BACKGROUND Prostate cancer(PCa)has high morbidity and mortality rates in elderly men.With a history of thousands of years,traditional Chinese medicine derived from insects could be an important source for developing c...BACKGROUND Prostate cancer(PCa)has high morbidity and mortality rates in elderly men.With a history of thousands of years,traditional Chinese medicine derived from insects could be an important source for developing cancer-targeted drugs to prevent tumorigenesis,enhance therapeutic effects,and reduce the risk of recurrence and metastasis.Multiple studies have shown that Coridius chinensis(Cc)has anticancer effects.AIM To elucidate the mechanism of action of Cc against PCa via network pharma-cology and molecular docking.METHODS Potential targets for Cc and PCa were predicted using ChemDraw 19.0 software,the PharmMapper database and the GeneCards database.Then,the STRING database was used to construct the protein–protein interaction network.Gene Ontology(GO),Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment and molecular docking analyses were subsequently conducted to identify the key targets,active ingredients and pathways involved.RESULTS GO and KEGG analyses indicated that the PI3K-Akt signalling pathway was the critical pathway(P value<1.0×10-8).Multiple targeting ingredients that can affect multiple pathways in PCa have been identified in Cc.Seven active compounds(asponguanosines A,asponguanine B,asponguanine C,aspong-pyrazine A,N-acetyldopamine,aspongadenine B and aspongpyrazine B)were selected for molecular docking with 9 potential targets,and the results revealed that aspongpyrazine A and asponguanosine A are the main components by which Cc affects PCa(affinity<-5 kcal/mol,hydrogen bonding),but more studies are needed.CONCLUSION We used network pharmacology to predict the bioactive components and important targets of Cc for the treatment of PCa,supporting the development of Cc as a natural anticancer agent.展开更多

The tonifying kidney recipe participates in protection of neural plasticity, brain development, immune regulation, and a variety of other physiological processes to act as an antidepressant that has great similarity as vitamin D axis. This review aims to sum up how Non-coding RNAs(ncRNAs)regulate the development of periodontitis and provides a new perspective for understanding the pathogenesis of periodontitis.We explored the ncRNA's dual role ...This review aims to sum up how Non-coding RNAs(ncRNAs)regulate the development of periodontitis and provides a new perspective for understanding the pathogenesis of periodontitis.We explored the ncRNA's dual role in the development of periodontitis by summarizing evidence from previous in vivo and in vitro studies as well as clinical samples.In our review,the downregulation of 18 miRNAs,22 lncRNAs and 10 circRNAs demonstrates protective roles in periodontitis.In contrast,the expression of other 11 miRNAs,7 lncRNAs and 6 circRNAs are upregulated in periodontitis,which promote the progression of periodontitis.These dysregulated ncRNAs exert their protective or destructive roles by mainly influencing cell proliferation,differentiation and apoptosis via cross-talking with various molecules or signaling pathways.Our findings suggested which and how ncRNAs promote or delay the progression of periodontitis,which may greatly contribute to diagnose and therapy development of periodontitis based on ncRNAs in the future.展开更多
Tartary buckwheat(Fagopyrum tataricum)is a dual-purpose medicinal and food crop grown for its high contents of functional compounds and abundant nutrients.Although studies have shown the differences of total flavonoid...Tartary buckwheat(Fagopyrum tataricum)is a dual-purpose medicinal and food crop grown for its high contents of functional compounds and abundant nutrients.Although studies have shown the differences of total flavonoid content in Tartary buckwheat at different ploidy levels,the composition of flavonoid and its regulatory mechanisms are largely unknown.In this study,the leaf metabolome and transcriptome of diploid and tetraploid accessions of Tartary buckwheat were analyzed to gain insight into the impact of polyploidization on comparative secondary metabolite composition and molecular regulatory mechanism.Based on a widely targeted metabolomics analysis,a total of 792 metabolites were identified,including 127 flavonoids.The accumulation of 127 metabolites and expression of 3871 genes differed significantly between diploid and tetraploid Tartary buckwheat.Integrated metabolomics and transcriptome analysis revealed that chromosome doubling up-regulated the expression of upstream genes in the flavonoid biosynthesis pathway to promote the accumulation of flavonoids.The present results contribute to elucidation of the molecular mechanism of phenotypic variation associated with polyploidy in Tartary buckwheat.The findings provide a reference for further studies on phenotypic traits in polyploid Tartary buckwheat,the cloning of crucial regulatory genes,and utilization of genetic engineering technologies in the breeding of Tartary buckwheat.展开更多

TLJN has been shown to protect primary cultured hippocampal neurons. How-ever, its mechanism of action remains unclear. In the present study, primary cultured hippocampal neurons treated with Aβ1-42 (10 μmol/L) signiifcantly increased the release of lactate dehydroge-nase, which was markedly reduced by TLJN (2 μL/mL), speciifcally by the component geniposide (26 μmol/L), but not ginsenoside Rg1 (2.5 μmol/L). hTe estrogen receptor inhibitor, ICI182780 (1 μmol/L), did not block TLJN-or geniposide-mediated decrease of lactate dehydrogenase under Aβ1-42-exposed conditions. However, the phosphatidyl inositol 3-kinase or mitogen-activated protein kinase pathway inhibitor, LY294002 (50 μmol/L) or U0126 (10 μmol/L), respectively blo cked the decrease of lactate dehydrogenase mediated by TLJN or geniposide. hTerefore, these results suggest that the non-classical estrogen pathway (i.e., phosphatidyl inositol 3-kinase or mitogen-activated protein kinase) is involved in the neuroprotective effect of TLJN, speciifcally its component, geniposide, against Aβ1-42-mediated cell death in primary cultured hippocampal neurons.展开更多

