It is divided in subgroups according to its morpho...Introduction: Diffuse large B-cell lymphoma (DLBCL), not otherwise specified, is a large B-cell lymphoma with a diffuse growth pattern and aggressive clinical course. It is divided in subgroups according to its morphology, immunophenotype, and primary site. Dissemination to bone marrow occurs in 11% to 35% of cases and can be of concordant or discordant morphology. Objective: To examine the association, the type of bone marrow involvement in relation to the primary site, morphology, immunohistochemistry of DLBCLs and to determine the cases of Epstein-Barr virus positive DLBCLs. Materials and Methods: We reviewed lymph node and extranodal biopsies as well as the respective bone marrow biopsies in all cases of DLBCL diagnosed in the Hospital General de México during the period from 2002 to 2010. We used immunohystochemistry for immunophenotype identification (Hans’s algorithm) and an in-situ hybridization technique to detect presence of Epstein Barr encoded RNA (EBER). Results: We included 108 patients with a mean age of 51.9 years, 59 (55%) were men. DLBCL involved lymph nodes in 60% of cases and palatine tonsils in 13%. The centroblastic variant predominated (80%) and 58% originated from activated B-cells. Infiltration of bone marrow was present in 30% of cases and was discordant in 55% of these cases. Correlation between morphology and bone marrow infiltration was statistically significant (P = 0.0003). Presence of Epstein-Barr virus was demonstrated in 15% of patients older than 50 years. Conclusions: Dissemination to bone marrow occurred in 30% of cases and discordant involvement was most common. DLBCL originating from activated B-lymphocytes predominated and the most common extranodal sites were palatine tonsils, suggesting that our population has a clinical behavior similar to Asiatic populations.展开更多
Objective To investigate the relationship between human cytomegalovirus(HCMV)infection and peripheral blood CD14+CD16+monocytes in the pathogenesis of coronary heart disease(CHD),and to elucidate the mechanism of path...Objective To investigate the relationship between human cytomegalovirus(HCMV)infection and peripheral blood CD14+CD16+monocytes in the pathogenesis of coronary heart disease(CHD),and to elucidate the mechanism of pathogenesis in CHD by analyzing the correlation between infection,inflammation,and CHD,to provide a basis for the prevention,evaluation,and treatment of the disease.Methods In total,192 patients with CHD were divided into three groups:latent CHD,angina pectoris,and myocardial infarction.HCMV-IgM and-IgG antibodies were assessed using ELISA;CD14+CD16+monocytes were counted using a five-type automated hematology analyzer;mononuclear cells were assessed using fluorescence-activated cell sorting;and an automatic biochemical analyzer was used to measure the levels of triglyceride,cholesterol,high-and low-density lipoprotein cholesterols,lipoprotein,hs-CRp and Hcy.Results The positive rates of HCMV-IgM and-IgG were significantly higher in the CHD groups than in the control group.HCMV infection affects lipid metabolism to promote immune and inflammatory responses.Conclusion HCMV infection has a specific correlation with the occurrence and development of CHD.The expression of CD14+CD16+mononuclear cells in the CHD group was increased accordingly and correlated with acute HCMV infection.Thus,HCMV antibody as well as peripheral blood CD14+CD16+mononuclear cells can be used to monitor the occurrence and development of CHD.展开更多

The three-dimensional visualization model of human body duct is based on virtual anatomical structure reconstruction with duct angiography,which realizes virtual model transferred from two-dimensional,planar and stati...The three-dimensional visualization model of human body duct is based on virtual anatomical structure reconstruction with duct angiography,which realizes virtual model transferred from two-dimensional,planar and static images into three-dimensional,stereoscopic and dynamic ones repectively.In recent years,the multi-duct segmentation and division of the same specimen(or organ) is the focus of attention shared by surgeons and clinical anatomists.On the basis of 4.22 g/cm3 body bone density,this study has screened out metal oxide contract agent with different density for infusion and modeling,as well as compared and analyzed the effects of three-dimensional image of CT virtual bronchoscopy(CTVB),three-dimensional image of CT maximum intensity projection and three-dimensional model.This experiment result showed synchronously infusing multi-duct of same specimen(or organ) with contrast agent in different densities could reconstruct three-dimensional models of all ducts once only and adjust threshold to develop single or multiple ducts.It was easier to segment and observe the duct structure,anastomosis,directions and crossing in different parts,which was beyond comparison with three-dimensional image of CTVB.Although the existing three-dimensional duct reconstruction techniques still cannot be applied in living bodies temporarily,this study focused on a creative design of ducts segmentation in different density,which proposed a new experimental idea for developing multi-duct three-dimensional model in living body in the future.It will play a significant role in disease diagnosis and individual design in surgical treatment program.Therefore,this study observes the three-dimensional status of human duct with the application of contrast agent fillers in different density,combined with three-dimensional reconstruction technology.It provides an innovative idea and method for constructing three-dimensional model of digital multi-duct specimen,and the ultimate goal is to develop the digitized virtual human and precise medical treatment better and faster.展开更多

