Objectives This paper aims to investigate the uterotrophic activities of lactational exposure to combination of soy isoflavones (SIF) and bisphenol A (BPA) and to examine estrogen receptor α (ERα) and estrogen receptor β (ERβ) expressions in hypothalamus-pituitary-ovary axis and uterus.Methods Maternal rats that were breeding about 8 litters were randomly divided into four groups with seven dams in each group.Dams in different treatment groups received corn oil (control),150 mg/kg BW of SIF,150 mg/kg BW of BPA or combination of 150 mg/kg BW of SIF and 150 mg/kg BW of BPA,respectively,from postnatal day 5 to 11 (PND5-11) by gavage.On PND12 and PND70,10 female litters were killed and hypothalamus,pituitary,ovary and uterus were collected.ERα and ERβ expressions in these organs were detected with Western blotting assay.And vaginal opening time and estrus cycle were examined in animals fed for PND70.Results On PND12,the relative uterine weight of rats treated with ISF or BPA or their combination was significantly higher than that of untreated rats (P〈0.05).But the relative uterine weight of rats in the co-exposure group was slightly lower than that in the group only exposed to SIF or BPA.On PND 70,however,the relative uterine weight in each treatment group was not statistically different from that in the control group (P〈0.05).Vaginal opening time and estrus cycle in groups treated with SIF or BPA or their combination were similar to those in the control group (P〈0.05).Exposure to SIF or BPA or their combination could up-regulate or down-regulate ERα and ERβ expressions in hypothalamus,pituitary,ovary and uterus on PND12 and PND70.These regulation patterns for ERα and ERβ were different in different organs at different time points.Conclusion Lactational exposure to ISF or BPA or their combination could induce uterotrophic responses in neonate rats,which disappeared in later life.But these data fail to suggest a possibility for synergic actions between SIF and BPA.It was also demonstrated that the uterotrophic effects of SIF and BPA exposure might,at least,involve modification of ERα or ERβ expressions in the hypothalamus-pituitary-ovary axis. 展开更多

An environmental capacity model for the petroleum hydrocarbon pollutions (PHs) in Jiaozhou Bay is constructed based on field surveys, mesocosm, and parallel laboratory experiments. Simulated results of PHs seasonal successions in 2003 match the field surveys of Jiaozhou Bay resaonably well with a highest value in July. The Monte Carlo analysis confirms that the variation of PHs concentration significantly correlates with the river input. The water body in the bay is reasonably subjected to self-purification processes, such as volatilization to the atmosphere, biodegradation by microorganism, and transport to the Yellow Sea by water exchange. The environmental capacity of PHs in Jiaozhou Bay is 1500 tons per year IF the seawater quality criterion (Grade Ⅰ/Ⅱ, 0.05 mgLl) in the region is to be satisfied. The contribution to self-purification by volatilization, biodegradation, and transport to the Yellow Sea accounts for 48%, 28%, and 23%, respectively, which make these three processes the main ways of PHs purification in Jiaozhou Bay. 展开更多

Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) was applied in the detection of the end point temperature (EPT) of thermal denatured protein in fish and meat in this study. It was also used in studying the thermal denatured temperature range of proteins in salmon and chicken meat. The results show that the temperature ranges of denatured proteins were from 65 ℃ to 75 ℃ , and these temperature ranges were influenced by the processing methods. Through SDS-PAGE, the features of repeated heating thermal denatured proteins under the same temperature and processing time were studied. The electrophoresis patterns of thermal denatured proteins determined through repeated heating at the same temperature did not exhibit any change. For the detection of cooked fish and meat samples, they were subjected to applying the SDS-PAGE method, which revealed an EPT ranging from 60 ℃ to 80 ℃ . 展开更多

Vibrio anguillarum is an important bacterial pathogen of aquatic organisms and a significant problem in aquatic farming. The rapid detection and identification of V. anguillarum, and other pathogens that infect marine organisms, is crucial to effective disease management. In this study, we developed a loop-mediated amplification (LAMP) assay to detect V. anguillarum in an hour in a single tube without the need for thermal cycling. Conserved regions of the metalloproteinase (empA) gene of V. anguillarum served as the targets for primer design. A fragment of the empA gene was amplified at 65℃ in the presence of the primer mixture and Bst DNA polymerase. In the optimized LAMP assay, 6.7 pg of V. anguillarum DNA could be detected. Six strains of V. anguillarum and 17 strains of non-V, anguillarum bacteria were used in this study to evaluate the species specificity of the primers. The six V. anguillarum strains gave a positive result in the LAMP assay. This method was also validated in V. anguillarum-infected fish. This LAMP method is more sensitive than PCR in the detection of V. anguillarum and shows good species specificity. The LAMP assay is therefore an effective method for the quick detection of V. anguillarum both in the laboratory and in the field. 展开更多