Objective:To observe the clinical efficacy and safety of oral administration of the traditional Chinese herb rhubarb to treat acute exacerbation of chronic obstructive pulmonary disease(AECOPD).Method:This was a multi...Objective:To observe the clinical efficacy and safety of oral administration of the traditional Chinese herb rhubarb to treat acute exacerbation of chronic obstructive pulmonary disease(AECOPD).Method:This was a multicenter randomized double-blinded placebo controlled study that took place in 7 provinces of China that enrolled 244 patients(aged 18e80 years)who had acute exacerbation of COPD with the traditional Chinese syndrome pattern of phlegm-heat obstructing lung.Participants were divided into experimental and control groups.The experimental group received 4.5 g of rhubarb granules twice daily and the control group received placebo granules.Both groups also received conventional Western therapy consisting of oxygen therapy,an antibiotic,expectorant,and a bronchodilator.Treatment lasted 10 days.Symptom scores for cough,sputum volume and color,wheezing and chest tightness before treatment and on days 3,5,7,and 10 during the treatment were recorded.Lung function,arterial blood gas and levels of serum inflammatory factors,interleukin-4(IL-4),interleukin-8(IL-8),and interleukin-10(IL-10)and tumor necrosis factor-alpha(TNF-a),before and after treatment were measured.Results:The sample size of the full analysis set(FAS)was 244 participants,and the sample size of per protocol set(PPS)was 235.Following 10 days’treatment,symptom scores of the experimental group were markedly lower than those of the placebo group(FAS:mean difference1.67,95%CI:e2.66 to0.69,P Z 0.001;PPS:mean difference1.55,95%CI:2.56 to0.54,P Z 0.003).Lung function in the experimental group was significantly higher than in the placebo group(FEV1,FAS:mean difference 0.12,95%CI:0.06 to 0.18;P<0.001;PPS:mean difference 0.12,95%CI 0.05 to 0.18;P<0.001.FVC:FAS:mean difference 0.16,95%CI:0.06 to 0.26;P Z 0.002;PPS:mean difference 0.16,95%CI 0.05 to 0.26;P Z 0.003.FEV1%,FAS:mean difference 5.95,95%CI:3.36 to 8.53;P<0.001;PPS:mean difference 5.92,95%CI 3.28 to 8.56;P<0.001.).PaO2,PaCO2,as well as serum inflammatory factors were also improved when compared to the placebo group.There were no significant differences in the incidence rate of adverse reaction between the two groups.Conclusions:Compared with placebo,rhubarb granules significantly reduced symptom scores,improved blood oxygen level,controlled systemic inflammatory response,without significant adverse effects.Thus,rhubarb may be a beneficial adjuvant method for treating the phlegm-heat obstructing the lung syndrome pattern of AECOPD.展开更多
Polyploid plants grow well,are stress tolerant,and are rich in nutrients and bioactive compounds.Thus,they are useful for improving crop quality and yield.In this study,we compared the seed characteristics and metabol...Polyploid plants grow well,are stress tolerant,and are rich in nutrients and bioactive compounds.Thus,they are useful for improving crop quality and yield.In this study,we compared the seed characteristics and metabolite profiles of diploid and tetraploid tartary buckwheat,which was developed via an artificially induced chromosome doubling event.The length,width,area,and thousand-grain weight were greater for the tetraploid seeds than for the diploid seeds.However,the germination rate decreased for the tetraploid seeds.Additionally,there was a gap between the shell and kernel of the tetraploid seeds.Moreover,the water absorption rate was higher for the tetraploid than for the diploid seeds.Chromosome doubling increased the seed total flavonoid content and deepened the seed color.A principal component analysis of the ultrahigh-pressure liquid chromatography-high resolution mass spectrometry data revealed the clear separation between the diploid and tetraploid samples.An orthogonal partial least squares-discriminant analysis and other multivariate statistical analyses identified 83 differentially abundant compounds,with most of the flavonoid metabolites more abundant in the tetraploid than in the diploid seeds.Research on tartary buckwheat polyploidy may result in enhanced germplasm resources and may clarify the mechanism underlying the biosynthesis of bioactive compounds.展开更多
Stress is an inevitable interference factor that seriously affects health.Listening to music is an economical,noninvasive,and highly accepted tool for easing stress.However,physiological studies investigating the abil...Stress is an inevitable interference factor that seriously affects health.Listening to music is an economical,noninvasive,and highly accepted tool for easing stress.However,physiological studies investigating the ability of music to reduce stress in daily life are limited.We established rat models of chronic immobilization stress(CIS)to observe changes in the hypothalamic arcuate nucleus(ARC)neurons involved in the regulation of food intake and the effect of comfortable classical music exposure.Twenty-one days of stress resulted in decreased food intake and delayed body weight gain,up-regulation of leptin receptor(Ob-R),cocaine-and amphetamine-regulated transcript(CART),proopiomelanocortin(POMC),and alpha-melanocyte-stimulating hormone(α-MSH)expression,and downregulation of neuropeptide Y(NPY)and agouti-related protein(AgRP)expression in the ARC.Thus,peripheral leptin entered the ARC under chronic stress conditions,bound to Ob-R,and affected downstream nerve pathways related to appetite,such as the NPY/AgRP and CART/POMC pathways.Gentle classical music played at 65 dB reversed the abnormal expression of Ob-R and NPY induced by chronic stress.Thus,listening to comfortable music improves changes in ARC neurons related to the regulation of food intake in CIS rats,and these results provide a reference for basic research regarding how music therapy alleviates stress and stress-related health issues.展开更多