Following treatment with traditional Chinese medicine or western medicine, β-amyloid expression decreased and neuronal morphology was normal in the rat hippocampal CA1 region, in addition to significantly shortened average latency in the Morris water navigation task. These findings suggested that compound prescription of Buyuan Congnao decoction, similar to the curative effects of piracetam, decreased hippocampal β-amyloid expression in a rat model of Alzheimer's disease, as well as improved learning and memory.展开更多

The aim of this study was...Oxidative stress has been implicated in the inflammatory process of Systemic Lupus Erythematosus (SLE), particularly by the formation of anti-DNA autoantibodies, which can lead to DNA damage. The aim of this study was to investigate, through comet assay, whether the level of DNA damage in SLE patients is different from that of healthy subjects. Twenty-five adult SLE patients with SLEDAI up to ten, and 25 healthy subjects were paired according to age, gender and Body Mass Index (BMI). Other anthropometric variables were also assessed. Comet assay was assessed as the marker of oxidative stress described as DNA Damage (DD) percentage. Waist Circumference (WC), Hip Circumference (HC) and BMI were also performed. Exclusion criteria for patients and controls comprised smoking and other chronic disorders. Level of damage index was remarkably higher in SLE patients than in controls, and no significant differences between the groups were found for age, BMI, WC and HC. No stratification concerning gender was performed, since there were just two males per group. No correlation was observed between BMI and DD (%). DD increased in SLE, which reflects the oxidant/antioxidant imbalance in these patients. These findings support an association between oxidative stress and SLE. This stronger correlation observed in patients with low disease activity may be useful in elucidating the mechanisms of disease pathogenesis.展开更多
OBJECTIVE:To investigate the mechanism of action of the Lingbao Huxin Dan(灵宝护心丹)in treating bradycardia arrhythmia with coronary heart disease(BACHD)by network pharmacology.METHODS:The active ingredients of the L...OBJECTIVE:To investigate the mechanism of action of the Lingbao Huxin Dan(灵宝护心丹)in treating bradycardia arrhythmia with coronary heart disease(BACHD)by network pharmacology.METHODS:The active ingredients of the Lingbao Huxin Dan were screened on the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform and Bioinformatics tools designed for the analysis of molecular mechanisms of Chinese medicine platform;target prediction was conducted with the SwissTargetPrediction database,and Cytoscape 3.8 was used to construct a drug ingredient-target network.The Genecards,Online Mendelian Inheritance in Man,and DrugB ank databases were searched for disease targets.Venn plots were used to display the common targets of BA-CHD and active ingredients.The STRING platform was used to construct a protein-protein interaction network.The Metascape data platform was used for Gene Ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis to construct a signaling pathway network of the active ingredients of the Lingbao Huxin Dan.RESULTS:There were 121 active ingredients,899 related targets,39 targets important in BA-CHD and 14 targets which intersected between the active ingredients and BA-CHD.There were 27 core therapeutic ingredients,153 biological processes,18 cell ingredients and 20 molecular functions obtained by GO enrichment analysis.The KEGG pathway analysis yielded 19 signaling pathways.CONCLUSION:RBA-CHD may treat BA-CHD by regulating adrenergic receptor beta-1,alpha 1-αadrenergic receptor,calcium voltage-gated channel subunit alpha1 C,alpha-1β-adrenergic receptor,nitric oxide synthase 2,beta-2 adrenergic receptor,voltagedependent calcium channel subunit alpha-2/delta-1,angiotensin-converting enzyme,Raf-1 proto-oncogene serine/threonine-protein kinase,and other targets,potentially by affecting adrenergic receptor binding and calcium channel opening,to regulate the activity of cardiomyocytes.展开更多
Many circular RNAs(circRNAs)are reported to be abnormally expressed during the progression of various tumors,and these circRNAs can be used as anti-tumor targets.Therefore,it is important to identify circRNAs that can...Many circular RNAs(circRNAs)are reported to be abnormally expressed during the progression of various tumors,and these circRNAs can be used as anti-tumor targets.Therefore,it is important to identify circRNAs that can be used effectively for the clinical diagnosis and treatment of colorectal cancer(CRC).Here,we report that hsa_Circ_0000826(Circ_0000826),a circ RNA with significantly reduced expression level in CRC tissues,is associated with a poor prognosis in patients.The silencing of Circ_0000826 promotes the proliferation of CRC cells.Conversely,the overexpression of Circ_0000826 restricted CRC cell proliferation both in vitro and in vivo.Furthermore,Circ_0000826 could target AU-rich element RNA-binding protein 1(AUF1).AUF1,known as heterogeneous nuclear ribonucleoprotein D(hnRNP D),could bind to the c-MYC 3’-UTR and promote c-MYC expression.When Circ_0000826 binds to AUF1,it competitively inhibits the binding of AUF1 to the c-MYC 3’-UTR,which inhibits the c-MYC expression and cell proliferation.These results provide novel insights into the functional mechanism of Circ_0000826 action in CRC progression and indicate its potential use as a therapeutic target in CRC.展开更多

