lasiodiplodia theobromae. This is the first report of stem rot in D. officinale caused by L. theobromae in China.展开更多

In order to study the remediation technology and effect of the black-odour water bodies,the treatment project of typical black-odour river was taken as an example,and the technical measures,such as sediment dredging,a...In order to study the remediation technology and effect of the black-odour water bodies,the treatment project of typical black-odour river was taken as an example,and the technical measures,such as sediment dredging,aeration,adding microbial preparation and constructing ecological floating bed,were adopted to treat and restore the water body.The results showed that the black and odorous phenomenon of the river had been basically eliminated and the water quality had been significantly improved after the 9-week operation.The highest removal rates of COD,NH3-N and TP were 82.5%,77.6%and 81.4%,respectively,and the water quality was improved from inferior class V to class IV.The engineering practice indicated that the technical scheme was feasible and could effectively remove the pollutants such as organic matters,ammonia nitrogen and total phosphorus in the water,which could provide basis and reference for the treatment project of similar black-odour water body.展开更多

Meanwhile, orthogonal experiment was conducted to get the optimal culture medium and cultivation condition. [Result] The optimal gene in the pigment of monascus pigment was 200 g/L of rice, 30 g/L of sweet potato powder, 10 g/L of glucose, 15 g/L of monosodium glutamate, 0.1% of zinc sulfate, and 0.1% of magnesium sulfate. The optimal culture condition was 30 ℃ and initial pH was 6.0. Fermentation time was 72 h, but when 24-L fermentation pot was used, culture time can last to 84 h. Soybean (Glycine max (L.) Merrill) is a salt-sensitive crop,and its production is severely affected by saline soils.Therefore,the response of soybean seeds to salt stress during germination was investigated at both physiological and proteomic levels.The salt-tolerant cultivar Lee68 and salt-sensitive cultivar N2899 were exposed to 100 mmol/L NaCl until radicle protrusion from the seed coat.In both cultivars,the final germination percentage was not affected by salt,but the mean germination times of Lee68 and N2899 were delayed by 0.3 and 1.0 d,respectively,compared with controls.In response to salt stress,the abscisic acid content increased,and gibberellic acid (GA1+3) and isopentenyladenosine decreased.Indole-3-acetic acid increased in Lee68,but remained unchanged in N2899.The proteins extracted from germinated seeds were separated using two-dimensional gel electrophoresis (2-DE),followed by Coomassie brilliant blue G-250 staining.About 350 protein spots from 2-DE gels of pH range 3 to 10 and 650 spots from gels of pH range 4 to 7 were reproducibly resolved,of which 18 protein spots showed changes in abundance as a result of salt stress in both cultivars.After matrix-assisted laser desorption ionization-time of flight-mass spectroscopy (MALDI-TOF-MS) analysis of the differentially expressed proteins,the peptide mass fingerprint was searched against the soybean UniGene database and nine proteins were successfully identified.Ferritin and 20S proteasome subunit β-6 were up-regulated in both cultivars.Glyceraldehyde 3-phosphate dehydrogenase,glutathione S-transferase (GST) 9,GST 10,and seed maturation protein PM36 were down-regulated in Lee68 by salt,but still remained at a certain level.However,these proteins were present in lower levels in control N2899 and were up-regulated under salt stress.The results indicate that these proteins might have important roles in defense mechanisms against salt stress during soybean seed germination.展开更多

Leech introduces the Politeness Principle to provide a reasonable explanation for this question, that is, for politeness. From the perspective of the purpose of communication, this article tries to argue that the Politeness Principle has the greater binding force in conversation and it should be given priority over the Cooperative Principle.

文摘With the rapid development of social network,public opinion monitoring based on social networks is becoming more and more important.Many platforms have achieved some success in public opinion monitoring.However,these platforms cannot perform well in scalability,fault tolerance,and real-time performance.In this paper,we propose a novel social-network-oriented public opinion monitoring platform based on ElasticSearch(SNES).Firstly,SNES integrates the module of distributed crawler cluster,which provides real-time social media data access.Secondly,SNES integrates ElasticSearch which can store and retrieve massive unstructured data in near real time.Finally,we design subscription module based on Apache Kafka to connect the modules of the platform together in the form of message push and consumption,improving message throughput and the ability of dynamic horizontal scaling.A great number of empirical experiments prove that the platform can adapt well to the social network with highly real-time data and has good performance in public opinion monitoring.
文摘Spark is a distributed data processing framework based on memory.Memory allocation is a focus question of Spark research.A good memory allocation scheme can effectively improve the efficiency of task execution and memory resource utilization of the Spark.Aiming at the memory allocation problem in the Spark2.x version,this paper optimizes the memory allocation strategy by analyzing the Spark memory model,the existing cache replacement algorithms and the memory allocation methods,which is on the basis of minimizing the storage area and allocating the execution area according to the demand.It mainly including two parts:cache replacement optimization and memory allocation optimization.Firstly,in the storage area,the cache replacement algorithm is optimized according to the characteristics of RDD Partition,which is combined with PCA dimension.In this section,the four features of RDD Partition are selected.When the RDD cache is replaced,only two most important features are selected by PCA dimension reduction method each time,thereby ensuring the generalization of the cache replacement strategy.Secondly,the memory allocation strategy of the execution area is optimized according to the memory requirement of Task and the memory space of storage area.In this paper,a series of experiments in Spark on Yarn mode are carried out to verify the effectiveness of the optimization algorithm and improve the cluster performance.

文摘Soybean (Glycine max (L.) Merrill) is a salt-sensitive crop,and its production is severely affected by saline soils.Therefore,the response of soybean seeds to salt stress during germination was investigated at both physiological and proteomic levels.The salt-tolerant cultivar Lee68 and salt-sensitive cultivar N2899 were exposed to 100 mmol/L NaCl until radicle protrusion from the seed coat.In both cultivars,the final germination percentage was not affected by salt,but the mean germination times of Lee68 and N2899 were delayed by 0.3 and 1.0 d,respectively,compared with controls.In response to salt stress,the abscisic acid content increased,and gibberellic acid (GA1+3) and isopentenyladenosine decreased.Indole-3-acetic acid increased in Lee68,but remained unchanged in N2899.The proteins extracted from germinated seeds were separated using two-dimensional gel electrophoresis (2-DE),followed by Coomassie brilliant blue G-250 staining.About 350 protein spots from 2-DE gels of pH range 3 to 10 and 650 spots from gels of pH range 4 to 7 were reproducibly resolved,of which 18 protein spots showed changes in abundance as a result of salt stress in both cultivars.After matrix-assisted laser desorption ionization-time of flight-mass spectroscopy (MALDI-TOF-MS) analysis of the differentially expressed proteins,the peptide mass fingerprint was searched against the soybean UniGene database and nine proteins were successfully identified.Ferritin and 20S proteasome subunit β-6 were up-regulated in both cultivars.Glyceraldehyde 3-phosphate dehydrogenase,glutathione S-transferase (GST) 9,GST 10,and seed maturation protein PM36 were down-regulated in Lee68 by salt,but still remained at a certain level.However,these proteins were present in lower levels in control N2899 and were up-regulated under salt stress.The results indicate that these proteins might have important roles in defense mechanisms against salt stress during soybean seed germination.