Microinjection, reverse transcriptase-PCR (RT-PCR), in situ hybridization and immunofluorescent staining were used in this research, and the morphology of the islet in normal zebrafish embryos, and in those treated with the siRNA specific to pdx-1 (siPDX-l) or siGFP was observed and compared. The expression of pdx-1 was detected in the stages of 1-cell, 2-cell, 4-cell, 8-cell, 16-cell, 16-hour by RT-PCT. The in situ hybridization and immunofluorescent staining results showed that siPDX-1 disturbed the formation of the islet in zebrafish embryos. Pdx-1 played multiple roles in maintaining the phenotype of the islet during embryogenesis in zebrafish.展开更多

BACKGROUND Post-traumatic stress disorder(PTSD)is a serious stress-related disorder.AIM To identify the key genes and pathways to uncover the potential mechanisms of PTSD using bioinformatics methods.METHODS Gene expr...BACKGROUND Post-traumatic stress disorder(PTSD)is a serious stress-related disorder.AIM To identify the key genes and pathways to uncover the potential mechanisms of PTSD using bioinformatics methods.METHODS Gene expression profiles were obtained from the Gene Expression Omnibus database.The differentially expressed genes(DEGs)were identified by using GEO2R.Gene functional annotation and pathway enrichment were then conducted.The gene-pathway network was constructed with Cytoscape software.Quantitative real-time polymerase chain reaction(qRT-PCR)analysis was applied for validation,and text mining by Coremine Medical was used to confirm the connections among genes and pathways.RESULTS We identified 973 DEGs including 358 upregulated genes and 615 downregulated genes in PTSD.A group of centrality hub genes and significantly enriched pathways(MAPK,Ras,and ErbB signaling pathways)were identified by using gene functional assignment and enrichment analyses.Six genes(KRAS,EGFR,NFKB1,FGF12,PRKCA,and RAF1)were selected to validate using qRT-PCR.The results of text mining further confirmed the correlation among hub genes and the enriched pathways.It indicated that these altered genes displayed functional roles in PTSD via these pathways,which might serve as key signatures in the pathogenesis of PTSD.CONCLUSION The current study identified a panel of candidate genes and important pathways,which might help us deepen our understanding of the underlying mechanism of PTSD at the molecular level.However,further studies are warranted to discover the critical regulatory mechanism of these genes via relevant pathways in PTSD.展开更多

Tartary buckwheat(Fagopyrum tataricum)is an important pseudocereal feed crop with medicinal and nutritional value.Drought is one of the main causes of reduced growth and yield in these plants.We investigated the growt...Tartary buckwheat(Fagopyrum tataricum)is an important pseudocereal feed crop with medicinal and nutritional value.Drought is one of the main causes of reduced growth and yield in these plants.We investigated the growth,physiological,and metabolic responses of the widely promoted Tartary buckwheat variety Chuan Qiao No.1 to polyethylene glycol(PEG)-mediated drought stress.Drought significantly decreased shoot length,shoot biomass and relative water content.Root length,malondialdehyde content,electrolyte leakage,activities of superoxide dismutase,peroxidase,catalase and amylase,and contents of soluble sugar,soluble protein and proline were increased by PEG-mediated drought.Untargeted metabolomics analysis identified 32 core metabolites in seedlings subjected to PEG-mediated drought,16 of which increased—including quercetin,isovitexin,cyanidin 3-O-beta-D-glucoside,L-arginine,and glycerophosphocholine,while the other 16 decreased—including 3-methoxytyramine,2,6-diaminopimelic acid,citric acid,UDP-alpha-D-glucose,adenosine,keto-D-fructose.The 32 core metabolites were enriched in 29 metabolic pathways,including lysine biosynthesis,citrate(TCA)cycle,anthocyanin biosynthesis,and aminoacyl-tRNA biosynthesis.Among them,taurine and hypotaurine metabolism,flavor and flavor biosynthesis,indole alkaline biosynthesis,and alanine,aspartate and glutamate metabolism were the four main metabolic pathways affected by drought.Our findings provide new insights into the physiological and metabolic response mechanisms of Tartary buckwheat to drought stress.展开更多
OBJECTIVE Lychee seed,a famous traditional Chinese medicine,recently were reported to improve the learning and memory abilities in mice.However,it is still unclear whether lychee seed saponins(LSS)can improve the cogn...OBJECTIVE Lychee seed,a famous traditional Chinese medicine,recently were reported to improve the learning and memory abilities in mice.However,it is still unclear whether lychee seed saponins(LSS)can improve the cognitive function and associated mechanisms.METHODS In present studies,we established the Alzheimer disease(AD)model by injecting Aβ25-35 into the lateral ventricle of rats.Then the spatial learning and memory abilities of LSS-treated rats were evaluated with the Morris water maze,meanwhile the protein expressions of AKT,GSK3β and Tau in the hippocampal neuron were analyzed by immunohistochemistry and Western blotting.RESULTS The results showed LSS can improve the cognitive functions of AD rats through shortening the escape latency,increasing the number across the platform,platform quadrant dwell time and the percentage of the total distance run platform quadrant.The protein expression of AKT was significantly up-regulated and that of GSK3β and Tau were decreased remarkably in the hippocampal CA1 area.CONCLUSION Our study is the first to show that LSS significantly improve the cognitive function and prevent hippocampal neuronal injury of the rats with AD by activation of the PI3K/AKT/GSK3βsignaling pathway,suggesting LSS may be developed into the nutrient supplement for the treatment of AD.展开更多