Firstly, we obtain the polypeptides of VAP by water extraction. Secondly, we observed the morphology, assayed the factors in the media by enzyme-linked immunosorbent assay, and detected the special neural molecules by immuno fl uorescence staining. NSCs were cultured on the cell climbing film. After neuronal differentiation, differentiated NSCs were mounted for immunocytochemistry with immunofluorescence technique.RESULTS: The differentiating cells look like neuron,some special factors, such as Glial cell line-derived neurotrophic factor, nerve growth factor, in the media can be detected while differentiated neuron-like cells can express the special neural molecules.CONCLUSION: Differentiation of NSCs towards neurons can be induced by velvet antler polypeptide.展开更多
OBJECTIVE: To assess the efficacy of Bushenjianpi prescription(BSJPP), a formula from Traditional Chinese Medicine, on a mouse model of autoimmune premature ovarian failure(POF) induced by mouse zona pellucida(ZP3) an...OBJECTIVE: To assess the efficacy of Bushenjianpi prescription(BSJPP), a formula from Traditional Chinese Medicine, on a mouse model of autoimmune premature ovarian failure(POF) induced by mouse zona pellucida(ZP3) and to investigate the mechanisms underlying the action.METHODS: After randomization, POF was induced in the model mice by immunization with ZP3. One week later, mice received low(8.1 mg/kg), moderate(16.2 mg/kg) and high(32.4 mg/kg) doses of BSJPP by gastrogavage once daily for 90 days. Premarin(0.03 mg/kg) served as the positive group. Serum samples were collected 1 week after the last dose and stored at-20 ℃ for analysis. After cervical dislocation, the uterus and ovaries were collected aseptically for evaluation by histological assessment, scanning electron microscopy, immunohistochemical staining, and Western blot and reverse transcription-polymerase chain reaction analyses.RESULTS: Serum E2 levels in POF model mice were decreased, whereas follicle-stimulating hormone(FSH) and luteinizing hormone(LH) levels were dramatically increased. Serum levels of E2,LH and FSH were reduced in POF model mice treated with BSJPP(moderate and high doses) and premarin. Anti-bone morphogenetic protein 15(BMP-15) and connexin 43(Cx43) were repressed in autoimmune POF model mice, whereas high expression was observed in control mice and those treated with BSJPP(moderate and high doses)and premarin.CONCLUSION: BSJPP is effective in treating ZP3-induced POF in mice and the increase in the expression of BMP-15 and Cx43 may be implicated in the mechanism underpinning the action.展开更多