MicroRNAs (miRNAs) are a class of newly identified, small, non-coding RNAs that play vital roles in regulation. Based on miRNAs unique features of expression pattern, evolutionary conservation, secondary structure and genetic requirements for biogenesis, computational predication strategy is adopted to predicate the novel miRNAs. In this research, potential miRNAs and their targets in grapevine (Vitis vinifera) were predicted. We used previously known plant miRNAs against grapevine genome sequence databases to search for potential miRNAs. A total of 81 potential miRNAs were detected following a range of strict filtering criteria. Using these potential miRNA sequences, we could further blast the mRNA database to find the potential targets in this species. Comparative analysis of miRNAs in grapevine and other species reveals that miRNAs exhibit an evolutional conservation, the number and function of miRNAs must have significantly expanded during the evolution of land plants. Furthermore divergence made versatile functions of miRNAs feasible. Cluster of miRNAs likely represents an ancient expression mechanism. Predicted target genes include not only transcription factors but also genes implicated in floral development, signal transduction, diseases and stress response. Till now, little is known about experimental or computational identification of miRNA in grapevine species. Increased knowledge of the biological mechanisms of the grapevine will allow targeted approaches to increase the quality of fruit and reduce the impact of parasites together with stress, which could enable a sustainable, environmentally-sound, farming policv. 展开更多

The aim is to develop a liquid chip technique to detect Taura syndrome virus( TSV) and yellow head disease virus( YHDV) on Penaeus orientalis simultaneously. The CP2 gene of TSV and N gene of YHDV in Gen Bank was analysed by using the software DNAStar 7. 0 to design the TSV-and YHDV-specific primers. The primers were labeled with biotin and subjected to amination modification. They were then coupled with fluorescence-coded microspheres and then used for hybridization with RT- PCR products of TSV and YHDV. The liquid chip detection technique for detection of TSV and YHDV was established by using BD FACSArray to detect fluorescence signal in the reaction system. This assay system had a high sensitivity to TSV and YHDV,with the detection of limit of 100 pg. Moreover,the assay was specific for the detection of TSV,YHDV and was not susceptible to cross with other viruses,including white spot syndrome virus( WSSV),spring viremia of carp virus( SVCV),infectious haematopoietic necrosis virus( IHNV). In conclusion,the liquid chip assay technique established in this study is highly sensitive and specific to TSV and YHDV detection. [Result] Using ATP bioluminescence magnetic enzyme immunoassay technique to detect standard solution for Shigella (ATCC 25931 ), result showed that correlation coefficient between relative light intensity detected by instrument and bacteria concentration detec- ted by culture counting method was 0.981 1. Moreover, relation curve between relative light intensity and Shigella concentration was drawn. This method was effective to detect artificially contaminated pet food. A new technique is introduced in this paper regarding red tide recognition with remotely sensed hyper-spectral images based on empirical mode decomposition (EMD), from an artificial red tide experiment in the East China Sea in 2002. A set of characteristic parameters that describe absorbing crest and reflecting crest of the red tide and its recognition methods are put forward based on general pictre data, with which the spectral information of certain non-dominant alga species of a red tide occurrence is analyzed for establishing the foundation to estimate the species. Comparative experiments have proved that the method is effective. Meanwhile, the transitional area between red-tide zone and non-red-tide zone can be detected with the information of thickness of algae influence, with which a red tide can be forecast. 展开更多

Fox is an important economic fur animal with a very promising prospect. The fox mitochondrial COl gene-specific primers and probes were used to opti- mize and develop a real time fluorescence PCR system for rapid detection of fox-derived components. The results showed that the real time fluorescence PCR gave a very high sensitivity, with the detection limit as low as 12 fg DNA, and it did not present any cross reaction with other common species like pig, cow, sheep, dog, soybean and maize. It could be concluded that this method can be used as an effective method for detecting the fox-derived components in food and feedstuff, and can also be applied for distinguishing the fox fur from adulterants. 展开更多

The extracting process and parameters optimization of Eulaliopsis binata fibers were studied.The components of Eulaliopsis binata were tested,the cellulose contents was about 45%,and the lignin content was as high as 18.5%.And the components of Eulaliopsis binata fibers were compared with some bast fibers.The analysis of physical and chemical properties related to acid,alkali,and auxiliary agent about cellulose and pectin,and lignin components in Eulaliopsis binata fibers were investigated.The chemical processing was employed to extract Eulaliopsis binata fibers in which NaOH as cooking agent and Na5P3O10 as auxiliary agent.The universal rotatable composite experiment design was used to get the highly precise regression equations,in which the NaOH concentration,the auxiliary agent Na5P3O10 concentration and the processing time were the main technological parameters.The residual gum content,fiber fineness,fiber strength,and fiber length of Eulaliopsis binata were the evaluation indexes.Based on the relation among parameters of extracting process,degumming effect and quality indexs of Eulaliopsis binata fibers,the optimum parameters of extracting process on Eulaliopsis binata fibers can be obtained.The fibers extracted from Eulaliopsis binata can be developed as one kind of natural cellulose fibers suitable for producing various textile products. 展开更多

[Objective] The paper was to establish pyrosequencing methods for detecting viral hemorrhagic septicemia virus (VHSV). [ Method ] One pair of PCR primers and one pyrosequencing primer of VHSV were designed. The pyrosequencing reaction system and conditions were optimized and the pyrosequencing method for detecting VHSV was established. [ Result] This method was only able to specifically detect the objective viruses in the eight fish viruses, and the method had the advantage of high sensitivity. The minimum detectable limit of nucleic acid was 82 copies/μL. The method was verified by detecting VHSV in 1 924 batches of samples collected from domestic and imported fishes. The detection results were consistent with that of traditional RT-PCR, and the specificity and sensitivity of the method could meet the detection requirement for aquatic animal diseases. [ Conclusion] The study provides a new detection method for monitoring and prevention and control of aquatic animal virus diseases. 展开更多