Therapeutic principles and therapeutic methods for healing many diseases are recorded in these ancient TCM books, providing a huge number of references for modern TCM physicians on conducting diagnosis and administering treatment for different diseases. The ancient TCM books can be dated back thousands of years, and this vast knowledge is recorded in different medical books in the form of text. However, it is difficult to systematically assimilate much information in ancient TCM books. At present, many researchers are applying advanced analytical techniques to analyze the text data in the ancient TCM books. Advanced techniques that have been applied include database construction, cognitive linguistic analysis, fuzzy logic, data mining, and artificial intelligence(AI) technology. There are different characteristics in these advanced analytical techniques. In this study, we comprehensively review recent advances in these techniques applied to the study of ancient TCM books. Furthermore, as AI technology is increasingly utilized in the medical field as well as in the study of ancient TCM books, we also review the application of AI technology to the study of ancient TCM books.展开更多
Objective:The objective of this study was to systematically evaluate the clinical efficacy of the integration method of heat-clearing, dampness-excreting,spleen-strengthening, and stasis-removing from traditional Chin...Objective:The objective of this study was to systematically evaluate the clinical efficacy of the integration method of heat-clearing, dampness-excreting,spleen-strengthening, and stasis-removing from traditional Chinese medicine (TCM) combined with Western medicine for the treatment of ulcerative colitis(UC).Materials and Methods:The databases China National Knowledge Infrastructure,China Biology Medicinedisc(CBMdisc),WANFANG, VIP, and PubMed were searched for randomized controlled trials investigating the integration of the TCM methods of clearing heat,draining dampness, invigorating the spleen, and removing stasis, combined with Western medicine to treat UC from January 2009 to March 2019. Two reviewers independently conducted literature searches, screenings, data extractions, and literature bias evaluations. A meta-analysis was conducted using RevMan 5.3 and Stata 13.0 software. Results:In total, 15 studies involving 1289 patients were included. The results of the meta-analysis showed that the total effective rate of treatment in the experimental groups was higher than that of the control groups (relative risk [RR]=1.27, 95%confidence interval [CI]:1.21, 1.35, Z=8.74, P <0.00001). In the subgroup analysis, the total effective rate of oral TCM combined with Western medicine was higher than that of the control groups (RR=1.24, 95%CI:1.15, 1.33, Z=5.88, P <0.00001). The total effective rate of oral TCM with enemas combined with Western medicine was higher than that of the control group (RR=1.30, 95%CI:1.12, 1.50, Z=3.52, P=0.0004).The comparison between Western medicine alone and oral TCM combined with enteroscopy and Western medicine showed that the effective rate of enteroscopy (RR=1.18, 95%CI:1.05, 1.33, Z=2.86, P=0.004) and the symptom scores before and after treatment all improved more in the combined treatment groups than in those of the Western medicine group alone, with statistically significant differences (RR=-4.23, 95%CI:-4.93,-3.53, Z=11.84, P <0.00001). Conclusion:The integration of the TCM methods of heat clearing, dampness excreting, spleen strengthening, and stasis removing combined with Western medicine can significantly improve the cure rate of UC, and is an effective method to treat UC.展开更多

However, few studies have reported the possible differences in the effect of obesity on outcomes between older adults with metabolic abnormalities and those without metabolic abnormalities. Methods A total of 3485 older men were included from 2000 to 2014. All-cause mortality and cardiovascular mortality were obtained during a mean follow-up of five years Metabolic abnormalities were defined as having established hypertension, diabetes, or dyslipidemia and taking the disease-related medications. All participants were stratified by the presence or absence of metabolic abnormalities. Results In the non-metabolic abnormalities group, all-cause and cardiovascular deaths were lowest in overweight participants and highest in obese participants. In the metabolic abnormalities group, mortality was also lowest in overweight participants but highest in participants with normal weight. After adjustment for covariates, hazard ratios (95% CI) for all-cause death and cardiovascular death were 0.68 (0.51, 0.92) and 0.59 (0.37, 0.93), respectively, in overweight participants with metabolic abnormalities. Furthermore, obesity was not associated with mortality risk in both groups. These findings were unchanged in stratified analyses. Conclusions Overweight was negatively associated with mortality risk in older men with metabolic abnormalities but not in those without metabolic abnormalities. Obesity did not increase death risk regardless of metabolic abnormalities. These findings suggest that the recommendation of pursuing a normal weight may be wrong in overweight/obese older men, especially for those with metabolic abnormalities.展开更多