Vitamin ...The H2-receptor antagonist cimetidine is an antiulcer drug also used for the treatment of cancer due to its antiangiogenic effect. However, this drug has caused structural changes in the seminiferous tubules. Vitamin B12 has been used as a therapeutic agent for the treatment of male infertility. The supplementation of rats with vitamin B12 during cimetidine treatment has recovered the damaged seminiferous tubules, but how this vitamin restores the seminiferous epithelium has not been clarified. In this study, we evaluated whether vitamin B12 improves the number of spermatogonia, spermatocytes, and sperm concentration in cimetidine-treated rats. Adult male rats were treated for 50 days as follows: cimetidine group received 100 mg kg^-1 b.w. of cimetidine, cimetidine-B12 group received cimetidine and 3 μg of vitamin B12-hydroxocobalamin, B12 group received only 3 μg of vitamin, and control group received saline. Sperm concentration was calculated and historesin-embedded testes sections were used for the quantitative analyses of spermatogonia (A; In/B) and spermatocytes. TUNEL method and PCNA immunofluorescence were performed. Cimetidine caused a significant reduction in sperm concentration. TUNEL-positive spermatogonia and spermatocytes were correlated to a significant reduction in the number of these cells. In cimetidine-B12 group, sperm concentration was higher than cimetidine group and a significant increase in the number of spermatogonia (stages Ⅱ-Ⅵ) was correlated to a high incidence of PCNA-immunolabeled spermatogonia and spermatocytes. The results show that the supplementation of rats with vitamin B12 during cimetidine treatment increases sperm concentration and exerts a potential effect in the recovery of spermatogonia and spermatocytes.展开更多

Comet assay was assessed as the marker of oxidative stress described as DNA Damage (DD) percentage. Waist Circumference (WC), Hip Circumference (HC) and BMI were also performed. Exclusion criteria for patients and controls comprised smoking and other chronic disorders. Level of damage index was remarkably higher in SLE patients than in controls, and no significant differences between the groups were found for age, BMI, WC and HC. No stratification concerning gender was performed, since there were just two males per group. No correlation was observed between BMI and DD (%). DD increased in SLE, which reflects the oxidant/antioxidant imbalance in these patients. These findings support an association between oxidative stress and SLE. This stronger correlation observed in patients with low disease activity may be useful in elucidating the mechanisms of disease pathogenesis.

文摘OBJECTIVE:To investigate the mechanism of action of the Lingbao Huxin Dan(灵宝护心丹)in treating bradycardia arrhythmia with coronary heart disease(BACHD)by network pharmacology.METHODS:The active ingredients of the Lingbao Huxin Dan were screened on the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform and Bioinformatics tools designed for the analysis of molecular mechanisms of Chinese medicine platform;target prediction was conducted with the SwissTargetPrediction database,and Cytoscape 3.8 was used to construct a drug ingredient-target network.The Genecards,Online Mendelian Inheritance in Man,and DrugB ank databases were searched for disease targets.Venn plots were used to display the common targets of BA-CHD and active ingredients.The STRING platform was used to construct a protein-protein interaction network.The Metascape data platform was used for Gene Ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis to construct a signaling pathway network of the active ingredients of the Lingbao Huxin Dan.RESULTS:There were 121 active ingredients,899 related targets,39 targets important in BA-CHD and 14 targets which intersected between the active ingredients and BA-CHD.There were 27 core therapeutic ingredients,153 biological processes,18 cell ingredients and 20 molecular functions obtained by GO enrichment analysis.The KEGG pathway analysis yielded 19 signaling pathways.CONCLUSION:RBA-CHD may treat BA-CHD by regulating adrenergic receptor beta-1,alpha 1-αadrenergic receptor,calcium voltage-gated channel subunit alpha1 C,alpha-1β-adrenergic receptor,nitric oxide synthase 2,beta-2 adrenergic receptor,voltagedependent calcium channel subunit alpha-2/delta-1,angiotensin-converting enzyme,Raf-1 proto-oncogene serine/threonine-protein kinase,and other targets,potentially by affecting adrenergic receptor binding and calcium channel opening,to regulate the activity of cardiomyocytes.

文摘Many circular RNAs(circRNAs)are reported to be abnormally expressed during the progression of various tumors,and these circRNAs can be used as anti-tumor targets.Therefore,it is important to identify circRNAs that can be used effectively for the clinical diagnosis and treatment of colorectal cancer(CRC).Here,we report that hsa_Circ_0000826(Circ_0000826),a circ RNA with significantly reduced expression level in CRC tissues,is associated with a poor prognosis in patients.The silencing of Circ_0000826 promotes the proliferation of CRC cells.Conversely,the overexpression of Circ_0000826 restricted CRC cell proliferation both in vitro and in vivo.Furthermore,Circ_0000826 could target AU-rich element RNA-binding protein 1(AUF1).AUF1,known as heterogeneous nuclear ribonucleoprotein D(hnRNP D),could bind to the c-MYC 3’-UTR and promote c-MYC expression.When Circ_0000826 binds to AUF1,it competitively inhibits the binding of AUF1 to the c-MYC 3’-UTR,which inhibits the c-MYC expression and cell proliferation.These results provide novel insights into the functional mechanism of Circ_0000826 action in CRC progression and indicate its potential use as a therapeutic target in CRC.