文摘BACKGROUND Prostate cancer(PCa)has high morbidity and mortality rates in elderly men.With a history of thousands of years,traditional Chinese medicine derived from insects could be an important source for developing cancer-targeted drugs to prevent tumorigenesis,enhance therapeutic effects,and reduce the risk of recurrence and metastasis.Multiple studies have shown that Coridius chinensis(Cc)has anticancer effects.AIM To elucidate the mechanism of action of Cc against PCa via network pharma-cology and molecular docking.METHODS Potential targets for Cc and PCa were predicted using ChemDraw 19.0 software,the PharmMapper database and the GeneCards database.Then,the STRING database was used to construct the protein–protein interaction network.Gene Ontology(GO),Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment and molecular docking analyses were subsequently conducted to identify the key targets,active ingredients and pathways involved.RESULTS GO and KEGG analyses indicated that the PI3K-Akt signalling pathway was the critical pathway(P value<1.0×10-8).Multiple targeting ingredients that can affect multiple pathways in PCa have been identified in Cc.Seven active compounds(asponguanosines A,asponguanine B,asponguanine C,aspong-pyrazine A,N-acetyldopamine,aspongadenine B and aspongpyrazine B)were selected for molecular docking with 9 potential targets,and the results revealed that aspongpyrazine A and asponguanosine A are the main components by which Cc affects PCa(affinity<-5 kcal/mol,hydrogen bonding),but more studies are needed.CONCLUSION We used network pharmacology to predict the bioactive components and important targets of Cc for the treatment of PCa,supporting the development of Cc as a natural anticancer agent.

文摘Accumulating data indicate that kidney tonifying recipe to treat depression patients may significantly reduce clinical symptoms, effectively improve the quality and prognosis of the patient's life. Clinical and experimental studies have suggested that tonifying kidney recipe has multiple targets, and thereby via multiple ways to accomplish antidepressant effect. The tonifying kidney recipe has been used in traditional Chinese medicine to control neurotrophic factor so as to promote the growth, differentiation, maturation of neurons. The tonifying kidney recipe participates in protection of neural plasticity, brain development, immune regulation, and a variety of other physiological processes to act as an antidepressant that has great similarity as vitamin D axis. In this article, we describe the current knowledge to introduct the relationship between kidney deficiency and vitamin D axis, and speculate about the molecular mechanism of tonifying kidney recipe in the treatment of depression by regulating vitamin D axis.

文摘This review aims to sum up how Non-coding RNAs(ncRNAs)regulate the development of periodontitis and provides a new perspective for understanding the pathogenesis of periodontitis.We explored the ncRNA's dual role in the development of periodontitis by summarizing evidence from previous in vivo and in vitro studies as well as clinical samples.In our review,the downregulation of 18 miRNAs,22 lncRNAs and 10 circRNAs demonstrates protective roles in periodontitis.In contrast,the expression of other 11 miRNAs,7 lncRNAs and 6 circRNAs are upregulated in periodontitis,which promote the progression of periodontitis.These dysregulated ncRNAs exert their protective or destructive roles by mainly influencing cell proliferation,differentiation and apoptosis via cross-talking with various molecules or signaling pathways.Our findings suggested which and how ncRNAs promote or delay the progression of periodontitis,which may greatly contribute to diagnose and therapy development of periodontitis based on ncRNAs in the future.

文摘Tartary buckwheat(Fagopyrum tataricum)is a dual-purpose medicinal and food crop grown for its high contents of functional compounds and abundant nutrients.Although studies have shown the differences of total flavonoid content in Tartary buckwheat at different ploidy levels,the composition of flavonoid and its regulatory mechanisms are largely unknown.In this study,the leaf metabolome and transcriptome of diploid and tetraploid accessions of Tartary buckwheat were analyzed to gain insight into the impact of polyploidization on comparative secondary metabolite composition and molecular regulatory mechanism.Based on a widely targeted metabolomics analysis,a total of 792 metabolites were identified,including 127 flavonoids.The accumulation of 127 metabolites and expression of 3871 genes differed significantly between diploid and tetraploid Tartary buckwheat.Integrated metabolomics and transcriptome analysis revealed that chromosome doubling up-regulated the expression of upstream genes in the flavonoid biosynthesis pathway to promote the accumulation of flavonoids.The present results contribute to elucidation of the molecular mechanism of phenotypic variation associated with polyploidy in Tartary buckwheat.The findings provide a reference for further studies on phenotypic traits in polyploid Tartary buckwheat,the cloning of crucial regulatory genes,and utilization of genetic engineering technologies in the breeding of Tartary buckwheat.

文摘Tongluojiunao (TLJN) is an herbal medicine consisting of two main components, geniposide and ginsenoside Rg1. TLJN has been shown to protect primary cultured hippocampal neurons. How-ever, its mechanism of action remains unclear. In the present study, primary cultured hippocampal neurons treated with Aβ1-42 (10 μmol/L) signiifcantly increased the release of lactate dehydroge-nase, which was markedly reduced by TLJN (2 μL/mL), speciifcally by the component geniposide (26 μmol/L), but not ginsenoside Rg1 (2.5 μmol/L). hTe estrogen receptor inhibitor, ICI182780 (1 μmol/L), did not block TLJN-or geniposide-mediated decrease of lactate dehydrogenase under Aβ1-42-exposed conditions. However, the phosphatidyl inositol 3-kinase or mitogen-activated protein kinase pathway inhibitor, LY294002 (50 μmol/L) or U0126 (10 μmol/L), respectively blo cked the decrease of lactate dehydrogenase mediated by TLJN or geniposide. hTerefore, these results suggest that the non-classical estrogen pathway (i.e., phosphatidyl inositol 3-kinase or mitogen-activated protein kinase) is involved in the neuroprotective effect of TLJN, speciifcally its component, geniposide, against Aβ1-42-mediated cell death in primary cultured hippocampal neurons.

文摘Traditional Chinese medicine(TCM)has been increasingly employed in the last decades in China for both preventing and treating a variety of cancers.3-epi-bufotalin is an active ingredient of TCM“Chanpi”with anti-tumor potential.However,the effect and mechanism of 3-epi-bufotalin on colorectal cancers were not well disclosed.The present study demonstrated that 3-epi-bufotalin could reduce viability,trigger apoptosis,and block the cell cycle at the G2/M stage in colorectal cancer cell lines HT29,RKO,and COLO205 in vitro.Moreover,3-epi-bufotalin inhibited the JAK1/STAT3 signaling pathway.These results indicated the anti-proliferation ability of 3-epi-bufotalin in colorectal cancer cells.

文摘Objective:To observe the clinical efficacy and safety of oral administration of the traditional Chinese herb rhubarb to treat acute exacerbation of chronic obstructive pulmonary disease(AECOPD).Method:This was a multicenter randomized double-blinded placebo controlled study that took place in 7 provinces of China that enrolled 244 patients(aged 18e80 years)who had acute exacerbation of COPD with the traditional Chinese syndrome pattern of phlegm-heat obstructing lung.Participants were divided into experimental and control groups.The experimental group received 4.5 g of rhubarb granules twice daily and the control group received placebo granules.Both groups also received conventional Western therapy consisting of oxygen therapy,an antibiotic,expectorant,and a bronchodilator.Treatment lasted 10 days.Symptom scores for cough,sputum volume and color,wheezing and chest tightness before treatment and on days 3,5,7,and 10 during the treatment were recorded.Lung function,arterial blood gas and levels of serum inflammatory factors,interleukin-4(IL-4),interleukin-8(IL-8),and interleukin-10(IL-10)and tumor necrosis factor-alpha(TNF-a),before and after treatment were measured.Results:The sample size of the full analysis set(FAS)was 244 participants,and the sample size of per protocol set(PPS)was 235.Following 10 days’treatment,symptom scores of the experimental group were markedly lower than those of the placebo group(FAS:mean difference1.67,95%CI:e2.66 to0.69,P Z 0.001;PPS:mean difference1.55,95%CI:2.56 to0.54,P Z 0.003).Lung function in the experimental group was significantly higher than in the placebo group(FEV1,FAS:mean difference 0.12,95%CI:0.06 to 0.18;P<0.001;PPS:mean difference 0.12,95%CI 0.05 to 0.18;P<0.001.FVC:FAS:mean difference 0.16,95%CI:0.06 to 0.26;P Z 0.002;PPS:mean difference 0.16,95%CI 0.05 to 0.26;P Z 0.003.FEV1%,FAS:mean difference 5.95,95%CI:3.36 to 8.53;P<0.001;PPS:mean difference 5.92,95%CI 3.28 to 8.56;P<0.001.).PaO2,PaCO2,as well as serum inflammatory factors were also improved when compared to the placebo group.There were no significant differences in the incidence rate of adverse reaction between the two groups.Conclusions:Compared with placebo,rhubarb granules significantly reduced symptom scores,improved blood oxygen level,controlled systemic inflammatory response,without significant adverse effects.Thus,rhubarb may be a beneficial adjuvant method for treating the phlegm-heat obstructing the lung syndrome pattern of AECOPD.

文摘Polyploid plants grow well,are stress tolerant,and are rich in nutrients and bioactive compounds.Thus,they are useful for improving crop quality and yield.In this study,we compared the seed characteristics and metabolite profiles of diploid and tetraploid tartary buckwheat,which was developed via an artificially induced chromosome doubling event.The length,width,area,and thousand-grain weight were greater for the tetraploid seeds than for the diploid seeds.However,the germination rate decreased for the tetraploid seeds.Additionally,there was a gap between the shell and kernel of the tetraploid seeds.Moreover,the water absorption rate was higher for the tetraploid than for the diploid seeds.Chromosome doubling increased the seed total flavonoid content and deepened the seed color.A principal component analysis of the ultrahigh-pressure liquid chromatography-high resolution mass spectrometry data revealed the clear separation between the diploid and tetraploid samples.An orthogonal partial least squares-discriminant analysis and other multivariate statistical analyses identified 83 differentially abundant compounds,with most of the flavonoid metabolites more abundant in the tetraploid than in the diploid seeds.Research on tartary buckwheat polyploidy may result in enhanced germplasm resources and may clarify the mechanism underlying the biosynthesis of bioactive compounds.