文摘OBJECTIVE: To investigate the neural differentiation capacity of water extraction of velvet antler.METHODS: Velvet antler(Cervus Nippon Temminck) polypeptide(VAP) was used to differentiate neural stem cells(NSCs) towards neurons in the study. Firstly, we obtain the polypeptides of VAP by water extraction. Secondly, we observed the morphology, assayed the factors in the media by enzyme-linked immunosorbent assay, and detected the special neural molecules by immuno fl uorescence staining. NSCs were cultured on the cell climbing film. After neuronal differentiation, differentiated NSCs were mounted for immunocytochemistry with immunofluorescence technique.RESULTS: The differentiating cells look like neuron,some special factors, such as Glial cell line-derived neurotrophic factor, nerve growth factor, in the media can be detected while differentiated neuron-like cells can express the special neural molecules.CONCLUSION: Differentiation of NSCs towards neurons can be induced by velvet antler polypeptide.

文摘OBJECTIVE: To assess the efficacy of Bushenjianpi prescription(BSJPP), a formula from Traditional Chinese Medicine, on a mouse model of autoimmune premature ovarian failure(POF) induced by mouse zona pellucida(ZP3) and to investigate the mechanisms underlying the action.METHODS: After randomization, POF was induced in the model mice by immunization with ZP3. One week later, mice received low(8.1 mg/kg), moderate(16.2 mg/kg) and high(32.4 mg/kg) doses of BSJPP by gastrogavage once daily for 90 days. Premarin(0.03 mg/kg) served as the positive group. Serum samples were collected 1 week after the last dose and stored at-20 ℃ for analysis. After cervical dislocation, the uterus and ovaries were collected aseptically for evaluation by histological assessment, scanning electron microscopy, immunohistochemical staining, and Western blot and reverse transcription-polymerase chain reaction analyses.RESULTS: Serum E2 levels in POF model mice were decreased, whereas follicle-stimulating hormone(FSH) and luteinizing hormone(LH) levels were dramatically increased. Serum levels of E2,LH and FSH were reduced in POF model mice treated with BSJPP(moderate and high doses) and premarin. Anti-bone morphogenetic protein 15(BMP-15) and connexin 43(Cx43) were repressed in autoimmune POF model mice, whereas high expression was observed in control mice and those treated with BSJPP(moderate and high doses)and premarin.CONCLUSION: BSJPP is effective in treating ZP3-induced POF in mice and the increase in the expression of BMP-15 and Cx43 may be implicated in the mechanism underpinning the action.
文摘The H2-receptor antagonist cimetidine is an antiulcer drug also used for the treatment of cancer due to its antiangiogenic effect. However, this drug has caused structural changes in the seminiferous tubules. Vitamin B12 has been used as a therapeutic agent for the treatment of male infertility. The supplementation of rats with vitamin B12 during cimetidine treatment has recovered the damaged seminiferous tubules, but how this vitamin restores the seminiferous epithelium has not been clarified. In this study, we evaluated whether vitamin B12 improves the number of spermatogonia, spermatocytes, and sperm concentration in cimetidine-treated rats. Adult male rats were treated for 50 days as follows: cimetidine group received 100 mg kg^-1 b.w. of cimetidine, cimetidine-B12 group received cimetidine and 3 μg of vitamin B12-hydroxocobalamin, B12 group received only 3 μg of vitamin, and control group received saline. Sperm concentration was calculated and historesin-embedded testes sections were used for the quantitative analyses of spermatogonia (A; In/B) and spermatocytes. TUNEL method and PCNA immunofluorescence were performed. Cimetidine caused a significant reduction in sperm concentration. TUNEL-positive spermatogonia and spermatocytes were correlated to a significant reduction in the number of these cells. In cimetidine-B12 group, sperm concentration was higher than cimetidine group and a significant increase in the number of spermatogonia (stages Ⅱ-Ⅵ) was correlated to a high incidence of PCNA-immunolabeled spermatogonia and spermatocytes. The results show that the supplementation of rats with vitamin B12 during cimetidine treatment increases sperm concentration and exerts a potential effect in the recovery of spermatogonia and spermatocytes.