文摘Stress is an inevitable interference factor that seriously affects health.Listening to music is an economical,noninvasive,and highly accepted tool for easing stress.However,physiological studies investigating the ability of music to reduce stress in daily life are limited.We established rat models of chronic immobilization stress(CIS)to observe changes in the hypothalamic arcuate nucleus(ARC)neurons involved in the regulation of food intake and the effect of comfortable classical music exposure.Twenty-one days of stress resulted in decreased food intake and delayed body weight gain,up-regulation of leptin receptor(Ob-R),cocaine-and amphetamine-regulated transcript(CART),proopiomelanocortin(POMC),and alpha-melanocyte-stimulating hormone(α-MSH)expression,and downregulation of neuropeptide Y(NPY)and agouti-related protein(AgRP)expression in the ARC.Thus,peripheral leptin entered the ARC under chronic stress conditions,bound to Ob-R,and affected downstream nerve pathways related to appetite,such as the NPY/AgRP and CART/POMC pathways.Gentle classical music played at 65 dB reversed the abnormal expression of Ob-R and NPY induced by chronic stress.Thus,listening to comfortable music improves changes in ARC neurons related to the regulation of food intake in CIS rats,and these results provide a reference for basic research regarding how music therapy alleviates stress and stress-related health issues.
文摘Pdx-1, an important transcription factor highlighting in the early pancreatic development, islet functions and pancreatic disorders, needs to be more investigated in zebrafish, and siRNA is still seldom applied in zebrafish embryo-related research.Our aim was to explore the role of pdx-l in pancreatic development of zebrafish embryos by using siRNA approach. Microinjection, reverse transcriptase-PCR (RT-PCR), in situ hybridization and immunofluorescent staining were used in this research, and the morphology of the islet in normal zebrafish embryos, and in those treated with the siRNA specific to pdx-1 (siPDX-l) or siGFP was observed and compared. The expression of pdx-1 was detected in the stages of 1-cell, 2-cell, 4-cell, 8-cell, 16-cell, 16-hour by RT-PCT. The in situ hybridization and immunofluorescent staining results showed that siPDX-1 disturbed the formation of the islet in zebrafish embryos. Pdx-1 played multiple roles in maintaining the phenotype of the islet during embryogenesis in zebrafish.
文摘Acrosin binding protein(ACRBP)is restrictedly expressed in sperm acrosomes and involved in many important reproductive processes.Here we investigated the immunocontraceptive effects of ACR BP peptide-based vaccines.B-cell epitope vaccines of mouse ACRBP were predicted by bioinformatics and prepared by vector coupling and adjuvant combination.Subsequently,female mice were actively immunized with the epitope vaccines subcutaneously,and the titers and specificity of anti-ACRBP antibodies were detected.Then the efficancy,safety and reversibility of epitope vaccines for contraception were evaluated in vivo and in vitro.

文摘BACKGROUND Post-traumatic stress disorder(PTSD)is a serious stress-related disorder.AIM To identify the key genes and pathways to uncover the potential mechanisms of PTSD using bioinformatics methods.METHODS Gene expression profiles were obtained from the Gene Expression Omnibus database.The differentially expressed genes(DEGs)were identified by using GEO2R.Gene functional annotation and pathway enrichment were then conducted.The gene-pathway network was constructed with Cytoscape software.Quantitative real-time polymerase chain reaction(qRT-PCR)analysis was applied for validation,and text mining by Coremine Medical was used to confirm the connections among genes and pathways.RESULTS We identified 973 DEGs including 358 upregulated genes and 615 downregulated genes in PTSD.A group of centrality hub genes and significantly enriched pathways(MAPK,Ras,and ErbB signaling pathways)were identified by using gene functional assignment and enrichment analyses.Six genes(KRAS,EGFR,NFKB1,FGF12,PRKCA,and RAF1)were selected to validate using qRT-PCR.The results of text mining further confirmed the correlation among hub genes and the enriched pathways.It indicated that these altered genes displayed functional roles in PTSD via these pathways,which might serve as key signatures in the pathogenesis of PTSD.CONCLUSION The current study identified a panel of candidate genes and important pathways,which might help us deepen our understanding of the underlying mechanism of PTSD at the molecular level.However,further studies are warranted to discover the critical regulatory mechanism of these genes via relevant pathways in PTSD.

文摘Tartary buckwheat(Fagopyrum tataricum)is an important pseudocereal feed crop with medicinal and nutritional value.Drought is one of the main causes of reduced growth and yield in these plants.We investigated the growth,physiological,and metabolic responses of the widely promoted Tartary buckwheat variety Chuan Qiao No.1 to polyethylene glycol(PEG)-mediated drought stress.Drought significantly decreased shoot length,shoot biomass and relative water content.Root length,malondialdehyde content,electrolyte leakage,activities of superoxide dismutase,peroxidase,catalase and amylase,and contents of soluble sugar,soluble protein and proline were increased by PEG-mediated drought.Untargeted metabolomics analysis identified 32 core metabolites in seedlings subjected to PEG-mediated drought,16 of which increased—including quercetin,isovitexin,cyanidin 3-O-beta-D-glucoside,L-arginine,and glycerophosphocholine,while the other 16 decreased—including 3-methoxytyramine,2,6-diaminopimelic acid,citric acid,UDP-alpha-D-glucose,adenosine,keto-D-fructose.The 32 core metabolites were enriched in 29 metabolic pathways,including lysine biosynthesis,citrate(TCA)cycle,anthocyanin biosynthesis,and aminoacyl-tRNA biosynthesis.Among them,taurine and hypotaurine metabolism,flavor and flavor biosynthesis,indole alkaline biosynthesis,and alanine,aspartate and glutamate metabolism were the four main metabolic pathways affected by drought.Our findings provide new insights into the physiological and metabolic response mechanisms of Tartary buckwheat to drought stress.

文摘OBJECTIVE Lychee seed,a famous traditional Chinese medicine,recently were reported to improve the learning and memory abilities in mice.However,it is still unclear whether lychee seed saponins(LSS)can improve the cognitive function and associated mechanisms.METHODS In present studies,we established the Alzheimer disease(AD)model by injecting Aβ25-35 into the lateral ventricle of rats.Then the spatial learning and memory abilities of LSS-treated rats were evaluated with the Morris water maze,meanwhile the protein expressions of AKT,GSK3β and Tau in the hippocampal neuron were analyzed by immunohistochemistry and Western blotting.RESULTS The results showed LSS can improve the cognitive functions of AD rats through shortening the escape latency,increasing the number across the platform,platform quadrant dwell time and the percentage of the total distance run platform quadrant.The protein expression of AKT was significantly up-regulated and that of GSK3β and Tau were decreased remarkably in the hippocampal CA1 area.CONCLUSION Our study is the first to show that LSS significantly improve the cognitive function and prevent hippocampal neuronal injury of the rats with AD by activation of the PI3K/AKT/GSK3βsignaling pathway,suggesting LSS may be developed into the nutrient supplement for the treatment of AD.

文摘The ancient books on traditional Chinese medicine(TCM) are the source of knowledge for TCM physicians. Therapeutic principles and therapeutic methods for healing many diseases are recorded in these ancient TCM books, providing a huge number of references for modern TCM physicians on conducting diagnosis and administering treatment for different diseases. The ancient TCM books can be dated back thousands of years, and this vast knowledge is recorded in different medical books in the form of text. However, it is difficult to systematically assimilate much information in ancient TCM books. At present, many researchers are applying advanced analytical techniques to analyze the text data in the ancient TCM books. Advanced techniques that have been applied include database construction, cognitive linguistic analysis, fuzzy logic, data mining, and artificial intelligence(AI) technology. There are different characteristics in these advanced analytical techniques. In this study, we comprehensively review recent advances in these techniques applied to the study of ancient TCM books. Furthermore, as AI technology is increasingly utilized in the medical field as well as in the study of ancient TCM books, we also review the application of AI technology to the study of ancient TCM books.

文摘Objective:The objective of this study was to systematically evaluate the clinical efficacy of the integration method of heat-clearing, dampness-excreting,spleen-strengthening, and stasis-removing from traditional Chinese medicine (TCM) combined with Western medicine for the treatment of ulcerative colitis(UC).Materials and Methods:The databases China National Knowledge Infrastructure,China Biology Medicinedisc(CBMdisc),WANFANG, VIP, and PubMed were searched for randomized controlled trials investigating the integration of the TCM methods of clearing heat,draining dampness, invigorating the spleen, and removing stasis, combined with Western medicine to treat UC from January 2009 to March 2019. Two reviewers independently conducted literature searches, screenings, data extractions, and literature bias evaluations. A meta-analysis was conducted using RevMan 5.3 and Stata 13.0 software. Results:In total, 15 studies involving 1289 patients were included. The results of the meta-analysis showed that the total effective rate of treatment in the experimental groups was higher than that of the control groups (relative risk [RR]=1.27, 95%confidence interval [CI]:1.21, 1.35, Z=8.74, P <0.00001). In the subgroup analysis, the total effective rate of oral TCM combined with Western medicine was higher than that of the control groups (RR=1.24, 95%CI:1.15, 1.33, Z=5.88, P <0.00001). The total effective rate of oral TCM with enemas combined with Western medicine was higher than that of the control group (RR=1.30, 95%CI:1.12, 1.50, Z=3.52, P=0.0004).The comparison between Western medicine alone and oral TCM combined with enteroscopy and Western medicine showed that the effective rate of enteroscopy (RR=1.18, 95%CI:1.05, 1.33, Z=2.86, P=0.004) and the symptom scores before and after treatment all improved more in the combined treatment groups than in those of the Western medicine group alone, with statistically significant differences (RR=-4.23, 95%CI:-4.93,-3.53, Z=11.84, P <0.00001). Conclusion:The integration of the TCM methods of heat clearing, dampness excreting, spleen strengthening, and stasis removing combined with Western medicine can significantly improve the cure rate of UC, and is an effective method to treat UC.

However, few studies have reported the possible differences in the effect of obesity on outcomes between older adults with metabolic abnormalities and those without metabolic abnormalities. Methods A total of 3485 older men were included from 2000 to 2014. All-cause mortality and cardiovascular mortality were obtained during a mean follow-up of five years Metabolic abnormalities were defined as having established hypertension, diabetes, or dyslipidemia and taking the disease-related medications. All participants were stratified by the presence or absence of metabolic abnormalities. Results In the non-metabolic abnormalities group, all-cause and cardiovascular deaths were lowest in overweight participants and highest in obese participants. In the metabolic abnormalities group, mortality was also lowest in overweight participants but highest in participants with normal weight. After adjustment for covariates, hazard ratios (95% CI) for all-cause death and cardiovascular death were 0.68 (0.51, 0.92) and 0.59 (0.37, 0.93), respectively, in overweight participants with metabolic abnormalities. Furthermore, obesity was not associated with mortality risk in both groups. These findings were unchanged in stratified analyses. Conclusions Overweight was negatively associated with mortality risk in older men with metabolic abnormalities but not in those without metabolic abnormalities. Obesity did not increase death risk regardless of metabolic abnormalities. These findings suggest that the recommendation of pursuing a normal weight may be wrong in overweight/obese older men, especially for those with metabolic abnormalities.
